Abstract Background Treatment on risk adapted intensive pediatric protocols has improved outcome for teenagers and young adults (TYA) with T‐cell acute lymphoblastic leukemia (T‐ALL). Understanding the biology of disease in this age group and the genetic basis of relapse is a key goal as patients with relapsed/refractory disease have poor outcomes with conventional chemotherapy and novel molecular targets are required. This study examines the question of whether TYA T‐ALL has a specific biological‐molecular profile distinct from pediatric or adult T‐ALL. Methods Genomic characterization was undertaken of a retrospective discovery cohort of 80 patients aged 15–26 years with primary or relapsed T‐ALL, using a combination of Genome‐Wide Human SNP Array 6.0, targeted gene mutation and promoter methylation analyses. Findings were confirmed by MLPA, real‐time quantitative PCR, and FISH. Whole Exome Sequencing was performed in 4 patients with matched presentation and relapse to model clonal evolution. A prevalence analysis was performed on a final data set of 1,792 individual cases to identify genetic lesions with age specific frequency patterns, including 972 pediatric (1–14 years), 439 TYA (15–24 years) and 381 adult (≥25 years) cases. These cases were extracted from 19 publications with comparable genomic data identified through a PubMed search. Results Genomic characterization of this large cohort of TYA T‐ALL patients identified recurrent isochromosome 7q i(7q) in our discovery cohort (n = 3). Prevalence analysis did not identify any age specific genetic abnormalities. Genomic analysis of 6 pairs of matched presentation – relapsed T‐ALL established that all relapses were clonally related to the initial leukemia. Whole exome sequencing analysis revealed recurrent, targetable, mutations disrupting NOTCH, PI3K/AKT/mTOR, FLT3, NRAS as well as drug metabolism pathways. Conclusions All genetic aberrations in TYA T‐ALL occurred with an incidence similar or intermediate to that reported in the pediatric and adult literature, demonstrating that overall TYA T‐ALL exhibits a transitional genomic profile. Analysis of matched presentation – relapse supported the hypothesis that relapse is driven by the Darwinian evolution of sub‐clones associated with drug resistance (NT5C2 and TP53 mutations) and re‐iterative mutation of known key T‐ALL drivers, including NOTCH1.

Laboratory-based sleep manipulations show asymmetries between positive and negative affect, but say little about how more specific moods might change. We report extensive analyzes of items from the Positive and Negative Affect Scale (PANAS) during days following nights of chronic sleep restriction (6 h sleep opportunity), during 40 h of acute sleep deprivation under constant routine conditions, and during a week-long forced desynchrony protocol in which participants lived on a 28-h day. Living in the laboratory resulted in medium effects sizes on all positive moods (Attentiveness, General Positive Affect, Joviality, Assuredness), with a general deterioration as the days wore on. These effects were not found with negative moods. Sleep restriction reduced some positive moods, particularly Attentiveness (also General Positive), and increased Hostility. A burden of chronic sleep loss also led to lower positive moods when participants confronted the acute sleep loss challenge, and all positive moods, as well as Fearfulness, General Negative Affect and Hostility were affected. Sleeping at atypical circadian phases resulted in mood changes: all positive moods reduced, Hostility and General Negative Affect increased. Deteriorations increased the further participants slept from their typical nocturnal sleep. In most cases the changes induced by chronic or acute sleep loss or mistimed sleep waxed or waned across the waking day, with linear or various non-linear trends best fitting these time-awake-based changes. While extended laboratory stays do not emulate the fluctuating emotional demands of everyday living, these findings demonstrate that even in controlled settings mood changes systematically as sleep is shortened or mistimed.

A recent report by the joint Informatics Europe & ACM Europe Working Group on Informatics Education emphasizes that: (1) computational thinking is an important ability that all people should possess; (2) informatics-based concepts, abilities and skills are teachable, and must be included in the primary and particularly in the secondary school curriculum. Accordingly, the "2013 Best Practices in Education Award" (organized by Informatics Europe) was devoted to initiatives promoting Informatics Education in Primary and Secondary Schools. In this paper we present one of the winning projects: "Multi-Sensory Informatics Education". We have developed effective multi-sensory methods and software-tools to improve the teaching-learning process of elementary, sorting and recursive algorithms. The technologically and artistically enhanced learning environment we present has also the potential to promote intercultural computer science education and the algorithmic thinking of both science- and humanities-oriented learners.

This article presents a detailed analytical evaluation and comprehensive description of a mathematical model designed to simulate blood flow within the human cardiovascular system. The primary objective of this research is to develop a computational model capable of accurately simulating blood flow dynamics and to assess the variations in results using different numerical methods for solving the Navier-Stokes equations, which govern fluid motion. To achieve this, the study begins with an in-depth examination of the anatomy of the cardiovascular system, including various cardiovascular diseases such as stenosis and atherosclerosis, which significantly affect blood flow. The model incorporates important characteristics of blood, treating it as a viscous fluid under laminar flow conditions. Using the Navier-Stokes equations, it was developed a Python-based model to simulate these flow conditions and solve for different flow variables, such as velocity and pressure fields, under both normal and pathological conditions. The computational model was developed using two numerical methods: the Euler method and the Alternating Direction Implicit (ADI) method, which were compared in terms of their computational efficiency and accuracy. The simulations generated insights into how plaque buildup (stenosis) affects blood flow by altering wall shear stress and velocity profiles. This model, while built on foundational fluid dynamics principles, serves as an essential step towards creating a virtual reality (VR) surgical simulator for cardiovascular procedures. This simulator aims to assist surgeons in visualizing and planning surgical interventions by providing an interactive and realistic environment for studying blood flow and related complications. We have investigated whether the lack of X-ray pulsations from most neutron star (NS) low-mass X-ray binaries (LMXBs) could be due to the extension of their inner disc to the NS surface. To estimate the inner disc radii, we have employed the model, recently proposed to account for the torque reversals of LMXBs. In this model, the inner disc radius depends on the spin period as well as the dipole moment and the mass inflow rate of the disc. Our model results indicate that most LMXBs have mass accretion rates above the minimum critical rates required for the inner disc to reach down to the NS surface and thereby quench the pulsed X-ray emission. For most sources X-ray pulsations are allowed when the period decreases below a certain critical value. For the same parameters, the model is also consistent with the observed X-ray luminosity ranges of the individual accreting millisecond X-ray pulsars (AMXPs). The paucity of AMXPs compared to the majority population of non-pulsing LMXBs is explained, as well as the fact that AMXPs are transient sources. [ABSTRACT FROM AUTHOR]

It examines historical power struggles between the West and non-Western countries, with a particular focus on China and India. The book argues that sustainable development requires economic and political sovereignty. Lama's analysis extends beyond the Western world, incorporating selected sequences from Japanese history to critique Western patterns and ideologies. The problem of establishing an effective solid waste management system is complex and still unresolved in Serbian municipalities, cities and settlements. Most landfills in Serbia do not meet basic sanitary standards and can often contain waste that is dangerous to human, plant and animal health, and waste that can be recycled is often dumped. The aim of this paper is to investigate the relationship between waste generation and private investments in waste management. Statistical method of multiple linear regression was used to examine the impact of three predictors - economic development, investments in waste management, and urban population, on the dependent variable - municipal solid waste generation. The results of the research reveal that investments in waste management in Serbia do not have an effect on the waste reduction because the financial resources for such investments are not adequately managed. There is a large number of scientific researches that have dealt with the analysis of the mentioned relationship, however, their number on the example of Serbia is rather scarce. The results of the research could be useful to the state administrations of the Republic of Serbia.

The article describes the IoT infrastructure, the hardware of the IoT system, considers the issue of security of the chosen LoRa data transmission technology. Data was received from sensors for gas, temperature and humidity, atmospheric pressure, as well as the location of the end device. At the same time, the standardized security features of the selected LoRa technology for transmitting data from sensors to the server were investigated. The article deals with LoRa bi-directional secure communication line, the security function requires devices/end devices to be configured through the LoRa gateway. Security research is devoted to the development of a security mechanism to increase its resilience. The payload was formed with a hash of the last bytes, and the entire payload was encrypted with AES for integrity and confidentiality. A method for assessing and visualizing atmospheric air pollution is given on the example of the city of Almaty, Kazakhstan. The process of numerical modeling of the study of emissions of harmful substances into the atmosphere is based on a mathematical model formed by the system of Navier-Stokes equations, consisting of the continuity equation, as well as the equations of motion and the k-epsilon turbulence model. To test the numerical methods for processing mixing and chemical reactions, a test problem was chosen – a jet in a transverse flow. Three-dimensional numerical simulation has been implemented. The use of the Internet of Things (IoT) and the acquisition of big data made it possible to simultaneously observe the concentrations of several pollutants in the atmosphere, calculate this concentration and analyze the state of the surface air layer. Modeling allows forecasting the possible concentration of pollutants in certain areas at certain times of the year.

The study aimed to evaluate the mediating roles of patient experiences on patient loyalty. The data were collected through an electronic questionnaire regarding feedback from 5732 patients received outpatient clinics. Patient loyalty was evaluated using the Net Promoter Score (NPS 11 ) that patients were asked whether they would like to recommend the hospital to their relatives or friends. Patient experiences with physicians, nurses, and waiting times were also asked in the questionnaire. After preliminary analysis, mediation analyses were performed to evaluate direct and indirect causal effects among variables for NPS 11. While patient experiences are used as possible mediators, Branch Groups in the first and Admission Time in the second model are independent variables. In the analyses, Surgical Medical Science ( p = 0.019) and Day Shift ( p = 0.000) have a direct mediating effect on NPS 11 . Nursing care experiences were found to be a mediator variable for NPS 11 in both models ( p = 0.000 for both). Patient loyalty was associated with Surgical Medical Science and Day Shift primarily whereas Nursing care experience had a mediating role.
