Abstract

Brown stem rot (BSR) caused by Cadophora gregata f. sp. adzukicola (syn. Phialophora gregata) is a serious soilborne disease of adzuki bean (Vigna angularis) in Japan. Cultivation of resistant cultivars is the most effective disease control method, therefore the selection of resistant lines is a priority for breeders. BSR-resistant adzuki bean lines have been screened in pathogen-infected fields. However, field selection using the pathogen and artificial inoculation methods is time-consuming and labor-intensive. In the present study, we used 105 F-3 lines derived from a cross between a BSR-resistant cultivar 'Syumari' and a susceptible cultivar 'Buchishoryukei-1' for BSR inoculation tests. Amplified fragment-length polymorphism (AFLP) analyses with 1024 primer sets revealed that six fragments were polymorphic between resistance and susceptible bulked groups. Five DNA markers (Pg77, Pg118, Pg138, Pg139 and Pg126) were developed from the nucleotide sequences of polymorphic AFLP markers and their flanking regions. Pg118, which was derived from E-ACT/M-ACT-118, was tightly linked to the resistance gene Pga1 and was converted into a codominant marker for its easier use in marker-assisted selection for adzuki bean BSR resistance. Finally, the applicability of the developed markers for BSR resistance was tested on 32 adzuki bean accessions or cultivars.

This study examined whether the representative bone absorption marker serum tartrate-resistant acid phosphatase 5b (TRACP-5b) or bone formation marker bone alkaline phosphatase (BAP) could estimate primary OP status and denosumab efficacy in a real-world setting. We retrospectively enrolled 114 female postmenopausal primary OP patients in Japan. Values and percent changes in TRACP-5b, BAP, lumbar 1–4 bone mineral density (L-BMD), and total hip BMD (H-BMD) were assessed before treatment and at 4, 8, and 12 months of therapy to identify the correlations between the percent changes in bone metabolic markers and BMD. We also established two sets of subgroups based on the upper limits of reference values in Japan for serum: TRACP-5b (<420 mU/dL) and (≥420 mU/dL) and BAP (<14.5 µg/L) and (≥14.5 µg/L). Negative correlations were observed for the percent changes of TRACP-5b and H-BMD at 4 months (r=–0.3476) and 8 months (r=–0.3880), for the percent changes of BAP and H-BMD at 8 months (r=–0.3354), and for the percent changes of BAP and L-BMD at 12 months (r=–0.3186). We observed a significant difference between the subgroups for the percent changes of L-BMD at 8 months (p=0.013) and 12 months (p=0.004) in BAP values. These results suggest that TRACP-5b and BAP had negative correlations with BMD, and that BAP represented a useful serum marker to evaluate L-BMD during denosumab therapy for OP. 166. Evaluation of the effects of five QTL regions on Fusarium head blight resistance and agronomic traits in spring wheat (Triticum aestivum L.)

Abstract

Fusarium head blight (FHB) is an important disease of wheat (Triticum aestivum L.). The aim of this study was to determine the effects of quantitative trait locus (QTL) regions for resistance to FHB and estimate their effects on reducing FHB damage to wheat in Hokkaido, northern Japan. We examined 233 F(1)-derived doubled-haploid (DH) lines from a cross between 'Kukeiharu 14' and 'Sumai 3' to determine their reaction to FHB during two seasons under field conditions. The DH lines were genotyped at five known FHB-resistance QTL regions (on chromosomes 3BS, 5AS, 6BS, 2DL and 4BS) by using SSR markers. 'Sumai 3' alleles at the QTLs at 3BS and 5AS effectively reduced FHB damage in the environment of Hokkaido, indicating that these QTLs will be useful for breeding spring wheat cultivars suitable for Hokkaido. Some of the QTL regions influenced agronomic traits: 'Sumai 3' alleles at the 4BS and 5AS QTLs significantly increased stem length and spike length, that at the 2DL QTL significantly decreased grain weight, and that at the 6BS QTL significantly delayed heading, indicating pleiotropic or linkage effects between these agronomic traits and FHB resistance.

Abstract

Wnt signaling is important for both skeletal development and bone disease, with Wnt inhibitory factors playing critical roles in bone metabolism. SHISA3 blocks the maturation and transportation of Frizzled receptors to the cell surface, thereby inhibiting the Wnt/β-catenin signaling pathway in lung cancer. However, the function of Shisa3 in bone biology remains uninvestigated. This study found that Shisa3 was strongly expressed in the calvarial bones of mice, especially in osteoblasts. In addition, adenovirus-mediated gene transfer of murine Shisa3 significantly inhibited Wnt3a-induced nuclear translocation of β-catenin and mRNA expression of the Wnt target gene Axin2. In bone phenotype assessments of Shisa3 knockout (Shisa3 KO) mice, micro-computed tomography, mRNA expressions of osteoblast markers, and skeletal preparations all displayed no significant differences compared with Shisa3 wild-type mice. mRNA expression analysis of canonical Wnt signaling target genes (Axin2, Lef1, Dkk1, and Tnfrsf11b) in calvarial bones at P0.5 also revealed no significant findings. In Axin2Cre/ERT2 knock-in mice, the number of Axin2-expressing cells in the calvariae of Shisa3 KO and control mice were comparable. Abstract

Appropriate management for osteoporosis in adult patients with Prader-Willi syndrome (PWS) has not been established. We report on a 21-year-old woman with PWS, who underwent denosumab treatment for osteoporosis. She presented with fractures and was shown to have very low bone mineral density (BMD), while she had been treated with supplementation of growth hormone for 7-14 years of age and estrogen from 15 years of age. BMD was monitored in the total hip region by dual-energy X-ray absorptiometry. Laboratory tests included bone-specific alkaline phosphatase, urinary type I collagen amino-terminal telopeptide, tartrate-resistant acid phosphatase 5b, 1-alpha, 25-dihydroxyvitamin D3, and parathyroid hormone. BMD and laboratory data were evaluated before and at 4, 8, and 13 months of treatment. After 13 months of denosumab therapy, BMD increased by 4.5%, and bone turnover markers notably improved. No fractures occurred. To the best of our knowledge, this is the first report to describe the clinical outcomes of denosumab treatment for osteoporosis in patients with PWS. Abstract

Osteoporosis-pseudoglioma (OPPG) syndrome is a very rare autosomal recessive disorder caused by mutations in the low-density lipoprotein receptor-related protein-5 (LRP5) gene. Treatment with bisphosphonates (BPs), particularly with pamidronate and risedronate, has been reported to have efficacy in this condition. However, there has been no consensus treatment strategy for OPPG. Moreover, there has been no report on the efficacy of denosumab in an OPPG patient to date. We enrolled an OPPG patient with osteoporosis treated by denosumab for a year. We report on a 19-year-old male patient with OPPG due to compound heterozygous mutations on the LRP5 gene (c.1145C > T,p.P382L and c.4600C > T,p.R1534*) who showed a good response after a 12-month period of denosumab therapy. Consequently, bone metabolism and bone mineral density were significantly improved after the treatment. In addition, no fracture or adverse effect was observed during the study period. Although BPs are considered to be administered in OPPG patients as a first-line therapy during early childhood, denosumab could be a good option to treat OPPG. [ABSTRACT FROM AUTHOR]
