Abstract

We report on molecular dynamics ( MD) simulations for predicting the density and isothermal compressibility of an H2O-NaCl fluid as a function of temperature (673-2000 K), pressure (0.2-2.0 GPa), and salt concentration (0.0-21.9 wt%). The atomistic behavior was analyzed via the hydration number of ions and number of ion pairs. Hydration numbers of Na+ and Cl− increased with increasing pressure and decreasing temperature. Conversely, the fraction of Na-Cl ion pairs increased with decreasing pressure and increasing temperature. This hydration and association behavior is consistent with the low dielectric constant of H2O under these conditions. The presence of polynuclear clusters of Na-Cl was confirmed at high temperatures, low pressures, and high salt concentrations. We propose a purely empirical equation of state (EoS) for H2O-NaCl fluids under high temperatures and pressures that should be useful for estimating the fluid distribution in Earth's crust and upper mantle in relation to effects on earthquakes and volcanic eruptions. Abstract

Introduction Given an increasing use of dipeptidyl peptidase-4 (DPP-4) inhibitors to treat patients with type 2 diabetes mellitus in the real-world setting, we conducted a prospective observational study (Japan-based Clinical Research Network for Diabetes Registry: J-BRAND Registry) to elucidate the safety and efficacy profile of long-term usage of alogliptin.Research design and methods We registered 5969 patients from April 2012 through September 2014, who started receiving alogliptin (group A) or other classes of oral hypoglycemic agents (OHAs; group B), and were followed for 3 years at 239 sites nationwide. Safety was the primary outcome. Symptomatic hypoglycemia, pancreatitis, skin disorders of non-extrinsic origin, severe infections, and cancer were collected as major adverse events (AEs). Efficacy assessment was the secondary outcome and included changes in hemoglobin A1c (HbA1c), fasting blood glucose, fasting insulin and urinary albumin.Results Of the registered, 5150 (group A: 3395 and group B: 1755) and 5096 (3358 and 1738) were included for safety and efficacy analysis, respectively. Group A patients mostly (>90%) continued to use alogliptin. In group B, biguanides were the primary agents, while DPP-4 inhibitors were added in up to ~36% of patients. The overall incidence of AEs was similar between the two groups (42.7% vs 42.2%). Kaplan-Meier analysis revealed the incidence of cancer was significantly higher in group A than in group B (7.4% vs 4.8%, p=0.040), while no significant incidence difference was observed in the individual cancer. Multivariate Cox regression analysis revealed that the imbalanced patient distribution (more elderly patients in group A than in group B), but not alogliptin usage per se, contributed to cancer development. The incidence of other major AE categories was with no between-group difference. Between-group difference was not detected, either, in the incidence of microvascular and macrovascular complications. HbA1c and fasting glucose decreased significantly at the 0.5-year visit and nearly plateaued thereafter in both groups.Conclusions Alogliptin as a representative of DPP-4 inhibitors was safe and durably efficacious when used alone or with other OHAs for patients with type 2 diabetes in the real world setting.

Abstract

ABSTRACT Forecasts of a Global Coupled Model for austral summer with a 1 month lead are downscaled to end-of-season maize yields and accumulated streamflow over the Limpopo Province and adjacent districts in northeastern South Africa through application of an MOS ( Model Output Statistics) approach applied over a 28 year period. Promising results, based on the hindcasts of the Global Models and historically observed yield and streamflow data, suggest potential for a commodity-orientated forecast system for application in agriculture in an operational environment. It also serves as a baseline study for inclusion of sophisticated crop or runoff models using GCM output data towards estimating potential yields and streamflows in the region. Abstract

We have used density functional theoryto predict the ion exchangeenergies for divalent cations Ni2+, Sr2+, Cd2+, and Pb2+into a calcite {10.4} surface in equilibriumwith water. Exchange energies were calculated for substitution intothe topmost surface layer, at the mineral–fluid interface,and into the second layer of the solid. This information can be usedas an indicator for cation substitution in the bulk phase, such asfor the uptake of toxic metals from the environment and the growthof secondary phases. In both the surface and in the second layer,Ni2+, Cd2+, and Pb2+substitute exothermicallyand Sr2+substitutes endothermically. Our results agreewith published experimental data that demonstrate trace metal coprecipitationwith calcite as a sink for Ni2+, Cd2+, and Pb2+, whereas Sr2+has a distribution constant significantlysmaller than 1. Ni2+substitution is favored at the mineral–fluidinterface compared with bulk substitution, which also agrees withexperimental data. Our results predict that Ni2+, Cd2+, and Pb2+form a stable solid solution with calcite.Successful prediction of the experimental results gives us confidencein our ability to predict the divalent cation preference for surfacesrather than for sites within the bulk crystal structure, which cannotbe directly derived from experiment.
