Abstract

Background: Glioblastoma (GBM) is the most common primary malignant tumor in the central nervous system. Paclitaxel (PTX) is a well‐established and highly effective anti‐cancer drug for peripheral solid tumors. However, the application of PTX in GBM is hindered by several limitations, including poor water solubility, restricted entry across the blood–brain barrier (BBB), and enhanced excretion by efflux transporters. P‐glycoprotein (P‐gp) is a crucial efflux transporter that is abundantly present in cerebral vascular endothelial cells and GBM cells. It plays a significant role in the exocytosis of PTX within tumor tissues. Methods: Recently, we have developed a novel technique for creating self‐assembled nanoparticles utilizing a range of natural bioactive molecules. These nanoparticles can encapsulate insoluble drugs and effectively cross the BBB. In additional, we revealed that certain nanoparticles have the potential to act as P‐gp inhibitors, thereby reducing the excretion of PTX. In this study, we conducted a screening of bioactive molecular nanoparticles to identify those that effectively inhibit the function of P‐gp transporters. Results: Among the candidates, we identified ursolic acid nanoparticles (UA NPs) as the P‐gp inhibitors. Furthermore, we prepared co‐assembled UA NPs embedded with paclitaxel, referred to as UA‐PTX NPs. Our results demonstrate that UA‐PTX NPs can enhance the blood concentration of PTX, facilitate its entry into the BBB, and inhibit the function of P‐gp, resulting in a decrease in the excretion of PTX. This discovery effectively addressed the above three issues associated with the use of PTX in glioma treatment. Conclusions: UA‐PTX NPs demonstrate strong anti‐tumor effects and show great potential for treating GBM. [ABSTRACT FROM AUTHOR]

Abstract

Objective: The diminished expression of microRNA‐146a (miR‐146a) in systemic lupus erythematosus (SLE) contributes to the aberrant activation of the interferon pathway. Despite its significance, the underlying mechanism driving this reduced expression remains elusive. Considering the integral role of enhancers in steering gene expression, our study seeks to pinpoint the SLE‐affected enhancers responsible for modulating miR‐146a expression. Additionally, we aim to elucidate the mechanisms by which these enhancers influence the contribution of miR‐146a to the activation of the interferon pathway. Methods: Circular chromosome conformation capture sequencing and epigenomic profiles were applied to identify candidate enhancers of miR‐146a. CRISPR activation was performed to screen functional enhancers. Differential analysis of chromatin accessibility was used to identify SLE‐dysregulated enhancers, and the mechanism underlying enhancer dysfunction was investigated by analyzing transcription factor binding. The therapeutic value of a lupus‐related enhancer was further evaluated by targeting it in the peripheral blood mononuclear cells (PBMCs) of patients with SLE through a CRISPR activation approach. Results: We identified shared and cell‐specific enhancers of miR‐146a in distinct immune cells. An enhancer 32.5 kb downstream of miR‐146a possesses less accessibility in SLE, and its chromatin openness was negatively correlated with SLE disease activity. Moreover, CCAAT/enhancer binding protein α, a down‐regulated transcription factor in patients with SLE, binds to the 32.5‐kb enhancer and induces the epigenomic change of this locus. Furthermore, CRISPR‐based activation of this enhancer in SLE PBMCs could inhibit the activity of interferon pathway. Conclusion: Our work defines a promising target for SLE intervention. We adopted integrative approaches to define cell‐specific and functional enhancers of the SLE critical gene and investigated the mechanism underlying its dysregulation mediated by a lupus‐related enhancer. 176. MiniMounter: A low‐cost miniaturized microscopy development toolkit for image quality control and enhancement.

Abstract

Head‐mounted miniaturized fluorescence microscopy (Miniscope) has emerged as a significant tool in neuroscience, particularly for behavioral studies in awake rodents. However, the challenges of image quality control and standardization persist for both Miniscope users and developers. In this study, we propose a cost‐effective and comprehensive toolkit named MiniMounter. This toolkit comprises a hardware platform that offers customized grippers and four‐degree‐of‐freedom adjustment for Miniscope, along with software that integrates displacement control, image quality evaluation, and enhancement of 3D visualization. Our toolkit makes it feasible to accurately characterize Miniscope. Furthermore, MiniMounter enables auto‐focusing and 3D imaging for Miniscope prototypes that possess solely a 2D imaging function, as demonstrated in phantom and animal experiments. Overall, the implementation of MiniMounter effectively enhances image quality, reduces the time required for experimental operations and image evaluation, and consequently accelerates the development and research cycle for both users and developers within the Miniscope community. 195. The Influence of Wind-Induced Response in Urban Trees on the Surrounding Flow Field

Abstract

In recent years, cities have experienced frequent climate changes and deteriorating wind environments. Urban vegetation has become an important measure to improve local microclimates with its flexible configuration. Leaves and branches also reorient with the direction of wind, affecting the airflow through the tree. However, trees are usually considered as stationary porous media areas and are not influenced by wind speed in existing numerical simulation studies. Therefore, by considering the response of a tree under natural wind, this study established a fitted relationship between porosity and wind speed by measuring the porosity of trees at different wind speeds in the field. A numerical model of the wind response of the tree was developed, and the tree drag coefficient was changed using the additional source term method to verify the feasibility of the model by measuring the wind environment behind the tree. To understand the effect of the wind-induced response on the surrounding flow field and its variation pattern, the surrounding flow fields of stationary tree (T-S) and wind-induced tree (T-D) at different wind speeds were compared and analyzed. The effect of porosity and height-to-width ratio under the wind-induced response of trees on the wind environment were quantified. It was found that at different wind speeds, as the wind speed increases, the tree porosity gradually increases and the drag coefficient decreases accordingly. The effective shading distance after wind response was 2.4H, which was 0.3H less compared to vertically fixed trees. The minimum wind speed increased linearly with plant porosity, and the minimum wind speed occurrence location and wind speed recovery distance were linearly and negatively correlated with tree height-to-width ratio. Therefore, the flow field around the tree was simulated to provide references for guiding tree planting and mitigating urban wind environments.

Abstract

In recent years, domain-specific knowledge bases (KBs) have attracted more attention in academics and industry because of their expertise and in-depth representation in a specific domain. However, when constructing a domain-specific KB, one needs to address not only the challenges in constructing a general KB, but also the difficulties raised by the nature of domain-specific raw data. Considering the usability of financial Knowledge Bases (KBs) in many downstream applications, such as financial risk analysis and fraud detection, we aim to build a Temporal Financial KB. However, the complex, time-varying financial knowledge and the volatile financial market evolution make construction quite challenging. Thus, we propose a Platform for Temporal Financial KB Construction, T-FinKB Platform 1. This platform consists of three fundamental modules, i.e., evolved knowledge extraction module, temporal record linkage and conflicts resolution module, and dynamic knowledge update module designed for financial knowledge. Generated temporal financial KBs (T-FinKBs) from T-FinKB Platform can be updated automatically. T-FinKB has been successfully applied to a downstream application, stock trend prediction with backtesting evaluation. In addition, T-FinKB Platform provides several APIs for visualization, queries and analytics. © 2023 IEEE.

Abstract

In recent years, the nitrogen-vacancy (NV) center in diamonds has been demonstrated to be a high-performance multiphysics sensor, where a lock-in amplifier (LIA) is often adopted to monitor photoluminescence changes around the resonance. It is rather complex when multiple resonant points are utilized to realize a vector or temperature-magnetic joint sensing. In this article, we present a novel scheme to realize multipoint lock-in detection with only a single-channel device. This method is based on a direct digital synthesizer (DDS) and frequency-shift keying (FSK) technique, which is capable of freely hopping frequencies with a maximum of 1.4 GHz bandwidth and encoding an unlimited number of resonant points during the sensing process. We demonstrate this method in experiments and show it would be generally useful in quantum multi-frequency excitation applications, especially in the portable and highly mobile cases.
