The paired box domain gene 5 (PAX5) is frequently altered in both childhood and adult B-cell acute lymphoblastic leukemia, and takes part in leukemogenesis. We analyzed data from the database of Gene Expression Omnibus (accession number: GSE11877) using bioinformatical and statistical methods. The results showed that cases of PAX5 alteration can cluster using unsupervised clustering algorithms, and one gene, zinc and ring finger 1 (ZNRF1), was characterized and validated by quantitative RT-PCR. ZNRF1 may be associated with leukemogenesis of ALL with PAX5 alteration.

The constitutive equation of semi-solid 6061 alloy was established according to the thermal simulation compression tests of semi-solid billet of 6061 alloy prepared by near-liquids semi-continuous casting method. Parameters of Bingham model, such as apparent viscosity, had been investigated based on the constitutive equation of semi-solid 6061 alloy. The software ANYCASTING was applied to simulate and analyze the semi-solid die-casting processes of semi-solid 6061 alloy. A well-designed runner and gating system of automobile aluminum alloy part were presented and the casting defect were investigated here utilizing these parameters by simulation. The results show that a well-designed runner and gating system is very important to secure good quality die castings through providing a homogenous mould filling pattern. Most defects of semi-solid casting products can be founded in the final solidification of the parts of casting.

We report on the fabrication of ridge waveguides in KTiOPOsub4/subnonlinear optical crystals through carbon ion irradiation followed by precise diamond blade dicing. The diced side-walls have low roughness, which allows for low propagation loss of ~1dB/cm in fabricated of ridges. The waveguide property investigation has been performed at 1064 nm as well as 532 nm, showing good guidance at both TE and TM polarizations. Based on type II phase matching configuration, efficient second harmonic generation of green light at room temperature has been realized. High conversion efficiencies of ~1.12%Wsup-1/supand ~12.4% have been obtained for frequency doubling under the pump of continuous-wave (CW) and pulsed fundamental waves at 1064 nm, respectively.

The nonlinear optical property of topological insulator bismuth selenide (Bi2Se3) is found to be well-tailored through ion irradiation by intentionally introducing defects. The increase of the optical modulation depth sensitively depends on the careful selection of the irradiation condition. By implementing the ion irradiated Bi2Se3 film as an optical saturable absorber device for the Q-switched wave-guide laser, an enhanced laser performance has been obtained including narrower pulse duration and higher peak power. Our work provides a new approach of tailoring the nonlinear optical properties of materials through ion irradiation, a well-developed chip-technology, which could find wider applicability to other layered two-dimensional materials beyond topological insulators, such as graphene, MoS2, black phosphours etc.

We report on the thermally manipulable waveguide laser emission by integrating the ultra-thin vanadium dioxide membrance with the waveguide as the saturable absorber. Combined with the study of unique optical features of the vanadium dioxide, the waveguide laser exhibits the significant laser operation switching between the picosecond laser and the continuous-wave laser. The saturable absorption of the vanadium dioxide exhibited the thermal hysteresis. Hence the continuous and picosecond laser can exist at the same temperature and can be respectively excited by the cooling and heating treatment.

Concomitance of the FLT3-ITD mutation is associated with poor prognosis in NPM1-mutated cytogenetically normal acute myeloid leukemia (CN-AML) patients, and precise studies on its role in leukemogenesis are needed; these may be elucidated at the molecular level by gene express profiling. In the present study, we built a gene-expression-based classifier using prediction analysis of microarray to characterize the FLT3-ITD signature in NPM1-mutated CN-AML patients, which comprised 10 annotated genes, and demonstrated an overall accuracy of 83.8 % in cross-validation. To characterize the signature in another way, differential expression was revealed for 34 genes by class comparison, and the up-regulation of LAPTM4B and MIR155HG was validated by quantitative RT-PCR in our small cohort of NPM1-mutated CN-AML samples, which appeared to be associated with this specific subtype. The 10-gene classifier and differentially expressed genes identified in this study indicate a potential utility for risk-assessed treatment stratification, and suggest new therapeutic targets for these high-risk AML patients.

Novel analytic tools are needed to elucidate the molecular basis of leukemia-relevant gene mutations in the post-genome era. We generated isogenic leukemia cell clones in which the FLT3 gene was disrupted in a single allele using TALENs. Isogenic clones with mono-allelic disrupted FLT3 were compared to an isogenic wild-type control clone and parental leukemia cells for transcriptional expression, downstream FLT3 signaling and proliferation capacity. The global gene expression profiles of mutant K562 clones and corresponding wild-type controls were compared using RNA-seq. The transcriptional levels and the ligand-dependent autophosphorylation of FLT3 were decreased in the mutant clones. TALENs-mediated FLT3 haplo-insufficiency impaired cell proliferation and colony formation in vitro. These inhibitory effects were maintained in vivo, improving the survival of NOD/SCID mice transplanted with mutant K562 clones. Cluster analysis revealed that the gene expression pattern of isogenic clones was determined by the FLT3 mutant status rather than the deviation among individual isogenic clones. Differentially expressed genes between the mutant and wild-type clones revealed an activation of nonsense-mediated decay pathway in mutant K562 clones as well as an inhibited FLT3 signaling. Our data support that this genome-editing approach is a robust and generally applicable platform to explore the molecular bases of gene mutations.

We report on a cladding-like waveguide structure in Nd:YAG crystal fabricated by the multiple carbon ion beam irradiation. After the designed multiple irradiation process, the cladding-like waveguide with triple refractive-index layers were constructed in the region near the surface of the crystal. With such a structure, the waveguiding core was compressed and refractive index profile was modified, resulting in a higher light intensity than that of the single ion-beam-irradiated monolayer waveguide. The waveguide lasing at wavelength of 1064 nm was achieved with enhanced performance in the cladding-like structures with both planar and ridge configurations by the optical pump at 810 nm.

Effects of deformation temperature and strain rate on deformation behavior of semi-solid ZL201 alloy were investigated on Gleeble3800 thermal-mechanical simulator. On basis of the experimental data of semi-solid ZL201, a model of was established to describe the relationship between the processing parameters and flow stress, which showed that the experimental results and calculation ones fitted well. The suitable conditions of this model for semi-solid ZL201 alloy were that the temperature range was 547°C-649°C and that of the strain rate was 0.1 s-1-5s-1.

Parameters of Bingham model had been verified according to the thermal simulation compression tests of semi-solid 6061 aluminum and the equations presented by Stefan. The effects of the mold filling temperature of semi-solid slurry of 6061 aluminum during thixo-diecasting were analyzed. The results showed that such conditions of the mold filling temperature about 625°C, the injection speed changing to 2 m/s from 0.4 m/s when 60% of die cavity had been filled, were ideal to gain solidification in sequence and beneficial to get fully filled semi-solid parts with clear contour. The thixo-diecasting experiments were performed under above optimized conditions.

Combined with the homogenization method and the least squares method, the filter function with the form of power function is studied in the Independent Continuum Map (ICM) method. Then, the numerical simulation method is used to study the relationship between a weight filter function and a stiffness filter function. Finally, the impact of the filtration function on the efficiency of topology optimization is showed by examples. It is important to further research structural topology optimization.

Applying man-machine-environment system engineering (MMESE) in vessels is a method to improve the effectiveness of the interaction between equipment, environment, and humans for the purpose of advancing operating efficiency, performance, safety, and habitability of a vessel and its subsystems. In the following research, the life cycle of vessels was divided into 9 phases, and 15 research subjects were also identified from among these phases. The 15 subjects were systemized, and then the man-machine-environment engineering system application model for vessels was developed using the ICAM definition method 0 (IDEF0), which is a systematical modeling method. This system model bridges the gap between the data and information flow of every two associated subjects with the major basic research methods and approaches included, which brings the formerly relatively independent subjects together as a whole. The application of this systematic model should facilitate the application of man-machine-environment system engineering in vessels, especially at the conceptual and embodiment design phases. The managers and designers can deal with detailed tasks quickly and efficiently while reducing repetitive work.

Industrial design is of a great importance for the new economy. The design education scenario is focused in a local region; with the questionnaire survey method is applied. The main issues discussed are concern about the course schema, the quality of staff, the teaching conditions, the international exchanges activities and the future professional career. The causes for existing problems are analyzed based on the data obtained, and general suggestions are proposed also.

The corrosion behaviors and mechanisms of 310S and AL-6XN stainless steels in MgCl2 molten salts were investigated at 800°C for 8-72h. The weight change kinetics, surface and cross-sectional morphologies as well as corrosion products and compositions were analyzed. The results showed that corrosion velocities of both steels were very high in the preliminary period, then diminished quickly and trended to zero afterward. The AL-6XN steels exhibited better corrosion resistance in MgCl2 molten salts than 310S. The predominant peaks of MgO were detected from both steels after corrosion in molten MgCl2 at 800°C, which could serve as the protective barriers between the alloy and the MgCl2 molten salts. The depletion of Cr was very serious throughout the corrosion layer for both steels due to their preferential oxidation for the formation of an outer corrosion layer. The other elements in alloy were corroded in succession through the outward diffusion with the progress of the corrosion. The outward migrations of the metal ions led to the coalescence of vacancies to form the voids. The two steels possessed the uniform corrosion behavior.

The Cu-10Cr-0.4Zr in situ composite microwires were prepared by cast and cold drawing procedure. Deformation processing and mechanical properties of Cu-10Cr-0.4Zr composites were investigated. The results showed that the additional 0.4wt. %Zr in the Cu-10Cr in situ composite microwires gave birth to smaller as-cast Cr phases, which led to refined filaments in the matrix at higher drawing strains. As the drawing strains increased, the Cr filaments were constrained to fold or twist (even overlapped together) on longitudinal sections, and the Cr filaments become homogeneity and refinement at the longitudinal sections at the same time. At η=6.2, the thickness of Cr filaments reached 250-300nm, and the ultimate strength of Cu-10Cr-0.4Zr composites reached 1089 MPa. And the predicted strength using Hall-Petch equation was 1037 MPa, which was in reasonably good agreement with the observed strength (1089 MPa).

The 6061 semi-solid aluminium alloy feedstocks prepared by near-liquidus casting were compressed in semi-solid state by means of Gleeble-3500 thermal-mechanical simulator. The relationship between the true stress and the true strain at different temperatures and strain rates was studied with the deformation degree of 70%. The microstructures during the deformation process were characterized. The deformation mechanism and thixo-forming properties of the semi-solid alloys were analyzed. The results show that the homogeneous and non-dendrite microstructures of semi-solid 6061Al alloy manufactured by near-liquidus casting technology could be transformed into semi-solid state with the microstructure suitable for thixo-forming which are composed of near-spherical grains and liquid phase with eutectic composition through reheating process. The deformation temperature and strain rate affect the peak stress significantly rather than steady flow stress. The resistance to deformation in semi-solid state decreases with the increase of the deformation temperature and decrease of the strain rate. At steady thixotropic deformation stage, the thixotropic property is uniform, and the main deformation mechanism is the rotating or sliding between the solid particles and the plastic deformation of the solid particles.

It is a complex but important issue to have a shower for the sailors in a luxury passenger liner whose working and living staff are all to be held in a closed environment. It is hard to decide what kind of showering mode can satisfy the sailors’ showering need, as well as using discrete event theory to handle it because of the mass of people and their behavior’s randomicity. A method based on the theory of discrete event to simulate the showering mode in the ship has been brought out by using the software of QUEST, and the design of the bath system of the ship has been proved to be with usability after simulations. What factors and how they affect the performance of the showering system have been found. At last, an implementation suggestion has been brought forward.

The service in dining-room for the sailors is a very important issue in a luxury passenger liner which affects the work’s performance, so as well as the ship’s. There has not too much research about this topic in China right now. A method to simulate and analyze the dining-room servicing mode which was based on the discrete event system using the software QUEST has been brought out, and used to estimate a design project by analyzing the six factors which affect the servicing mode while releasing a final implement suggestion.

To study the effect of flavonoids isolated from aerial parts of Scutellaria baicalensis Georgi (SSF) on cerebral damage induced by okadaic acid (OA) in rats.OA was microinjected into the right lateral ventricle of male rats at a dose of 200 ng kg(-1) twice with a 3-day interval between injections to establish a model of Alzheimer's-disease-like cerebral damage. Neuronal morphology was observed with thionin staining and the expressions of glial fibrillary acidic protein (GFAP) and β-amyloid peptide 1-40 (Aβ1-40) were monitored via immunohistochemistry. The level of malondialdehyde (MDA) and the activities of glutathione peroxidase (GSH-Px) and lactate dehydrogenase (LDH) were measured using spectrophotometry.The results showed that OA-treated rats exhibited marked neuronal damage accompanied by increased levels of Aβ1-40 peptide and MDA accumulation, decreased GFAP protein expression and reduced GSH-Px and LDH activity in the brain. SSF at three doses (25, 50 and 100 mg kg(-1)) dramatically reversed the OA-induced changes in the brains of rats.SSF-mediated amelioration of OA-induced neuronal damage in rats provides a rationale for assessing SSF as a means of to reducing tau hyperphosphorylation and Aβ expression in the treatment of Alzheimer's disease.

It has recently been shown that JARID2 contributes to the malignant character of solid tumors, such as epithelial-mesenchymal transition in lung and colon cancer cell lines, but its role in leukemia progression is unexplored. In this study, we explored the effect and underlying molecular mechanism of JARID2 on leukemia cell proliferation. Real-time PCR and Western assay were carried out to detect JARID2 and CCND1 expression. Cell number and cell cycle change were detected using hemocytometer and flow cytometry, and a ChIP assay was utilized to investigate JARID2 and H3K27me3 enrichment on the CCND1 promoter. JARID2 is down-regulated in B-chronic lymphocytic leukemia (B-CLL) and acute monocytic leukemia (AMOL), and knockdown of JARID2 promotes leukemia cell proliferation via acceleration of the G1/S transition. Conversely, ectopic expression of JARID2 inhibits these malignant phenotypes. Mechanistic studies show that JARID2 negatively regulates CCND1 expression by increasing H3K27 trimethylation on the CCND1 promoter. Our findings indicate that JARID2 is a negative regulator of leukemia cell proliferation, and functions as potential tumor suppressor in leukemia.

We report on the first experimental study of the layer-to-layer compression and enhanced optical properties of few-layer graphene nanosheet by applying ion irradiation. The deformation of graphene layers is investigated both theoretically and experimentally. It is observed that after the irradiation of energetic ion beams, the space between separate graphene layers is reduced due to layer-to-layer compression, resulting in tighter contact of the graphene sheet with the surface of the substrate. This processing enables enhanced interaction of the graphene with the evanescent-field wave near the surface, which induces reinforced polarization-dependent light absorption of the graphene. Utilizing the ion-bombarded graphene nanosheets as saturable absorbers, we have realized efficient Q-switched waveguide lasing with enhanced performance through the interaction of the graphene and evanescent field.

Previous studies reported that the total flavonoids from the stems and leaves of Scutellaria baicalensis Georgi (TFSS) could enhance and improve learning and memory abilities in experimental animals, and reduce the neuronal pathologic alterations induced by some reagents in mice. The present study examined whether TFSS can improve memory dysfunction, neuronal damage, and abnormal free radicals induced by permanent cerebral ischemia in rats. The permanent cerebral ischemic model in rats was produced by bilateral ligation of the common carotid arteries. The influence of permanent cerebral ischemia on learning and memory was determined in the Morris water maze. The neuronal damage in the hippocampus and cerebral cortex was assessed by the neuronal morphologic observations. The contents of malondialdehyde (MDA) and nitric oxide (NO), and the activities of superoxide dismutase (SOD) and catalase (CAT) in the hippocampus and cerebral cortex were measured using thiobarbituric acid, nitrate reductase, xanthine-xanthine oxidase, and ammonium molybdate spectrophotometric methods, respectively. In learning and memory performance tests, cerebral ischemic rats always required a longer latency time to find the hidden platform and spent a shorter time in the target quadrant in the Morris water maze. TFSS 17.5-70 mg.kg(-1) daily orally administered to ischemic rats for 20 d, from day 16-35 after operation differently reduced the prolonged latency and increased swimming time spent in the target quadrant. In neuronal morphologic observations, daily oral TFSS 17.5-70 mg.kg(-1) for 21 d, from day 16-36 after operation markedly inhibited the ischemia-induced neuronal damage. In addition, the increased contents of MDA and NO, and SOD activity, and the decreased activity of CAT in the hippocampus and cerebral cortex induced by cerebral ischemia were differently reversed. The reference drug piracetam (140 mg.kg(-1) per day for 20-21 d) similarly improved impaired memory and neuronal damage but had no significant effects on free radicals in ligated rats. TFSS can improve memory deficits and neuronal damage in rats after permanent cerebral ischemia, which may be beneficial in the treatment of cerebrovascular dementia.

To monitor the temporal and spatial characteristics of particulate matter and water vapor in the aerosol pollution and haze episodes, the polarization Raman Mie lidar system has been developed. The lidar system includes four detection channels and it can measure the extinction coefficient and depolarization ratio of particulate matter as well as water vapor mixing ratio. The extinction coefficient indicates the visibility of atmosphere and it associates with the concentration of particulate matter. The depolarization ratio demonstrates the nonsphericity of particulate matter and is useful to distinguish the dust and pollution aerosol. The water vapor mixing ratio denotes the content of water vapor in the air and it is an important factor to influence of the hygroscopic growth on the pollution aerosol. The lidar system can operate in the automatic and continuous modes through a window on the roof of the observation room regards of the weather, and it takes continuous measurement from 20 November 2013 to 6 February 2014 over Hefei, China. During the experiment, the typical results of particulate matter measured with lidar in clear air, aerosol pollution and haze, and dust episodes are analyzed and given. The lidar observations are also compared with the air quality data and the meteorological data on the ground.

To investigate neural-reproductive hormonal basis of liver yang rising (LYR), liver qi stagnation (LQS) premenstrual syndrome (PMS), and to develop standardized diagnostic criteria for PMS.HPLC, HPLC-MC, ELISA and radioimmunoassay were used to compare levels of serum hormones, plasma neurotransmitters and neurosteroids between LYR PMS patients, LQS PMS patients and healthy controls (30 subjects in each group).Of the measures, all three groups exhibited no significant differences during the follicular phase. In contrast, during the luteal phase, LYR PMS testosterone levels tended to be higher than controls, while dopamine and 5-HT of the LYR PMS group were significantly higher. Conversely, γ-aminobutyric acid in the LYR PMS group was significantly lower than controls (p 0.05). On the other hand, epinephrine and norepinephrine levels in both PMS groups were significantly higher than controls (p 0.05), while pregnenolone and allopregnanolone of LYR and LQS groups were significantly lower than controls, with dehydroepiandrosterone (DHEA) being significantly higher than controls (p 0.05). The ratios of DHEA/allopregnanolone and DHEA/pregnenolone of both PMS groups were significantly higher than the control group, with the LYR PMS group ratios being significantly higher than in the LQS PMS group (p 0.05).The decrease in pregnenolone and allopregnenolone, increase in DHEA, DHEA/allopregnanolone and DHEA/pregnenolone during the luteal phase may be one of the biological bases for anger in LYR PMS patients and depression in LQS PMS patients.

The purpose of this study is to investigate the effects of flavonoid, isolated from aerial parts of Scutellaria baicalensis Georgi (SSF), on memory deficits, neuronal degeneration and abnormal energy metabolism induced by permanent global ischemia in rats. The global ischemia was produced in female Sprague-Dawley rats by permanent occlusion of the bilateral common carotid arteries. The permanent global ischemia in rats resulted in a significantly increased latency of the rat to find the hidden platform and a decreased swimming distance from the target quadrant in the Morris water maze task. The pathological changes in the neurons of ischemic rats, observed in the hippocampus and cerebral cortex, included neuron loss, neuron swelling, nuclear shrinkage or disappearance, neuronophagia and reduced density of Nissl bodies in the neuron. Moreover, the levels of lactate and ATPase activity in ischemic rats were notably increased and decreased, respectively, in the hippocampus and cerebral cortex as compared with sham-operated rats. Daily oral administration of SSF (35 mg/kg, 19-20 days) dramatically reduced the decrease in learning and memory, attenuated neuronal injury and improved abnormality of energy metabolites in rats induced by global ischemia. These findings suggest that SSF may be beneficial for the treatment of vascular dementia.

Based on the independent, continuous and mapping (ICM) method and homogenization method, a research model is constructed to propose and deduce a theorem and corollary from the invariant between the weight filter function and the corresponding stiffness filter function of the form of power function. The efficiency in searching for optimum solution will be raised via the choice of rational filter functions, so the above mentioned results are very important to the further study of structural topology optimization.

SummaryBackground: Although many epidemiologic studies have investigated the methylenetetrahydrofolate reductase (MTHFR) gene polymorphisms and their association with acute lymphoblastic leukemia (ALL), definitive conclusions cannot be drawn. To clarify the effects of MTHFR polymorphisms on the risk of ALL, a meta-analysis was performed in a Chinese population. Materials and Methods: A computerized literature search was carried out in PubMed, the Chinese Biomedicine (CBM) database, China National Knowledge Infrastructure (CNKI) platform, and the Wanfang database (Chinese) to collect relevant articles. Results: A total of 11 articles including 1,738 ALL cases and 2,438 controls were included in this meta-analysis. Overall, a significantly decreased association was found between the MTHFR C677T polymorphism and ALL risk when all studies in Chinese populations were pooled into the meta-analysis. In subgroup analyses stratified by age, ethnicity, and source of controls, the same results were observed in children, in population-based studies, and in people with no stated ethnicity. However, a significantly increased association was also found for MTHFR C677T in hospital-based studies, and for MTHFR A1298C in people with no stated ethnicity. Conclusion: Our results suggest that the MTHFR C677T and A1298C polymorphisms may be potential biomarkers for ALL risk in Chinese populations, and studies with a larger sample size and wider population spectrum are required before definitive conclusions can be drawn.

The aim of this study was to determine whether tumor manipulation enhances cancer cell release from the primary tumor in HCC patients and which surgical approach, open surgery or laparoscopic resection, is superior with respect to preventing tumor cells from scattering in the blood.A total of 26 HCC patients were prospectively randomized to receive either open surgery (n = 14) or laparoscopic surgery (n = 12). Blood samples were obtained at three time points: preoperative, postoperative, and 24 h after surgery. The CD45(-)/CD44(+)/CD90(+) cells were obtained and counted using quantitative flow cytometry. The serum levels of interleukin-6 (IL-6), interleukin-8 (IL-8), and tumor necrosis factor (TNF-α) were also compared between the two groups.There was no significant difference between the laparoscopic and open groups in terms of patient characteristics. The levels of CCSCs increased immediately after surgical manipulation, and the laparoscopy group released fewer tumor cells into the blood stream. The amount of CCSCs in both groups decreased to reach a similar level 24 h after surgery. Both IL-6 and IL-8 increased after surgery, and the mean postoperative increases in IL-6 and IL-8 serum levels were significantly less in the laparoscopic group than in the open group. The TNF-α levels showed no differences at any time point.Our results showed that patients with laparoscopic surgery have lower IL-6, IL-8 secretion and less CTCs, which may suggest an advantage by restricting CTCs release and a preserved immune response. Further studies are needed to investigate the relationship between the number of CCSCs after surgery and long-term survival rates.

Background We conducted a systematic review and meta-analysis to clarify the incidence and risk of cardiotoxicity associated with bortezomib in cancer patients. Methods Databases from PubMed, Web of Science and abstracts presented at ASCO meeting up to July 31, 2013 were searched to identify relevant studies. Eligible studies included prospective phase II and III trials evaluating bortezomib in cancer patients with adequate data on cardiotoxicity. Statistical analyses were conducted to calculate the summary incidence, odds ratio (OR) and 95% confidence intervals (CIs) by using either random effects or fixed effect models according to the heterogeneity of included studies. Results A total of 5718 patients with a variety of malignancies from 25 clinical trials were included in our analysis. The incidence of all-grade and high-grade cardiotoxicity associated with bortezomib was 3.8% (95%CI: 2.6–5.6%) and 2.3% (1.6–3.5%), with a mortality of 3.0% (1.4–6.5%). Patients treated with bortezomib did not significantly increase the risk of all-grade (OR 1.15, 95%CI: 0.82–1.62, p = 0.41) and high-grade (OR 1.13, 95%CI: 0.58–2.24, p = 0.72) cardiotoxicity compared with patients treated with control medication. Sub-group analysis showed that the incidence of cardiotoxicity varied with tumor types, treatment regimens and phases of trials. No evidence of publication bias was observed. Conclusions The use of bortezomib does not significantly increase the risk of cardiotoxicity compared to control patients. Further studies are recommended to investigate this association and risk differences among different tumor types, treatment regimens and phases of trials.

The best preoperative examination in Graves’ disease with thyroid cancer still remains uncertain. The objectives of the present study were to investigate the prevalence of thyroid cancer in Graves’ disease patients, and to identify the predictive factors and ultrasonographic features of thyroid cancer that may aid the preoperative diagnosis in Graves’ disease. This retrospective study included 423 patients with Graves’ disease who underwent surgical treatment from 2002 to 2012 at our institution. The clinical features and ultrasonographic findings of thyroid nodules were recorded. The diagnosis of thyroid cancer was determined according to the pathological results. Thyroid cancer was discovered in 58 of the 423 (13.7 %) surgically treated Graves’ disease patients; 46 of those 58 patients had thyroid nodules, and the other 12 patients were diagnosed with incidentally discovered thyroid carcinomas without thyroid nodules. Among the 58 patients with thyroid cancer, papillary microcarcinomas were discovered in 50 patients, and multifocality and lymph node involvement were detected in the other 8 patients. Multivariate regression analysis showed younger age was the only significant factor predictive of metastatic thyroid cancer. Ultrasonographic findings of calcification and intranodular blood flow in thyroid nodules indicate that they are more likely to harbor thyroid cancers. Because the influencing factor of metastatic thyroid cancers in Graves’ disease is young age, every suspicious nodule in Graves’ disease patients should be evaluated and treated carefully, especially in younger patients because of the potential for metastasis.

This study was purpose to analyze the frequency and of isocitrate dehydrogenase 2 (IDH2) gene mutation in acute myeloid leukemia (AML) and its clinic significance. The multiplex polymerase chain reaction (PCR) and sequencing were performed to screen 192 AML patients for exon 4 of the IDH2 gene. FLT3, NPM1, CEBPA, c-kit and WT1 mutations were also included in analysis. The results showed that IDH2 mutation was found in 14 (7.29%) of 192 patients. There were 9 AML patients with R140Q mutation, 1 patient with R140W mutation, and 1 patient with R172K mutation. IDH2 aberrations significantly more were detected in French-American-British (FAB) M5 (P0.005) than other types. There was no statistical difference in age, sex, WBC, platelet count, bone marrow blasts count, hemoglobin as compared with IDH2 wild-type. For immunotype analysis, IDH2 mutation patients were more likely to express CD34 and CD13, less CD36. IDH2 mutation combined with FLT3/ITD mutation was found in 7 cases, with CEBPA mutation in 4 cases, with NPM1 mutation in 4 cases, with Dnmt3a mutation in 5 cases, neither with c-kit, IDH1 or WT1 mutation for no one, which revealed a significant interaction between IDH2 mutation and the FLT3/ITD positive genotype, Dnmt3a mutated, and IDH1 wild-type. IDH2 mutation was detected in 5 (8.47%) of 59 CN-AML. There was no significant difference of IDH2 mutation incidence between the normal and abnormal karyotype. The CR rate was higher in IDH2 R140 mutated patients than wild-type ones, but there was no significant in the two group. It is concluded that the rate of IDH2 mutation is 7.29% in Chinese AML patients and 7.81% in CN-AML. IDH2 mutation is significantly associated with AML-M5, FLT3/ITD, Dnmt3a, IDH1 wild-type and fusion gene wild-type, but not with age, leucocyte and platelet counts in peripheral blood, karyotype, NPM1, CEBPA, c-kit or WT1 mutation. And IDH2 R140 mutation has no impact on CR rate.

To investigate the roles of E-cadherin (E-cad) in enhancing the in vitro differentiation of hepatocytes from murine embryonic stem cells (ESCs).Exogenous E-cad was transfected into BALB/c murine ESCs to enable its stable expression. Then hepatic differentiation from E-cad-ESCs was induced by such growth factors as hepatocyte growth factor (HGF), fibroblast growth factor (FGF) and transforming growth factor (TGF). And the expressions of hepatic markers ALB, TAT, Cyp7a1 and urea were detected. The morphology of hepatic differentiation was observed under microscopy.E-cad expression gradually decreased in normal ESC differentiation, but was stably expressed in E-cad-ESCs. In E-cad-ESC group, hepatic markers ALB, TAT and CYP7a1 were expressed earlier or higher than that in normal ESC group, and the concentrations of ALB and urea were significantly higher than that in normal ESC group. The adhesion of the differentiated E-cad-ESCs was significantly enhanced compared with the normal ESCs. They maintained close connections and multidimensional growth. Cell number of hepatocytes from ESC increased significantly in E-cad-ESC group.E-cad enhances the hepatic differentiation of ESC by increasing the number of differentiated cells and increasing the synthetic capacity of ALB and urea.

This study was purposed to detect the mutation of isocitrate dehydrogenase 1 (IDH-1) gene in patients with acute myeloid leukemia (AML) and to explore its clinical significance. The genomic DNA was extracted from mononuclear cells (MNC) of bone marrow or peripheral blood in 205 adult AML patients, the exon 4 of IDH1 gene was amplified by PCR, then the sequencing and comparison were performed. The results showed that IDH1 mutation was detected in 9 (4.39%) of 205 AML patients. There were 6 cases of R132H mutation, 1 of R132L mutation, 1 of R132G mutation and 1 of R132S mutation. Significantly more IDH1 aberrations were detected in AML-M2 (P = 0.002) than other types. And the 9 patients with IDH1 mutation were characterized by low platelet count which was lower than patients with wild type IDH1 (P = 0.003). IDH1 mutation combined with FLT3/ITD mutation was found in 5 cases, c-kit mutation in 1, NPM1 mutation in 2, and IDH1 mutation with CEBPA or WT1 mutation was not found, which revealed a significant interaction between IDH1 mutation and the FLT3/ITD positive genotype or the CEBPA wild-type. IDH1 mutation were detected in 4 of 71 (5.63%) CN-AML. There was no significant difference of IDH1 mutation incidence between the normal and abnormal karyotypes. It is concluded that the rate of IDH1 mutation was 4.39% in Chinese AML patients. IDH1 mutation is significantly associated with AML-M2, lower platelet counts in peripheral blood, FLT3/ITD mutation and CEBPA wild-type, but not with age, white blood cell count in peripheral blood, karyotype, NPM1, c-kit or WT1 mutation.

This work demonstrates the realization of a lasing in scattering media, which contains dispersive solution of ZnO nanoparticles (NPs) and laser dye 4-dicyanomethylene-2-methyle-6-(p-dimethylaminostyryl)-4H-pyran(DCM) in negative liquid crystals (LCs) that was injected into a cell. The lasing intensity of the dye-doped negative LC laser can be tuned from low to high if the NPs concentration is increased. The tunability of the laser is attributable to the clusters-sensitive feature in effective refractive index of the negative LCs. Such a tunable negative liquid crystal laser can be used in the fabrication of new optical sources, optical communication, and liquid crystal laser displays.

This work reports on the passively Q-switched waveguide laser system based on Nd:YAG crystal and MoS2 satuable absorber. A depressed cladding waveguide with circular cross-sectional geometry has been produced in Nd:YAG crystal by direct femtosecond laser writing at low-repetition rate. The confocal microscopic investigation of the structure reveals the well-preserved microphotoluminescence features in the waveguide core. With chemical-vapor-deposition (CVD) MoS2 membrane as satuable absorber, the passive Q-switching of the Nd:YAG waveguide system has been achieved under optical pump, reaching maximum average output power of 85.2 mW, corresponding to single-pulse energy of 112 nJ, at wavelength of 1064 nm. The repetition rate of the pulsed waveguide laser system is tunable from 0.51 to 1.10 MHz, and the obtained minimum pulse duration is 203 ns., The work is supported by the National Natural Science Foundation of China (No. 11274203) and Junta de Castilla y León under Project SA086A12-2. Patients with FLT3-ITD (mut) /NPM1 (-) cytogenetically normal acute myeloid leukemia (CN-AML), as high-risk molecular group in CN-AML, are associated with a worse prognosis than other CN-AML patients. It is beneficial to generate xenotransplantation model of FLT3-ITD (mut) /NPM1 (-) CN-AML to better understand the pathogenesis and therapeutic strategies of such AML subtype. The purpose of present study was to establish the xenotransplantation model in NOD/SCID mice with FLT3-ITD (mut) /NPM1 (-) CN-AML primary cells. The FLT3-ITD (mut) /NPM1 (-) CN-AML primary cells from 3 of 7 cases were successfully transplanted into NOD/SCID mice, and human CD45 positive cells were detected in the peripheral blood, spleen and bone marrow of mice by using flow cytometry. Infiltration of human leukemia cells in various organs of mice was observed by using immunohistochemistry. Gene analysis confirmed sustained FLT3/ITD mutation without NPM1 mutation in mice. By performing serial transplantation, it was found that characteristics of the leukemia cells in secondary and tertiary generation models remained unchanged. Moreover, in vivo cytarabine administration could extend survival of NOD/SCID mice, which was consistent with clinical observation. In conclusion, we successfully established xenotransplantation model of human FLT3-ITD (mut) /NPM1 (-) CN-AML in NOD/SCID mice. The model was able to present primary disease and suitable to evaluate the curative effects of new drugs or therapy strategies.

Recurrent mutations of isocitrate dehydrogenase isoforms 1 and 2 (IDH1 and IDH2) have recently been studied in adult patients with acute myeloid leukemia (AML). A meta-analysis was performed to demonstrate their controversial prognostic impacts. The associations of IDH1 or IDH2 mutations with other molecular abnormalities were also investigated. In patients with AML, IDH1/2 mutations were significantly associated with normal karyotype and isolated trisomy 8 (p < .05). After adjusting for well-studied prognostic factors, IDH1 mutation seems to be associated with subtle but significantly inferior event-free survival (EFS) (p = 0.02) and possible adverse overall survival (OS) (p = 0.13) in patients with AML, especially in the favorable genotype subset with mutated NPM1 but without FLT3-ITD mutation (p < 0.05). Longer OS (p = 0.01) and better EFS tendency (p = 0.18) are implicated in patients with IDH2 mutations, which suggests that IDH2 mutations appear to confer a favorable prognosis. Moreover, IDH1 and IDH2 mutations may play a more important role in abnormal cytogenetics subgroups such as isolated trisomy 8. Screening of IDH1/2 mutations could help to identify patients at high risk within some subsets of AML.

This study was aimed to evaluate IKAROS6 expression in patients with chronic myelogenous leukemia (CML) and its clinical significance. cDNAs from 73 CML patients were amplified by PCR and sequenced for IKAROS expression to elucidate clinical characteristics in IKAROS6 positive patients. The results showed that there was no IKAROS6 gene expression in 8 healthy controls and 15 CML patients in chronic phase and accelerated phase, and 15 cases (35.71%) were IKAROS6 positive in lymphoblast crisis samples among 42 newly diagnosed CML; however, none was found in myeloblast crisis of 16 newly diagnosed CML. Among 42 lymphoblast crisis of CML, the complete remission (CR) rate of IKAROS6 expression positive patients reached 40% (6/15), which was obviously lower than that in IKAROS6 negative patients (85.19%, 23/27) (p0.01), IKAROS6 positive patients relapsed after CR for 15 (2 - 18) months with relapse rate 66.7% (4/6), which was higher than that in expressed wild type IKAROS gene patients (21.74%, 5/23) (p0.05). It is concluded that abnormal expression of IKAROS gene dominated by IKAROS6 isoforms can be detected in lymphoblast crisis samples of CML patients. Abnormal expression of IKAROS gene may be an important factor in lymphoblast crisis of CML. Therefore, detection of IKAROS gene expression may be important for target therapy and evaluation of clinical prognosis of CML patients.

To investigate the diagnostic value of FICTION (Fluorescence Immunophenotyping and Interphase Cytogenetics as a Tool for the Investigation of Neoplasms) technique, combining immunofluorescence and fluorescence in situ hybridization (FISH), to detect genetic aberrations in multiple myeloma (MM).Bone marrow samples were collected from 18 MM and 2 plasma cell leukemia (PCL) patients. Probes targeting IgH and MMSET were prepared using a Nick Translation Kit from Bacterial artificial chromosome (BAC) clones. The immunophenotyping was achieved via the CD138 tyramide signal amplification (TSA)-mediated immunofluorescence, followed by FISH with the prepared probes \[t(4;14), t(11;14), t(14;16)\] and the commercial deletion probes (13q and p53) to detect common genetic aberrations in MM.All the 20 samples were assayed with the probes mentioned above, and revealed 4 cases with t(4;14), 6 with t(11;14), 1 with t(14;16), 3 with p53 deletion; and 8 with 13q deletion. The remaining 4 cases had none of the 5 aberrations.FICTION technique facilitates the detection of genetic abnormalities of MM in situ; enhances both efficiency and sensitivity of positive detection, thus, could be used as the screening test of molecular diagnosis of MM to guide coming-up risk-adapted therapy and evaluate prognosis.

UHF RFID is becoming the main stream of passive RFID applications. Reader is a key component of a RFID application system. Based on the ARM9 chip S3C2440, the UHF reader ASIC AS3990 and a battery charger 9-channel programmable DC-DC power management chip SMB122, a low-power handheld UHF RFID reader is designed and implemented in this paper. In addition to long enough read range, the power consumption is also important for a handheld RFID reader. A low-power control program is analysed and designed in detail in this paper.

Radio Frequency Identification (RFID) has many perfect features such as completely non-contact, high recognition rate, bulk-storage memory, securely access and easily be integrated with existing management information system, etc. It has widely been applied in many fields especially in logistics and product quality tracking systems. The research on the application of RFID in agricultural seed quality management was studied in this paper, which included the cases from the seed's cultivation, storage, transportation to it's quality supervision by related enforcement and supervision departments, even to quality confirmation and plant guide for the last end user - farmer, also analyzed several key technologies for the using of RFID in seed quality tracking system, such as the function design, tag type choosing, frequency picking, protocol selecting, data security design, anti-collision technology planning etc. Finally, several valuable conclusions were achieved.

The electronic information display console is an important device displaying integrated information on some type of large-scale ship. The human-computer interface (HCI) design that is not good enough will cause inefficiency or injury to the operators. If the deficiency is discovered while working, it will cost much labor, money and time. The virtual simulation method is tried to use evaluating the HCI design. Set the evaluation criteria on the basis of ergonomics theories such as the characteristic of human motion. Establish the models on CATIA platform, set the parameters in the Human Model Module and run the simulation. Analyze the results of the simulation according to the ergonomics theories such as the dynamic body measurement, the standards of the workspace design, and bring out the suggestions such as reducing the height by approximately 230mm of the screen. This method could be well used to evaluate the HCI design of other similar type of devices in the control room.

To study the therapeutic effect of splenic autotransplantation combined with lower esophagus transaction anastomosis in the treatment of liver cirrhosis induced portal hypertension.Thirty-six patients admitted from January 2003 to December 2006 were randomly divided into splenic autotransplantation group undergoing splenic autotransplantation after splenectomy combined with lower esophagus transaction anastomosis, and splenectomy group only undergoing splenectomy combined with lower esophagus transaction anastomosis. The general conduction, splenic scanning, liver function, and the level of serum Tuftsin and IgM of each patient were observed before and after operation.The levels of Tuftsin and IgM in splenic autotransplantation group were significant higher than that of splenectomy group 2 months after the operation, and the liver function showed no significant difference between these two groups. Splenic tissue was detected in the retroperitoneal space by 99mTc-DRBC 2 months after operation.Splenic autotransplantation combined with lower esophagus transaction anastomosis is a safe and effective treatment strategy for patients with liver cirrhosis induced portal hypertension, and the spleen tissue transplanted into the retroperitoneal space can partially preserve the immune function.

To study the effects of flavonoid, isolated from aerial parts of Scutellaria baicalensis Georgi (SSF), on memory impairment, neuronal damage, free radicals and energy metabolite disorders in aged rats.Approximately 25-month-old rats were used to establish the ageing model. The cognition of the rats was determined using the Morris water maze, neuronal morphology was observed by light/electron microscope, malondialdehyde (MDA) levels and the activity of superoxide dismutase (SOD), lactate dehydrogenase (LDH) and Na(+)-K(+)-ATPase were measured by spectrophotometry.In the Morris water maze task, the aged rats always took longer latency to find the hidden platform and spent less time swimming in the target quadrant than those of young control rats. The light/electron microscopic observations found significant neuropathological changes in the aged rats' brain. In addition, the production of MDA and the activity of SOD, LDH and Na(+)-K(+)-ATPase in the hippocampus and cerebral cortex of the aged rats showed critical abnormal changes. However, pre-treatment of the aged rats with SSF (35-140 mg kg(-1)) for 16-21 days dramatically improved cognitive dysfunction, neuropathological changes and biochemical abnormalities.These results indicate that the beneficial effects of SSF on memory impairment and neuronal damage in aged rats may be important for the treatment of senile dementia and for delaying the ageing processes.
