Streaming data applications are becoming more common due to the ability of different information sources to continuously capture or produce data, such as sensors and social media. Although there are recent advances, most visualization approaches, particularly Dimensionality Reduction (DR) techniques, cannot be directly applied in such scenarios due to the transient nature of streaming data. A few DR methods currently address this limitation using online or incremental strategies, continuously updating the visualization as data is received. Despite their relative success, most impose the need to store and access the data multiple times to produce a complete projection, not being appropriate for streaming where data continuously grow. Others do not impose such requirements but cannot update the position of the data already projected, potentially resulting in visual artifacts. This paper presents Xtreaming, a novel incremental DR technique that continuously updates the visual representation to reflect new emerging structures or patterns without visiting the high-dimensional data more than once. Our tests show that in streaming scenarios where data is not fully stored in-memory, Xtreaming is competitive in terms of quality compared to other streaming and incremental techniques while being orders of magnitude faster.

Diffuse intrinsic pontine glioma (DIPG) remains a fatal brainstem tumor demanding innovative therapies. As B7-H3 (CD276) is expressed on central nervous system (CNS) tumors, we designed B7-H3–specific chimeric antigen receptor (CAR) T cells, confirmed their preclinical efficacy, and opened BrainChild-03 (NCT04185038), a first-in-human phase I trial administering repeated locoregional B7-H3 CAR T cells to children with recurrent/refractory CNS tumors and DIPG. Here, we report the results of the first three evaluable patients with DIPG (including two who enrolled after progression), who received 40 infusions with no dose-limiting toxicities. One patient had sustained clinical and radiographic improvement through 12 months on study. Patients exhibited correlative evidence of local immune activation and persistent cerebrospinal fluid (CSF) B7-H3 CAR T cells. Targeted mass spectrometry of CSF biospecimens revealed modulation of B7-H3 and critical immune analytes (CD14, CD163, CSF-1, CXCL13, and VCAM-1). Our data suggest the feasibility of repeated intracranial B7-H3 CAR T-cell dosing and that intracranial delivery may induce local immune activation.Significance:This is the first report of repeatedly dosed intracranial B7-H3 CAR T cells for patients with DIPG and includes preliminary tolerability, the detection of CAR T cells in the CSF, CSF cytokine elevations supporting locoregional immune activation, and the feasibility of serial mass spectrometry from both serum and CSF.This article is highlighted in the In This Issue feature, p. 1

Supplementary Data from Intraventricular B7-H3 CAR T Cells for Diffuse Intrinsic Pontine Glioma: Preliminary First-in-Human Bioactivity and Safety

Multivariate or multidimensional visualization plays an essential role in exploratory data analysis by allowing users to derive insights and formulate hypotheses. Despite their popularity, it is usually users' responsibility to (visually) discover the data patterns, which can be cumbersome and time-consuming. Visual Analytics (VA) and machine learning techniques can be instrumental in mitigating this problem by automatically discovering and representing such patterns. One example is the integration of classification models with (visual) interpretability strategies, where models are used as surrogates for data patterns so that understanding a model enables understanding the phenomenon represented by the data. Although useful and inspiring, the few proposed solutions are based on visual representations of so-called black-box models, so the interpretation of the patterns captured by the models is not straightforward, requiring mechanisms to transform them into human-understandable pieces of information. This paper presents multiVariate dAta eXplanation (VAX), a new VA method to support identifying and visual interpreting patterns in multivariate datasets. Unlike the existing similar approaches, VAX uses the concept of Jumping Emerging Patterns, inherent interpretable logic statements representing class-variable relationships (patterns) derived from random Decision Trees. The potential of VAX is shown through use cases employing two real-world datasets covering different scenarios where intricate patterns are discovered and represented, something challenging to be done using usual exploratory approaches.

I periodi dell’Avvento e quello successivo del tempo di Natale offrono uno spaccato complesso di quella che era la coscienza folclorica e liturgico-musicale dei territori istriani, quarnerini e dalmati. Alla sovrapposizione delle residue credenze pagane si era sovrapposta la grande tradizione cristiana nella sua declinazione cattolicoromana e aquileiese, che aveva modellato per secoli, sovente in osmosi anche con le tradizioni d’Oltre Mare (Veneto e Venezia), repertori e costumanze sociali legate alle celebrazioni rituali del Natale. Attraverso la registrazione e la conservazione delle ultime testimonianze, avviatasi a partire dall’inizio del Novecento scorso per iniziativa di singoli studiosi, si è potuto dare testimonianza a un quadro di tradizioni rituali alquanto ricco e complesso anche nelle sue connessioni, ulteriore prova della grande ricchezza culturale d’impronta latino-veneta un tempo offerta dalle popolazioni delle coste dell’Adriatico Orientale, mosaico non piccolo che contribuisce anch’esso all’identità collettiva della nazione italiana., Razdoblje došašća i razdoblje nakon Božića nude složen presjek nekadašnje folklorne i liturgijsko-glazbene svijest istarskog, kvarnerskog i dalmatinskog područja: preklapanja zaostalih paganskih vjerovanja nadograđena su velikom kršćanskom tradicijom u rimokatoličkom i akvilejskom izričaju, koja se stoljećima oblikovala repertoare i društvene običaje vezane uz obredne proslave Božića, vrlo često u osmozi i s prekomorskim tradicijama (Veneto i Venecija). Zahvaljujući snimkama i čuvanju posljednjih svjedočanstava, čije je prikupljanje započelo početkom prošlog dvadesetog stoljeća na inicijativu pojedinih znanstvenika, bilo je moguće prenijeti svjedočanstvo o prilično bogatim i složenim ritualnim tradicijama, čak i u njihovim međusobnim vezama, što je još jedan dokaz velikog kulturnog bogatstva s latinskovenetskim pečatom koje je nekada nudilo stanovništvo istočne obale Jadrana, što je nemali mozaični doprinos kolektivnom identitetu talijanske nacije. Vrijeme pripreme za Božić u Istri bilo je prije svega pojava čarobne dimenzije u kojoj nalazimo životinje koje govore, ukleta bića i mjesta poput krsnika i raskrižja (cavalcanti i crosère). Bilo je to i vrijeme darivatelja, prikazanih u popularnim likovima Sv. Nikole i Sv. Lucije. U tom periodu u Istri održalo se i praznovjerje u predviđanja po kalendama. U liturgiji adventskog doba obnovljeni su obredi mise zornice i pjevanje drevnih narodnih hvalospjeva: takozvane tempora invernali, Božićna Devetnica u gregorijanskom napjevu, Missus u obližnjem Friuliju i u brdskom predjelu Veneta, a priprema jaslica i božićnog drvca upotpunili su sliku punu sugestivnih proslava i obiteljskih tradicija.
