As neoplasias mieloproliferativas (NMPs) BCR-ABL1 negativas compreendem a mielofibrose primária (PMF), trombocitemia essencial (TE) e a policitemia vera (PV). A patogênese e progressão dessas NMPs não estão completamente elucidadas. As metaloproteinases de matriz (MMPs) degradam a matriz extracelular, ativando citocinas e fatores de crescimento que, por sua vez, participam da tumorigênese e angiogênese. O objetivo deste estudo foi avaliar a relação da expressão gênica das MMPs, TIMPs, HIF1-α e SPARC com os marcadores angiogênicos bFGF e VEGFA em pacientes com MF e TE, considerando o status mutacional; bem como avaliar a regulação desses genes em camundongos submetidos à hipóxia, e em modelos HIF1-α(-/-) e VHL(-/-). Foram incluídos 21 pacientes com MF, 21 com MF pós-TE, 6 com MF pós-PV, 23 com TE e 78 indivíduos controle. As análises realizadas foram: dosagem sérica e expressão de RNAm de MMP2, MMP9, TIMP1, TIMP2 e SPARC, hemograma, determinação da proteína C reativa ultrassensível, determinação das concentrações de VEGFA e bFGF e avaliação das mutações nos genes JAK2, cMPL e CALR. A avaliação da densidade microvascular da medula óssea foi feita em 30 dos pacientes incluídos. A exposição excessiva à radiação Ultravioleta (UV) resulta em manifestações clínicas à pele humana como queimaduras, fotoenvelhecimento e câncer. A radiação UVA, preferencialmente, induz à formação de espécies reativas de oxigênio, enquanto que a radiação UVB é absorvida diretamente pelo DNA. Apesar de mecanismos endógenos auxiliarem na prevenção/reparação dos danos causados pela radiação UV, quando o dano excede a capacidade de reparação celular, diversos efeitos lesivos ocorrem na pele como alterações da matriz dérmica, resposta inflamatória e desidratação do estrato córneo. O uso de compostos fenólicos com atividade antioxidante pode auxiliar na prevenção das consequências patológicas da exposição à radiação UV. O presente trabalho teve como objetivo estudar em cultura de células da pele (HaCaT -queratinócito humano imortalizado e FHPD - fibroblasto humano primário dermal) exposta às radiações UVA e UVB a atividade fotoprotetora de 3 compostos fenólicos, ácido cafeico (AC), clorogênico (ACG) e rosmarínico (AR). Inicialmente, células HaCaT e FHPD cultivadas em monocamada foram expostas às doses crescentes de radiação UVA ou UVB e, após 24 horas, foram analisadas quanto a viabilidade, marcadores de morte celular, mediadores inflamatórios, presença de aquaporina e lesões de DNA. HaCaT quando exposta às radiações UVA e UVB são conduzidas à morte por apoptose, com aumento de Caspases 3 e 9, p53 e redução de PARP. Após a exposição à radiação UVA, HaCaT responde com aumento na liberação de IL-6, TNF-α e COX-2, internalização/redução de AQP3 da membrana, redução na liberação de MMP-2 e 9, aumento na liberação de MMP-1 e na produção de ERO. Quando expostos à radiação UVB, HaCaT aumenta a liberação de IL-6 e COX-2, promove internalização/redução de AQP3 na membrana e redução na liberação de MMP-2 e 9. FHPD são menos sensíveis à exposição a ambas as radiações, mostrando redução de viabilidade com parada de ciclo apenas frente à radiação UVA. Além disto, FHPD exposto a radiação UVA responde com aumento na liberação de IL-6 e danos no DNA do tipo 8-oxo-dG. Dentre os compostos, o ACG apresentou melhor atividade fotoquimioprotetora perante ambas as radiações UVA e UVB, pois foi capaz de reverter em HaCaT a morte celular induzida por ambas as radiações e de reverter a parada de ciclo em FHPD expostos à radiação UVA. HaCaT tratado com ACG e exposto à radiação UVA responde com aumento na expressão de AQP3 e PARP, aumento na expressão gênica de AQP3, redução na expressão gênica de CDKN1A e na liberação de MMP-1, 2 e 9. Após a radiação UVB, o tratamento com ACG aumenta a expressão gênica de AQP3, reduz a expressão gênica de CDKN1A, reduz a produção de COX-2 e aumenta a liberação de MMP-2 e 9. O tratamento com o AR apresentou atividade fotoquimioprotetora frente à radiação UVA, com HaCaT respondendo a radiação com aumento na população de células viáveis, aumento na expressão de AQP3 e PARP e na expressão gênica de AQP3, redução na liberação de MMP-1 e 9 e redução na produção de COX-2. FHPD tratados com AR apresentaram aumento na população em fase G1, na expressão de p21, e redução de danos de DNA tipo 8-oxo-dG. O tratamento de HaCaT com AC foi capaz de reverter a morte celular, aumentar a expressão de p53 e aumentar a liberação de MMP-2 e 9 frente à radiação UVB e de reduzir a produção de ERO, a expressão de p21 e a liberação de MMP-1, 2 e 9 frente à radiação UVA. Para FHPD, o tratamento com AC foi capaz apenas de reduzir a formação de danos de DNA tipo 8-oxo-dG. Os resultados indicam que o modelo proposto foi capaz de discriminar a atividade fotoprotetora dos compostos frente à radiação UVA e UVB. Além disto, foi possível demonstrar que os compostos antioxidantes se comportam de maneira distinta enquanto fotoprotetores no modelo empregado. Excessive exposure to Ultraviolet radiation (UV) results in clinical manifestations in human skin such as burns, photo-aging and cancer. UVA radiation preferentially induces formation of reactive oxygen species, while UVB radiation is absorbed directly by the DNA. Although endogenous mechanisms are able to prevent/repair cellular damages caused by UV radiation, excess cellular damage retains cells repair capacity and also results on diverse harmful effects on skin, such as, changes in the dermal matrix, inflammatory response and dehydration of the stratum corneum. The use of phenolic compounds with antioxidant activity may help preventing pathological conditions caused by UV radiation. This work aimed to study the photoprotective activity of three phenolic compounds, caffeic (CA), chlorogenic (CGA) and rosmarinic acid (RA) in human skin cells (HaCaT - immortalized human keratinocytes and HDSF - human dermal skin fibroblast) exposed to UVA and UVB radiation. Initially, HDSF and HaCaT cells were exposed to increasing doses of UVA and UVB radiation. After 24 hours of exposure, we evaluated cell viability, cell death, inflammatory mediators, aquaporin and DNA damage. Exposure to UVA and UVB radiation in HaCaT cells results on apoptotic cell death, with an increase of caspases 3 and 9, p53 and reduction of PARP. HaCaT cells when exposed to UVA radiation resulted on increased levels of IL-6, TNF-α and COX-2, internalization of the membrane AQP3, reduction of MMP-2 and MMP-9 release, increase of MMP-1 and ROS production. After UVB radiation, HaCaT cells resulted on an increase of IL-6 and COX-2 production, it also promoted internalization of membrane AQP3 and reduced release of MMP-2 and 9. HDSF were less sensitive to both radiations. Moreover, HDSF resulted in cell viability decrease and cell cycle arrest only after UVA radiation. Furthermore, HDSF when exposed to UVA radiation resulted on an increase of IL-6 production and in DNA damage (8-oxo-dG). Among the studied compounds, CGA presented better photochemiprotective activity towards UVA and UVB radiation. Also, this compound was able to reverse cell death in HaCaT after exposure to both radiations and inhibited cell cycle arrest in HDSF after UVA radiation exposure. HaCaT cells treated with CGA and exposed to UVA radiation resulted on an increase in AQP3 and PARP expression, increased in AQP3 gene expression, reduction in CDKN1A gene expression and reduction in MMP-1, 2 and 9 release. After UVB radiation, GCA treatment increases AQP3 gene expression, reduces CDKN1A gene expression, reduces COX-2 production and increase MMP-2 and 9 releases. The AR treatment showed photochemiprotective activity towards the effects of UVA radiation, with HaCaT responding with an increase on cells viability, increased in PARP and AQP3 expression and in AQP3 gene expression, decreased MMP-1 and 9 releases and reduced COX-2c production. HDSF when treated with AR showed an increase in G1 phase population, in p21 expression and reduced DNA damage-type 8-oxo-dG. HaCaT cells treated with AC reversed cell death, increased p53 expression and increased MMP-2 and 9 releases after UVB radiation and reduced ROS production, p21 expression and MMP -1, 2, 9 release after UVA radiation. HDSF treated with AC was only able to reduce the formation of 8-oxodG DNA damage. These results indicated that the proposed model was able to discriminate the photochemiprotective activity of the studied compounds against the UVA and UVB radiation. In addition, it was demonstrated that the each studied antioxidant have different photoprotective mode of action.

A psoríase é uma doença inflamatória crônica, de alta incidência mundial, caracterizada por lesões de pele e prurido. Achados prévios do grupo mostraram que a administração i.p. do doador lento de sulfeto de hidrogênio (H2S), GYY4137, inibiu significativamente a inflamação cutânea e coceira em animais com psoríase. Assim, neste estudo avaliou-se o efeito terapêutico de uma nanodispersão tópica contendo GYY4137 sobre a psoríase induzida por imiquimode em camundongos e, comparou-se farmacologicamente o efeito do GYY4137 versus dexametasona. Animais controle ou psoríase foram tratados (1 e 2x/dia) com a nanodispersão (65 mg) contendo GYY4137, dexametasona ou apenas veículo. A aplicação tópica da nanodispersão com o GYY4137 (4%) 2x/dia, foi mais efetiva e reduziu de forma significativa o rubor, espessura, células sanguíneas totais, MPO e níveis de IL-6 e IL-1β. Os efeitos farmacológicos promovidos pelo H2S foram semelhantes aos da dexametasona, porém, a nanodispersão contendo GYY4137 demonstrou efeitos sistêmicos menos pronunciados quando comparada ao glicocorticóide. Psoriasis is a chronic inflammatory disease, with high incidence worldwide and, characterized by skin lesions and intense itching. Previous findings of this group showed that systemic administration of the slow donor release of hydrogen sulfide (H2S), GYY4137, significantly inhibited skin inflammation and itching in mice with psoriasis. Thus, this study evaluated the therapeutic effect of a topical nanodispersion containing GYY4137 on the Imiquimod-induced psoriasis mice model and compared pharmacologically the effect of GYY4137 versus dexamethasone. Control or psoriasis animals were treated (1 and 2x/day) with nanodispersion (65 mg) containing GYY4137, dexamethasone, or just vehicle. Topical application of the nanodispersion with GYY4137 (4%) 2x/day was more effective and significantly reduced redness, thickness, total blood cells, MPO, and IL-6 and IL-1β levels. The pharmacological effects caused by H2S were similar to those of dexamethasone, however, the nanodispersion containing GYY4137 demonstrated less systemic effects when compared to glucocorticoid.

A glicação não enzimática das proteínas é um fator comum para a fisiopatologia de uma série de transtornos relacionados ao envelhecimento e a doenças como o diabetes mellitus (DM). O geração dos produtos de glicação, os AGEs (do inglês: Advanced Glycation End Products) se dá através de reações de glicação da mariz extracelular (MEC) na derme e têm sido apontado como um dos fatores responsáveis pela perda de elasticidade e deficiência de cicatrização da pele. A permeação cutânea de compostos anti-AGE é uma limitação importante para eficiência terapêutica de compostos que devem atingir camadas mais profundas da pele. Modelos de pele reconstruída contendo equivalente dérmico glicado são estruturas tridimensionais geradas in vitro que mimetizam a pele humana e representam um eficiente modelo para o estudo de células e modificações provocadas na MEC no processo de envelhecimento e DM. O modelo 3D de pele reconstruída tem características metabólicas, de permeabilidade e atividade semelhantes à da pele original, potencializando seu papel nas investigações sobre permeabilidade de drogas, toxicidade, irritação, eficácia e segurança de compostos e diferenciação de queratinócitos. Uma série de compostos naturais ou sintéticos inibidores de AGEs têm sido descobertos e apresentados recentemente e podem representar inovação terapêutica no tratamento de modificações causadas pela a formação e acúmulo destes AGEs também na pele. Este estudo avaliou o desenvolvimento da pele reconstruída glicada e posteriormente, a avaliação da eficácia e toxicidade de compostos anti-glicação como aminoguanidina e carnosina em modelo de pele reconstruída glicada. Em perspectiva, este estudo contribuiu para o desenvolvimento de uma nova tecnologia in vitro, a pele reconstruída glicada, que auxiliará a compreensão da biologia da interação célula-MEC mimetizando processos fisiopatológicos importantes como o envelhecimento e o DM. The Advanced Glycation End Products (AGEs) of proteins is a common factor to the pathophysiology of a number of disorders related to aging and diseases such as diabetes mellitus (DM). The generation of the AGEs products on skin occurs mainly through non-enzymatic glycation reactions of the dermal extracellular matrix and has been touted as one of the factors responsible for loss of elasticity and disability of skin healing. The skin permeation of compounds is an important limitation for therapeutic/cosmetic efficacy of anti-AGE compounds, which must reach the deepest layers of the skin. Reconstructed skin model containing dermal equivalent modified by in vitro glycation is able to mimic the elderly human skin and represent an efficient model for the study of cells interactions and changes in extracellular matrix induced by aging and diabetes. The 3D reconstructed skin model has metabolic characteristics, permeability and activity similar to the original skin, reinforcing its role in drug permeability of investigations toxicity, irritation, safety and efficacy evaluation of compounds and differentiation of keratinocytes. A number of natural or synthetic AGEs inhibitor compounds have been recently discovered and displayed and can represent therapeutic innovation for the treatment of changes caused by the aging of the skin. In this study we performed the development of reconstructed glycated skin model and evaluated the efficacy and toxicity of anti-glycation compounds such as aminoguanidine and carnosine. In perspective, this study has contributed to the development of a new technology in vitro, and for the understanding cell-extracellular matrix interaction during the aging of skin.

Os pacientes com melanomas, em geral, apresentam extrema quimiorresistência e prognóstico ruim, com taxa de sobrevida de aproximadamente seis meses; portanto, novas estratégias terapêuticas são necessárias. As células deste tipo de tumor acumulam alterações na expressão gênica que contribuem para a proliferação descontrolada, evasão de senescência e inibição de morte celular em múltiplas rotas intracelulares. No Capítulo 1, foi explorado o controle epigenético do supressor tumoral RECK. Esse gene é largamente expresso em tecidos normais, com correlação de sua expressão com melhor prognóstico. Em tumores, RECK é inibido, incluindo em melanoma. Neste estudo, sua inibição por hipermetilação do promotor e remodelamento da cromatina por HDACs não foi o único fator inibitório nas linhagens de melanoma analisadas. Mesmo após a utilização a remoção de marcas epigenéticas associadas ao silenciamento gênico, a expressão proteica não pôde ser recuperada nas linhagens utilizadas neste trabalho. No Capítulo 2, isolou-se subpopulações clonais ao acaso a fim de modelar a heterogeneidade tumoral intrínseca. Neste modelo experimental, caracterizamos a presença de dois perfis tumorais subclonais: uma mais proliferativa, invasiva e sensível a vemurafenibe; e outra menos proliferativa, mais resistente e com maior expressão de RECK e fatores ligados a EMT. Nossos resultados, em conjunto, mostraram que as linhagens celulares parentais utilizadas apresentam diferenças entre si quanto à viabilidade celular, indução de apoptose e fosforilação de ERK após o tratamento com vemurafenibe. Segundo o nosso modelo, a resistência intrínseca ao vemurafenibe está presente e reflete a heterogeneidade do tumor inicial. Com estes resultados, pretendemos contribuir para o conhecimento sobre a composição clonal presente na heterogeneidade tumoral, além de contribuir para a função de RECK na biologia do melanoma. Patients with melanoma often present extreme chemoresistance and poor prognosis, with survival rates of approximately six months; therefore, new therapeutic strategies are necessary. Melanoma cells accumulate genotypic changes that contribute to uncontrolled proliferation, evasion of senescence and cell death inhibition in multiple cellular pathways. In Chapter 1, we have explored the epigenetic control of the tumor suppressor RECK. This gene is widely expressed in normal tissues, with correlation of its expression with better prognosis. In tumors, RECK is inhibited, including melanoma. Here, RECK inhibition by promoter hypermethylation and chromatin remodeling by HDACs was not the only inhibiting factor in the melanoma cell lines analyzed. Even after removing the epigenetic marks associated to gene expression silencing, the protein expression could not be recovered in the cell lines used in this study. In Chapter 2, it has been isolated several tumor clonal subpopulations in order to modulate the intrinsic tumor heterogeneity. In this experimental model, we have characterized the presence of two tumor subclonal profiles: a more proliferative, invasive and sensitive to Vemurafenib; and other less proliferative, less sensitive to Vemurafenib and presenting more expression of RECK and factors associated to EMT. Our results together have showed the parental cell lines used here differed from each other in terms of cell viability, induction of apoptosis, and ERK phosphorylation after treatment with Vemurafenib. According to our model, the intrinsic resistance to Vemurafenib is present and reflects the heterogeneity of the initial tumor. With these results, we intend to contribute to the knowledge of the tumor clonal subpopulations composition in tumor heterogeneity, and contributes to the RECK function in melanoma biology.

As neoplasias induzidas pela forma oncogênica da KRAS são doenças muito comuns, para as quais não existem terapias efetivas. Uma inibição direta da KRAS falhou em ensaios clínicos e esforços intensos tem sido feitos para identificar alvos de KRAS importantes para a oncogênese. Uma via promissora regulada por KRAS, que tem sido pouco explora a é a via dos microRNAs (miRNAs). Nossa meta foi identificar miRNAs regulados pela KRAS em células pulmonares e pancreáticas, que possam contribuir para o fenótipo oncogênico. Para alcançar esta meta nós usamos duas abordagens: (1) Nós investigamos o miRNA 486-5p como um alvo de KRAS em câncer de pâncreas e de pulmão. A expressão deste miRNA havia sido correlacionada positivamente com a presença de mutações em KRAS em pacientes portadores de câncer de cólon; (2) Nós usamos uma plataforma de microarranjo para identificar miRNAs diferencialmente expressos entre células humanas primárias imortalizadas pulmonares ou pancreáticas e suas linhagens isogênicas transformadas por KRAS. Na primeira abordagem, conseguimos mostrar que a expressão do miRNA 486-5p está correlacionada ao status de KRAS em células primárias pulmonares, mas não em células primárias pancreáticas. Além disso, geramos células pulmonares tanto com ganho e perda de função de KRAS e demonstramos que KRAS regula a expressão do miRNA 486-5p. Também de terminamos uma correlação negativa entre a expressão de KRAS e a expressão do alvo do miRNA 486-5p FoxO1, um supressor tumoral. Para avaliar como o miRNA 486-5p afeta as propriedades oncogênicas induzidas por KRAS, nós transfectamos oligonucleotídeos inibitórios para o miRNA 486-5p em células pulmonares positivas para mutações em KRAS. A inibição da expressão do miRNA 486-5p levou a uma redução da clonogenicidade e viabilidade celulares. Esta redução não está associada a um aumento de morte celular, mas a uma redução da proliferação celular. Interessantemente, a transfecção de oligonucleotídeos mímicos do miRNA 486-5p em células pulmonares negativas para mutações em KRAS ou em células com perda de função de KRAS por RNAi levou a um aumento da proliferação e clonogenicidade. Estes dados indicam que o miRNA 486-5p, não só é um alvo de KRAS em câncer de pulmão, mas também age como um oncomiR contribuindo para a proliferação celular induzida por KRAS. Na nossa segunda abordagem, nós identificamos 17 miRNAs com expressão aumentada e 3 com expressão diminuída em células primárias pancreáticas expressando KRAS oncogênica. Destes, 9 miRNAs foram foram também identificados por metanálise de dados de microarranjo publicados comparando amostras tumorais pancreáticas com amostras não tumorais. Apesar do experimento de microarranjo com as linhagens primárias pulmonares não ter produzido resultados estatisticamente significativos após a correção por FDR, uma tendência à expressão diferencial foi observada para vários miRNAs e nós validamos por qPCR a expressão diferencial dos miRNAs 720 e 139-3p. Em conclusão, nós conseguimos identificar miRNAs regulados pela KRAS tanto em células pulmonares, quanto pancreáticas. Um melhor entendimento das suas funções biológicas, bem como dos alvos por eles regulados nestes contextos, pode revelar novas vias para a exploração terapêutica. KRAS-induced lung cancer is a very common disease, for which there are currently no effective therapies. Direct targeting of KRAS has failed in clinical trials and intense efforts are underway to identify KRAS targets that play a crucial role in oncogenesis. One promising KRAS-regulated pathway that has so far been overlooked is the micro RNA (miRNA) pathway. Our goal was to identify miRNAs regulated by oncogenic KRAS in lung and pancreatic cells that could contribute to the oncogenic phenotype. In order to achieve this goal we used two different approaches: (1) We investigated miRNA 486-5p as a KRAS target in lung and pancreatic cancer. The expression of this miRNA had been correlated to the presence of KRAS mutations in colon cancer patients; (2) we used a microarray platform to identify differentially expressed miRNAs between immortalized human primary pulmonary or pancreatic epithelial cell lines and their isogenic K-Ras-transformed counterparts. In our first approach, we were able to show that mi486-5p expression correlates with KRAS status in lung primary cells, but not in pancreatic primary cells. Furthermore, we generated lung cancer cells with either gain-of-function or loss-of-function of KRAS and demonstrated that KRAS regulates miRNA 486-5p in these cells. We also found, in all lung cell models analyzed, a negative correlation between expression of KRAS and expression of miR-486-5p target FoxO1, a tumor suppressor. In order to evaluate how miR-486-5p affects KRAS-induced oncogenic properties, we transfected miR-486-5p inhibitor oligonucleotides into KRAS-positive lung cancer cell lines. Inhibition of miR-486-5p expression leads to reduced clonogenic growth and viability. This reduction is not associated with increased cell death, but with decreased cell proliferation. Interestingly, transfection of miR-486-5p double-stranded RNA mimic oligonucleotides in to KRAS negative lung cancer cell lines or into cells with loss-of-function of KRAS by RNAi leads to enhanced proliferation and clonogenicity. These results indicate, not only that miR-486-5p is a KRAS target in lung cancer, but also that miR-486-5p acts as an oncomiR contributing to KRAS-induced cell proliferation. In our second approach, we identified 17 upregulated microRNAs and 3 downregulated microRNAs in the primary pancreatic cell line expressing KRAS. Of these, 9 miRNAs were also identified by a metanalysis of published microarray datasets comparing pancreatic cancer patient samples to non-cancerous pancreatic tissues. Even though our array experiment in the primary pulmonary cells did not produce statistically significant results after FDR correction, differential expression trends were seen for many miRNAs and we validated miRNAs 720 and 139-3p as differentially expressed. In conclusion we were able to identify miRNAs regulated by KRAS both in lung and pancreatic cancer cells. Further understanding of their biological function, as well as the targets they regulate in these settings, could uncover novel pathways for therapy design.

Gliomas são tumores altamente invasivos, resistentes aos tratamentos disponíveis atualmente e com alta taxa de mortalidade. A superexpressão de RECK na linhagem de glioma humano T98G comprometeu a capacidade das células de migrar e invadir in vitro, com rearranjo do citoesqueleto e alteração na distribuição espacial de FAK fosforilado. Entretanto, o possível mecanismo envolvido na inibição da migração mediada por RECK não foi desvendado. Para estudarmos os mecanismos envolvidos nesta alteração da capacidade migratória, as células T98G foram transfectadas com o vetor plasmidial pCXN2-hRECK (RECK+). A via das integrinas, a atividade de alguns membros da família das RhoGTPases e elementos do citoesqueleto foram avaliados através de imunoblotting, imunomarcação e ensaios de pull-down para as células RECK+ em comparação com células T98G não-transfectadas (WT), células T98G transfectadas com vetor pCXN2 na ausência do gene RECK (vetor) e fibroblastos primários humanos (FF287). Nossos resultados mostram um aumento na expressão de integrina β1 e uma diminuição da fosforilação de FAK no sítio de auto-fosforilação Tyr397 que, juntamente com o aumento das fibras de estresse e a diminuição dos lamelipódios, sugerem um fenótipo menos migratório da célula. Porém, quando avaliada a atividade de Rac1, esta se mostrou aumentada, embora uma das vias de ativação de Rac1 seja através da fosforilação de FAK levando à formação dos lamelipódios. A hipótese é que RECK inibe a quebra das adesões focais que participam do processo de migração, dificultando a mobilidade celular. Como as células continuam recebendo o estímulo para migrar, estas ativam Rac1 através de uma via independente de FAK. Além disso, a imunomarcação de paxilina mostrou um aumento no tamanho das adesões focais nas células RECK+, indicando que RECK pode influenciar nas estruturas responsáveis pelo contato célula-matriz. Gliomas are highly invasive, treatment-resistant and lethal tumors. Overexpression of RECK in human glioma cell line T98G decreased cell migration and invasion in vitro, lead to cytoskeleton rearrangement and caused changes in phospho-FAK distribution. However, the pathway involved in RECK-mediated inhibition of cell migration has not been elucidated yet. To study the mechanisms by which RECK affects cell motility, T98G cells were transfected with pCXN2-hRECK vector (RECK+). Some proteins involved in the integrin pathway, activity of some proteins of RhoGTPase family and cytoskeleton proteins were analyzed through immunoblotting, immunostaining and pull-down assay in RECK+ cells and compared with non-transfected T98G cells, T98G transfected with pCXN2 without RECK gene and human primary fibroblasts (FF287). Our results showed an increase in integrin β1 expression and a decrease in FAK phosphorylation in the Tyr397 site, which together with the increase of stress fibers and decrease of lamellipodia, suggest a less migratory phenotype. Despite this, Rac1 activity was increased even though one of Rac activation pathways is through phospho-FAK, leading to lamellipodium formation. Our hypotheses is that RECK affects focal adhesion turnover, diminishing cell motility. As cells are still receiving a positive signal to migrate, they activate Rac1 through a FAK-independent pathway. Besides that, paxillin immunostaining showed that focal adhesions are larger in RECK+ cells, indicating that RECK can influence structures related with cell-matrix contact.

Cell culture is characterized by maintaining live cells in the laboratory regardless of the organism in which they originated. This technique has contributed to better understanding of molecular cell mechanisms, permitting important scientific advances, for example, concerning vaccine production and tumor cell biology. Three-dimensional (3D) cell culture initially derived from commonly used cell cultures (monolayer cell cultures). As a particularity, a 3D cell culture permits cells to explore the three dimensions of the space thereby increasing cell-cell interactions, as well as interaction with the environment. When grown in this system, cells form structures known as multicelullar spheroids. The interior of these spheroids present cell heterogeneity, microenvironment formation and different exposure to factors, such as nutrients and oxygen. Owing to the fact that these characteristics are very similar to those of in vivo avascular tumors, 3D cell culture advanced in various research lines, thus becoming a widely used model in radiology and chemotherapy essays. In studies related to breast cancer biology, spheroids are becoming widely used in the aim to comprehend luminal space morphogenesis. In this context, the objective here was to develop a 3D culture model that made it possible to analyze spheroid formation using 3 characteristics that have not yet been described in the literature: without addition of basement membrane mimetic, use of tumor cell lines and, lastly, cell exposure for periods longer than 30 days in 3D cell culture. We showed that, in the aforementioned conditions, MCF-7 cells reorganize themselves in tubular and acinar structures. In both situations, lumen formation was accompanied by the establishment of a layer of polarized cells, an arrangement that is very similar to that of breast glands. The presented results suggest the existence of an MCF cell line population not completely committed to the tumor phenotype. When maintained as differentiated, MCF-7 cell spheroids can be a new model for studies regarding lumen formation, thereby exploring the role of diiferent pathways, such as those related to cell apoptosis, autophagy, differentiation and survival.

Gliomas são tumores do sistema nervoso central e o glioblastoma multiforme (GBM) é sua forma mais comum e de pior prognóstico. A proliferação desordenada, migração estão entre os processos biológicos que conferem ao GBM um comportamento altamente agressivo. O objetivo deste estudo foi analisar in vitro o papel de PGE2 na proliferação, migração e apoptose da linhagem de glioma humano T98G através de tratamento com ácido gama-linolênico (GLA), Ibuprofeno (IBU) e adição de prostaglandina E1 (PGE1) e prostaglandina E2 (PGE2) exógeno. Nossos resultados demonstraram através uma diminuição nas taxas de proliferação e de migração e uma indução de apoptose nas células T98G tratadas com GLA ou IBU. Por outro lado, a adição exógeno de PGE1 ou PGE2 resultou num aumentou da proliferação e da migração e numa inibição da apoptose. Esses resultados demonstram a influência de PGE2 na linhagem T98G. Gliomas are tumors of the central nervous system and glioblastoma multiforme (GBM) is the most common form with the worst prognosis. Uncontrolled cell proliferation, migration are among the biological processes that give GBM a highly aggressive behavior. The aim of this study was to analyze in vitro the role of PGE2 the proliferation, migration and apoptosis of the T98G human glioma cell line through treatment with gamma-linolenic acid (GLA), ibuprofen (IBU) and the addition of exogenous prostaglandin E1 (PGE1) and prostaglandin E2 (PGE2). Our results demonstrated through various tests a decrease in the rates of proliferation and migration and an induction of apoptosis in T98G cells treated with GLA or IBU. On the other hand, the addition of exogenous PGE1 or PGE2 resulted in increased proliferation and migration and inhibition of apoptosis. These results demonstrate the influence of PGE2 in the T98G cell line.

Introdução: As CYP3A4 e CYP3A5 são enzimas do citocromo P450 responsáveis pela biotransformação de esteróides endógenos e vários fármacos, entre eles as estatinas. Polimorfismos nos genes CYP3A4 e CYP3A5 (CYP3A4*1B, CYP3A5*3C e CYP3A5*1D) foram associados com diferenças na resposta hipolipemiante de indivíduos tratados com atorvastatina e sinvastatina. Neste estudo foram avaliados os efeitos de hipolipemiantes sobre a expressão e a atividade de CYP3A4 e CYP3A5, em linhagens celulares HepG2 e Caco-2 e em CMSP de indivíduos hipercolesterolêmicos, e sua relação com variantes de CYP3A4 e CYP3A5. Métodos: Foram analisados 99 indivíduos normolipidêmicos (NL) e 139 hipercolesterolêmicos (HC). Os HC foram tratados com atorvastatina (10 mg/dia/4 semanas). A genotipagem das variantes CYP3A4*1B, CYP3A5*3C e CYP3A5*1D foi feita por PCR-RFLP ou sequenciamento. A análise da expressão de RNAm de CYP3A4 e CYP3A5 foi avaliada por PCR em tempo real quantitativo (PCRq). As proteínas totais de HepG2 foram avaliadas por Western Blotting. A atividade de CYP3A4 e CYP3A5 in vivo foi avaliada pela relação entre cortisol e seu metabólito, 6β-hidróxicortisol, na urina (razão 6βOH-cortisol/cortisol), por CLAE. Resultados: O perfil de expressão basal de RNAm de CYP3A4 e CYP3A5 é diferente entre HepG2 e Caco-2. Caco-2 expressa 31 vezes mais CYP3A4 e 122 vezes mais CYP3A5 que HepG2. Em células HepG2 tratadas por 12 h, a atorvastatina 20 µM aumentou a expressão de CYP3A4 em 10 vezes, em relação ao controle (p=0,006). Após 24 h de tratamento, atorvastatina (1-20 µM) aumentou a expressão de CYP3A4 em 5 a 8 vezes, nas HepG2 (p< 0,001). Para CYP3A5, a exposição por 12 h à atorvastatina 20 µM aumentou a expressão em 4 vezes em relação ao controle ( p 0,05). Os indivíduos NL apresentam atividade de CYP3A4 e CYP3A5 basal maior que os indivíduos HC- (p0,05). As variantes gênicas estudadas (CYP3A4*1B, CYP3A5*3C e CYP3A5*1D) como grupos haplotípicos não afetaram a resposta ao tratamento, a expressão de RNAm ou a atividade de CYP3A4 e CYP3A5, embora o haplótipo AGT tenha expressão basal de RNAm de CYP3A5 menor que os portadores de haplótipos GAT e GAC (p0.05). CYP3A4/5 activity was higher in NL subjects than in HC (p0.05). Um dos fatores que regula o córtex adrenal é o hormônio adrenocorticotrópico, ACTH, no entanto, o fator de crescimento de fibroblastos do tipo 2, FGF2, e os peptídeos N-terminais da pro-opiomelanocortina, N-POMC, também podem estar envolvidos. As vias de sinalização das proteínas quinases: ERK, JNK e p38, juntamente com outras vias como PKA, PKC e PI3K/Akt são importantes para a definição trófica das células. Nós analisamos a importância destas vias de sinalização e sua influência na viabilidade, proliferação e morte celular, induzidas pelo ACTH, FGF2 e N-POMC, utilizando inibidores farmacológicos e moleculares em culturas primárias de células adrenocorticais, células glomerulosa e fasciculadas/reticulares. Nossos resultados mostram que as vias mediadoras envolvidas na resposta proliferativa do FGF2 e da N-POMC são, respectivamente, as vias ERK/JNK e ERK/JNK/Akt. Por outro lado, a resposta pró-apoptótica promovida pelo ACTH é mediada pela via p38, provavelmente associada à ausência de ativação das vias relacionadas com a sobrevivência, como as vias ERK e JNK. One of the factors that regulate adrenal cortex is the adrenocorticotrophic hormone, ACTH, however, the fibroblast growth factor type 2, FGF2) and pro-opiomelanocortin N-terminal peptides, N-POMC, might also be involved. The mitogen-activated protein kinase pathways: ERK, JNK and p38, together with other signaling pathways such as PKA, PKC and PI3K/Akt are important for cells trophic definition. We analyze the importance of these pathways and their influence in viability, proliferation and cell death stimulated by ACTH, FGF2 and N-POMC, using pharmacological and molecular inhibitors in primary culture of adrenocortical cells, glomerulosa and fasciculata/reticularis cells. Our results show that the mediating signaling pathways involved in FGF2 and N-POMC proliferative effects are, respectively, ERK/JNK and ERK/JNK/Akt. On the other hand, the pro-apoptotic response promoted by ACTH is through p38 signaling, probably associated with the absence of activation of other pathways involved with cell survival, like ERK and JNK.

Os modelos de reconstrução do microambiente são úteis para investigar as propriedades biológicas dos melanócitos humanos com a matriz e como plataforma para testes de novos fármacos. Existe uma demanda crescente para a utilização de pele e derme reconstruídas em laboratório, em ensaios in vitro de citotoxicidade, viabilidade celular, crescimento celular, irritabilidade e avaliação dos constituintes da matriz extracelular. Caracterizamos, em equivalente dérmico, alguns mecanismos de viabilidade e invasão de melanoma metastático humano quando na presença da matriz, a fim de ampliar o conhecimento a respeito de novas terapias contra o melanoma e entender os mecanismos que favorecem seu potencial invasivo, mimetizando com mais fidelidade o que ocorre in vivo. Através da validação deste modelo, constatamos que ele é essencial para resgatar a fisiopatologia do tumor, pois o melanoma, na presença de equivalente dérmico, torna-se capaz de evadir mecanismos de morte e aumentar a secreção de metaproteinases e citocinas que favorecerão a evolução tumoral. Avaliamos também as propriedades antineoplásicas do ácido clorogênico, que já foram relatadas em um grande número de tumores, porém os mecanismos que levam à sua ação antitumoral ainda não foram bem elucidados. Diante dos trabalhos enfatizando o efeito quimioprotetor dos polifenóis, referentes à sua ação antioxidante em células neoplásicas e com potencial metastático, não há na literatura estudos que comprovem a eficácia do ácido clorogênico sobre células de melanoma metastático humano. Portanto, este estudo visou recriar a estrutura dérmica in vitro e, a partir disso, comparar a resposta de fármacos em células de melanoma cultivada em equivalente dérmico, a fim de avaliar se há modulação diferencial nesse substrato, sugerindo, portanto, um protocolo mais eficaz para a prospecção de fármacos antimelanoma. The microenvironment reconstruction models are useful for investigating the biological properties of human melanocytes onto the matrix and as platform for testing new drugs. There is a growing demand for the use of reconstructed skin and dermis in the laboratory for in vitro assays of cytotoxicity, viability, cell growth, irritability, and evaluation of the extracellular matrix. We characterize in dermal equivalent some mechanisms for cell viability and invasion of human metastatic melanoma in the presence of matrix, the order to increase knowledge about new therapies against melanoma and to understand the mechanisms that favor its invasive potential, to more closely mimic what occurs in vivo. By means of this model, we find that it is essential to rescue the tumor physiopathology as melanoma, in the presence of dermal equivalent, it is able to evade death mechanisms and increase the expression of metalloproteinases and cytokines which favor the tumor evolution. We also evaluated the antineoplastic properties of chlorogenic acid, that have been reported in a large number of tumors, but the mechanisms that lead to its antitumor action has not been well elucidated. Before the work emphasizing the chemoprotective effect of polyphenols related to its antioxidant action in neoplastic cells and with metastatic potential, there is no literature studies confirming the effectiveness of chlorogenic acid on human metastatic melanoma cells. Therefore, this study aims to rebuild the dermal structure in vitro, and from this, to compare the response to drugs in melanoma cells dermal equivalent cultured in order to evaluate whether modulation differential on the substrate, thus suggesting a protocol more effective for prospecting antimelanoma drugs.

O Papilomavírus Humano (HPV), da família Papilomaviridae, são vírus epiteliotrópicos que provocam lesões de pele ou mucosa. O carcinoma do colo uterino é uma das principais causas de morte de mulheres em todo o mundo, sendo a infecção pelo HPV o principal fator de risco para o desenvolvimento do carcinoma cervical. Durante o processo maligno, a migração descontrolada de células neoplásicas, é uma característica fundamental da invasão tumoral, sendo que as metaloproteinases de matriz (MMPs) participam largamente deste processo pois são responsáveis pela clivagem de proteínas da matriz extracelular. O projeto propôs investigar a expressão de MMP-9 total em amostras cérvico-vaginais com inflamação, lesão e tumor. Foram coletadas duas amostras cérvico-vaginais como esfregaços para diagnóstico citológico e imunocitoquímica. A coleta foi realizada em 630 pacientes da Penitenciária Feminina de Sant´ana, no período de agosto de 2009 a agosto de 2010. As lâminas foram submetidas à imunocitoquímica para detecção da expressão de MMP-9. Os resultados indicaram um aumento da expressão de MMP-9 total diretamente proporcional com o aumento do grau das lesões nos esfregaços, corroborando com dados de histologia da literatura. Portanto, tal método de correlação de MMP-9 total com esfregaços cérvico-vaginais poderá auxíliar o prognóstico em esfregaços. The Human Papillomavirus (HPV) are epitheliotropic viruses from family Papillomaviridae, which cause skin or mucous lesions. Carcinoma of the cervix is a major cause of death in women worldwide, and HPV infection a major risk factor for development of cervical carcinoma. During the malignant process, cell migration is a fundamental characteristic of tumor invasion, and the matrix metalloproteinases (MMPs) are responsible for the cleavage of extracellular matrix proteins. This project aims to investigate the expression of MMP-9 in different degrees of injury in the smear. We collected two cervical smears for cytological diagnosis and immunocytochemistry. Data collection was conducted in 630 women of the Sant´ana Women\'s Penitentiary, during the period August 2009 to August 2010. The slides were submitted to immunocytochemistry to detect the expression of MMP-9 and noticed an increase in protein expression in parallel with the increased lesions grade, which corroborates the histology of the literature and could be a method prognostic aid in vaginal smears.

O Metotrexato (MTX) é um fármaco utilizado como anti-inflamatório no tratamento da artrite reumatóide (AR). O risco de doença cardiovascular em pacientes com AR é menor quando tratados com MTX. Apesar dessa evidência, há poucos relatos da utilização de MTX para o tratamento da aterosclerose. Foi desenvolvida em nosso laboratório uma nanopartícula rica em colesterol (LDE), a qual é reconhecida pelos receptores da lipoproteína de baixa densidade (LDLr) após injeção na corrente sangüínea. A LDE concentra-se em células com hiperexpressão de LDLr, em processos proliferativos como a aterosclerose. Dessa maneira, a LDE pode ser utilizada como veículo para o direcionamento de fármacos contra essas células. A molécula de MTX foi latenciada e a modificação do fármaco aumentou a sua incorporação à LDE. A proposta desse estudo foi avaliar a eficácia de MTX associado à LDE (LDE-MTX) no tratamento da aterosclerose em coelhos além de investigar o efeito desse complexo na expressão de genes inflamatórios que participam do processo aterogênico. Para realização do estudo foram utilizados quatro grupos de 10 coelhos (raça New Zealand) cada, sendo que todos foram submetidos a uma dieta rica em colesterol por 8 semanas. Após as primeiras 4 semanas de dieta, os grupos foram tratados com LDE-MTX (grupo LDE-MTX), MTX comercial (grupo MTX comercial), LDE (grupo controle LDE) ou solução salina (grupo controle Salina), via endovenosa por 4 semanas. O grupo LDE-MTX não apresentou toxicidade ao longo do tratamento de acordo com os parâmetros utilizados, enquanto que o grupo MTX comercial apresentou uma queda acentuada de eritrócitos ao final do tratamento (p

Para caracterizar os potenciais evocados auditivos de crianças com distúrbio fonológico e verificar a evolução dos resultados destes potenciais frente à terapia fonoaudiológica, foram avaliadas por meio do potencial evocado auditivo de tronco encefálico, potencial evocado auditivo de média latência e potencial cognitivo, 24 crianças sem distúrbio fonológico e 23 crianças com distúrbio fonológico. Os resultados demonstraram que crianças com distúrbio fonológico apresentam diferença estatisticamente significante no potencial evocado auditivo de tronco encefálico e no potencial cognitivo antes da terapia fonoaudiológica, observando melhora nestes potenciais pós terapia fonoaudiológica In order to characterize the auditory evoked potentials of children with phonological disorder and to verify these potentials results improvement after speech therapy, 24 children without phonological disorder and 23 children with phonological disorder were evaluated through brainstem auditory evoked potential, middle latency response and cognitive potential. The results showed that children with phonological disorder present a statistically significant difference in the brainstem auditory evoked potential and in the cognitive potential before the speech therapy, with an improvement of those potential after speech therapy

A invasão de células tumorais para o tecido cerebral sadio é a marca patológica dos gliomas e contribui para o fracasso das modalidades terapêuticas atuais (cirurgia, radioterapia e quimioterapia). As células da glia transformadas apresentam os padrões clássicos do processo invasivo, incluindo adesão celular aos componentes da matriz extracelular (MEC), locomoção celular e a capacidade de remodelar o espaço extracelular. As metaloproteases de matriz (MMPs) são essenciais para o remodelamento adequado da MEC e para a invasão. O proteína supressora de tumor e metástase RECK regula pelo menos três diferentes membros da família das MMPs, especificamente: MMP-2, MMP-9 e MMP-14. Com o propósito de mimetizar o processo invasivo in vivo, as linhagens celulares de glioma humano A172 e T98G, respectivamente não invasiva e invasiva, foram plaqueadas em plástico (controle), colágeno tipo 1 ou Matrigel - membrana basal reconstituída, e incubadas por 3 e 7 dias, para estabelecer o processo invasivo. Nossos resultados mostram uma diminuição das taxas de proliferação e alterações morfológicas quando estas células foram cultivadas na presença de colágeno ou Matrigel. Microscopia eletrônica de transmissão das células T98G, cultivadas por 7 dias em colágeno, evidenciam invaginações de membrana similares ao que foi recentemente descrito como podossomos. Este novo tipo de estrutura é encontrado tipicamente em células que precisam cruzar barreiras teciduais, já que são sítios de degradação da MEC. A presença destas estruturas reforça o caráter invasivo da linhagem T98G. Ensaios de PCR em tempo real revelaram maior expressão de mRNA de RECK nas células A172, quando comparadas às células T98G, nas três condições de cultivo. Interessantemente as células A172 apresentaram maior expressão de RECK no colágeno tipo 1, enquanto a T98G demonstra uma tendência de aumento na expressão de RECK para colágeno tipo 1 e Matrigel. As MMPs são mais expressas e possuem maior atividade nas células T98G, e também são induzidas pelo substrato de colágeno. Estes resultados sugerem: 1) a expressão de RECK é diminuída pelo caráter invasivo apresentado por T98G, 2) o uso de substratos de MEC, como colágeno tipo 1 (A172 e T98G) e Matrigel (T98G), permite modular a expressão de RECK nestas linhagens de glioma. Como foi estabelecida uma correlação positiva entre a expressão de RECK e a taxa de sobrevida de pacientes para vários tipos tumorais, nossos resultados podem contribuir para o esclarecimento dos complexos mecanismos do processo invasivo no modelo de gliomas The invasion of neoplastic cells into healthy brain tissue is a pathologic hallmark of gliomas and contributes to the failure of current therapeutic modalities (surgery, radiation and chemotherapy). Transformed glial cells display the common attributes of the invasion process, including cell adhesion to extracellular matrix (ECM) components, cell locomotion, and the ability to remodel the extracellular space. Matrix metalloproteinases (MMPs) are essential for proper ECM remodeling and invasion. The tumor and metastasis suppressor RECK protein regulates at least three members of the matrix metalloproteinase (MMPs) family, namely: MMP-2, MMP-9 and MMP-14. In order to mimic the in vivo invasion process, A172 and T98G, respectively, non-invasive and invasive human glioma cell lines were plated onto either plastic (control), or collagen type I gel or Matrigel™-basement membrane reconstituted, and incubated for 3 and 7 days, to establish the invasion process. Our results indicate decreased growth rates and morphological alterations regarding the invasive phenotype when these cell lines are cultured onto collagen gel and Matrigel™. Electronic transmission microscopy of T98G cells, cultured for 7 days onto collagen, pointed out membrane invaginations that are similar to what was recently described as podosomes. These new structures are typically found in cells that have to cross tissue boundaries, since they are sites of ECM degradation. The presence of these structures reinforces the invasive behavior of T98G cell line. Real time PCR assays revealed higher RECK mRNA expression in A172 cells, when compared to T98G cells in all three different substrate conditions. Interestingly, A172 cells displayed the upregulation of RECK expression in collagen gel, while in T98G high RECK expression was observed both in collagen and in Matrigel™. MMPs appear more expressed and more active for T98G cells, and are also upregulated by collagen. These results suggest that: 1) RECK expression is downregulated in the invasion process displayed by T98G cells, 2) coating of the substrate with ECM elements, such as collagen in the case of A172, and Matrigel™ and Collagen for T98G cells, can modulate RECK expression in glioma cell lines. Since positive correlation between RECK expression and survival of patients has been noted in several types of tumors, our preliminary results can contribute to elucidate the complex mechanisms of the glioma invasiveness process.

Atualmente existe uma forte tendência mundial para o desenvolvimento de testes in vitro que supram o uso de animais em ensaios de avaliação de segurança. Em 1986 a Diretiva Europeia 86/609/EEC induziu a discussão mundial de redução de testes em animais e a 7ª emenda desta Diretiva, publicada em 2004, determinou a transferência dos ensaios para a avaliação de risco para sistema in vitro. Diferentes abordagens foram desenvolvidas para suprir essa necessidade como, por exemplo, a geração de equivalentes de epiderme, que foi considerada a mais promissora e extensamente investigada. Dois modelos de equivalentes de epiderme (EpiDermTM-MATTEK e EPISKINTM LORÉAL) comercialmente disponíveis foram validados pelo ECVAM (Centro Europeu para a Validação de Métodos Alternativos) e outros modelos de epiderme reconstruída estão sendo desenvolvidos em diferentes laboratórios de pesquisa do mundo. Esses dois modelos são recomendados no Guia 431 (Organização para cooperação e desenvolvimento econômico - OECD) que avalia in vitro a corrosão da pele. No Brasil esta prática ainda é inexistente, porém deverá ser implementada rapidamente para que atenda aos conceitos humanitários internacionais. Sendo assim, este projeto teve como objetivo criar, dentro dos princípios do Guia 431, um equivalente de epiderme e posterior otimização deste processo através da incorporação de uma membrana biopolimérica (PET e colágeno I) como suporte celular. O modelo de epiderme humana reconstruída consiste de um tecido epidérmico diferenciado tridimensionalmente a partir de queratinócitos humanos normais cultivados em um meio quimicamente definido na interface ar-líquido. A validação deste modelo para ensaios de avaliação do potencial corrosivo de substâncias foi realizada seguindo os princípios do Guia 431 da OECD, sendo que quatro substâncias da lista indicada pelo Guia foram testadas: duas corrosivas (Ácido Lático e Ácido Octanóico) e duas não corrosivas (2-bromo Etilbenzeno e Benzilacetona), assim como controles positivo (Acido acético) e negativo (NaCl 0,9%). Os resultados de viabilidade celular após 3 minutos e 1 hora de exposição às substâncias teste demostraram que o nosso modelo permite distinguir as substâncias entre corrosivas e não corrosivas, no entanto não permite a subcategorização das corrosivas em 1A ou 1B/C. Assim, nós demonstramos o potencial do nosso modelo como um método de teste relevante e confiável que pode ser usado para pesquisa e investigação de risco químico. Visando a otimização do modelo, testamos a incorporação de uma membrana biopolimérica eletrofiada de PET e colágeno tipo I como suporte para crescimento celular em substituição ao transwell. A análise histológica das membranas demonstrou que as mesmas são biocompativeis, permitindo adequada adesão e proliferação dos queratinócitos e dos fibroblastos, no entanto, não se observou uma diferenciação adequada com a formação de uma epiderme semelhante à humana (estratos basal, espinhoso, granuloso e córneo). Além disso, a falta de uniformidade e reprodutibilidade nos resultados indicam que é necessário uma otimização dos processos de fabricação das membranas e geração do equivalente epidérmico. Em perspectiva, este equivalente epidérmico poderá ser utilizado para pesquisa e avaliação de risco de fármacos, medicamentos, cosméticos, praguicidas, etc., contribuindo para a geração de métodos alternativos aos testes em animais e inserindo o Brasil na tendência internacional do desenvolvimento de testes para avaliação do risco. Currently there is a strong global trend towards the development of in vitro tests that fulfill the use of animals in safety evaluation tests. In 1986, the European Directive 86/609/EEC led to the discussion of global reduction of animal testing and the 7th amendment of this Policy, published in 2004, ordered the transfer of the tests for risk assessment for in vitro system. Different approaches have been taking into account to achieve this goal, for example, the development of epidermal equivalents models, which have been considered the most promising and widely studied model. ECVAM (European Centre for the Validation of Alternative Methods) has validated two models of reconstructed epidermis (EpiDermTM - Mattek and EPISKINTM - L\'Oréal) that are commercially available and other models are being developed in different research laboratories in the world. These two models are recommended in Guide 431 (Organisation for Economic Cooperation and Development - OECD) for in vitro evaluation of skin corrosion. In Brazil, this practice is still lacking, but should be implemented quickly so that meets international humanitarian concepts. Therefore, this project aimed the creation, within the principles of Guide 431, of an epidermal equivalent and the optimization of this process through the incorporation of a biopolymer membrane (PET and collagen I) as cellular support. The reconstructed human epidermis (RHE) consists of a differentiated three-dimensional epidermal tissue reconstructed from normal human keratinocytes in a chemically defined medium and air liquid interface growth. The validation of this model for the assessment of skin corrosion potential of substances was performed following the principles of the OECD Guide 431, hereupon four substances from the list indicated by the Guide were tested: two corrosive (lactic acid and octanoic acid) and two non-corrosive (2-phenylethyl bromide and benzylacetone) and positive (acetic acid) and negative (NaCl 0.9%) controls. The cell viability results after 3 minutes and 1 hour of exposure to the test substances indicates that our model is capable of distinguishing the substances among corrosive and non-corrosive. Although they do not, allow the sub-categorization of corrosive in 1A or 1B/C. Thus, we have demonstrated that our model is a potentially relevant and a reliable test method that could be used in research and specifically for the investigation of chemical hazard. To optimize the model, we tested the incorporation of a biopolymer membrane composed of PET and type I collagen as support for cell growth, as a replacement of the transwell. Histological examination of membranes has shown that they are biocompatible, enabling adequate adhesion and proliferation of keratinocytes and fibroblasts. However, there was no adequate differentiation with the formation of a similar human epidermis (stratum basale, spinosum, granulosum and corneum). Moreover, the lack of uniformity and reproducibility of the results indicates that it is still necessary to optimize the membranes production process and the generation of the epidermal equivalent using the membranes as support. In perspective, the developed epidermal equivalent may be used for research and risk assessment of pharmaceuticals, medicines, cosmetics, chemicals, pesticides, among others. This directly contributes to the development of alternative methods able to substitute animal testing in Brazil and follows the international trend of development tests for risk assessment.

O carbaril (metilcarbamato de naftila), um inseticida de amplo espectro, foi recentemente associado ao desenvolvimento de melanoma cutâneo em estudo epidemiológico de coorte com trabalhadores agrícolas norte-americanos, expostos também à radiação solar, o principal fator etiológico para o desenvolvimento de tumores cutâneos. Apesar de abrangente e bem planejado, aquele estudo epidemiológico não é suficiente para caracterizar a contribuição direta do inseticida e da radiação solar na melanomagênese. Diversos estudos têm explorado o efeito sinérgico de determinadas substâncias químicas à radiação UV, potencializando seus efeitos deletérios sobre a pele, e possivelmente contribuindo para o desenvolvimento de tumores. A hipótese deste trabalho é de que a exposição ao carbaril associada à radiação solar possa estimular a transformação de melanócitos. Esse estudo visou caracterizar melanócitos humanos após exposição individual ou combinada ao carbaril (100uM) e à radiação solar (375 mJ/ cm2). Em ensaio de microarray, o carbaril, mas não a radiação solar, induziu uma importante resposta a estresse oxidativo, evidenciada pelo aumento da expressão de genes antioxidantes, como o Hemeoxigenase-1 (HMOX1), e pela diminuição da expressão do gene MiTF, regulador da atividade melanocítica; os resultados foram confirmados por qRT-PCR. Além disso, tanto o carbaril quanto a radiação solar induziram respostas que sugerem dano ao DNA e alteração de ciclo celular. A expressão dos genes nestas categorias, como p21 e BRCA1/2, foi notavelmente mais intensa no grupo de tratamento combinado e de fato, ensaios por citometria de fluxo demonstraram parada de ciclo celular na fase S, redução do número de células em apoptose e indução mais rápida de lesões do tipo CPD neste grupo experimental. Nossos dados sugerem que o carbaril é genotóxico para melanócitos humanos, especialmente quando associado à radiação solar Carbaryl (1-naphthyl-methylcarbamate), a broad spectrum insecticide, has recently been associated with the development of cutaneous melanoma in an epidemiological cohort study with U.S. farm workers also exposed to ultraviolet radiation, which is known to be the main etiologic factor for skin carcinogenesis. Although comprehensive and well designed, the epidemiological study is not sufficient to characterize the direct contribution of the insecticide and solar radiation in melanomagenesis. Several studies have explored the synergistic effect of certain chemicals with UV radiation, increasing its deleterious effects on the skin, possibly contributing to tumor development. We hypothesized that Carbaryl exposure associated with UV solar radiation may induce melanocyte transformation. This study aims to characterize human melanocytes after individual or combined exposure to Carbaryl (100uM) and solar radiation (375 mJ/ cm2). In a microarray analysis, Carbaryl, but not solar radiation, induced an important oxidative stress response, evidenced by the upregulation of antioxidant genes, such as Hemeoxygenase-1 (HMOX1), and downregulation of MiTF, the main regulator of melanocytic activity; results were confirmed by qRT-PCR. Moreover, both Carbaryl and solar UV induced a gene response that suggests DNA damage and cell cycle alteration. The expression of genes in these categories, such as p21 and BRCA1/2, was notably more intense in the combined treatment group in an additive manner and in fact, flow cytometry assays demonstrated cell cycle arrest in S phase, reduced apoptosis induction and faster induction of CPD lesions in this experimental group. Our data suggests that carbaryl is genotoxic to human melanocytes, especially when associated with solar radiation

Os melanomas são o tipo mais mortal de câncer de pele, apesar da baixa incidência, 80% das mortes de câncer de pele são devem-se ao melanoma metastático. Novas abordagens farmacológicas e a busca por novos compostos para a terapêutica do melanoma, em aplicações isolados ou em combinação com outros fármacos é imprescindível. Esta busca ocorre principalmente no campo das terapias de alvos específicos, devido à aquisição de resistência tumoral e recidiva. O resveratrol (RES) é um polifenol com atividade anti-oxidante, e seu efeito anti-tumoral foi mostrado pela indução de morte celular, porém o seu estudo não foi aprofundado pela inviabilidade do uso de altas doses in vitro para observação de efeitos celulares. Outro composto, o 2-methoxiestradiol (2ME) é um metabólito do estrógeno cujo efeitos anti-câncer já foi demonstrado em melanoma, porém sem elucidação das vias de sinalização envolvidas. O efeito em células com resistência adquirida também nunca foram testados. Neste estudo ampliamos o painel de linhagens celulares de melanoma humano, e demonstramos que o 2ME induz morte celular, inibe a proliferação destas células sendo que esta inibição está associada a indução de senescência. Pela primeira vez foi observada a inibição de proliferação pelo 2ME em células com a mutação BRAF V600E resistentes ao vemurafenibe (inibidor de BRAF) e duplo resistentes ao vemurafenibe e trametinibe (inibidor de MEK). A inibição de proliferação foi acompanhada pela modulação de p21Cip1, Ciclina B1, pRb, proteínas envolvidas na regulação do ciclo celular. A exposição prolongada ao 2ME inibiu a formação de colônias em todas as linhagens de melanoma (não resistentes e resistentes), mas não teve o mesmo efeito em fibroblastos primários, mostrando efeito seletivo. Em modelo tridimensional de esferóides, foi observado que as linhagens resistentes (Sk-Mel-28R) e duplo resistentes (Sk- Mel-28RT) são mais invasivas que a parental (Sk-Mel-28). Neste modelo, o 2ME foi capaz de inibir a invasão e viabilidade destas células. No modelo de pele reconstituída, na ausência de tratamento, observa-se invasão das células de melanoma pela derme, porém este fenômeno é diminuído quando as peles são tratadas com 2ME. Estes resultados demonstram que o 2ME é um efetivo agente anti-melanoma, independente de sua resistência. Melanomas are the deadliest type of skin cancer, and in spite of the low incidence, 80% of the skin cancer associated death cases are due to metastatic melanoma. New pharmacological approaches and the search of new compounds for melanoma therapeutics, for monotherapy or combination therapy, are essential. This search occurs mainly in the targeted therapy field because of the melanoma acquisition of resistance to the current treatments. Resveratrol (RES) is a polyphenol with anti-oxidant activity, and its anti-tumor effect has been shown through the induction of cell death. However, the study of this compound has been discontinued in this work due to the impossibility of using high doses in vitro for the observation of cellular effects. Another compound, 2-methoxyestradiol (2ME) is a metabolite from estrogen, and its anti-cancer effects has already been shown in melanoma, but with no elucidation of the signaling pathways involved. Furthermore, the effects in cells with acquired resistance have never been shown. In this study we used a broader panel of human melanoma cell lines and demonstrated that 2ME induces cell death, inhibits proliferation of these cells, and this inhibition is associated with the induction of cell senescence. The inhibition of proliferation caused by 2ME was observed for the first time in BRAF V600E cells that are resistant to Vemurafenib (BRAF inhibitor) and double resistant to Vemurafenib and Trametinib (MEK inhibitor). The proliferation inhibition was related to the modulation of p21Cip1,Cyclin B1 and pRb, which are proteins involved in cell cycle regulation. Long exposure to 2ME in colony formation assay showed the inhibition of colony in all melanoma cell lines (regardless of resistance and mutational status), but not in primary fibroblasts, showing selective effect. In three-dimensional spheroid model, it was observed that the resistant (Sk-Mel- 28R) and double-resistant (Sk-Mel-28RT) cell lines were more invasive than the parental cell line (Sk-Mel-28). In this model, 2ME was able to inhibit cell invasion and cell viability. In the skin reconstruct model, in the absence of treatment, melanoma cell invasion can be observed in the dermis layer. However, after the treatment with 2ME these cell invasion foci are inhibited. Altogether, these effects demonstrate that 2ME is an effective anti-melanoma agent, regardless of resistance.

A cicatrização deficiente é uma complicação do Diabetes Mellitus (DM), associada ao estresse oxidativo promovido pela hiperglicemia crônica. O objetivo deste estudo foi avaliar os efeitos da administração sistêmica (por via oral, vitaminas E+C) e tópica (utilizando nanopartículas lipídicas sólidas NLSs, vitamina E) de vitaminas antioxidantes na cicatrização de feridas cutâneas em camundongos diabéticos. Uma ferida dorsal foi realizada e os animais tratados com antioxidantes por via oral ou topicamente, diariamente. Os resultados mostraram um atraso na cicatrização das feridas nos diabéticos, menor intensidade da reação inflamatória no 3º dia. No 14º dia, observou-se persistência da reação inflamatória, atraso na reepitelização e menor conteúdo de colágenos fibrilares. O tratamento com vitaminas antioxidante, tanto sistêmico quanto tópico, foi efetivo na aceleração da cicatrização das feridas, por meio da modulação do estresse oxidativo e da reação inflamatória. Concluímos que o tratamento com vitaminas antioxidantes favorece a cicatrização cutânea em diabéticos. Deficient cutaneous wound healing is a common complication of diabetes mellitus (DM), is associated with the oxidative stress caused by chronic hyperglycemia. The aim of this study was to evaluate the effects of systemic administration (oral, vitamins E + C) and topical (solid lipid nanoparticles SLN, vitamin E) of antioxidant vitamins in skin wound healing in diabetic mice. After 30 days, a dorsal wound was performed and the mice were treated orally with antioxidants or topically, daily. The results showed a delay in wound healing in diabetic animals, less severe inflammatory reactions on the 3rd and increased oxidative stress. On 14th there was a persistence of the inflammatory reactions, delayed reepithelization and lower content of fibrillar collagens in diabetic animals. Treatment with antioxidant vitamins, both systemic and topical, was completely effective in accelerating wound healing by modulating the oxidative stress and inflammatory reactions. We concluded that treatment with antioxidant vitamins E and C favors cutaneous wound healing in diabetics.

A terapia fotodinâmica (TFD) é uma modalidade clínica para tratar uma variedade de neoplasias, doenças de pele e representa um promissor tratamento estético. O presente trabalho avaliou os aspectos histopatológicos e moleculares do tratamento pela TFD mediada por azul de metileno (TFD-AM) no modelo experimental in vivo do Carcinoma Espinocelular Cutâneo (CEC) e na pele sadia de camundongos Swiss. O protocolo da TFD foi de uma única sessão, com aplicação da solução de AM a 1% seguido por irradiação com laser diodo na dose total de 24 J/cm2 nos tecidos tumorais e sadios. Os animais foram sacrificados em dois períodos, 24 horas e 15 dias após TFD. Alterações morfológicas foram pouco marcantes nos tecidos tumorais tratados, entretanto, foram mais pronunciadas nos tecidos sadios. Podemos concluir que os efeitos de uma única sessão da TFD mediada pelo AM na dose aplicada não conferiu melhora no tratamento do CEC. Estes resultados motivam novos estudos com ajustes no protocolo para melhorar a eficácia desta terapia. Photodynamic therapy (PDT) is a clinical method for treating a variety of tumors, skin disorders and represents a promising cosmetic treatment. This study evaluated the histopathological and molecular aspects of the treatment by PDT mediated by methylene blue (PDT-MB) in vivo experimental model of cutaneous squamous cell carcinoma (SCC) and in healthy skin of swiss mice. The PDT protocol was a single session with the application of MB 1% solution followed by irradiation with diode laser at a total dose of 24 J/cm2 in tumor and healthy tissue. The animals were sacrificed at two periods, 24 hours and 15 days after PDT. Morphological changes were less marked in the tumor tissues treated, however, were more pronounced in healthy tissues treated. We can conclude that the effects of a single session of PDT mediated by MB in applied dose conferred no improvement in the treatment of SCC. These results motivate further studies with adjustments in the protocol to improve the effectiveness of this therapy.

As indolinonas são moléculas farmacologicamente importantes, conhecidas por sua atividade inibidora de proteínas quinases, exibindo efeitos promissores no tratamento do câncer e outras doenças. Apesar das diferentes abordagens para a terapia e prevenção do câncer, esta doença continua a ser uma das principais causas de morte no mundo. Seus tratamentos atuais apresentam efeitos indesejados, fazendo com que novos compostos e terapias sejam necessários. A fim de contribuir para a solução deste problema, o presente estudo visou avaliar o potencial citotóxico, antiproliferativo e genotóxico de uma nova indolin-3-ona, assim como seu efeito na expressão de genes relacionados ao câncer. O composto apresentou efeito citotóxico sobre eritrócitos humanos apenas a partir da concentração de 32 µg/mL nos tempos de 24, 48, 72 e 96 horas de tratamento. Foi observado que os efeitos citotóxicos e antiproliferativo da indolin-3-ona sobre a linhagem celular HepG2, mensurados pelo ensaio do MTT, foram dose-tempo-dependentes, se intensificando de acordo com a variável em questão. O ensaio do cometa mostrou que a indolin-3-ona possui atividade genotóxica nas concentrações de 16 e 24 µg/mL, contudo, este ensaio detecta danos primários ao DNA que ainda podem ser reparados pelo sistema de reparo de DNA da célula. A partir dos ensaios de RT-qPCR foi possível observar que a indolin-3-ona modula a expressão de genes envolvidos no processo de carcinogênese, reprimindo a expressão gênica, principalmente de proto-oncogenes, também de forma dose-tempo-dependente. Estes resultados apontam para a indicação desta indolinona como uma nova molécula com potencial utilização na terapia do câncer. The indolinones are pharmacologically important molecules known for their inhibitory activity of protein kinases, showing promising effects in treatment of cancer and other diseases. Despite the different approaches for therapy and prevention of cancer, it remains a major cause of death worldwide. Its current treatments have unwanted effects, making new compounds and therapies needed. In order to solve this problem, the present study aimed to evaluate the cytotoxic, antiproliferative and genotoxic potential of a new indolin-3-one and its effect on expression of genes related to cancer. The compound showed cytotoxic effect on human erythrocytes only at concentrations above 32 µg/mL at times of 24, 48, 72 and 96 hours of treatment. The indolinone cytotoxic and antiproliferative effects on HepG2 cells, measured by the MTT assay, were dose-time-dependent, intensifying according to the variable in question. The comet assay revealed that indolinone has genotoxic activity at concentrations of 16 and 24 µg/mL, however, this assay detects only primary DNA damages, which can still be repaired by the DNA repair system of the cell. In RT-qPCR assays we could observe that indolinone modulates the expression of genes involved in the carcinogenesis process, suppressing the gene expression, specially of proto-oncogenes, also in a dose-time-dependent manner. These results point to the indication of this indolinone as a new molecule with potential use in cancer therapy.

Existe um grande interesse das indústrias farmacêuticas em encontrar um tratamento adequado para os pacientes com câncer que apresentaram resistência aos medicamentos comumente utilizados para o combate dessa neoplasia. Sendo assim, se faz necessário a busca de novos marcadores genéticos que estejam associados à predisposição do paciente a reagir ao efeito da droga de maneira diferente do esperado. Para isso, vários estudos vêm sendo desenvolvidos, baseados nos medicamentos anticancerígenos. Entre esses medicamentos podemos destacar os agentes quimioterápicos a base de platina, como a carboplatina, cisplatina, oxaliplatina e a nedaplatina, que agem eliminando as células cancerosas através da formação de adutos nas bases de purina do DNA nuclear e a rebecamicina, um antitumoral que possui a capacidade de inibir a ação catalítica das topoisomerases I, ainda em teste clínico. Desse modo, este estudo teve a perspectiva de encontrar mutantes de leveduras resistentes a carboplatina e análogos de rebecamicina. Como resultado, identificamos 19 genes associados à resistência a carboplatina, sendo que 6 deles possuem genes ortólogos em humanos associados principalmente ao mecanismo de reparo de DNA e ao transporte celular. Foram também identificadas 9 linhagens que estavam associadas a sensibilidade a carboplatina sendo que 3 delas possuem deleções em genes com ortólogos humanos, que estão associados ao transporte endossomal, ao ribossomo e ao bloqueio do ciclo celular na fase G1. Na busca por um novo composto antitumoral derivado de rebecamicina, identificamos Reb C como um análogo promissor, já que apresentou 9 genes associados a resistência, dos quais 2 deles possuem ortólogos humanos relacionados ao ribossomo e ao proteassoma There is a strong interest from pharmaceutical companies in finding an appropriate treatment for patients with cancer who had resistance to drugs commonly used to fight this disease. Therefore, it is necessary to search for new genetic markers that are associated with the patient\'s predisposition to react to the drug differently than expected. Aiming this, several studies have been developed based on anticancer drugs. Among these drugs can be highlighted the chemotherapeutic agents based on platinum, such as carboplatin, cisplatin, oxaliplatin and nedaplatin, which act by eliminating cancer cells through the formation of adducts with the purine bases of nuclear DNA and the rebeccamycin, an antitumor that has the ability to inhibit the catalytic action of topoisomerase I, still in clinical trial. This study aimed to find yeast resistant mutants to carboplatin and rebeccamycin analogs. As a result, we identified 19 genes associated with resistance to carboplatin, 6 of them presenting orthologous genes in humans, which are mainly associated to the mechanism of DNA repair and to cellular transport. 9 strains were also identified which are associated with carboplatin sensitivity, 3 possessing their orthologous in humans associated with endosomal transport, ribosome and cell cycle arrest in the G1 phase. In a search for a new antitumor compound derived from rebeccamycin, we identified the Reb C analog as promising, since it featured nine genes associated with resistance to it, among 2 of them have human counterparts related to the ribosome and the proteasome.

Considerando o melanoma a forma mais agressivas de câncer de pele e mais resistente aos tratamentos convencionais, o presente estudo teve como objetivo avaliar a atividade de um novo complexo de platina (II) com derivado de hidantoína (CX42) em células de melanoma humano e murino. Foram utilizados também para comparação células da pele (fibroblastos, queratinócitos e melanócitos) e os compostos cisplatina (CIS) e complexo de hidantoína isolado (NN10). Ensaios de viabilidade, ciclo e morte celular foram realizados. Investigou-se também a atividade da enzima tirosinase, principal enzima que regula a síntese de melanina durante o processo conhecido como melanogênese. Como resultados, obteve-se a diminuição da viabilidade celular e parada de ciclo na fase G0/G1 nas células de melanoma, principalmente na linhagem murina B16F10, frente ao composto CX42. Em células B16F10, foi possível observar estímulo na melanogênese, com aumento da atividade da enzima tirosinase. O CX42 apresentou uma atividade com efeito citostático nas células de melanoma, não sendo observado efeito citotóxico nas células da pele. Ainda, com a prévia estimulação da melanogênese, o CX42 apresentou-se mais efetivo, aumentando a morte celular em 40% por apoptose. Portanto, conclui-se que o CX42 diminuiu a viabilidade celular e seu efeito foi mais intenso quando as células apresentavam-se pigmentadas, demonstrando indução da tirosinase e da morte celular, atributo importante para potenciais novos fármacos anti-melanoma. Since melanoma is one of the most aggressive forms of skin cancer and more resistant to conventional treatments, the present study aimed to evaluate the activity of a new platinum complex (II) with hydantoin derivative (CX42) in melanoma cells human and murine. Skin cells (fibroblasts, keratinocytes and melanocytes) were used as control and the compounds cisplatin (CIS) and hydantoin compound alone (NN10) were also evaluated. Viability assays, cell cycle analysis and cell death characterization were performed. Likewise, the activity of tyrosinase, the key enzyme that regulates melanin synthesis during the process known as melanogenesis, was also investigated. The results demonstrated a reduction in cell viability and cell cycle arrest in G0/G1 phase of murine melanoma B16F10 treated with CX42. In B16F10 cells, it was also observed melanogenesis stimulation with increased activity of tyrosinase. The CX42 showed a selective cytostatic effect on melanoma cells, however no toxic effects were observed in skin cells. In addition, with prior stimulation of melanogenesis, the CX42 demonstrated to be more effective, increasing apoptosis cell death in 40%. Therefore, the results demonstrated that CX42 decreased cell viability and the intensity of its effect is associated with cell pigmentation, demonstrating an association between tyrosinase high activity and induction of cell death which is an important attribute for new potential anti - tumoral drugs.

O câncer cervical é o segundo mais comum em mulheres em países em desenvolvimento, sendo causado por infecção persistente por Papilomavírus Humano (HPV). Quando esta persistência ocorre, entre outros fatores, está relacionada a mecanismos de evasão do sistema imune apresentados pelo vírus. A frequência de macrófagos aumenta com a progressão da lesão cervical e há aumento de células mielóides no baço de camundongos com tumor. Este trabalho tem como objetivo observar os efeitos sistêmicos de tumores associados ao HPV sobre a proliferação e recrutamento de células do sistema imune e identificar fatores que tenham papel nesses mecanismos. Utilizando modelos de tumor in vivo, observou-se que tumores associados ao HPV recrutam mais células para o tumor e induzem maior proliferação celular. Também avaliamos o perfil de expressão de citocinas nas linhagens tumorais e o perfil geral de expressão de proteínas através de eletroforese 2D. Com isto, demonstramos que linhagens tumorais positivas para HPV apresentam maior expressão de IL-6, IL-8, CXCL1, sICAM e Serpina E1. Cervical cancer is the second most common type of cancer in women in developing countries. Its main etiologic factor is persistent infection with high risk human papillomavirus (HPV). This persistence occurs only in some cases and, among other factors, is related to mechanisms of immune evasion displayed by the virus. There is an increase in the frequency of macrophages proportional to cervical intraepithelial lesion grade and an increase of myeloid cells in the spleen of tumor bearing mice. This work aims to observe the systemic effects of HPV associated tumors on the proliferation and recruitment of immune cells, and identify factors that have a role in these mechanisms. Using in vivo tumor models, we found that HPV positive tumors recruit a higher percentage of cells and induce cellular proliferation. We also studied cytokine expression profiles of tumor cell lines, and performed proteomic assay with tumor cells transduced with HPV oncogenes. Our data shows that HPV associated tumor cell lines display higher expression of IL-6, IL-8, CXCL1, sICAM and Serpin E1.

O glioblastoma multiforme (GBM) é um dos gliomas mais comuns, classificado como um glioma de grau IV (Organização Mundial da Saúde - OMS) e notoriamente difícil de ser tratado. O tratamento recomendado consiste na ressecção cirúrgica seguida de radio e quimioterapia, e a sobrevida média dos pacientes é de apenas 12 meses após o diagnóstico. Portanto, novas terapias que focam a redução do volume do tumor e o aumento da morte das células tumorais são urgentemente necessárias. Estudos pré-clínicos sugerem que a inibição de COX-1 e 2 com Anti-inflamatórios não esteroidais (AINEs), como o Ibuprofeno (IBP), inibiram significantemente a proliferação e a migração celular em diferentes tumores. Assim, o presente estudo teve como objetivo avaliar in vitro o efeito da inibição de COX-1 e COX-2 através do tratamento com IBP, e também com inibidores específicos como SC-560 e NS-398 respectivamente, na migração e na proliferação da linhagem celular de glioma humano U251-MG. O presente estudo demonstrou através de diversos ensaios que a inibição de COX-1 e COX-2 através do IBP, do SC-560 e do NS-398 inibiu significativamente a migração e a proliferação celular da linhagem celular U251-MG. Dessa maneira, concluímos que a PGE2 está envolvida na migração e proliferação celular das células desta linhagem celular. Glioblastoma Multiforme (GBM) is the most common glioma, classified as grade IV (World Health Organization WHO) and notoriously difficult to treat. The recommended treatment consists of surgery followed by radiotherapy and chemotherapy, and the median survival is ten to twelve months. Preclinical studies suggest that inhibiton of cyclooxygenase-2 by treatment with non-steroidal anti-inflammatory drugs, such as ibuprofen, significantly blocked the proliferation of different tumors including gliomas. Thus this study aimed to evaluate the migration of glioma cell line U251-MG after inhibition of cyclooxygenases 1 and 2 by treatment with ibuprofen (IBP), and specifics inhibitors such as SC-560 for COX-1 and NS-398 for COX-2. The present study demonstrated by various assays where inhibition of COX-1 and COX-2 by IBP, SC-560 and NS-398, significantly inhibited migration and cell proliferation of U251-MG cell line. Thus, we conclude that PGE2 is involved in this cell line migration and cell proliferation.

Na etapa inicial do desenvolvimento de novos fármacos, a avaliação do metabolismo e da toxicidade é fundamental para definir seu potencial emprego como candidato a fármaco. Nestes estudos, diversos modelos in vitro são empregados, dentre eles linhagens de hepatoma humano. Entretanto, uma grande limitação ao uso deste modelo in vitro é a baixa expressão das enzimas do sistema citocromo P450. O carotenóide bixina, componente majoritário do anato (urucum), apresentou em estudos in vivo, a capacidade de induzir algumas isoformas do sistema citocromo P450, com a vantagem de apresentar baixa toxicidade. Neste trabalho, a fração lipossolúvel do anato (bixina) e hidrossolúvel (norbixina) foram avaliadas como indutores do sistema citocromo P450 em linhagens de hepatoma humano. Ensaios de MTT, empregando as linhagens HepG2, C3A e SK-HEP-1 indicaram que bixina e norbixina em concentrações abaixo de 0,22 mM são seguras quanto à citotoxicidade. A expressão dos genes CYP 1A1, 1A2, 2C9, 2B6, 2E1 e 3A4 foi avaliada, através de ensaios de RT-PCR em tempo real, em linhagens de hepatoma humano submetidas a tratamento com os compostos bixina e norbixina. Os resultados mostraram que células HepG2 e C3A tratadas com bixina nas concentrações de 0,05 e 0,1 mM, por períodos de 24 e 48 horas, apresentaram aumento de expressão da CYP 1A1 e CYP 1A2. Porém, a exposição de células HepG2 e C3A ao composto norbixina não resultou em aumento de expressão das isoformas avaliadas neste estudo. Os resultados deste trabalho indicaram o potencial emprego de bixina como agente indutor das CYPs 1A1 e 1A2, em linhagens de hepatoma humano utilizadas como modelo in vitro, para estudo de compostos cuja metabolização envolva uma destas vias, entretanto, estudos adicionais são fundamentais, a fim de avaliar a ação deste composto sobre outras isoformas do sistema citocromo P-450, bem como outros sistemas enzimáticos. In the early development stage of the new drugs, the pharmacological and toxicological properties are critical to define the potential use of the candidate drug. During this stage, several in vitro models systems are employed, including human hepatoma cell lines. However, the main limitation of the use of cell lines as in vitro model is the low expression level of cytochrome P450 enzymes. A carotenoid knowed as bixin, the main pigment in the annatto (urucum), it has been reported to induce some isoforms of cytochrome P450 in rats, with the advantage of its low toxicity. In this work, the oil-soluble (bixin) and aqueous soluble extracts (norbixin) were evaluated as inducers of the cytochrome P450 system in human hepatoma cell lines (HepG2, C3A, SK-HEP-1). The results of MTT assays showed that bixin concentrations below 0.22 mM were not cytotoxic in HepG2, C3A and SK-HEP-1 cell lines. Expression changes in CYP 1A1, 1A2, 2C9, 2B6, 2E1 and 3A4 were evaluated, by real time RT-PCR and the results showed that the exposition to 0,05 mM and 0,1 mM bixin, for 24 and 48 hours of treatment, lead to an increase in CYP 1A1 and CYP 1A2 expression level. By contrast, the cytochrome P450 isoforms were not affected by the exposition to norbixin. In conclusion, this work indicated the potential use of bixin induced hepatoma cell lines as in vitro model for studies of biotransformation and toxicity of drugs involving CYP 1A, however, further studies are necessary to evaluate the effect of bixin on the other cytochrome P450 isoforms as well as other enzymatic systems.

The identification of high-risk (HR) Human Papillomavirus (HPV) types offers the possibility of improving the efficiency of cervical screening programs. Therefore, there is a continuing need for the identification of factors that modulate the risk of disease progression, and which might serve as predictive biomarkers. The aim of the present study was determine by polymerase chain reaction (PCR) the distribution of HPV genotypes and the E2 gene disruption of the HPV16/18 to assess whether viral pure integration is correlated with the degree of cervical abnormalities in South Brazilian women. A total of 137 women were studied between November 2009 and August 2011, cervical samples were collected in the colposcopy clinic at União da Vitória city, south/Brazil. The mean age was 33.60±30.72 years (range of 16-78 years), and those with invasive carcinoma (IC) were older (p < 0.05). The prevalence of total DNA- HPV, HR-HPV and multiple infections in all women was 56.2%, 38.7% and 12.4%, respectively and were higher detected in those with high-grade lesions (HSIL) and invasive carcinomas (IC) (p < 0.05). The most common HPV detected types was HPV-16 (17.5%; p < 0.05). The low-risk (LR)-HPV types were detected in 29.2%, being HPV-72 the most common (15.3%) and HPV-6 and -11 were not detected. For multiple infections, the most common occurred between HPV-72 and 16 (64.7% the all co-infections and 8.0% of the women). The overall HPV-18 E2 gene disruption was 100.0% and this infection was evenly distributed in atypia (ASC-H), low-grade lesions (LSIL), HSIL and IC (0.7% each). For HPV-16 E2 gene, the overall disruption was 54.2%, and occurred in 50.0% in ASC, 66.7% in LSIL, 53.8% in HSIL and 50% in IC (p < 0.05). Considering the similar results for pure integration of HPV-16 among different degrees of cytological abnormalities since ASC to IC, the use of PCR does not seem to be applicable to distinguish women with greater propensity for progression of the lesions. Os gliomas são o tipo de tumor primário cerebral mais comum em adultos. A sobrevida média dos pacientes é de cerca de um ano para pacientes de glioblastoma multiforme, o maior grau de malignidade. As terapias disponíveis atualmente, que incluem cirurgia, radioterapia e quimioterapia, não têm sido eficientes devido a vários fatores, em particular a capacidade invasiva das células tumorais. RECK (reversion-inducing-cysteine-rich protein with Kazal motifs) é um importante gene supressor de tumor cuja atividade anti-tumoral têm sido associada à sua atividade inibitória sobre algumas MMPs. A perda da função de RECK compromete a integridade tecidual, em parte causada pela atividade aumentada das MMPs. A linhagem celular T98G, derivada de glioblastoma multiforme (GBM), apresenta grande capacidade invasiva e níveis elevados de MMP-2 e -9. A fim de avaliar o efeito de RECK na contenção da invasão no modelo de glioma humano, foi feita a superexpressão de RECK nesta linhagem. A expressão de RECK, MMP-2, MMP-9 a MT1-MMP foram avaliadas por qPCR e por western blotting nas células T98G/RECK+ (células T98G superexpressando RECK após transfecção estável utilizando o vetor pCXN2). O potencial invasivo e migratório das células T98G/RECK+foi inibido, verificado por ensaio transwell. Foram observadas nas células T98G/RECK+ alterações importantes no arranjo do citoesqueleto, mas não nas células controle. Arranjos de actina na forma de \"stress fibers\" presentes no clone positivo podem ser responsáveis pela alteração observada na migração. A distribuição de FAK foi avaliada por imunocitoquímica e sua expressão por Western blot, mostrando que RECK só altera a distribuição desta proteína no citoesqueleto celular. Quanto ao potencial de inibição de MMPs, observou-se uma diminuição significativa dos níveis gênicos de MMP-9, mas não em termos protéicos ou de atividade de MMPs. Assim, este trabalho contribui para a discussão do papel de RECK na migração de células do modelo de glioma humano, uma das características responsáveis pela ausência de terapia efetiva deste tipo de tumor. Malignant gliomas are the most common type of primary brain tumors in adults. Patient survival is less than one year in average for glioblastoma, the most malignant glioma. Therapies available today, which include surgery, radiotherapy and chemotherapy, have not been successful due to several factors, specially the invasiveness of the tumor. RECK (reversion-inducing-cysteine-rich protein with Kazal motifs) is an important tumor suppressor gene whose anti-tumoral activity is associated to its anti-MMPs activity. Lack of functional RECK compromises tissue integrity, in part due to elevated MMPs activity. T98G cells, derived from a human multiform glioblastoma (GBM), were described as a highly invasive glioma cell line, which displays high levels of MMPs 2 and 9. In order to evaluate the effect of RECK in restraining glioma invasion, we overexpressed RECK in these invasive cell line derived from GBM. The expression of RECK, MMP-2, MMP-9 and MT1-MMP was evaluated by qPCR and by western blotting in T98G/RECK+ (T98G cells overexpressing RECK cells, where RECK gene was cloned into the pCXN2 vector generating a stable transfection). The invasion and migration capacity of T98G/RECK+ cells was inhibited in transwell assay. Important cytoskeleton modifications were also observed in T98G/RECK+ cells but not on the control cells. Actin arrangements representing stress fibers on the positive clones may be responsible for motility alteration patterns observed. FAK distribution was assessed through imunocytochemical staining, and its expression evaluated by western blot analyses, showing that RECK forced expression changed the distribution pattern but not FAK expression. Concerning MMPs inhibition, a significant inhibition of MMP-9 gene expression was observed in T98G/RECK+, but neither protein levels nor protein activity were affected. Thus, the present study improves the discussion about RECK role in the migration of glioma cells, an important feature for the failure of current therapies for this kind of tumor.

Um dos mais versáteis mecanismos de reparo de DNA é o reparo por excisão de nucleotídeos (nucleotide excision repair- NER). Defeitos genéticos associados a esta via podem gerar diferentes síndromes com deficiência de reparo. Dentre essas, Xeroderma pigmentosum (XP) é a que apresenta maior sensibilidade à luz solar, resultando em um grande aumento na incidência de tumores em regiões expostas da pele e, em alguns casos, degeneração neurológica progressiva e envelhecimento prematuro. Na primeira parte deste projeto é apresentado o uso de adenovírus recombinantes portando genes da via de NER para identificar a deficiência gênica de três pacientes portadores de XP. Na segunda parte do trabalho os estudos de reparo de DNA são estendidos a modelos animais, com deficiências nos mesmos genes carregados pelos vetores adenovirais. A expressão gênica do vetor foi avaliada pela detecção de proteína e por visualização da fluorescência de EGFP na pele dos animais infectados. Em resumo, este trabalho apresenta o uso eficiente de vetores adenovirais portando genes de reparo em ensaios in vitro e in vivo, e descreve duas mutações deletérias no gene XPC de pacientes XP brasileiros, incluindo uma mutação nova. One of the most versatile mechanisms of DNA repair is the nucleotide excision repair (NER). Genetic defects in NER can generate different syndromes. Among these, Xeroderma pigmentosum) presents the highest sensitivity to sunlight, resulting in a large increase in the incidence of skin cancer, especially in areas exposed to the sunlight, and in some cases, progressive neurological degeneration and premature aging. In the first part of this project, adenoviral vectors carrying NER genes were used to identify genetic deficiency of three XP patients. The second part of work was extended to animal models, deficient for the same XP genes carried by adenoviral vectors. The genetic expression of vector was evaluated by detection of protein and EGFP fluorescence visualization in the skin of animals transduced. In summary, this work presents the use of adenovirus, carrying DNA repair genes for in vitro in vivo studies reports two deleterious mutations in Brazilian XP patients, including a new mutation.

O objetivo do presente trabalho foi avaliar a influência da aplicação de bevacizumab (Avastin®) sobre a angiogênese corneal. Para tanto ratos Wistar, machos, com idade entre 8 e 10 semanas, pesando 300 a 350g, foram submetidos a cauterização química com nitrato de prata por 10 segundos. Após realização da lesão, cada grupo de 5 animais foi tratado com injeções subconjuntivais de 0,02 ml de bevacizumab (Avastin ®) pela via subconjuntival no momento da lesão, no 3° e 5° dia e submetidos à eutanásia ao 7° dia após cauterização corneal. A rede vascular neoformada foi quantificada após preenchimento do leito vascular com Tinta da China e análise das imagens em sistema computadorizado (Image Pro-Plus®). Para a segunda etapa do experimento, avaliou-se a atividade das metaloproteinases MMP-2 e MMP-9 utilizando-se do mesmo modelo de angiogênese e dos mesmos tempos de tratamento. Transcorridos 7 dias de lesão corneal os animais foram eutanasiados e suas córneas submetidas a zimografia. Os resultados mostraram haver uma inibição da angiogênese quando se compara o grupo controle aos grupos tratados nos diferentes períodos de tempo. Quando a densidade vascular é comparada entre os tempos de aplicação, não se observa diferença estatisticamente significante. Estes resultados em conjunto indicam que o tempo de aplicação não influencia a inibição da angiogênese e que o bevacizumab foi eficiente na redução da formação de vasos quando se compara a densidade vascular do grupo controle. O modelo experimental produziu considerável aumento das metaloproteinases MMP-2 e MMP-9. Houve uma tendência a diminuição da atividade das MMPs quando bevacizumab foi aplicado no momento da lesão e quando foi aplicado ao 3° dia após cauterização. A atividade das MMPs mostrou-se aumentada quando o tratamento foi realizado no 5° dia. Desta forma pode-se concluir que o bevacizumab foi capaz de inibir a angiogênese corneal, independentemente do período de tratamento e ainda que houve uma tendência a redução da atividade das MMPs carecendo, por enquanto, de mais investigações que possam elucidar a interação deste fármaco com a inibição de sua atividade. The purpose of this study was to evaluate the effects of the use of the subconjunctival injection of bevacizumab (Avastin®) on angiogenesis and metalloproteinases activities in the rat cornea after cauterization. Wistar male rats, aging 8 to 10 weeks, were used. The animals were divided in four groups: control group (GC) that received subconjunctivally 0,02 ml of 0,9% saline solution; group GO that received subconjunctivally 0,02 ml of bevacizumab just after the lesion; group G3 that received bevacizumab at day 3 after lesion and G5 that received bevacizumab at day 5 after lesion. The animals were euthanized at day 7 after lesion. The new formed vessels were quantified after the China Ink perfusion and photographs were obtained and analyzed in computadorized system (Image Pro-Plus®). The activities of metalloproteinases MMP-2 and MMP-9 were evaluated by zimography in a pool of corneas obtained from animals submitted to the same model and treatment schedule. The results showed an inhibition of angiogesenis when the control group were compared with all treated groups. There were no statistic differences when vascular density was compared between the treated groups. These results may indicate that bevacizumab was able to inhibit corneal angiogenesis and that there was no influence of the injection day. This angiogenesis model promoted an increase of matrix metalloproteinases MMP-2 and MMP-9 activity. Zymography also showed a slight reduction in matrix metalloproteinases activity when bevacizumab were administrated at the injury moment and most significant at day 3. At day 5 zymography showed an increase of the activity of these metalloproteinases. Together these results may indicate that the subconjunctival injection of bevacizumab may have a therapeutic potential as an antiangiogenic agent and that matrix metalloproteinases activity decreased, but not at significant levels, demanding more investigations regarding to the interaction of bevacizumab and activity of matrix metalloproteinases.

A ação das metaloproteinases de matriz (MMP-2, MMP-9 e MT1-MMP) é necessária para degradação da membrana basal em carcinomas da cérvice uterina. O objetivo deste trabalho consistiu na avaliação da expressão das metaloproteinases MMP -2, -9 e MT1-MMP, do gene supressor de metástase RECK e do inibidor tecidual de MMPs (TIMP-2) em modelo de células de neoplasia da cérvice-uterina cultivadas em substratos de matriz extracelular. As linhagens celulares de carcinoma de cérvice uterina SiHa, CaSki, ambas HPV 16 positivas, e C33A, HPV negativa, foram cultivadas em gel de colágeno tipo I, Matrigel e plástico. Avaliou-se o crescimento, invasão, expressão gênica, através de ensaios de real-time PCR, e atividade de metaloproteinases, através de ensaios de zimografia. Os resultados demonstraram que estas linhagens de carcinoma cervical quando cultivadas em gel de colágeno tipo I apresentaram uma diminuição no crescimento, morfologia modificada na presença de substrato de matriz extracelular, e que nas linhagens HPV positivas há um aumento da expressão de MMP-2, MT1-MMP e TIMP-2 e da atividade de pró-MMP-2 em relação à linhagem HPV negativa. Observou-se também que, RECK apresentou maior expressão gênica em CaSki associada à atividade de pró-MMP-2. MMP-9 apresentou muito baixa expressão gênica em todas as linhagens e condições estudadas. Quando analisamos as linhagens separadamente, observamos que o Matrigel influenciou a expressão gênica de MMP-2, e que o gel de colágeno tipo I aparece como indutor da atividade de pró-MMP-2 em todas as linhagens. Em conclusão, nossos resultados mostram que a expressão de MMP-2, MT1-MMP e TIMP-2 e que a atividade de pró-MMP-2 estão aumentadas nas células HPV positivas, sugerindo que o HPV está associado com a expressão de MMPs e TIMP-2. Matrix metalloproteinases (MMPs) -2, -9, and MT1-MMP are required for basement membrane degradation in cervical carcinoma. We evaluated the expression and activity of MMPs and their inhibitors RECK and TIMP-2 in three human invasive cervical carcinoma cell lines. Two HPV-16- positive cell lines (SiHa and CaSki), HPV-negative cell line (C33A) were cultured either onto type-I collagen gel, Matrigel or plastic, in order to recreate their three-dimensional growth environment and evaluate the growth and invasion of the cells and expression of these genes using quantitative real-time PCR (Q-PCR). We also analyzed the gelatinolytic activity of MMP-2 and -9 by zymography. We found that the growth curves carcinoma cells are decreased and cells morphology are modified in ECM substrate. HPV-positive cell lines expressed higher levels of MMP-2, MT1-MMP and TIMP-2 than the HPV negative cell line. In addition, MMP-9 was expressed at very low levels in both HPV-negative and HPV-positive cell lines. We also observed that the expression of the RECK gene is higher in CaSki cells, being associated with pro-MMP-2 activity. The Matrigel substrate influence MMP-2 expression for SiHa and CaSki cells. On the other hand, we found that type-I collagen gel, but not Matrigel, can enhance pro-MMP-2 activity in all cell lines. Our results suggest that the presence of HPV is related to increased expression of MMP -2, MT1-MMP and TIMP-2 and pro-MMP-2 activity in HPV-positive cells than in HPV-negative cells.

O efeito anti-aterogênico da HDL foi sugerido ser devido, parcialmente, à ação da paraoxonase (PON) associada à HDL. Três SNPs em PON1 (L55M e Q192R) e PON2 (S311C) têm sido implicados como fatores de risco independentes para doença arterial coronariana (DAC) em alguns, mas não todos os estudos. O efeito destes três polimorfismos do gene PON na DAC e na concentração sérica de lipídios, apolipoproteínas A-I e B (apo A-I e apo B) foi investigado em 221 indivíduos sem vínculos genéticos. Os três polimorfismos genéticos (L55M, Q192R e S311C) do gene PON foram analisados por PCR-RFLP. Não houve diferença entre a distribuição genotípica e a freqüência relativa dos alelos dos polimorfismos L55M, Q192R e S311C do gene PON entre pacientes e controles. Não houve associação entre os polimorfismos da PON e a DAC na população estudada. As freqüências dos haplótipos dos pacientes foram similares às encontradas no grupo controle. O polimorfismo L55M do gene PON1 está associado com variações na concentração sérica de apo A-I dos pacientes. O polimorfismo S311C do gene PON2 não está associado com variações na concentração sérica de lipídios, apo A-I e apo B dos pacientes. The anti-atherogtenic effect of the HDL has been suggested to be due, partially, to the action of the HDL-associated paroxonase(PON). Three SNPs in PON1 (L55M e Q192R) and PON2 (S311C) have been involved as independent risk factors for coronary artery disease (CAD), in some, but not all studies. The effect of these three polymorphisms of the PON gene on CAD and on the levei of lipids, apolipoproteins A-I e B (apo A1 and apo B) was investigated in 221 genetically unrelated individuais. The three gene polymorphismis (L55M, Q192R e S311C) of the PON gene were analyzed by PCR-RFLP. Is no significant difference between distribuition of genotype and allele frequencies of the L55M, Q192R and S311C polymorfisms of the PON gene between patient group and controls. Is no association between polymorphisms of the PON gene and CAD in the population studied. The frequencies of the haplotypes in the patients were similar to those found in the control group. The L55M polymorphism of the PON1 gene is associated with variations of the level of polipoprotein A-I at patients. The S311C polymorphism of the PON2 is not associated with variations of the level of lipids, apo A-I and apo B at patients.
