Abstract

Survey researchers have carefully modified their data collection operations for various reasons, including the rising costs of data collection and the ongoing Coronavirus disease (COVID-19) pandemic, both of which have made in-person interviewing difficult. For large national surveys that require household (HH) screening to determine survey eligibility, cost-efficient screening methods that do not include in-person visits need additional evaluation and testing. A new study, known as the American Family Health Study (AFHS), recently initiated data collection with a national probability sample, using a sequential mixed-mode mail/web protocol for push-to-web US HH screening (targeting persons aged 18–49 years). To better understand optimal approaches for this type of national screening effort, we embedded two randomized experiments in the AFHS data collection. The first tested the use of bilingual respondent materials where mailed invitations to the screener were sent in both English and Spanish to 50 percent of addresses with a high predicted likelihood of having a Spanish speaker and 10 percent of all other addresses. We found that the bilingual approach did not increase the response rate of high-likelihood Spanish-speaking addresses, but consistent with prior work, it increased the proportion of eligible Hispanic respondents identified among completed screeners, especially among addresses predicted to have a high likelihood of having Spanish speakers. The second tested a form of nonresponse follow-up, where a subsample of active sampled HHs that had not yet responded to the screening invitations was sent a priority mailing with a $5 incentive, adding to the $2 incentive provided for all sampled HHs in the initial screening invitation. We found this approach to be quite valuable for increasing the screening survey response rate.

Abstract

The control of hydrogen concentration and diffusion in metal alloys is critical for advancing clean energy technologies. A quantitative understanding of the composition–property relationship can accelerate the design of hydrogen storage materials, structural hydrogen-resistant alloys, and materials for energy-efficient nanoelectronics. In this work, we employed Density Functional Theory simulations to investigate the energy landscape of hydrogen absorption and mobility for a wide spectrum of high-entropy alloys and intermetallic compounds. Our study sheds light on the origin of hydrogen stability also providing quantitative guidance for compositional considerations and the design of technologically useful materials. The developed analytical model uses physically intuitive metrics characterizing the local environment, such as electronic structure features, volume of interstitial voids, and lattice vibrational modes, to predict the energy landscape of hydrogen in metal alloys based on simple calculations of bulk properties. The developed model exhibits remarkable accuracy in predicting hydrogen binding energy, achieving a low mean absolute error of less than 0.1 eV. Utilizing this model, we successfully predicted hydrogen absorption energies and migration barriers demonstrating that these quantities are linearly correlated, akin to the Brønsted–Evans–Polanyi principle. Abstract

Responsive survey design is a technique aimed at improving the efficiency or quality of surveys by using incoming data from the field to make design changes. The technique was pioneered on large national surveys, but the tools can also be applied on the smaller-scale surveys most commonly used by sociologists. We demonstrate responsive survey design in a small-scale, list-based sample survey of students on the topic of sexual misconduct. We investigate the impact of individual incentive levels and a two-phase responsive design with changes to mode of contact as approaches for limiting the potential of nonresponse bias in data from such surveys. Our analyses demonstrate that a two-phase design introducing telephone and face-to-face reminders to complete the survey can produce stronger change in response rates and characteristics of those who respond than higher incentive levels. Abstract

In push-to-web surveys that use postal mail to contact sampled cases, participation is contingent on the mail being opened and the survey invitations being delivered. The design of the mailings is crucial to the success of the survey. We address the question of how to design invitation mailings that can grab potential respondents' attention and sway them to be interested in the survey in a short window of time. In the household screening stage of a national survey, the American Family Health Study, we experimentally tested three mailing design techniques for recruiting respondents: (1) a visible cash incentive in the initial mailing, (2) a second incentive for initial nonrespondents, and (3) use of Priority Mail in the nonresponse follow-up mailing. We evaluated the three techniques' overall effects on response rates as well as how they differentially attracted respondents with different characteristics. We found that all three techniques were useful in increasing the screening response rates, but there was little evidence that they had differential effects on sample subgroups that could help to reduce nonresponse biases. Abstract

Microfluidics-based sorting of cells after encapsulation in water-in-oil or water/oil/water droplet emulsions has great potential to significantly accelerate the design-build-test cycle for production strain engineering. This is especially true for non-conventional hosts and products that are secreted, as screening can be performed with minimal prior biological knowledge (relative to rational engineering) and entrapment of secreted product with the responsible producer cell can enable high throughput screening directly for extracellular product concentration. Microfluidic technology for droplet sorting has advanced and become more available in recent years, but gaps remain, including the limited number of droplet-compatible methods for transducing extracellular product titers into sortable fluorescence signals, and a lack of direct, matched comparisons to more established techniques such as fluorescence activated cell sorting (FACS). This research used yeast-based systems (Saccharomyces cerevisiae and Yarrowia lipolytica) as test beds for focusing on and addressing three areas that are related to these limitations. In the first focus area, we develop RNA aptamers in droplets (RAPID) screening, a method that expands the generality of high throughput microfluidic droplet screening. Using RNA aptamers, we transduce extracellular titer into fluorescence, enabling the screening of millions of yeast variants and ultimately enhancing secretion of a recombinant protein (streptavidin) from yeast. For the second focus area, we undertook a comparative analysis of traditional single cell fluorescence activated cell sorting (single cell FACS) and microdroplet-enabled FACS (droplet FACS) using water/oil/water (w/o/w) emulsions. To do so, we engineered and evolved the non-conventional yeast Y. lipolytica for high extracellular production of riboflavin (vitamin B2), an innately fluorescent product. In a parity-matched comparison of single cell FACS and droplet FACS, both techniques led to significant increases in total riboflavin. However, single cell FACS favored intracellular accumulation, while droplet FACS favored extracellular product accumulation and yielded higher total riboflavin production. In the third focus area, a droplet-based bacterial-fungal consortium/co-culture approach was applied, wherein Escherichia coli biosensors were used to screen for improved extracellular polyketide production from Y. lipolytica. This droplet sorting strategy was used to screen genome-wide piggyBac transposon libraries, leading to the identification of several alleles for enhancing triacetic acid lactone (TAL) production.
