Abstract

BackgroundUnintentional home injuries are a leading cause of preventable death in young children. Safety education and equipment provision improve home safety practices, but their impact on injuries is less clear. Between 2009 and 2011, a national home safety equipment scheme was implemented in England (Safe At Home), targeting high-injury-rate areas and socioeconomically disadvantaged families with children under 5. This provided a ‘natural experiment’ for evaluating the scheme’s impact on hospital admissions for unintentional injuries.MethodsControlled interrupted time series analysis of unintentional injury hospital admission rates in small areas (Lower Layer Super Output Areas (LSOAs)) in England where the scheme was implemented (intervention areas, n=9466) and matched with LSOAs in England and Wales where it was not implemented (control areas, n=9466), with subgroup analyses by density of equipment provision.Results57 656 homes receiving safety equipment were included in the analysis. In the 2 years after the scheme ended, monthly admission rates declined in intervention areas (−0.33% (−0.47% to −0.18%)) but did not decline in control areas (0.04% (−0.11%–0.19%), p value for difference in trend=0.001). Greater reductions in admission rates were seen as equipment provision density increased. Effects were not maintained beyond 2 years after the scheme ended.ConclusionsA national home safety equipment scheme was associated with a reduction in injury-related hospital admissions in children under 5 in the 2 years after the scheme ended. Providing a higher number of items of safety equipment appears to be more effective in reducing injury rates than providing fewer items.

Abstract

Many accounts of Anglo-American relations during the early 1970s stress the tensions with Washington that marked the premiership of Edward Heath, as Britain moved to become a member of the European Economic Community, with particular attention given to the Year of Europe dispute during 1973. Over the issue of the American war in Vietnam, however, Heath’s Conservative Government stood out from other West European states in offering a grateful Nixon administration wholehearted support, even in the face of domestic political criticism, when American air power was used during 1972 with an intensity and scope not seen before. Although prepared to offer firm diplomatic backing for American actions, British officials were still keen to avoid any direct involvement, repudiating the kind of role they had played in the settlement of the French Indochina War in 1954. By early 1973 the Nixon administration, influenced by Heath’s recent public support over Vietnam, looked upon the British Government as a firm ally, a context which helps in understanding the disappointment experienced over the subsequent Year of Europe dispute.

155. Particle or Cluster: On the Atomic Structure of the Seeds Used in Gold Nanoparticle Synthesis

Abstract

Seed-mediated synthesis strategies, in which small nanoparticle precursors are added to a growth solution to initiate heterogeneous nucleation, are among the most prevalent, simple, and productive methodologies for generating well-defined colloidal anisotropic nanostructures. Here, we identify the majority component in the seed solution as an atomically-precise gold nanocluster, consisting of a 32 atom Au core with 8 halide ligands and 12 neutral ligands constituting a bound ion pair between a halide and the cationic surfactant: Au32X8[AQA+•X-]12 (X = Cl, Br; AQA = alkyl quaternary ammonium). Ligand exchange is dynamic and versatile, occurring on the order of minutes and allowing for the formation of 48 distinct Au32 clusters with AQAX (alkyl quaternary ammonium halide) ligands. Anisotropic nanoparticle syntheses seeded with solutions enriched in Au32X8[AQA+•X-]12 show narrower size distributions and fewer impurity particle shapes, indicating the importance of this cluster as a precursor to the growth of well-defined nanostructures.

Abstract

Background:Thyroid gland malignancies are rare in pediatric patients (0.7% of tumors); only 1.8% are observed in patients aged MethodsInstitutional data were collected from eight international surgical oncology centers for pediatric patients with thyroid cancer between 2000 and 2020. Statistical analyses were performed using the GraphPad Prism software.ResultsAmong 255 total cases of thyroid cancer, only 13 (5.1%) were secondary tumors. Primary thyroid malignancies were more likely to be multifocal in origin (odds ratio [OR] 1.993, 95% confidence interval [CI].7466–5.132, p = 0.2323), have bilateral glandular location (OR 2.847, 95% CI.6835–12.68, p = 0.2648), and be metastatic at first diagnosis (OR 1.259, 95% CI.3267–5.696, p > 0.999). Secondary tumors showed a higher incidence of disease relapse (OR 1.556, 95% CI.4579-5.57, p = 0.4525) and surgical complications (OR 2.042, 95% CI 0.7917–5.221, p = 0.1614), including hypoparathyroidism and recurrent laryngeal nerve injury. The overall survival (OS) was 99% at 1 year and 97% after 10 years. No EFS differences were evident between the primary and secondary tumors (chi-square 0.7307, p = 0.39026).ConclusionsThis multicenter study demonstrated excellent survival in pediatric thyroid malignancies. Secondary tumors exhibited greater disease relapse (15.8 vs. 10.5%) and a higher incidence of surgical complications (36.8 vs. 22.2%).

159. Predicting patient treatment response and resistance via single-cell transcriptomics of their tumors

Abstract

Tailoring the best treatments for individual cancer patients is an important open challenge. Here, we build a precision oncology computational pipeline for PERsonalized single-Cell Expression-based Planning for Treatments In ONcology (PERCEPTION). Our approach capitalizes on recently published matched bulk and single-cell (SC) transcriptome profiles of large-scale cell-line drug screens to build treatment response models from patients' SC tumor transcriptomics. We start by showing that PERCEPTION successfully predicts the response to monotherapy and combination treatments in screens performed in cancer and patient-tumor-derived primary cells based on their SC-expression profiles. Our key result is that PERCEPTION successfully stratifies responders to combination therapy based on the patients’ tumor’s SC-expression, as tested in two recently published clinical trials, including multiple myeloma and breast cancer. Thirdly, studying the emergence of resistance via a recent SC non-small cell lung cancer (NSCLC) patients’ cohort, we show that PERCEPTION successfully captures and quantifies the development of patients’ resistance during treatment with tyrosine kinase inhibitors. Notably, PERCEPTION predictions markedly outperform that of bulk expression-based predictors in all these cohorts. In sum, this study provides a first-of-its-kind conceptual and computational method demonstrating the feasibility of predicting patients' response from SC gene expression of their tumors.

Abstract

Missing values in proteomic data sets have real consequences on downstream data analysis and reproducibility. Although several imputation methods exist to handle missing values, no single imputation method is best suited for a diverse range of data sets, and no clear strategy exists for evaluating imputation methods for clinical DIA-MS data sets, especially at different levels of protein quantification. To navigate through the different imputation strategies available in the literature, we have established a strategy to assess imputation methods on clinical label-free DIA-MS data sets. We used three DIA-MS data sets with real missing values to evaluate eight imputation methods with multiple parameters at different levels of protein quantification: a dilution series data set, a small pilot data set, and a clinical proteomic data set comparing paired tumor and stroma tissue. We found that imputation methods based on local structures within the data, like local least-squares (LLS) and random forest (RF), worked well in our dilution series data set, whereas imputation methods based on global structures within the data, like BPCA, performed well in the other two data sets. We also found that imputation at the most basic protein quantification level-fragment level-improved accuracy and the number of proteins quantified. With this analytical framework, we quickly and cost-effectively evaluated different imputation methods using two smaller complementary data sets to narrow down to the larger proteomic data set's most accurate methods. This acquisition strategy allowed us to provide reproducible evidence of the accuracy of the imputation method, even in the absence of a ground truth. Overall, this study indicates that the most suitable imputation method relies on the overall structure of the data set and provides an example of an analytic framework that may assist in identifying the most appropriate imputation strategies for the differential analysis of proteins.

Abstract

This paper presents the first archaeobotanical results on plant macroremains other than charcoal from the Early and Middle Epipalaeolithic site of Kharaneh IV in the Azraq basin, one of the largest Epipalaeolithic sites in the southern Levant and one of the few with evidence for multiple phases of occupation. The analysis of the substantial archaeobotanical assemblage from the site provides new insights into the local environmental conditions and how these changed throughout occupation, potentially affecting the use of the site, and it further contributes to debates about hunter-gatherer lifeways during the earlier Epipalaeolithic. A variety of potential food plant resources was identified, including several starch-rich seeds and tissues, fruits and various other wild seeds and grains. Comparison of the Kharaneh IV archaeobotanical assemblage with those from other Epipalaeolithic sites in the southern Levant reveals a number of similarities and differences. These comparisons, and especially with the contemporary nearby site of Wadi Jilat 6, support the emerging picture of an ecological mosaic within the Epipalaeolithic Azraq basin, and a general pattern of local resource use across the wider region.

Abstract

Current approaches to stage chronic liver diseases have limited utility to predict liver cancer risk. Here, we employed single nucleus RNA sequencing (snRNA-seq) to characterize the cellular microenvironment of healthy and pre-malignant livers using two distinct mouse models. Downstream analyses unraveled a previously uncharacterized disease-associated hepatocyte transcriptional state (daHep). These cells were absent in healthy livers but increasingly prevalent as chronic liver disease progressed. Copy number variation (CNV) analysis of microdissected tissue demonstrated that daHep enriched regions are riddled with structural variants, suggesting these cells represent a pre-malignant intermediary. Integrated analysis of three recent human snRNA-seq datasets confirmed the presence of a similar phenotype in human chronic liver disease and further supported its enhanced mutational burden. Importantly, we show that high daHep levels precede carcinogenesis and predict a higher risk of hepatocellular carcinoma development. These findings may change the way chronic liver disease patients are staged, surveilled and risk-stratified.

Abstract

Kruschke’s EXIT model (Kruschke, 2001b) has been very successful in explaining a variety of learning phenomena by means of selective attention. In particular, EXIT produces learned predictiveness effects (Le Pelley & McLaren, 2003), the inverse base rate effect (Kruschke, 1996; Medin & Edelson, 1988), inattention after blocking (Beesley & Le Pelley, 2011; Kruschke & Blair, 2000), differential cue use across the stimulus space (Aha & Goldstone, 1992) and conditional learned predictiveness effects (Uengoer, Lachnit, Lotz, Koenig, & Pearce, 2013). We dissect EXIT into its component mechanisms (error-driven learning, selective attention, attentional competition, rapid attention shifts and exemplar mediation of attention) and test whether simplified versions of EXIT can explain the same experimental results as the full model. Most phenomena can be explained by either rapid attention shifts or attentional competition, without the need for combining them as in EXIT. There is little evidence for exemplar mediation of attention when people learn linearly separable category structures (e.g. Kruschke & Blair, 2000; Le Pelley & McLaren, 2003); whether or not it is needed for non-linear categories depends on stimulus representation (Aha & Goldstone, 1992; Uengoer et al., 2013). On the whole, we believe that attentional competition—embodied in a model which we dub CompAct—offers the simplest explanation for the experimental results we examine.

172. Announcing Phase 4 of PAGES 2k: Hydroclimate of the Common Era

Abstract

Understanding the climate of the past two millennia (2k) remains vital for developing our wider comprehension of the climate system, including the nature and scale of recent and future anthropogenic change. Phase 4 of the PAGES 2k network will build on previous phases and take us to a new level of understanding and science-policy integration.During previous phases, PAGES 2k members compiled global networks of proxy measurements, extending records beyond the instrumental period by more than an order of magnitude, reconstructing past climate and developing new knowledge of past variability and processes. Through data-model integration with state-of-the-art Earth systems models, proxy system modelling and data assimilation, we took key steps towards a more comprehensive understanding of climate dynamics.Phase 4 will take us even further, challenging our community to turn its focus primarily towards the hydroclimate of the Common Era: performing new reconstructions and improving the interoperability, extent and scope of our data and model products. In doing so, we also seek to facilitate the translation of our science into evidence-based policy outcomes. Our overarching aim is to reconstruct hydroclimate variability over the Common Era from local to global spatial scales, at sub-annual to multi-centennial time scales. We propose to achieve this through new community-led data curation efforts and the development of new data-driven tools and practices to maximise the interoperability of convenient, efficient and widespread model/data products. We will aim for a process-level understanding of past hydroclimate events and variability by evaluating and constraining Earth system models and through data assimilation.Our coordination team places a strong emphasis on respect and inclusion, aiming to foster a diverse and equitable community. Through a ‘hub and spoke’ structure, our team will provide a facilitation, coordination and support role (the hub) for Pages 2k working groups (the spokes). We are actively seeking participation of those engaging in climate policy and climate services. Welcome to Phase 4! We warmly invite your collaborations and contributions!

Abstract

SUMMARYCurrent approaches to stage chronic liver diseases have limited utility to directly predict liver cancer risk. Here, we employed single nucleus RNA sequencing (snRNA-seq) to characterize the cellular microenvironment of healthy and chronically injured pre-malignant livers using two distinct mouse models. Analysis of 40,748 hepatic nuclei unraveled a previously uncharacterized disease-associated hepatocyte transcriptional state (daHep). These cells were absent in healthy livers, but were increasingly prevalent as chronic liver disease progressed towards hepatocarcinogenesis. Gene expression deconvolution of 1,439 human liver transcriptomes from publicly available datasets revealed that daHep frequencies highly correlate with current histopathological liver disease staging systems. Importantly, we show that high daHep levels precede carcinogenesis in mice and humans and predict a higher risk of hepatocellular carcinoma (HCC) development. This novel transcriptional signature with diagnostic and, more importantly, prognostic significance has the potential to change the way chronic liver disease patients are staged, surveilled and risk-stratified.

174. Global attribution of anthropogenic and lightning fires

Abstract

Fires can have anthropogenic or lightning origins. The spatiotemporal niches of anthropogenic and lightning fires are different. Lightning fires usually occur during a discrete apex in seasonal lightning occurrence. Conversely, anthropogenic fires have an expanded temporal niche and occur throughout the year. In addition, lightning and anthropogenic fires occupy different parts of the landscape. While human accessibility is a key determinant of anthropogenic ignitions, lightning ignitions prevail in remote landscapes.We used these differing temporal and spatial niches between anthropogenic and lightning fires to construct random forest models that attribute causes, lightning vs. anthropogenic, to global fire activity. We built two separate models. The first model predicts the fraction of lightning fires, whereas the second model predicts the fraction of burned area from lightning. Our model ingests two geospatial predictor variables that quantify the differences between the temporal and spatial niches of lightning and anthropogenic fires. The first predictor is the seasonal correlation between lightning and burned area. The second predictor is the fraction of low impact land. These fire cause predictors capture 47 % of the spatial variability in ignition cause, and 40 % of the spatial variability in burned area cause, compared to reference data from six different parts of the world.Our global fire cause attribution contrasts savannas and agricultural lands with human-dominated fire regimes from temperate and boreal forests with lightning-dominated fire regimes. Our global fire cause attribution can be implemented in fire and Earth system models to further optimize projections of future fire activity under changing socio-economic and climatological conditions.

Abstract

OBJECTIVE: Medication maintenance is critical in the management of asthma. We investigated the differences in electronic health record (EHR) documentation of medication refills for Spanish- and English-speaking Latino children and non-Hispanic white children by examining rates of albuterol rescue inhaler refills from 2005 to 2017, and and inhaled corticosteroid refills from 2015 to 2017 in a multi-state network of community health centers (CHCs). METHODS: We used data from the ADVANCE network of CHCs. Our sample consisted of children aged 3–17, with a diagnosis of asthma and either albuterol or inhaled corticosteroid prescriptions (n = 39,162; n = 4,738 children, respectively). Negative binomial regression was used to calculate rates of refills per prescription adjusted for relevant patient-level covariates. Analyses stratified by asthma severity were also conducted. RESULTS: English-speaking Latino children had lower rates of albuterol refills compared with non-Hispanic white children (rate ratio [RR] = 0.88, 95% confidence interval [CI]: 0.80–0.98), a trend that persisted among children with moderate/severe persistent asthma severity (RR = 0.85, 95% CI: 0.76–0.95). Spanish-speaking Latino and non-Hispanic white children had similar albuterol refills. Inhaled corticosteroid refill rates were comparable between all groups. CONCLUSIONS: In a multi-state network, these findings suggest that CHCs deliver equitable asthma care related to prescription refills between their Latino and white patients, but there is still opportunity for providers to ensure that their English-speaking Latino patients have access to necessary emergency asthma medication.

Abstract

Funder: Funding details are provided in the Supplementary Material, Polygenic risk scores (PRS) for epithelial ovarian cancer (EOC) have the potential to improve risk stratification. Joint estimation of Single Nucleotide Polymorphism (SNP) effects in models could improve predictive performance over standard approaches of PRS construction. Here, we implemented computationally-efficient, penalized, logistic regression models (lasso, elastic net, stepwise) to individual level genotype data and a Bayesian framework with continuous shrinkage, ���select and shrink for summary statistics��� (S4), to summary level data for epithelial non-mucinous ovarian cancer risk prediction. We developed the models in a dataset consisting of 23,564 non-mucinous EOC cases and 40,138 controls participating in the Ovarian Cancer Association Consortium (OCAC) and validated the best models in three populations of different ancestries: prospective data from 198,101 women of European ancestry; 7,669 women of East Asian ancestry; 1,072 women of African ancestry, and in 18,915 BRCA1 and 12,337 BRCA2 pathogenic variant carriers of European ancestry. In the external validation data, the model with the strongest association for non-mucinous EOC risk derived from the OCAC model development data was the S4 model (27,240 SNPs) with odds ratios (OR) of 1.38(95%CI:1.28���1.48,AUC:0.588) per unit standard deviation, in women of European ancestry; 1.14(95%CI:1.08���1.19,AUC:0.538) in women of East Asian ancestry; 1.38(95%CI:1.21-1.58,AUC:0.593) in women of African ancestry; hazard ratios of 1.37(95%CI:1.30���1.44,AUC:0.592) in BRCA1 pathogenic variant carriers and 1.51(95%CI:1.36-1.67,AUC:0.624) in BRCA2 pathogenic variant carriers. Incorporation of the S4 PRS in risk prediction models for ovarian cancer may have clinical utility in ovarian cancer prevention programs.

Abstract

Traditional efforts around improving assessment often center on the teacher as the evaluator of work rather than the students. These assessment efforts typically focus on measuring learning rather than stimulating, promoting, or producing learning in students. This paper summarizes a study of a large sample of undergraduate students (n = 550) in an entry-level design-thinking course who engaged with Adaptive Comparative Judgment (ACJ), a form of assessment, as a learning mechanism. Following random assignment into control and treatment sections, students engaged in identical activities with the exception of a 20-minute intervention we call learning by evaluating (LbE). Prior to engaging in a Point Of View (POV) creation activity, treatment group students engaged in LbE by viewing pairs of previously-collected POV statements through ACJ; in each case they viewed two POV statements side-by-side and selected the POV statement they believed was better. Following this experience, students created their own POV statements and then the final POV statements, from both the control and treatment students, were collected and evaluated by instructors using ACJ. In addition, qualitative data consisting of student comments, collected during ACJ comparisons, were coded by the researchers to further explore the potential for the students to use class knowledge while engaging in the LbE review of peer work. Both the quantitative and qualitative data sets were analyzed to investigate the impact of the LbE activity. Consistent with other ACJ research findings, significant positive learning gains were found for students who engaged in the intervention. Researchers also noted that these findings did not indicate the actual quality of the assignments, meaning the while students who engaged in the LbE intervention were better than their peers, they were not necessarily “good” at the assignment themselves. Discussion of these findings and areas for further inquiry are presented.

Abstract

Glacier-fed lakes on the Tibetan Plateau (TP) have undergone rapid expansions since the late 1990s, concurrent with the changing climate. However, the dominant cause(s) of lake area increases is still debated. To identify the drivers of lake expansion, we studied Aweng Co, a glacier-fed lake in the western TP, where surface area has increased (0.74 km2 year−1) since the late 1970s and most rapidly (0.998 km2 year−1) since the late 1990s. A water balance model was used to clarify the reasons for increased lake water volume, supported by stable isotope hydrology and the δ18O change recorded in recent sediments. Results showed that glacial meltwater probably had the biggest impact on changes in Aweng Co lake level in recent decades, but that precipitation was also an important contributor. Our study shows that δ18O of carbonate (δ18Ocarb) has great potential for indicating source changes of water supply in such lakes, but there is a need to be cautious when interpreting δ18Ocarb due to the influence of multiple hydrological factors, which can change in dominance over time.

Abstract

AIMS Necrotising enterocolitis (NEC) is a disease associated with high mortality and morbidity, low birthweight and prematurity are risk factors. This study reports outcomes of babies having emergency laparotomy for NEC, examining institutional trends and exploring impact of multiple variables on mortality at 30 days and 1 year post-operatively. METHODS Case records of babies with ICD coding for NEC were examined from 2000 to 2015. After exclusions, 243 cases were identified-confirmed by operative findings and histology. Cohort demographics and trends in mortality were investigated, and the relationship of common variables to mortality was modelled with univariate and multivariate logistic regression to generate a mortality prediction tool. RESULTS Mean gestational age was 28 + 4 weeks. A 30-day mortality was 18.9%. Gestation, birthweight and area of bowel affected were significant of outcome (mortality), and the presence of pre-operative pneumoperitoneum was strongly correlated. Year of surgery and congenital cardiac pathology requiring intervention were not significant. Using multivariate regression modelling, a mortality outcome prediction tool has been developed. CONCLUSION Good survival following operation for NEC (>70%) is feasible, even in those babies born extremely premature ( 32 weeks), mortality is uncommon.

Abstract

Reconstructions of global hydroclimate during the Common Era (CE; the past ∼2000 years) are important for providing context for current and future global environmental change. Stable isotope ratios in water are quantitative indicators of hydroclimate on regional to global scales, and these signals are encoded in a wide range of natural geologic archives. Here we present the Iso2k database, a global compilation of previously published datasets from a variety of natural archives that record the stable oxygen (δ18O) or hydrogen (δ2H) isotopic compositions of environmental waters, which reflect hydroclimate changes over the CE. The Iso2k database contains 759 isotope records from the terrestrial and marine realms, including glacier and ground ice (210); speleothems (68); corals, sclerosponges, and mollusks (143); wood (81); lake sediments and other terrestrial sediments (e.g., loess) (158); and marine sediments (99). Individual datasets have temporal resolutions ranging from sub-annual to centennial and include chronological data where available. A fundamental feature of the database is its comprehensive metadata, which will assist both experts and nonexperts in the interpretation of each record and in data synthesis. Key metadata fields have standardized vocabularies to facilitate comparisons across diverse archives and with climate-model-simulated fields. This is the first global-scale collection of water isotope proxy records from multiple types of geological and biological archives. It is suitable for evaluating hydroclimate processes through time and space using large-scale synthesis, model–data intercomparison and (paleo)data assimilation. Abstract

Background and Objectives In gerontological research and practice, an increasing amount of attention is being paid to lesbian, gay, bisexual, transgender, and queer (LGBTQ) older people and how their experiences differ from their heterosexual and cisgender counterparts. However, LGBTQ older adults themselves are not a homogenous group. Moreover, as the immigrant populations in industrialized nations age, the number of LGBTQ older adults from ethnic minority backgrounds will only grow. This systematic review hence investigates the experiences of LGBTQ ethnic minority older adults. Research Design and Methods Following the Preferred Reporting Items for Systematic Reviews and Meta-Analysis guidelines, we conducted a systematic search in 5 databases for English peer-reviewed studies. The retrieved articles were coded and analyzed inductively using an intersectional framework to tease out the varying influences of ethnicity, age, gender, and sexual identity on the LGBTQ ethnic minority older adults’ experiences. Results A total of 30 articles across 21 studies (13 qualitative, 7 quantitative, and 1 mixed-methods) were identified. Six key themes emerged from the studies: stigma and discrimination; isolation, support and belonging; interactions with services and institutions; self-acceptance, resilience, and agency; mental health and well-being; and uncertain futures. Discussion and Implications The experiences of LGBTQ ethnic minority older adults echo those of LGBTQ ethnic majority older adults when they are shaped by gender and sexual identity factors. Nevertheless, significant differences in experiences—both positive and negative—emerge when cultural and ethnicity-related factors come to the fore. These findings emphasize the need for intersectional aging policies and services that go beyond catering for LGBTQ older adults to include the diversity within this subpopulation.

Abstract

To examine the usefulness of the Edinburgh Cognitive and Behavioral Amyotrophic Lateral Sclerosis (ALS) Screen (ECAS) as a cognitive screening tool for the detection of behavioral variant frontotemporal dementia (bvFTD). A secondary aim was to determine whether people with FTD combined with ALS (ALS-FTD) exhibit a similar ECAS profile to that of people with bvFTD alone. Methods: Patients with ALS-FTD and bvFTD and healthy controls were recruited. Participants were administered the ECAS, which comprises tests of language, verbal fluency, executive functions, memory, and visual-spatial functions. They also carried out analogous, full-length cognitive tests that examine naming, spelling, sentence completion, and social cognition skills. Results: The study cohort comprised 20 ALS-FTD patients, 23 with bvFTD, and 30 controls. Highly significant group differences were elicited for all cognitive domains, reflecting poorer performance in patients compared to controls. No significant differences in overall test scores were found between ALS-FTD and bvFTD patients, although ALS-FTD patients showed a higher frequency of impairment on verbal fluency. Correlative analyses revealed inter-relationships in patients (but not controls) between scores in different domains, most marked in bvFTD. There were strong correlations between performance on ECAS subtests and analogous cognitive tasks. Conclusion: The ECAS is a sensitive and valuable tool for the assessment of FTD. Executive, language and behavioral breakdown may, however, compromise performance in other cognitive domains, reducing the specificity of the ‘frontotemporal’ cognitive profile. Subtle differences observed between ALS-FTD and bvFTD raise questions regarding the precise relationship between bvFTD with and without ALS.

Abstract

This study detected, mapped and analysed lineaments from Landsat-8 satellite images of the coastal area of Kribi-Campo, Cameroon. Kribi, a fast-growing coastal city with a youthful population whose main industry is agriculture, is currently under great pressure due to the influx of internally displaced persons, victims of the current socio-political crisis in the western part of Cameroon. With the growing demand for food and water resources, it has been reported that saline soils and groundwater are affecting the agricultural productivity and domestic water supply of the area. This paper investigates the lineaments network in this area to understand its spatial distribution, its extent and the potential effect on inland intrusion of saline water. The datasets, including Landsat-8 imagery, geological data and a digital elevation model were processed using principal component analysis and filtering techniques. Linear features were enhanced based on textural changes of false colour composite images to describe fractures, faults and hydrology features. 390 effective lineaments were extracted automatically and manually from those images and analysed based on length, density and orientation using statistical analysis. But before this processing, non-effective lineaments that may correspond to terraces, linear valleys, cliffs, or tonal contrast as well as to human activities have been deleted. The effective lineaments range in length from 95 to 4910 m with an average of 1171 m. Although the lineaments are not isotropic, their perpendicular orientations to the coast suggest potential intrusion of saline water into regional groundwater resources and farmlands. The paper presents a combined technique of geospatial and statistical analysis in the interpretation of lineament effects which have the potential to inform on salinity intrusion in coastlands. The electrical conductivity of 50 groundwater samples collected in the study area are used to validate the impact of lineaments on seawater intrusion.

Abstract

Background Working effectively with Aboriginal and Torres Strait Islander people is important for maximising the effectiveness of a health care interaction between and Aboriginal and Torres Strait Islander patients and a health professional. This paper presents a framework to guide health professional practice in Aboriginal and Torres Strait Islander health. Methods This qualitative study was based in a social constructionist epistemology and was guided by a critical social research methodology. Two methods were employed: interviews with Aboriginal health workers and allied health professionals about their experiences of working together in Aboriginal health, and an auto-ethnography conducted by the researcher, a non-Aboriginal dietitian and researcher who worked closely with two Aboriginal communities while undertaking this research. Results Interviews were conducted with 44 allied health professionals and Aboriginal health workers in 2010. Critical Social research, which involves the deconstruction and reconstruction of data, was used to analyse data and guided the evolution of themes. Strategies that were identified as important to guide practice when working respectfully in Aboriginal health included: Aboriginal and non-Aboriginal people working with Aboriginal health workers, using appropriate processes, demonstrating commitment to building relationships, relinquishing control, having an awareness of Aboriginal history, communication, commitment, flexibility, humility, honesty, and persistence. Reciprocity and reflection/reflexivity were found to be cornerstone strategies from which many other strategies naturally followed. Strategies were grouped into three categories: approach, skills and personal attributes which led to development of the Framework. Conclusions The approach, skills and personal attributes of health professionals are important when working in Aboriginal health. The strategies identified in each category provide a Framework for all health professionals to use when working with Aboriginal and Torres Strait Islander people.

Abstract

Objectives To explore what actually happens in relation to physical activity promotion in primary and junior schools within one unitary authority and to relate this to Public Health England (PHE) promising principles of practice to promote physical activity in schools. Study design A qualitative approach was undertaken to explore practice in all primary and junior schools in the unitary authority of Southampton. Methods All primary (n = 36) and junior (n = 8) schools in Southampton were involved in the study. Publicly available primary physical education and sport premium (PESP) funding reports (n = 36) alongside a school survey (n = 14) were collated. The collated qualitative data set was semantically coded and then a multilayered approach including identification, reviewing, defining and naming meaningful and important themes were inductively developed. The inductively developed themes were then fitted in relation to PHE eight promising principles. Results There was evidence of practice across all eight promising principles although this varied in depth and scale. There was one set of data that did not fit appropriately within the PHE eight promising principles and warranted its own category broadly termed ‘rewards to recognise physical activity’. There was widespread evidence of PESP funding providing increased provision, variety and quality of sport opportunities but limited evidence of physical activity practice or programmes targeting the least active. Two different approaches in relation to ensuring a skilled workforce materialised continuing professional development which reflected in impact statements linked to increased confidence to deliver and quality of physical education versus outsourcing to specialists with little impact cited other than offering specialist or diverse sports. Conclusions The study demonstrated that the PHE eight promising principles of practice was a useful framework in relation to current practice, although a ninth promising principle of rewarding physically active behaviour should be considered. The two key themes that need to be addressed for the ambitions established in the new school sport and activity action plan to be deliverable, with PESP funding as a driver, are skilled workforce and development of a wider understanding of what physical activity is.

Abstract

Thermogravimetric analysis (TGA) is a technique which can probe chemisorption of substrates onto metal organic frameworks. A TGA method was developed to examine the catalytic oxidation of S-nitrosoglutathione (GSNO) by the MOF H3[(Cu4Cl)3(BTTri)8] (abbr. Cu-BTTri; H3BTTri = 1,3,5-tris(1H-1,2,3-triazol-5-yl)benzene), yielding glutathione disulfide (GSSG) and nitric oxide (NO). Thermal analysis of reduced glutathione (GSH), GSSG, GSNO, and Cu-BTTri revealed thermal resolution of all four analytes through different thermal onset temperatures and weight percent changes. Two reaction systems were probed: an aerobic column flow reaction and an anaerobic solution batch reaction with gas agitation. In both systems, Cu-BTTri was reacted with a 1 mM GSH, GSSG, or GSNO solution, copiously rinsed with distilled-deionized water (dd-H2O), dried (25 °C, < 1 Torr), and assessed by TGA. Additionally, stock, effluent or supernatant, and rinse solutions for each glutathione derivative within each reaction system were assessed by mass spectrometry (MS) to inform on chemical transformations promoted by Cu-BTTri as well as relative analyte concentrations. Both reaction systems exhibited chemisorption of glutathione derivatives to the MOF by TGA. Mass spectrometry analyses revealed that in both systems, GSH was oxidized to GSSG, which chemisorbed to the MOF whereas GSSG remained unchanged during chemisorption. For GSNO, chemisorption to the MOF without reaction was observed in the aerobic column setup, whereas conversion to GSSG and subsequent chemisorption was observed in the anaerobic batch setup. These findings suggest that within this reaction system, GSSG is the primary adsorbent of concern with regards to strong binding to Cu-BTTri. Development of similar thermal methods could allow for the probing of MOF reactivity for a wide range of systems, informing on important considerations such as reduced catalytic efficiency from poisoning, recyclability, and loading capacities of contaminants or toxins with MOFs.

Abstract

This paper reports on a search for an extension to the scalar sector of the standard model, where a new CP-even (odd) boson decays to a Z boson and a lighter CP-odd (even) boson, and the latter further decays to a b quark pair. The Z boson is reconstructed via its decays to electron or muon pairs. The analysed data were recorded in proton-proton collisions at a center-of-mass energy $$ \sqrt{s} $$ s = 13 TeV, collected by the CMS experiment at the LHC during 2016, corresponding to an integrated luminosity of 35.9 fb−1. Data and predictions from the standard model are in agreement within the uncertainties. Upper limits at 95% confidence level are set on the production cross section times branching fraction, with masses of the new bosons up to 1000 GeV. The results are interpreted in the context of the two-Higgs-doublet model.

Abstract

This article explores the development of an integrated programme of initial teacher education premised upon a clinical practice approach that aims to develop student teachers as research-informed, reflective practitioners. The article outlines key decisions in developing an integrated approach to a new programme of initial teacher education that is both research-infused and research-informed. It highlights the centrality of a Partnership approach to the design of the programme and outlines the co-construction process that underpinned it. The article also explains how University subject experts are integral to the ITE programme providing both subject and research expertise. The main features of this integrated programme of ITE are outlined, and the article concludes by offering some reflections and considerations about an approach to initial teacher education that is genuinely research informed and grounded in teacher enquiry.

Abstract

Culverts reduce connectivity for aquatic animals by being both a hydraulic and physical barrier. However, altered light intensity may also be a behavioural barrier to fish movement, especially for diurnal species that have adapted to moving when it is light. We propose that knowledge of optical physiology and fish behaviour, two important mechanisms underpinning movement, can inform efforts to improve fish movement through culverts. We firstly review the sensory systems of fish with reference to visual sense and explore how this affects fish movement. We then highlight theoretical knowledge that can help us understand fish behaviour and the potential mismatch between the conditions under which fish have evolved and altered conditions within culverts. We describe potential knowledge gaps and directions for future research to improve our understanding of how culverts may affect fish movement. Finally, we explore the potential costs and ecological benefits of different mitigation options to identify those with the most promise for managing the light environment in culverts to facilitate movement. For researchers and managers exploring this subject, we suggest an approach that: (a) identifies light requirements for movement by different species, (b) tests movement under different light conditions and (c) considers an integrative assessment method for testing fish behaviour around culverts. Understanding how optical physiology, fish behaviour and culvert design influence fish movement can improve connectivity for a range of species.

Abstract

Context As many as 75% of patients with polycystic ovary syndrome (PCOS) are estimated to be unidentified in clinical practice. Objective Utilizing polygenic risk prediction, we aim to identify the phenome-wide comorbidity patterns characteristic of PCOS to improve accurate diagnosis and preventive treatment. Design, Patients, and Methods Leveraging the electronic health records (EHRs) of 124 852 individuals, we developed a PCOS risk prediction algorithm by combining polygenic risk scores (PRS) with PCOS component phenotypes into a polygenic and phenotypic risk score (PPRS). We evaluated its predictive capability across different ancestries and perform a PRS-based phenome-wide association study (PheWAS) to assess the phenomic expression of the heightened risk of PCOS. Results The integrated polygenic prediction improved the average performance (pseudo-R2) for PCOS detection by 0.228 (61.5-fold), 0.224 (58.8-fold), 0.211 (57.0-fold) over the null model across European, African, and multi-ancestry participants respectively. The subsequent PRS-powered PheWAS identified a high level of shared biology between PCOS and a range of metabolic and endocrine outcomes, especially with obesity and diabetes: “morbid obesity”, “type 2 diabetes”, “hypercholesterolemia”, “disorders of lipid metabolism”, “hypertension”, and “sleep apnea” reaching phenome-wide significance. Conclusions Our study has expanded the methodological utility of PRS in patient stratification and risk prediction, especially in a multifactorial condition like PCOS, across different genetic origins. By utilizing the individual genome–phenome data available from the EHR, our approach also demonstrates that polygenic prediction by PRS can provide valuable opportunities to discover the pleiotropic phenomic network associated with PCOS pathogenesis.

Abstract

A series of well-defined mono- and dimeric Zn(II)-complexes were prepared and fully characterised by X-ray crystallography and NMR spectroscopy. Their application to the ROP of rac-LA to produce biocompatible atactic PLA was demonstrated in both solution and under industrially preferred melt conditions at 180 °C with a [rac-LA] : [Init] : [BnOH] of 3000 : 1 : 10. Exceptional activity was observed in all instances, albeit with generally poor molecular weight control (Mn) and broad dispersities (Đ), with dimers outperforming their monomeric counterparts. The propensity of all Zn(II)-complexes to facilitate PLA degradation into methyl lactate (Me-LA) under mild conditions is also demonstrated. Zn(2–3)2 were identified as the outstanding candidates, achieving full conversion to Me-LA within 8 h at 80 °C in THF. Further degradation kinetic analysis revealed Zn(2–3)2 to have kapp values of 0.63 ± 0.051 and 0.44 ± 0.029 h−1 for the rate of consumption of PLA respectively.
