Decomposition‐based solution algorithms for optimization problems depend on the underlying latent block structure of the problem. Methods for detecting this structure are currently lacking. In this article, we propose stochastic blockmodeling (SBM) as a systematic framework for learning the underlying block structure in generic optimization problems. SBM is a generative graph model in which nodes belong to some blocks and the interconnections among the nodes are stochastically dependent on their block affiliations. Hence, through parametric statistical inference, the interconnection patterns underlying optimization problems can be estimated. For benchmark optimization problems, we show that SBM can reveal the underlying block structure and that the estimated blocks can be used as the basis for decomposition‐based solution algorithms which can reach an optimum or bound estimates in reduced computational time. Finally, we present a general software platform for automated block structure detection and decomposition‐based solution following distributed and hierarchical optimization approaches. Background: Regorafenib improves progression-free survival (PFS) and overall survival (OS) in patients with refractory metastatic colorectal cancer (mCRC). Here, we report the treatment patterns of regorafenib in the third- or late-line setting for mCRC in four centers in China. Patients and Methods: Patients with refractory mCRC in four centers in China administered regorafenib from February 1, 2018 to June 31, 2021 were enrolled. Patients were grouped into 3 cohorts, namely, the monotherapy (regorafenib alone), chemo (regorafenib plus chemotherapy), and immune [regorafenib plus anti-PD1 (programmed cell death 1) antibodies] groups. Demographic, clinical, survival and safety data were retrospectively analyzed. Results: A total of 177 patients were included in this study. Of them, 116 (65.5%) were treated with regorafenib alone, while 28 (15.9%) and 33 (18.6%) were administered regorafenib plus chemotherapy and anti-PD1 antibodies, respectively. The median followed-up time was 9.2 months. The disease control rate (DCR) was 40.7%. The median PFS (mPFS) was 2.43 months and the median OS (mOS) was 12.2 months. The immune group had longer median PFS (3.5 m vs. 2.2 m, p = 0.043) compared with the monotherapy group. Patients administered regorafenib plus chemotherapy had longer median OS (15.9 m vs. 8.4 m, p = 0.032) compared with the monotherapy group. Patients who began regorafenib treatment at 120 mg had longer median PFS and OS compared with those who began at 80 mg (PFS: 3.7 m vs. 2.0 m; p <0.001; OS: 13.4 m vs. 10.2 m; p = 0.005). Patients with a final dose of 120 mg had longer median PFS and OS compared with the 80 mg or less group (PFS: 5.0 m vs. 2.3 m; p = 0.045; OS: UR (unreach) vs. 10.9 m; p = 0.003). There were 87.0% (154/177) patients who experienced AEs. Three groups had similar rates of AEs (86.2% vs. 89.3% vs. 87.9%; p = 0.89). Conclusion: Patients administered regorafenib alone or regorafenib in combination with other agents were relieved to some extent, with a disease control rate of 40.7%. Regorafenib plus anti-PD1 antibodies showed better PFS, while regorafenib plus chemotherapy had the most benefit in OS. There was no significant difference among three groups in terms of AEs. Women with premature ovarian insufficiency (POI) may be more vulnerable to a variety of health risks. To seek a new method to treat the disease, the effects of low-intensity pulsed ultrasound (LIPUS) on promoting repair of ovarian injury in female SD rats induced by 4-vinylcyclohexene diepoxide (VCD) were explored in this research. A total of 24 female SD rats were subjected to intraperitoneal injection of VCD to induce POI. Successful modeling was achieved in 22 rats, which were then randomized into VCD + LIPUS group (n = 13) and VCD group (n = 9). The control group (n = 5) was injected with equal normal saline. Hematoxylin and eosin staining, enzyme-linked immunosorbent assay, Western blot analysis, scanning electron microscope, immunohistochemistry, and terminal deoxynucleotidyl transferase-mediated nick end labeling assay were applied to detect the results. The results indicated that rats in the VCD group showed disorder in the estrous cycle, the number of atresia follicles and apoptosis granulosa cells increased (p < .05). After the LIPUS treatment, the estrous cycle recovered, the number of follicles increased (p < .05), the level of E2 and anti-Müllerian hormone enhanced (p < .05), and the follicle-stimulating hormone decreased (p < .05). The expression of NF-κB p65, TNFα, Bax, ATF4, and caspase-3 in ovarian tissue was significantly decreased (p < .05). These findings showed that LIPUS could promote the repair of the VCD-induced ovarian damage in SD rats, which has the potential to be further applied in the clinic. [ABSTRACT FROM AUTHOR]

The global amplitude of the westward propagating quasi-16-day waves (16DW) with wavenumber 1 (Q16W1), the strongest component of 16DW, are derived from the European Centre for Medium-Range Weather Forecasts ERA-Interim reanalysis temperature and zonal wind data sets from February 1979 to January 2018. In terms of temperature and zonal wind, strong climatologically average amplitudes of Q16W1 appear in the upper stratosphere at mid–high latitudes in both hemispheres, and the wave amplitude is stronger in the Northern Hemisphere (NH) than in the Southern Hemisphere (SH). Multivariate linear regression is separately applied to calculate the responses of the Q16W1 temperature and zonal wind amplitudes to the QBO (quasi-biennial oscillation), ENSO (El Niño-Southern Oscillation), solar activity and linear trends of the Q16W1 amplitude. The QBO signatures of the Q16W1 temperature and zonal wind amplitudes are mainly located in the stratosphere. The Q16W1 has significant QBO responses at low latitudes. In addition, only the temperature amplitude presents a larger QBO signature in its strongest climatological amplitude region. No significant responses to ENSO and solar activity are observed in temperature and zonal wind amplitudes. The linear trends of the monthly mean Q16W1 temperature and zonal wind amplitude are generally positive, especially in the mid-upper stratosphere. The trend is asymmetric about the equator and significantly stronger in the NH than in the SH. The seasonal variation in the trend of the temperature amplitude is studied and illustrated to be stronger in winter and weaker in spring and autumn. Further investigation suggests that the background and local instability trends contribute most of the increasing trend of the Q16W1 amplitude. In winter in both hemispheres, a weakening trend of eastward zonal wind provides more favourable background wind for Q16W1 upward propagation, in autumn and winter in the NH and in spring, autumn and winter in the SH, and the increasing trend of local instability may enhance wave excitation.
