Alignment of 36 MinC sequences revealed four completely conserved C-terminal glycines. As MinC inhibits cytokinesis in Neisseria gonorrhoeae and Escherichia coli, the functional importance of these glycines in N. gonorrhoeae MinC (Min[C.sub.Ng]) and E. coli MinC (Min[C.sub.Ec]) was investigated through amino acid substitution by using site-directed mutagenesis. Each mutant was evaluated for its ability to arrest cell division and to interact with itself and MinD. In contrast to overexpression of wild-type MinC, overexpression of mutant proteins in E. coli did not induce filamentation, indicating that they lost functionality. Yeast two-hybrid studies showed that Min[C.sub.Ec] interacts with itself and Min[D.sub.Ec]; however, no interactions involving Min[C.sub.Ng] were detected. Therefore, a recombinant MinC protein, with the N terminus of Min[C.sub.Ec] and the C terminus of Min[C.sub.Ng], was designed to test for a Min[C.sub.Ng]-Min[D.sub.Ng] interaction. Each MinC mutant interacted with either MinC or MinD but not both, indicating the specificity of glycine residues for particular protein-protein interactions. Each glycine was mapped on the C-terminal surfaces (A, B, and C) of the solved Thermotoga maritima MinC structure. We found that Min[C.sub.Ec] G161, residing in close proximity to the A surface, is involved in homodimerization, which is essential for MinC function. Glycines corresponding to Min[C.sub.Ec] G135, G154, and G171, located within or adjacent to the B-C surface junction, are critical for MinC-MinD interactions. Circular dichroism revealed no gross structural perturbations of the mutant proteins, although the contribution of glycines to protein flexibility and stability cannot be discounted. Using molecular modeling, we propose that exposed conserved MinC glycines interact with exposed residues of the [alpha]-7 helix of MinD.

The elliptic-flow ratio of neutrons with respect to protons or light complex particles in reactions of heavy ions at pre-relativistic energies has been proposed as an observable sensitive to the strength of the symmetry term of the nuclear equation of state at supra-saturation densities. In the ASY-EOS experiment at the GSI laboratory, flows of neutrons and light charged particles were measured for 197Au+197Au, 96Zr+96Zr and 96Ru+96Ru collisions at 400 MeV/nucleon with the Large Area Neutron Detector LAND as part of a setup with several additional detection systems used for the event characterization. Flow results obtained for the Au+Au system, in comparison with predictions of the UrQMD transport model, confirm the moderately soft to linear density dependence of the symmetry energy deduced from the earlier FOPI-LAND data.

The mitochondrial Human Proteome Project aims at understanding the function of the mitochondrial proteome and its crosstalk with the proteome of other organelles. Being able to choose a suitable and validated enrichment protocol of functional mitochondria, based on the specific needs of the downstream proteomics analysis, would greatly help the researchers in the field. Mitochondrial fractions from ten model cell lines were prepared using three enrichment protocols and analyzed on seven different LC-MS/MS platforms. All data were processed using neXtProt as reference database. The data are available for the Human Proteome Project purposes through the ProteomeXchange Consortium with the identifier PXD007053. The processed datasets were analysed using a suite of R routines to perform a statistical analysis and to retrieve subcellular and sub-mitochondrial localizations. Although the overall number of identified total and mitochondrial proteins was not significantly dependent on the enrichment protocol, specific line to line differences were observed. Moreover, the protein lists were mapped to a network representing the functional mitochondrial proteome, encompassing mitochondrial proteins and their first interactors. More than 80% of the identified proteins resulted nodes of this network but with a different ability in co-isolating mitochondria-associated structures for each enrichment protocol/cell line pair.

Come muta la relazione che lega corpi e tecnologie? Come si modificano gli spazi che attraversa? Come leggere da una prospettiva intersezionale e di genere le trasformazioni in atto? Il libro affronta queste domande a partire da tre ambiti distinti ma costantemente intrecciati: Spazio inteso come ambito di riflessione intorno al quale sviluppare ragionamenti in tema di autodeterminazione, mobilità, attivismo; Bios per affrontare il rapporto tra tecnologie e vita attraverso le forme della sua ri/produzione; Media per guardare criticamente i processi di soggettivazione innescati in particolar modo dalle tecnologie digitali. Per scriverlo abbiamo pensato al corpo come a una somateca, un archivio di finzioni politiche vive che in nessun modo possono costituire un unico corpus. Le tecnologie occupano oggi uno spazio molto ampio di questo archivio, per gli ineluttabili (poiché voluti, ma anche subiti) legami che i nostri corpi hanno intessuto con esse. Nelle riflessioni sulla relazione tra corpi e tecnologie si pone spesso l’accento sui processi di disincarnazione, di smaterializzazione, da un lato con i toni dell’entusiasmo, dall’altro con quelli della catastrofe. Questo libro è incarnato, si sottrae al binarismo, si insinua negli spazi in beetween, là dove i margini non sono confini. Gli interventi che proponiamo si muovono tra accademia, esperienza e attivismo, provano a elaborare nuove teorie e pratiche di critica radicale al tecnocapitalismo, discutono tra loro concordando o scontrandosi, uniti da un filo conduttore: smascherare i dispositivi di potere e i loro complicati intrecci.

The future opportunities for high-density QCD studies with ion and proton beams at the LHC are presented. Four major scientific goals are identified: the characterisation of the macroscopic long wavelength Quark-Gluon Plasma (QGP) properties with unprecedented precision, the investigation of the microscopic parton dynamics underlying QGP properties, the development of a unified picture of particle production and QCD dynamics from small (pp) to large (nucleus--nucleus) systems, the exploration of parton densities in nuclei in a broad ($x$, $Q^2$) kinematic range and the search for the possible onset of parton saturation. In order to address these scientific goals, high-luminosity Pb-Pb and p-Pb programmes are considered as priorities for Runs 3 and 4, complemented by high-multiplicity studies in pp collisions and a short run with oxygen ions. High-luminosity runs with intermediate-mass nuclei, for example Ar or Kr, are considered as an appealing case for extending the heavy-ion programme at the LHC beyond Run 4. The potential of the High-Energy LHC to probe QCD matter with newly-available observables, at twice larger center-of-mass energies than the LHC, is investigated.

We analyse the hadron-quark phase transition in neutron stars by confronting the hadronic Equation of State (EoS) obtained according to the microscopic Brueckner-Hartree-Fock many body theory, with the quark matter EoS derived within the Field Correlator Method. In particular, the latter EoS is only parametrized in terms of the gluon condensate and the large distance quark-antiquark potential, so that the comparison of the results of this analysis with the most recent measurements of heavy neutron star masses provides some physical constraints on these two parameters.

The elliptic-flow ratio of neutrons with respect to protons or light complex particles in reactions of heavy-ions at pre-relativistic energies has been proposed as an observable sensitive to the strength of the symmetry term of the nuclear equation of state at supra-saturation densities. The results obtained from the existing FOPI/LAND data for 197Au+197Au collisions at 400 MeV/nucleon in comparison with the UrQMD model simulations favoured a moderately soft symmetry term, but suffer from a considerable statistical uncertainty. These results have been confirmed by an independent analysis based on the Tübingen QMD simulations. In order to obtain an improved data set for Au+Au collisions and to extend the study to other systems, a new experiment was carried out at the GSI laboratory by the ASY-EOS collaboration. A current goal of relativistic heavy ion collisions experiments is to understand the impact of initial non-equilibrium on final observables. A Color Glass Condensate (CGC) as the limiting state of QCD matter at very high density implies initial non-thermal distribution at least for momenta below the saturation scale. In viscous hydrodynamics simulations, a standard Glauber initial condition leads to estimate 4πη/s ~ 1, while employing the Kharzeev-Levin-Nardi (KLN) modeling of the CGC leads to at least a factor of 2 larger η/s. Within a kinetic theory approach based on a relativistic Boltzmann-like transport simulation, our main result is that the out-of-equilibrium initial distribution in p-space reduces the effciency in building-up the elliptic flow. At RHIC energy we find the available data on υ2 are in agreement with a 4πη/s ~ 1 also for KLN initial conditions.

In this talk, we report on our results about the computation of isotropization and thermalization times of the quark-gluon plasma produced in relativistic heavy ion collisions, as well as of the elliptic flow. Simulation of the evolving fireball is achieved by solving the relativistic Boltzmann equation for the parton distribution function tuned at a fixed shear viscosity to entropy density ratio η/s. Our results show that the saturation in the initial spectrum reduces the efficiency in building-up the elliptic flow. Moreover both thermalization and isotropization times are quite small, approximately of 1 fm/c, if the system is in a strong coupling regime.

- Abstract
Prosthetic joint replacement failure has a huge impact on quality of life and hospitalization costs. A leading cause of prosthetic joint infection is bacteria-forming biofilm on the surface of orthopedic devices. Staphylococcus epidermidis is an emergent, low-virulence pathogen implicated in chronic infections, barely indistinguishable from aseptic loosening when embedded in a mature matrix. The literature on the behavior of quiescent S. epidermidis in mature biofilms is scarce. To fill this gap, we performed comparative analysis of the whole proteomic profiles of two methicillin-resistant S. epidermidis strains growing in planktonic and in sessile form to investigate the molecular mechanisms underlying biofilm stability. After 72-h culture of biofilm-forming S. epidermidis, overexpression of proteins involved in the synthesis of nucleoside triphosphate and polysaccharides was observed, whereas planktonic bacteria expressed proteins linked to stress and anaerobic growth. Cytological analysis was performed to determine why planktonic bacteria unexpectedly expressed proteins typical of sessile culture. Images evidenced that prolonged culture under vigorous agitation can create a stressful growing environment that triggers microorganism aggregation in a biofilm-like matrix as a mechanism to survive harsh conditions. The choice of a unique late time point provided an important clue for future investigations into the biofilm-like behavior of planktonic cells. Our preliminary results may inform comparative proteomic strategies in the study of mature bacterial biofilm. Finally, there is an increasing number of studies on the aggregation of free-floating bacteria embedded in an extracellular matrix, prompting the need to gain further insight into this mode of bacterial growth.

- Abstract
There is increasing evidence to suggest that, in addition to their regenerative effect, mesenchymal stromal cells (MSCs), and their secretome have an anti-inflammatory and antimicrobial role in the innate immune response in conditions such as sepsis. However, there is no published information on the effect of MSCs in bovine mastitis. Mastitis often results in extensive tissue damage due to multi-microorganism co-infection. This study investigated the ability of amniotic-derived conditioned medium (CM), in vitro and in vivo, to counteract microbial action and restore healthy tissue capable of milk production. Following determination of a dose-response curve, 10,000 colony-forming units (CFU) of Staphylococcus aureus (S. aureus) were inoculated into bovine mammary epithelial cell culture with and without 10% CM (supplemented either at the time of bacteria inoculation or after 4 h). Acridine orange staining was used to assess cell viability/apoptosis. Additionally, an in vivo study was performed using 48 dairy cows with acute and chronic mastitis, treated with CM (treated group) or antibiotics (control group). In vitro results showed that CM can attenuate bacterial growth, as evaluated by the number of CFU. After 24 h of culture with S. aureus, 89.67% of mammary epithelial cells treated with CM were still alive, whereas all cells cultured without CM were dead. Rates of epithelial cell survival (60.67%) were similar when CM was added 4 h after bacteria inoculation. There was no difference in somatic cell count between cases of acute mastitis in the CM-treated or control group in the in vivo study. However, relapses in chronic mastitis were less common in the group receiving CM. Our results show that CM is able to mitigate bacterial growth in vitro and may be particularly useful in the treatment of chronic mastitis, aiding restoration of milk production in cows that would otherwise be removed from the production cycle.

This document was prepared by the community that is active in Italy, within INFN (Istituto Nazionale di Fisica Nucleare), in the field of ultra-relativistic heavy-ion collisions. The experimental study of the phase diagram of strongly-interacting matter and of the Quark-Gluon Plasma (QGP) deconfined state will proceed, in the next 10-15 years, along two directions: the high-energy regime at RHIC and at the LHC, and the low-energy regime at FAIR, NICA, SPS and RHIC. The Italian community is strongly involved in the present and future programme of the ALICE experiment, the upgrade of which will open, in the 2020s, a new phase of high-precision characterisation of the QGP properties at the LHC. As a complement of this main activity, there is a growing interest in a possible future experiment at the SPS, which would target the search for the onset of deconfinement using dimuon measurements. On a longer timescale, the community looks with interest at the ongoing studies and discussions on a possible fixed-target programme using the LHC ion beams and on the Future Circular Collider., Comment: 99 pages, 56 figures

- Abstract
Neutrinoless double beta decay (0 nu beta beta) is considered the best potential resource to access the absolute neutrino mass scale. Moreover, if observed, it will signal that neutrinos are their own anti-particles (Majorana particles). Presently, this physics case is one of the most important research “beyond Standard Model” and might guide the way towards a Grand Unified Theory of fundamental interactions. Since the 0 nu beta beta decay process involves nuclei, its analysis necessarily implies nuclear structure issues. In the NURE project, supported by a Starting Grant of the European Research Council (ERC), nuclear reactions of double charge-exchange (DCE) are used as a tool to extract information on the 0 nu beta beta Nuclear Matrix Elements. In DCE reactions and beta beta decay indeed the initial and final nuclear states are the same and the transition operators have similar structure. Thus the measurement of the DCE absolute cross-sections can give crucial information on beta beta matrix elements. In a wider view, the NUMEN international collaboration plans a major upgrade of the INFN-LNS facilities in the next years in order to increase the experimental production of nuclei of at least two orders of magnitude, thus making feasible a systematic study of all the cases of interest as candidates for 0 nu beta beta.

- Abstract
The article describes the main achievements of the NUMEN project together with an updated and detailed overview of the related R&D activities and theoretical developments. NUMEN proposes an innovative technique to access the nuclear matrix elements entering the expression of the lifetime of the double beta decay by cross section measurements of heavy-ion induced Double Charge Exchange (DCE) reactions. Despite the fact that the two processes, namely neutrinoless double beta decay and DCE reactions, are triggered by the weak and strong interaction respectively, important analogies are suggested. The basic point is the coincidence of the initial and final state many-body wave functions in the two types of processes and the formal similarity of the transition operators. First experimental results obtained at the INFN-LNS laboratory for the 40Ca(18O,18Ne)40Ar reaction at 270MeV give an encouraging indication on the capability of the proposed technique to access relevant quantitative information. The main experimental tools for this project are the K800 Superconducting Cyclotron and MAGNEX spectrometer. The former is used for the acceleration of the required high resolution and low emittance heavy-ion beams and the latter is the large acceptance magnetic spectrometer for the detection of the ejectiles. The use of the high-order trajectory reconstruction technique, implemented in MAGNEX, allows to reach the experimental resolution and sensitivity required for the accurate measurement of the DCE cross sections at forward angles. However, the tiny values of such cross sections and the resolution requirements demand beam intensities much larger than those manageable with the present facility. The on-going upgrade of the INFN-LNS facilities in this perspective is part of the NUMEN project and will be discussed in the article. © 2018, SIF, Springer-Verlag GmbH Germany, part of Springer Nature.
