Abstract Azulene-based conjugated systems are of great interests due to their unusual structures and photophysical properties. Incorporation of a transition metal into azulene skeleton presents an intriguing opportunity to combine the d π-p π and p π-p π conjugated properties. No such metallaazulene skeleton however has been reported to date. Here, we describe our development of an efficient [5 + 2] annulation reaction to rapid construction of a unique metal-containing [5-5-7] scaffold, termed metalla-dual-azulene (MDA), which includes a metallaazulene and a metal-free organic azulene intertwined by sharing the tropylium motif. The two azulene motifs in MDA exhibit distinct reactivities. The azulene motif readily undergoes nucleophilic addition, leading to N-, O- and S-substituted cycloheptanetrienyl species. Demetalation of the metallaazulene moiety occurs when it reacts with n Bu4NF, which enables highly selective recognition of fluoride anion and a noticeable color change. The practical [5 + 2] annulation methodology, facile functional-group modification, high and selective fluoride detection make this new π-conjugated polycyclic system very suitable for potential applications in photoelectric and sensing materials.

Abstract Background To investigate the diagnostic performance of parameters derived from monoexponential, biexponential, and stretched-exponential diffusion-weighted imaging models in differentiating tumour progression from pseudoprogression in glioblastoma patients. Methods Forty patients with pathologically confirmed glioblastoma exhibiting enhancing lesions after completion of chemoradiation therapy were enrolled in the study, which were then classified as tumour progression and pseudoprogression. All patients underwent conventional and multi-b diffusion-weighted MRI. The apparent diffusion coefficient (ADC) from a monoexponential model, the true diffusion coefficient (D), pseudodiffusion coefficient (D*) and perfusion fraction (f) from a biexponential model, and the distributed diffusion coefficient (DDC) and intravoxel heterogeneity index (α) from a stretched-exponential model were compared between tumour progression and pseudoprogression groups. Mitreola quanruii , a new species from Guangxi, China, is described and illustrated in this study. It is morphologically similar to M. liuyanii because of the terete stems, creeping and branched at the base, the leaves which are pilose on both surfaces and the bilobed capsules with two erect horns. The new species can be distinguished from M. liuyanii by its taller habit, up to 20–50 cm tall, its linear leaves, 4–18 × 0.3–1 cm with acuminate apex and 8–10 pairs of lateral veins, its narrowly triangular stipules, its linear bracts, ca. 1.0 cm long and glabrous and its glabrous calyx. Mitreola quanruii is temporarily assessed as data deficient (DD) according to IUCN. The habitat of Mitreola quanruii is extremely fragile. Therefore, this species deserves close attention and protection.

Currently, many cancer patients with bone defects are still threatened by tumor recurrence, postoperative bacterial infection, and massive bone loss. Many methods have been studied to endow bone implants with biocompatibility, but it is difficult to find an implant material that can simultaneously solve the problems of anticancer, antibacterial and bone promotion. Here, a multifunctional gelatin methacrylate/dopamine methacrylate adhesive hydrogel coating containing 2D black phosphorus (BP) nanoparticle protected by polydopamine (pBP) is prepared by photocrosslinking to modify the surface of poly (aryl ether nitrile ketone) containing phthalazinone (PPENK) implant. The multifunctional hydrogel coating works in conjunction with pBP, which can deliver drug through photothermal mediation and kill bacteria through photodynamic therapy at the initial phase followed by promotion of osteointegration. In this design, photothermal effect of pBP control the release of doxorubicin hydrochloride loaded via electrostatic attraction. Meanwhile, pBP can generate reactive oxygen species (ROS) to eliminate bacterial infection under 808 nm laser. In the slow degradation process, pBP not only effectively consumes excess ROS and avoid apoptosis induced by ROS in normal cells, but also degrade into PO43− to promote osteogenesis. In summary, nanocomposite hydrogel coatings provide a promising strategy for treatment of cancer patients with bone defects.

Based on historical documents and field investigations, this article analyzes the origin and development of the concept and form of the Chinese courtyard “TianJing” in vernacular residences to address common misunderstandings in previous studies. Focused on the scale standard in YingZaoFaYuan traditional building technology in the JiangNan region, TianJing is closely related to the main hall in dwellings reflecting the construction customs in different regions. Through the statistical analysis of the measured dimension data, this study shows the scale characteristics, demonstrating the cultural correlations of vernacular residences in different regions based on similarities. It explores the causes, rules and effects of and changes in traditional construction wisdom reflected by the historical geographic comparative background.

Abstract Background SPOCK3 is a secreted extracellular matrix proteoglycan. This study aimed to investigate the effect of SPOCK3 on the malignant progression of prostate cancer and to construct a prognostic model to predict DFS of patients with prostate cancer. Methods Clinical and transcriptome sequencing data for prostate cancer were download from the TCGA and GEO databases. The survival curve showed that SPOCK3 has prognostic significance. GO, KEGG, and GSEA enrichment analysis were used to investigate how SPOCK3 affects the malignant progression of prostate cancer. Based on ESTIMATE and ssGSEA, the relationship between SPOCK3 and immune cell infiltration in prostate cancer tissue was clarified. Univariate and multivariate COX regression analysis was used to identify the independent prognostic factors of prostate cancer OS and to construct a nomogram. The calibration curve and ROC curves were drawn to assess the nomogram's predictive power. Results The survival curve revealed that patients in the low-expression group of SPOCK3 had a poor prognosis. According to enrichment analysis, SOPCK3-related genes were enriched in collagen-containing extracellular matrix, PI3K-Akt, and MAPK signaling pathway. ESTIMATE analysis revealed that SPOCK3 expression was positively correlated with the interstitial score, immune score, and ESTIMATE score. The results of ssGSEA analysis revealed that the infiltration levels of Mast cells, NK cells, and B cells were higher in the SPOCK3 high expression group. Cox regression analysis showed that SPOCK3 expression level, T and Gleason score were independent risk factors of patient prognosis, and a nomogram was constructed. The ROC curve showed the AUCs of DFS at 2, 3, and 5 years. Conclusion SPOCK3 is a protective factor for DFS in prostate cancer patients. SPOCK3 is significantly associated with immune cell infiltration. The prognostic model constructed based on SPOCK3 has excellent predictive performance.

Background/Purpose: Acute oral mucositis (OM) is a painful complication of concurrent chemoradiotherapy (CCRT). This severe adverse symptom may impact on patient's quality of life, lead to malnutrition. Thus, finding more effective methods in OM management is very important. The purpose of this study is to evaluate the efficacy of polyacrylate silver salt/Polyvinylpyrrolidone-based liquid oral gel (named as polyacrylate silver salt oral gel) in improving the symptomatic relief of CCRT-induced oral mucositis and oral dysfunction in neck and head cancer patients. Methods: In this study, 24 oral cancer patients underwent CCRT and having OM grade 2 or higher were randomly assigned into the test group and the control group. Both groups followed Multinational Association of Supportive Care in Cancer and International Society of Oral Oncology (MASCC/ISOO) clinical practice guidelines for the management of mucositis, but adding rinsing with 15 g oral gel right after oral hygiene treaded the test group. Clinical OM and oral function were assessed weekly for 4 consecutive weeks till 5–10 days after the completion of radiotherapy. For evaluation, Common Terminology Criteria for Adverse Events (CTCAE) v3.0 was used for collecting the data of OM grade. Abstract Severe disturbances in a power network can cause the system frequency to exceed the safe operating range. As the last defensive line for system emergency control, under frequency load shedding (UFLS) is an important method for preventing a wide range of frequency excursions. This paper proposes a hierarchical UFLS scheme of “centralized real-time decision-making and decentralized real-time control” for inter-connected systems. The centralized decision-layer of the scheme takes into account the importance of the load based on the equivalent transformation of kinetic energy (KE) and potential energy (PE) in the transient energy function (TEF), while the load PE is used to determine the load shedding amount (LSA) allocation in different loads after faults in real-time. At the same time, the influence of inertia loss is considered in the calculation of unbalanced power, and the decentralized control center is used to implement the one-stage UFLS process to compensate for the unbalanced power. Simulations are carried out on the modified New England 10-generator 39-bus system and 197-bus system in China to verify the performance of the proposed scheme. The results show that, compared with other LSA allocation indicators, the proposed allocation indicators can achieve better f nadir and t d . At the same time, compared with other multi-stage UFLS schemes, the proposed scheme can obtain the maximum f nadir with a smaller LSA in scenarios with high renewable energy sources (RES) penetration.

Dielectric ceramics are promising in large-scale commercial millimeter-wave communication technology, such as 5G and the upcoming 6G, thanks to their excellent frequency selection characteristics and environmental stability. In this work, various contents of SbO bonds were introduced into the MgTa2O6 lattice using the solid-phase reaction method to investigate the effects on the lattice and microwave dielectric properties. XRD confirms that Sb ions successfully occupy Ta sites in the lattice and cause lattice shrinkage and crystallinity deterioration, which leads to a slight decrease in the quality factor. Furthermore, DFT calculations reveal that the doping leads to electron-biased aggregation toward O atoms, causing higher SbO ionicity, but also attenuates the degree of ionization of Ta and Mg ions, which makes the dielectric constant of the doped samples vary non-monotonically with gradient doping. Satisfactorily, Sb doping substantially enhanced the thermal stability of the ceramics, with TCF values reduced from 36 × 10−6 °C-1 to 15 × 10−6 °C-1.

Abstract Ferroptosis is an iron-dependent form of regulated cell death characterized by lipid peroxidation. Colorectal cancer (CRC) cells evade ferroptosis despite their requirement of substantial iron and reactive oxygen species (ROS) to sustain active metabolism and extensive proliferation. However, the underlying mechanism is unclear. Herein, we report the role of lymphoid-specific helicase (LSH), a chromatin-remodeling protein, in suppressing erastin-induced ferroptosis in CRC cells. We demonstrate that erastin treatment leads to dose- and time-dependent downregulation of LSH in CRC cells, and depletion of LSH increases cell sensitivity to ferroptosis. Mechanistically, LSH interacts with and is stabilized by ubiquitin-specific protease 11 (USP11) via deubiquitination; this interaction was disrupted by erastin treatment, resulting in increased ubiquitination and LSH degradation. Moreover, we identified cytochrome P450 family 24 subfamily A member 1 (CYP24A1) as a transcriptional target of LSH. LSH binds to the CYP24A1 promoter, promoting nucleosome eviction and reducing H3K27me3 occupancy, thus leading to transcription of CYP24A1. This cascade inhibits excessive intracellular Ca2+ influx, thereby reducing lipid peroxidation and ultimately conferring resistance to ferroptosis. Importantly, aberrant expression of USP11, LSH, and CYP24A1 is observed in CRC tissues and correlates with poor patient prognosis. Taken together, our study demonstrates the crucial role of the USP11/LSH/CYP24A1 signaling axis in inhibiting ferroptosis in CRC, highlighting its potential as a therapeutic target in CRC treatment.

Remote patient-monitoring systems are helpful since they can provide timely and effective healthcare facilities. Such online telemedicine is usually achieved with the help of sophisticated and advanced wearable sensor technologies. The modern type of wearable connected devices enable the monitoring of vital sign parameters such as: heart rate variability (HRV) also known as electrocardiogram (ECG), blood pressure (BLP), Respiratory rate and body temperature, blood pressure (BLP), respiratory rate, and body temperature. The ubiquitous problem of wearable devices is their power demand for signal transmission; such devices require frequent battery charging, which causes serious limitations to the continuous monitoring of vital data. To overcome this, the current study provides a primary report on collecting kinetic energy from daily human activities for monitoring vital human signs. The harvested energy is used to sustain the battery autonomy of wearable devices, which allows for a longer monitoring time of vital data. This study proposes a novel type of stress- or exercise-monitoring ECG device based on a microcontroller (PIC18F4550) and a Wi-Fi device (ESP8266), which is cost-effective and enables real-time monitoring of heart rate in the cloud during normal daily activities. In order to achieve both portability and maximum power, the harvester has a small structure and low friction. Neodymium magnets were chosen for their high magnetic strength, versatility, and compact size. Due to the non-linear magnetic force interaction of the magnets, the non-linear part of the dynamic equation has an inverse quadratic form. Electromechanical damping is considered in this study, and the quadratic non-linearity is approximated using MacLaurin expansion, which enables us to find the law of motion for general case studies using classical methods for dynamic equations and the suitable parameters for the harvester. The oscillations are enabled by applying an initial force, and there is a loss of energy due to the electromechanical damping. A typical numerical application is computed with Matlab 2015 software, and an ODE45 solver is used to verify the accuracy of the method.

In this paper, based on Meyerhof’s theory of homogeneous foundation, the limit equilibrium analysis method and unified logarithmic spiral sliding surface assumption are used to derive the theoretical formula for the ultimate bearing capacity of a layered foundation when the foundation is completely rough. It should be noted that this formula is only applicable to strip foundations of upper soft clay and lower sandy soil. In addition, a comparative analysis is conducted between theoretical formulas and semiempirical formulas for layered foundations. On the basis of verifying the reliability of the theoretical formula results, numerical simulation is carried out to further explore and analyze the influence of the width to depth ratio of the foundation, the strength parameters of the double-layer soil, and the thickness of the upper soft soil on the bearing capacity of the foundation. Research has shown that the formula for the bearing capacity of a layered foundation derived in this paper has a certain degree of error compared to Meyerhof’s semiempirical formula, but it is in good agreement with numerical simulation results and Hansen’s weighted average method results. The ratio of the width to depth of the foundation, the ratio of the cohesive force of the double-layer soil, and the tangent ratio of the internal friction angle have a significant positive correlation with the ultimate bearing capacity of the foundation. The increase in thickness of the overlying cohesive soil has a negative impact on the ultimate bearing capacity of the foundation, and the thicker the soil, the smaller the foundation’s bearing capacity.

Due to the limited availability of fault samples and the expensive nature of marking fault samples in Electric Multiple Unit (EMU) gearbox monitoring data, a study was conducted to simulate the degradation process of key components in the CRH5 gearbox using rigid–flexible coupling dynamics. Vibration acceleration data from the simulation were utilized to create a six-dimensional hybrid feature domain representing the degradation process. By leveraging the capabilities of the Hidden Markov Model (HMM) for handling hidden transitive probabilities in temporal data and Gated Recurrent Unit (GRU) for addressing long-distance and high-dependence temporal data, a GRU-HMM fault prediction model was developed. This model was validated using monitoring data and the six-dimensional hybrid feature domain from the CRH5 gearbox and compared against actual maintenance records. The findings indicated that the GRU-HMM fault prediction model can effectively recognize the degradation patterns of multiple components, offering higher accuracy in fault prediction compared to traditional models. These research outcomes are expected to optimize EMU maintenance schedules based on usage conditions, enhance EMU utilization rates, and reduce operational and maintenance costs, thereby providing valuable theoretical support.

The WRKY gene family is a key transcription factor family for plant development and the stress response. However, few studies have investigated the WRKY gene family in Chinese rose (Rosa chinensis). In this study, 68 RcWRKY genes were identified from the Chinese rose genome and classified into three primary groups and five subgroups based on the structural and phylogenetic characteristics. The analysis of the conserved domains, motifs, and gene structure revealed that the RcWRKY genes within the same group had the same exon–intron organization and composition. Chromosome mapping and gene duplication revealed that the RcWRKY genes were randomly dispersed across seven chromosomes. Fragment duplication and refined selection may have influenced the evolution of the WRKY gene family in Chinese rose. The cis-acting elements in the WRKY promoter region revealed that the RcWRKY genes contained numerous abiotic stress response elements. The results of qRT-PCR revealed that the expression of RcWRKY was tissue-specific, with high expression being observed under drought, heat, and salt stress. Notably, RcWRKY49′s expression increased more than fivefold following salt stress, indicating that it is a crucial gene mediating the salt stress response of Chinese rose. These findings shed light on the regulatory role of RcWRKY in the growth and development of Chinese rose, and they serve as a foundation for future molecular breeding programs and gene discovery.

NASH (non-alcoholic steatohepatitis) is a severe liver disease characterized by hepatic chronic inflammation that can be associated with the gut microbiota. In this study, we explored the therapeutic effect of Gynostemma pentaphyllum extract (GPE), a Chinese herbal extract, on methionine- and choline-deficient (MCD) diet-induced NASH mice. Based on the peak area, the top ten compounds in GPE were hydroxylinolenic acid, rutin, hydroxylinoleic acid, vanillic acid, methyl vanillate, quercetin, pheophorbide A, protocatechuic acid, aurantiamide acetate, and iso-rhamnetin. We found that four weeks of GPE treatment alleviated hepatic confluent zone inflammation, hepatocyte lipid accumulation, and lipid peroxidation in the mouse model. According to the 16S rRNA gene V3–V4 region sequencing of the colonic contents, the gut microbiota structure of the mice was significantly changed after GPE supplementation. Especially, GPE enriched the abundance of potentially beneficial bacteria such as Akkerrmansia and decreased the abundance of opportunistic pathogens such as Klebsiella. Moreover, RNA sequencing revealed that the GPE group showed an anti-inflammatory liver characterized by the repression of the NF-kappa B signaling pathway compared with the MCD group. Ingenuity Pathway Analysis (IPA) also showed that GPE downregulated the pathogen-induced cytokine storm pathway, which was associated with inflammation. A high dose of GPE (HGPE) significantly downregulated the expression levels of the tumor necrosis factor-α (TNF-α), myeloid differentiation factor 88 (Myd88), cluster of differentiation 14 (CD14), and Toll-like receptor 4 (TLR4) genes, as verified by real-time quantitative PCR (RT-qPCR). Our results suggested that the therapeutic potential of GPE for NASH mice may be related to improvements in the intestinal microenvironment and a reduction in liver inflammation.

Officers on Watch (OOWs) of the ship’s bridge play a vital role in maritime navigation safety, monitoring the ship’s navigational status, and ensuring maritime safety. The status of inactive watch officers, such as fatigued driving and negligence on lookout, is one of the main causes of accidents. Intelligent technology for real-time perception and state evaluation of ship OOWs significantly reduces accidents caused by human factors. The traditional computer vision method is difficult to adapt to the complex environment of a ship bridge, and carries strong privacy risks. With the development of Internet of Things technology, sensing technology based on ubiquitous Wi-Fi devices provides a new way to accurately monitor the status of ship OOWs. In this paper, we use commercial off-the-shelf (COTS) Wi-Fi devices to propose a ship driving activity state detection method based on beamforming feedback information (BFI). Using wireless sensing data to sense the number of OOWs and their driving behavior realizes low-cost and high-precision detection of the behavioral status of the ship’s bridge watchkeeper. Experiments were conducted in a ship-driving simulation laboratory and on a real-world Yangtze River cruise ship. The experimental results demonstrate that our proposed method achieves 92.4% and 98.1% accuracy for tracking active status and estimating the number of OOWs, respectively.

In order to study the adsorption process and adsorption characteristics of shale oil at the macro scale, the isothermal adsorption experiments of illite and kerogen on a heptadecane (oil phase) solution were carried out by infrared spectrophotometry and gas chromatography–mass spectrometry. Based on the adsorption isotherm model and adsorption thermodynamic model, the characteristics of heptadecane adsorbed by illite and kerogen at different temperatures and oily solution concentrations were studied. The experimental results show that the concentration and temperature of the alkane solution help to enhance the adsorption and increase the saturated adsorption capacity. The difference is that the concentration will have a certain effect on the adsorption rate, while the temperature will not. Based on the three adsorption isotherm models, it was found that Langmuir and Freundlich were more suitable for describing the adsorption process of the heptadecane solution by illite and kerogen, and the adsorption characteristics of heptadecane molecules at different temperatures and adsorbents were evaluated. Heating leads to an increase in the collision efficiency between adsorbate molecules and adsorbents, thereby accelerating the migration rate of alkanes. Therefore, increasing temperature helps to enhance the adsorption capacity of rocks and increase the saturated adsorption capacity of minerals. The research results clarify the adsorption characteristics of shale oil heavy components from the macro level and fill the research gap in the application of solid–liquid isothermal adsorption physical experiments on the adsorption and occurrence of shale oil.

Single-shot dual-wavelength interferometry offers a promising avenue for surface profile measurement of dynamic objects. However, current techniques employing pixel multiplexing or color cameras encounter challenges such as complex optical alignment, limited measurement range, and difficulty in measuring rough surfaces. To address these issues, this study presents a novel approach to single-shot dual-wavelength interferometry. By utilizing separated polarization illumination and detection, along with a monochromatic polarization camera and two slightly different wavelengths, this method enables the simultaneous recording of two frames of separated interferometric patterns. This approach facilitates straightforward optical alignment, expands measurement ranges, accelerates data acquisition, and simplifies data processing for dual-wavelength interferometry. Consequently, it enables online shape measurement of large dynamic samples with rough surfaces.

Rabies virus (RABV) is a neurotropic virus that causes fatal neurological disease, raising serious public health issues and attracting extensive attention in society. To elucidate the molecular mechanism of RABV-induced neuronal damage, we used hematoxylin–eosin staining, transmission electron microscopy, transcriptomics analysis, and immune response factor testing to investigate RABV-infected neurons. We successfully isolated the neurons from murine brains. The specificity of the isolated neurons was identified by a monoclonal antibody, and the viability of the neurons was 83.53–95.0%. We confirmed that RABV infection induced serious damage to the neurons according to histochemistry and transmission electron microscope (TEM) scanning. In addition, the transcriptomics analysis suggested that multiple genes related to the pyroptosis pathway were significantly upregulated, including gasdermin D (Gsdmd), Nlrp3, caspase-1, and IL-1β, as well as the chemokine genes Ccl2, Ccl3, Ccl4, Ccl5, Ccl7, Ccl12, and Cxcl10. We next verified this finding in the brains of mice infected with the rRC-HL, GX074, and challenge virus standard strain-24 (CVS-24) strains of RABV. Importantly, we found that the expression level of the Gsdmd protein was significantly upregulated in the neurons infected with different RABV strains and ranged from 691.1 to 5764.96 pg/mL, while the basal level of mock-infected neurons was less than 100 pg/mL. Taken together, our findings suggest that Gsdmd-induced pyroptosis is involved in the neuron damage caused by RABV infection.

In response to the inefficiencies and high costs associated with manual buoy inspection, this paper presents the design and testing of an Autonomous Navigation Unmanned Surface Vehicle (USV) tailored for this purpose. The research is structured into three main components: Firstly, the hardware framework and communication system of the USV are detailed, incorporating the Robot Operating System (ROS) and additional nodes to meet practical requirements. Furthermore, a buoy tracking system utilizing the Kernelized Correlation Filter (KCF) algorithm is introduced. Secondly, buoy image training is conducted using the YOLOv7 object detection algorithm, establishing a robust model for accurate buoy state recognition. Finally, an improved Line-of-Sight (LOS) method for USV path tracking, assuming the presence of an attraction potential field around the inspected buoy, is proposed to enable a comprehensive 360-degree inspection. Experimental testing includes validation of buoy image target tracking and detection, assessment of USV autonomous navigation and obstacle avoidance capabilities, and evaluation of the enhanced LOS path tracking algorithm. The results demonstrate the USV’s efficacy in conducting practical buoy inspection missions. This research contributes insights and advancements to the fields of maritime patrol and routine buoy inspections.

Space–time adaptive processing (STAP) is an effective technology in clutter suppression and moving target detection for airborne radar. When working in the heterogeneous environment, the number of training samples that satisfy independent and identically distributed (IID) conditions is insufficient, making it difficult to ensure the estimation accuracy of the clutter plus noise covariance matrix for traditional STAP methods. Sparse recovery–based STAP (SR–STAP) methods have received widespread attention in the past few years. The accurate estimation of the clutter plus noise covariance matrix can be achieved using only a few training samples. The iterative adaptive approach (IAA) can quickly and accurately estimate the power spectrum, but applying this method directly to the STAP method cannot produce good performance. In this paper, a fast IAA–based SR–STAP method is proposed. Based on the weighted l1 problem, the IAA spectrum is used as a weighted term to obtain a good approximation. In order to obtain an analytical solution, we use the weighted l2 norm to approximate the weighted l1 norm without loss of performance. Compared with the IAA–STAP method, the proposed method is more robust to errors. Moreover, the proposed method has a fast computational speed. The effectiveness of the proposed method is demonstrated by simulations.

At present, most studies use the direct method to analyze the oscillation problem of modern power systems. However, these studies often only simplify the external characteristics of the wind turbine and lack an in-depth understanding of its internal refined energy structure. In this paper, based on the direct-drive permanent magnetic synchronous generator’s detailed model (D-PMSG), combined with the dynamic energy of its port, layers of analysis are performed on the wind turbine’s internal connections, and a detailed model of the energy structure is created. Then, the interaction mechanism of each control link in the wind turbine is analyzed by combining the energy function of the wind turbine with the improved perturbation method. Finally, this paper constructs a sub-synchronous oscillation (SSO) scenario of weak damping and a forcing type and proves the accuracy and effectiveness of the traceability method based on the refined energy of D-PMSG. This traceability method based on refined energy is expected to provide a new solution to the stability problem caused by the integration of new energy.

Abstract Objective and design An accumulating body of evidence has shown that gut microbiota is involved in regulating inflammation; however, it remains undetermined if and how gut microbiota plays an important role in modulating deep venous thrombosis (DVT), which is an inflammation-involved thrombotic event. Subjects Mice under different treatments were used in this study. Methods and treatment We induced stenosis DVT in mice by partially ligating the inferior vena cava. Mice were treated with antibiotics, prebiotics, probiotics, or inflammatory reagents to modulate inflammatory states, and their effects on the levels of circulating LPS and DVT were examined. Results Antibiotic-treated mice or germ-free mice exhibited compromised DVT. Treatment of mice with either prebiotics or probiotics effectively suppressed DVT, which was accompanied with the downregulation of circulating LPS. Restoration of circulating LPS in these mice with a low dose of LPS was able to restore DVT. LPS-induced DVT was blocked by a TLR4 antagonist. By performing proteomic analysis, we identified TSP1 as one of the downstream effectors of circulating LPS in DVT. Conclusion These results suggest that gut microbiota may play a nonnegligible role in modulating DVT by leveraging the levels of LPS in circulation, thus shedding light on the development of gut microbiota-based strategies for preventing and treating DVT.

Abstract Background Targeted therapy combined with immunotherapy is the current first-line treatment for metastatic renal cell carcinoma (mRCC), but patients with tumor thrombus (TT) may suffer from lower limb edema or even sudden cardiac death, so the purpose of this study is to investigate the efficacy and safety of surgical treatment in patients with mRCC and TT and explore worse factors to affect the prognosis in this series of patients. Patients and methods A total of 85 mRCC patients with TT who received cytoreductive nephrectomy and thrombectomy at our medical center from 2014 to 2023 are included. All patients received postoperative systemic therapy. Overall survival (OS) is defined as the time from surgery to death due to any reason or the last follow-up. Kaplan–Meier analysis was performed to evaluate OS and differences among groups were tested by log-rank. Multivariable Cox proportional hazards analysis was performed to ascertain independent relationships between clinicopathological factors and OS. Results The median age of patients was 58 years old. Eleven patients (12.9%) had no symptoms, 39 patients (45.9%) had local symptoms, 15 patients (17.6%) had systemic symptoms, and 20 patients (23.5%) had both. Mayo grade of TT was 0, 1, 2, 3, and 4 for 12, 27, 31, 7, and 8 patients respectively. Fifty-five patients had lung metastasis, 23 had bone metastasis, 16 had liver metastasis, 13 had adrenal metastasis, and 9 had lymph node metastasis. Of all patients, 17 patients had multiple metastases. The median operation time is 289 min and the median intraoperative hemorrhage is 800 ml. Twenty-eight patients experienced postoperative complications, 8 of which were serious complications of modified Clavien grade III or higher. The median OS of all patients was 33 months and median follow up time was 26 months. In multivariate analysis, systemic symptom (p = 0.00753), pathological type (p = 0.0166), sarcomatous degeneration (p = 0.0334), and perirenal fat infiltration (p = 0.0202) are independent predictors of OS. Conclusion Cytoreductive nephrectomy and thrombectomy is relatively safe and effective for patients with mRCC accompanied by TT. In this series of patients, the worse prognosis is associated with systemic symptoms, non-clear cell carcinoma, sarcomatous degeneration and perirenal fat infiltration.

ABSTRACTWe for the first time demonstrate ground-based remote sensing of Nitrous Oxide (N2O) over Hefei in eastern China from high resolution Fourier Transform Infra-Red (FTIR) solar spectra. We have retrieved Column-averaged Abundance of N2O ([Formula: see text]) from both Near-Infrared (NIR, 4000 to 11,000 cm−1) and Mid-Infrared (MIR, 2400 to 3200 cm−1) solar spectra and inspected their agreement. Generally, NIR and MIR measurements agree well with a correlation coefficient of 0.86 and an average difference of (1.33 ± 4.05) ppbv (NIR – MIR). By correcting the bias of the two datasets, we combine the NIR and MIR measurements to investigate seasonality and inter-annual trend of [Formula: see text] over Hefei. The observed monthly mean time series of [Formula: see text] minimize in June and maximize in September, with values of (316.55 ± 12.22) ppbv and (322.05 ± 12.93) ppbv, respectively. The [Formula: see text] time series from 2015 to 2020 showed an inter-annual trend of (0.53 ± 0.10) %/year over Hefei, China. We also compared the FTIR [Formula: see text] observations with GEOS-Chem model [Formula: see text] simulations. They are in reasonable agreement with a correlation coefficient (R) of 0.71, but GEOS-Chem model underestimated the seasonality of the observations. This study can enhance current knowledge of ground-based high-resolution FTIR remote sensing of N2O in the atmosphere and contribute to generating a new reliable N2O dataset for climate change research.

Abstract The transcription factor hypoxia-inducible factor-1α (HIF-1α), as a master regulator of adaptive responses to hypoxia, possesses two transcriptional activation domains [TAD, N-terminal (NTAD), and C-terminal (CTAD)]. Although the roles of HIF-1α NTAD in kidney diseases have been recognized, the exact effects of HIF-1α CTAD in kidney diseases are poorly understood. Here, two independent mouse models of hypoxia-induced kidney injury were established using HIF-1α CTAD knockout (HIF-1α CTAD−/−) mice. Furthermore, hexokinase 2 (HK2) and mitophagy pathway are modulated using genetic and pharmacological methods, respectively. We demonstrated that HIF-1α CTAD−/− aggravated kidney injury in two independent mouse models of hypoxia-induced kidney injury, including ischemia/reperfusion-induced kidney injury and unilateral ureteral obstruction-induced nephropathy. Mechanistically, we found that HIF-1α CTAD could transcriptionally regulate HK2 and subsequently ameliorate hypoxia-induced tubule injury. Furthermore, it was found that HK2 deficiency contributed to severe renal injury through mitophagy inhibition, while mitophagy activation using urolithin A could significantly protect against hypoxia-induced kidney injury in HIF-1α C-TAD−/− mice. Our findings suggested that the HIF-1α CTAD-HK2 pathway represents a novel mechanism of kidney response to hypoxia, which provides a promising therapeutic strategy for hypoxia-induced kidney injury.

The application of commercially pure titanium (CP–Ti) has long been limited by its insufficient strength and plastic instability. However, strength enhancement typically comes at the expense of ductility since the two important mechanical properties are often mutually exclusive. In the present work, multi-element Fe–Cr–Ni–Mo in the form of 316L steel was added into CP-Ti by additive manufacturing (AM), so as to realize the simultaneous enhancement of strength and uniform ductility. The localized homogenization of CP-Ti and 316L alloy melts during laser melting deposition produced the microstructure comprised of basket-weave ɑ laths, discontinuous ɑGB, and retained β phases, which endowed Ti-1.35Fe-0.36Cr-0.24Ni-0.05Mo alloy with related higher ultimate tensile strength (635 MPa) and larger uniform ductility (12%). This approach could provide guidance for designing high-performance metal materials starting with elements powder through additive manufacturing.

AIM: To compare the efficacy and safety of intravitreal injection of aflibercept combined with posterior sub-fascial injection of triamcinolone acetonide in the treatment of wet age-related macular degeneration(ARMD)with poor response to anti-vascular endothelial growth factor drugs.METHODS: Retrospective cohort study. From June 2018 to May 2020, a total of 60 patients(60 eyes)with refractory ARMD who had poor response to the treatment of anti VEGF drug ranibizumab were randomly divided into the control group of aflibercept and the observation group of triamcinolone acetonide combined with aflibercept, with 30 patients(30 eyes)in each group. Once a month, the patients in the two groups received intravitreal injection of aflibercept alone or intravitreal injection of aflibercept combined with posterior sub-fascial injection of triamcinolone acetonide for three consecutive times. Abstract Objective This study aimed to examine the effect of exercise intervention for antenatal depression using meta-analysis and to propose the best exercise intervention program. Methods Review Manager 5.3 was used to analyze 17 papers with 2224 subjects by setting five moderators, including type, time, frequency, period, and format of exercise intervention, and a random-effects model was used to test for overall effect, heterogeneity, and publication bias. Results (1) The effect size of the exercise intervention on antenatal depression was d = -0.56, which reached a good effect and was statistically significant; b (2) The effect size of the exercise type on antenatal depression was Yoga and a combination of aerobic exercise in order of intervention; (3) the single intervention duration of 10–75 min all had a good effect on antenatal depression, and 30–60 min had the best effect; (4) the intervention frequency of 3 to 5 times/week had the greatest amount of intervention effect on maternal depression; (5) exercise lasting 6–10 weeks had a good intervention effect on antepartum depression, and the amount of effect decreased gradually with the extension of time; (6) In terms of exercise format, the amount of intervention effect on maternal depression was in the order of group exercise, individual + group exercise. Conclusions Exercise intervention can significantly alleviate antenatal depression symptoms. The best exercise program for exercise intervention for antenatal depression is: Yoga and a combination of aerobic exercise intervention effects are more prominent, and the intervention effect of Yoga is the best. The use of group exercise 3–5 times per week for 30–60 min for 6–10 weeks was more likely to achieve the desired intervention effect of improving antenatal depression.

Abstract The survival of tumor cells in the bloodstream, and vasculature adhesion at metastatic sites are crucial for tumor metastasis. Perivascular invasion aids tumor cell self-renewal, survival, and formation of metastases by facilitating readily available oxygen, nutrients, and endothelial-derived paracrine factors. Renal cell carcinoma (RCC) is among the most prevalent tumors of the urinary system, and the formation of venous tumor thrombus (VTT) is a characteristic feature of RCC. We observed high expression of L1CAM in the VTT with vessel wall invasion. L1CAM promotes the adhesion, migration, and invasion ability of RCC and enhances metastasis by interacting with ITGA5, which elicits activation of signaling downstream of integrin α5β1. L1CAM promotes ADAM17 transcription to facilitate transmembrane ectodomain cleavage and release of soluble L1CAM. In response to soluble L1CAM, vascular endothelial cells release several cytokines and chemokines. Endothelial-derived CXCL5 and its receptor CXCR2 promote the migration and intravasation of RCC toward endothelial cells suggesting that crosstalk between endothelial cells and tumor cells has a direct guiding role in driving the metastatic spread of RCC. LICAM plays a crucial role in the invasive ability of RCC, and regulation of L1CAM expression may contribute therapeutically to preventing RCC progression.

Paired SpCas9 nickases (SpCas9n) are an effective strategy to reduce off-target effect in genome editing. However, this approach is not efficient with 3′-overhanging ends, limiting its applications. In order to expand the utility of paired SpCas9n in genome editing, we tested the effect of the TREX2 3′-5′ exonuclease on repair of 3′-overhanging ends. We found ectopic overexpression of Trex2 stimulates the efficiency of paired SpCas9n in genome disruption with 3′-overhanging ends up to 400-fold with little stimulation of off-target editing. TREX2 overexpressed preferentially deletes entire 3′ overhangs but has no significant effect on 5′ overhangs. Trex2 overexpression also stimulates genome disruption by paired SpCas9n that potentially generate short 3′-overhanging ends at overlapping SpCas9n target sites, suggesting sequential nicking of overlapping target sites by SpCas9n. This approach is further simplified with improved efficiency and safety by fusion of TREX2 and particularly its DNA-binding-deficient mutant to SpCas9n. Junction analysis at overlapping targets revealed the different extent of end resection of 3′ single-stranded DNA (ssDNA) by free TREX2 and TREX2 fused to SpCas9n. SpCas9n-TREX2 fusion is more convenient and safer than overexpression of free TREX2 to process 3′-overhanging ends for efficient genome disruption by paired SpCas9n, allowing practical use of this TREX2-based strategy in genome editing.

Objectives: This study aims to identify the key senescence genes and potential regulatory mechanisms that contribute to the etiology of intervertebral disc degeneration (IDD).Method: We analyzed GSE34095 and GSE70362 datasets, identifying key senescence-related differentially expressed genes (DEGs) in IDD using lasso regression. Risk scores classified patients into high- and low-risk groups. We compared pathways, functions, and immune infiltration between these groups. Diagnostic ability was assessed using ROC curves and a nomogram predicted IDD incidence. In single-cell dataset GSE165722, we evaluated expression of key senescence-related DEGs.Results: We identified 12 key senescence-related DEGs distinguishing high- and low-risk IDD patients. Enrichment analysis revealed cellular stress response, apoptotic signaling pathway, and protein kinase activation differences. Immune cell analysis showed elevated eosinophils in low-risk group and increased effector memory CD8 T, central memory CD4 T, myeloid-derived suppressor, natural killer, monocyte, Type 1 T helper, plasmacytoid dendritic, and natural killer T cells in high-risk group. A nomogram using AUC >0.75 genes (CXCL8, MAP4K4, MINK1, and TNIK) predicted IDD incidence with good diagnostic power. High senescence scores were observed in neutrophils.Conclusion: Our diagnostic model, based on key senescence-related DEGs and immune cell infiltration, offers new insights into IDD pathogenesis and immunotherapy strategies.

Background: The 18F-fluoroestradiol positron emission tomography/computed tomography (18F-FES PET/CT) technique provides a convenient method to evaluate the overall estrogen receptor (ER) expression in metastatic breast cancer (MBC) patients. There are long debates on the characteristics and treatment strategy of patients with positive primary ER lesions but negative ER expression in metastatic disease. 18F-FES PET offers an opportunity to answer this question. Objectives: This study aimed to characterize the primary ER-positive patients with advanced-stage FES negativity and investigate the real-world treatment decisions made by physicians subsequently, and compare the efficacy between different regimens. Design: This observational cohort study was conducted at Fudan University Shanghai Cancer Center, enrolling breast cancer patients with ER-positive primary tumors who showed advanced-stage FES negativity. Methods: Descriptive statistics were used in clinicopathologic characteristics and compared with a chi-square test or t -test. In addition, progression-free survival (PFS) was estimated by the Kaplan-Meier method and compared by log-rank test. Results: 16.6% (52/314) of patients with an ER-positive primary tumor had negative ER expression assessed by 18F-FES for MBC prior to receiving first-line systemic therapy, among whom adjuvant endocrine therapy was prevalently utilized (86.5%, 45/52). The rate of FES negativity in the advanced stage was negatively correlated with levels of ER expression of primary tumors. Chemotherapy (83.3%, 40/48) was the most common treatment strategy afterward, among which capecitabine monotherapy (62.5%, 25/40) was a dominant alternative. PFS was significantly prolonged with capecitabine alone versus other chemotherapy (median PFS: 13.14 versus 6.21 months, p = 0.029). Conclusion: Negative conversion of ER in MBC detected by 18F-FES occurred frequently. Patients with lower ER expression in the primary lesion were more likely to have negative ER expression in the metastasis. In real-world clinical practice, most physicians primarily opted for chemotherapy, with capecitabine monotherapy being a commonly selected regimen. Introduction We explored the relationship and the predictive value of serum fibroblast growth factor 21 (FGF21) with all-cause mortality, major adverse cardiovascular events (MACEs) and pneumonia in hemodialysis (HD) patients.Methods A total of 388 Chinese HD patients from two HD centers were finally enrolled in this prospective cohort study (registration number: ChiCTR 1900028249) between January 2018 and December 2018. Serum FGF21 was detected. Patients were followed up with a median period of 47 months to record the MACEs and pneumonia until death or 31 December 2022.Results The incidence of all-cause mortality, MACEs and pneumonia in HD patients were 20.6%, 29.6%, and 34.8%, respectively. The optimal cutoffs for FGF21 to predict all-cause mortality, MACEs and pneumonia were 437.57 pg/mL, 216.99 pg/mL and 112.79 pg/mL. The city wall is a military defense building of an ancient city and a unique landscape architecture of a modern country that has extremely important historical, artistic, and scientific values. Most ancient Chinese walls were built of bricks and have been severely damaged and degraded by environmental and human influences. To protect these historical building monuments, it is necessary to determine scientifically the material properties of the bricks used in the ancient walls. In this study, the physical and mechanical properties, chemical element composition, mineral composition, micromorphology, and pore structure of 20 ancient fired clay bricks from four different regions, including white and blue wall bricks from Nanjing, blue wall bricks from Xi'an and Linqing, and the Great Wall bricks from Beijing, were compared using various modern analysis and testing methods. The results show that talc could be one of the raw materials to produce white bricks and the firing temperature of some white bricks could exceed 1200 ℃. The ancient blue bricks were mainly composed of quartz and feldspar, whereas the ancient white bricks were mainly composed of quartz and long prismatic and rod-like enstatite. The solid density and compressive strength of the white bricks were higher than that of the blue bricks. The proportion of pores larger than 10 µm in all ancient bricks is less than 10%. The pores smaller than 0.1 µm is less than 6% at higher firing temperatures. The results of this study can provide scientific guidance for the restoration and conservation of fired clay bricks of ancient city wall.

Abstract Background and aim Huntington's disease (HD) is an autosomal dominant inherited neurodegenerative disorder caused by CAG repeats expansion. Cognitive decline contributes to the loss of daily activity in manifest HD. We aimed to examine the cognition status in a Chinese HD cohort and explore factors influencing the diverse cognitive domains. Methods A total of 205 participants were recruited in the study with the assessment by neuropsychological batteries, including the mini–mental state examination (MMSE), Stroop test, symbol digit modalities test (SDMT), trail making test (TMT), verbal fluency test (VFT), and Hopkins verbal learning test–revised, as well as motor and psychiatric assessment. IntroductionContrast-induced nephropathy (CIN) is a common complication of percutaneous coronary intervention (PCI). Identifying patients at high CIN risk remains challenging. The triglyceride-glucose (TyG) index may help predict CIN but evidence is limited. We conducted a meta-analysis to evaluate the diagnostic value of TyG index for CIN after PCI.MethodsA systematic literature search was performed in MEDLINE, Cochrane, and EMBASE until August 2023 (PROSPERO registration: CRD42023452257). Observational studies examining TyG index for predicting CIN risk in PCI patients were included. This diagnostic meta-analysis aimed to evaluate the accuracy of the TyG index in predicting the likelihood of CIN. Secondary outcomes aimed to assess the pooled incidence of CIN and the association between an elevated TyG index and the risk of CIN.ResultsFive studies (Turkey, n=2; China, n=3) with 3518 patients (age range: 57.6 to 68.22 years) were included. The pooled incidence of CIN was 15.3% [95% confidence interval (CI) 11-20.8%]. A high TyG index associated with increased CIN risk (odds ratio: 2.25, 95% CI 1.82-2.77). Pooled sensitivity and specificity were 0.77 (95% CI 0.59-0.88) and 0.55 (95% CI 0.43-0.68) respectively. Analysis of the summary receiver operating characteristic (sROC) curve revealed an area under the curve of 0.69 (95% CI 0.65-0.73). There was a low risk of publication bias (p = 0.81).ConclusionThe TyG index displayed a noteworthy correlation with the risk of CIN subsequent to PCI. However, its overall diagnostic accuracy was found to be moderate in nature. While promising, the TyG index should not be used in isolation for CIN screening given the heterogeneity between studies. In addition, the findings cannot be considered conclusive given the scarcity of data. BackgroundSystemic inflammation is one of the underlying mechanisms of cognitive impairment. The neutrophil-to-lymphocyte ratio (NLR) has emerged as a systemic inflammation indicator. This meta-analysis aimed to evaluate the association between high NLR and cognitive impairment (CI) risk.MethodA comprehensive systematic search was conducted to identify eligible studies published until May 30, 2023. The reference group comprised patients with the lowest NLR level, whereas the exposure group comprised those with the highest NLR level. The main outcome was to examine the relationship between NLR and CI risk. The secondary outcome included the association between patient characteristics or comorbidities and CI risk.ResultsThis meta-analysis included 11 studies published between 2018 and 2023, involving 10,357 patients. The incidence of zoonotic diseases, such as coronavirus disease 2019 and Ebola virus disease, is increasing worldwide. However, drug and vaccine development for zoonotic diseases has been hampered because the experiments involving live viruses are limited to high-containment laboratories. The Ebola virus minigenome system enables researchers to study the Ebola virus under BSL-2 conditions. Here, we found that the addition of the nucleocapsid protein of human coronaviruses, such as severe acute respiratory syndrome coronavirus 2, can increase the ratio of green fluorescent protein-positive cells by 1.5–2 folds in the Ebola virus minigenome system. Further analysis showed that the nucleocapsid protein acts as an activator of the Ebola virus minigenome system. Here, we developed an EBOV MiniG Plus system based on the Ebola virus minigenome system by adding the SARS-CoV-2 nucleocapsid protein. By evaluating the antiviral effect of remdesivir and rupintrivir, we demonstrated that compared to that of the traditional Ebola virus minigenome system, significant concentration-dependent activity was observed in the EBOV MiniG Plus system. Taken together, these results demonstrate the utility of adding nucleocapsid protein to the Ebola virus minigenome system to create a powerful platform for screening antiviral drugs against the Ebola virus.

Background The presence of intraventricular hemorrhage (IVH) was extensively investigated and was associated with poor outcome in patients with intracerebral hemorrhage (ICH). However, the effect of the speed of ventricular bleeding on outcomes is unknown. Methods and Results We prospectively included patients with ICH who had baseline computed tomography scans within 6 hours after ictus between January 2016 and October 2021. The clinical characteristics were compared between patients with and without early neurologic deterioration (END). Ultraearly IVH growth (uIVHG) was defined as baseline IVH volume by onset‐to‐imaging time. The association between uIVHG and outcomes was assessed by using multivariable logistic regression analysis. We established the ultraearly IVH growth (uIVH) score and compared the areas under the receiver operating characteristic curves of the existing scores for predicting END. A total of 299 patients were finally enrolled. Of those, 38 patients (12.7%) experienced END at 24 hours and 89 patients (29.8%) had poor outcomes at 90 days. After adjustment for confounding factors, uIVHG (odds ratio, 1.061 [95% CI, 1.011–1.113]; P=0.016) was independently associated with END in multivariable analysis. A prediction score was developed on the basis of the logistic model. The uIVH score was developed as a sum of individual points (0–6) based on age, hematoma volume, National Institutes of Health Stroke Scale, hematoma expansion, and uIVHG ≥2.5 mL/h. In comparison with the ICH score and modified Emergency Department ICH Scale, the uIVH score exhibited best performance in the prediction of END. Conclusions uIVHG is associated with early neurologic deterioration and poor functional outcome in patients with ICH.

Historical aflatoxin (AFL) and fumonisin (FUM) mycotoxin contamination data of corn, daily weather data, satellite data, dynamic geospatial soil properties, and land usage parameters were modeled to identify factors significantly contributing to the outbreaks of mycotoxin contamination of corn grown in Illinois (IL), AFL >20 ppb, and FUM >5 ppm. Two methods were used: a gradient boosting machine (GBM) and a neural network (NN). Both the GBM and NN models were dynamic at a state-county geospatial level because they used GPS coordinates of the counties linked to soil properties. GBM identified temperature and precipitation prior to sowing as significant influential factors contributing to high AFL and FUM contamination. AFL-GBM showed that a higher aflatoxin risk index (ARI) in January, March, July, and November led to higher AFL contamination in the southern regions of IL. Higher values of corn-specific normalized difference vegetation index (NDVI) in July led to lower AFL contamination in Central and Southern IL, while higher wheat-specific NDVI values in February led to higher AFL. FUM-GBM showed that temperature in July and October, precipitation in February, and NDVI values in March are positively correlated with high contamination throughout IL. Furthermore, the dynamic geospatial models showed that soil characteristics were correlated with AFL and FUM contamination. Greater calcium carbonate content in soil was negatively correlated with AFL contamination, which was noticeable in Southern IL. Greater soil moisture and available water-holding capacity throughout Southern IL were positively correlated with high FUM contamination. The higher clay percentage in the northeastern areas of IL negatively correlated with FUM contamination. NN models showed high class-specific performance for 1-year predictive validation for AFL (73%) and FUM (85%), highlighting their accuracy for annual mycotoxin prediction. Our models revealed that soil, NDVI, year-specific weekly average precipitation, and temperature were the most important factors that correlated with mycotoxin contamination. These findings serve as reliable guidelines for future modeling efforts to identify novel data inputs for the prediction of AFL and FUM outbreaks and potential farm-level management practices.

BackgroundMild cognitive impairment (MCI) is an intermediate stage between normal ageing and dementia. The early identification of MCI is important for timely intervention. The visual cognitive assessment test (VCAT) is a brief language-neutral screening tool for detecting MCI/mild dementia. This meta-analysis evaluated the diagnostic efficacy of the VCAT for MCI/mild dementia.MethodsMedline, Embase, Google Scholar, and Cochrane Library were searched from their inception until August 2023 to identify studies using VCAT to diagnose MCI/mild dementia. The primary outcome was to assess the diagnostic accuracy of the VCAT for detecting MCI/mild dementia through area under the receiver operating characteristic curve (AU-ROC) analysis. The secondary outcome was to explore the correlation between VCAT scores and MCI/mild dementia presence by comparing scores among patients with and without MCI/mild dementia. Pooled sensitivity, specificity, and area under the curve (AUC) were calculated.ResultsFive studies with 1,446 older adults (mean age 64–68.3 years) were included. The percentage of participants with MCI/mild dementia versus controls ranged from 16.5% to 87% across studies. All studies were conducted in Asian populations, mostly Chinese, in Singapore and Malaysia. BackgroundChatGPT is a powerful pretrained large language model. It has both demonstrated potential and raised concerns related to knowledge translation and knowledge transfer. To apply and improve knowledge transfer in the real world, it is essential to assess the perceptions and acceptance of the users of ChatGPT-assisted training. ObjectiveWe aimed to investigate the perceptions of health care trainees and professionals on ChatGPT-assisted training, using biomedical informatics as an example. MethodsWe used purposeful sampling to include all health care undergraduate trainees and graduate professionals (n=195) from January to May 2023 in the School of Public Health at the National Defense Medical Center in Taiwan. Subjects were asked to watch a 2-minute video introducing 5 scenarios about ChatGPT-assisted training in biomedical informatics and then answer a self-designed online (web- and mobile-based) questionnaire according to the Kirkpatrick model. The survey responses were used to develop 4 constructs: “perceived knowledge acquisition,” “perceived training motivation,” “perceived training satisfaction,” and “perceived training effectiveness.” The study used structural equation modeling (SEM) to evaluate and test the structural model and hypotheses. ResultsThe online questionnaire response rate was 152 of 195 (78%); 88 of 152 participants (58%) were undergraduate trainees and 90 of 152 participants (59%) were women. The ages ranged from 18 to 53 years (mean 23.3, SD 6.0 years). There was no statistical difference in perceptions of training evaluation between men and women. Most participants were enthusiastic about the ChatGPT-assisted training, while the graduate professionals were more enthusiastic than undergraduate trainees. Nevertheless, some concerns were raised about potential cheating on training assessment. The average scores for knowledge acquisition, training motivation, training satisfaction, and training effectiveness were 3.84 (SD 0.80), 3.76 (SD 0.93), 3.75 (SD 0.87), and 3.72 (SD 0.91), respectively (Likert scale 1-5: strongly disagree to strongly agree). Knowledge acquisition had the highest score and training effectiveness the lowest. In the SEM results, training effectiveness was influenced predominantly by knowledge acquisition and partially met the hypotheses in the research framework.
