Abstract

Nicht-photorealistisches Rendering (NPR) hat in den vergangenen 15 Jahren eine hohe Aufmerksamkeit erlangt. Im Gegensatz zur photorealistischen Computergrafik, deren Basis die physikalisch-korrekte Simulation des Lichttransportes und der Materialeigenschaften ist, umfasst NPR vielseitige Werkzeuge zur Erzeugung ausdrucksstarker und persönlicher Interpretationen mittels unterschiedlichster Darstellungsstile - Informationen und Inhalte können dadurch fokussiert und effizient vermittelt werden. NPR ist durch ästhetische Ansprüche, traditionelles Wissen des Kunsthandwerks und Erkenntnissen auf dem Gebiet der visuellen Perzeption geprägt.Diese Dissertation untersucht Aspekte einer Rendering-Pipeline, die für die computer-generierte Darstellung von Aquarell-Illustrationen konzipiert wurde. Die digitale Nachbildung dieses komplexen Mediums erfordert eine ganzheitliche Betrachtung des Zeichenprozesses - dies beinhaltet die Emulation des Mediums an sich als auch gestalterische Aspekte, wie das Extrahieren wesentlicher Elemente, das Platzieren von Akzenten oder die Verwendung von Farbkontrasten. Wir sehen unseren wissenschaftlichen Beitrag in der Verbesserung der visuellen Ästhetik und Glaubwürdigkeit von computer-generierten Aquarellen durch die Kombination von traditionellen Gesetzmäßigkeiten des Kunsthandwerks und effektiven Techniken zur Wiedergabe der Zeichenwerkzeuge und des Zeichenmediums.Wir stellen wissenschaftliche Beiträge zu folgenden Themengebieten vor: Es werden zwei empirisch-basierte Ansätze formuliert, die die Nachbildung der visuellen Eigenschaften von Wasserfarbe erlauben. Im Gegensatz zu physikalischkorrekten Simulationen basieren die Ansätze auf Algorithmen der Bildverarbeitung, wodurch das effiziente Rendern von 2d und 3d Daten mit einem Aquarellstil erreicht wird. Des Weiteren werden stilistische Mittel innerhalb des Zeichenprozesses reproduziert, wodurch allgemeingültige Erkenntnisse des traditionellen Kunsthandwerks in die computer-generierte, künstlerische Darstellung überführt werden können. Dadurch wird eine verbesserte visuelle und ästhetische Qualität und Glaubwürdigkeit erreicht.Letztendlich präsentieren wir Zusammenarbeiten, die unsere Erkenntnisse und Algorithmen in Projekten der Film- und Grafikindustrie anwenden. Das erste Projekt zielte auf die Integration eines Aquarell-Stiles in eine 3d Animations-Pipeline. Eine zweite Zusammenarbeit konzentrierte sich auf das Ziel, eine einfache, so genannte "one-click" Lösung zu erarbeiten, die eine überzeugende Aquarell-Illustration innerhalb eines bestehenden CAD Systems ausgeben kann.

Abstract

The Snapshot Painter is a proof-of-concept for the modular structure of the library. This module processes 2d image data such as photographs or renderings, turning them into a computer generated artwork. The algorithms are currently adopted to a watercolor style combined with loose and sketchy strokes. As the intent is to communicate our vision of animated npr content respecting issues of temporal coherence, the image we generate is embedded within a dynamic artwork: the forms evolve in fluid patterns that turn the single image to a dynamic temporal process.

Abstract

We describe a project that integrates a non-photorealistic rendering pipeline into an existing CAD system. The pipeline enables the customers of the CAD company to create aesthetically appealing renderings in addition to the classical CAD output. Since these customers are operating in the high-quality segment of furniture and interior design, the visual style of the sales brochure is highly important. We concentrate on watercolor drawings, since they are a traditional medium to present artistic interpretations of architectural and CAD data. To create a convincing and aesthetic reproduction of this style, we propose two techniques: First, we introduce a tone-based lighting to separate the traditional shading of a rendering into tone and intensity, which supports traditional color palettes for watercolor paintings, and second, we exploit ambient occlusion information to render traditional stylistic means such as abstraction and indication.

Abstract

This paper presents an approach for vivid representations of foliage based on implicit surfaces. It approximates the complex lighting interaction within the foliage and enables a clear illustration of the general shape and the local density of the foliage, thus supporting the three-dimensional depth cue of the viewer. Due to its straightforward implementation as a preprocessing step that only adjusts the normal vectors of the geometry, this method has no additional memory requirements during the rendering process, and is especially applicable to real-time visualizations. Furthermore, it may be applied to photorealistic as well as nonphotorealistic visualizations, which both benefit from the modified normal vector information.

We present algorithms that produce a natural watercolor appearance of 3D-scenes and render them in realtime. Our approach imitates the most important natural effects of watercolor and incorporates two essential painting techniques: the wet-in-wet and the wet-on-dry painting. We apply our technique to different scenes, and show how to segment and abstract the content.

Abstract

Previous epidemiologic studies showed an increased risk of neutralizing antibody (NAb) development against Interferon beta in multiple sclerosis patients who smoke. Cotinine is an easily detectable metabolite of nicotine and, therefore, can be used as an objective surrogate marker for smoking status. We measured cotinine levels in NAb-positive and NAb-negative patients to find a potential association of nicotine consumption and NAb development. Cotinine was measured in 37 patients with known smoking status and in 123 patients with unknown smoking status, all of whom were routinely tested for NAb. Cotinine was detected by an enzyme-linked immunosorbent assay, inhibition assay. We compared cotinine levels by NAb status and tested for the strength of association between cotinine and NAb status. We found a discrepancy between smoking status stated by patients and status defined by cotinine levels in 7 of 37 patients. In both cohorts, together with and without previously known smoking status ( n = 160), we found 34% and 39% smokers, respectively, as defined by cotinine levels in NAb-negative and NAb-positive patients ( P = 0.511). In our analysis, smoking was not associated with higher risk of NAb development. Moreover, smoking habits stated by patients do not always correlate with cotinine levels. [ABSTRACT FROM AUTHOR]

Diese Arbeit stellt ein Rendering-System vor, das die Echtzeitdarstellung von gezeichneten Gesichtern erlaubt. Auf Basis einer dreidimensionalen Beschreibung eines Kopfes werden signifikante Linien extrahiert und auf zeichnerische Weise dargestellt. Details, die nicht oder unzureichend geometrisch modelliert sind, können manuell mit Hilfe von Surface- und Anchor-Strokes ergänzt werden. Auf diese Weise können charakteristische, detaillierte Gesichter in Echtzeit dargestellt und animiert werden, wodurch sich interessante Anwendungsgebiete im Bereich virtueller Avatare ermöglichen.

Abstract

SF3B1, SRSF2, and U2AF1 are the most frequently mutated splicing factor genes in the myelodysplastic syndromes (MDS). We have performed a comprehensive and systematic analysis to determine the effect of these commonly mutated splicing factors on pre-mRNA splicing in the bone marrow stem/progenitor cells and in the erythroid and myeloid precursors in splicing factor mutant MDS. Using RNA-seq, we determined the aberrantly spliced genes and dysregulated pathways in CD34+ cells of 84 patients with MDS. Splicing factor mutations result in different alterations in splicing and largely affect different genes, but these converge in common dysregulated pathways and cellular processes, focused on RNA splicing, protein synthesis, and mitochondrial dysfunction, suggesting common mechanisms of action in MDS. Many of these dysregulated pathways and cellular processes can be linked to the known disease pathophysiology associated with splicing factor mutations in MDS, whereas several others have not been previously associated with MDS, such as sirtuin signaling. We identified aberrantly spliced events associated with clinical variables, and isoforms that independently predict survival in MDS and implicate dysregulation of focal adhesion and extracellular exosomes as drivers of poor survival. Aberrantly spliced genes and dysregulated pathways were identified in the MDS-affected lineages in splicing factor mutant MDS. Functional studies demonstrated that knockdown of the mitosis regulators SEPT2 and AKAP8, aberrantly spliced target genes of SF3B1 and SRSF2 mutations, respectively, led to impaired erythroid cell growth and differentiation. This study illuminates the effect of the common spliceosome mutations on the MDS phenotype and provides novel insights into disease pathophysiology.

Abstract

We tested the hypothesis that proliferative activity of hematopoietic stem cells has impact on survival in newly diagnosed patients with myelodysplastic syndrome (MDS) and secondary acute myeloid leukemia (AML). RNA expression profiles of CD34+ cells were analyzed in 125 MDS patients and compared to healthy controls. Prognostic impact on overall survival (OS) of mRNA proliferation signatures established for solid tumor cells was analyzed retrospectively. For validation on the protein level, immunofluorescence and immunohistochemistry analyses in bone marrow (BM) biopsies were performed, and an independent cohort of 223 MDS and secondary AML patients was investigated. Lower proliferative activity correlated with the expression of cyclin-dependent kinase inhibitor 1C ( CDKN1C) and with shorter OS ( p < 0.001). In multivariable analysis, higher CDKN1C expression was associated with worse OS ( p = 0.02). On the BM level, a total of 84 (38%) patients showed CDKN1C protein expression before start of treatment. Patient, disease and treatment characteristics did not differ between CDKN1C-positive and -negative patients. Positive CDKN1C BM status was associated with shorter OS in multivariable analysis (HR 1.54, p = 0.04). There was an interaction between CDKN1C BM status and subsequent treatment with negative impact on OS being most pronounced in patients receiving conventional cytotoxic chemotherapy ( n = 83, 2-year OS 30% versus 58%, p = 0.002). In conclusion, low-proliferative phenotype and CDKN1C expression were associated with shorter OS. CDKN1C protein expression in the BM of newly diagnosed, treatment-naïve MDS and secondary AML patients was identified as a prognostic factor for poor survival in patients treated with antiproliferative chemotherapy. Abstract

The main task of product developers is to meet the ever increasing demands on quality, cost and functionality of technical products by setting the appropriate characteristics in order to realize the product properties desired by the customer. The overall objective of the authors is to develop a suitable procedure model for property-based product development, which guides the developers purposefully through the development process. An essential part of this procedure model is the matrix-based product description, which is in the focus of this paper. The matrix-based product description is basically a so-called multi domain matrix. By using this matrix-based product description, dependencies between requirements, behaviors of the product and the product's properties and characteristics can be mapped and analyzed systematically during the product development process. Thereby, for example, the effects of characteristic changes on product properties become visible and traceable and also differential requirements are set into relationship with properties and characteristics of the product. However, it has been shown that the manual filling of the matrix-based product description is very time-consuming. Hence, a computer-aided support is indispensable. For this reason, the software Loomeo is used in this paper to investigate its potential for IT support for the matrix-based product description. The interaction of dependencies between characteristics, properties and the resulting behavior is demonstrated in this paper through the matrix-based product description using the example of a front wheel suspension of a car. Thus, the strengths and weaknesses as well as opportunities and risks of Loomeo regarding the suitability of the IT-based matrix-based product description can be determined. The findings from both the application as well as from the evaluation of the software are the basis for the improvement and extension of the matrix-based product description. [ABSTRACT FROM AUTHOR]
