154. Iconic Sign Comprehension in Older Adults: The Role of Cognitive Impairment and Text Enhancement

Abstract

Sign comprehension is critical for effective driving, responses to warnings, and way-finding. Signs that are poorly comprehended by older people increase accident risk and may compromise independence. This study sought to determine whether iconic sign comprehension suffers in healthy aging and in the presence of cognitive impairment. Additionally, we examined whether the addition of text to iconic signage would increase comprehension in older adults. In Experiment 1, young adults, healthy older adults, and older adults with varying levels of cognitive impairment were asked the meaning of 65 signs used for driving, warnings, and way-finding. Healthy older adults were generally good at sign comprehension but had difficulty with way-finding signs. Older adults with cognitive impairment had poorer sign comprehension overall and particular difficulty with way-finding icons and signs that had icons only. In Experiment 2, healthy older adults were asked the meaning of signs containing icons only, or icons and text. A significant improvement in comprehension was found when text was added. An important implication of this work is that the assessment of sign comprehension needs to involve a broad and heterogeneous sample of older adults reflecting the range of perceptual and cognitive abilities represented in the population.

Differential diagnosis of dysphonia comprehends many different etiologies and many causative agents (neoplasms, inflammations, traumatic injuries) that can occur in a large anatomical space (from the encephalic trunk to the upper mediastinum). It is fundamental to remember that vascular etiologies are responsible for some rare cases of dysphonia. In the radiological database of two urban academic hospitals, from 2012 to 2020, we sought patients who underwent CT or MRI for dysphonia, selecting only the ones with an underlying clinically confirmed vascular etiology. We present three emblematic cases with different vascular etiologies: a ductus arteriosus aneurysm, a left internal carotid artery dilatation, a laryngeal arteriovenous malformation (AVM). Vascular causes of dysphonia are rare, but especially in these cases an accurate and prompt diagnosis is fundamental, in particular considering that the underlying cardiovascular anomaly can often pose a higher risk for the patient than the hoarseness itself. Diagnostic imaging plays a fundamental role in detecting the most common causes of dysphonia but it is very important that radiologists take the vascular causes into account so as not to miss them and to obtain a correct diagnosis.Keywords: dysphonia, thoracic aorta, arteriovenous malformation, carotid artery, patent ductus arteriosus

156. Aging Research across Disciplines: A Student-Mentor Partnership Using the United Nations Principles for Older Persons

Abstract

A grant from the Canadian Institutes of Health Research for training on communication and social interaction in healthy aging was used to support the collaboration of three students and one program mentor from various age-related backgrounds (e.g., vision, hearing, cognition, and social work) to develop a transdisciplinary and interinstitutional research program. The first project focused on integrating the United Nations Principles for Older Persons into a research agenda to gauge awareness of the principles in 100 seniors-related organizations in Ontario, Canada. The questionnaire elicited both quantitative and qualitative data related to the principles, and these data are discussed herein. Questionnaire, United Nations Principles for Older Persons, and cover letter sent to participants are appended.

157. Interdisciplinary Research Education in Communication and Social Interaction among Healthy Older Adults

Abstract

An innovative gerontology education program was developed to advance research on aging that is interdisciplinary and promotes the translation of knowledge from lab to life. The program focuses on communication and social interaction in healthy aging. It brings together faculty mentors, graduate students, and post-doctoral fellows from six different postsecondary institutions in Canada. The program unifies basic laboratory research in hearing, vision, and cognition with applied research in audiology, biomedical engineering, optometry, psychology, speech-language pathology, human factors, and social work. The design, implementation, and evaluation of the first year of the program are described and future directions are suggested.

Abstract

Huntington’s disease (HD) is caused by an expanded CAG trinucleotide repeat in exon 1 of the huntingtin (HTT) gene. We report the design of a series of HTTpre-mRNA splicing modulators that lower huntingtin (HTT) protein, including the toxic mutant huntingtin (mHTT), by promoting insertion of a pseudoexon containing a premature termination codon at the exon 49–50 junction. The resulting transcript undergoes nonsense-mediated decay, leading to a reduction of HTTmRNA transcripts and protein levels. The starting benzamide core was modified to pyrazine amide and further optimized to give a potent, CNS-penetrant, and orally bioavailable HTT-splicing modulator 27. This compound reduced canonical splicing of the HTTRNA exon 49–50 and demonstrated significant HTT-lowering in both human HD stem cells and mouse BACHD models. Compound 27is a structurally diverse HTT-splicing modulator that may help understand the mechanism of adverse effects such as peripheral neuropathy associated with branaplam.

177. Obesity and cardiovascular disease: Risk assessment, physical activity, and management of complications

Abstract

The patient with obesity is at risk of developing cardiovascular disease and risk factors. Obesity negatively impacts prognosis and increases cardiovascular morbidity and mortality. Therefore, a comprehensive risk assessment is needed to define the cardiovascular risk of the patient and, thus, a tailored management and treatment. Chronic and successful management of these patients involves the evaluation of the various therapeutic strategies available (comprehensive lifestyle intervention, weight-loss medications, and bariatric surgery) and the diagnosis and treatment of cardiovascular complications (coronary artery disease, heart failure, and atrial fibrillation). Cardiac rehabilitation in patients with obesity is showing beneficial effect and a positive impact on weight loss, cardiovascular risk factors, mental health, functional capacity, and adherence to lifestyle interventions and pharmacological treatment. Long-term weight loss and maintenance represent a key objective during the management of the patient with obesity to reduce the risk of future adverse events. Multidisciplinary management and interventions are necessary to prevent and reduce overall cardiovascular risk and mortality. The aim of our review is to propose a comprehensive, critical and updated overview regarding risk assessment, physical activity, and the management of cardiovascular complications in patient with obesity.

183. International experience and imitation of location choices: The role of experience interpretation and assessment and its board‐level microfoundations

Abstract

Drawing on the information‐based imitation and information‐processing perspectives, we examine how experience interpretation and assessment—and in particular its board‐level microfoundations—affects the relationship between a firm's international experience and its decision to imitate the market leader's location choices. Our results show that the negative relationship between international experience and imitation of location choices is positively moderated by board turnover, board age, and board equity ownership but not influenced by board gender diversity. These findings advance our understanding of the interplay between information‐based motives for imitation and firms' information processing and organizational learning. Specifically, we contribute to research on the effect of international experience on firms' mimetic behavior by pointing out the relevance of experience interpretation and assessment from a microfoundations perspective. Our study provides indications for executives attempting to predict competitors' global strategy. When it comes to location choices, we find that companies with less international experience are more likely to follow the market leader, while those internationally experienced are more likely to follow their own path. Moreover, lower board turnover, relatively younger directors, and smaller equity ownership can favor the articulation and exploitation of the lessons offered by prior international experiences, thus further reducing the company's inclination to imitate the leader's location choices. Firms seeking an independent path toward internationalization can therefore use corporate governance—and in particular board‐level factors—to enhance their ability to interpret and assess their international experience.

Abstract

Hypervariable DNA markers, which are referred to as VNTRs (variable number of tandem repeats), are powerful tools in both genetic studies and forensic science because of their high polymorphic information [...], Linkage disequilibria in the apolipoprotein B (APOB) gene (EcoRI RFLP/3′ APOB VNTR) and in the thyroid peroxidase (TPO) gene (AcyI RFLP/TPO VNTR) were investigated in a sample of 100 individuals from southern Italy. By recoding multiallelic data as diallelic data, each RFLP-VNTR system showed linkage disequilibrium significantly different from zero (EcoRI RFLP/3′ APOB VNTR: p < 0.001; AcyI RFLP/TPO VNTR: p < 0.025), thus suggesting that the VNTR arrays are stable. Furthermore, the relationship between the 3′ APOB (2p24-p23) and TPO (2pter-p24) VNTR multiallelic systems was also analyzed. The two VNTR polymorphisms were found to be in linkage equilibrium, thus indicating that they can be used together in forensic casework. KEY WORDS: VNTR, TPO, 3′ APOB

Abstract

Even though the use of DNA markers containing a variable number of tandem repeats (VNTRs) is a recent development in population genetics, an increasing amount of data indicates their usefulness [...], Genotype and allele frequencies of the DYS19, D12S67, and D1S80 highly polymorphic loci were determined in population samples from southern Italy (103 subjects) and Greece (84 subjects) using the amplified fragment length polymorphism (AFLP) technique (polymerase chain reaction followed by native PAGE and silver staining). Five, eleven, and eighteen alleles were found at the DYS19, D12S67, and D1S80 loci, respectively. PIC values ranged from 0.55 (DYS 19 locus in Italians) to 0.79 (D12S67 locus in Italians). The distribution of D12S67 and D1S80 genotypes conformed to Hardy-Weinberg equilibrium, as confirmed by three statistics. Heterogeneity G tests, carded out on allele frequency distributions, showed a significant difference between the samples at the DYS19 locus, whereas no difference was found with regard to the other polymorphisms. Using data from the literature, we widened the comparison to other European groups analyzed for the same markers. All the polymorphisms were found to distinguish between populations of the same main ethnic group. In particular, D1S80 allele frequencies distinguished the Finns from other European groups (Spanish, German, Italian, and Greek samples). The reduced assay time, the high polymorphism level, and the ability to distinguish between populations indicate that these markers have potential value in population genetic studies.
