Abstract Lysosomal biogenesis plays a vital role in cell fate. Under certain conditions, excessive lysosomal biogenesis leads to susceptibility for lysosomal membrane permeabilization resulting in various pathological conditions including cell death. In cancer cells apoptosis machinery becomes dysregulated during the course of treatment, thus allows cancer cells to escape apoptosis. So it is therefore imperative to identify cytotoxic agents that exploit non-apoptotic mechanisms of cell death. Our study showed that pancreatic cancer cells treated with SDS-203 triggered an incomplete autophagic response and a nuclear translocation of transcriptional factor TFEB. This resulted in abundant biosynthesis and accumulation of autophagosomes and lysosomes into the cells leading to their death. It was observed that the silencing of autophagy genes didn’t alter the cell fate, whereas siRNA-mediated silencing of TFEB subdued SDS-203 mediated lysosomal biogenesis and associated cell death. Further mouse tumors treated with SDS-203 showed a significant reduction in tumor burden and increased expression of lysosomal markers. Taken together this study demonstrates that SDS-203 treatment triggers non-apoptotic cell death in pancreatic cancer cells through a mechanism of lysosome over accumulation.

Hybrid nanofluid, which is a combination of Propylene Glycol (PG) – Water (H2O) admixture and paraffin wax and sand, may be utilized as a standby for PG and (H2O) blend in solar thermal framework. Objective of this article is the exploration of the dissipative flow propylene-glycol and water mixture based hybrid nanofluid by a sphere with chemical reaction and heat source parameters. MATLAB in-built solver bvp4c is utilized to exhibit the impacts of various parameters on regular profiles including temperature. Correlation coefficient is utilized to elucidate the impact of pertinent parameters on engineering parameters of concern, such as, surface friction factor. Main findings of this work are magnetic field is having a negative association with friction factor and chemical reaction is consuming a significant positive relationship with Sherwood number. It is witnessed that heat source and Eckert number are useful to meliorate the fluid temperature. Furthermore, validation is performed among our results and earlier published outcomes. Good agreement is detected.

Obesity has reached epidemic proportions, emphasizing the importance of reliable biomarkers for detecting early metabolic alterations and enabling early preventative interventions. However, our understanding of the molecular mechanisms and specific lipid species associated with childhood obesity remains limited. Therefore, the aim of this study was to investigate plasma lipidomic signatures as potential biomarkers for adolescent obesity. A total of 103 individuals comprising overweight/obese (n = 46) and normal weight (n = 57) were randomly chosen from the baseline ORANGE (Obesity Reduction and Noncommunicable Disease Awareness through Group Education) cohort, having been followed up for a median of 7.1 years. Plasma lipidomic profiling was performed using the UHPLC-HRMS method. We used three different models adjusted for clinical covariates to analyze the data. Clustering methods were used to define metabotypes, which allowed for the stratification of subjects into subgroups with similar clinical and metabolic profiles. We observed that lysophosphatidylcholine (LPC) species like LPC.16.0, LPC.18.3, LPC.18.1, and LPC.20.3 were significantly (p < 0.05) associated with baseline and follow-up BMI in adolescent obesity. The association of LPC species with BMI remained consistently significant even after adjusting for potential confounders. Moreover, applying metabotyping using hierarchical clustering provided insights into the metabolic heterogeneity within the normal and obese groups, distinguishing metabolically healthy individuals from those with unhealthy metabolic profiles. The specific LPC levels were found to be altered and increased in childhood obesity, particularly during the follow-up. These findings suggest that LPC species hold promise as potential biomarkers of obesity in adolescents, including healthy and unhealthy metabolic profiles. Ever since lightweight cryptography emerged as one of the trending topics in symmetric key cryptography, optimizing the implementation cost of MDS matrices has been in the center of attention. In this direction, various metrics like d-XOR, s-XOR and g-XOR have been proposed to mimic the hardware cost. Consequently, efforts also have been made to search for the optimal MDS matrices for dimensions relevant to cryptographic applications according to these metrics. However, finding the optimal MDS matrix in terms of hardware cost still remains an unsolved problem. In this paper, we settle the question of the optimal 4 x 4 MDS matrices over GL(n, F2) under the recently proposed metric sequential XOR count based on words (sw-XOR). We prove that the sw-XOR of such matrices is at least 8n + 3, and the bound is tight as matrices with sw-XOR cost 35 and 67 for the values of n = 4 and 8, respectively, were already known. Moreover, the lower bound for these values of n matches with the known lower bounds according to s-XOR and g-XOR metrics.

The density (ρ) and speed of sound (u) of binary solutions containing protic ionic liquids such as N‑butyl ethanolammonium acetate [NBEA][Ace] and N‑butyl ethanolammonium propionate [NBEA][Pro] with Dimethyl Sulfoxide (DMSO) were determined over a temperature range of (293.15 to 318.15) K at intervals of 5 K at 0.1 MPa pressure. The apparent molar volume (Vφ), isentropic compressibility (ks) and apparent molar isentropic compressibility (ks,φ) were estimated using the measured data. The apparent molar volume (Vφ∞) and apparent molar isentropic compressibility (kφ∞) at infinite dilution, as well as the related empirical parameters (Sv, Bv, Sk and Bk), were calculated using the Redlich-Mayer type equation. The solute-solute and solute-solvent interactions are indicated by the obtained limiting apparent molar volume and limiting apparent molar isentropic compressibility. Furthermore, the apparent molar expansibility at infinite dilution was calculated using temperature dependent apparent molar volume at infinite dilution (Eφ∞).

The trial was undertaken in a completely randomized design to to evaluate the synergistic effect of probiotic (Pro), chicory root powder (CRP) and coriander seed powder (CSP) on the performance of broiler chicken. A total of 240 day-old broiler chicks were randomly allotted to six dietary treatments with 8 replicates of 5 birds in each. Treatment groups included T1 as control, i.e. basal diet (BD) without any growth promoter and T2 - BD + antibiotic (BMD @ 500 gm/ton). In the remaining experimental diets, T3 - pro (@ 10 gm/100 kg) + CRP (@ 1.0%), T4 - pro (@ 10 gm/100 kg) + CSP (@ 1.5%), T5 - CRP (@ 1.0%) + CSP (@ 1.5%) and T6 - pro (@ 10 gm/100 kg) + CRP (@ 1.0%) + CSP (@ 1.5%). The results revealed that supplementation of pro + CRP significantly increased the body weight gain during prestarter, starter and finisher phase and improved the feed conversion ratio during finisher phase compared to other treatment groups. Supplementation of different dietary groups did not show any significant effect on feed intake and various slaughter parameters of broilers. Whereas, the humoral immune response to ND vaccine and immune organ weights were significantly higher in all the test diets (T3 to T6) compared to control and antibiotic groups. Supplementation of probiotic with chicory combination group significantly reduced the serum total cholesterol and recorded higher returns over feed cost compared to other treatment groups. It can be concluded that combination of probiotic (10 g) with chicory root powder (1.0%) proved more effective than combinations of other additives.

BackgroundThe leading edge of the global problem of antibiotic resistance necessitates novel therapeutic strategies. This study develops a novel systems biology driven approach for killing antibiotic resistant pathogens using benign metabolites.ResultsControlled laboratory evolutions established chloramphenicol and streptomycin resistant pathogens of Chromobacterium. These resistant pathogens showed higher growth rates and required higher lethal doses of antibiotic. Growth and viability testing identified malate, maleate, succinate, pyruvate and oxoadipate as resensitising agents for antibiotic therapy. Resistant genes were catalogued through whole genome sequencing. Intracellular metabolomic profiling identified violacein as a potential biomarker for resistance. The temporal variance of metabolites captured the linearized dynamics around the steady state and correlated to growth rate. A constraints-based flux balance model of the core metabolism was used to predict the metabolic basis of antibiotic susceptibility and resistance.ConclusionsThe model predicts electron imbalance and skewed NAD/NADH ratios as a result of antibiotics - chloramphenicol and streptomycin. The resistant pathogen rewired its metabolic networks to compensate for disruption of redox homeostasis. We foresee the utility of such scalable workflows in identifying metabolites for clinical isolates as inevitable solutions to mitigate antibiotic resistance.

Wnt/β-catenin (WβC) signaling pathway is an important signaling pathway for the maintenance of cellular homeostasis from the embryonic developmental stages to adulthood. The canonical pathway of WβC signaling is essential for neurogenesis, cell proliferation, and neurogenesis, whereas the noncanonical pathway (WNT/Ca2+ and WNT/PCP) is responsible for cell polarity, calcium maintenance, and cell migration. Abnormal regulation of WβC signaling is involved in the pathogenesis of several neurodegenerative diseases such as Alzheimer’s disease (AD), Parkinson’s disease (PD), Huntington’s disease (HD), amyotrophic lateral sclerosis (ALS), multiple sclerosis (MS), and spinal muscular atrophy (SMA). Hence, the alteration of WβC signaling is considered a potential therapeutic target for the treatment of neurodegenerative disease. In the present review, we have used the bibliographical information from PubMed, Google Scholar, and Scopus to address the current prospects of WβC signaling role in the abovementioned neurodegenerative diseases.

The current situation needs to be reflected timelier. With the rise of social communities, community users’ comments have gradually become an important reference for other community members. Twitter is one of the most popular social media in the world. During the COVID-19 pandemic, people were restricted by rules and government policies, such as wearing masks, maintaining social distancing, and avoiding crowding. This led people to spend time on devices. By using devices, most people are involved in social media activities. This study aims to discover the awareness Indonesians display in the text they upload to Twitter. Using the Twitter crawling technique, we collected data. We also analyzed the text with text mining techniques and latent semantic analysis (LSA) with semiotic methods. The crisis communication was classified, and the definition of crisis terminology was improved in social media.

Programmatic advertising uses big data to spread personalized marketing materials to target audiences, which is a major driving force for the growth of digital advertising. Among them, in-application advertisements (in-app ads) are an important part of programmatic advertising. In in-app advertising, which is highly related to application revenue, ads are delivered to customers through mobile devices at any time and place based on personal needs. Due to the power of electronic Word-of-Mouth (e-WOM), text comments from social media are becoming a new mode of advertising, influencing consumers’ purchase behavior. Text reviews on social media are more powerful than traditional ads. However, relatively little research has studied this issue. Therefore, using text mining and latent semantic analysis techniques, we aimed to discover the advertising elements of text reviews in the social community. Based on the results, suggestions were made to advertising companies to improve the performance of text reviews when employing key opinion leaders (KOL) to write commercial comments that promote products or services.

Highly metastatic breast cancers, such as triple-negative subtypes (TNBC), require the most effective treatments. Since interleukin-13 receptor (IL-13R)α2 is reportedly over-expressed in some cancers, we investigated here its expression and the feasibility of therapeutically targeting this receptor in breast cancer using a novel hybrid cytolytic peptide (Pep-1-Phor21) consisting of IL-13Rα2-binding (Pep-1) and cytolytic (Phor21) domains. This study demonstrates that particularly TNBC tissues and cells display the prominent expression of IL-13Rα2. Furthermore, Pep-1-Phor21 induced the rapid necrosis of tumor cells expressing cell-surface IL-13Rα2. Notably, IL-13Rα2 expression was found to be epigenetically regulated in breast cancer cells in that the inhibition of histone deacetylase (HDAC) or DNA methyltransferase (DNMT) upregulated IL-13Rα2 expression, thereby sensitizing them to Pep-1-Phor21. IL-13Rα2-negative non-malignant cells were refractory to these epigenetic effects. Consistent with its cytolytic activity, Pep-1-Phor21 readily destroyed IL-13Rα2-expressing breast cancer spheroids with HDAC or DNMT inhibition, further enhancing cytolytic activity. Therefore, the Pep-1-Phor21-mediated targeting of IL-13Rα2 is a potentially novel therapeutic strategy for TNBC. Given that tumor cells can be selectively sensitized to Pep-1-Phor21 via the epigenetic up-regulation of IL-13Rα2, a combined adjuvant approach involving Pep-1-Phor21 and epigenetic inhibitors may be an effective strategy.

In this paper,we introduce and study a new subclass of meromorphic functions associated with a certain differential operator on Hilbert space. For this class, we obtain several properties like the coefficient inequality, growth and distortion theorem, radius of close-to-convexity, starlikeness and meromorphically convexity and integral transforms. Further, it is shown that this class is closed under convex linear combinations.

A study was carried out for a period of 90 days during the summer season with the inclusion of chromium propionate and vitamin E with selenium, in the feed to investigate their effect on 24 uniparous lactating Holstein Friesian cows in an early phase of lactation. Cows were randomly assigned to four supplemental doses of 0 (T0); 500 IU/animal/day of vitamin E and 0.3 mg/kg DM of selenium (T1); 0.5 mg/kg DM of chromium propionate (T2) and 0.5 mg/kg DM of chromium propionate, 500 IU/animal/day of vitamin E and 0.3 mg/kg DM of selenium (T3). The mean THI value in the morning was 75.10±0.42 and 80.01±0.64 in the afternoon indicating that the animals were under mild to moderate heat stress. Increase in the DMI was found in the supplemented groups with a significant difference between control and T3 group. Milk yield differed significantly between the control and supplemented groups. The hematological profile did not vary significantly among the groups. Supplementation of chromium along with vitamin E and selenium decreased the cortisol level and glutathione peroxidase activity signigicantly in the T3 group. Chromium supplemented groups (T2 and T3) showed greater increase in mean total protein, albumin concentration and A/G ratio. However, the mean values for thyroid profile, liver enzymes, superoxide dismutase, catalase, globulin, total cholesterol, triglycerides, glucose, urea, creatinine were comparable among the four groups. Statistical analysis revealed that the rectal temperature, respiratory rate, and pulse rate in supplemented groups were significantly reduced. The findings showed that chromium supplementation plays a significant role in alleviating thermal stress and is further more beneficial when supplemented with proven antioxidants vitamin E and selenium.

Productivity of cropland soils, especially in arid and semi-arid tropics of India rigorously declined due to increasing losses of soil organic carbon (SOC) over the past decades. In the present study, we examined a 16-years long term experiment with continuous mono cropping rainfed groundnut (Arachis hypogaea) to quantify the influence of fertilization and manuring on yield sustainability and C sequestration potential on rainfed Vertisols of Western India. The treatments include, i) T1 = control; ii) T2 = 100% recommended dose of fertilizer [RDF-20:40:40 kg ha−1 of N:P2O5: K2O]; iii) T3 = integrated nutrient management [INM- 50% RDF + compost 6 Mg ha−1 + biofertilizers (BF)]; iv) T4 = organic nutrient source [compost 6 Mg ha−1 + vermicompost (VC) 2 Mg ha−1 + castor neem cake (CNC) 250 kg ha−1 + BF + mulching]. Maximum mean groundnut pod yield (1.17 Mg ha−1) was recorded in T2 which was statistically similar to T4 (1.16 Mg ha−1). However, Mann-Kendall test for yield trend suggests the consistency in yield increase under T3 treatment during the last 16 years. The rate of pod yield enhancement was 27 kg ha−1 for every Mg increase in profile SOC stock. The mean SOC concentration (g kg−1) of 1-m soil depth increased from 4.0 to 5.6 (40%) in T4 over control and the mean SOC sequestration rate was 0.63 Mg C ha−1 yr−1. A minimum of 1.22 Mg C ha−1 yr−1 input was needed to maintain SOC stock at its antecedent level (zero change). We conclude that combined use of chemical fertilizers along with locally available organic resources is essential for enhancing SOC storage while achieving sustainable crop productivity in semi-arid agro-ecosystem.

Abstract Metabolic reprogramming and its molecular underpinnings are critical to unravel the duality of cancer cell function and chemo-resistance. Here, we use a constraints-based integrated approach to delineate the interplay between metabolism and epigenetics, hardwired in the genome, to shape temozolomide (TMZ) resistance. Differential metabolism was identified in response to TMZ at varying concentrations in both the resistant neurospheroidal (NSP) and the susceptible (U87MG) glioblastoma cell-lines. The genetic basis of this metabolic adaptation was characterized by whole exome sequencing that identified mutations in signaling pathway regulators of growth and energy metabolism. Remarkably, our integrated approach identified rewiring in glycolysis, TCA cycle, malate aspartate shunt, and oxidative phosphorylation pathways. The differential killing of TMZ resistant NSP by Rotenone at low concentrations with an IC50 value of 5 nM, three orders of magnitude lower than for U87MG that exhibited an IC50 value of 1.8 mM was thus identified using our integrated systems-based approach.

Heart failure is a common, complex clinical syndrome with high morbidity and mortality. Hemodynamic parameter evaluation is useful for early detection, clinical outcome monitoring, timely treatment, and the overall prognosis of heart failure patients. Therefore, continuous monitoring of hemodynamic parameters helps in the evaluation of patients with suspected heart failure. The hemodynamic parameters change with respect to the contraction and expansion of the heart. Hence, in this research, two circuit-less 30 mm spherical receiver coils were implanted in both the left and right sides of the heart and an external transceiver coil was placed above the chest. The changes in the reflection coefficient of the transceiver coil at the resonant frequency changed depending on the distance between the implanted coils, allowing the contraction and expansion of the heart to be determined. This work was carried out at 13.56 MHz, considering the safety limits imposed by the FCC. The proposed reflection coefficient monitoring technique may distinguish healthy patients from heart failure and heart attack patients. The reflection coefficients at a maximum distance of 50 mm for simulation and measurement are −10.3 dB and −10.6 dB, respectively, at the resonant frequency.

Coronavirus disease COVID-19, which is caused by severe acute respiratory syndrome coronavirus SARS-CoV-2, has become a worldwide pandemic in recent years. In addition to being a respiratory disease, COVID-19 is a ‘vascular disease’ since it causes a leaky vascular barrier and increases blood clotting by elevating von Willebrand factor (vWF) levels in the blood. In this study, we analyzed in vitro how the SARS-CoV-2 spike protein S1 induces endothelial cell (EC) permeability and its vWF secretion, and the underlying molecular mechanism for it. We showed that the SARS-CoV-2 spike protein S1 receptor-binding domain (RBD) is sufficient to induce endothelial permeability and vWF-secretion through the angiotensin-converting enzyme (ACE)2 in an ADP-ribosylation factor (ARF)6 activation-dependent manner. However, the mutants, including those in South African and South Californian variants of SARS-CoV-2, in the spike protein did not affect its induced EC permeability and vWF secretion. In addition, we have identified a signaling cascade downstream of ACE2 for the SARS-CoV-2 spike protein-induced EC permeability and its vWF secretion by using pharmacological inhibitors. The knowledge gained from this study could be useful in developing novel drugs or repurposing existing drugs for treating infections of SARS-CoV-2, particularly those strains that respond poorly to the existing vaccines.

The presence of microplastics and nanoplastics (MNPs) in soils is becoming pervasive in most agroecosystems. The recent estimates suggest that the soil burden of MNPs in the agroecosystems is more than 0.5 megatons (Mt) annually. In all the agroecosystems, the transformation, migration, and transferring of MNPs, along with other contaminants, and the trophic transfer of MNPs can threaten the food web. MPs can exhibit negative and positive effects, or none, on the physical/chemical properties of soil, soil microbiota, invertebrates, and plant systems, depending on the polymer compositions, additives, and exposure time. Difficulties in comparing the studies on the effects of MNPs, as well as the discrepancies among them, are mostly due to variations in the methods followed for sampling, detection, quantification, and the categorization of particles, abundance, and exposure time. Since agricultural soils are important environmental reservoirs for diverse chemicals and contaminants, they provide milieus for several types of interactions of MNPs with soil biota. The present review critically examines the sources and transformation of MNPs in agricultural soils, the release and fate of additives, as well as their role as vectors of other potential contaminants and influence on soil physical/chemical properties, toxicities to soil biota (i.e., microorganisms, invertebrates, and plants), current regulatory guidelines for the mitigation of MNPs, and future research directions.

The IL-13Rα2 cell surface receptor is highly expressed in tumours such as prostate cancer. In this report, we evaluated the hypothesis that prostate cancer cells with enhanced IL-13Rα2 expression are a suitable target for the hybrid lytic peptide (Pep-1-Phor21) peptide, which is generated by fusing the IL-13Rα2 specific ligand (Pep-1) and a cell membrane disrupting lytic peptide (Phor21). The expression of IL-13Rα2 mRNA and protein in prostate cancer tissues and cell lines was assessed via real-time PCR (RT-PCR) and immunoblotting. The effect of Pep-1-Phor21 on the viability of prostate cancer cells grown in monolayers (2D) and microtissue spheroids (3D) was assessed via CellTox green cytotoxic assay. IL-13Rα2 expression and Pep-1-Phor21-mediated killing were also determined in the cells treated with epigenetic regulators (Trichostatin A (TSA) and 5-aza-2 deoxycytidine (5-Aza-dC)). The hybrid lytic peptide cytotoxic activity correlated with the expression of IL-13Rα2 in prostate cancer cell lines cultured as monolayers (2D) or 3D spheroids. In addition, TSA or 5-Aza-dC treatment of prostate cancer cells, particularly those with low expression of IL-13Rα2, enhanced the cells’ sensitivity to the lytic peptide by increasing IL-13Rα2 expression. These results demonstrate that the Pep-1-Phor21 hybrid lytic peptide has potent and selective anticancer properties against IL-13Rα2-expressing prostate cancer cells.

In the current work, the morphological and mechanical features of a hybrid epoxy matrix composite created by hand layup and reinforced with fibers from rice husk, sisal, and kenaf are predicted by using Watershed segmentation algorithm. To forecast the effect of coating composition on physical performance of a composite, Tensile and hardness tests were used to define the resulting hybrid composites' mechanical characteristics. The objective of the empirical investigation presented in this work is to understand the mechanical behavior of hybrid natural fiber composites. The production of rose husk-sisal-kenaf-epoxy hybrid samples is done by hand layup, with the planned plies being alternately stacked and the weight of the matrix and fibers being kept between 40% and 60%. Samples are sliced from a laminate that has been manufactured in accordance with ASTM specifications in order to undertake various experiments. Dog bone and flat bar shapes were used to cut specimens for tensile test & flexural test. Tensile strength and flexural strength were measured following execution of experiment under UTM and contrasted with base values of used epoxy polymer to identify constant change in strength.

It became a bottleneck for the Machine Learning (ML) researchers to select/develop a sustainable model for a particular problem. Hence, there is a need for an approach to prepare a model with all constraints of the software system. The proposed approach is based on Object Constraint Language (OCL) which is a declarative language for writing constraints on software artifacts, it is widely used for effective representation of Functional Requirements (FR’s) and Non-Functional Requirements (NFR’s). In the proposed system, the paddy leaf disease identification system is considered and proposed Leaf Identification Constraints (LIC) and Leaf Disease Identification Constraints (LDIC) based on OCL, for the proposed constraints the Convolutional Neural Network (CNN) is chosen, as it can handle diverse range of input data and large volume of concurrent requests. To satisfy other constraints of the system, the Auto encoders are used along with CNN and the input data was take in the form of thermal imaging. This system was evaluated with test data and validation data and obtained the accuracy of 90.6%. And 84.8 was attained by earlier researchers before this approach.

Nowadays, industry 4.0 plays a tremendous role in the manufacturing industries for increasing the amount of data and accuracy in modern manufacturing systems. Thanks to artificial intelligence, particularly machine learning, big data analytics have dramatically amended, and manufacturers easily exploit organized and unorganized data. This study utilized hybrid optimization algorithms to find friction stir welding and optimal hardness value at the nugget zone. A similar AA 6262 material was used and welded in a butt joint configuration. Tool rotational speed (RPM), tool traverse speed (mm/min), and the plane depth (mm) are used as controllable parameters and optimized using Taguchi L9, Random Forest, and XG Boost machine learning tools. Analysis of variance was also conducted at a 95% confidence interval for identifying the significant parameters. The result indicated that the coefficient of determination from Taguchi L9 orthogonal array is 0.91 obtained while Random Forest and XG Boost algorithm imparted 0.62 and 0.65 respectively. Internet of Things (IoT) innovation is one of the fastest growing fields in various regions or aspects which include irrigation.
