Abstract Background Single-agent chemotherapy using methotrexate or actinomycin D is the first-line treatment for patients with low-risk gestational trophoblastic neoplasia. Various methotrexate-based and actinomycin D-based single-agent regimens can be used. However, there is insufficient evidence to determine the superior regimen. To guide doctors in selecting a single-agent chemotherapy regimen for patients with low-risk gestational trophoblastic neoplasia, we will compare two regimens. Methods We will conduct a multicentre, randomized, prospective clinical trial. Selected low-risk gestational trophoblastic neoplasia patients (FIGO score 0–4) will be randomized 1:1 to a biweekly single-dose actinomycin D group or a multiday methotrexate therapy group. The actinomycin D group will receive IV pulse actinomycin D (1.25 mg/m2) every 14 days, and the methotrexate group will receive methotrexate (50 mg) intramuscularly on days 1, 3, 5, and 7 (4 doses per cycle) and leucovorin (15 mg) intramuscularly on days 2, 4, 6, and 8. This process will be repeated every 14 days. The primary endpoints will include the complete remission rate by single-agent therapy and the overall complete remission rate. The secondary endpoints will include the duration needed to achieve complete remission after single-agent chemotherapy, number of courses needed to achieve complete remission after single-agent chemotherapy, incidence and severity of adverse effects, effects on menstrual conditions and ovarian function based on the anti-Mullerian hormone level, and patient-reported quality of life. Discussion Previous clinical trials comparing biweekly single-dose actinomycin D with multiday methotrexate therapy for treating low-risk gestational trophoblastic neoplasia patients failed to meet the expected case number. Through this multicentre study, the complete remission ratio and efficacy difference between biweekly single-dose actinomycin D and multiday methotrexate therapy will be obtained. This study will also provide the basis for formulating a preferred regimen for treating patients with low-risk gestational trophoblastic neoplasia. Objective: To find the material basis of antioxidant activity of Sedum aizoon L. based on the study of spectrum-effect relationship, which provided a certain reference for its quality control. Method: The fringerprint of Sedum aizoon L. from different areas were established by high performance liquid chromatography-electrochemical detection (HPLC-ECD) and the similarity of the samples were evaluated. At the same time, the spectrum-effect relationship between HPLC-ECD fingerprint of Sedum aizoon L. and antioxidant activity was studied by partial least squares regression analysis and grey correlation analysis. Results: The HPLC-ECD fingerprint of Sedum aizoon L was established. The similarity of the samples was high and 17 common peaks were obtained. Among them, the 12 components were identified with the standard solution, including gallic acid, protocatechuic acid, tryptophan, methyl gallate, epigallocatechin, caffeic acid, epigallocatechin gallate, iriflophene, myricitrin, isoquercitrin, rosmarinic acid and quercitrin. The spectrum relationship showed that the variables important in projection (VIP) scores of gallic acid, protocatechuic acid, epigallocatechin, epigallocatechin gallate, rosmarinic acid and quercitrin were greater than 1. Among them, the grey correlation degree of gallic acid, rosmarinic acid and quercitrin was nearly 0.8. Furthermore, epigallocatechin gallate, myricitrin, isoquercitrin, rosmarinic acid and quercitrin had higher libdock score in molecular docking. Therefore, epigallocatechin gallate, myricitrin, isoquercitrin, rosmarinic acid and quercitrin were the primary components contributing to the antioxidant activity of Sedum aizoon L.. Conclusion: The basis of antioxidant activity of Sedum aizoon L. was determined by the study of spectrum effect relationship with chemometrics, which provides a certain scientific basis for the quality evaluation of Sedum aizoon L..

Oilseed rape (Brassica napus L.) is an allotetraploid (AACC, 2n ​= ​38) crop, valued for its edible oil and protein content. seed yield and nutritional composition of rapeseed are influenced by its yield and oil quality traits. However, the genetic basis of yield-related and oil-quality traits remain ambiguous. A panel of 266 diversified oilseed rape accessions was genotyped using 223 simple sequence repeat (SSR) markers covering all 19 chromosomes to identify significant markers associated with yield and quality traits. To achieve a hierarchical and quantitative evaluation of grid-wide power quality in the distribution network, reflecting the overall power quality level of the distribution network, a comprehensive evaluation method for power quality in a grid-wide system based on complex network theory is proposed. Firstly, based on the propagation characteristics of power quality disturbances, a power quality evaluation index system is constructed. Secondly, to reflect the constraint effect of the local power quality level of nodes on the overall power quality level of the distribution system, corresponding indices such as improved node degree, improved node electrical betweenness, and node self-healing capability are proposed based on complex network theory, and the power quality influence degree of nodes is calculated. Then, the GRA-ANP (Grey Relational Analysis–Analytic Network Process) subjective weight calculation method is improved by introducing grey relational analysis to address the impact of differences in different decision-making results. Based on power quality monitoring data, the entropy weight method is used for objective weighting. To avoid the partiality of a single weight evaluation result, the game equilibrium algorithm is employed to calculate the comprehensive weight of each power quality index. Subsequently, considering the correlation and dependency among indices, the VIKOR (VIseKriterijumska Optimizacija I Kompromisno Resenje) method is used to obtain the power quality grade of each node. Combining this with the calculation of the power quality influence degree of nodes, the overall power quality grade of the distribution network is determined, achieving a hierarchical and quantitative evaluation of power quality in the entire distribution system. Finally, through a case study analysis of an improved 13-node distribution network, it is verified that the proposed method can fully extract data information and produce comprehensive and accurate power quality assessment results by comparing it with other methods. This provides strong support for the safe and stable operation of the distribution system and the subsequent optimization and management of power quality.

Abstract A growing body of evidence links gut microbiota changes with inflammatory bowel disease (IBD), raising the potential benefit of exploiting metagenomics data for non-invasive IBD diagnostics. The sbv IMPROVER metagenomics diagnosis for inflammatory bowel disease challenge investigated computational metagenomics methods for discriminating IBD and nonIBD subjects. Participants in this challenge were given independent training and test metagenomics data from IBD and nonIBD subjects, which could be wither either raw read data (sub-challenge 1, SC1) or processed Taxonomy- and Function-based profiles (sub-challenge 2, SC2). A total of 81 anonymized submissions were received between September 2019 and March 2020. Most participants’ predictions performed better than random predictions in classifying IBD versus nonIBD, Ulcerative Colitis (UC) versus nonIBD, and Crohn’s Disease (CD) versus nonIBD. However, discrimination between UC and CD remains challenging, with the classification quality similar to the set of random predictions. We analyzed the class prediction accuracy, the metagenomics features by the teams, and computational methods used. These results will be openly shared with the scientific community to help advance IBD research and illustrate the application of a range of computational methodologies for effective metagenomic classification.

AbstractBackground The prognostic factors for patients with pure ovarian immature teratoma (POIT) and the role of adjuvant chemotherapy in stage IA G2-G3 and IB-IC POIT remains controversial.Methods We conducted a retrospective study of 155 POIT patients treated in our hospital between 2000 and 2022. The recurrence-free survival (RFS), disease-specific survival (DSS), and potential prognostic factors of POIT patients were evaluated. Subgroup analysis was conducted in stage I other than stage IA G1 POIT.Results The median age at diagnosis was 23.0 years (range: 4.0 − 39.0), and 126 (81.3%), 2 (1.3%), 26 (16.8%), and 1 (0.6%) patients had FIGO stage I, stage II, stage III, and stage IV disease, respectively. Twenty-three patients relapsed and five died of the diseases after a median follow-up of 7.6 years, with a 5-year RFS and DSS rate of 86.0% and 97.0%, respectively. Multivariate analysis showed that positive postoperative tumour markers (TM) were the risk factor for recurrence in the overall cohort (hazard ratio [HR] 4.058, 95% CI 1.175 − 14.019, p = 0.027) and subgroup (HR 10.237, 95% CI 2.175 − 48.179, p = 0.003), and FIGO stage II–IV was the only factor for DSS in overall cohort (HR 7.751, 95% CI 1.281 − 46.895, p = 0.026). In 110 patients subjected to subgroup analysis, 29 patients received surveillance without chemotherapy and 81 patients were administered adjuvant chemotherapy. Fractional vegetation coverage (FVC) is an important ecological parameter reflecting the growth of regional plants. Existing FVC estimation is often based on vegetation indices, especially the Normalized Difference Vegetation Index (NDVI). However, NDVI can be oversaturated and easily affected by problems such as ‘shadow’ in images, which leads to a decrease in the precision of FVC estimation. In this study, the Normalized Shaded Vegetation Index (NSVI) was used to comprehensively compare the estimation ability of FVC with NDVI, and the differences in FVC estimation ability between NSVI and NDVI were explored. Based on two dimensions of ‘bright’ and ‘shadow’ hierarchies and FVC ranks, four evaluation systems of signal-to-noise ratio (SNR), index range, statistical model and dimidiate pixel model were selected from Sentinel-2A MSI, Landsat-8 OLI and Resource-1-02D (ZY1-02D) images and covered a variety of topographic landscapes. The results showed that: (1) the ability of NSVI to resist soil background is slightly smaller than NDVI; (2) the range of NSVI and NDVI in hyperspectral images is slightly larger than multispectral images, and the ability of NSVI to detect vegetation information in medium-high-rank and high-rank FVC areas has obvious advantages; (3) in the same kind of regression model, the goodness of fit of NSVI, NDVI and FVC was slightly higher in bright areas than in shaded areas, and the goodness of fit of the five regression models obtained from NSVI showed a significant advantage in bright areas compared with NDVI, with the best fit of the cubic curve model; (4) the estimation accuracy of the dimidiate pixel model based on NSVI and NDVI is slightly higher in bright areas than in shaded areas, and the estimation ability of NSVI is slightly better than that of NDVI in bright areas with medium-high-rank and high-rank FVC. NSVI and NDVI have their own advantages in SNR, index range, statistical model and dimidiate pixel model, so it is suggested that when remote sensing estimation of FVC is carried out, NSVI should be preferred in medium-high-rank and high-rank FVC areas, and NDVI should be preferred in low-rank FVC areas in shaded areas.

The imperfection of measurements in real-world scenarios can compromise the performance of device-independent quantum key distribution (DIQKD) protocols. In this paper, we focus on a specific DIQKD protocol that relies on the violation of the Svetlichny’s inequality (SI), considering an eavesdropper utilizing the convex combination attack. Our analysis covers both the three-party DIQKD case and the general n-party scenario. Our main result is the relationship between the measurement accuracy and the extractable secret-key rate in all multi-party scenarios. The result demonstrates that as measurement accuracy improves, the extractable secret-key rate approaches 1, reaching its maximum value when the measurement accuracy is perfect. We reveal that achieving positive extractable secret-key rates requires a threshold measurement accuracy that is consistently higher than the critical measurement accuracy necessary to violate the SI. We depict these thresholds for n-party scenarios ranging from n=3 to n=10, demonstrating that as the number of parties (n) increases, both thresholds exhibit a rapid and monotonic convergence towards unity. Furthermore, we consider a scenario involving a non-maximally entangled state with imperfect measurements, where the emission of the initial GHZ state undergoes noise during transmission, resulting in a Werner state. The study further quantifies and demonstrates the relationship between the extractable secret-key rate, the visibility of the Werner state, and the measurement accuracy, specifically emphasizing the three-party scenario. This study aims to illuminate the influence of imperfect measurement accuracy on the security and performance of multi-party DIQKD protocols. The results emphasize the importance of high measurement accuracy in achieving positive secret-key rates and maintaining the violation of the SI.

Objective: To clarify the effect of controlled atmosphere on preservation of water bamboo. Methods: Cold storage under air was used as control, the sensory quality, texture （firmness, chewiness and shear force）, color （L* value and WH value）, the contents of soluble protein, lignin and cellulose, as well as the activities of phenylalanine ammonia lyase （PAL） and polyphenoloxidase （PPO） were determined under four different gas components （CA1: 2% O2+7% CO2+91% N2, CA2: 5% O2+10% CO2+85% N2, CA3: 5% O2+7% CO2+88% N2, CA4: 2% O2+10% CO2+88% N2）, then stored at 0 ℃ with the relative humidity （RH） of 85% ~ 90%. Results: Controlled atmosphere （CA） treatment could significantly slow down the decline of sensory quality, L* value and WH value of water bamboo and the consumption of soluble protein （P＜0.05）. Among these CA treatments, the effects of CA2 treatment on preservation was the best. After 90 d storage, the PAL and PPO activities of CA2 treatment were significantly lower than those of control group, CA1 and CA3 （P＜0.05）, but there was no significant difference with CA4. Moreover, the accumulation of lignin and cellulose were inhibited, but the inhibition effect of CA2 treatment on lignin and cellulose were lower than that of CA3. The firmness and chewiness of CA1 treatment were significantly higher than other treatments （P＜0.05）, but the shear force was lower than that of CA2. Conclusion: The gas components of 5% O2+10% CO2+85% N2 effectively inhibited the quality deterioration of water bamboo, showing the best effect on commodity quality after 90 days of storage, providing theoretical basis and technical support for storage of water bamboo.

The United States' large-scale poultry meat industry is energy and water intensive, and opportunities may exist to improve sustainability during the broiler chilling process. By USDA regulation, after harvest the internal temperature of the chicken must be reduced to 40°F or less within 16 h to inhibit bacterial growth that would otherwise compromise the safety of the product. This step is accomplished most commonly by water immersion chilling in the United States, while air chilling methods dominate other global markets. A comprehensive understanding of the differences between these chilling methods is lacking. Therefore, we assessed the meat quality, shelf-life, microbial ecology, and techno-economic impacts of chilling methods on chicken broilers in a university meat laboratory setting. We discovered that air chilling methods resulted in superior chicken odor and shelf-life, especially prior to 14 days of dark storage. Moreover, we demonstrated that air chilling resulted in a more diverse microbiome that we hypothesize may delay the dominance of the spoilage organism Pseudomonas Finally, a techno-economic analysis highlighted potential economic advantages to air chilling compared to water chilling in facility locations where water costs are a more significant factor than energy costs.IMPORTANCE As the poultry industry works to become more sustainable and to reduce the volume of food waste, it is critical to consider points in the processing system that can be altered to make the process more efficient. In this study, we demonstrate that the method used during chilling (air versus water chilling) influences the final product microbial community, quality, and physiochemistry. Notably, the use of air chilling appears to delay the bloom of Pseudomonas spp. that are the primary spoilers in packaged meat products. By using air chilling to reduce carcass temperatures instead of water chilling, producers may extend the time until spoilage of the products and, depending on the cost of water in the area, may have economic and sustainability advantages. As a next step, a similar experiment should be done in an industrial setting to confirm these results generated in a small-scale university lab facility.

Although high-throughput RNA sequencing (RNA-seq) has greatly advanced small non-coding RNA (sncRNA) discovery, the currently widely used complementary DNA library construction protocol generates biased sequencing results. This is partially due to RNA modifications that interfere with adapter ligation and reverse transcription processes, which prevent the detection of sncRNAs bearing these modifications. Here, we present PANDORA-seq (panoramic RNA display by overcoming RNA modification aborted sequencing), employing a combinatorial enzymatic treatment to remove key RNA modifications that block adapter ligation and reverse transcription. PANDORA-seq identified abundant modified sncRNAs-mostly transfer RNA-derived small RNAs (tsRNAs) and ribosomal RNA-derived small RNAs (rsRNAs)-that were previously undetected, exhibiting tissue-specific expression across mouse brain, liver, spleen and sperm, as well as cell-specific expression across embryonic stem cells (ESCs) and HeLa cells. Using PANDORA-seq, we revealed unprecedented landscapes of microRNA, tsRNA and rsRNA dynamics during the generation of induced pluripotent stem cells. Importantly, tsRNAs and rsRNAs that are downregulated during somatic cell reprogramming impact cellular translation in ESCs, suggesting a role in lineage differentiation.

This study was conducted to evaluate the effect of red macroalgae Asparagopsis taxiformis supplementation for cattle on the shelf life of fresh beef steaks (longissimus dorsi). Three treatment groups (seven steers per treatment) included: 1) Control diet, 2) Control diet + 0.25% of macroalgae inclusion (low dose, LD), and 3) Control + 0.5% of macroalgae inclusion (high dose, HD). After the animals were harvested, the strip loins from all animals were collected and aged for 14 days at the meat lab of the University of California, Davis. Then the strip loins were cut into steaks, packaged, and placed on a retail display case for 6 days. During a retail display, instrumental color (L*, a*, and b*) of lean muscle and external fat surfaces were measured every 12 h. Bacterial counts for total aerobic mesophilic bacteria (AMB), aerobic psychrotrophic bacteria (APB), and lactic acid bacteria (LAB) were assessed on days 0, 3, and 6 of retail display. The thiobarbituric acid reactive substances (TBARS) analysis was conducted to measure the lipid oxidation and the pH was also assessed on days 0, 3, and 6. No interactive effect between treatments and time on the shelf life of steaks was observed. The HD red macroalgae supplement decreased (P < 0.05) the lightness (L*) of the surface muscle of the steaks, while the lightness of the external fat was not affected (P < 0.05) by treatments throughout the retail display. The external fat yellowness of the steaks was lower (P < 0.05) in LD and HD treatment groups compared with the control group. An increase (P < 0.05) in counts of AMB, APB, and LAB was observed on the steaks from the steers in the HD treatment group while steaks in Control and LD group had similar bacterial numbers throughout the retail display. The results indicated that the shelf life of steaks from cattle in LD group remained the same as that of the Control group, but the HD of A. taxiformis caused a darker color of steaks with higher microbial counts, which may lead to a shortened shelf life due to undesirable appearance and faster microbial spoilage.
