208. KLF4 targets RAB26 and decreases 5-FU resistance through inhibiting autophagy in colon cancer

Abstract

ABSTRACTBackgroundAccumulating studies demonstrated that resistance of colon cancer (CC) to 5-fluorouracil (5-FU) contributes to adverse prognosis. We investigated how Kruppel-like factor 4 (KLF4) affected 5-FU resistance and autophagy in CC cells.MethodsKLF4 expression and its downstream target gene RAB26 in CC tissues was analyzed by bioinformatics analysis, and the effect of abnormal KLF4 expression on prognoses of CC patients was predicted. Luciferase reporter assay detected the targeted relationship between KLF4 and RAB26. The viability and apoptosis of CC cells were analyzed by CCK-8 and flow cytometry. The formation of intracellular autophagosomes was detected by confocal laser scanning microscopy and immunofluorescence staining. The mRNA and protein levels were assayed by qRT-PCR and western blot. A xenograft animal model was constructed to verify the function of KLF4. Rescue assay was employed to verify whether KLF4/RAB26 could affect 5-FU resistance in CC cells through autophagy.ResultsKLF4 and RAB26 were lowly expressed in CC. KLF4 correlated with patients’ survival. KLF4 was down-regulated in 5-FU resistant CC cells. KLF4 overexpression suppressed the proliferation and 5-FU resistance of CC cells, and inhibited LC3 II/I expression and autophagosome formation. Autophagy activator Rapamycin or sh-RAB26 treatment reversed the impact of KLF4 overexpression on 5-FU resistance. In vivo assay verified that KLF4 inhibited 5-FU resistance in CC cells. Rescue experiments revealed that KLF4 targeted RAB26 to inhibit CC cell autophagy, resulting in decreasing the resistance to 5-FU.ConclusionKLF4 strengthened the sensitivity of CC cells to 5-FU by targeting RAB26 to restrain autophagy pathway.

Abstract

Strain-hardening mechanism of high strain line pipe was investigated and discussed in terms of microstructure and grain orientation. It is concluded that X70 high strain line pipe does not fit to the Hollimon formula in the entire deforming process, but the normal X70 line pipe does. At the beginning of plastic deformation, the strain-hardening capacity of X70 high strain line pipe is excellent. It decreases rapidly as the stain increases, then becomes stable after 2.0% total strain. The high strain line pipe is composed of soft phase (polygonal ferrite) and hard phase (bainite). When plastic deformation initiates, the strain-hardening capacity of high strain line pipe is better because of the ferrite deformation firstly ahead of the bainite, due to strain-hardening capacity of the ferrite is much better than that of the bainite. With ferrite deformation decreasing and bainite deformation increasing, the strain-hardening capacity declines until the micro-deformation is homogeneous. Besides, it is obvious that the large angle grain boundaries of more than 15° are in the majority of the microstructure of high strain line pipe without deformation, so the strain-hardening capacity is highest at the beginning of plastic deformation. 214. Quantitative Analysis of the Roots, Stems, and Leaves of <italic>Polygonum multiflorum</italic> by Ultra-High-Performance Liquid Chromatography-Mass Spectrometry.

Abstract

The concentrations of 2,3,5,4′-tetrahydroxystilbene-2-O-β-d-glucoside, quercetin, catechin, emodin, and polygoacetophenoside in roots, stems and leaves of Polygonum multiflorum, a traditional Chinese medicinal herb, were simultaneously determined by ultra-performance liquid chromatography equipped with electrospray ionization and quadrupole-time-of-flight mass spectrometry (UPLC/ESI-Q-TOF-MS). The established quantitative analysis method has good precision (<2.94%), accuracy (97.21-101.37%), linear correlation coefficient (<0.990), and high sensitivity (0.46 ng/mL). The concentrations of 2,3,5,4′-tetrahydroxystilbene-2-O-β-d-glucoside and emodin were 34.86 ± 10.58 and 31.09 ± 8.26 mg/g, respectively, in the roots, much higher than that in the stems and leaves, while the concentration of catechin (29.62 ± 7.76) was the highest in stems. However, polygoacetophenoside and quercetin are the main components in the leaves: 50.54 ± 12.24 and 28.35 ± 1.88 mg/g, respectively. The results show that these concentration differences in roots, stems and leaves of P. multiflorum may be associated with the therapeutic effects of this traditional Chinese medicinal herb. Abstract

Strain-hardening capacity is the most effective way to improve the deformability of line pipe. In this paper, four kinds of new X70 high strain line pipe have been produced by four manufactures respectively. Microstructure, strain-hardening capacity, and characterization of the X70 high strain line pipe have been analyzed. It is concluded that X70 high strain line pipe steel does not fit to the Hollimon formula in the entire deforming process, and a stable strain-hardening exponent is not available. At the beginning of plastic deformation, the strain-hardening capacity of X70 high strain line pipe steel is excellent. It decreases rapidly as the stain increases then becomes stable after 2.0% total strain. The stress ratios of Rt₁.₅/Rt₀.₅, Rt₂.₀/Rt₁.₀ and Rt₅.₀/Rt₁.₀ are proved to characterize the strain-hardening capacity of X70 high strain line pipe steel. When Rt₁.₅/Rt₀.₅≥1.070, Rt₂.₀/Rt₁.₀≥1.025, and Rt₅.₀/Rt₁.₀≥1.050, the average strain over the gauge length of 2 times the outer diameter, centered at the wrinkle on intrados peak, is above 1.3%.

Abstract

Bead-based assays have drawn more and more attention in biomedical fields. Herein, we proposed a novel approach to achieve multiplex analysis on a single porous hydrogel bead (PHB) with Raman dyes (RDs) encoded core–shell surface-enhanced Raman scattering (SERS) nanotags. Because of the amplified signal of RDs by core–shell metal structure of the nanotag and the high surface area to volume ratio (SVR) of the PHB, the sensitivity and linear dynamic range (LDR) of the as-proposed multiplex analysis method are significantly improved. We anticipate this approach to be widely used in the multiplex assays and biosensors.
