Abstract Oxepinone rings represent one of structurally unusual motifs of natural products and the biosynthesis of oxepinones is not fully understood. 1,5-Seco-vibralactone (3) features an oxepinone motif and is a stable metabolite isolated from mycelial cultures of the mushroom Boreostereum vibrans. Cyclization of 3 forms vibralactone (1) whose β-lactone-fused bicyclic core originates from 4-hydroxybenzoate, yet it remains elusive how 4-hydroxybenzoate is converted to 3 especially for the oxepinone ring construction in the biosynthesis of 1. In this work, using activity-guided fractionation together with proteomic analyses, we identify an NADPH/FAD-dependent monooxygenase VibO as the key enzyme performing a crucial ring-expansive oxygenation on the phenol ring to generate the oxepin-2-one structure of 3. The crystal structure of VibO reveals that it forms a dimeric phenol hydroxylase-like architecture featured with a unique substrate-binding pocket adjacent to the bound FAD. Computational modeling and solution studies provide insight into the likely VibO active site geometry, and suggest possible involvement of a flavin-C4a-OO(H) intermediate.

In the granite regions of southern China, benggang poses a substantial threat to the ecological environment due to significant soil erosion. This phenomenon also imposes constraints on economic development, necessitating substantial investments in restoration efforts in recent decades. Despite these efforts, there remains a notable gap in comprehensive risk assessment that integrates both the erosion risk and disaster risk associated with benggang. This study focuses on a representative benggang area in Wuhua County, Guangdong province, employing transformer methods and high-resolution imagery to map the spatial pattern of the benggang. The integrated risk of benggang was assessed by combining soil-erosion risk and disaster risk, and cultivated land, residential land, and water bodies were identified as key disaster-affected entities. The machine-learning Segformer model demonstrated high precision, achieving an Intersection over Union (IoU) of 93.17% and an accuracy (Acc) of 96.73%. While the number of large benggang is relatively small, it constitutes the largest area proportion (65.10%); the number of small benggang is more significant (62.40%) despite a smaller area proportion. Prioritization for benggang management is categorized into high, medium, and low priority, accounting for 17.98%, 48.34%, and 33.69%, respectively. These priorities cover areas of 30.27%, 42.40%, and 27.33%, respectively. The findings of this study, which offer benggang management priorities, align with the nature-based solutions approach. Emphasizing the importance of considering costs and benefits comprehensively when formulating treatment plans, this approach contributes to sustainable solutions for addressing the challenges posed by benggang.

This study presents a biosensor fabricated based on integrated passive device (IPD) technology to measure microbial growth on solid media in real-time. Yeast (Pichia pastoris, strain GS115) is used as a model organism to demonstrate biosensor performance. The biosensor comprises an interdigital capacitor in the center with a helical inductive structure surrounding it. Additionally, 12 air bridges are added to the capacitor to increase the strength of the electric field radiated by the biosensor at the same height. Feasibility is verified by using a capacitive biosensor, and the change in capacitance values during the capacitance detection process with the growth of yeast indicates that the growth of yeast can induce changes in electrical parameters. The proposed IPD-based biosensor is used to measure yeast drop-added on a 3 mm medium for 100 h at an operating frequency of 1.84 GHz. The resonant amplitude of the biosensor varies continuously from 24 to 72 h due to the change in colony height during vertical growth of the yeast, with a maximum change of 0.21 dB. The overall measurement results also fit well with the Gompertz curve. The change in resonant amplitude between 24 and 72 h is then analyzed and reveals a linear relationship with time with a coefficient of determination of 0.9844, indicating that the biosensor is suitable for monitoring yeast growth. Thus, the proposed biosensor is proved to have potential in the field of microbial proliferation detection.

Spinal fusion, a common surgery performed for degenerative lumbar conditions, often uses recombinant human bone morphogenetic protein 2 (rhBMP-2) that is associated with adverse effects. Mesenchymal stromal/stem cells (MSCs) and their extracellular vesicles (EVs), particularly exosomes, have demonstrated efficacy in bone and cartilage repair. However, the efficacy of MSC exosomes in spinal fusion remains to be ascertained. This study investigates the fusion efficacy of MSC exosomes delivered via an absorbable collagen sponge packed in a poly Ɛ-caprolactone tricalcium phosphate (PCL–TCP) scaffold in a rat posterolateral spinal fusion model. Herein, it is shown that a single implantation of exosome-supplemented collagen sponge packed in PCL–TCP scaffold enhanced spinal fusion and improved mechanical stability by inducing bone formation and bridging between the transverse processes, as evidenced by significant improvements in fusion score and rate, bone structural parameters, histology, stiffness, and range of motion. This study demonstrates for the first time that MSC exosomes promote bone formation to enhance spinal fusion and mechanical stability in a rat model, supporting its translational potential for application in spinal fusion.

Chalkophomycin is a novel chalkophore with antibiotic activities isolated from Streptomyces sp. CB00271, while its potential in studying cellular copper homeostasis makes it an important probe and drug lead. The constellation of N-hydroxylpyrrole, 2H-oxazoline, diazeniumdiolate, and methoxypyrrolinone functional groups into one compact molecular architecture capable of coordinating cupric ions draws interest to unprecedented enzymology responsible for chalkophomycin biosynthesis. To elucidate the biosynthetic machinery for chalkophomycin production, the chm biosynthetic gene cluster from S. sp. CB00271 was identified, and its involvement in chalkophomycin biosynthesis was confirmed by gene replacement. The chm cluster was localized to a ~31 kb DNA region, consisting of 19 open reading frames that encode five nonribosomal peptide synthetases (ChmHIJLO), one modular polyketide synthase (ChmP), six tailoring enzymes (ChmFGMNQR), two regulatory proteins (ChmAB), and four resistance proteins (ChmA′CDE). A model for chalkophomycin biosynthesis is proposed based on functional assignments from sequence analysis and structure modelling, and is further supported by analogy to over 100 chm-type gene clusters in public databases. Our studies thus set the stage to fully investigate chalkophomycin biosynthesis and to engineer chalkophomycin analogues through a synthetic biology approach.

Background: Exploring the neural encoding mechanism and decoding of motion state switching during flight can advance our knowledge of avian behavior control and contribute to the development of avian robots. However, limited acquisition equipment and neural signal quality have posed challenges, thus we understand little about the neural mechanisms of avian flight. Methods: We used chronically implanted micro-electrode arrays to record the local field potentials (LFPs) in the formation reticularis medialis mesencephali (FRM) of pigeons during various motion states in their natural outdoor flight. Subsequently, coherence-based functional connectivity networks under different bands were constructed and the topological features were extracted. Finally, we used a support vector machine model to decode different flight states. Results: Our findings indicate that the gamma band (80–150 Hz) in the FRM exhibits significant power for identifying different states in pigeons. Specifically, the avian brain transmitted flight related information more efficiently during the accelerated take-off or decelerated landing states, compared with the uniform flight and baseline states. Finally, we achieved a best average accuracy of 0.86 using the connectivity features in the 80–150 Hz band and 0.89 using the fused features for state decoding. Conclusions: Our results open up possibilities for further research into the neural mechanism of avian flight and contribute to the understanding of flight behavior control in birds.

Since different quantities of white blood cells (WBCs) in solution possess an adaptive osmotic pressure of cells, the WBCs themselves and in solution have similar concentrations, resulting in them having similar dielectric properties. Therefore, a microwave sensor could have difficulty in sensing the quantity variation when WBCs are in solution. This paper presents a highly sensitive, linear permittivity-inspired microwave biosensor for WBCs, counting through the evaporation method. Such a measurement method is proposed to record measurements after the cell solution is dripped onto the chip and is completely evaporated naturally. The proposed biosensor consists of an air-bridged asymmetric differential inductor and a centrally located circular fork-finger capacitor fabricated on a GaAs substrate using integrated passive fabrication technology. It is optimized to feature a larger sensitive area and improved Q-factor, which increases the effective area of interaction between cells and the electromagnetic field and facilitates the detection of their changes in number. The sensing relies on the dielectric properties of the cells and the change in the dielectric constant for different concentrations, and the change in resonance properties, which mainly represents the frequency shift, corresponds to the macroscopic change in the concentration of the cells. The microwave biosensors are used to measure biological samples with concentrations ranging from 0.25 × 106 to 8 × 106 cells per mL in a temperature (26.00 ± 0.40 °C) and humidity (54.40 ± 3.90 RH%) environment. The measurement results show a high sensitivity of 25.06 Hz/cells·mL−1 with a highly linear response of r2 = 0.99748. In addition, a mathematical modeling of individual cells in suspension is performed to estimate the dielectric constant of individual cells and further explain the working mechanism of the proposed microwave biosensor.

Tobacco is an ideal model plant in scientific research. G-quadruplex is a guanine-rich DNA structure, which regulates transcription and translation. In this study, the prevalence and potential function of G-quadruplexes in tobacco were systematically analyzed. In tobacco genomes, there were 2,924,271,002 G-quadruplexes in the nuclear genome, 430,597 in the mitochondrial genome, and 155,943 in the chloroplast genome. The density of the G-quadruplex in the organelle genome was higher than that in the nuclear genome. G-quadruplexes were abundant in the transcription regulatory region of the genome, and a difference in G-quadruplex density in two DNA strands was also observed. The promoter of 60.4% genes contained at least one G-quadruplex. Compared with up-regulated differentially expressed genes (DEGs), the G-quadruplex density in down-regulated DEGs was generally higher under drought stress and salt stress. The G-quadruplex formed by simple sequence repeat (SSR) and its flanking sequence in the promoter region of the NtBBX (Nitab4.5_0002943g0010) gene might enhance the drought tolerance of tobacco. This study lays a solid foundation for further research on G-quadruplex function in tobacco and other plants.

Pigeons have natural advantages in robotics research, including a wide range of activities, low energy consumption, good concealment performance, strong long-distance weight bearing and continuous flight ability, excellent navigation, and spatial cognitive ability, etc. They are typical model animals in the field of animal robot research and have important application value. A hot interdisciplinary research topic and the core content of pigeon robot research, altering pigeon motor behavior using brain stimulation involves multiple disciplines including animal ethology, neuroscience, electronic information technology and artificial intelligence technology, etc. In this paper, we review the progress of altering pigeon motor behavior using brain stimulation from the perspectives of the neural basis and neuro-devices. The recent literature on altering pigeon motor behavior using brain stimulation was investigated first. The neural basis, structure and function of a system to alter pigeon motor behavior using brain stimulation are briefly introduced below. Furthermore, a classified review was carried out based on the representative research achievements in this field in recent years. Our summary and discussion of the related research progress cover five aspects including the control targets, control parameters, control environment, control objectives, and control system. Future directions that need to be further studied are discussed, and the development trend in altering pigeon motor behavior using brain stimulation is projected.

Deep eutectic solvents (DESs) are novel solvents with physicochemical properties similar to those of ionic liquids, and they have attracted extensive attention for the extraction of bioactive compounds from different plant materials in the context of green chemistry and sustainable development. In this study, seven DESs with different polarities were explored as green extraction solvents for cembratrien-diols (CBT-diols) from waste tobacco flowers. The best solvent, DES-3 (choline chloride: lactic acid (1:3)), which outperformed conventional solvents (methanol, ethanol, and ethyl acetate), was selected and further optimized for microwave-assisted DES extraction using the response surface methodology. The maximum yield of CBT-diols (6.23 ± 0.15 mg/g) was achieved using a microwave power of 425 W, microwave time of 32 min, solid/liquid ratio of 20 mg/mL, and microwave temperature of 40 °C. Additionally, the isolated CBT-diols exhibited strong antimicrobial activity against Salmonella, Staphylococcus aureus, Escherichia coli, Bacillus subtilis, and Pseudomonas aeruginosa and antitumor activity in the human liver cancer HepG2 and SMMC-7721 cell lines. This study highlights the feasibility of recovering CBT-diols from tobacco flower waste using DESs and provides opportunities for potential waste management using green technologies.

Unreasonable cultivation methods and management measures have led to widespread obstacles in tobacco continuous cropping in planting areas, resulting in reduced tobacco yield and soil degradation. Therefore, intercropping tobacco with other crops is an effective strategy to improve continuous cropping barriers. In this study, flue-cured tobacco NC102 and conventional planting varieties of Salvia miltiorrhiza were used as materials, and four treatments of flue-cured tobacco monoculture (CK), flue-cured tobacco, and Salvia miltiorrhiza at a ratio of 1:1 (TS11), 2:2 (TS22), and 2:3 (TS23), respectively, were set up to study their effects on soil microorganisms, physical and chemical properties, and yield and quality of flue-cured tobacco. The results showed that intercropping Salvia miltiorrhiza increased the number of soil bacteria and actinomycetes, decreased the number of fungi, and increased the activity of urease and sucrase. The content of available nitrogen and available phosphorus in intercropping Salvia miltiorrhiza soil was significantly higher than that of the flue-cured tobacco monoculture, while the content of available potassium was lower than that of the flue-cured tobacco monoculture. The soil environment was more conducive to the growth of flue-cured tobacco. Compared with the flue-cured tobacco monoculture, the proportion of superior tobacco in intercropping Salvia miltiorrhiza increased by 2.2–3.4%, and the ratio of potassium to chlorine in leaves of different parts of flue-cured tobacco increased by 12.3–75.0%. The content of total sugar and soluble sugar in middle and upper leaves of intercropping flue-cured tobacco was higher than that of the flue-cured tobacco monoculture, which improved the quality of flue-cured tobacco. From the analysis of the chemical composition of tobacco leaves, TS11 (flue-cured tobacco and Salvia miltiorrhiza intercropping row ratio of 1:1) had the best treatment effect, potassium content, total sugar, reducing sugar content, and potassium chloride ratio of flue-cured tobacco were the highest, the chlorine content was the lowest, and the quality was better than other treatments. From the analysis of total output value, the total output value of TS22 (flue-cured tobacco and Salvia miltiorrhiza intercropping row ratio of 2:2) was higher than that of other intercropping treatments. In 2018 and 2019, the total output value increased by 21.3% and 22.4%, respectively, compared with the flue-cured tobacco monoculture. The intercropping advantage was obvious, and the treatment effect was the best.

AbstractCordyceps fumosorosea is an important species in the genus of Cordyceps, containing a variety of bioactive compounds, including fumosorinone (FU). This study was a ground-breaking assessment of FU levels in liquid and solid cultures. The present study focused on the impacts of solid-state fermentation (SSF) using solid substrates (wheat, oat, and rice), as well as the effects of fermentation parameters (pH, temperature, and incubation period), on the generation of FU. All the fermentation parameters had significant effects on the synthesis of FU. In a study of 25 °C, 5.5 pH, and 21 days of incubation period combinations calculated -to give maximal FU production, it was found that the optimal values were 25 °C, 5.5 pH, and 21 days, respectively. In a solid substrate medium culture, FU could be produced from SSF. At 30 days, a medium composed of rice yielded the most FU (798.50 mg/L), followed by a medium composed of wheat and oats (640.50 and 450.50 mg/L), respectively. An efficient method for increasing FU production on a large scale could be found in this approach. The results of this study might have multiple applications in different industrial fermentation processes.

Waldenstrom’s macroglobulinemia (WM) is a lymphoplasmacytic lymphoma characterized by the infiltration of the bone marrow by clonal lymphoplasmacytic cells that produce monoclonal immunoglobulin M as defined by the World Health Organization Classification of hematological malignancies. Historically, the treatment options for WM were limited to alkylating agents and purine analogs. The introduction of immune therapy, including CD20 targeted therapy, proteasome inhibitors, and immune modulators, has provided benefit to those patients and has now become the standard of care. As WM patients become long-term survivors, treatment’s late toxicities have become more apparent. Here, we report a case of a 74-year-old female who presented to the hospital with fatigue and was diagnosed with WM. She was treated with bortezomib, doxorubicin, and bendamustine, followed by rituximab. After a remission period of 15 years, the patient had a relapse of WM, and bone marrow biopsy findings were consistent with intermediate-risk t-MDS with complex cytogenetics, presenting us with a treatment dilemma. We decided to treat WM, and the patient went into VGPR with residual lymphoma cells. Despite having dysplasia and complex cytogenetics, she did not have any cytopenia. Currently, she is under observation anticipating the progression of her MDS, given her intermediate I risk status. This case features the occurrence of t-MDS after therapy with bendamustine, cladribine, and doxorubicin. This highlights the need for closer monitoring and consideration of long-term adverse effects when treating patients with indolent lymphomas, especially WM. Late complications need to be considered, and risk versus benefit analysis needs to be carefully evaluated, especially in younger patients with WM.

In 2020, diseased seedlings of winter oilseed rape (Brassica napus) with cankered taproots as well as abundant sclerotium-like structures in the soil surrounding the roots were found in Longxi County of Gansu province of northwestern China. A fungus with production of pycnidia was isolated from the diseased roots, and it was identified based on morphological characteristics, molecular phylogeny (ITS, LSU) and PCR detection with the specific primers. The fungus was identified as Leptosphaeria sclerotioides Gruyter, Aveskamp & Verkley [anamorph: Phoma sclerotioides (Preuss) ex Sacc.]. Re-inoculation of isolates of P9 and P10 of L. sclerotioides on winter oilseed rape (B. napus cultivar ‘Zhongshuang No. 9’) in Wuhan caused formation of abundant sclerotium-like structures in soil surrounding the roots, but failed to produce root cankers as those observed in Gansu possibly due to lack of long peroid of low-temperature conditioning in Wuhan. In spite of this, plant height, pod number and seed yield of oilseed rape were significantly reduced in the treatment with L. sclerotioides P9 and P10, compared to the control treatment. To the best of our knowledge, this is the first report of L. sclerotioides on B. napus in China and the finding broadened our understanding about the natural distribution of this psychrophilic fungus.

Abstract Background and aims Few studies have examined the relationship between malnutrition, as defined by the Geriatric Nutrition Risk Index (GNRI), and all-cause mortality and cardiovascular mortality events, particularly in persons with diabetes. The study aimed at the association between GNRI and all-cause mortality and cardiovascular mortality in older Americans with diabetes. Methods Data from this retrospective study were obtained from the National Health and Nutrition Examination (NHANES) 1999–2016. Using data from The NHANES Public-Use Linked Mortality Files to assess all-cause mortality (ACM) and cardiovascular mortality (CVM). After excluding participants younger than 60 years and without diabetes, and with missing follow-up data, 4400 cases were left in this study. Abstract This paper investigates low‐latency offloading strategy in a non‐orthogonal multiple access aided mobile edge computing (NOMA‐MEC) system consisting of K edge servers, one mobile user and one cloud server. An intelligent edge server selection strategy (IESSS) based on Markov decision process (MDP) is proposed to select an edge server, in order to reduce the task completion latency of this system. When an edge server is selected by the proposed IESSS, a joint optimization problem of power allocation and task scheduling factors is formulated to minimize the task completion latency of the hybrid NOMA‐MEC system. To solve the formulated non‐convex optimization problem with coupled variables, a low‐complexity adaptive power‐task resource allocation iterative (APTRAI) algorithm is proposed. Simulation results demonstrate the advantages of the proposed IESSS and verify the convergence and time complexity of the proposed APTRAI algorithm.

Abstract Nicotiana alata Link et Otto, widely used in landscaping, is not only of great ornamental value but also of high commercial and medical value. The global potential habitat of N. alata and the environmental factors affecting its distribution are not that clear at present. To provide a reference for the reasonable and extensive planting of N. alata now and in the future, the MaxEnt model was used to predict its global suitable habitats under current and future climate conditions, respectively, based on global geographic distribution data of N. alata and the current and future world bioclimatic variables. The results showed that mean temperature of the driest quarter (bio9), precipitation of driest month (bio14), precipitation seasonality (bio15) and max temperature of warmest month (bio5), were the key bioclimatic variables governing the distribution of N. alata. The global suitable habitats of N. alata were mainly distributed in Europe, the United States, southeastern South America, and China under current climate conditions. Compared with current climate conditions, the future climate decreased suitable habitats of N. alata under SSP1-2.6, and SSP2-4.5 scenario and increased suitable habitats of N. alata under SSP3-7.0, and SSP5-8.5 climatic scenarios. The results provided valuable information and theoretical reference for the reasonable planting of N. alata.

Introduction: The effects of isoform-specific histone deacetylase inhibitors (HDACIs) and the non-selective HDACI on sepsis have been profoundly reported. However, the best HDAC classes have not been fully evaluated. Therefore, this study aimed to determine which HDACIs are responsible for survival and beneficial for organ injury. Methods: Experiment I, SD rats were subjected to cecal ligation and puncture and randomly assigned to the no treatment, dimethyl sulfoxide (DMSO) only, MS-275, LMK-235, tubastatinA (TubA), trichostatin-A (TSA), and sirtinol groups (n = 5). Survival was monitored for 48 h. Experiment II, the animals were monitored for 12 h, then, blood and tissues sample were collected. Interleukin (IL)-6, IL-1β, tumour necrosis factor (TNF)-α, alanine aminotransferase (ALT), aspartate aminotransferase (AST), creatine kinase (CK) and lactate dehydrogenase (LDH) expressions were evaluated using ELISA. Liver, heart and lung tissues were analysed via hematoxylin and eosin staining. Liver and heart tissue lysates were analysed for acetylated histones H3, H4, a-tubulin and nuclear factor kappa B (NF-κB), IL-6, IL-1β, and TNF-α using western blotting. Splenocytes were examined via flow cytometry to analyse the immune cell population. Results: MS-275, TubA and TSA treatments significantly prolonged survival. MS-275, LMK-235, TubA and TSA significantly reduced the histopathological scores and AST, ALT, CK, LDH, IL-6, IL-1β and TNF-α levels, significantly increased acetylated of NF-κB and changed the immune cell proportion. Conclusion: Our results indicated that HDACI classes I and IIb and non-selective HDACI can significantly prolong survival. Moreover, non-selective and isoform-specific class I and IIa/IIb HDACIs can attenuate inflammation and organ injury.

Backgrounds: Hypertension stands as the predominant global cause of mortality. A notable deficiency exists in terms of predictive models for mortality among individuals with hypertension. We aim to devise an effective nomogram model that possesses the capability to forecast all-cause mortality within hypertensive populations.Methods: The data for this study were drawn from nine successive cycles of the National Health and Nutrition Examination Survey (NHANES) spanning the years from 1999 to 2016. The dataset was partitioned into training and validation sets at a 7:3 ratio. We opted for clinical practice-relevant indicators, applied the least absolute shrinkage and selection operator (LASSO) regression to identify the most pertinent variables, and subsequently built a nomogram model. We also employed concordance index, receiver operating characteristic (ROC) curves, calibration curves and decision curve analysis (DCA) to assess the model’s validity.Results: A total of 17,125 hypertensive participants were included in this study with a division into a training set (11,993 individuals) and a validation set (5,132 individuals). LASSO regression was applied for the training set to obtain nine variables including age, monocytes, neutrophils, serum albumin, serum potassium, cardiovascular disease, diabetes, serum creatinine and glycated hemoglobin (HbA1C), and constructed a nomogram prediction model. To validate this model, data from the training and validation sets were used for validation separately. The concordance index of the nomogram model was 0.800 (95% CI, 0.792–0.808, p < 0.001) based on the training set and 0.793 (95% CI, 0.781–0.805, p < 0.001) based on the validation set. The ROC curves, calibration curves, and DCA curves all showed good predictive performance.Conclusion: We have developed a nomogram that effectively forecasts the risk of all-cause mortality among American adults in hypertensive populations. Clinicians may use this nomogram to assess patient’s prognosis and choose a proper intervention in a timely manner.

Microorganisms present on the surface of tobacco leaves play a significant role in shaping the composition of the tobacco microbial ecosystem, which undergoes continuous changes throughout the curing process. In the present study, a total of four distinct tobacco curing periods were selected for sampling, namely the fresh, yellowing, leaf-drying, and stem-drying stages. The bacterial 16S rRNA gene sequences of the collected samples were subsequently analyzed to identify operational taxonomic units (OTUs). The findings indicated that the complete dataset of leaf microbial samples was clustered, resulting in the identification of 1,783 operational taxonomic units (OTUs). Furthermore, the analysis of diversity revealed a pattern of initially increasing and subsequently decreasing community diversity. Redundancy Analysis (RDA) and weighted gene correlation networks for analysis (WGCNA) were employed in conjunction with environmental factors to assign OTUs to 22 modules for functional analysis. Additionally, a classification model utilizing the random forest algorithm was utilized to identify seven marker microorganisms (Escherichia coli, Faecalibacterium prausnitzii, Faecalibacterium, Escherichia-Shigella, Peptostreptococcaceae, Peptostreptococcales-Tissierellales, and Proteobacteria) that exhibited discriminative characteristics across different time periods. This study aimed to investigate the dynamic changes in the bacterial community throughout the curing process and their impact on the community’s function. Additionally, certain bacteria were identified as potential markers for detecting changes in the curing stage. These findings offer a novel opportunity to accurately regulate the curing environment, thereby enhancing the overall quality of tobacco leaf curing.

Summary: Sclerotinia sclerotiorum and Botrytis cinerea are necrotrophic plant-pathogenic fungi, causing substantial economic losses on many crops. So far, resistant cultivars against these pathogens are unavailable in most crops. Here, we show that the serine protease CmSp1 of Coniothyrium minitans, a well-characterized mycoparasite of S. sclerotiorum, contributed to suppressing the petal-mediated infection by S. sclerotiorum in rapeseed. Application of recombinant CmSp1 proteins facilitates the bulk degradation of S. sclerotiorum proteins and inhibits spore germination and hyphal growth of S. sclerotiorum and B. cinerea, thereby preventing the development of both diseases. Stable transgenic rapeseed plants with tissue-specific expression of CmSp1 in flower petals inhibit the petal-mediated infection by both S. sclerotiorum and B. cinerea, and resulting transgenic plants have no adverse effect on other agronomic traits. Thus, our findings provide a novel mechanism by which a mycoparasite inhibits fungal pathogens and an environmentally friendly disease management strategy.

IntroductionTai Chi standing meditation (Zhan Zhuang, also called pile standing) is characterized by meditation, deep breathing, and mental focus based on theories of traditional Chinese medicine. The purpose of the present study was to explore prefrontal cortical hemodynamics and the functional network organization associated with Tai Chi standing meditation by using functional near-infrared spectroscopy (fNIRS).MethodsTwenty-four channel fNIRS signals were recorded from 24 male Tai Chi Quan practitioners (54.71 ± 8.04 years) while standing at rest and standing during Tai Chi meditation. The general linear model and the SPM method were used to analyze the fNIRS signals. Pearson correlation was calculated to determine the functional connectivity between the prefrontal cortical sub-regions. The small world properties of the FC networks were then further analyzed based on graph theory.ResultsDuring Tai Chi standing meditation, significantly higher concentrations of oxygenated hemoglobin were observed in bilateral dorsolateral prefrontal cortex (DLPFC), ventrolateral prefrontal cortex (VLPFC), frontal eye field (FEF), and pre-motor cortex (PMC) compared with the values measured during standing rest (p < 0.05). Simultaneously, significant decreases in deoxygenated hemoglobin concentration were observed in left VLPFC, right PMC and DLPFC during Tai Chi standing meditation than during standing rest (p < 0.05). Functional connectivity between the left and right PFC was also significantly stronger during the Tai Chi standing meditation (p < 0.05). The functional brain networks exhibited small-world architecture, and more network hubs located in DLPFC and VLPFC were identified during Tai Chi standing meditation than during standing rest.DiscussionThese findings suggest that Tai Chi standing meditation introduces significant changes in the cortical blood flow and the brain functional network organization.

The application of plant growth-promoting rhizobacteria (PGPR) is a novel and an efficient strategy for improving soil degradation and productivity. However, the effect of PGPR on tobacco (Nicotiana tabacum L.) seedling growth under salt stress remains unclear. Here, microcosm experiments were designed to verify the effects of Bacillus cereus TC012 (BC), Bacillus methylotrophicus TC023 (BM), and Bacillus amyloliquefacien TC037 (BA) on tobacco grown in salt-affected soil. The results showed that BC, BM, and BA treatments significantly increased the height of tobacco plants by 38.65%, 91.94%, and 90.66%, respectively. Furthermore, the growth of various components of tobacco plant, such as stem girth, seedling biomass, carotenoid, and chlorophyll were stimulated in salt-affected soils. The changes in the salinity of the tobacco plant mostly relies on the improvement of proline, soluble protein, soluble sugar content, plant protective enzymatic activity, and K+/Na+ ratios. Increases in indole-3-acetic acid, zeatin riboside and gibberellic acid also promoted tobacco growth. Additionally, inoculation with PGPR enhanced the enzymatic activity of laccase, urease, neutral protease, acid phosphatase, and sucrase in soil samples and had positive effects on the physicochemical properties. The soil bacterial communities significantly improved after inoculation with PGPR. In particular, the relative abundance of Pseudomonas and Bacillus significantly increased. Overall, PGPR inoculation has great potential to alleviate salt damage in tobacco plants and may have far reaching benefits to the agricultural community.

The impact of large reservoirs on regional climate extremes, such as heatwaves, is not yet well understood, primarily due to the complex interactions of heat, moisture, and momentum over mountainous terrains and the dynamic nature of water surfaces. In this study, we investigate the role of the Miyun Reservoir (MYR), one of the largest reservoirs in northern China, covering an extensive water surface area of 180 km2, in influencing a severe heatwave event that occurred southwest of MYR from 2nd to 6th July 2010. Using high-resolution simulations from the Weather Research and Forecasting Model, our findings reveal contrasting thermodynamic effects of MYR on near-surface air temperatures, with up to three degrees of warming during night-time and up to five degrees of cooling during daytime, with a spatial extent of impact reaching up to 20 km away from the reservoir. These impacts are attributed to a combination of synergized land-lake breeze and mountain-valley winds during the night, leading to the transport of warm air plumes downwind towards the plain and intensifying the heatwave. Furthermore, we observe an increase in surface water vapor brought to higher atmospheric levels due to strong vertical mixing, resulting in lowered surface-specific humidity. Our study emphasizes the dual role of large reservoirs in exacerbating remote heatwaves while providing cooling effects in their proximity. Gaining a comprehensive understanding of the intricate interactions between reservoirs, topography, and regional climate extremes is of paramount importance for accurate climate risk assessments and the formulation of effective mitigation strategies..

Background and objectivesThe relationship between the tumor microenvironment and the network of key signaling pathways in cancer plays a key role in the occurrence and development of tumors. Tumor-associated macrophages (TAMs) are important inflammatory cells in the tumor microenvironment and play an important role in tumorigenesis and progression. Macrophages in malignant tumors, mainly the M2 subtype, promote tumor progression by producing cytokines and down-regulating anti-inflammatory immune responses. Several articles have investigated the effect of macrophages on the sensitivity of cancer chemotherapeutic agents, but few such articles have been reported in cholangiocarcinoma, so we investigated the effect of M2 macrophage on the sensitivity of cholangiocarcinoma cells to Lenvatinib compared to M1.MethodsTHP-1 monocytes were polarized to M0 macrophage by phorbol 12-myristate 13-acetate (PMA) and then induced to differentiate into M1 and M2 macrophages by LPS, IFN-γ and IL-4 and IL-13, respectively. Macrophages and cholangiocarcinoma cells were co-cultured prior to 24 hours of Lenvatinib administration, cancer cell apoptosis was detected by western-blot, FACS analysis of Annexin V and PI staining. Furthermore, we use xCELLigence RTCA SP Instrument (ACEA Bio-sciences) to monitor cell viability of Lenvatinib administration in co-culture of cholangiocarcinoma cells and macrophages. After tumorigenesis in immunodeficient mice, Lenvatinib was administered, and the effects of M2 on biological characteristics of cholangiocarcinoma cells were investigated by immuno-histochemistry.ResultsmRNA and protein expression of M1 and M2 markers confirmed the polarization of THP-1 derived macrophages, which provided a successful and efficient model of monocyte polarization to TAMs. Lenvatinib-induced apoptosis of cholangiocarcinoma cells was significantly reduced when co-cultured with M2 macrophage, whereas apoptosis of cholangiocarcinoma cells co-cultured with M1 macrophage was increased. In the CDX model, Lenvatinib-induced cancer cell apoptosis was markedly reduced, and proliferative cells increased in the presence of M2 macrophages. Angiogenesis related factors was significantly increased in cholangiocarcinoma cells co-cultured with M2.ConclusionCompared with M1, M2 macrophages can inhibit the anti-tumor effect of Lenvatinib on cholangiocarcinoma through immune regulation, which may be related to the tumor angiogenesis factor effect of M2 macrophage.
