A recombinant inbred line population, derived from a cross between a high light-tolerant wheat Triticum aestivum cv. Xiaoyan 54 and a high yielding, but high light-sensitive variety, Jing 411, was used to explore the genetic relation between photosynthesis and grain yield-related traits. The net CO2 assimilation rate, chlorophyll content, chlorophyll a fluorescence parameters, leaf area index, plant height, spike number, biomass, grain yield, and harvest index were evaluated in the field across two consecutive years. The results reveal that a total of 57 quantitative trait loci (QTL) are found to be associated with the investigated traits. They distributed on almost all 21 chromosomes, except for chromosomes 5D, 6D, 7A, and 7D. The phenotypic variance explained by a single QTL ranged from 9.3% to 39.9% depending on traits and QTL. Of these QTL, 12 QTL clusters were found to regulate at least 2 of the investigated traits, which distributed on 8 chromosomes, 1A, 1B, 2A, 2B, 2D, 3A, 3B, 4B and 5A. Seven QTL clusters were associated with both photosynthesis and grain yield-related traits, indicative of their genetic relation. Two QTL clusters on 2D and 4B were co-located with two reduced-height genes, Rht8 and Rht-B1b, respectively. These QTL clusters may be used as potential targets for wheat radiation use efficiency improvement in the future.

There is increasing interest in furfurylated wood, but the polymerization mechanism between its internal polyfurfuryl alcohol (PFA) and lignin is still uncertain. This paper investigated the distribution of PFA and the feasibility of the polymerization of PFA with lignin in furfurylated balsa wood. The wood first immersed in the furfuryl alcohol (FA) solution followed by in situ polymerization and the distribution of PFA was characterized by Raman, fluorescence microscopy, SEM, and CLSM. Then, the mill wood lignin (MWL) of balsa wood and lignin model molecules were catalytically polymerized with PFA, respectively, studying the mechanism of interaction between PFA and lignin. It was concluded that PFA was mainly deposited in cell corner with high lignin concentration, and additionally partly deposited in wood cell cavity due to high concentration of FA and partial delignification. TGA, FTIR, and NMR analysis showed that the cross-linked network structure generated by the substitution of MWL aromatic ring free position by PFA hydroxymethyl enhanced the thermal stability. New chemical shifts were established between PFA and C5/C6 of lignin model A and C2/C6 of model B, respectively. The above results illustrated that lignin-CH2-PFA linkage was created between PFA and lignin in the wood cell wall.

Abstract Introduction With the need for hearing screenings increasing across multiple populations, a need for automated options has been identified. This research seeks to evaluate the hardware requirements for automated hearing screenings using a mobile application. Objective Evaluation of headphone hardware for use with an app-based mobile screening application. Methods For the purposes of this study, hEAR, a Bekesy-based mobile application designed by the research team, was compared with pure tone audiometric tests administered by an audiologist. Both hEAR and the audiologist's test used 7 frequencies (125 Hz, 250 Hz, 500 Hz, 1,000 Hz, 2000 Hz, 4,000 Hz and 8,000 Hz) adopting four different sets of commercially available headphones. The frequencies were regarded as the independent variable, whereas the sound pressure level (in decibels) was the dependent variable. Thirty participants from a university in Texas were recruited and randomly assigned to one of two groups, whose only difference was the order in which the tests were performed. Data were analyzed using a generalized estimating equation model at α = 0.05. Results Findings showed that, when used to collect data with the mobile app, both the Pioneer HDJ-2000 (Pioneer, Bunkyo, Tokyo, Japan) (p > 0.05) and the Sennheiser HD280 Pro (Sennheiser, Wedemark, Hanover, Germany) (p > 0.05) headphones presented results that were not statistically different from the audiologist's data across all test frequencies. Analyses indicated that both headphones had decreased detection probability at 4kHz and 8kHz, but the differences were not statistically significant. Conclusion Data indicate that a mobile application, when paired with appropriate headphones, is capable of reproducing audiologist-quality data.

Objective: To explore the emotional characteristics of counterfactual thinking (CT)-related emotion responses in patients with major depressive disorder (MDD) via the “counterfactual thinking gambling task (CTGT).”Method: Twenty-five patients with MDD (the MDD group) and twenty-five healthy controls (the HC group) with matched demographic features were included. The 17-item Hamilton Depression Scale (HAMD) and the 14-item Hamilton Anxiety Rating Scale (HAMA) were used to assess the severity of depression and anxiety symptoms. The counterfactual thinking gambling task was applied to assess the situation-focused- and behavior-focused-CT-related emotion responses in the MDD group and the HC group.Results: There was no significant difference in general demographic data between the two groups (p > 0.05). Compared with the HC group, the MDD group experienced higher levels of “disappointment” and lower levels of “joy” in the situation-focused CT paradigm (p < 0.05). However, the experience of “regret” and “relief” in the behavior-focused CT paradigm were not significantly different between the two groups (p > 0.05).Conclusions: MDD is associated with an impaired situation-focused-CT-related emotion responses, and is often accompanied by increased disappointment and decreased joy; however, behavior-focused-CT-related emotion responses are not significantly affected in MDD. This pattern may represent the characteristic CT-related emotion responses of MDD.

Background: Previous studies have indicated that childhood maltreatment (CM) may potentially influence the clinical symptomatology of obsessive-compulsive disorder (OCD). Here, we aimed to quantify the relationship between CM and obsessive-compulsive symptoms (OCS) and depressive symptoms in OCD through a meta-analysis.Method: We searched PubMed, Embase, Cochrane Library, and PsycARTICLES databases for articles reporting the association between CM and OCD on April 15, 2020. Random-effect models were used to quantify the relationship between CM and the severity of OCS and depressive symptoms in OCD.Results: Ten records with 1,611 OCD patients were included in the meta-analysis. The results revealed that CM is positively correlated with the severity of OCS [r = 0.10, 95%Confidence Interval (CI): 0.01–0.19, P = 0.04] as well as depressive symptoms in OCD (r = 0.15, 95%CI: 0.07–0.24, P = 0.0002). For the subtypes of CM, childhood emotional abuse (CEA) and childhood sexual abuse (CSA) was related with the severity of OCS (r = 0.11, 95%CI: 0.03–0.19, P = 0.009) and obsession (r = 0.13, 95%CI: 0.03–0.23, P = 0.01), respectively.Conclusion: Our meta-analysis indicates that OCD patients who suffered more CM may exhibit more severe OCS and depressive symptoms.

Strong optical confinement and light–matter interaction within semiconductor nanostructures are crucial to modulate the emission property. A fundamental understanding of these interesting phenomena is important to scientific and industrial communities. In this paper, we synthesized the surface oxidized CdS micro/nano-wires by post-oxidation of CdS micro/nano-wires at 475 °C and investigated systematically their novel emission properties. Scanning electron microscopy, energy dispersive x-ray spectrometry, and Raman spectroscopy confirmed the formation of the oxidized surface without changing the structure and morphology of CdS micro/nano-wires greatly. In situ micro-photoluminescence showed the continuous modulation of multi-sub-band emission of both near bandgap and defect state simultaneously with an ultra-broad spectrum range from 490 nm to 750 nm. According to the excitation wavelength-, excitation power-, and temperature-dependent PL spectra, the sub-band near bandgap emission originated from strong light–matter interaction such as photon–exciton coupling, while the whispering gallery mode (WGM) micro-cavity played an important role in the sub-band emission of the defect state. The spatial-resolved mapping of emission modes confirmed the sub-band near bandgap and defect WGM micro-cavity emission from the core and oxidized surface of CdS micro-wires, respectively. The multi-exponential lifetime dynamics further indicated the complex recombination process of both near bandgap and defect state emission. These results are different from the one-step synthesized CdS@CdO core/shell micro-wires and reveal an interesting exciton–photon coupling with enhanced optical cavity confinement in the post-treated CdS micro/nano-wires, representing the rational tailoring of surface/interface relevant properties to control the light emission for application in nanophotonic devices.

Infections caused by the opportunistic pathogen Pseudomonas aeruginosa can be difficult to treat due to innate and acquired antibiotic resistance and this is exacerbated by the emergence of multi-drug resistant strains. Unfortunately, no licensed vaccine yet exists to prevent Pseudomonas infections. Here we describe a novel subunit vaccine that targets the P. aeruginosa type III secretion system (T3SS). This vaccine is based on the novel antigen PaF (Pa Fusion), a fusion of the T3SS needle tip protein, PcrV, and the first of two translocator proteins, PopB. Additionally, PaF is made self-adjuvanting by the N-terminal fusion of the A1 subunit of the mucosal adjuvant double-mutant heat-labile enterotoxin (dmLT). Here we show that this triple fusion, designated L-PaF, can activate dendritic cells in vitro and elicits strong IgG and IgA titers in mice when administered intranasally. This self-adjuvanting vaccine expedites the clearance of P. aeruginosa from the lungs of challenged mice while stimulating host expression of IL-17A, which may be important for generating a protective immune response in humans. L-PaF’s protective capacity was recapitulated in a rat pneumonia model, further supporting the efficacy of this novel fusion vaccine.

Chronic liver diseases are global health problems representing a significant source of morbidity and mortality worldwide. Cholestasis is one of the most common clinical manifestations of almost all types of liver diseases regardless of etiologies and promotes the pathogenesis of liver. Artemisiae Scopariae Herba (ASH, or Yin Chen) is the most commonly used Traditional Chinese Medicine (TCM) herbal for the treatment of chronic liver diseases, especially cholestatic liver diseases. Various bioactive ingredients have been isolated and identified in ASH, however, most of them were not well characterized for their pharmacological effects. In the current review, we summarize clinical evidence demonstrating the promising therapeutic effects of ASH. We then introduce the profile of bioactive ingredients found in ASH and highlight the change of their abundance during different harvest times of ASH. We further thoroughly review recent advances in the studies of major bioactive ingredients in ASH, including scoparone, capillarisin, chlorogenic acid, and representative flavonoids, for their pharmacological effects, including choleretic, antioxidant, anti-inflammation, anti-fibrosis and other hepatoprotective activities. Underlying molecular targets and signaling networks are also outlined. In conclusion, ASH-based TCM prescriptions are promising strategies for the treatment of cholestatic liver diseases and are more effective than conventional therapies. The future advancement of pharmacology and toxicology researches of ASH and its bioactive ingredients will further contribute to the discovery of novel anti-cholestasis and hepatoprotective drugs.

Considerable attention has been paid to the roles of lipid metabolism in virus infection due to its regulatory effects on virus replication and host antiviral immune response. However, few literature has focused on whether lipid metabolism is involved in the life cycle of lower vertebrate viruses. Singapore grouper iridovirus (SGIV) is the causative aquatic virus that extensively causes fry and adult groupers death. Here, the potential roles of cellular de novo fatty acid synthesis in SGIV infection was investigated. SGIV infection not only increased the expression levels of key enzymes in fatty acid synthesis in vivo/vitro, including acetyl-Coenzyme A carboxylase alpha (ACC1), fatty acid synthase (FASN), medium-chain acyl-CoA dehydrogenase (MCAD), adipose triglyceride lipase (ATGL), lipoprotein lipase (LPL) and sterol regulatory element-binding protein-1 (SREBP1), but it also induced the formation of lipid droplets (LDs), suggesting that SGIV altered de novo fatty acid synthesis in host cells. Using the inhibitor and specific siRNA of ACC1 and FASN, we found that fatty acid synthesis was essential for SGIV replication, evidenced by their inhibitory effects on CPE progression, viral gene transcription, protein expression and virus production. Moreover, the inhibitor of fatty acid β-oxidation could also reduce SGIV replication. Inhibition of fatty acid synthesis but not β-oxidation markedly blocked virus entry during the life cycle of SGIV infection. In addition, we also found that inhibition of ACC1 and FASN increased the IFN immune and inflammatory response during SGIV infection. Together, our data demonstrated that SGIV infection in vitro regulated host lipid metabolism and, in that process, cellular fatty acid synthesis might exert crucial roles during SGIV infection via regulating virus entry and host immune response.

Exploring efficient methods to enhance leaching efficiency is critical for bioleaching technology to deal with sulfide concentrate. In our study, a novel artificial microbial community was established to augment the bioleaching efficiency and recovery of copper (Cu) and zinc (Zn). The optimum parameters in bioleaching experiments were explored according to compare a series of conditions from gradient experiments: the pH value was 1.2, temperature was 45 °C, and rotation speed was 160 r/min, which were different with pure microorganism growth conditions. Under optimal conditions, the result of recovery for Cu and Zn indicated that the average leaching rate reached to 80% and 100% respectively, which almost increased 1.8 times and 1.2 times more than control (aseptic condition) group. Therefore, this method of Cu and Zn recovery using a new-type artificial microbial community is expected to be an environmentally-friendly and efficient bioleaching technology solution, which has the potential of large-field engineering application in the future.

MicroRNAs (miRNAs) are indispensable for the regulation of skeletal muscle. We performed RNA sequencing (RNA-seq) to establish a comprehensive miRNA profiling of goats in seven stages, namely, 45- (F45), 65- (F65), 90- (F90), 120- (F120), and 135-day (F135) fetuses, newborn (B1), and 90-day-old (B90) kids. In total, 421 known miRNAs and 228 goat novel miRNAs were identified in the data, and the average abundance of 19 miRNAs in seven stages exceeds 10,000 reads per million. Furthermore, 420 differentially expressed miRNAs (DEmiRNAs) were identified in all comparison group at seven stages, 80 of which were uniquely differentially expressed in the B1 and B90 comparison groups. Pathway analysis indicated that this group was associated with the release of muscle hypertrophy and regulation of myoblast proliferation. Besides, 305 DEmiRNAs were clustered into three significantly enriched profiles (profiles 11, 16, and 19). Function analysis revealed that profile 16 was related to muscle hypertrophy and differentiation. Profile 11 was involved in multiple enzyme activities and metabolic processes in muscle cells. And profile 19 was involved in material transport and structural stability. Two highly expressed miRNAs and three key miRNAs (chi-miR-328-3p, chi-miR-767, and chi-miR-150) of these profiles were verified to be consistent with the data by quantitative real-time PCR. These results provided a catalog of goat muscle-associated miRNAs, allowing us to better understand the transformation of miRNA roles during mammalian muscle development.

Esophageal cancer micro environment factor WNT2 was critical in cancer metastasis. However, very little is known about WNT2 receptors and their role in the malignant progression of ESCC. The clinical significance and underlying molecular mechanisms of FZD2, one of the receptors of WNT2, was further investigated in ESCC. We found that FZD2 expression was positively correlated with WNT2 levels in clinical ESCC specimens through database analysis. Upregulated FZD2 expression was detected in 69% (69/100) of the primary ESCC cases examined, and increased FZD2 expression was significantly correlated with poor prognosis (P < 0.05). Mechanistically, FZD2 induced the migration and invasion of ESCC cells by regulating the FZD2/STAT3 signaling. In vivo xenograft experiments further revealed the metastasis-promoting role of FZD2 in ESCC. Moreover, we found that the WNT2 ligand could stabilize and phosphorylate the FZD2 receptor by attenuating FZD2 ubiquitination, leading to the activation of STAT3 signaling and the initiation of ESCC cell metastasis. Collectively, our data revealed that a novel non-canonical WNT2/FZD2/STAT3 signaling axis is critical for ESCC progression. Strategies targeting this specific signaling axis might be developed to treat patients with ESCC.

Backgroud It has been shown that aberrant expression of microRNAs (miRNAs) and transcriptional factors (TFs) is tightly associated with the development of HCC. Therefore, in order to further understand the pathogenesis of HCC, it is necessary to systematically study the relationship between the expression of miRNAs, TF and genes. In this study, we aim to identify the potential transcriptomic markers of HCC through analyzing common microarray datasets, and further establish the differential co-expression network of miRNAs–TF–mRNA to screen for key miRNAs as candidate diagnostic markers for HCC. Method We first downloaded the mRNA and miRNA expression profiles of liver cancer from the GEO database. After pretreatment, we used a linear model to screen for differentially expressed genes (DEGs) and miRNAs. Further, we used weighed gene co-expression network analysis (WGCNA) to construct the differential gene co-expression network for these DEGs. Next, we identified mRNA modules significantly related to tumorigenesis in this network, and evaluated the relationship between mRNAs and TFs by TFBtools. Finally, the key miRNA was screened out in the mRNA–TF–miRNA ternary network constructed based on the target TF of differentially expressed miRNAs, and was further verified with external data set. Results A total of 465 DEGs and 215 differentially expressed miRNAs were identified through differential genes expression analysis, and WGCNA was used to establish a co-expression network of DEGs. One module that closely related to tumorigenesis was obtained, including 33 genes. Next, a ternary network was constructed by selecting 256 pairs of mRNA–TF pairs and 100 pairs of miRNA–TF pairs. Network mining revealed that there were significant interactions between 18 mRNAs and 25 miRNAs. Finally, we used another independent data set to verify that miRNA hsa-mir-106b and hsa-mir-195 are good classifiers of HCC and might play key roles in the progression of HCC. Conclusion Our data indicated that two miRNAs—hsa-mir-106b and hsa-mir-195—are identified as good classifiers of HCC.

Artificial intelligence based on artificial neural networks, which are originally inspired by the biological architectures of the human brain, has mostly been realized using software but executed on conventional von Neumann computers, where the so-called von Neumann bottleneck essentially limits the executive efficiency due to the separate computing and storage units. Therefore, a suitable hardware platform that can exploit all the advantages of brain-inspired computing is highly desirable. Based upon micromagnetic simulation of the magnetization dynamics, we demonstrate theoretically and numerically that recurrent neural networks consisting of as few as 40 magnetic tunnel junctions can generate and recognize periodic time series after they are trained with an efficient algorithm.

The mechanism of the formation of the hydrophobic agglomerate in fine scheelite flotation was studied using zeta potential measurement, contact angle measurement, optical microscope measurement, and atomic force microscopy (AFM) colloid probe technology. Zeta potential measurement results confirmed the adsorption of sodium oleate on scheelite, fluorite, and calcite surface and surface potential difference at different pH values of ultrapure water. Contact angle measurement results confirmed the surface of nature scheelite, fluorite, and calcite was hydrophilic, and the surface after thread by sodium oleate solution was hydrophobic. The optical microscope measurement results confirmed the agglomerates could really form in ultrapure water of pH 8 or 10 and in 1 mM sodium oleate solution of pH 10. The agglomerations were empty and not tight in ultrapure water. On the contrary, the hydrophobic agglomerations were larger and denser after treated with sodium oleate solution than that of in ultrapure water. According to the AFM experiment results, the interaction forces on hydrophilic scheelite-scheelite and scheelite-fluorite were repulsive at pH 5.6 and attractive at pH 8 or 10. However, the interaction forces on hydrophilic scheelite-calcite were attractive at pH 5.6, 8 or 10. The interaction forces on hydrophobic scheelite-scheelite, scheelite-fluorite, and scheelite-calcite were attractive strongly due to the existence of hydrophobic force. The measurement results of the interaction forces were in good agreement with the changes of zeta potential and contact angle at different conditions. The combined results could be beneficial to understand the interaction force in fine scheelite flotation.

Abstract Background The first-line agents comprising antihistamines for chronic urticaria, are not completely satisfactory. Tripterygium wilfordii Hook F (TwHF), a Chinese herb, has been developed into several Tripterygium agents and have definite effects on autoimmune and inflammatory diseases. In chronic urticaria, however, their values of practical application remain unclear. The aim of this study was to investigate the efficacy and safety of TwHF in patients with chronic urticaria. Methods Several databases were systematically searched including PubMed, Embase, Cochrane Central Register of Controlled Trials, China Network Knowledge Infrastructure, Chinese Scientific Journals Database, Wan Fang Database, and Chinese Biomedicine. Randomized controlled trials comparing antihistamines with TwHF or Tripterygium agents in combination with antihistamines were included. Revman5.3 was utilized to calculate risk ratios (RR) with 95% confidence intervals (CI). This study was registered with PROSPERO, number CRD42018091595. Results Twenty-one trials with 2565 participants were included in this analysis. Meta-analysis showed that, when antihistamines were combined with TwHF and Tripterygium agents, the curative effect in cases of chronic urticaria was superior to that of antihistamines alone (RR: 1.40; 95% CI: 1.33–1.46). The incidence rates of gastrointestinal disorder (RR: 2.91; 95% CI: 1.70–4.99) and menstrual disorder (RR: 6.00; 95% CI: 1.79–20.13) in drug combination groups were higher than those in controls, while other adverse events were similar between the two groups. After treatment, Dermatology Life Quality Index (RR: 1.23; 95% CI: 1.09–1.40), quality of sleep (RR: 1.50; 95% CI: 1.07–2.12), and daily activity (RR: 1.49; 95% CI: 1.25–1.78) were all improved. Furthermore, drug combination groups demonstrated less relapse (RR: 0.34; 95% CI: 0.25–0.45). Conclusions TwHF and Tripterygium agents, in combination with antihistamines, appear to be more effective than antihistamines alone. Nevertheless, adverse events cannot be ignored. Large sample, multi-center, high-quality clinical studies are needed to verify the exact effects and safety of TwHF and Tripterygium agents in treatment of chronic urticaria.

Pre-implantation development is a dynamic, complex and precisely regulated process that is critical for mammalian development. There is currently no description of the role of the long noncoding RNAs (lncRNAs) during the pre-implantation stages in the goat. The in vivo transcriptomes of oocytes (n = 3) and pre-implantation stages (n=19) at seven developmental stages in the goat were analyzed by RNA sequencing (RNA-Seq). The major zygotic gene activation (ZGA) event was found to occur between the 8- and 16-cell stages in the pre-implantation stages. We identified 5,160 differentially expressed lncRNAs (DELs) in developmental stage comparisons and functional analyses of the major and minor ZGAs. Fourteen lncRNA modules were found corresponding to specific pre-implantation developmental stages by weighted gene co-expression network analysis (WGCNA). A comprehensive analysis of the lncRNAs at each developmental transition of high correlation modules was done. We also identified lncRNA-mRNA networks and hub-lncRNAs for the high correlation modules at each stage. The extensive association of lncRNA target genes with other embryonic genes suggests an important regulatory role for lncRNAs in embryonic development. These data will facilitate further exploration of the role of lncRNAs in the developmental transformation in the pre-implantation stage.

Abstract Culture-independent analysis of microbial communities frequently relies on amplification and sequencing of the prokaryotic 16S ribosomal RNA gene. Typical analysis pipelines group sequences into operational taxonomic units (OTUs) to infer taxonomic and phylogenetic relationships. Here, we present HmmUFOtu, a novel tool for processing microbiome amplicon sequencing data, which performs rapid per-read phylogenetic placement, followed by phylogenetically informed clustering into OTUs and taxonomy assignment. Compared to standard pipelines, HmmUFOtu more accurately and reliably recapitulates microbial community diversity and composition in simulated and real datasets without relying on heuristics or sacrificing speed or accuracy.

Abstract Developing greener batteries with new chemistries is a formidable challenge, and a major focus for years to come. Redox flow batteries are receiving increasing research interest for grid-scale electrochemical energy storage owing to their unique architecture. However, challenges still remain by their low energy density as well as corrosive and/or toxic electrolytes. An anolyte based on aprotic Zn deep-eutectic-solvent, which uses low cost, abundant and environmentally benign materials, exhibits a utilizable concentration of Zn2+ ca. 1.7 M, resulting in a reversible volumetric capacity of ca. 90 A h·L−1. Combined with high efficiencies and relatively low redox potential of −1.12 V vs. Ag/AgCl, such an anolyte provides an alternative way to explore a family of anolytes using new chemistries for rechargeable Zn batteries that meet the criteria for grid-scale electrical energy storage.

Background/Aims: Qiliqiangxin (QL), a traditional Chinese medicine, has been demonstrated to be effective and safe for the treatment of chronic heart failure. Left ventricular (LV) remodeling causes depressed cardiac performance and is an independent determinant of morbidity and mortality after myocardial infarction (MI). Our previous studies have shown that QL exhibits cardiac protective effects against heart failure after MI. The objective of this study was to explore the effects of QL on myocardial fibrosis in rats with MI and to investigate the underlying mechanism of these effects. Methods: A rat model of acute myocardial infarction was induced by ligating the left anterior descending coronary artery. The rats were treated with QL (1.0 g/kg/day) for 4 weeks after surgery. Echocardiography and histology examination were performed to evaluate heart function and fibrosis, respectively. Protein levels of transforming growth factor-β1 (TGF-β1), phosphorylated Smad3 (p-Smad3), phosphorylated Smad7 (p-Smad7), collagen I (Col- I), alpha smooth muscle actin (a-SMA), tumor necrosis factor-α (TNF-α), interleukin 6 (IL-6), nuclear factor κB (NF-κB), and phosphorylated inhibitor of kappa B alpha (p-IκBα) were measured by western blot analysis. Results: QL treatment ameliorated adverse cardiac remodeling 8 weeks after AMI, including better preservation of cardiac function, decreased inflammation, and reduced fibrosis. In addition, QL treatment reduced Col-I, a-SMA, TGF-β1, and p-Smad3 expression levels but increased p-Smad7 levels in postmyocardial infarct rat hearts. QL administration also reduced the elevated levels of cardiac inflammation mediators, such as TNF-α and IL-6, as well as NF-κB and p-IκBα expression. Conclusions: QL therapy exerted protective effects against cardiac remodeling potentially by inhibiting TGF-β1/Smad3 and NF-κB signaling pathways, thereby preserving cardiac function, as well as reducing myocardial inflammation and fibrosis.

Abstract Background The skin harbors complex communities of resident microorganisms, yet little is known of their physiological roles and the molecular mechanisms that mediate cutaneous host-microbe interactions. Here, we profiled skin transcriptomes of mice reared in the presence and absence of microbiota to elucidate the range of pathways and functions modulated in the skin by the microbiota. Results A total of 2820 genes were differentially regulated in response to microbial colonization and were enriched in gene ontology (GO) terms related to the host-immune response and epidermal differentiation. Innate immune response genes and genes involved in cytokine activity were generally upregulated in response to microbiota and included genes encoding toll-like receptors, antimicrobial peptides, the complement cascade, and genes involved in IL-1 family cytokine signaling and homing of T cells. Our results also reveal a role for the microbiota in modulating epidermal differentiation and development, with differential expression of genes in the epidermal differentiation complex (EDC). Genes with correlated co-expression patterns were enriched in binding sites for the transcription factors Klf4, AP-1, and SP-1, all implicated as regulators of epidermal differentiation. Finally, we identified transcriptional signatures of microbial regulation common to both the skin and the gastrointestinal tract. Conclusions With this foundational approach, we establish a critical resource for understanding the genome-wide implications of microbially mediated gene expression in the skin and emphasize prospective ways in which the microbiome contributes to skin health and disease.

The past few years have seen a surge of novel applications of the Luria-Delbrück fluctuation assay protocol in bacterial research. Appropriate analysis of fluctuation assay data often requires computational methods that are unavailable in the popular web tool FALCOR. This paper introduces an R package named rSalvador to bring improvements to the field. The paper focuses on rSalvador’s capabilities to alleviate three kinds of problems found in recent investigations: (i) resorting to partial plating without properly accounting for the effects of partial plating; (ii) conducting attendant fitness assays without incorporating mutants’ relative fitness in subsequent data analysis; and (iii) comparing mutation rates using methods that are in general inapplicable to fluctuation assay data. In addition, the paper touches on rSalvador’s capabilities to estimate sample size and the difficulties related to parameter nonidentifiability.

To determine the cutaneous effects of in utero and lactational exposure to the AHR ligand 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD), pregnant C57BL/6J mice were exposed by gavage to a vehicle or 5 μg TCDD/kg body weight at embryonic day 12 and epidermal barrier formation and function were studied in their offspring from postnatal day 1 (P1) through adulthood. TCDD-exposed pups were born with acanthosis. This effect was AHR-dependent and subsided by P6 with no evidence of subsequent inflammatory dermatitis. The challenge of adult mice with MC903 showed similar inflammatory responses in control and treated animals, indicating no long-term immunosuppression to this chemical. Chloracne-like sebaceous gland hypoplasia and cyst formation were observed in TCDD-exposed P21 mice, with concomitant microbiome dysbiosis. These effects were reversed by P35. CYP1A1 and CYP1B1 expression in the skin was increased in the exposed mice until P21, then declined. Both CYP proteins co-localized with LRIG1-expressing progenitor cells at the infundibulum. CYP1B1 protein also co-localized with a second stem cell niche in the isthmus. These results indicate that this exposure to TCDD causes a chloracne-like effect without inflammation. Transient activation of the AhR, due to the shorter half-life of TCDD in mice, likely contributes to the reversibility of these effects.

Trait anxiety is a vulnerable personality factor for anxiety and depression. High levels of trait anxiety confer an elevated risk for the development of anxiety and other psychiatric disorders. There is evidence that 5-hydroxytryptamine receptor 1B (5-HT1B) gene polymorphisms play an important role in emotional disorders. Genotyping for four single-nucleotide polymorphisms (SNP) (rs11568817, rs130058, rs6297, and rs13212041) was conducted for 388 high trait anxious (HTA) individuals and 463 low traitanxious (LTA) individuals in Chinese Han college subjects. The results showed that the frequencies of the C-allele and TC + CC genotype of rs13212041 in the LTA individuals were higher than that in the HTA individuals (p = 0.025 and p = 0.014, respectively). Both the C-allele and TC + CC genotype were associated with trait anxiety decreasing (OR = 0.771 and OR = 0.71, respectively). Furthermore, different gene model analysis also showed that the C allele was a protective factor for trait anxiety in Chinese Han college subjects. These findings suggest that 5-HT1B rs13212014 may play a role in trait anxiety among China Han college subjects. The rs13212014 polymorphism may be involved in decreasing the risk of trait anxiety. These results also provide a novel insight into the molecular mechanism underlying trait anxiety.

Ischemic stroke is the main cause of death/disability, posing a great menace to human health. Though efforts to search for therapeutic drugs are ongoing, few of them have succeeded. Adenosine A1 receptor (A1R) activation could ameliorate ischemic injury, representing a very tempting target for stroke treatment. Tetrahydroxy stilbene glycoside (TSG), a potent antioxidant from the well-known Chinese herb Polygonum multiflorum Thunb., has been reported to have notable neuroprotective activities but the underlying mechanisms are elusive. This study investigated the mechanism of TSG focusing on A1R. TSG markedly decreased mortality, neurological deficit score, cerebral infarct size and brain water content of MCAO rats, and ameliorated the disorders in purine metabolism, energy metabolism and antioxidative defense system. TSG helped the survival of SH-SY5Y cells in OGD/R by alleviating oxidative stress and glutamate release, and by maintaining calcium homeostasis. TSG effects were abolished by A1R antagonist DPCPX. Docking and binding assays confirmed the binding of TSG with A1R. In addition, TSG upregulated the A1R level lowered by MCAO and OGD/R. The downstream signals of A1R activation, ERK1/2, HIF-1α and NF-κB contributed to the neuroprotection of TSG. Moreover, void of “well-known” cardiovascular side effects of classical A1R agonists, TSG showcased its great potential for stroke treatment.

Second language (L2) speakers often experience difficulties in learning words with L2-specific phonemes due to the unfaithful lexical encoding predicted by the fuzzy lexical representations hypothesis. Currently, there is limited understanding of how allophonic variation in the first language (L1) influences L2 phonological and lexical encoding. We report how the Mandarin Chinese L1 phonemic inventory and allophonic variation subject to phonotactic constraints predict phonological encoding problems for novel L2 English words with the /v/-/w/ contrast. L1 English and L1 Chinese participants speaking two varieties of Mandarin Chinese differing as to the presence of [[voiced labiodental approximant]]-[w] allophonic variation for the /w/ phoneme participated in a vocabulary learning task. The novel L2 words with the /v/-/w/ contrast were systematically less robustly encoded than the control words on the day of training and 24 hours later. The degree of fuzziness in lexical representations was jointly predicted by L1 allophonic variation subject to phonotactic constraints and L2 phonological categorization.

Although academic resilience is of great concern to contemporary educational practitioners, there is no consensus on its measurement. Furthermore, protective factors characterized by East Asian societal contexts remain ambiguous. This systematic review aims to offer an overview of the operational definitions, statistical methodology, and protective factors of academic resilience identified in East Asian countries/economies. With a focus on large-scale assessment (LSA) research, three databases (i.e., Web of Science, CNKI, and AiritiLibary) were searched and returned 31 peer-reviewed studies over the last decade. Results indicated that the definition-driven method was commonly adopted in international LSA studies (e.g., Programme for the International Student Assessment) to measure academic resilience, and the research conducted in national/regional LSA (e.g., China Education Panel Survey) tended to use the process-driven approach. Logistic regression was the most frequently data analysis technique utilized in the definition-driven approach, while structural equation modeling and mediation/moderation analyses accounted for the largest proportion of the process-driven methods. Our study shed light on the methodological issues of academic resilience in LSA. Additionally, it highlights the aspiration of educational researchers to identify Asian-specific protective factors from the social-ecological perspective to propose appropriate interventions fostering academic resilience.

Collaborative problem-solving competence (CPSC) is a core skill in the twenty-first century. As an innovative literacy, CPSC was assessed for the first time in the Program for International Student Assessment (PISA) 2015. Our study aims to identify the effects of student's perceptions toward interpersonal relationships on CPSC in PISA 2015. Multiple linear regression was used to examine these effects across Hong Kong, Macao, Taiwan, and the four affluent communities in China. After controlling student's gender and socio-economic status, the results indicated that: (a) Student's external perceptions such as teacher unfairness and disciplinary climate, and student's internal perceptions such as valuing relationships and valuing teamwork significantly predicted CPSC; (b) Disciplinary climate was the strongest predictor among all the external perceptions analyzed; (c) Regarding the internal perceptions, valuing relationships positively predicted CPSC, while valuing teamwork negatively predicted CPSC. These findings enlighten educational practitioners to help students develop their CPSC from the perspective of interpersonal relationships.

Several international large-scale assessments were conducted at the turn of the new century, and during the past two decades the Programme for International Student Assessment (PISA) completed seven cycles of assessment to facilitate practitioners' policy debates and governance. This study reviews PISA-related articles published in English and Chinese. Three literature databases were searched, with a focus on SSCI, CSSCI, and TSSCI journal publications. The frequency of publication was analyzed according to the author's country affiliation, type of journal, and research categories/themes. Findings indicate that research on student-, school- and system-level indicators with a focus on students' learning processes and outcomes as well as critiques of technical matters on PISA were frequent topics in the literature during the past two decades. Issues of equality and equity examined in the publications have implications for enhancing practitioners' capabilities in terms of policy making and governance.

With the rapid growth of education data in large-scale assessment, machine learning techniques are crucial to the interdisciplinary development of education and information. Although data mining tools are increasingly used to predict overall student performance, resilient students in the digital world remain unstudied. Our study aims to comprehensively identify key features in the classification of academically resilient students (ARS) and non-academically resilient students (NRS) in digital reading. With a sample of 11,496 disadvantaged students from seven high-performing Asian economies, data drawn from the Programme for International Student Assessment (PISA) 2018 were analyzed through Support Vector Machine (SVM). Results indicated that 20 key features were selected from 105 contextual features at the individual, home, and school levels, which demonstrated a high predictive ability of the model. Personal experience, especially the use of metacognitive strategies in digital reading and reading enjoyment were predominant features. Interestingly, information and communication technology (ICT) resources and usage showed mixed effects on resilient students. This study provides significant implications for cultivating resilient students in online learning environments.

Abstract Circular RNAs (circRNAs) represent a class of endogenous noncoding RNAs that have recently been recognized as important regulators of gene expression and pathological networks. However, their transcriptional activities and functional mechanisms in cancer remain largely unknown. Here, we present results from a global circRNA expression and functional analysis of patients with hepatocellular carcinoma (HCC). Using a circRNA microarray, we identified 226 differentially expressed circRNAs, of which 189 were significantly upregulated and 37 were downregulated. High expression of circRNA_100338, one of the upregulated circRNAs in HCC, is closely correlated with a low cumulative survival rate and metastatic progression in HCC patients with hepatitis B. Furthermore, our in silico and experimental analyses identified miR-141-3p as a direct target of circRNA_100338. Thus, circRNA_100338 functions as an endogenous sponge for miR-141-3p in HCC. In addition, we identified the crucial antagonistic roles of circRNA_100338 and miR-141-3p in the regulation of invasive potential in liver cancer cells. Overall, the differential expression of multiple circRNAs in HCC tissues and their clinical significance in hepatitis B-related HCC patients as revealed by our study suggests that circRNA_100338 is a potentially valuable biomarker for HCC diagnosis and target for HCC therapeutics.

Abstract Background Axon growth inhibitory factors NogoA/Nogo receptor (NgR) and its signaling pathways RhoA/Rho kinase (ROCK) play a critical role in the repair of nerve damage in multiple sclerosis (MS). Bu Shen Yi Sui Capsule (BSYSC) is an effective Chinese formula utilized to treat MS in clinical setting and noted for its potent neuroprotective effects. In this study, we focus on the effects of BSYSC on promoting nerve repair and the underlying mechanisms in mice with experimental autoimmune encephalomyelitis (EAE), an animal model of MS. Methods The EAE mouse model was induced by injecting subcutaneously with myelin oligodendrocyte glycoprotein (MOG) 35–55 supplemented with pertussis toxin. BSYSC was orally administrated at dose of 3.0 g/kg once a day for 40 days. The levels of protein gene product (PGP) 9.5, p-Tau, growth associated protein (GAP) -43, KI67 and Nestin in the brain or spinal cord on 20 and 40 day post-induction (dpi) were detected via immunofluorescence and Western blot analysis. Furthermore, NogoA/NgR and RhoA/ROCK signaling molecules were studied by qRT-PCR and Western blot analysis. Results Twenty or 40 days of treatment with BSYSC increased markedly PGP9.5 and GAP-43 levels, reduced p-Tau in the brain or spinal cord of mice with EAE. In addition, BSYSC elevated significantly the expression of KI67 and Nestin in the spinal cord 40 dpi. Further study showed that the activation of NogoA/NgR and RhoA/ROCK were suppressed by the presence of BSYSC. Conclusions BSYSC could attenuate axonal injury and promote repair of axonal damage in EAE mice in part through the down-regulation of NogoA/NgR and RhoA/ROCK signaling pathways.

Abstract Background Multiple sclerosis (MS) as an autoimmune disorder is a common disease occurring in central nervous system (CNS) and the remyelination plays a pivotal role in the alleviating neurological impairment in the MS. Catalpol, an effective component extracted from the Chinese herb Radix Rehmanniae, which has been proved protective in cerebral diseases. Methods To determine the protective effects and mechanisms of Catalpol on MS, the mice with experimental autoimmune encephalomyelitis (EAE) were induced by myelin oligodendrocyte glycoprotein (MOG) 35–55, as a model for human MS. Th17 cells were counted by flow cytometric (FCM). The expressions of nerve-glial antigen (NG) 2 and myelin basic protein (MBP) were measured by immunohistochemical staining. Olig1+ and Olig2+/BrdU+ cells were counted by immunofluorescence. Olig1 and Olig2 gene expressions were detected by real-time fluorescent quantitative reverse transcription (qRT) -PCR. Results The results showed that Catalpol improved neurological function, reduced inflammatory cell infiltration and demyelination. It could decrease Th17 cells in the peripheral blood. It increased the protein expressions of NG2 and MBP in mice brains, up-regulated markedly protein and gene expressions of Olig1 and Olig2 in terms of timing, site and targets. Conclusions These data demonstrated that Catalpol had a strong neuroprotective effect on EAE mice. Catalpol also plays a role in remyelination by promoting the expressions of Olig1 and Olig2 transcription factors.

End-to-end transmission is an essential pattern in current Internet. It had brought obvious conveniences and potential hazard simultaneously. New network technology provides plenty of opportunities to avoid the harmful influences. After reviewing the identifier-based network, a candidate solution of future network, we proposed a smart collaborative connection management scheme for improving the dependability. Comparing with the conventional approaches, our scheme can protect the hosts via intermediate network equipments. During transmission process, the current usage pattern will not be influenced. Traditional unicast can be easily and smoothly enabled. The details of the scheme design are presented via four perspectives of connection identifier, i.e., communication mode, generation, distribution, and deployment. Then the supporting approaches of multipath transport, service migration, network mobility, P2P distribution, and application layer multicast are carefully investigated. The results of implementation and validation illustrate that the performances of new scheme are acceptable. We expect that this paper could provide a compatible and effective solution for both current and future Internet.

Forsythia suspensa is an important medicinal plant and native to China. In this article, the complete chloroplast genome of F. suspensa was studied and annotated. The chloroplast genome of F. suspensa is 156,404 bp in length, which has a large single-copy region (LSC) of 87,150 bp, a small single-copy region (SSC) of 17,812 bp, and two of inverted-repeat regions (IRs) of 25,716 bp. The overall nucleotide composition is: 30.8% (48,103 bp) of A, 31.4% (49,193 bp) of T, 19.2% (30,066 bp) of C and 18.6% (29,042 bp) of G, and the total content of the chloroplast genome A + T content of 62.2% and G + C content of 37.8%. The chloroplast genome of F. suspensa contains 133 genes, including 88 protein-coding genes (PCGs), 37 transfer RNA (tRNAs), and 8 ribosome RNA (rRNAs). The phylogenetic tree result shows that F. suspensa is closely related to F. intermedia in the phylogenetic relationship by the maximum-likelihood (ML) method.

The aim of this study was to develop an intravenous clarithromycin lipid emulsion (CLE) with good stability and excellent antibacterial activity. The CLE was prepared by the thin-film dispersed homogenization method. The interaction between clarithromycin (CLA) and cholesteryl hemisuccinate (CHEMS) was confirmed by DSC, FT-IR and 1H NMR analysis. The interfacial drug loading, thermal sterilization, freeze–thaw stability, and in vitro and in vivo antibacterial activity were investigated systematically. DSC, FT-IR and 1H NMR spectra showed that CHEMS (CLA: CHEMS, M ratio 1:2) could interact with CLA through H-bonding and a hydrogen-bonded ion pair. The CHEMS was found necessary to maintain the stability of CLE. Ultracentrifugation showed that almost 88% CLA could be loaded into the interfacial layer. The optimized CLE formulation could withstand autoclaving at 121 °C for 10 min and remain stable after three freeze–thaw cycles. The in vitro susceptibility test revealed that the CLA–CHEMS ion-pair and CLE have similar activity to the parent drug against many different bacterial strains. The in vivo antibacterial activity showed that the ED50 of intravenous CLE was markedly lower than that of CLA solution administrated orally. CLE exhibited pronounced antibacterial activity and might be a candidate for a new nanocarrier for CLA with potential advantages over the current commercial formulation.

Axonal damage is recognized as an important pathological feature in the chronic progressive neurological disorder multiple sclerosis (MS). Promoting axonal regeneration is a critical strategy for the treatment of MS. Our clinical and experimental studies have shown that the Bu Shen Yi Sui formula (BSYS) promotes axonal regeneration in MS and experimental autoimmune encephalomyelitis (EAE), an animal model of MS, but the exact mechanism has not been thoroughly elucidated to date. In this study, we investigated the effects of BSYS and its two decomposed formulas—the Bu Shen formula (BS) and the Hua Tan Huo Xue formula (HTHX)—on brain-derived neurotrophic factor (BDNF)/TrkB and related signaling pathways to explore the mechanism by which axonal regeneration is promoted in vitro and in vivo. Damaged SH-SY5Y cells incubated with low serum were treated with BSYS-, BS-, and HTHX-containing serum, and EAE mice induced by the myelin oligodendrocyte glycoprotein (MOG)35–55 peptide were treated with BSYS. The results showed that the BSYS-containing serum markedly increased cell viability and increased the levels of growth associated protein (GAP)-43, phosphorylated (p)-cAMP-response element binding protein (CREB), BDNF, TrkB, and p-PI3K. The BS and HTHX treatments also induced the protein expression of GAP-43 and p-extracellular signal-regulated kinase (ERK) in the cells. Furthermore, the effects of BSYS on cell viability, GAP-43, p-CREB, and neurite outgrowth were clearly inhibited by LY294002, a specific antagonist of the PI3K signaling pathways. The addition of U0126 and U73122, antagonists of the ERK and PLCγ pathway, respectively, significantly inhibited cell viability and GAP-43 protein expression. Moreover, BSYS treatment significantly increased the expression of the 68-, 160-, and 200-kDa neurofilaments (NFs) of proteins and the BDNF, TrkB, PI3K, and Akt mRNA and proteins in the brain or spinal cord of mice at different stages. These results indicated that BSYS promotes nerve regeneration, and its mechanism is mainly related to the upregulation of the BDNF/TrkB and PI3K/Akt signaling pathways. BS and HTHX also promoted nerve regeneration, and this effect involved the ERK pathway.

Diarrheal diseases are a major cause of morbidity and mortality worldwide. They are most prevalent in settings with inadequate sanitation, poor hygiene and contaminated water. An important diarrheal pathogen in such settings is Shigella. No commercially available vaccine exists against shigellosis and immunity to the pathogen is serotype-restricted. We have previously shown that a polypeptide fusion of the Type Three Secretion Apparatus (T3SA) proteins IpaB and IpaD (named DBF) was efficacious as a vaccine against Shigella. Vaccination using different administration routes indicated that protection conferred by DBF did not fully correlate with antibodies. To define the immune responses involved in protection, we studied cellular responses to intranasal immunization with the DBF and the adjuvant dmLT. We found dendritic cell (DC) activation at the nasal associated lymphoid tissue (NALT). Activation markers CD86 and MHCII significantly increase in cells from immunized mice. Antigen exposure in vitro further confirmed the upregulation of CD80 and CD40 in primary dendritic cells. Animals immunized with antigen-primed dendritic cells were protected against Shigella infection, at levels comparable to the efficacy of immunization with the protein vaccine formulation. Therefore, we show that antigen-primed DCs are enough to provide immunity, and propose a mechanism of protection against Shigella spp. based on DC-mediated antigen presentation to T cells.

Mammalian cell expression systems are the most commonly used platforms for producing biotherapeutic proteins. However, development of recombinant mammalian cell lines is often hindered by the unstable and variable transgene expression associated with random integration. We have developed an efficient strategy for site-specific integration of genes of interest (GOIs). This method enables rapid and precise insertion of a gene expression cassette at defined loci in mammalian cells, resulting in homogeneous transgene expression. We identified the Hipp11 (H11) gene as a "safe harbor" locus for gene knock-in in CHO-S cells. Using clustered regularly interspaced short palindromic repeats (CRISPR)/Cas9 mediated homologous recombination, we knocked in a DNA cassette (the landing pad) that includes a pair of PhiC31 bacteriophage attP sites and genes facilitating integrase-based GOI integration. A master cell line, with the landing pad inserted correctly in the H11 locus, was established. This master cell line was used for site-specific, irreversible recombination, catalyzed by PhiC31 integrase. Using this system, an integration efficiency of 97.7% was achieved with green fluorescent protein (GFP) after selection. The system was then further validated in HEK293T cells, using an analogous protocol to insert the GFP gene at the ROSA26 locus, resulting in 90.7% GFP-positive cells after selection. In comparison, random insertion yielded 0.68% and 1.32% GFP-positive cells in the CHO-S and HEK293T cells, respectively. Taken together, these findings demonstrated an accurate and effective protocol for generating recombinant cell lines to provide consistent protein production. Its likely broad applicability was illustrated here in two cell lines, CHO-S and HEK293T, using two different genomic loci as integration sites. Thus, the system is potentially valuable for biomanufacturing therapeutic proteins.

Chitosan (CS), the second most abundant polysaccharide in nature, has been widely developed as a nanoscopic drug delivery vehicle due to its intriguing characteristics. In this work, a positively charged CS-based nanogel was designed and synthesized to inhibit the proliferation of breast cancer cell lines. The model drug of 10-hydroxycamptothecin (HCPT) was entrapped into the core via a facile diffusion to form CS/HCPT. The characteristics of CS/HCPT were evaluated by assessing particle size, drug loading content, and drug loading efficiency. Furthermore, cell internalization, cytotoxicity, and apoptosis of CS/HCPT were also investigated in vitro. The present investigation indicated that the positively charged CS-based nanogel could be potentially used as a promising drug delivery system.

In this study, furfural was used as a crosslinking agent to enhance the water resistance of lignin-phenol-formaldehyde (LPF) resin. The effect of the furfural content on the physicochemical properties of the adhesives was explored, and the possible synthesis mechanism of the furfural-modified lignin-phenol-formaldehyde (LPFF) resin adhesives was investigated. Compared with the LPF adhesive, the LPFF adhesive with 15% furfural content and 50% lignin substituent exhibited outstanding properties in all considered aspects; it had a high wet shear strength (1.30 MPa), moderate solid content (54.51%), and low viscosity (128 mPa∙s), which were 38.0% higher, 3.6% higher, and 37.5% lower than those of the LPF adhesive. Analyses via nuclear magnetic resonance and Fourier transform infrared (FTIR) spectroscopy confirmed that the furfural content improved water resistance of the lignin-based adhesive; this improvement was due to the formation of new chemical bonds between furfural and lignin to construct a dense crosslinked network structure. In addition, the decrease in viscosity and the increase in solid content enabled the adhesive to better penetrate into the wood porous structure, showing stronger adhesion. Therefore, the LPFF adhesive has superior water resistance, high strength, and good thermal stability; thus, it has a great potential for industrial applications.

Microbial fuel cells (MFCs) are of increasing interest as bioelectrochemical systems for decomposing organic materials and converting chemical energy into electricity. The main challenge for this technology is that the low power and voltage of the devices restricts the use of MFCs in practical applications. In this paper, a power management system (PMS) is developed to store the energy and export an increased voltage. The designed PMS successfully increases the low voltage generated by an individual MFC to a high potential of 5 V, capable of driving a wireless temperature and humidity sensor based on nRF24L01 data transmission modules. With the PMS, MFCs can intermittently power the sensor for data transmission to a remote receiver. It is concluded that even an individual MFC can supply the energy required to power the sensor and telemetry system with the designed PMS. The presented PMS can be widely used for unmanned environmental monitoring such as wild rivers, lakes, and adjacent water areas, and offers promise for further advances in MFC technology.

Predictive models play a central role in decision making. Penalized regression approaches, such as least absolute shrinkage and selection operator (LASSO), have been widely used to construct predictive models and explain the impacts of the selected predictors, but the estimates are typically biased. Moreover, when data are ultrahigh-dimensional, penalized regression is usable only after applying variable screening methods to downsize variables. We propose a stepwise procedure for fitting generalized linear models with ultrahigh dimensional predictors. Our procedure can provide a final model; control both false negatives and false positives; and yield consistent estimates, which are useful to gauge the actual effect size of risk factors. Simulations and applications to two clinical studies verify the utility of the method.

Fluorescence immunochromatographic assay (FICA) is a rapid immunoassay technique that has the characteristics of high precision and sensitivity. Although image FICA strip readers have the advantages of high portability and easy operation, the use of high-precision complementary metal oxide semiconductor (CMOS) image sensors leads to an increase in overall cost. Considering the popularity of CMOS image sensors in smartphones and their powerful processing functions, this work developed a smartphone-based FICA strip reader. An optical module suitable for the test strips with different fluorescent markers was designed by replacing the excitation light source and the light filter. An android smartphone was used for image acquisition and image denoising. Then, the test and control lines of the test strip image were recognized by the sliding window algorithm. Finally, the characteristic value of the strip image was calculated. A linear detection range from 10 to 5000 mIU/mL (R2 = 0.95) was obtained for human chorionic gonadotrophin with the maximum relative error less than 9.41%, and a linear detection range from 5 to 4000 pg/mL (R2 = 0.99) was obtained for aflatoxin B1, with the maximum relative error less than 12.71%. Therefore, the smartphone-based FICA strip reader had high portability, versatility, and accuracy.

The precise timing, petrogenesis, and geodynamic significance of three granitoid bodies (Beidao granite, Caochuanpu granite, Yuanlongzhen granite, and the Roche type rock) of the Tianshui area in the Western Qinling Orogen, central China, are poorly constrained. We performed an integrated study of petrology, geochemistry, and zircon U-Pb dating to constrain their genesis and tectonic implication. Petrographic investigation of the granites shows that the rocks are mainly monzogranites. The Al saturation index (A/CNK versus SiO2) of the granitoid samples indicates meta-aluminous to peraluminous I-type granites. Their magmas were likely generated by the partial melting of igneous protoliths during the syn-collisional tectonic regime. Rare-earth-elements data further support their origin from a magma that was formed by the partial melting of lower continental crust. The Beidao, Caochuanpu, and Yuanlongzhen granites yielded U-Pb zircon weighted mean ages of 417 ± 5 Ma, 216 ± 3 Ma, and 219 ± 3 Ma, respectively. This study shows that the Beidao granite possibly formed in syn- to post-collision tectonic settings due to the subduction of the Proto-Tethys under the North China Block, and can be linked to the generally reported Caledonian orogeny (440–400 Ma) in the western segment of the North Qinling belt, whereas Yuanlongzhen and Caochuanpu granites can be linked to the widely known Indosinian orogeny (255–210 Ma). These granitoids formed due to the subduction of the oceanic lithospheres of the Proto-Tethyan Qinling and Paleo-Tethyan Qinling. The Roche type rock, tourmaline-rich, was possibly formed from the hydrothermal fluids as indicated by the higher concentrations of boron leftover during the late-stages of magmatic crystallization of the granites.

In this paper, we report on a type of SU-8 microstructure with vertical sidewalls used for polydimethydiloxane (PDMS) microchannels. Multi-layer lithography using focal plane changing approach is proposed to expose the SU-8 photoresist based on a digital micromirror device (DMD) maskless lithography system. We used a light-emitting diode source with a wavelength of 405 nm. The thickness of the SU-8 is divided into multi-layers according to the depth of focus. Each layer corresponds to a depth of focus, and then, a virtual mask is designed for the layer. Finally, each layer is exposed to changes in the focal plane. The results indicate that the actual profile of the SU-8 mold shows good agreement with the design profile without any T-profiles. Additionally, there is better linewidth in the proposed method compared with multi-exposure by a single fixed focal plane. The PDMS microchannels result also demonstrate the stability of the SU-8 mold.

Stalk lodging resistance, which is mainly measured by stem diameter (SD), stalk bending strength (SBS), and rind penetrometer resistance (RPR) in maize, seriously affects the yield and quality of maize (Zea mays L.). To dissect its genetic architecture, in this study multi-locus genome-wide association studies for stalk lodging resistance-related traits were conducted in a population of 257 inbred lines, with tropical, subtropical, and temperate backgrounds, genotyped with 48,193 high-quality single nucleotide polymorphisms. The analyses of phenotypic variations for the above traits in three environments showed high broad-sense heritability (0.679, 0.720, and 0.854, respectively). In total, 423 significant Quantitative Trait Nucleotides (QTNs) were identified by mrMLM, FASTmrEMMA, ISIS EM-BLASSO, and pLARmEB methods to be associated with the above traits. Among these QTNs, 29, 34, and 48 were commonly detected by multiple methods or across multiple environments to be related to SD, SBS, and RPR, respectively. The superior allele analyses in 30 elite lines showed that only eight lines contained more than 50% of the superior alleles, indicating that stalk lodging resistance can be improved by the integration of more superior alleles. Among sixty-three candidate genes of the consistently expressed QTNs, GRMZM5G856734 and GRMZM2G116885, encoding membrane steroid-binding protein 1 and cyclin-dependent kinase inhibitor 1, respectively, possibly inhibit cell elongation and division, which regulates lodging resistance. Our results provide the further understanding of the genetic foundation of maize lodging resistance.
