Abstract

In recent years, increasing attention has been paid to the pharmacological efficacy of tannins. Tannic acid (TA), the simplest hydrolysable tannin that has been approved by the FDA as a safe food additive, is one of the most important components of these traditional medicines. Studies have shown that TA displays a wide range of pharmacological activities, such as anti-inflammatory, neuroprotective, antitumor, cardioprotective, and anti-pathogenic effects. Here, we summarize the known pharmacological effects and associated mechanisms of TA. We focus on the effect and mechanism of TA in various animal models of inflammatory disease and organ, brain, and cardiovascular injury. Moreover, we discuss the possible molecular targets and signaling pathways of TA, in addition to the pharmacological effects of TA-based nanoparticles and TA in combination with chemotherapeutic drugs.

207. Evaluation of the efficacy and safety of Abelmoschus manihot alone and in combination with irbesartan for reduction of albuminuria in patients with type 2 diabetes and diabetic kidney disease.

Abstract

Objective To evaluate the efficacy and safety of Abelmoschus manihot (A. Manihot) alone and in combination with irbesartan, for reduction of albuminuria in patients with type 2 diabetic kidney disease. Methods A multicenter randomized double-blind and parallel controlled clinical trial was performed in 9 hospitals of Jiangsu Province (Affiliated Hospital of Nanjing University of Chinese Medicine, First People's Hospital of Changzhou, First People's Hospital of Xuzhou, Wuxi People's Hospital, Affiliated Hospital of Nantong University, Taizhou Hospital of Traditional Chinese Medicine, First Affiliated Hospital of Soochow University, Zhongda Hospital, Southeast University, Changzhou Hospital of Traditional Chinese Medicine) from May 2017 to March 2021. Huangkui capsule, as a traditional Chinese medicine, is made from the ethanol extract of flowers in A. Manihot. All enrolled patients were randomly assigned to the irbesartan group [irbesartan tablets (150 mg/dose, 1 dose/day) +Huangkui capsule simulant (2.5 g/dose, 3 doses/day)], Huangkui capsule group [Huangkui capsule (2.5 g/dose, 3 doses/day) +irbesartan simulant (150 mg/dose, 1 dose/day)], and combined treatment group [irbesartan tablets (150 mg/dose, 1 dose/day) +Huangkui capsule (2.5 g/dose, 3 doses/day)]. The duration of intervention was 24 weeks. Urinary creatinine and urinary albumin were detected at baseline and 24 weeks after treatment, and the urinary albumin-to-creatinine ratio was calculated to observe the change value and rate of UACR compared with baseline. The occurrence of adverse events, adverse reactions, serious adverse events, serious adverse events, and adverse events leading to withdrawal were recorded and the incidence was calculated. One-way analysis of variance (ANOVA), χ² test or Fisher's exact test were used to compare among groups. Results A total of 413 patients with type 2 diabetic kidney disease were included, including 138 in the irbesartan group, 137 in the Huangkui capsule group and 138 in the combined treatment group. After 24 weeks of treatment, the UACR changes of three groups were (-89.07±51.17) mg/g in the irbesartan group, (-146.06±45.52) mg/g in the Huangkui capsule group, and (-262.31±39.08) mg/g in the combined treatment group. The rate of UACR change was (-5.21±6.12)% in the irbesartan group, (-11.89±5.75)% in the Huangkui capsule group, and (-28.56±4.65)% in the combined treatment group, respectively. There were significant differences between the combined treatment group and the irbesartan group in the change value and rate of UACR (P<0.001), while there were no statistical differences between the Huangkui capsule group and irbesartan group (P>0.05). After 24 weeks of treatment, there were no significant differences in the incidence of adverse events, adverse reactions, serious adverse events and serious adverse reactions among the 3 groups (P>0.05). Conclusion The combination of irbesartan and Huangkui capsule can reduce the level of UACR in patients with type 2 diabetic kidney disease, and it shows good efficacy and safety in the treatment of albuminuria in patients with type 2 diabetic kidney disease. Abstract

Accumulating evidence demonstrates that Macleaya cordata extract exerts antiviral and anti-inflammatory effects in various diseases. The present study aimed to investigate the potential effects of M. cordata on rotavirus SA11-induced diarrhea in mice. Diarrhea severity, levels of inflammatory cytokines, histological changes in the small intestine and the underlying mechanisms were evaluated in rotavirus-stimulated mice treated with 1, 2 and 4 mg/kg/day M. cordata or 4 mg/kg/day ribavirin (positive control). M. cordata treatment effectively ameliorated rotavirus-induced diarrhea in a dose-dependent manner by decreasing viral RNA levels. In addition, M. cordata reduced the release of pro-inflammatory cytokines including migration inhibitory factor, interleukin (IL)-8, IL-β, interferon-γ and tumor necrosis factor-α, and elevated the secretion of the anti-inflammatory cytokine IL-10 following rotavirus infection. M. cordata inhibited intestinal epithelial cell apoptosis and improved intestinal inflammation after rotavirus infection. The study also revealed that M. cordata exerted antiviral and anti-inflammatory effects on rotavirus-induced diarrhea by suppressing the Janus kinase 2 (JAK2)/STAT3 pathway, as reflected by decreased protein expression of phosphorylated (p)-JAK2 and p-STAT3. Overall, M. cordata effectively inhibited the inflammation caused by rotavirus, which was closely associated with the suppression of JAK2/STAT3 phosphorylation. These data suggested that M. cordata may be applied as a treatment for rotavirus-induced diarrhea.

Abstract

A growing body of research showed that diet management, such as promoting protein and vitamin intake and food restriction play a crucial role in extending time to morbidity and mortality in patients undergoing hemodialysis. However, the current dietary recommendations in nutrition guidelines lack examination of cultural factors. The study aimed to understand the diet influenced by culture in Chinese patients undergoing hemodialysis. Semi-structured interviews were conducted for 23 patients, aged 23–75 years, undergoing hemodialysis in a local tertiary hospital. Interview questions mainly focused on patients' real experience about diet, and their perceptions and attitudes toward diet. Each interview was digitally recorded, and conventional content analysis was used to attain information. The majority of patients reserved Chinese traditional dietary habits about salt and calcium intake. Although Chinese herbal medicine was not consumed, dietary therapy including brown sugar and dates was included in the participants' diet. Eggs, broth, and seafood were three prominent preferences and taboos profoundly impacted by culture. Additionally, Chinese social culture influenced patients' dietary behavior of eating at home and knowledge attainment. Diet in Chinese patients undergoing hemodialysis was still strongly influenced by culture. Culturally sensitive interventions regarding the improvement of diet intake are urgently needed.

The main objective of this work is to develop a new oily nanosuspension to improve bioavailability and stability of CS formulations.Methods: After screening the formulations, cefquinome sulfate oily nanosuspension (CS-NSP) was prepared by mortar grinding, using propylene glycol dicaprolate/dicaprate (Labrafac™ PG) as oil medium and caprylocaproyl polyoxyl-8 glycerides (Labrasol®) as stabilizer. The properties of CS-NSP were investigated by testing its physicochemical characteristics, stability, in vitro release, hemolysis, and muscle irritation. The in vivo pharmacokinetics of CS-NSP was studied using rats.Results: Results show that CS-NSP presents suitable stability, physicochemical properties and safety. 229. Evaluation and Empirical Research on Eco-Efficiency of Financial Industry Based on Carbon Footprint in China

Abstract

Since finance is the core of economic development, the green development of the financial industry is an essential driving force not only for achieving the dual “carbon” goal of China but also for economic and social sustainable development. An accurate understanding of the ecological efficiency of the financial industry is of great importance for guiding sustainable economic development. In this paper, we first calculate the carbon footprint of China’s financial industry in 2012 and 2017 based on the life cycle theory and the input–output analysis method. Second, we analyze the primary sources and final flows of the carbon footprint of the financial industry in each province from the perspectives of the industrial chain and final demand. Finally, we estimate the ecological efficiency, emission reduction, and value-added potential of the financial industry by using the radially adjusted slack variable DEA model (SRAM-DEA) under two assumptions, natural disposability and managerial disposability. The results show that (1) the ecological efficiency of the financial industry in most provinces is low, and the regional differences are significant; (2) the overall ecological efficiency of the financial industry in 2017 was better than that in 2012; (3) technological innovation of financial products and the upgrading of capital supervision play an essential role in promoting the improvement of ecological efficiency. Especially, under managerial disposability, the ecological efficiency of the financial industry in each province has a greater potential for emission reduction and added value.

Abstract

Disordered inflammation and apoptosis are closely related to diseases, and inflammation can also promote cell apoptosis, where growing evidence has shown that circular RNAs (circRNAs) play important roles. Lipopolysaccharide (LPS) is the main component of the cytoderm of gram-negative bacterium, which can cause inflammatory responses in macrophages. We constructed an inflammatory model by exposing chicken macrophage cell lines (also known as HD11) to LPS for in vitro experiments. In this study, we validated a novel circRNA—circNFIC—which was dramatically up-regulated in tissues infected by coccidia and cells exposed to LPS. Besides, circNFIC could significantly promote the expression levels of pro-inflammation factors, including (IL-1β, TNFα, and IFNγ) and pro-apoptosis maker genes (caspase 3 and caspase 8) in HD11 exposed to LPS or not. In terms of mechanism, circNFIC exerted notable effects on DENND1B to regulate cell inflammation and apoptosis by sponging miR-30e-3p. The molecular functions played by miR-30e-3p and DENND1B have been explored, respectively. In addition, the effects of circNFIC knockdown suppressing the expression of pro-inflammatory and pro-apoptosis functions could be reversed by a miR-30e-3p inhibitor. On the whole, circNFIC promoted cell inflammation and apoptosis via the miR-30e-3p/DENND1B axis.

Abstract

The fimbrin protein family contains a variety of proteins, among which Plastin1 (PLS1) is an important member. According to recent studies, variations in the coding region of the PLS1 gene are associated with the development of deafness. However, the molecular mechanism of deafness caused by PLS1 gene variants remains unknown. Whole‐exome sequencing was performed on hearing‐impaired family members and hearing family members to identify pathogenic variants, followed by Sanger sequencing. A minigene assay was conducted to investigate the effect of the variant on PLS1 mRNA splicing. The pathogenicity of the variant was further investigated in zebrafish. RNA‐sequencing (RNA‐seq) was performed to analyze the dysregulation of downstream signaling pathways caused by knockdown of PLS1 expression. We identified a novel variant, PLS1 c.981+1G>A, in a large Chinese family with hearing loss and showed that the variant is responsible for the occurrence of hearing loss by inducing exon 8 skipping. The variant caused abnormal inner ear phenotypes, characterized by decreases in the mean otolith distance, anterior otolith diameter, posterior otolith diameter, cochlear diameter, and swimming speed and distance in zebrafish. Furthermore, silencing PLS1 expression significantly upregulated the expression of genes in the PI3K‐Akt signaling pathway, including Col6a3, Spp1, Itgb3 and hepatocyte growth factor (Hgf). PLS1 c.981+1G>A is a novel pathogenic variant causing hearing loss by inducing exon 8 skipping.
