Nanopore sequencing from Oxford Nanopore Technologies (ONT) is a promising third-generation sequencing (TGS) technology that generates relatively longer sequencing reads compared to the next-generation sequencing (NGS) technology. A basecaller is a piece of software that translates the original electrical current signals into nucleotide sequences. The accuracy of the basecaller is crucially important to downstream analysis. Bonito is a deep learning-based basecaller recently developed by ONT. Its neural network architecture is composed of a single convolutional layer followed by three stacked bidirectional gated recurrent unit (GRU) layers. Although Bonito has achieved state-of-the-art base calling accuracy, its speed is too slow to be used in production. We therefore developed Fast-Bonito, by using the neural architecture search (NAS) technique to search for a brand-new neural network backbone, and trained it from scratch using several advanced deep learning model training techniques. The new Fast-Bonito model balanced performance in terms of speed and accuracy. Fast-Bonito was 153.8% faster than the original Bonito on NVIDIA V100 GPU. When running on HUAWEI Ascend 910 NPU, Fast-Bonito was 565% faster than the original Bonito. The accuracy of Fast-Bonito was also slightly higher than that of Bonito. We have made Fast-Bonito open source, hoping it will boost the adoption of TGS in both academia and industry.

Omics data can be used to identify biological characteristics from genetic to phenotypic levels during the life span of a living being, while molecular interaction networks have a fundamental impact on life activities. Integrating omics data and molecular interaction networks will help researchers delve into comprehensive information hidden in the data. Here, we propose a new multimodal method — AutoGGN — to integrate multi-omics data with molecular interaction networks based on graph convolutional neural networks (GCNs). We evaluated AutoGGN using three classification tasks: single-cell embryonic developmental stage classification, pan-cancer type classification, and breast cancer subtyping. On all three tasks, AutoGGN showed better performance than other methods. This means AutoGGN has the potential to extract insights more effectively by means of integrating molecular interaction networks with multi-omics data. Additionally, in order to provide a better understanding of how our model makes predictions, we utilized the SHAP module and identified the key genes contributing to the classification, providing insight for the design of downstream biological experiments.

Abstract Background Periprosthetic joint infection (PJI) is the most serious complication of joint replacement surgery. Further comorbidities include bedsore, deep vein thrombosis, reinfection, or even death. An increasing number of researchers are focusing on this challenging complication. The aim of the present study was to estimate global PJI research based on bibliometrics from meta-analysis studies. Methods A database search was performed in PubMed, Scopus, and Web of Science. Relevant studies were assessed using the bibliometric analysis. Results A total of 117 articles were included. The most relevant literature on PJI was found on Scopus. China made the highest contributions to global research, followed by the USA and the UK. The institution with the most contributions was the University of Bristol. The journal with the highest number of publications was The Journal of Arthroplasty, whereas the Journal of Clinical Medicine had the shortest acceptance time. Furthermore, the top three frequently used databases were Embase, MEDLINE, and Cochrane. The most frequent number of authors in meta-analysis studies was four. Most studies focused on the periprosthetic hip and knee. The alpha-defensin diagnostic test, preventive measures on antibiotics use, and risk factors of intra-articular steroid injections were the most popular topic in recent years. Conclusion Based on the results of the present study, we found that there was no single database that covered all relevant articles; the optimal method for bibliometric analysis is a combination of databases. The most popular research topics on PJI focused on alpha-defensin, antibiotic use, risk factors of intra-articular steroid injections, and the location of prosthetic hip and knee infection.

In order to analyze the response law of the coal permeability to the angle of the bedding in Baode Mining Area, coal samples with different bedding angles were prepared. The QTS-2 coal rock permeation tester was used to test the permeability of coal under different confining pressures and inlet pressures. The results show that the coal body permeability in Baode Mining Area has obvious response to the bedding angle. With the increase of airflow direction and bedding angle, the permeability of coal body decreases linearly. The permeability of 0° angle differs from 90° angle coal sample by 1 to 2 orders of magnitude. With the increase of confining pressure, the permeability of coal body decreases exponentially. The permeability of coal sample reflects the difficulty of gas flow along the flow direction. In the actual gas drainage project, when the drilling is arranged parallel to the bedding, the direction of the coal gas flow is perpendicular to the bedding, and the permeability is minimal; when the drilling is arranged perpendicular to the bedding, the direction of coal gas flow is parallel to the bedding, and the permeability is maximal.

Abstract Background There is a concern regarding the use of a closed-suction drain (CSD) in two-stage exchange arthroplasty for periprosthetic joint infection as it may decrease the antibiotic concentrations in the joint fluids. The purpose of this study was to identify whether the use of a CSD could reduce local antibiotic concentrations following spacer implantation. Methods A prospective, randomized, controlled trial was conducted at our institution between January 2018 and November 2018. We enrolled 32 patients undergoing two-stage exchange arthroplasty for periprosthetic hip infection with an interim cement spacer containing 4-g vancomycin and 2-g meropenem per 40-g methyl-methacrylate cement polymer. Patients were randomized and evenly divided into the study group (non-CSD) and control group (CSD group) by sealed envelopes. Drainage samples of joint fluids (n = 160) were collected every 24 h for the first five days following spacer implantation. The antibiotic concentrations of drainage samples were measured by high-performance liquid chromatography, and the bioactivities of the drainage samples against methicillin-sensitive and methicillin-resistant Staphylococcus aureus (MSSA and MRSA) and E. coli were assessed. Results There was no significant difference in the decrease of vancomycin (study group vs. control group: 163.20 ± 77.05 vs. 162.39 ± 36.31; p = 0.917) and meropenem concentration (123.78 ± 21.04 vs. 117.27 ± 19.38; P = 0.548) between the two groups during the first five days following spacer implantation. All joint drainage samples in each group exhibited antibacterial activity against MSSA, MRSA and E. coli. Conclusions The use of CSD following the implantation of an antibiotic-loaded cement spacer does not reduce the effectiveness of such a spacer in two-stage exchange arthroplasty. Abstract Background Periprosthetic joint infection (PJI) is a challenging complication following total joint arthroplasty (TJA), and the diagnostic criteria remains controversial. The 2018 new definition proposed in May 2018 consists of new diagnostic criteria for PJI. We conducted a retrospective study and demonstrated that the new definition could improve the diagnostic efficiency in Chinese patients. However, missing data led to bias in the previous retrospective study. Therefore, this prospective study is designed to further validate the feasibility of 2018 new definition (and its modified version) for Chinese patients. Methods/design This is a single-centre, prospective diagnostic study with 1 year of follow-up. The patients enrolled in the trial will be divided into a PJI group and an Aseptic group based on the eligibility criteria. We will recruit at least 70 patients for each group from October 2019 to October 2020. Blood samples, synovial fluid samples and intraoperative variables of all the included patients will be collected to assess various indicators. We will integrate the results of the various tests and examine the diagnostic efficiency (sensitivity and specificity) of five diagnostic criteria. Discussion We design the prospective study in the hope of reducing the bias caused by missing data. Therefore, the prospective study will further support the conclusion of our preceding retrospective study. The results of this study will be submitted to a peer-reviewed journal for publication. Conclusion Through this prospective study, we will validate the feasibility of the 2018 new PJI definition (and its modified version) for Chinese patients and determine the optimal method of PJI diagnosis. Abstract Purpose The diagnosis of persistent infection at reimplantation of two-stage revision arthroplasty for periprosthetic joint infection (PJI) remains challenging. Several studies have shown the benefit of serum interleukin-6 (IL-6) in diagnosing periprosthetic joint infection (PJI). Recent data indicated serum IL-6 could be promising in differentiating persistent infection. The purpose of this study was to validate the efficacy of serum IL-6 in diagnosing persistent infection at reimplantation. Methods A retrospective review of 86 PJI patients with a two-stage exchanged hip arthroplasty from 2013 to 2017 was conducted. Persistent infection was defined using the modified Musculoskeletal Infection Society (MSIS) criteria combined with follow-up results. Serum IL-6 at reimplantation were collected and compared among patients with or without persistent infection. Receiver operating characteristic (ROC) curves were generated to evaluate the diagnostic performance and optimal cut-off value of serum IL-6 at reimplantation. Results Sixteen cases were diagnosed as persistent infection at reimplantation. There was no significant difference in serum IL-6 levels between cases with persistent infection and controls (7.89 pg/ml vs. 5.56 pg/ml; P = 0.179). The area under the ROC curve (AUC) for serum IL-6 in diagnosing persistent infection at reimplantation was 0.59 (95% confidential interval [CI] 0.40–0.77). With the calculated threshold set at 8.12 pg/ml, the corresponding sensitivity, specificity, positive predictive value, and negative predictive values were 38%, 88%, 38%, and 87%, respectively. Conclusion Serum IL-6 is inadequate in diagnosing persistent infection at reimplantation for two-stage revision arthroplasty. With the serum IL-6 threshold set at 8.12 pg/ml, the specificity to rule out persistent infection is high, but the sensitivity to predict persistent infection is not satisfactory.

Increasing effort has been devoted to restoring coastal ecosystems to counteract their degradation globally. Restoration success of coastal ecosystems often relies on harnessing biotic interactions that shape the performance of foundation plant species. Crabs acting as essential ecosystem engineers and consumers are commonly present in coastal saltmarshes. Previous work has focused on the grazing effect of crabs on plants. In line with the idea of top-down control, plant performance (in terms of biomass or height) is often used as indicators of plant-animal interactions. However, crabs could also produce facilitative effects through non-consumptive behavior. It remains unclear if and to what extent the net effect of crabs on coastal plants is dependent on environmental context. Therefore, systematic assessments are needed to test if the reliability of plant performance as an indicator of plant-crab interactions is context-dependent. Here, we conducted field survey in two similar intertidal saltmarsh ecosystems along the Yellow Sea coast, eastern China, and examined the effects of crab burrowing, soil properties, and nutrient availability on plant (Suaeda salsa) performance using multiple regression and structural equation models. We found that crab burrow density was significantly correlated with plant height, which signals strong plant-crab interactions. Surprisingly, we observed opposite signs (positive vs. negative) of plant-crab relationships between the two study sites that have highly similar composition and structure of plant communities. A possible explanation is that soil compactness can mediate crab burrowing behavior, resulting in differential facilitative effects on the plants. Thus, the usefulness of plant height serving as an indicator of plant-crab interactions depends on environmental context to a substantial degree. More comprehensive indicators taking into account soil compactness may facilitate robust inference of plant-crab interactions. Our results highlight landscape-scale spatial heterogeneity and context-dependency of plant-animal interactions in coastal saltmarshes, and provide a useful implication to inferring nuanced biotic interactions using compressive biotic and abiotic indicators in coastal ecosystems.

The prevalence and role of malnutrition in periprosthetic joint infection (PJI) remain unclear. This study aimed to use measurable nutritional screening tools to assess the prevalence of malnutrition in PJI patients during two-stage exchange arthroplasty and to explore the association between malnutrition and treatment failure. Our study retrospectively included 183 PJI cases who underwent 1st stage exchange arthroplasty and had available nutritional parameters, of which 167 proceeded with 2nd stage reimplantation. The recently proposed Musculoskeletal Infection Society (MSIS) Outcome Reporting Tool was used to determine clinical outcomes. The Controlling Nutritional Status (CONUT), Nutritional Risk Index (NRI), and Naples Prognostic Score (NPS) were used to identify malnutrition at 1st and 2nd stage exchange, respectively. Multivariate logistic regression analyses were performed to determine the association between malnutrition and treatment failure. Restricted cubic spline models were further used to explore the dose–response association. Additionally, risk factors for moderate-to-severe malnutrition were evaluated. Malnourished patients identified by CONUT, NPS, and NRI accounted for 48.1% (88/183), 98.9% (181/183), and 55.7% (102/183) of patients at 1st stage, and 9.0% (15/167), 41.9% (70/167), and 43.1% (72/167) at 2nd stage, indicating a significant improvement in nutritional status. We found that poorer nutritional status was a predictor of treatment failure, with CONUT performing best as a predictive tool. Moderate-to-severe malnutrition at 1st stage identified by CONUT was significantly related to treatment failure directly caused by PJI (odds ratio [OR] = 5.86), while the OR was raised to 12.15 at 2nd stage (OR = 12.15). The linear dose–response associations between them were also confirmed (P for nonlinearity at both 1st and 2nd stage > 0.05). As for total treatment failure, moderate-to-severe malnutrition as determined by CONUT was associated with a 1.96-fold and 8.99-fold elevated risk at the 1st and 2nd stages, respectively. Age ≥ 68 years (OR = 5.35) and an increased number of previous surgeries (OR = 2.04) may be risk factors for moderate-to-severe malnutrition. Overall, the prevalence of malnutrition in PJI patients is very high. Given the strong association between moderate-to-severe malnutrition identified by CONUT and PJI treatment failure, COUNT could be a promising tool to evaluate the nutritional status of PJI patients to optimize treatment outcomes.

Background:. Patients undergoing total joint arthroplasty (TJA) following septic arthritis are at higher risk for developing periprosthetic joint infection (PJI). Minimal literature is available to guide surgeons on the optimal timing of TJA after completing treatment for prior native joint septic arthritis. This multicenter study aimed to determine the optimal timing of TJA after prior septic arthritis and to examine the role of preoperative serology in predicting patients at risk for developing PJI. Methods:. A total of 207 TJAs were performed after prior septic arthritis from 2000 to 2017 at 5 institutions. Laboratory values, prior treatment, time from the initial infection, and other variables were recorded. Bivariate analyses were performed to identify the association between the time from septic arthritis to TJA and the risk of developing subsequent PJI. A subanalysis was performed between patients who underwent TJA in 1 setting (n = 97) compared with those who underwent 2-stage arthroplasties (n = 110). Receiver operating characteristic (ROC) curve analysis was performed for serum markers prior to TJA in predicting the risk of a subsequent PJI. Results:. The overall PJI rate was 12.1%. Increasing time from septic arthritis treatment to TJA was not associated with a reduction of PJI, whether considering time as a continuous or categorical variable, for both surgical treatment cohorts (all p > 0.05). Although the ROC curve analysis found that the optimal threshold for timing of TJA from the initial treatment was 5.9 months, there was no difference in the PJI rate when the overall cohort was dichotomized by this threshold and when stratified by 1-stage compared with 2-stage TJA. There was no significant difference in erythrocyte sedimentation rate (ESR) and C-reactive protein (CRP) level prior to conversion TJA between patients who subsequently developed PJI and those who did not. Conclusions:. Serum markers have limited value in predicting subsequent PJI in patients who undergo TJA after prior septic arthritis. There was no optimal interim period between septic arthritis treatment and subsequent TJA; thus, delaying a surgical procedure does not appear to reduce the risk of PJI. Level of Evidence:. Rapid and accurate estimation of maize biomass is critical for predicting crop productivity. The launched Sentinel-1 (S-1) synthetic aperture radar (SAR) and Sentinel-2 (S-2) missions offer a new opportunity to map biomass. The selection of appropriate response variables is crucial for improving the accuracy of biomass estimation. We developed models from SAR polarization indices, vegetation indices (VIs), and biophysical variables (BPVs) based on gaussian process regression (GPR) and random forest (RF) with feature optimization to retrieve maize biomass in Changchun, Jilin province, Northeastern China. Three new predictors from each type of remote sensing data were proposed based on the correlations to biomass measured in June, July, and August 2018. The results showed that a predictor combined by vertical-horizontal polarization (VV), vertical-horizontal polarization (VH), and the difference of VH and VV (VH-VV) derived from S-1 images of June, July, and August, respectively, with GPR and RF, provided a more accurate estimation of biomass (R2 = 0.81–0.83, RMSE = 0.40–0.41 kg/m2) than the models based on single SAR polarization indices or their combinations, or optimized features (R2 = 0.04–0.39, RMSE = 0.84–1.08 kg/m2). Among the S-2 VIs, the GPR model using a combination of ratio vegetation index (RVI) of June, normalized different infrared index (NDII) of July, and normalized difference vegetation index (NDVI) of August achieved a result with R2 = 0.83 and RMSE = 0.39 kg/m2, much better than single VIs or their combination, or optimized features (R2 of 0.31–0.77, RMSE of 0.47–0.87 kg/m2). A BPV predictor, combined with leaf chlorophyll content (CAB) in June, canopy water content (CWC) in July, and fractional vegetation cover (FCOVER) in August, with RF, also yielded the highest accuracy (R2 = 0.85, RMSE = 0.38 kg/m2) compared to that of single BPVs or their combinations, or optimized subset. Overall, the three combined predictors were found to be significant contributors to improving the estimation accuracy of biomass with GPR and RF methods. This study clearly sheds new insights on the application of S-1 and S-2 data on maize biomass modeling.

Climate change has posed serious challenges to food production and sustainable development. We evaluated crop yields, N2O emissions, and soil organic carbon (SOC) in a typical wheat–corn rotation system field on the North China Plain on a 50-year scale using the Denitrification–Decomposition (DNDC) model and proposed adaptive strategies for each climate scenarios. The study showed a good consistency between observations and simulations (R2 > 0.95 and nRMSE < 30%). Among the twelve climate scenarios, we explored ten management practices under four climate scenarios (3 °C temperature change: P/T−3 and P/T+3; 30% precipitation change: 0.7P/T and 1.3P/T), which have a significant impact on crop yields and the net greenhouse effect. The results revealed that changing the crop planting time (CP) and using cold-resistant (CR) varieties could reduce the net greenhouse effect by more than 1/4 without sacrificing crop yields under P/T−3. Straw return (SR) minimized the negative impact on yields and the environment under P/T+3. Fertigation (FG) and Drought-Resistant (DR) varieties reduced the net greenhouse effect by more than 8.34% and maintained yields under 0.7P/T. SR was most beneficial to carbon sequestration, and yields were increased by 3.87% under 1.3P/T. Multiple adaptive strategies should be implemented to balance yields and reduce the environmental burden under future climate change.

Summary: The recent mass mortality event of more than 330 African elephants in Botswana has been attributed to biotoxins produced by cyanobacteria; however, scientific evidence for this is lacking. Here, by synthesizing multiple sources of data, we show that, during the past decades, the widespread hypertrophic waters in Southern Africa have entailed an extremely high risk and frequent exposure of cyanotoxins to the wildlife within this area, which functions as a hotspot of mammal species richness. The hot and dry climatic extremes have most likely acted as the primary trigger of the recent and perhaps also of prehistoric mass mortality events. As such climate extremes are projected to become more frequent in Southern Africa in the near future, there is a risk that similar tragedies may take place, rendering African megafauna species, especially those that are already endangered, in risk of extinction. Moreover, cyanotoxin poisoning amplified by climate change may have unexpected cascading effects on human societies. Seen within this perspective, the tragic mass death of the world's largest terrestrial mammal species serves as an alarming early warning signal of future environmental catastrophes in Southern Africa. We suggest that systematic, quantitative cyanotoxin risk assessments are made and precautionary actions to mitigate the risks are taken without hesitation to ensure the health and sustainability of the megafauna and human societies within the region. Public Summary: • Cyanotoxin is the most likely cause for the massive death of elephants in Botswana. • Mammal species hotspot in Africa is under increasing risk of cyanotoxin poisoning. • This tragic event serves as an alarming early warning signal of future catastrophes. • Systematic assessments on cyanotoxin risk are strongly suggested in Southern Africa.

A novel acoustic metamaterial structure, made up of both resonator and multiple channels, is proposed to insulate low-frequency broadband noises. Three bandgaps with a total width of 658 Hz can be obtained below 900 Hz when the structure appears as a single negative characteristic within specific frequency ranges. These bandgaps are caused by the standing-wave characteristics under the coupling action of the interior and exterior cavities and the multiple channels. By changing structural parameters and materials, the low-frequency broadband sound insulation performance of the structure can be improved. Experimental results further prove the sound insulation characteristics. The results obtained herein can be used as a guide toward achieving low-frequency broadband sound insulation using a lightweight structure.

Abstract Background The treatment strategy for evolutive septic arthritis (SA) with coexistent degenerative joint disease is not well established. The purposes of this study were to 1) investigate treatment outcome and potential risk factors of treatment failure in patients with evolutive SA following two-stage procedure, including insertion of an antibiotic-loaded spacer at the first stage and subsequent implantation of a new prosthesis; and 2) determine the performance of serum erythrocyte sedimentation rate (ESR), C-reactive protein (CRP), and Interleukin-6 (IL-6) in predicting persisting infection at second-stage procedure. Methods We retrospectively reviewed 74 patients with evolutive SA of hips and knees who underwent a two-stage TJA between 2008 and 2015. The treatment success was defined according to the modified Delphi criteria and Kaplan-Meier survivorship curves were constructed to determine treatment success. A Cox regression model was performed to identify risk factors for treatment failure. Receiver operating characteristic (ROC) curves were generated to determine the prognostic value of ESR, CRP, and IL-6 in predicting persistent infection before second-stage prostheses implantation. Results Overall, the treatment success rate was 93% for hips and 100% for knees after the first-stage surgery. The treatment success rate was 89% for hips and 84% for knees after second-stage prosthesis implantation with a mean follow-up of 4.7 (range, 2.2 to 10.8) years. Older age (Hazard ratio [HR] [per 10-year increase], 1.20; 95% confidential interval [CI], 1.11 to 1.62), higher preoperative CRP level (HR [per 1-mg/dL increase], 1.15; 95% CI, 1.04 to 1.28) and resistant organism (HR, 13.96; 95% CI, 3.29 to 19.20) were associated with an increased risk of treatment failure. All serologic tests presented limited values in predicting persisting infection, with the area under ROC curve of ESR, CRP, IL-6 and combination of the three markers was 57.8, 61.6, 60.3, and 62.1%, respectively. Conclusions Two-stage TJA is an adequate management of infection control in patients with evolutive SA. The three potential risk factors (old age, high preoperative CRP, and resistant organism profile) may predict treatment failure following a two-stage procedure for evolutive SA. Additionally, serum ESR, CRP, and IL-6 had no benefit in predicting persisting infection before second-stage prostheses implantation. These findings may be useful when treating patients with evolutive SA.

Abstract Purpose The optimal timing of reimplantation of two-stage exchange arthroplasty for periprosthetic joint infection remains unknown. The purpose of the study was to (1) evaluate performance of combination of serum erythrocyte sedimentation rate (ESR), C-reactive protein (CRP), and frozen section in predicting persistent infection at the time of second-stage hip reimplantation and (2) compare accuracies of 5 and 10 polymorphonuclear neutrophils (PMNs) per high power field (HPF) as the threshold of frozen section. Methods We retrospectively reviewed 97 two-stage exchange hip arthroplasties from 2012–2016. Persistent infection at time of reimplantation was diagnosed using the Musculoskeletal Infection Society (MSIS) criteria. Two diagnostic models were developed. Model 1 utilized ESR, CRP, and > 5 PMNs/HPF on frozen section. Model 2 utilized ESR, CRP, and > 10 PMNs/HPF. Receiver operating characteristic (ROC) curves of the two models were generated, and areas under the curves (AUCs) were compared. A set of sensitivity analysis, using the Delphi-based consensus criteria for treatment success, was conducted to verify the accuracy of our models. Results The overall rate of infection at reimplantation was 14.4%. AUCs for models 1 and 2 were 0.709 (95% confidence interval [CI], 0.557–0.852) and 0.697 (95% CI, 0.529–0.847), respectively. Sensitivity, specificity, positive predictive value (PPV), and negative predictive value (NPV) were 57.1%, 88.0%, 44.4%, and 92.4%, respectively, in model 1 and 42.9%, 96.4%, 66.7%, and 90.9%, respectively, in model 2. Models 1 and 2 had no significant difference in predictive values (p = 0.821). Results remained robust in the sensitivity analysis. Conclusions This study reveals that the combination of serum ESR, CRP, and frozen section has limited diagnostic value in predicting persistent infection at reimplantation. Additionally, no significant difference in accuracies between 5 and 10 PMNs/HPF as the threshold of frozen section were found. There is a need for timely biomarkers with higher accuracy in diagnosing infection before reimplantation.

Abstract Background The diagnosis of persistent infection before reimplantation in two-stage exchange arthroplasty for periprosthetic joint infection (PJI) remains challenging. Currently, several studies suggested coagulation-related markers, such as D-dimer and fibrinogen, may be promising in diagnose of PJI. The purpose of the study was to investigate the predictive values of plasma D-dimer and fibrinogen for assessment of persistent infection before reimplantation hip arthroplasty. Methods We retrospectively reviewed 129 hips that treated with two-stage exchange arthroplasty for PJI from 2012 to 2016 in our institution. The persistent infection before reimplantation was based on a modified Musculoskeletal Infection Society (MSIS) criteria. After exclusion, 102 hips were included in the final analysis. Receiver operating characteristic (ROC) curves were generated to determine the prognostic value of plasma D-dimer and fibrinogen in predicting persistent infection before reimplantation. Results The area the under ROC curves (AUC) for fibrinogen (0.773; 95% confidential interval [CI], 0.569–0.905) was significantly higher than that of D-dimer (0.565; 95% CI, 0.329–0.777). With the calculated threshold of fibrinogen set at 3.61 g/L, the sensitivity, specificity, positive predictive value (PPV), and negative predictive value (NPV) was 87.5%, 62.8%, 16.7%, and 98.3%, respectively. With the threshold value of D-dimer set at 0.82 μg/mL, the sensitivity, specificity, PPV, and NPV was 83.3%, 41.9%, 21.7%, and 92.9%, respectively. Conclusions In conclusion, the current study reveals that the plasma fibrinogen may be a promising biomarker in predicting persistent infection before reimplantation. Further prospective studies with larger cohorts are needed to validate predictive values and optimal thresholds of coagulation-related markers.

Abstract Background Unmanned aerial vehicle (UAV)-based remote sensing provides a flexible, low-cost, and efficient approach to monitor crop growth status at fine spatial and temporal resolutions, and has a high potential to accelerate breeding process and improve precision field management. Method In this study, we discussed the use of lightweight UAV with dual image-frame snapshot cameras to estimate aboveground biomass (AGB) and panicle biomass (PB) of rice at different growth stages with different nitrogen (N) treatments. The spatial–temporal variations in the typical vegetation indices (VIs) and AGB were first investigated, and the accuracy of crop surface model (CSM) extracted from the Red Green Blue (RGB) images at two different stages were also evaluated. Random forest (RF) model for AGB estimation as well as the PB was then developed. Furthermore, variable importance and sensitivity analysis of UAV variables were performed to study the potential of improving model robustness and prediction accuracies. Results It was found that the canopy height extracted from the CSM (Hcsm) exhibited a high correlation with the ground-measured canopy height, while it was unsuitable to be independently used for biomass assessment of rice during the entire growth stages. We also observed that several VIs were highly correlated with AGB, and the modified normalized difference spectral index extracted from the multispectral image achieved the highest correlation. RF model with fusing RGB and multispectral image data substantially improved the prediction results of AGB and PB with the prediction of root mean square error (RMSEP) reduced by 8.33–16.00%. The best prediction results for AGB and PB were achieved with the coefficient of determination (r2), the RMSEP and relative RMSE (RRMSE) of 0.90, 0.21 kg/m2 and 14.05%, and 0.68, 0.10 kg/m2 and 12.11%, respectively. In addition, the result confirmed that the sensitivity analysis could simplify the prediction model without reducing the prediction accuracy. Conclusion These findings demonstrate the feasibility of applying lightweight UAV with dual image-frame snapshot cameras for rice biomass estimation, and its potential for high throughput analysis of plant growth-related traits in precision agriculture as well as the advanced breeding program.

Flexible production is a typical representative of high-end manufacturing and is also a manifestation of a country’s national production capability. Compared with other production modes, the key distinction of flexible production is that all four dimensions of production (i.e., machines, operations, products, and orders) can be scheduled dynamically. Although many studies have investigated the flexible job shop scheduling problem, most have limited dynamic support and cannot deal with multidimensional dynamic production. This study, therefore, proposed a fine-grained system state description model, which was used to analyze the maximum production completion time. In the presence of a dynamic event, the model was able to quickly assign priorities to products according to the cost loss of each product. The system can therefore dynamically respond to events in a timely manner while reducing production costs and losses. Finally, we used a large number of orders to evaluate the proposed algorithm, which demonstrated millisecond-level response capability and low-cost maintenance capability. Compared with existing algorithms, the proposed algorithm reduced cost loss by up to 11%.

Soil invertebrates and microorganisms are two major drivers of litter decomposition. Even though the importance of invertebrates and microorganisms in biogeochemical soil cycles and soil food webs has been studied, the effects of invertebrates on fungi are not well understood compared to other organisms. In this work, we investigated the effects of soil invertebrates on fungi as a factor that cannot be ignored in the study of nutrient cycling. The result showed the grazing of isopods on white-rot fungi was transitive and persistent. The grazed fungi appeared “compensatory” growing. The biomass of fungi increased after grazing. The activities of enzymes associated with nutrient cycling were increased under grazing. The zymography images showed the enzyme hotspots and activities also increased significantly in the grazing area. The results suggest that invertebrate grazing can significantly increase the fungal biomass and enzyme activity, accelerating litter decomposition in the unreached grazer area. The grazing effects of invertebrate plays an important role in promoting the nutrient cycling of the forest ecosystem. We believe that this study will be a good reference related to showing the relationship between soil invertebrates, fungi and soil biogeochemical cycles.

Abstract Background Pigmented villonodular synovitis (PVNS) is a relatively rare, locally aggressive, and potentially recurrent synovial disease of large joints. The purpose of this study was to investigate (1) the disease recurrence rate and (2) the treatment outcomes including Harris hip scores, complications, and revision following cementless total hip arthroplasty (THA) with ceramic-on-ceramic (CoC) articulation in patients with PVNS. Methods Twenty-two patients (14 females and 8 males) with histologically confirmed PVNS underwent cementless THA using CoC bearings between 2000 and 2013. Three patients with less than 5-year follow-up were excluded. The mean age was 35.2 years (range, 22–58 years) with a mean follow-up of 8.6 years (range, 6.9–10.8 years). A control group was matched in a 2:1 ratio with the PVNS group for age, sex, body mass index (BMI), year of surgery, and American Society of Anesthesiologists score (ASA). Postoperative outcome variables included disease recurrence, Harris Hip Scores (HHS) at the latest follow-up, complications (dislocation, squeaking, ceramic fracture), and any-cause revision. A Kaplan-Meier implant survivorship curve with 95% confidence interval (CI) of the two groups was generated. Results No recurrence of PVNS was noted in the follow-up period. The HSS in the PVNS group was 92.6 ± 5.5, which was similar to the control group (93.4 ± 4.6, p = 0.584) at the last follow-up visit. No patients sustained dislocation, osteolysis, or any ceramic fracture within the study duration. One patient in the PVNS group had a complication of squeaking, but did not require revision. Another patient in the PVNS group underwent revision surgery due to aseptic loosening. There was no significant difference in revision rates between the two groups (p = 1.000). The implant survivorship free of any revision was 90.0% (95% CI, 73.2% to 100%) in the PVNS group and 92.5% (95% CI, 82.6% to 100%) in the control group at 10 years (p = 0.99). Conclusions For young and active patients with end-stage PVNS of the hips, cementless THA using CoC bearing has similar functional outcome scores, a low complication rate, and similar implant survivorship compared to the control group.

Anxiety is a common comorbidity associated with chronic pain, which results in chronic pain complexification and difficulty in treatment. Electroacupuncture (EA) is commonly used to treat chronic pain and anxiety. However, the underlying mechanisms of the EA effect are largely unknown. Here, we showed that a circuitry underlying chronic pain induces anxiety disorders, and EA can treat them by regulating such circuitry. Using chemogenetic methods, we found that chemogenetic activation of the rostral anterior cingulate cortex (rACC) glutamatergic output to the thalamus induced anxiety disorders in control rats. Then, chemogenetic inhibition of the rACC-thalamus circuitry reduced anxiety-like behavior produced by intraplantar injection of the complete Freund’s adjuvant (CFA). In this study, we examined the effects of EA on a rat model of CFA-mediated anxiety-like behaviors and the related mechanisms. We found that chemogenetic activation of the rACC-thalamus circuitry effectively blocked the effects of EA on chronic pain-induced anxiety-like behaviors in CFA rats. These results demonstrate an underlying rACC-thalamus glutamatergic circuitry that regulates CFA-mediated anxiety-like behaviors. This study also provides a potential mechanistic explanation for EA treatment of anxiety caused by chronic pain.

There is a rising prospective in harvesting energy from the environment, as in situ energy is required for the distributed sensors in the interconnected information society, among which the water flow energy is the most potential candidate as a clean and abundant mechanical source. However, for microscale and unordered movement of water, achieving a sustainable direct-current generating device with high output to drive the load element is still challenging, which requires for further exploration. Herein, we propose a dynamic PN water junction generator with moving water sandwiched between two semiconductors, which outputs a sustainable direct-current voltage of 0.3 V and a current of 0.64 μA. The mechanism can be attributed to the dynamic polarization process of water as moving dielectric medium in the dynamic PN water junction, under the Fermi level difference of two semiconductors. We further demonstrate an encapsulated portable power-generating device with simple structure and continuous direct-current voltage output of 0.11 V, which exhibits its promising potential application in the field of wearable devices and the IoTs.
