OBJECTIVE: Our objective was to examine the association between single nucleotide polymorphisms of interleukin (IL)-8 (rs4073 and rs2227306) and IL-10 (rs1800871 and rs1800872) genes, and clinical effects of transcatheter arterial chemoembolization (TACE) and subsequent prognosis in patients with liver cancer. PATIENTS AND METHODS: 115 patients with liver cancer underwent TACE. Venous blood specimens were collected for genomic DNA extraction. The restriction fragment length polymorphism-polymerase chain reaction (RFLP-PCR) analysis was used to detect the above variants of IL-8 and IL-10 genes. In addition, blood levels of alpha fetal protein (AFP) were quantified by radioimmunoassay. Patients were followed up to uncover the association of the above genotypes with treatment efficacy and survival. RESULTS: Patients with the homozygous genotype AA or homozygous genotype TT (respectively, -251 and +781 sites) of IL-8 gene, and wild-type genotype TT or homozygous genotype AA (respectively, -819 and -592 sites) of IL-10 gene showed the best effectiveness of TACE. Furthermore, these patients also exhibited the lowest AFP levels and the longest survival after the treatment. CONCLUSIONS: Clinical efficacy of TACE and patient survival in liver cancer are associated with specific variants of IL-8 and IL-10 genes. [ABSTRACT FROM AUTHOR]

The grass carp, Ctenopharyngodon idella (Valenciennes), is one of the most extensively aquacultured freshwater fish in China. However, because of the lack of effective control measures and the high-density culture environment, considerable economic losses are caused by infection of C. idella with the parasitic ciliate, Ichthyophthirius multifiliis. The major histocompatibility ( MH) DAB gene belongs to antigen-presented genes in the class II genomic region, which is associated with parasite resistance. To understand the relationship of the DAB gene with I. multifiliis infection in grass carp, the expression profiles of MH II-DAB were studied in tissues using real-time quantitative polymerase chain reaction. The results showed that expression of the MH II-DAB gene was up-regulated in head kidney after I. multifiliis infection, and the expression peak appeared earlier in the study (case) group than in the control group. The obvious up-regulation peak of MH II- DAB gene was found at days 2 and 4 in skin; at 12 h to day 4 in spleen; at 12 h and days 1 and 6 in gill; and at day 10 in blood, whereas the MH II-DAB gene was down-regulated in liver and intestines after I. multifiliis infection. The interface between steel bar and concrete plays an important role in retaining the strength of reinforced concrete structures. When the interface is damaged by cracking, de-bonding between the two materials and/or other kinds of damage, significant degradation of the structural performances will occur owing to loss of composite actions. In this study, wave propagation on several steel bars embedded in two concrete plates with different de-bonding scenarios is tested. Piezoelectric actuators and sensors are attached to steel bars for recording input and response signals. This paper uses the wavelet transform combined with the wavelet packet decomposition, component energy and Shannon entropy to analyse the experimental results. The results demonstrate that both damage index and relative entropy are sensitive to the existence of de-bonding damage and change linearly with de-bonding length when there is only a single de-bonding damage, while energy and entropy distribution are largely influenced by the property of the excitation signal. Shear connectors are widely used in composite bridges that provide composite action. Their damage will reduce the load-carrying capacity of the structure. In this study, a novel method based on Kullback-Leibler distance (KLD) was developed to assess the integrity of the shear connectors. A bridge model was constructed in the laboratory and some removable anchors were specially designed and fabricated to link the beams and slab that were cast separately. Each anchor consists of a threaded bar that penetrates through the soffit of the beam and ties up into an embedded nut cap to simulate a shear connector in the real bridges. Different damage scenarios were introduced by pulling out some connectors. Vibration tests were carried out in each damage scenario. Various damage detection methods have been applied and results show that the method was able to detect all the assumed damage scenarios successfully and consistently. [ABSTRACT FROM AUTHOR]

In the present study, the antibodies to Toxoplasma gondii in 191 farm-bred and 83 house-bred geese ( Anser domestica) were assessed for the prevalence of T. gondii infection in southern China with the modified agglutination test. Antibodies to T. gondii (MAT ≥ 1 : 5) were found in 27 (14.14%) of farm-bred geese and 14 (16.87%) of house-bred geese. Chinese chestnut trees ( Castanea mollissima BL.) planted in a suburb of Beijing, China developed symptoms including yellowing, leaf crinkling, little leaf, shortened internodes, and empty burrs. Transmission electron microscopy revealed presence of phytoplasma cells in phloem sieve elements of the symptomatic chestnut trees. Molecular cloning and sequence analysis of PCR-amplified near-full length 16S rRNA gene indicated that the phytoplasma associated with the Chinese chestnut yellow crinkle disease is closely related to Japanese chestnut witches'-broom phytoplasma. Quantitative condition assessment of structures has been traditionally using proof load test leading to an indication of the load-carrying capacity. Alternative approaches using ultrasonic, dynamics etc. are based on the unloaded state of the structure and anomalies may not be fully mobilized in the load resisting path and thus their effects are not fully included in the measured responses. This paper studies the effect of the load carried by a reinforced concrete beam on the assessment result of the crack damage. This assessment can only be performed with an approach based on static measurement. The crack damage is modelled as a crack zone over an area of high tensile stress of the member, and it is represented by a damage function for the simulation study. An existing nonlinear optimization algorithm is adopted. The identified damage extent from a selected high level load and a low load level are compared, and it is concluded that accurate assessment can only be obtained at a load level close to the one that creates the damage. The aim of this study was to investigate the changes in cerebral blood flow velocity (CBFV) and its relation to apolipoprotein E (apoE) ℇ4 allele in mild cognitive impairment (MCI). Thirty MCI and 30 controls were assessed using Mini-Mental State Examination (MMSE) and Cambridge Cognitive Examination-Chinese version (CAMCOG-C), and then insonated in the anterior (ACA), middle (MCA) and basilar (BA) cerebral arteries using transcranial Doppler ultrasonography. Compared with controls, MCI showed significant decreases in the mean ( Vm), systolic ( Vs) and diastolic ( Vd) CBFV, bilaterally in MCA and ACA ( P < 0.05–0.001), but not in BA. Compared with 17 apoE ℇ4 allele non-carriers, 13 carriers in MCI showed significant CBFV decreases, bilaterally in MCA ( P < 0.05–0.001). Our findings, the decreased CBFV in apoE ℇ4 allele carriers with MCI, suggest that a large sample and longitudinal study in CBFV and cognitive changes may have the implications on early diagnosis of Alzheimer's disease. Aims To investigate the prevalence of the Gly482Ser polymorphism of the PPARGC1 gene in a northern Chinese population and to clarify the susceptibility of individuals with the Gly482Ser polymorphism to insulin resistance and related diseases. Methods We studied the association of the Gly482Ser polymorphism identified in the PPARGC1 gene with Type 2 diabetes mellitus (T2DM) in 390 unrelated patients with T2DM and 525 control subjects with normal glucose tolerance. Clinical parameters and measures of insulin resistance were recorded. Genotypes were determined by the polymerase chain reaction—restriction fragment length polymorphism (PCR-RFLP) method, which was further confirmed by direct sequencing in 20 randomly selected cases. Results The Gly482Ser polymorphism was common in the northern Chinese population. Univariate analysis indicated no statistically significant differences in allele frequencies or genotype frequencies of the Gly482Ser polymorphism in diabetic and control subjects (minor 482Ser allele frequency 44.4 vs. 41.4%, P = 0.169). However, logistic regression analysis demonstrated a positive 1.645-fold higher risk of the Ser/Ser genotype for T2DM (P = 0.039, 95% CI = 1.026-2.632). After stratification by gender, the risk of Type 2 diabetes in men was increased 1.852-fold (95% CI = 1.125-3.049) in those with the Ser/X genotype compared with those with the Gly/Gly genotype (P = 0.015). No associations were observed between the Gly482Ser polymorphism and parameters of insulin resistance, obesity and hypertension. Conclusion The Gly482Ser variant of the PPARGC1 gene might contribute to susceptibility to T2DM in northern Chinese subjects. The Ser/X genotype of the Gly482Ser polymorphism in the PPARGC1 gene appears to be a risk factor for T2DM in northern Chinese men. [ABSTRACT FROM AUTHOR]

A new method to identify moving loads on a beam using measured vibration responses is studied for its possible application to identify axle loads when applied to the real bridge/vehicle interaction problem with road surface roughness and incomplete vehicle speed. Results from numerical studies on both a single and multi-span bridges and from laboratory experiment show that the method can identify individual axle loads travelling at non-uniform speed with small error. It can even identify the weight of the vehicle accurately when the vehicle brakes on top of the bridge deck. [Copyright &y& Elsevier]

The dynamic behavior of an orthotropic plate simply supported on a pair of parallel edges and under a system of moving loads is analyzed based on Lagrange equation and modal superposition. Thin plate theory is assumed for the plate model and no restriction is placed on the type of loading. Parameters of the plate affecting its dynamic behavior are discussed, and a new classification of the plates for computing the mode shapes and natural frequencies is proposed. The impact factors and the dynamic responses of a typical bridge deck are studied using the proposed method. Preliminary results indicate that the effect of eccentric loads on the impact factor depends on the proportion ratio between the flexural and torsional rigidities of the bridge deck, and the multilane loading case is less critical than a single-lane loading case. Several methods have been developed in recent years to identify moving loads on top of a continuous beam using measured vibration responses. The methods can identify the forces with some accuracy, but they have not been tested under field measurement conditions with a bridge–vehicle system. This paper discusses the weaknesses and merits of two methods when applied to a single-span bridge deck. The influence, on the moving load identification, of practical aspects such as measurement noise, sampling frequency, a small number of measured response modes, a small number of measuring points, road surface roughness and non-uniform velocity or braking of vehicle is studied in simulations and experiment. Results show that finite element approach with orthogonal function approximation of the responses give more accurate results, in general, than the exact solution approach for all the studies presented in this paper. The road surface roughness and a large variation in the speed are identified as the two main obstacles leading to erroneous results. [Copyright &y& Elsevier]

The dynamic loading on a multi-lane continuous bridge deck due to vehicles moving on top at a constant velocity is investigated. The bridge is modelled as a multi-span continuous orthotropic rectangular plate with line rigid intermediate supports. The vehicle is simulated as a two-axle three-dimensional vehicle model with seven degrees of freedom according to the H20-44 vehicle design loading (AASHTO LRFD Bridge Design Specifications 1998 American Association of State Highway and Transportation Officials [1]). The dynamic behavior of the bridge deck under single and several vehicles moving in different lanes is analyzed using the orthotropic plate theory and modal superposition technique. The dynamic loading is studied in terms of the dynamic impact factor of the bridge deck. The impact factor is found varying in an opposite trend as the dynamic responses for the different loading cases under study. [Copyright &y& Elsevier]

This study examined genetic variability within Spirometra erinacei (Cestoda: Pseudophyllidea) from different host species and geographical origins in Australia using a polymerase chain reaction (PCR)-based mutation detection approach, followed by DNA sequencing. Part of the cytochrome c oxidase subunit 1 gene (p cox 1) was amplified by PCR, scanned for sequence variation by single-strand conformation polymorphism (SSCP), and representative samples from different host species were selected for DNA sequencing. While no variation in SSCP profiles was detected among S. erinacei samples from dog, fox, cat, tiger snake and python, they differed in profile from 5 specimens from the green tree frog (Litoria caerulea). This was supported by sequence data which demonstrated that p cox 1 sequences of samples from the latter host species differed at 8 of 393 (2%) nucleotide positions from those from the non-amphibian host. Using a nucleotide difference in the p cox 1 sequence, a PCR-linked restriction fragment length polymorphism (RFLP) could be employed to unequivocally delineate between samples from non-amphibian and amphibian hosts. These findings demonstrate the existence of at least two genotypes within S. erinacei, which may have important implications for studying the epidemiology, ecology and systematics of this cestode. The objective of the present investigation was to determine the seroprevalence of Coxiella burnetii infection in free-range yaks in China. A total of 552 serum samples were collected from yaks in Gansu province, northwest China between April 2013 and January 2014, and antibodies against C. burnetii were evaluated using enzyme-linked immunosorbent assay (ELISA). Overall, 13.59% (75/552, 95% CI: 10.73-16.45) of the examined animals were positive for C. burnetii antibodies. There was no significant difference in C. burnetii seroprevalence between female yaks (13.78%, 95% CI: 10.36-17.19) and male yaks (13.13%, 95% CI: 7.89-18.36) (P>0.05). Coxiella burnetii seroprevalence in yaks in different age groups ranged from 10.88% to 15.26%, but the difference was not statistically significant (P>0.05). Coxiella burnetii seroprevalence in yaks sampled in different seasons ranged from 12.06% (autumn) to 18.33% (summer), but the difference was not statistically significant (P>0.05). This is the first report of C. burnetii seroprevalence in free-range yaks in China, indicating the need for measures to be taken to control C. burnetii infection in free-range yaks in China.
