In addition to its well-characterized function as a sequence specific transcriptional activator, there is growing evidence that the p53 tumor suppressor protein is also a sequence-specific transcriptional repressor. The concept that a transcription factor can exist as both an activator and a repressor of transcription is not new. In fact, it is the rare transcription factor that can perform only one of these functions. The initial challenges for individuals studying the repression function of p53 have been met; that is, a set of genes whose expression is decreased following p53 induction has been identified. These include the genes encoding alpha-fetoprotein, bcl-2, cyclin B, cdc2, cdc25, Map4, Mdr1, presenilin-1, and survivin, as well as others. The promoters for many of these genes have been cloned, and p53 has been found to bind to sites within these promoters using assays that measure binding in vivo, such as chromatin immunoprecipitation. The p53 binding sites in these promoters have been identified, and in many cases the mechanism whereby p53 represses transcription, for example by promoter occlusion or by recruitment of histone deacetylases, has been elucidated. The current challenge is to create mutant forms of p53 that are capable of repressing transcription but not activating it (or vice versa), such that the contribution of this activity to tumor suppression by p53 can be effectively delineated. Differential display is a reverse-transcription polymerase chain reaction (RT-PCR) technique that was introduced in 1992 by Liang and Pardee (1). In this technique, messenger RNA 3′ termini are amplified using an anchored oligo-dT primer and a series of arbitrary 13-mers; a single round of cDNA synthesis is followed by PCR amplification in the presence of radiolabeled nucleotide, resulting in the amplification of subsets of mRNAs expressed in the cell. This complex PCR product is resolved on a denaturing polyacrylamide gel; following autoradiography, gene expression is detected as a ladder of cDNAs that differ in size by a single nucleotide. Theoretically, the use of different sets of primers from both directions in discrete RT-PCR reactions allows for the examination of the majority of expressed genes within the cell. This chapter will focus first on a brief review of the use of differential display to identify p53 target genes, as well as secondary tumor suppressor genes important for tumor progression. This will be followed by an overview of the methodology of differential display, along with recent advances in this technique; a more comprehensive analysis can be found in Methods in Molecular Biology, Volume 85: Differential Display Methods and Protocols, edited by Liang and Pardee (2). An outline of the methodology involved in differential display is provided in Fig. 1, and individual steps are described below; typically these steps follow the protocol described by Liang and Pardee (1,2), but in some instances changes have been made and noted. - Abstract
There is substantial evidence in the literature that, in addition to functioning as an activator of transcription, the p53 tumor suppressor protein can also function as a sequence-specific transcriptional repressor of a separate set of genes. However, elucidation of the mechanism whereby p53 functions as a transcriptional repressor has been obscured by the use of artificial assays to measure this activity; these assays include transient transfection analyses, where both p53 and target promoters are overexpressed. This chapter describes alternative approaches for the definition of sequence elements that mediate transcriptional repression by p53. These include the McKay (immunobinding) assay, which measures the in vitro binding of large fragments of DNA, as well as chromatin immunoprecipitations (ChIPs), which measure in vivo binding. The use of such assays should better define the mechanism of transcriptional repression by p53 and should aid in the elucidation of the contribution of this activity to p53-dependent growth arrest and programmed cell death (apoptosis). The p53 tumor suppressor gene has a common polymorphism at codon 72 that alters its function. We previously reported that the proline 72 polymorphic variant of p53 (P72) demonstrates increased ability to transactivate a subset of genes, relative to arginine 72 (R72); one of these genes is macrophage colony stimulating factor (CSF1). At present, the mechanism(s) underlying the increased transcriptional activity of P72 toward genes like CSF1 have not been completely elucidated. Additionally, the consequences of increased transcription of genes like CSF1 by the P72 variant to the downstream p53 pathway are unknown. In this report, we address these issues. We show that the CSF1 gene contains a conserved binding site for p53, and interestingly that the P72 variant shows increased ability to bind to this site. Moreover, we show that increased CSF1/CSF1R signaling in P72 cells feeds back on the p53 pathway to enhance p53 phosphorylation, levels, and transactivation of target genes, particularly the cyclin-dependent kinase inhibitor p21 (CDKN1A). This leads to an increase in p53-mediated growth arrest, along with a concomitant decrease in apoptosis. Notably, the CSF1/CSF1R signaling axis is overexpressed in several epithelial cancers, and there is clinical evidence that this pathway plays a role in radio-resistance of some cancers. We show that cells expressing CSF1 and CSF1R are indeed radio-resistant, and further, that this effect requires p53. These combined data are the first to implicate the CSF1/CSF1R pathway in the decision between p53-mediated growth arrest and apoptosis. They are also the first to highlight a cytokine as influential in cell fate determined by p53 in epithelial cells. Finally, these data may explain the association of the P72 variant and the CSF1/CSF1R pathway with increased senescence and radio-resistance in some epithelial tumor types. [ABSTRACT FROM AUTHOR]

Tumor suppressor p53 is crucial for embryonic implantation through transcriptional up-regulation of uterine leukemia inhibitory factor (LIF). This article reports that p53 and estrogen receptor α were activated in endometrial tissues during implantation to coordinately regulate LIF production. By using human p53 knockin (Hupki) mice carrying a single nucleotide polymorphism (SNP) at codon 72 (arginine/proline), the arginine allele was demonstrated to produce higher uterine LIF levels during implantation than the proline allele. In humans, the diversity of haplotypes of the p53 gene has decreased during evolution, because the arginine allele, existing in only a subset of haplotypes, is under positive selection. This observation is consistent with previous results showing that the proline allele is enriched in patients undergoing in vitro fertilization (IVF). Studies with p63- and p73-knockout mice have demonstrated the involvement of p63 and p73 in female reproduction and their roles in egg formation and apoptosis (p63) and spindle checkpoint (p73) in female mice. Here, the role of p63 and p73 in human reproduction was investigated. Selected alleles of SNPs in p63 and p73 genes were enriched in IVF patients. The tumor suppressor protein p53 plays an important role in maternal reproduction in mice through transcriptional regulation of leukemia inhibitory factor (LIF), a cytokine crucial for blastocyst implantation. To determine whether these observations could be extended to humans, a list of single-nucleotide polymorphisms (SNPs) in the p53 pathway that can modify the function of p53 was assembled and used to study their impact on human fertility. The p53 allele encoding proline at codon 72 (P72) was found to be significantly enriched over the allele encoding arginine (R72) among in vitro fertilization (IVF) patients. The P72 allele serves as a risk factor for implantation failure. LIF levels are significantly lower in cells with the P72 allele than in cells with the R72 allele, which may contribute to the decreased implantation and fertility associated with the P72 allele. Selected alleles in SNPs in LIF, Mdm2, Mdm4, and Hausp genes, each of which regulates p53 levels in cells, are also enriched in IVF patients. Interestingly, the role of these SNPs on fertility was much reduced or absent in patients older than 35 years of age, indicating that other functions may play a more important role in infertility in older women. The association of SNPs in the p53 pathway with human fertility suggests that p53 regulates the efficiency of human reproduction. These results also provide a plausible explanation for the evolutionary positive selection of some alleles in the p53 pathway and demonstrate the alleles in the p53 pathway as a good example of antagonistic pleiotropy.
