Abstract

International audience; The 5‐alkoxyoxazole ring system has proved to be a useful and versatile building block that is involved in a variety of chemical transformations (e.g., hetero‐Diels–Alder reactions, formal 1,3‐dipolar cycloadditions, photochemical [2+2] cycloadditions, Claisen rearrangements, and ring‐opening processes). 5‐Alkoxyoxazoles have provided synthetic access to heteroaromatic scaffolds such as pyridines, furans, oxadiazolines, pyrrolidines, and significant linear chemical patterns, including amino acid derivatives. This review focuses on the broad range of reactivity of 5‐alkoxyoxazoles and their subsequent applications to the syntheses of naturally occurring compounds and biologically active molecules.

156. In vitro evaluation of uncharged oximes for organophosphorus nerve agent poisoning

Abstract

Organophosphorus nerve agents (OPNA ; e.g. sarin, cyclosarin, tabun, VX) are lethal compounds that act on cholinergic nervous system by irreversible inhibition of acetylcholinesterase (AChE, E.C. 3.1.1.7) and butyrylcholinesterase (BChE, E.C. 3.1.1.8). Such an inhibition causes accumulation of the synaptic acetylcholine (ACh) and overstimulation of muscarinic and nicotinic receptors in the peripheral and central nervous system (CNS). Poisoning leads to respiratory failure and seizures, while those who survive experience long-term neurological and neuropsychiatric disorders. In light of the growing risk of terrorist attacks in Europe and worldwide, a growing need for adequate protection and proper safety plans has arisen. Current therapeutic approach consists of antimuscarinic atropine, oxime reactivator of acetylcholinesterase and anticonvulsive diazepam. Standard oximes (e.g. 2-PAM, HI-6, obidoxime) are not efficient for every OPNA and they do not possess a possibly preferred pharmacokinetic profile, i.e. their penetration across brain-blood barrier (BBB) is not more than 10 % of the blood concentration. Since standard oxime reactivators are charged, novel uncharged oximes have been synthesised to obtain more efficient reactivators with enhanced CNS penetration capabilities. Four out of seven novel oximes (JR585, JR595, RM048, and GM415) showed a highly promising in vitro reactivation profile of AChE and/or BChE- inhibited by different OPNA. The best results were obtained in the reactivation of VX- inhibited AChE/BChE, sarin-inhibited AChE and cyclosarin-inhibited BChE. Their increased lipophilicity (ClogP = 3.24 – 4.99) and other in silico determined molecular descriptors suggest a better penetration across plasma membranes and a higher volume of distribution (Vd), which implies potential and beneficial reactivation of AChE in the brain. However, a higher incidence of adverse in vivo effects is also predicted by computational studies. Therefore, further evaluation of their in vivo efficacy will include both antidotal and toxicity testing in mice. International audience; A shorter and more versatile synthetic route has been developed to prepare tetrahydropyrido[2,1‐a]isoindolone and superior analogues. The key step of this synthesis is the formation of the lactam ring by a cyclization using Shibasaki's conditions.The isoindolone scaffold is present in many biologically active compounds. Here, we have developed a shorter and more efficient synthesis of tetrahydropyrido[2,1‐a]isoindolone. The key step of this approach is a cyclization to form the γ‐lactam ring under Shibasaki's conditions. Thus, tetrahydropyrido[2,1‐a]isoindolone and superior analogues, namely hexahydroazepino[2,1‐a]isoindolone and hexahydroazocino[2,1‐a]isoindolone, have been prepared in only three steps in 39, 25, and 19 % overall yields, respectively. This novel strategy offers a shorter alternative to existing procedures.

Abstract

International audience; The phenol residue of water‐soluble 2‐(2′‐hydroxy‐5′‐dimethylaminobenzyl)‐4,6‐dimethylamino‐1,3,5‐triazine is masked by an enzyme‐labile protecting group constructed with a self‐immolative para‐hydroxybenzyl alcohol. The fluorescence of the dyes is restored with lipases under physiological conditions without aggregation or precipitation of the fluorescent dyes.Derivatives of 2‐(2′‐hydroxy‐5′‐dimethylaminobenzyl)‐4,6‐dimethylamino‐1,3,5‐triazine were synthesized by an original multistep protocol that afforded, after quaternization of the N,N‐dimethylaminobenzyl moiety, water‐soluble fluorescent dyes. These fluorophores exhibited excited‐state intramolecular proton transfer and large Stokes shifts (ΔSS > 10000 cm–1). The phenol residue was masked by an enzyme‐labile protecting group based on self‐immolative para‐hydroxybenzyl alcohol. Lipases were used to unveil the fluorescence under physiological conditions (phosphate‐buffered saline, pH 7.2) without aggregation or precipitation of the fluorescent dyes.

176. Assessing the efficacy of novel uncharged oximes as reactivators of phosphylated cholinesterases

Abstract

A set of newly synthesized uncharged oximes was characterised through detailed in silico and in vitro studies as potential therapy in organophosphorus compound (OP) poisoning. We analysed the oximes’ interaction with cholinesterases (ChE) and assessed their efficacy in reactivating cholinesterases inhibited by different organophosphorus compounds. The results showed that the ChE binding affinity for oximes is comparable to that for similarly charged pyridinium oximes, Ki ranging from 5‐150 μM. The observed reactivation potency was most pronounced in the case of VX‐inhibited ChE, reflected in a high reactivation maximum of 80 % obtained within 5‐60 min. Such a result indicated that the studied uncharged oximes could be considered potential centrally acting antidotes for OP‐poisoning and could be promising candidates for in vivo studies. However, as in the case of the charged oximes, their relatively tight binding to uninhibited ChE could imply their toxicity in vivo. By means of molecular modelling, we analysed the positioning of the tested oximes in the OP‐inhibited ChE active site and evaluated possibilities for further oxime structure refinements. Abstract

Defining an efficient therapy for highly toxic organophosphorus (OP) compounds poisoning is an ongoing challenge. Our recent investigations, ranging from in silico and in vitro to in vivo experiments, set guidelines for the synthesis of new compounds to act as more efficient antidotes in OP compound poisoning. A part of our research indicated that the potential benefits could be observed by introducing uncharged compounds with especially designed structures to comply with the specificities of the cholinesterase active site. Here, we present data obtained in in vitro studies on one set of newly-synthesized uncharged oximes. To assess the strengths and weaknesses of their antidotal potencies, we defined their physiological and chemical properties and linked them to their interactions with the human acetylcholinesterase (AChE) and butyrylcholinesterase (BChE). Furthermore, we performed detailed kinetics studies on the reactivation of different OP-inhibited AChE and BChE by these oximes to see whether any of them could be pointed out as a promising candidate for in vivo studies. Abstract

Reported herein is a nitroso‐based fluorescence turn‐on probe that displays an almost instantaneous and strong fluorescence response towards hydrogen sulfide.Since the recent discovery that hydrogen sulfide plays a key role in many biological and physiological processes, the development of novel methods to efficiently detect this molecule has become an intensive field of research. In this context, fluorimetric strategies have become one of the methods of choice. However, very few different reducible chemical functionalities have been successfully used in the design of reduction‐based probes for the fluorescence detection of hydrogen sulfide. Reported herein is the first nitroso‐based fluorescence turn‐on probe that displays an almost instantaneous and strong fluorescence response towards hydrogen sulfide. A comparative study carried out with the hydroxylamino‐based probe analogue showed a significant difference in fluorescence response.

Abstract

Organophosphates (OP) inhibit acetylcholinesterase (AChE, EC 3.1.1.7), both in peripheral tissues and the central nervous system, causing adverse and sometimes fatal effects due to the accumulation of the neurotransmitter acetylcholine. The currently used therapy, which focuses on the reactivation of inhibited AChE, is limited to peripheral tissues because the commonly used quaternary pyridinium oxime reactivators do not cross the blood brain barrier at therapeutically relevant levels. Three new uncharged oximes 1 [E)-3-hydroxy-6-(4-morpholinobutyl)picolinaldehyde oxime], 2 [(E)-6-(5-(diethylamino)pentyl)-3-hydroxypicolinaldehyde oxime], and 3[(Z)-3-hydroxy-6-(5-(piperidin-1-yl)pentyl)picolinaldehyde oxime] have been synthesized. These novel compounds exhibited in vitro reactivation potencies toward VX-phosphorylated human acetylcholinesterase equal or superior to those of pyridinium oximes (HI-6, 2PAM, etc.), which are currently used as antidotes against nerve agents. The evaluation of individual reactivation constants revealed that the potent reactivation exhibited by oxime 1 referred to its efficient interaction with the phosphonate group of the VX-conjugated AChE through forming a transition state reflected in an up to 2-times faster k+2 than HI-6. Interestingly, the binding affinity of the other two oximes was very similar to that of HI-6, meaning that the structural requirements of novel compounds to react with phosphorylated AChE were fulfilled. In conclusion, we obtained potential antidotes for OP-poisoning that could act centrally, reactivating brain OP-phosphorylated AChE.
