Mit verschiedenen Industriepartnern und dem Fraunhofer AGP wurde ein Fertigungsverfahren zur Herstellung von Stabtragwerken aus faserverstärktem Kunststoff (FVK) entwickelt. Es erlaubt, beliebige Tragwerksstrukturen als ein integrales und besonders leichtes Bauteil direkt aus den Halbzeugen Roving und Matrixharz ressourceneffizient sowie kostengünstig herzustellen. Das Verfahren ist weitgehend automatisierbar. Zu den Produktbeispielen zählen unter anderem Masten, Brückenstrukturen und Rotorblätter von Windkraftanlagen.

Die humane Peptidyl-Prolyl-cis/trans-Isomerase Pin1 bindet spezifisch an phosphorylierte Serin-Prolin- und phosphorylierte Threonin-Prolin-Reste (pSer/Thr-Pro) und katalysiert die Prolylisomerisierung in diesen Motiven. Im Rahmen dieser Arbeit wurde das Protein DNA replication licensing factor MCM3, eine Untereinheit des „minichromosome maintenance“-Komplexes MCM2-7, als neuer Interaktionspartner von Pin1 identifiziert. Die Interaktion wird über die WW-Domäne von Pin1 vermittelt und erfordert die Phosphorylierung von Ser/Thr-Pro-Motiven in MCM3. Pin1 unterstützt in HeLa-Zellen die Ausbildung des MCM2-7-Komplexes und beeinflusst die Assemblierung von MCM3 am Chromatin in Abhängigkeit von der Zellzyklusphase. Bei der Untersuchung von neuen unterschiedlich substituierten Arylhetarylketonen und Thioketonen als Inhibitoren wurden Verbindungen mit Inhibitionskonstanten im mikromolaren und submikromolaren Bereich gegenüber Pin1 gefunden. Zudem wurde eine Methode zur Isolierung von Cyclophilinen aus humanem Serum entwickelt., The human peptidyl prolyl cis/trans isomerase Pin1 specifically binds phosphorylated serine-proline and phosphorylated threonine-proline moieties (pSer/Thr-Pro) and catalyzes the prolyl isomerization within these motifs. In this work the DNA replication licensing factor MCM3 which is a subunit of the minichromosome maintenance protein complex MCM2-7 was identified as a new Pin1 interaction partner. The interaction is mediated by the WW domain of Pin1 and requires phosphorylation of Ser/Thr-Pro motifs within MCM3. In HeLa cells Pin1 facilitates the formation of the MCM2-7 complex and influences the binding of MCM3 to chromatin in a cell cycle dependent manner. During investigation of differently substituted new aryl hetaryl ketones and thioketones as inhibitors compounds with inhibitor constants towards Pin1 ranging in the micromolar and submicromolar range were detected. Furthermore a method to isolate cyclophilins from human serum was developed.

This approach provides for end-to-end verification and validation of process workflows, which is needed for process workflows that are intended for use in developing and maintaining high-integrity systems. We demonstrate the technical feasibility of our approach by applying it on the development of the US government’s process workflow for implementing, certifying, and accrediting cross-domain computer security solutions. Our approach involves identifying human-in-the-loop decision points in the process activities and then modeling these via statechart assertions. We developed techniques to specify and enforce workflow hierarchies, which was a challenge due to the existence of concurrent activities within complex workflow processes. Some of the key advantages of our approach are: it results in development of a model that is executable, supporting both upfront and runtime checking of process-workflow requirements; aids comprehension and communication among stakeholders and process engineers; and provides for incorporating accountability and risk management into the engineering of process workflows.

Processes are a fundamental component of most activities undertaken by humans. In software engineering and information assurance, in particular, it is important that processes be understandable, documented, and repeatable so as to ensure that the process outcomes are consistent and predictable. This dissertation provides a novel approach to process creation, documentation, checking, and maintenance that applies mathematical formalism to the engineering of processes that rely in large measure on human decision-making to advance the process flow. However, the modeling approach is sufficiently general for application to any process. This dissertation advances the state-of-the-art in software engineering by providing a formal computer-assisted end-to-end way to conduct requirements engineering. This dissertation advances the state-of-the-art in information assurance by developing a systematic approach that makes the creation of security processes precise and uses formal methods to allow upfront validation and runtime verification of modeled processes.
