Aridity and drought, which are determined by climatic and temporary water scarcity, respectively, are important limiting factors for plant gross primary production. These phenomena are commonly assessed and/or monitored by means of weather indices, most of which are based on observations of precipitation and potential evapotranspiration. The estimation of such indices over large areas can be carried out using multiple datasets, i.e., those derived from weather stations, satellite images, and ground radars. The possibility of using interpolated or remotely sensed datasets in place of ground measurements was currently investigated for Tuscany, a region in Central Italy, showing complex and heterogeneous environmental features. The former weather datasets were first evaluated versus corresponding ground measurements. Next, the basic weather variables were combined and cumulated over 30–60 days to yield synthetic indicators of water deficit, which were assessed in the same way. Finally, these indicators were evaluated to predict the soil water conditions of a meadow and an olive grove during the 2021 summer period. The results obtained indicate that the use of the multi-source weather datasets induces only a minor deterioration of the water stress indicators and is therefore efficient to monitor the water status of different ecosystems with high spatial (200 m) and temporal (daily) details.

Objective Emerging evidence demonstrates that excessive accumulation of senescent cells is associated with some chronic diseases and suggests a pathogenic role of cellular senescence in fibrotic processes, such as that occurring in ageing or in SSc. Recently we demonstrated that parvovirus B19 (B19V) activates normal human dermal fibroblasts and induces expression of different profibrotic/pro-inflammatory genes. This observation prompted us to investigate whether it is also able to induce fibroblast senescence as a potential pathogenetic mechanism in B19V-induced fibrosis. Methods Primary cultures of fibroblasts were infected with B19V and analysed for the acquisition of senescence markers, such as morphological modifications, senescence-associated β-galactosidase (SA-β-gal) activity, DNA damage response and expression of senescence-associated secretory phenotype (SASP)-related factors. Results We demonstrated that B19V-infected fibroblasts develop typical senescence features such as enlarged and flat-shaped morphology and SA-β-gal activity similar to that observed in SSc skin fibroblasts. They also developed an SASP-like phenotype characterized by mRNA expression and release of some pro-inflammatory cytokines, along with activation of the transcription factor nuclear factor κB. Moreover, we observed B19V-induced DNA damage with the comet assay: a subpopulation of fibroblasts from B19V-infected cultures showed a significantly higher level of DNA strand breaks and oxidative damage compared with mock-infected cells. An increased level and nuclear localization of γH2AX, a hallmark of DNA damage response, were also found. Conclusions B19V-induced senescence and production of SASP-like factors in normal dermal fibroblasts could represent a new pathogenic mechanism of non-productive B19V infection, which may have a role in the fibrotic process.

During the night between 9 and 10 September 2017, multiple flash floods associated with a heavy-precipitation event affected the town of Livorno, located in Tuscany, Italy. Accumulated precipitation exceeding 200 mm in two hours was recorded. This rainfall intensity is associated with a return period of higher than 200 years. As a consequence, all the largest streams of the Livorno municipality flooded several areas of the town. We used the limited-area weather research and forecasting (WRF) model, in a convection-permitting setup, to reconstruct the extreme event leading to the flash floods. We evaluated possible forecasting improvements emerging from the assimilation of local ground stations and X- and S-band radar data into the WRF, using the configuration operational at the meteorological center of Tuscany region (LaMMA) at the time of the event. Simulations were verified against weather station observations, through an innovative method aimed at disentangling the positioning and intensity errors of precipitation forecasts. A more accurate description of the low-level flows and a better assessment of the atmospheric water vapor field showed how the assimilation of radar data can improve quantitative precipitation forecasts.

uPAR is a globular protein, tethered to the cell membrane by a GPI-anchor involved in several cancer-related properties and its overexpression commonly correlates with poor prognosis and metastasis. We investigated the consequences of uPAR irreversible loss in human melanoma and colon cancer cell lines, knocking out its expression by CRISPR/Cas9. We analyzed through flow cytometry, western blotting and qPCR, the modulation of the most known cancer stem cells-associated genes and the EGFR while we observed the proliferation rate exploiting 2D and 3D cellular models. We also generated uPAR “rescue” expression cell lines as well as we promoted the expression of only its 3’UTR to demonstrate the involvement of uPAR mRNA in tumor progression. Knocking out PLAUR, uPAR-encoding gene, we observed an inhibited growth ratio unexpectedly coupled with a significant percentage of cells acquiring a stem-like phenotype. In vivo experiments demonstrated that uPAR loss completely abrogates tumorigenesis despite the gained stem-like profile. Nonetheless, we proved that the reintroduction of the 3’UTR of PLAUR gene was sufficient to restore the wild-type status validating the hypothesis that such a region may act as a “molecular sponge”. In particular miR146a, by binding PLAUR 3’ UTR region might be responsible for uPAR-dependent inhibition of EGFR expression.

Purpose Hyponatraemia is frequently observed in cancer patients and can be due to the syndrome of inappropriate anti-diuresis (SIAD), related to ectopic vasopressin secretion, particularly in small cell lung cancer (SCLC). Hyponatraemia is associated with a worse outcome in cancer patients. The vasopressin receptor antagonist tolvaptan effectively corrects hyponatraemia secondary to SIAD and there is in vitro evidence that it has also an antiproliferative effect in cancer cells. The purpose of this study was i) to analyse the effect of low serum sodium concentrations ([Na+]) in SCLC cells and ii) to determine whether tolvaptan counteracts tumor progression. Methods We evaluated cell proliferation, cell cycle, apoptosis, oxidative stress, invasivity in low [Na+] as well as after exposure to tolvaptan. We also analysed the intracellular signalling pathways involved. Results In reduced [Na+] cell proliferation was significantly increased compared to normal [Na+] and cells were mostly distributed in the G2/M phase. Apoptosis appeared reduced. In addition, the ability to cross matrigel-coated membranes markedly increased. As observed in other cancer cell models, the expression of the heme-oxigenase-1 gene was increased. Finally, we found that in cells cultured in low [Na+] the RhoA/ROCK1/2 pathway, which is involved in the regulation of actin cytoskeleton, was activated. On the other hand, we found that tolvaptan effectively inhibited cell proliferation, anchorage-independent growth, invasivity and promoted apoptosis. Accordingly, the RhoA/ROCK-1/2 pathway was inhibited. Conclusions These findings demonstrate for the first time that low [Na+] favours tumor progression in SCLC cells, whereas tolvaptan effectively inhibits cell proliferation, survival and invasivity.

Dal 2017 il lancio del satellite gemello SENTINEL 2B, il tempo di rivisitazione si è dimezzato, permettendo agli utenti di ottenere una immagine sullo stesso territorio ogni 5 giorni. Il consorzio LaMMA acquisisce sull’intera regione Toscana tali immagini calcolandone vari indici di vegetazione e archiviano solo tali prodotti (NDVI Normalized difference vegetation index e NBR Nearinfrared Burned Ratio). Per ogni acquisizione disponiamo quindi di una informazione relativa allo stato di vegetazione sull’intero territorio regionale. L’indice NDVI è utilizzato per studi relativi alla vegetazione attiva presente sul territorio ed una analisi condotta verificando la variazione di tale indice nel tempo, fornisce in prima approssimazione, una indicazione sul cambiamento dell’uso e copertura del suolo.

This chapter gives an overview of some of the key concepts in the field of discrimination. It starts by distinguishing between direct and indirect discrimination in legal definitions. Next, we define the interrelated concepts of multiple discrimination and intersectionality, which increasingly are used in both legal studies and the social sciences, before giving an account of the interrelated concepts of organizational, institutional, and systemic discrimination. The chapter ends by reflecting on the complex relationship between discrimination and the endurance of categorical inequalities in societies where all members formally enjoy the principle of equality.

Recent investigations have supported the possibility of predicting soil water content (SWC) through a simplified soil water balance (WB) model fed with remotely sensed actual evapotranspiration (ETa) estimates. This approach, however, requires information on main soil features (i.e. depth, wilting point, field capacity) which are generally difficult to retrieve over large regions. The current paper proposes an alternative model which directly predicts SWC relying on the same logic of the recently proposed ETa estimation method, i.e. the combination of meteorological and normalized difference vegetation index (NDVI) datasets. The theoretical bases of the old and new SWC estimation methods are first described. Both methods are then applied in Central Italy using ancillary and Moderate Resolution Imaging Spectroradiometer (MODIS) NDVI data having a spatial resolution of 250 m; the outputs obtained are assessed versus SWC measurements taken both continuously and instantaneously. In the former case three ecosystems are analyzed (i.e. the grassland of Amplero and the forests of Barbialla and Amiata), where SWC was measured by probes during three different years (2003, 2012 and 2018, respectively). The latter experiment concerns 362 sample sites where SWC gravimetric measurements were taken during a similar time span (2000–2018). In both cases SWC is first normalized into relative SWC (RSWC), which is more directly influential on vegetation conditions. The results of both experiments indicate that the two simulation methods perform similarly when the former is driven by adequate information on soil features. The main limitations of the two simulation approaches are due to the spatial resolution mismatch between SWC measurements and estimates, which has a relatively minor impact on the first three homogeneous areas but is decisive for the other sampled sites. In general, the new simulation method is capable of predicting RSWC with relatively high spatial and temporal resolution without the use of specific soil information.

Discrimination and stigmatization are costly for the society by lowering economic growth, by reinforcing ethnic inequalities, by fueling political conflicts and by jeopardizing social cohesion. Moreover, victims of unfair treatment pay a high price as discrimination and stigmatization. Far from being passive victims, however, many members of minority groups develop and deploy individual and collective strategies to meet such challenges. This chapter focuses on the impact of discrimination – for economy and society, but mainly focusing on the consequences of discrimination for the targeted individuals and groups. The chapter also addresses responses to experiences of exclusion and disadvantage by reviewing recent research of how awareness of the repercussions of unfair treatment lead both individuals and groups to protect themselves and seek strategies for overcoming future barriers.
