The properties of inverted repeat (foldback) sequences in Drosophila melanogaster DNA have been studied by HAP chromatography and electron microscope methods. Electron microscope observations show that there is a broad distribution of lengths of the duplex regions of the inverted repeats from very short to greater than 15 kb, with number and weight average values of 1.35 kb and 5.0 kb respectively. About 20% of the inverted repeats are separated by a single-strand spacer with lengths too short to observe, but the other 80% have spacers, P, with lengths ranging from 0.5 kb to greater than 30 kb. The number average and weight average spacer lengths for the total sample are 2.7 kb and 6.1 kb. With respect to the lengths of the spacers, P, between inverted repeats, the Drosophila genome differs from that of most organisms which have been studied where the spacers P are mostly too short to be measured. EM and HAP studies suggest that the average center-to-center spacing between sets of inverted repeats is 40-80 kb. The HAP studies show that there is a broad range of thermal stabilities for the duplexes formed by reassociation of inverted repeat sequences. Kinetic analysis shows that all of the frequency components of the Drosophila genome are present in the inverted repeats, the loops P, and the flanking sequences. There is a somewhat larger proportion of middle repetitive DNA in those inverted repeat duplexes which are resistant to digestion by Mung Bean Endonuclease I. These enzyme resistant duplexes comprise about 3% of the entire genome. It is estimated that there are approximately 2000-4000 inverted repeat pairs in the entire genome.

Intense exertion is an occupational hazard inherent to fire fighting. This study was designed to look at the exertion levels that fire fighters attain during a fire fighting exercise when using (1) no self-contained breathing apparatus (SCBA), (2) light SCBA, and (3) heavy SCBA. Exertion levels were measured as a function of the heart rate increase relative to the maximum predicted heart rate determined by a standard treadmill exercise test. Five fire fighters wore electrocardiographic monitors during a routine fire fighting exercise. Heart rates increased rapidly to 70% to 80% of maximum within the first minute and then plateaued at 90% to 100% until the attack on the fire was completed. There was no significant difference between exertion levels when using no SCBA, light SCBA, and heavy SCBA (split-plot analysis of variance, p greater than .25). These results suggest that fire fighters attain an intense level of physical activity quickly and maintain that level as long as they are actively engaged in fighting fire. These results also suggest that regardless of the weight of the SCBA, if employed, fire fighters exert themselves from 85% to 100% of their maximum and adjust their work output to maintain that near-maximal level.

Using a heterologous rat cDNA probe, we have identified a 14.7 kbp Drosophila melanogaster genomic clone containing the X-linked gene Pgd+, which encodes the enzyme 6-phosphogluconate dehydrogenase (6PGD). We used in situ hybridization to larval polytene chromosomes, a somatic transient expression assay for enzyme activity, and the rescue of the lethal Pgd- phenotype by germline transformation to verify the identity of the gene. A 7.4 kbp fragment including the gene and approximately 1.2 kbp of upstream and 1.8 kbp of downstream sequences was relocated to autosomal ectopic sites by germline transformation; this transduced gene exhibits levels of enhanced activity in males comparable to those of the indigenous gene at its normal X chromosome locus. We conclude that the sequences responsible for dosage compensation of Pgd+ are included in this fragment.

The protozoa Crithidia and Trypanosoma contain within a mitochondrion a mass of DNA known as kinetoplast DNA (kDNA) which consists mainly of an association of thousands of small circular molecules of similar size held together by topological interlocking. Using kDNA from Crithidia acanthocephali, Crithidia luciliae, and Trypanosoma lewisi, physicochemical studies have been carried out with intact associations and with fractions of covalently closed single circular molecules, and of open single circular and unit length linear molecules obtained from kDNA associations by sonication, sucrose sedimentation, and cesium chloride-ethidium bromide equilibrium centrifugation. Buoyant density analyses failed to provide evidence for base composition heterogeneity among kDNA molecules within a species. The complementary nucleotide strands of kDNA molecules of all three species had distinct buoyant densities in both alkaline and neutral cesium chloride. For C. acanthocephali kDNA, these buoyant density differences were shown to be a reflection of differences in base composition between the complementary nucleotide strands. The molar ratios of adenine: thymine:guanine:cytosine, obtained from deoxyribonucleotide analyses were 16.8:41.0:28.1:14.1 for the heavy strand and 41.6:16.6:12.8:29.0 for the light strand. Covalently closed single circular molecules of C. acanthocephali (as well as intact kDNA associations of C. acanthocephali and T. lewisi) formed a single band in alkaline cesium chloride gradients, indicating their component nucleotide strands to be alkaline insensitive. Data from buoyant density, base composition, and thermal melting analyses suggested that minor bases are either rare or absent in Crithidia kDNA. The kinetics of renaturation of 32P labeled C. acanthocephali kDNA measured using hydroxyapatite chromatography were consistent with at least 70% of the circular molecules of this DNA having the same nucleotide sequence. Evidence for sequence homologies among the kDNAs of all three species was obtained from buoyant density analyses of DNA in annealed mixtures containing one component kDNA strand from each of two species.

The LSP1-alpha gene of Drosophila melanogaster is located on the X chromosome at 11B yet is not dosage compensated. In order to determine if this gene is inherently incapable of dosage compensation or if it does not compensate because the appropriate regulatory cis-acting sequences are absent from its chromosomal domain, we have undertaken to relocate it to ectopic sites on the X chromosome. To differentiate between the transcripts produced by the transduced gene and those produced by the indigenous gene, we inserted a 500-bp sequence of mouse DNA into the LSP1-alpha clone prior to using it for transformation. Our results show that the LSP1-alpha gene exhibits equivalent levels of transcripts in the two sexes when it is relocated to either an autosomal site or to an ectopic site on the X chromosome. We conclude that the LSP1-alpha gene is capable of dosage compensation.

An enzyme linked immune sorbent assay (ELISA) was developed to measure bacterial surface lipoteichoic acid (LTA). Numerous strains of oral streptococci belonging to the 'viridans' group were examined on three separate occasions. The results show that, under these cultural conditions, oral streptococci do not normally express LTA on the cell surface. Occasionally strains produced amounts of LTA detectable using the ELISA but this was not a reproducible phenomenon.

Cot analysis shows that the haploid Drosophila genome contains 12 percent rapidly reassociating, highly reiterated DNA, 12 percent middle repetitive DNA with an average reiteration frequency of 70, and 70 percent single-copy DNA. The distribution of the middle repetitive sequences in the genome has been studied by an examination in the electron microscope of the structures obtained when middle repetitive sequences present on large DNA strands reassociate and by the hydroxyapatite binding methods developed by Davidson et al. (1973). At least one third by weight of the middle repetitive sequences are interspersed in single-copy sequences. These interspersed middle repetitive sequences have a fairly uniform distribution of lengths from less than 0.5 to 13 kb, with a number average value of 5.6 kb. The average distance between middle repetitive sequences is greater than 13 kb. The data do not exclude the possibility that essentially all of the middle repetitive sequences have the interspersion pattern described above; however, it is possible that some of the middle repetitive sequences of Drosophila are clustered in stretches of length much greater than 13 kb. The interspersion pattern of the middle repetitive sequences in Drosophila is quite different from that which occurs in the sea urchin, in Xenopus, in rat, and probably many other higher eucaryotes.

In previous studies we identified a 500 bp DNA fragment from the genome of Trypanosoma cruzi which encoded epitopes present in an 85 kd trypomastigote-specific surface antigen. A unique feature of this DNA insert was the presence of a 27 bp tandem repeat unit within the putative coding region. The findings presented here show that the gene which encodes this particular surface protein is a member of a multigene family, and that the 27 bp repeat unit defines a subset of this family. Only four separate members of the family contain sequences homologous to the 27 bp repeat unit. Of these, three have been cloned and shown by direct nucleotide sequence analysis not to contain the original 500 bp fragment. By restriction enzyme analysis, the 500 bp fragment is inferred to be present in a 5.4 kb EcoRI genomic DNA fragment that is refractory to isolation by standard cloning procedures. Preferential sensitivity of this fragment to digestion with Bal31 nuclease indicates that it is likely to be telomeric, thus explaining the inability to obtain it in several different recombinant DNA libraries. In order to determine which of the four members were transcribed, 26 cDNA recombinants having sequence homology with the 27 bp repeat were examined. Restriction enzyme maps and nucleotide sequence analysis of these cDNAs indicate that transcription occurs almost exclusively from the telomeric member of the family.

A new method for gene mapping at the chromosome level using in situ hybridization and scanning electron microscopy is described and has been applied to mapping the rRNA genes of Drosophila melanogaster. Biotin is covalently attached to Drosophila rRNA via a cytochrome c bridge at a ratio of one cytochrome-biotin per 130 nucleotides by a chemical procedure. Polymethacrylate spheres with a diameter of ca. 60 nm are prepared by emulsion polymerization and are covalently attached to the protein avidin at a ratio of 5-20 avidins per sphere. The biotin-labeled rRNA is hybridized to denatured DNA in a chromosome squash. Upon incubation with a sphere solution, some of the biotin sites become labeled with spheres because of the strong non-covalent interaction between biotin and avidin. The chromosome squash is examined in the scanning electron microscope (SEM). Polymer spheres, which are visible in the SEM, are observed to label the nucleolus, where the rRNA genes are located.

The proliferative response of medial smooth muscle cells to balloon-induced endothelial denudation was measured in coronary arteries of normal and von Willebrand's disease swine, a strain of swine with a genetic defect affecting platelet-vessel wall interactions. Animals were sacrificed 48 hours following endothelial injury. Incorporation of a 1-hour pulse label of 3H-thymidine was determined by measuring DNA specific activity. Autoradiography of coronary artery step segments was also performed. The ratio of DNA specific activity in ballooned vessels to that in nonballooned vessels ranged from 1.2 to 26.2. No statistically significant difference in 3H-thymidine incorporation between the two phenotypes was documented (p greater than 0.25). Similarly, DNA incorporation of 3H-thymidine as determined by autoradiography was accelerated to the same degree in ballooned vessels in both phenotypes. The results suggest that von Willebrand factor-mediated platelet activation is not essential for the release of platelet-derived growth factor or that in vivo factors other than platelet-derived growth factor play a significant role in the response of medial smooth muscle cells to acute intimal injury causing endothelial denudation.

The hydrophobicity of human oral streptococci was measured with the hexadecane assay modified by the incorporation of polyethylene glycol 6000. Large variability in the hydrophobicity between cultures of some strains grown on different occasions was observed whereas other strains were less variable. The variation in hydrophobicity was significantly reduced by growing the cells in continuous culture in a chemostat under glucose-limiting conditions. The Streptococcus mutans strains used all had low hydrophobicity and the mean hydrophobicity of this species was significantly lower (P less than 0.05) than the mean hydrophobicity of Strep. salivarius, Strep. sanguis Type I and Strep. sanguis Type II strains. This finding supports the view that hydrophobicity is a contributing factor in the adhesion of viridans streptococci to oral surfaces.

The parasitic protozoan Trypanosoma cruzi can establish infection in humans and other vertebrate hosts through direct penetration of host cells by trypomastigotes transmitted by the insect vector. Although the molecular processes involved in trypomastigote interiorization of vertebrate cells are unknown, several studies suggest that surface glycoproteins are involved. It is likely that the proteins involved are specific to the trypomastigote stage of the parasite, since only trypomastigotes found in both the insect vector and the vertebrate host bloodstream are capable of invading vertebrate cells. In contrast, the epimastigote stage, found exclusively in the vector, and the amastigote stage, an intracellular stage in the vertebrate host, cannot penetrate the cell directly. We have therefore concentrated our efforts on trypomastigote surface proteins and, along with others, have identified two trypomastigote-specific surface glycoproteins of relative molecular mass (Mr) 90,000 (90K) and 85,000 (85K). Antibody neutralization experiments indicate that the 85K glycoprotein is necessary for efficient interiorization of trypomastigotes in mammalian cells. Here we describe the molecular cloning of a genomic DNA fragment that encodes antigenic determinants present in the 85K trypomastigote surface antigen. The polypeptide fragment encoded by the cloned DNA is recognized by serum from a T. cruzi-infected host and is inferred by DNA sequence analysis to contain a nonapeptide unit that is tandemly repeated five times. Also, the messenger complementary to the cloned DNA fragment is present only in the trypomastigote stage of the parasite.

The comparative lack of sexual dimorphism of apricot and other [sex-linked] genes studied is due to "sex-limitation", i.e., to a compensatory influence of the dosage difference between the sexes in respect to other genes in the X-chromosome. The facts are of particular interest from an evolutionary standpoint.

The surface polypeptides of epimastigotes and tissue culture-derived trypomastigotes of Trypanosoma cruzi have been isolated free of most cytosolic components by use of the 2-iminobiotin-avidin interaction. Polypeptides of the trypomastigote stage obtained by this technique are recognized by serum antibodies from Chagasic patients and T. cruzi-infected mice. These polypeptides have been used as the detecting antigen for the identification of hybridoma cells producing monoclonal antibodies against the surface proteins of the trypomastigote stage of T. cruzi. These experiments document a practical approach for obtaining T. cruzi surface proteins in sufficient quantity and purity for use in immunological studies.

Chloroplast DNA of the protozoan flagellate, Euglena gracilis, exists as circular molecules, 40 mum in contour length, as shown by electron microscopy and buoyant density analyses.
