Nicotinamide mononucleotide (NMN) is a natural antioxidant approved as a nutritional supplement and food ingredient, but its protective role in silicosis characterized by oxidative damage remains unknown. In this study, we generated a silicosis model by intratracheal instillation of silica, and then performed histopathological, biochemical, and transcriptomic analysis to evaluate the role of NMN in silicosis. We found that NMN mitigated lung damage at 7 and 28 days, manifested as a decreasing coefficient of lung weight and histological changes, and alleviated oxidative damage by reducing levels of reactive oxygen species and increasing glutathione. Meanwhile, NMN treatment also reduced the recruitment of inflammatory cells and inflammatory infiltration in lung tissue. Transcriptomic analysis showed that NMN treatment mainly regulated immune response and glutathione metabolism pathways. Additionally, NMN upregulated the expression of antioxidant genes Gstm1, Gstm2, and Mgst1 by promoting the expression and nuclear translocation of nuclear factor-erythroid 2 related factor 2 (Nrf2). Gene interaction analysis showed that Nrf2 interacted with Gstm1 and Mgst1 through Gtsm2. Promisingly, oxidative damage mediated by these genes occurred mainly in fibroblasts. In summary, NMN alleviates silica-induced oxidative stress and lung injury by regulating the endogenous glutathione metabolism pathways. This study reveals that NMN supplementation might be a promising strategy for mitigating oxidative stress and inflammation in silicosis.

The recrystallization and texture evolution of cold-rolled FeCrAl−0.65Nb and FeCrAl−1.2Nb alloys thin-wall tubes annealed at 600–900 °C for 1–600 min were investigated. The microstructures were characterized by electron back scattering diffraction, electron probe micro-analyzer and transmission electron microscopy. The Vickers hardness and room temperature tensile properties were tested. The results showed that the hardness of fully recrystallized FeCrAl−1.2Nb alloy was higher and more likely to recrystallize than FeCrAl−0.65Nb alloy. The weak texture strength of annealing sample was obtained and the proportion of //ND texture increased. The fine Laves phase distributed uniformly in FeCrAl−0.65Nb alloy had good pinning effect and inhibited recrystallization. Higher Nb content had little effects on tensile properties of thin-wall tube, and induced the formation of larger Laves phase. There was less fine Laves phase pinning in the large area adjacent to the blocky Laves phase, which resulted in easy recrystallization in FeCrAl−1.2Nb alloy.

Objective To analyze the clinicopathological features and immunohistochemical expression of meningiomas in the Tibetan population in Tibet,and improve the understanding of meningiomas. Methods The clinical and pathological data of all the meningiomas diagnosed by pathology in Tibet Autonomous Region People's Hospital from April 2013 to March 2021 were analyzed retrospectively.All the cases underwent immunohistochemical staining of trimethylation of lysine 27 on histone H3 (H3K27me3),mucin 4 (MUC4),somatostatin receptor 2 (SSTR2),progesterone receptor,epithelial membrane antigen,glial fibrillary acidic protein,vimentin,S-100,P53,and Ki-67.The histopathological features and the staining results were observed under a light microscope. Results A total of 116 cases of meningiomas were included in this study,with the male-to-female ratio of 1.0∶2.6 and the age of 4-73 years.The main clinical symptom was headache.The imaging examination showed that 114 cases had single lesions and 2 cases had multiple lesions.The tumors were located in the cranium (108 cases) and spinal canal (8 cases).The maximum diameter of the tumors ranged from 0.3 cm to 10.0 cm,with a mean of (5.7±2.2) cm.In terms of microscopic grading and histological types,the 116 cases included 111 cases of WHO grade Ⅰ (including 53 cases of fibrous type,20 cases of meningothelial type,24 cases of transitional type,10 cases of psammomatous type,etc.),4 cases of WHO grade Ⅱ (3 cases of atypical type and 1 case of clear cell type),and 1 case of WHO grade Ⅲ (papillary type).The immunohistochemical staining showed H3K27me3 expression absent in 9 cases (9/116,7.8%),MUC4 positive in 64 cases (64/116,55.2%),SSTR2 positive in 101 cases (101/116,87.1%).Eighty cases had follow-up results,among which 71 cases had no recurrence,while 9 cases recurred. Conclusions Meningioma is the most common tumor in the central nervous system in the pathological file of Tibet.It mainly attacks the middle-aged female patients,occupying the parasagittal sinus,falx,and convex surface of the brain.Fibrous meningioma of WHO grade Ⅰ is common,while the meningiomas of WHO grades Ⅱ and Ⅲ are rare.The expression degree of MUC4 is higher in meningothelial and transitional meningiomas but lower in fibrous meningiomas.There may be no correlation between the absence of H3K27me3 expression and prognosis.

On the basis of the lanthanide metalloligand [Ln(ODA)3]3- (H2ODA = oxydiacetic acid), three new Na-Ln heterometallic coordination polymers, [Ln(ODA)3Na2]n [Ln = Eu (1) and Gd (2)] and [Tb(ODA)3Na3(H2O)2]n (3), had been assembled by adjusting the concentration of Na+ ions in the reaction system. The investigations of fluorescence sensing showed that 1 could be a ratiometric probe to detect tetracycline (TC) and oxytetracycline (OTC) with high sensitivity and low detection limits, 71.92 ppb for the former and 45.54 ppb for the latter, and 3 could selectively sense 4-(phenylazo)aniline through the turn-off pathway with 14.59 ppb of detection limits. Moreover, the competing and circulating experiments indicated that both 1 and 3 had satisfactory antiinterference and recyclability for the corresponding analytes. All of these results implied that 1 and 3 should be potential fluorescent sensors for the detection of TC/OTC and 4-(phenylazo)aniline, and the possible sensing mechanism had also been discussed in depth.

In order to study the influence of the axial compression ratio and steel ratio on the shear-carrying capacity of steel-truss-reinforced beam-column joints, five shear failure interior joint specimens were designed. The effect of different coaxial pressure ratios (0.1, 0.2, and 0.3) and steel contents on the strain, ultimate bearing capacity, seismic performance, and failure pattern of cross-inclined ventral and chord bars in the joint core area was investigated. The experimental results show that the load-displacement hysteretic curves of all test specimens exhibit a bond-slip phenomenon. With the increase of the axial compression ratio, the ultimate bearing capacity of the joint core increases by 3.4% and 5.9%, respectively. While the ductility decreases by 10.3% and 13.1%, and the energy consumption capacity decreases by 3.2% and 5.8%, respectively. The shear capacity and ductility of the member with cross diagonal ventral steel angle in the joint core are increased by 12.9% and 13.4%, respectively. The shear capacity and ductility of the joint can be significantly improved by increasing the amount of steel in the core area. The expression of shear capacity suitable for this type of joint is obtained by fitting analysis, which can be used as a reference for engineering design.

SF3B1, an essential component of the U2 snRNP, is frequently mutated in cancers. Cancer-associated SF3B1 mutation causes aberrant RNA splicing, mostly at 3' splice sites (3'ss). RNA splicing of DVL2, a regulator of Notch signaling, is affected by SF3B1 mutation. Here, we report that the mutated SF3B1 use an alternative branchpoint sequence (BPS) for the aberrant splicing of DVL2, which has a higher affinity to U2 snRNA than the BPS for the canonical splicing of DVL2. Swapping the position of the alternative BPS with the position of the canonical BPS decreased the aberrant splicing of DVL2, suggesting that the mutated SF3B1 prefers to use BPS with high affinity to U2 snRNA for splicing. Additionally, swapping the positions of two BPSs associated with the canonical splicing of DVL2 demonstrated that both the affinity to the U2 snRNA and the distance to the 3'ss are important to the selection of BPS. Importantly, the aberrant splicing of DVL2 does not require the canonical 3'ss and the canonical polypyrimidine tract, which reveals a novel type of aberrant splicing induced by SF3B1 mutation. These findings provide a more comprehensive understanding of the mechanisms underlying aberrant splicing induced by SF3B1 mutation in cancer.

Glucose performs many essential functions associated with metabolic processes in the living system, and is closely related to many diseases such as diabetes and hypoglycemia. Most of the existing glucose concentration detection methods require complex instruments, which undoubtedly limit its widespread use. Here, we have designed a glucose colorimetric detection system composed of glucose, glucose oxidase (GOD), manganese dioxide nanosheets (MnO2 NSs) and 3,3',5,5'-tetramethylbenzidine (TMB) to achieve colorimetric detection with the naked eye. Compared with the single-color change of the colorimetric method in previous studies, multiple-color changes have been realized. MnO2 NSs, as a kind of nanomaterial imitating oxidase, can directly oxidize TMB to oxTMB. Because oxTMB showed a dark yellow color when strongly oxidized and light blue when weakly oxidized, this feature can achieve multiple-color changes rather than a single-color change, which is helpful for colorimetric observation with the naked eye. Finally, we successfully used MnO2 NSs for colorimetric detection of glucose and realized multiple-color changes, making it easier to achieve colorimetric observation with the naked eye. The linear detection range is 0-4000 μM and limit of detection is 5.0 μM. This is not only useful for glucose, but also has an important significance for other experiments considering colorimetric experiments with the naked eye.

BackgroundFibrosis is a core pathological factor of ligamentum flavum hypertrophy (LFH) resulting in degenerative lumbar spinal stenosis. Autophagy plays a vital role in multi-organ fibrosis. However, autophagy has not been reported to be involved in the pathogenesis of LFH.MethodsThe LFH microarray data set GSE113212, derived from Gene Expression Omnibus, was analyzed to obtain differentially expressed genes (DEGs). Potential autophagy-related genes (ARGs) were obtained with the human autophagy regulator database. Functional analyses including Gene Ontology (GO), Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment, Gene Set Enrichment Analysis (GSEA), and Gene Set Variation Analysis (GSVA) were conducted to elucidate the underlying biological pathways of autophagy regulating LFH. Protein-protein interaction (PPI) network analyses was used to obtain hub ARGs. Using transmission electron microscopy, quantitative RT-PCR, Western blotting, and immunohistochemistry, we identified six hub ARGs in clinical specimens and bipedal standing (BS) mouse model.ResultsA total of 70 potential differentially expressed ARGs were screened, including 50 up-regulated and 20 down-regulated genes. According to GO enrichment and KEGG analyses, differentially expressed ARGs were mainly enriched in autophagy-related enrichment terms and signaling pathways related to autophagy. GSEA and GSVA results revealed the potential mechanisms by demonstrating the signaling pathways and biological processes closely related to LFH. Based on PPI network analysis, 14 hub ARGs were identified. Using transmission electron microscopy, we observed the autophagy process in LF tissues for the first time. Quantitative RT-PCR, Western blotting, and immunohistochemistry results indicated that the mRNA and protein expression levels of FN1, TGFβ1, NGF, and HMOX1 significantly higher both in human and mouse with LFH, while the mRNA and protein expression levels of CAT and SIRT1 were significantly decreased.ConclusionBased on bioinformatics analysis and further experimental validation in clinical specimens and the BS mouse model, six potential ARGs including FN1, TGFβ1, NGF, HMOX1, CAT, and SIRT1 were found to participate in the fibrosis process of LFH through autophagy and play an essential role in its molecular mechanism. These potential genes may serve as specific therapeutic molecular targets in the treatment of LFH.

As the most important domestic fish in the Antarctic Ocean, Antarctic toothfish (鳞头犬牙南极鱼为南极最为重要的底层鱼类,具有重要的生态作用和极高的商业价值,鱼类耳石形态随着生活史过程的推移有所差异,故耳石形态分析可用于推断鳞头犬牙南极鱼的生活史过程。本研究利用采集自南极罗斯海、阿蒙森海、威德尔海及拉扎列夫海120尾分属4个生活史阶段的鳞头犬牙南极鱼耳石,结合传统测量分析与椭圆傅里叶分析两种形态学研究方法,对其各生活史阶段耳石形态的差异进行研究。结果表明: 鳞头犬牙南极鱼各生活史阶段耳石形态存在差异,随着生活史过程的推移,耳石的变化趋势为整体上由轮廓平滑、复杂度较低向轮廓曲折、高度复杂化转变,且纵轴方向增长速度低于横轴方向,翼叶等特征部位变化显著。针对耳石形态的线性判别分析结果显示,椭圆傅里叶分析判别值较高,为85.4%,而传统测量分析判别值较低,为71.9%,表明相较于传统测量分析,椭圆傅里叶分析更具优势。.

The development of multifunctional nano-drug carriers has become one of the most effective and prevailing approaches to overcome the non-selectivity and low cell uptake efficiency of drugs, as well as various side effects of traditional chemotherapy drugs. Herein, we reported novel hollow mesoporous silica nanoparticles (HMSNs) coated with polydopamine (PDA) and poly(poly(ethylene glycol) methacrylate) (PPEGMA) (HMSNs@PDA-s-s-PPEGMA) for the treatment of cancer. The HMSNs mainly served as a warehouse loading anticancer drug doxorubicin (DOX); the covering of PDA acted as pH-sensitive gatekeepers to control the diffusion and/or release of DOX from HMSNs in different pH stimuli environments, whilst the grafting of PPEGMA on the surface of HMSNs@PDA by disulfide bonds and atom transfer radical polymerization (ATRP) increased the stability under physiological conditions and showed reductant sensitive responsiveness. The HMSNs@PDA-s-s-PPEGMA had a high loading capacity and entrapment efficiency of drugs through loading twice but low cytotoxicity. The in vitro release results indicated that the HMSNs@PDA-s-s-PPEGMA exhibited a high sensitivity to pH and glutathione (GSH) due to the PDA coating and disulfide bonds, which could control DOX release as intelligent "switches". The drug-loaded formulations could better be taken up by the cells, formed a higher concentration of DOX in the cells, and produced more remarkable apoptosis, thus achieving better treatment of tumors with high therapy efficacy. Therefore, the developed multifunctional materials could be used as a promising nano-drug carrier for cancer treatment.

AIM: To investigate the early corneal morphological changes and the possible influenced factors of small incision lenticule extraction(SMILE)combined with corneal collagen cross-linking(CXL)in the treatment of refractive error.METHODS: This was a retrospective case study. The patients(76 eyes)with ametropia who had undergone SMILE surgery in our hospital from September 2018 to March 2019 were small age, high degree of severity, thin cornea or irregular shape. The patients were divided into the SMILE combined with CXL group with 17 patients(32 eyes)and the SMILE group with 22 patients(44 eyes). The corneal morphological parameters of the two groups were measured preoperatively and 1mo postoperatively. RESULTS: There were significant statistically differences in the changes of K1, K2, Km, central corneal thickness, posterior corneal elevation, anterior corneal elevation parameters, posterior corneal elevation parameters, the D value and index of vertical asymmetry(IVA)between these two groups(PP>0.05). The changes of some parameters of the corneal anterior surface morphology of the two groups were correlated with the corneal cutting depth, spherical equivalent and optical zone diameter(PP>0.05). There was no apparent correlation between the changes of corneal surface morphology and the cross-linking depthin SMILE combined with CXL group(P>0.05). CONCLUSION: SMILE combined with CXL was applied to the high-risk patients with small age, high degree of severity, thin cornea or irregular shape, which increased its safety and effectiveness. SMILE combined with CXL didn't make significant effects on the posterior surface morphology of the cornea, but it influenced the anterior surface morphology. The influenced factors may be related to the corneal cutting depth, the spherical equivalent and the optical zone diameter.

The sound insulation performance of railway car body structures is critical for the control of rail vehicle interior noise. In sound transmission loss (STL) measurements, a niche with a large depth is necessary to allow for mounting the wide range of thicknesses of railway car body panels and for the mechanical isolation of the two rooms. In this study, two typical interior floor panels are tested in a series of mounting conditions and mechanical boundary conditions. The change of STL results during measurement is also predicted by an STL prediction model based on the finite element method. At lower frequencies, the STL results are influenced by both the mounting positions and the mechanical boundary conditions. At higher frequencies, the STL results are mainly influenced by the mechanical boundary conditions. Differences between the panel in the infinite baffle and niches at the resonance and off-resonance frequencies are different. Considering both the effects of mounting positions and mechanical boundary conditions, the existence of the cavity amplifies the STL difference caused by the mechanical boundary conditions.

Objective To investigate the clinicopathological features and immunohistochemical phenotypes of brain metastatic carcinoma in Tibetan patients. Methods The clinical and pathological data of all patients with brain metastases from 2014 to 2020 in Tibet Autonomous Region People's Hospital were retrospectively analyzed,including 13 cases of brain metastatic carcinoma.All cases were diagnosed and classified by immunohistochemical staining. Results 13 cases(9 males and 4 females)of brain metastatic carcinoma,aged 26-62 years old,present with headache,dizziness,nausea and vomiting clinically.Four patients had a medical history of tumor,and among the 9 patients with no history of tumor,7 present space occupying lesions in both the brain and other organs.Imaging data could be found in 10 cases,including 4 cases of single lesion and 6 cases of multiple lesions.Primary tumors were identified in 11 cases(8 located in the lung,including 4 cases of adenocarcinoma,3 cases of small cell carcinoma,and 1 case of squamous cell carcinoma;1 case of urothelial carcinoma of the renal pelvis;1 case of thyroid papillary carcinoma;1 case of uterine choriocarcinoma),whereas the primary tumor was unknown for the other 2 cases(1 case of small cell carcinoma and 1 case of adenocarcinoma). Conclusions Brain metastatic carcinoma are more common among middle-aged and elderly people in Tibet.Most of the cases have no history of tumor,with the initial site at the brain metastatic lesions.The most common primary site is the lung,and the primary site of some cases is unknown.Multiple lesions are common in brain metastatic carcinoma,especially in the cerebral hemisphere.For older patients with multiple brain space occupying lesions,the possibility of brain metastatic carcinoma increases.

Time-delay reservoir computing uses a nonlinear node associated with a feedback loop to construct a large number of virtual neurons in the neural network. The clock cycle of the computing network is usually synchronous with the delay time of the feedback loop, which substantially constrains the flexibility of hardware implementations. This work shows an asynchronous reservoir computing network based on a semiconductor laser with an optical feedback loop, where the clock cycle (20 ns) is considerably different to the delay time (77 ns). The performance of this asynchronous network is experimentally investigated under various operation conditions. It is proved that the asynchronous reservoir computing shows highly competitive performance on the prediction task of Santa Fe chaotic time series, in comparison with the synchronous counterparts.

We survey the related theories and burgeoning literature on the consequences of institutional investors’ holdings and shareholder activism campaigns. Agency theory, monitoring theory, and limited attention theory are widely used in institutional investors studies. About the economic consequences of institutional investors’ holdings, we summarize that institutional holdings could improve corporate disclosure quality and frequency, for both mandatory and voluntary information disclosure. Additionally, higher ownership concentration of institutional investors is related to higher performance sensitivity of management compensation and lower compensation level. Furthermore, we propose that investor holdings could also affect dividend policy, firm performance and firm innovation. Lastly, we further discussed about shareholder activism campaign. We find that shareholder activism has both positive effect on firm performance and corporate governance, and negative side on executives and threats of exit.

Colorectal cancer (CRC) is a malignant tumor with the second highest morbidity and the third highest mortality in the world, while the therapeutic options of targeted agents remain limited. Here, mucosa-associated lymphoid tissue lymphoma translocation protein 1 (MALT1), known as the upstream of the NF-κB signaling pathway, was identified to be highly upregulated in CRC tumors and cell lines. Furthermore, the downregulation of MALT1 or inhibition of its proteolytic function by MI-2 suppressed the cell proliferation and migration of CRC cells.

Progranulin (PGRN) is a secreted factor involved in inflammatory diseases. However, the function of PGRN in silica-induced lung inflammation has not been elucidated. In this study, we demonstrated that PGRN in serum and lung tissues was markedly increased in silicosis mouse model. And immunohistochemistry results showed that PGRN was mainly expressed in alveolar macrophages, which was further confirmed in silica-treated alvelar macrophages cell line (MH-S) in vitro. PGRN promoted pro-inflammatory cytokines transcription such as interleukin (Il)-6, tumor necrosis factor-α (Tnf-α) and Il-1β in MH-S cells, and the increasing of Il-6 was most obvious. Knockdown of PGRN blocked the silica-induced elevation of intracellular Il-6 in MH-S cells. Furthermore, we also found that PGRN could increase the phosphorylation of Cyclic AMP-responsive element-binding protein 1 (Creb1), a transcriptional regulator of Il-6. Inhibition of p-Creb1 by the phosphorylation inhibitor of Creb1 (666-15) decreased PGRN-induced intracellular Il-6 production in MH-S cells. In conclusion, PGRN was highly increased in silicosis mouse model and upregulated inflammatory cytokines expression. These findings suggested that PGRN might be a key mediator in silica-induced inflammation and provided a new clue for the diagnosis and drug therapy of silicosis.

Ascoviruses are fatal double-stranded DNA viruses with a special pathogenesis in which cells are converted into vesicles with virions. Several closely related ascovirus isolates that shared more than 90% genomic DNA identity showed different pathogenic courses in previous studies. To investigate the pathogenic differences between the related ascovirus isolates, Heliothis virescens ascovirus 3i (HvAV-3i) and Heliothis virescens ascovirus 3j (HvAV-3j) were used to inoculate four noctuid pest species (Helicoverpa armigera, Mythimna separata, Spodoptera frugiperda, and Spodoptera litura), and the pathogenic indexes were recorded. The mortality of HvAV-3i infected H. armigera and S. frugiperda was approximately 60%, while the other HvAV-infected larvae had mortality rates above 90%. Bridges and tunnels are built across lakes, especially in China, to improve road connectivity for transportation. However, their environmental impacts and risks have not received adequate attention. In this study, the magnitude of bridges and tunnels across 142 lakes in China were investigated. The investigation revealed that 37 bridges and 10 tunnels (a total length of 56.82 km) were built across 26 lakes during 2000-2020. From 2011 to 2020, the construction rate of bridges and tunnels across lakes in China was ∼6 times higher than the average value worldwide. Their environmental impacts and risks on lakes were summarised by analyzing previous publications screened from the Web of Science platform. The environmental impacts and risks during the bridge/tunnel construction period generally include decrease in water exchange, habitat destruction, biological reduction, increase in suspended solids, and water quality pollution. Tunnel construction may also affect the ecological conditions of groundwater due to its disturbances in the underground area. During the bridge/tunnel operation period, environmental impacts and risks were mainly induced by rainfall events and water pollution accidents. The impacts and risks were highly related to the construction location, bridge/tunnel properties, moving vehicles, fixing activities, and atmospheric deposition. Owing to the existing weaknesses in management practices, a framework, aiming to reduce the environmental impacts and risks caused by bridges and tunnels across lakes, was proposed. The framework identified the critical issues and their corresponding management strategies during the bridge/tunnel construction and operation periods and enabled the generation of the best management strategy for each specific period.

This paper mainly investigates the tracking control problem for a mobile robot with visibility constraints. The dynamics of a two-wheeled nonholonomic mobile robot is firstly established. By considering the objective of tracking a moving target with unknown velocities, an adaptive controller is designed by utilizing backstepping technique. Furthermore, the prescribed performance bound (PPB) technique is adopted to guarantee that the moving target is always forced within the visible range of the on-board camera. Simulation results are provided to validate the theoretical results.

Liposomes are a useful carrier for delivering drugs but rarely make a poorly water-soluble drug (PWSD) realize its therapeutic potential. A key barrier lies in that, by conventional methods, PWSD is mainly loaded just in liposome bilayer membranes, which rarely provide sufficient room to accommodate drugs satisfying clinical therapy. In this investigation, a novel procedure of temperature up-down cycle (TUDC) was developed for loading PWSDs into the liposome interiors instead of bilayer membranes to hold enough agents. In particular, the TUDC procedure renders PWSDs such as curcumin (Cur) entrapment purposely controllable, as evidenced by the encapsulation efficiency (EE) of Cur varies nearly from 0% to 100% in response to changes the determinant factors of the procedure. In addition, several mathematical equations that could calculate the loading efficiency by TUDC were established and proved, when combined with thermodynamic process, able to successfully predict the loading results through including thermodynamic parameters, such as temperature and deduced drug solubility, thus remarkably cutting down the laborious experiments and enhancing liposome development efficiency. Cryo-TEM, SAXS, XRD and DSC tests proved that TUDC is feasible to load a PWSD into PEG-liposomes but rendering the drug in the amorphous state. Thus, the novel TUDC procedure and the established mathematical and thermodynamic process may provide a useful tool to promote the development of liposome products.
