Abstract

Intermittent high glucose enhances apoptosis in human umbilical vein endothelial cells in culture. Am J Physiol Endocrinol Metab 281: E924-E930, 2001.--To explore the effect of fluctuating glucose on endothelial cells, human umbilical vein endothelial cells were incubated for 14 days in media containing different glucose concentrations: 5 mmol/l, 20 mmol/l, or a daily alternating 5 or 20 mmol/l glucose. Apoptosis was studied by different methods: viability assay, cell cycle analysis, DNA fragmentation, and morphological analysis. Furthermore, the levels of Bcl-2 and Bax, well known proteins involved in apoptosis, were evaluated. Stable high glucose induced apoptosis in human umbilical vein endothelial cells, a phenomenon accompanied by a significant decrease of Bcl-2 and a simultaneous increase of Bax expression. However, apoptosis was enhanced in human umbilical vein endothelial cells exposed to intermittent, rather than constant, high glucose concentration. In this condition, Bcl-2 was not detectable, whereas Bax expression was significantly enhanced. These findings suggest that variability in glycemic control could be more deleterious to endothelial cells than a constant high concentration of glucose. intermittent glucose; endothelium; apoptosis; Bcl-2 protein; Bax protein

Abstract

The Italian waste sector continues to make a growing trend, both in terms of costs and opportunities for the community, showing difficulties by the public administration to ensure an environmental improvement and development strategy, thereby determining negative externalities in the social and economic implications of the territory. The paper aims to implement an integrated waste management system, through which define innovative solutions in the relationship between economic processes and environmental sustainability, using the network analysis as a basis of inquiry. Therefore, the methodology used is based on the analysis of the issues which contributing to immobility and inefficiency of the waste management Italian system, considering the main European regulatory elements and using the different indicators related to the singular driver associated (public health, environmental, resource scarcity, value of waste). From this analysis it has been possible to identify governance guidelines aimed to improvement the treatment and disposal waste system and, consequently, to reduce, in terms of quantity, the transfer to landfill. The paper does not seek to provide a "one best way" in the waste management process because, as shown by previous studies, exist a high dependence between the management system and local specificities.
