Abstract

This essay argues that one of the factors holding back civilization-wide transitions to renewable energy is the widespread tendency to render petroleum and other hydrocarbons abject and abstract. Fossil fuel industry representations do this by hiding the true costs of petroculture behind the virtualization Energy; environmentalist framings do it by relying too much on petroaesthetics of doom (i.e., apocalyptic imagery) and gloom (i.e., Gothic visualizations of oil spills and rusting extractive infrastructure). The scarcity of representations of hydrocarbons that acknowledge both their life-giving and life-destroying properties, their powerful nonhuman agency in mediating practically every human and nonhuman relationship in the modern world, makes it hard to imagine alternatives to petroculture. Recently, artists have begun subverting petroaesthetic conventions in ways that counter the abstraction and abjection of hydrocarbons, including by using crude oil as an artistic medium in its own right. The oddly playful bitumen sands photographs of Louis Helbig, the bitumen-based “petrographs” of Warren Cariou, and the weird, enthralling “oilscapes” of Kathleen Thum are interpreted as meaningful challenges to the petroaesthetic status quo—provocations that matter in every sense of the word. Beyond merely promoting energy transitions, these images perform transitions as they empower viewers to see hydrocarbons as media with which all living things are deeply entangled.

Abstract

Foot-and-Mouth Disease (FMD) is one of the most important animal diseases hindering livestock production in Thailand. In this study, a temporal and spatial analysis at the subdistrict level was performed on FMD outbreak reports in Thailand from 2011 to 2018. Risk factors associated with FMD outbreaks were investigated using generalised estimating equations. The results showed that the incidence of FMD outbreaks was the highest in 2016 and was affected by season, with a peak in FMD outbreaks occurring in the rainy-winter season. The occurrences of FMD were mostly distributed as a number of small clusters within a few subdistricts. Some high-risk areas with repetitive outbreaks were detected in the central regions. Risk factors significantly associated with FMD outbreaks included the size of the dairy, beef, and pig populations in each subdistrict, the distance to the nearest subdistrict with an outbreak, the percentage of forest area, international border contact, the presence of a livestock market, and the occurrence of an FMD outbreak in a neighbouring subdistrict in the previous month. This study helped to identify high-risk areas and periods of FMD outbreaks in Thailand. Together with the identified risk factors, its results can be used to optimise the FMD control program in Thailand and in other countries having a similar livestock industry and FMD situation.

Abstract

Summary Background Among asymptomatic patients with severe carotid artery stenosis but no recent stroke or transient cerebral ischaemia, either carotid artery stenting (CAS) or carotid endarterectomy (CEA) can restore patency and reduce long-term stroke risks. However, from recent national registry data, each option causes about 1% procedural risk of disabling stroke or death. Comparison of their long-term protective effects requires large-scale randomised evidence. Methods ACST-2 is an international multicentre randomised trial of CAS versus CEA among asymptomatic patients with severe stenosis thought to require intervention, interpreted with all other relevant trials. Patients were eligible if they had severe unilateral or bilateral carotid artery stenosis and both doctor and patient agreed that a carotid procedure should be undertaken, but they were substantially uncertain which one to choose. Patients were randomly allocated to CAS or CEA and followed up at 1 month and then annually, for a mean 5 years. Procedural events were those within 30 days of the intervention. Intention-to-treat analyses are provided. Analyses including procedural hazards use tabular methods. Analyses and meta-analyses of non-procedural strokes use Kaplan-Meier and log-rank methods. The trial is registered with the ISRCTN registry, ISRCTN21144362. Findings Between Jan 15, 2008, and Dec 31, 2020, 3625 patients in 130 centres were randomly allocated, 1811 to CAS and 1814 to CEA, with good compliance, good medical therapy and a mean 5 years of follow-up. Overall, 1% had disabling stroke or death procedurally (15 allocated to CAS and 18 to CEA) and 2% had non-disabling procedural stroke (48 allocated to CAS and 29 to CEA). Kaplan-Meier estimates of 5-year non-procedural stroke were 2·5% in each group for fatal or disabling stroke, and 5·3% with CAS versus 4·5% with CEA for any stroke (rate ratio [RR] 1·16, 95% CI 0·86–1·57; p=0·33). Combining RRs for any non-procedural stroke in all CAS versus CEA trials, the RR was similar in symptomatic and asymptomatic patients (overall RR 1·11, 95% CI 0·91–1·32; p=0·21). Interpretation Serious complications are similarly uncommon after competent CAS and CEA, and the long-term effects of these two carotid artery procedures on fatal or disabling stroke are comparable. 217. Assessment of wall elasticity variations on intraluminal haemodynamics in descending aortic dissections using a lumped-parameter model.

Abstract

Descending aortic dissection (DAD) is associated with high morbidity and mortality rates. Aortic wall stiffness is a variable often altered in DAD patients and potentially involved in long-term outcome. However, its relevance is still mostly unknown. To gain more detailed knowledge of how wall elasticity (compliance) might influence intraluminal haemodynamics in DAD, a lumped-parameter model was developed based on experimental data from a pulsatile hydraulic circuit and validated for 8 clinical scenarios. Next, the variations of intraluminal pressures and flows were assessed as a function of wall elasticity. In comparison with the most rigid-wall case, an increase in elasticity to physiological values was associated with a decrease in systolic and increase in diastolic pressures of up to 33% and 63% respectively, with a subsequent decrease in the pressure wave amplitude of up to 86%. Moreover, it was related to an increase in multidirectional intraluminal flows and transition of behaviour as 2 parallel vessels towards a vessel with a side-chamber. The model supports the extremely important role of wall elasticity as determinant of intraluminal pressures and flow patterns for DAD, and thus, the relevance of considering it during clinical assessment and computational modelling of the disease.

Abstract

This paper reports evidence that an individual meeting with a study counselor at high school significantly improves the quality of choice of tertiary educational field, as self-assessed 18 months after graduation from college. To address endogeneity, we explore the variation in study counseling practices between schools as an instrumental variable (IV). Following careful scrutiny of the validity of the IV, our results indicate a significant and positive influence of study counseling on the quality of educational choice, foremost among males and those with low educated parents. The overall result is stable across a number of robustness checks.

Abstract

This paper studies the extent to which young children develop their cognitive ability in high and low quality schools. We use a representative panel data set containing cognitive test scores of 4-6 year olds in Dutch schools. School quality is measured by the school's average achievement test score at age 12. Our results indicate that children in high-quality schools develop their skills substantially faster than those in low-quality schools. The results remain robust to the inclusion of initial ability, parental background, and neighborhood controls. Moreover, using proximity to higher-achieving schools as an instrument for school choice corroborates the results. The robustness of the results points toward a causal interpretation, although it is not possible to erase all doubt about unobserved confounding factors.

222. Abstract 3696: GS-9716: A potent, selective and orally bioavailable small molecule inhibitor of MCL1 for the treatment of cancer

Abstract

MCL1 is an anti-apoptotic member of the BCL2 family of proteins. These anti-apoptotic proteins prevent caspase-mediated cell apoptosis by binding to the pro-apoptotic proteins BIM, BAK and BAX, thereby inhibiting the formation of pores on the outer mitochondrial membrane. In cancer, MCL1 is upregulated to overcome such stress-induced effects, promoting cell survival, therapy resistance, and tumor progression. Here, we describe the in vitro and in vivo activity of GS-9716, a potent and selective MCL1 small molecule inhibitor that binds directly to MCL1 and induces rapid apoptosis in cancer cells by activating the mitochondrial apoptotic pathway. GS-9716 displayed potent and selective disruption of human MCL1-BIM protein dimer in vitro. In hematological and solid tumor cell line models, GS-9716 led to dose dependent reduction of MCL1-BIM and MCL1-BAK protein dimers. MCL1 protein dimer (BAK and BIM) reduction correlated with dose-dependent increases in cleaved caspase (apoptosis marker). GS-9716 potently reduced cellular viability across panels of hematological (Median GI50 = 30 nM) and breast cancer cell lines (Median GI50 = 470 nM). GS-9716 exhibits favorable pharmacokinetics properties with good oral bioavailability across species. GS-9716 dosed daily, once per week, or 7-day cycles of 2 days on/5 days off, resulted in significant tumor growth inhibition against models of multiple myeloma (H929), several TNBC cell line xenografts (HCC1187 and HCC70) and patient-derived xenografts. Also, in these models, GS-9716 demonstrated dose-dependent MCL1 protein dimer (BIM and BAK) reduction and activation of apoptosis in tumors. In addition to single agent activity, GS-9716 showed strong synergy with multiple anti-cancer therapies; robust in vitro synergy was observed with venetoclax in the AML models and as well as with paclitaxel in the TNBC models. The combination treatment of paclitaxel (dosed IV, QW) with GS-9716 (dosed orally, 2 days on/5 days off) in the TNBC HCC1187 xenograft model achieved complete tumor regression. Similar responses were also observed when GS-9716 was dosed with paclitaxel in the TNBC PDX models. GS-9716 is currently in Phase 1 clinical trial evaluating safety, tolerability and pharmacokinetics as monotherapy and in combination with anticancer therapies in adults with advanced solid tumors (NCT05006794). Abstract

Microbubbles-assisted ultrasound (USMB) has shown promise in improving local drug delivery. The formation of transient membrane pores and endocytosis are reported to be enhanced by USMB, and they contribute to cellular drug uptake. Exocytosis also seems to be linked to endocytosis upon USMB treatment. Based on this rationale, we investigated whether USMB triggers exocytosis resulting in the release of extracellular vesicles (EVs). USMB was performed on a monolayer of head-and-neck cancer cells (FaDu) with clinically approved microbubbles and commonly used ultrasound parameters. At 2, 4, and 24 h, cells and EV-containing conditioned media from USMB and control conditions (untreated cells, cells treated with microbubbles and ultrasound only) were harvested. EVs were measured using flow cytometric immuno-magnetic bead capture assay, immunogold electron microscopy, and western blotting. After USMB, levels of CD9 exposing-EVs significantly increased at 2 and 4 h, whereas levels of CD63 exposing-EVs increased at 2 h. At 24 h, EV levels were comparable to control levels. EVs released after USMB displayed a heterogeneous size distribution profile (30–1200 nm). Typical EV markers CD9, CD63, and alix were enriched in EVs released from USMB-treated FaDu cells. In conclusion, USMB treatment triggers exocytosis leading to the release of EVs from FaDu cells.

Abstract

Melanopsin-containing retinal ganglion cells play an important role in the non-image forming effects of light, through their direct projections on brain circuits involved in circadian rhythms, mood and alertness. Individual differences in the functionality of the melanopsin-signaling circuitry can be reliably quantified using the maximum post-illumination pupil response (PIPR) after blue light. Previous protocols for acquiring PIPR relied on the use of mydriatics to dilate the light-exposed eye. However, pharmacological pupil dilation is uncomfortable for the participants and requires ophthalmological expertise. Hence, we here investigated whether an individual’s maximum PIPR can be validly obtained in a protocol that does not use mydriatics but rather increases the intensity of the light stimulus. In 18 participants (5 males, mean age ± SD: 34.6 ± 13.6 years) we evaluated the PIPR after exposure to intensified blue light (550 µW/cm2) provided to an undilated dynamic pupil. The test-retest reliability of the primary PIPR outcome parameter was very high, both between day-to-day assessments (Intraclass Correlation Coefficient (ICC) = 0.85), as well as between winter and summer assessments (ICC = 0.83). Compared to the PIPR obtained with the use of mydriatics and 160 µW/cm2 blue light exposure, the method with intensified light without mydriatics showed almost zero bias according to Bland-Altman plots and had moderate to strong reliability (ICC = 0.67). In conclusion, for PIPR assessments, increasing the light intensity is a feasible and reliable alternative to pupil dilation to relieve the participant’s burden and to allow for performance outside the ophthalmological clinic.
