Abstract

Microbathymetry data, in-situ observations, and sampling along the 13°20’N and 13°20’N oceanic core complexes (OCCs) reveal mechanisms of detachment fault denudation at the seafloor, links between tectonic extension and mass wasting, and expose the nature of corrugations, ubiquitous at OCCs. In the initial stages of detachment faulting and high angle fault scarps show extensive mass-wasting that reduces their slope. Flexural rotation further lowers scarp slope, hinders mass wasting, resulting in morphologically complex chaotic terrain between the breakaway and the denuded corrugated surface. Extension and drag along the fault plane uplifts a wedge of hangingwall material (apron). The detachment surface emerges along a continuous moat that sheds rocks and covers it with unconsolidated rubble, while local slumping emplaces rubble ridges overlying corrugations. The detachment fault zone is a set of anostomosed slip planes, elongated in the along-extension direction. Slip planes bind fault rock bodies defining the corrugations observed in microbathymetry and sonar. Fault planes with extension-parallel stria are exposed along corrugation flanks, where the rubble cover is shed. Detachment fault rocks are primarily basalt fault breccia at 13°20‘N OCC, and gabbro and peridotite at 13°30’N, demonstrating that brittle strain localization in shallow lithosphere form corrugations, regardless of lithologies in the detachment zone. Finally, faulting and volcanism dismember the 13°30’N OCC, with widespread present and past hydrothermal activity (Semenov fields), while the Irinovskoe hydrothermal field at the 13°20’N core complex suggests a magmatic source within the footwall. These results confirm the ubiquitous relationship between hydrothermal activity and oceanic detachment formation and evolution.

Abstract

KatG is a bifunctional, heme-dependent enzyme in the front-line defense of numerous bacterial and fungal pathogens against H2O2-induced oxidative damage from host immune responses. Contrary to the expectation that catalase and peroxidase activities should be mutually antagonistic, peroxidatic electron donors (PxEDs) enhance KatG catalase activity. Here, we establish the mechanism of synergistic cooperation between these activities. We show that at low pH values KatG can fully convert H2O2 to O2 and H2O only if a PxED is present in the reaction mixture. Stopped-flow spectroscopy results indicated rapid initial rates of H2O2 disproportionation slowing concomitantly with the accumulation of ferryl-like heme states. These states very slowly returned to resting (i.e. ferric) enzyme, indicating that they represented catalase-inactive intermediates. We also show that an active-site tryptophan, Trp-321, participates in off-pathway electron transfer. A W321F variant in which the proximal tryptophan was replaced with a non-oxidizable phenylalanine exhibited higher catalase activity and less accumulation of off-pathway heme intermediates. Finally, rapid freeze-quench EPR experiments indicated that both WT and W321F KatG produce the same methionine–tyrosine–tryptophan (MYW) cofactor radical intermediate at the earliest reaction time points and that Trp-321 is the preferred site of off-catalase protein oxidation in the native enzyme. Of note, PxEDs did not affect the formation of the MYW cofactor radical but could reduce non-productive protein-based radical species that accumulate during reaction with H2O2. Our results suggest that catalase-inactive intermediates accumulate because of off-mechanism oxidation, primarily of Trp-321, and PxEDs stimulate KatG catalase activity by preventing the accumulation of inactive intermediates.

Abstract

Properly assessing the extent and magnitude of fault ruptures associated with large earthquakes is critical for understanding fault behavior and associated hazard. Submarine faults can trigger tsunamis, whose characteristics are defined by the geometry of seafloor displacement, studied primarily through indirect observations (e.g., seismic event parameters, seismic profiles, shipboard bathymetry, coring) rather than direct ones. Using deep-sea vehicles, we identify for the first time a marker of coseismic slip on a submarine fault plane along the Roseau Fault (Lesser Antilles), and measure its vertical displacement of ∼0.9 m in situ. We also map recent fissuring and faulting of sediments on the hangingwall, along ∼3 km of rupture in close proximity to the fault's base, and document the reactivation of erosion and sedimentation within and downslope of the scarp. These deformation structures were caused by the 2004 Mw 6.3 Les Saintes earthquake, which triggered a subsequent tsunami. Their characterization informs estimates of earthquake recurrence on this fault and provides new constraints on the geometry of fault rupture, which is both shorter and displays locally larger coseismic displacements than available model predictions that lack field constraints. This methodology of detailed field observations coupled with near-bottom geophysical surveying can be readily applied to numerous submarine fault systems, and should prove useful in evaluating seismic and tsunamigenic hazard in all geodynamic contexts. © 2016

Two Holstein heifers were referred for non-weight bearing lameness. The physical examination and radiographic findings were diagnostic of tarsal luxation. Treatment and outcome are reported. A closed reduction was successfully performed in 1 heifer. The second animal was euthanized after attempts to reduce and stabilize the joint.

Abstract

Detection of a gamma-ray source above 300 GeV is reported, confirming the unidentified source MGRO J1908+06, discovered by the Milagro collaboration at a median energy of 20 TeV. The source was observed during 27 h as part of the extension of the H.E.S.S. Galactic plane survey to longitudes >30degr. HESS J1908+063 is detected at a significance level of 10.9 s with an integral flux above 1 TeV of (3.76+-0.29stat+-0.75sys)x10^-12 ph cm^-2 s^-1, and a spectral photon index Gamma = 2.10+-0.07stat+-0.2sys. The positions and fluxes of HESS 1908+063 and MGRO J1908+06 are in good agreement. Possible counterparts at other wavelengths and the origin of the gamma-ray emission are discussed. The nearby unidentified GeV source, GRO J1908+0556 (GeV) which also remains unidentified and the new Fermi pulsar 0FGL J1907.5+0617, may be connected to the TeV source., Accepted for publication on Astron. Abstract

Aside from abasic sites and ribonucleotides, the DNA adduct N7-methyl deoxyguanosine (N7-CH3 dG) is one of the most abundant lesions in mammalian DNA. Because N7-CH3 dG is unstable, leading to deglycosylation and ring-opening, its miscoding potential is not well-understood. Here, we employed a 2'-fluoro isostere approach to synthesize an oligonucleotide containing an analog of this lesion (N7-CH3 2'-F dG) and examined its miscoding potential with four Y-family translesion synthesis DNA polymerases (pols): human pol (hpol) η, hpol κ, and hpol ι and Dpo4 from the archaeal thermophile Sulfolobus solfataricus. We found that hpol η and Dpo4 can bypass the N7-CH3 2'-F dG adduct, albeit with some stalling, but hpol κ is strongly blocked at this lesion site, whereas hpol ι showed no distinction with the lesion and the control templates. hpol η yielded the highest level of misincorporation opposite the adduct by inserting dATP or dTTP. Moreover, hpol η did not extend well past an N7-CH3 2'-F dG:dT mispair. MS-based sequence analysis confirmed that hpol η catalyzes mainly error-free incorporation of dC, with misincorporation of dA and dG in 5-10% of products. We conclude that N7-CH3 2'-F dG and, by inference, N7-CH3 dG have miscoding and mutagenic potential. The level of misincorporation arising from this abundant adduct can be considered as potentially mutagenic as a highly miscoding but rare lesion. [ABSTRACT FROM AUTHOR]

Abstract

Background: Inherently, preceptorship, is challenging and stressful for nursing students. To date, there is a dearth of literature concerning the challenges and opportunities experienced by members of the preceptorship triad namely the nursing student, their faculty advisor, and the preceptor. Purpose: The purpose of this study was to explore the challenges and opportunities associated with rural preceptorship by nursing students, their faculty advisors and preceptors. Method: Photovoice was drawn on as a creative approach to participatory action research (PAR). This method has been found to empower and engage participants as co-researchers with the ultimate goal of implementing change derived from the priorities of the community, in this case, the teaching and learning experiences of nursing students (n=9), their preceptors and their faculty advisors (n=5). Findings: As participants described their experiences throughout the rural preceptorship placement, belongingness emerged throughout the data as a predominant theme. Moreover, the experience of belonging had a significantly positive impact on student learning and overall rural preceptorship experience. Conclusions: Relationships between the nursing student and members of the health care team were found to be critical to preceptorship success in rural communities and the motivation among new graduates, to seek permanent employment in the rural community post-graduation. Abstract

Disease eradication as a public health strategy was discussed at international meetings in 1997 and 1998. In this article, the ongoing poliomyelitis eradication initiative is examined using the criteria for evaluating candidate diseases for eradication proposed at these meetings, which covered costs and benefits, biological determinants of eradicability (technical feasibility) and societal and political considerations (operational feasibility). The benefits of poliomyelitis eradication are shown to include a substantial investment in health services delivery, the elimination of a major cause of disability, and far-reaching intangible effects, such as establishment of a "culture of prevention". The costs are found to be financial and finite, despite some disturbances to the delivery of other health services. The "technical" feasibility of poliomyelitis eradication is seen in the absence of a non-human reservoir and the presence of both an effective intervention and delivery strategy (oral poliovirus vaccine and national immunization days) and a sensitive and specific diagnostic tool (viral culture of specimens from acute flaccid paralysis cases). The certification of poliomyelitis eradication in the Americas in 1994 and interruption of endemic transmission in the Western Pacific since March 1997 confirm the operational feasibility of this goal. When the humanitarian, economic and consequent benefits of this initiative are measured against the costs, a strong argument is made for eradication as a valuable disease control strategy. Abstract

This article provides a framework for the design of future eradication programmes so that the greatest benefit accrues to health systems development from the implementation of such programmes. The framework focuses on weak and fragile health systems and assumes that eradication leads to the cessation of the intervention required to eradicate the disease. Five major components of health systems are identified and key elements which are of particular relevance to eradication initiatives are defined. The dearth of documentation which can provide "lessons learned" in this area is illustrated with a brief review of the literature. Opportunities and threats, which can be addressed during the design of eradication programmes, are described and a number of recommendations are outlined. Abstract

Catalase-peroxidase (KatG) is found in eubacteria, archaea, and lower eukaryotae. The enzyme from Mycobacterium tuberculosis has received the greatest attention because of its role in activation of the antitubercular pro-drug isoniazid, and the high frequency with which drug resistance stems from mutations to the katG gene. Generally, the catalase activity of KatGs is striking. It rivals that of typical catalases, enzymes with which KatGs share no structural similarity. Instead, catalatic turnover is accomplished with an active site that bears a strong resemblance to a typical peroxidase (e.g., cytochrome c peroxidase). Yet, KatG is the only member of its superfamily with such capability. It does so using two mutually dependent cofactors: a heme and an entirely unique Met-Tyr-Trp (MYW) covalent adduct. Heme is required to generate the MYW cofactor. The MYW cofactor allows KatG to leverage heme intermediates toward a unique mechanism for H2O2 oxidation. This review evaluates the range of intermediates identified and their connection to the diverse catalytic processes KatG facilitates, including mechanisms of isoniazid activation.

192. Ultrasonographic screening for subclinical osteochondrosis of the femoral trochlea in foals (28–166 days old): a prospective farm study.

Abstract

Summary: Background: Extensive osteochondritis dissecans (OCD) lesions of the lateral ridge of the trochlea of the femur (LRTF), the most common OCD‐affected site in the stifle, have a poor outcome with surgical debridement and can be career ending. The early detection of osteochondrosis lesions and their conservative management holds the promise to enhance outcome. We hypothesise that ultrasonography is a valuable field screening tool to detect and monitor early subclinical LRTF osteochondrosis. Objectives: The goals were to 1) describe the normal ultrasonographic features of the LRTF in foals of different ages and 2) screen a foal cohort at the farm for early subclinical osteochondrosis lesions. Study design: Prospective cohort study. Methods: The LRTF of both hindlimbs of Thoroughbred foals (n = 46, 27–166 days old) were imaged once with ultrasonography and radiography (lateromedial and caudolateral‐craniomedial oblique views). Cartilage thickness, ossification front indentation of the chondro‐osseous junction and epiphyseal vascularisation were assessed on ultrasonography. Follow‐up radiographs were taken as yearlings. Results: The cartilage thickness, ossification front indentation and epiphyseal vascularisation significantly decreased with advancing maturity. Subclinical osteochondrosis lesions, characterised by semicircular indentations in the ossification front (indirect evidence of focal failure of ossification and retained cartilage) were detected in six foals (28–145 days old), both with radiography and ultrasonography. Ultrasonography provided a better overall subjective assessment of the osteochondrosis lesion topography (length, depth and the width) compared with radiography. Main limitations: Post‐mortem validation of lesions was not possible. Conclusion: Ultrasonography of the LRTF is a practical, inexpensive and reliable technique to discriminate physiological from pathological events at the LRTF in young foals. It revealed the complex topography of the chondro‐osseous junction permitting a rapid, comprehensive assessment of the subclinical osteochondrosis lesions in very young foals. [ABSTRACT FROM AUTHOR]

Abstract

Aims. Previous observations with the HESS telescope array revealed the existence of extended very-high-energy (VHE; E > 100 GeV) γ-ray emission, HESS J1023–575, coincident with the young stellar cluster Westerlund 2. At the time of discovery, the origin of the observed emission was not unambiguously identified, and follow-up observations have been performed to further investigate the nature of this γ-ray source. Methods. The Carina region towards the open cluster Westerlund 2 has been re-observed, increasing the total exposure to 45.9 h. The combined dataset includes 33 h of new data and now permits a search for energy-dependent morphology and detailed spectroscopy. Results. A new, hard spectrum VHE γ-ray source, HESS J1026–582, was discovered with a statistical significance of 7σ. It is positionally coincident with the Fermi LAT pulsar PSRJ1028–5819. The positional coincidence and radio/γ-ray characteristics of the LAT pulsar favors a scenario where the TeV emission originates from a pulsar wind nebula. The nature of HESS J1023–575 is discussed in light of the deep HESS observations and recent multi-wavelength discoveries, including the Fermi LAT pulsar PSRJ1022–5746 and giant molecular clouds in the region. Abstract

Context: The detection of gamma-rays in the very-high-energy (VHE) range (100 GeV-100 TeV) offers the possibility of studying the parent population of ultrarelativistic particles found in astrophysical sources, so it is useful for understanding the underlying astrophysical processes in nonthermal sources. Aim: The discovery of the VHE gamma-ray source HESS J1507-622 is reported and possibilities regarding its nature are investigated. Methods: The H.E.S.S. array of imaging atmospheric Cherenkov telescopes (IACTs) has a high sensitivity compared with previous instruments (~1% of the Crab flux in 25 hours observation time for a 5 sigma point-source detection) and has a large field of view (~5 deg in diameter). HESS J1507-622 was discovered within the ongoing H.E.S.S. survey of the inner Galaxy, and the source was also studied by means of dedicated multiwavelength observations. Results: A Galactic gamma-ray source, HESS J1507-622, located ~3.5 deg from the Galactic plane was detected with a statistical significance > 9 sigma. Its energy spectrum is well fitted by a power law with spectral index \Gamma = 2.24 +/- 0.16_{stat} +/- 0.20_{sys} and a flux above 1 TeV of (1.5 +/- 0.4_{stat} +/- 0.3_{sys}) X 10^{-12} cm^{-2} s^{-1}. Possible interpretations (considering both hadronic and leptonic models) of the VHE gamma-ray emission are discussed in the absence of an obvious counterpart., Comment: Accepted for publication in A&A

Abstract

Increasing interest in science education research has focused on ways of representing cognitive structure in graphical and quantitative terms. A method is presented for displaying the sequential and multirelational ideation of scientific narrative elicited from respondents. The flow map provides a figural representation of the flow of information, the points in the flow where multirelational and recurrent linkages are made, and the time required to retrieve and express the information at major intervals in the sequence and in total. In keeping with constructivist models of information analysis, the elicitation of responses requires minimal intervention by the interviewer, and flow-map representation requires low inference for its construction, providing a convenient diagram of the sequential and multirelational thought patterns expressed by the respondent. The method is illustrated by analyzing interviews from students varying in academic achievement under two interview conditions that varied in emphasis on recall of multiple relationships. The illustrative data show that students of increasing academic ability produce flow maps of more complex patterns with more cross-relational linkages. These kinds of linkages, on the whole, increase with increased emphasis in the interview on recall of relationships.

Abstract

The inner 10 pc of our galaxy contains many counterpart candidates of the very high energy (VHE; > 100 GeV) gamma-ray point source HESS J1745-290. Within the point spread function of the H.E.S.S. measurement, at least three objects are capable of accelerating particles to very high energies and beyond, and of providing the observed gamma-ray flux. Previous attempts to address this source confusion were hampered by the fact that the projected distances between those objects were of the order of the error circle radius of the emission centroid (34", dominated by the pointing uncertainty of the H.E.S.S. instrument). Here we present H.E.S.S. data of the Galactic Centre region, recorded with an improved control of the instrument pointing compared to H.E.S.S. standard pointing procedures. Stars observed during gamma-ray observations by optical guiding cameras mounted on each H.E.S.S. telescope are used for off-line pointing calibration, thereby decreasing the systematic pointing uncertainties from 20" to 6" per axis. The position of HESS J1745-290 is obtained by fitting a multi-Gaussian profile to the background-subtracted gamma-ray count map. A spatial comparison of the best-fit position of HESS J1745-290 with the position and morphology of candidate counterparts is performed. The position is, within a total error circle radius of 13", coincident with the position of the supermassive black hole Sgr A* and the recently discovered pulsar wind nebula candidate G359.95-0.04. It is significantly displaced from the centroid of the supernova remnant Sgr A East, excluding this object with high probability as the dominant source of the VHE gamma-ray emission., 7 pages, 2 figures, accepted for publication in MNRAS
