Abstract

NK group 2, member D (NKG2D) is one of the most critical activating receptors expressed by natural killer (NK) cells. There is growing evidence that acute myeloid leukemia (AML) cells may evade NK cell-mediated cell lysis by expressing low or no ligands for NKG2D (NKG2D-Ls). We hypothesized that CCAAT/enhancer-binding protein α (C/EBPα), one of the most studied lineage-specific transcription factors in hematopoiesis, might influence the expression of NKG2D-Ls. To test this hypothesis, we first examined the endogenous expression of wild-type C/EBPα (C/EBPα-p42) in human AML cell lines and demonstrated that its expression level was highly relevant to the sensitivity of AML cells to NK cell cytotoxicity. Induction of C/EBPα-p42 in the low endogenous CEBPA-expressing AML cell line increased the sensitivity to NK-induced lysis. Moreover, decreased expression of C/EBPα-p42 by RNA interference in AML cells abrogated NK-mediated cytotoxicity. We further showed that the increase in NK susceptibility caused by C/EBPα-p42 occurred through up-regulation of the NKG2D-Ls ULBP2/5/6 in AML cells. More importantly, chromatin immunoprecipitation (ChIP) coupled with high-throughput sequencing captured C/EBPα motif signatures at the enhancer regions of the ULBP 2/5/6 genes. Whilst, the AML-associated C/EBPα C-terminal mutant and N-terminal truncated mutant (C/EBPα-p30) diminished ULBP2/5/6 transcription. Finally, we identified that histone demethylase lysine-specific demethylase 1 (LSD1) inhibition can restore the expression of ULBPs via induction of CEBPA expression in AML cells, which may represent a novel therapeutic strategy for CEBPA-mutated AML. 178. A bioinspired floatable system with a 3D sandwich-type triphase interface for highly efficient nitrogen fixation.

Abstract

Photocatalytic nitrogen fixation is an ecologically friendly technique for synthesizing ammonia, and a key factor in the improvement of efficiency is the effective mass transfer of nitrogen and water simultaneously. Here, by anchoring BiVO4 on PDMS-modified Janus melamine sponge (BiVO4@PDMS@MS), we have prepared a floatable system that forms a 3D sandwich-type triphase reaction interface in the air phase, allowing nitrogen and water to reach the reaction active sites from separate diffusion pathways in different directions, effectively ensuring mass transfer of nitrogen and water simultaneously so that the performance improvement of photocatalytic nitrogen fixation can be guaranteed in kinetics. In Janus BiVO4@PDMS@MS, BiVO4 disperses uniformly on each surface of MS micropores, where many microreactors form, leading to a large triphase interface area that exposes a considerable number of active sites. In addition, the triphase interface is constructed above the water surface, and sunlight can irradiate the reaction interface directly and cause multiple scattering in the macropores of MS, enhancing light utilization. BiVO4@PDMS@MS demonstrated an ammonia synthesis rate of 624.87 μmol gcat−1 h−1 with high selectivity and stability, which is approximately 1.67 times and 353 times higher than that of the related diphase system (BiVO4@MS) and the powder system (BiVO4), respectively. We employed the triphase interface that forms in the gas phase for photocatalytic nitrogen fixation for the first time, effectively ensuring nitrogen and water mass transfer to improve photoactivity and selectivity, offering a promising option in optimizing highly efficient mass transfer for gas-consuming reactions. Abstract

This study examines whether the CEO uses share repurchases to sell her equity grants at inflated stock prices, a concern regularly voiced in politics and media. We find that the timing of buyback programs and equity compensation, i.e., the granting, vesting, and selling of equity, is largely determined by the corporate calendar through blackout periods and earnings announcement dates. As a consequence, share repurchases and equity compensation are positively correlated. This correlation disappears once we account for the corporate calendar and should thus not be interpreted causally. Our results do not support the conclusion that CEOs systematically misuse share repurchases at the expense of shareholders. To the contrary, equity compensation increases the propensity to launch a buyback program when buying back shares is beneficial for long-term shareholder value.

Abstract

Ovomucin (OVM) is a natural glycoprotein with various biological activities but poor solubility. This study aimed to enhance the solubility of OVM by using an ultrasonic-assisted method. The effect of ultrasound (US) on the structure, thermal stability and biological functions of OVM aggregates was evaluated. It was found that insoluble OVM aggregates were dissociated and the solubility increased significantly to 90.0 % after US under 400 W for 45 min. US also improved the onset temperature (To) and denaturation temperature (Td) of OVM. More importantly, the cholesterol binding capacity of both OVM and its digestion products were significantly improved after US (p < 0.05). The gastrointestinal digestion products of US-OVM also showed higher α-amylase and α-glucosidase inhibition than native OVM aggregates. US-induced dissociation of OVM aggregates and the conversion of β-sheet and β-turn to random coil, resulting in the exposure of hydrophobic binding sites may be an important reason for the enhanced stability and adsorption capacity. These findings suggested that US was an effective method for preparing soluble OVM and improved its adsorption capacity, which can further facilitate the application of OVM in the food industry.
