The authors report the results obtained from the application of electron microscopy techniques to the cytology of fine-needle-aspirated samples of neoplastic lesions from various body sites. These results show that the tissue structure, which is usually lost during the squashing necessary for light microscopy cytology, is preserved when the samples are processed for ultrastructural analysis. Electron microscopy also allows a highly detailed study of the cell's inner structures. Thus, when this technique is applied, fine needle-aspirated samples can be regarded as actual microbiopsies. However, because of the high cost of ultrastructural techniques, we suggest that actual analysis be performed only in selected cases, whereas fixation and inclusion for electron microscopy could be done routinely.

In the seminiferous tubules of patients at early puberty we have observed Sertoli cells showing an homogeneous aspect. In the cytoplasm of some of these elements dense bodies, which present a particular structure and shape, have been recognized. As far as the germ cells, we can find spermatogonia, spermatocytes and spermatids at various developmental stages. The presence of intercellular bridges among the germ cells and of binucleated spermatids with only one acrosome is a characteristic and fairly frequent finding in our specimens, obtained from normal patients.

Bioptic specimens of 10 normal human parathyroid glands taken during thyroidectomies were studied both by light microscopy, using a histochemical technique, and electron microscopy. All the glands exhibited regions of different sizes in which follicles made up the tissue architecture. The follicles contained a homogeneous colloid-like substance. Our results seem to exclude the amyloid nature of the follicular content suggested by some Authors, though no further clarification as to its real make-up is provided. The cells lining the follicles belong to a type not as yet described in the normal human parathyroid. These cells appear to be large and clear and are also found in the non-follicular parenchyma. On the basis of their morphologic features we have classified the large clear cells as a particular, deviated stage of the actively secreting chief-cell. The origin and significance of the follicles are also discussed.

The ultrastructural features of normal human thymocytes were analyzed at various stages of immunologic differentiation as defined by a panel of OKT monoclonal antibodies visualized by colloidal gold labelled antimouse IgG. The study shows that first stage thymocytes (OKT10+, OKT9+) have a typical morphology not found in more mature stages and describes the critical ultrastructural change which they undergo in becoming second stage thymocytes (OKT10+, OKT6+, OKT8+, OKT4+). No change in thymocyte fine morphology has been found in the progression from second to third stage (OKT10+, OKT3+, OKT8+ or OKT10+, OKT3+, OKT4+). Cells with ultrastructural features intermediate between those of the first and second stage have been found to react with monoclonal antibodies characterizing the first and the second stage. Ultrastructural similarity and homogeneity found in OKT8+ and OKT4+ thymic subpopulations suggests the acquisition by T lymphocytes of morphologic structures correlate with immunologic features previously found in peripheral blood T cell subpopulations to be a post-thymic process.

Our research is about the ultrastructural aspects of Paneth cells obtained from human jejunal mucosa. The cells appear columnar in shape and show a roundish nucleus; their cytoplasm is characterized by a great amount of R.E.R. and by typical large granules in sopranuclear position. Our results confirm those previously described by other authors about the morphology of Paneth cells. In our specimens we have found moreover a variable number of particular cytoplasmic structures; these may show a fibrillar or granular matrix and are always surrounded by a membrane.

In the present study some aspects of Sertoli cells of testicular-biopsy specimens of children from 0 to 8 years old are examined. We can distinguish two main morphological situations. In the first one, Sertoli cells with monomorphic aspect can be seen; in the second can be shown Sertoli cells with various aspects. In this polimorphic situation we can distinguish three Sertoli cell types differing in cellular shape, cytoplasme electron-density and amount of RER and Golgi complex.
