208. Isolates of Meloidogyne hapla with Distinct Mitochondrial Genomes

Abstract

Because two conflicting reports of the structure of the Meloidogyne hapla mitochondrial genome exist, we compared the mitochondrial DNA (mtDNA) purified from two isolates of M. hapla: one from San Bernardino County in southern California (BRDO) and the other from England. The authenticity of the BRDO isolate in particular was confirmed by examination of morphological characters, isoenzyme analysis, and differential host range tests. Restriction analysis revealed that mtDNA from the BRDO and English isolates corresponded to only the structure first reported, although significant differences between the two isolates were apparent. Southern blots probed with cloned, cytochrome oxidase I (cox-l) DNA from Romanomermis culicivorax mtDNA confirmed that the analyzed DNA was of mitochondrial origin. Thus, M. hapla has at least two distinct but presumably related mitchondrial genomes, plus at least one very different structure. These data are discussed with reference to recent molecular diagnostic and phylogenetic analyses of Meloidogyne.

Abstract

The Dunes anomaly is a water-dominated geothermal system in the alluvium of the Salton Trough, lacking any surface expression. It was discovered by shallow-temperature gradient measurements. A 612-meter-deep test well encountered several temperature-gradient reversals, with a maximum of 105 C at 114 meters. The program involves surface geophysics, including electrical, gravity, and seismic methods, down-hole geophysics and petrophysics of core samples, isotopic and chemical studies of water samples, and petrological and geochemical studies of the cores and cuttings. The aim is (1) to determine the source and temperature history of the brines, (2) to understand the interaction between the brines and rocks, and (3) to determine the areal extent, nature, origin, and history of the geothermal system. These studies are designed to provide better definition of exploration targets for hidden geothermal anomalies and to contribute to improved techniques of exploration and resource assessment.

Abstract

The advanced control technology is considered that is necessary to cope with the medium STOL transport landing problem and, in particular, the necessity to decouple with active control techniques. It is shown that the need to decouple is independent of the powered lift concept but that the provisioning for decoupling is most greatly dependent on the preassumed piloting technique. The implications of decoupling and active control techniques with respect to pilot technique options, handling quality criteria, flight control mechanization, and the use of piloted simulation as a design tool, are also discussed.

Abstract

Apart from the Bibiani mine on its eastern boundary, the Sefwi belt has produced no significant gold. Indications of gold mineralisation at Yamfo originated in a regional geochemical soil sampling traverse and were confirmed by a small soil sampling survey. A number of gold-bearing shear zones have now been recognised on licence areas straddling the NW contact zone between the Sefwi volcanic belt and the Sunyani basin sediments. Exploration to locate new, large, open-pittable deposits has been by means of stream sediment sampling, soil sampling, trenching, geophysical surveys and exploration drilling. By mid-1998, resources totalled 25 700 000 tonnes grading 2.2 g/t Au at Yamfo/Centenary, 9 300 000 t grading 2.6 g/t Au at Subenso, 7 900 000 t grading 2.0 g/t Au at Bosumkese and 19 900 000 t grading 2.6 g/t Au at Kenyase.
