Background\ud \ud In patients with severe brain injury, withdrawal of life-sustaining measures (WLSM) is common in intensive care units (ICU). WLSM constitutes a dilemma: instituting WLSM too early could result in death despite the possibility of an acceptable functional outcome, whereas delaying WLSM could unnecessarily burden patients, families, clinicians, and hospital resources. We aimed to describe the occurrence and timing of WLSM, and factors associated with timing of WLSM in European ICUs in patients with traumatic brain injury (TBI).\ud \ud \ud \ud Methods\ud \ud The CENTER-TBI Study is a prospective multi-center cohort study. 211. Could immune-related hepatitis rapidly progress to immune-related cirrhosis?

- Abstract
BACKGROUND: Immune-related hepatitis is one of the prevalent adverse events associated with immunotherapy, especially immune checkpoint inhibitors (ICIs). For patients without a history of liver disease, autoimmune disease, or alcohol consumption, it is not clear whether immune-related hepatitis could rapid progress to immune-related cirrhosis. CASE REPORT: We report the case of a 54-year-old female with stage IIIB primary pulmonary lymphoepithelioma-like carcinoma (PLELC) diagnosed with immune-related hepatitis. After 15 months, a liver biopsy demonstrated the rapid progression of liver cirrhosis although systematic corticosteroid administration. CONCLUSIONS: Long-term immune activation caused by ICIs may exacerbate the process of cirrhosis. Great attention should be paid to the rapid progression to liver cirrhosis of immune-related hepatitis in the clinic. [ABSTRACT FROM AUTHOR]

Methods: RT-qPCR was used to detect miR-129-5p expression in tumor tissues and adjacent normal tissues from patients with prostate cancer. The cell proliferation assay and colony forming assay were used to study the role of miR-129-5p in mediating prostate cancer cell growth. Bioinformatic analysis and dual luciferase assay were performed to predict and confirm ETV1 as a target gene of miR-129-5p. Results: We found that miR-129-5p levels were decreased significantly in human prostate cancer tissues compared with matched normal tissues from patients with prostate cancer. Overexpression of miR-129-5p suppressed prostate cancer cell growth while antagonist of miR-129-5p promoted cell proliferation in immortal prostate cell line RWPE-1. In addition, elevation of miR-129-5p decreased ETV1 expression in prostate cancer cells while downregulation of miR-129-5p increased ETV1 expression in RWPE-1. Mechanistically, ETV1 is confirmed a direct target of miR-129-5p in prostate cancer cells. Through repression of ETV1 expression, miR-129-5p could inactivate YAP signaling in prostate cancer cells. In addition, overexpression of ETV1 attenuated miR-129-5p induced cell proliferation in prostate cancer cells. Correlation analysis further revealed that there was a negative correlation between miR-129-5p levels and ETV1 mRNA levels in tumor tissues from patients with prostate cancer. First, survival analysis was performed, and IGF-1’s effects on long-term cognition and depressive emotion were assessed. Second, the characteristics of IGF-1 function in cognition were evaluated. Finally, cytochrome C, caspase-9, tumor necrosis factor receptor (TNFR), and caspase-8 levels as well as cell apoptosis in the hippocampus were evaluated.Results: IGF-1 did not reduce mortality or alleviate depressive symptoms in septic rats, but improved the memory of noxious stimulation and spatial learning and memory. These effects were observed only when IGF-1 was administered within 0–6 hours after CLP. Moreover, cytochrome C and caspase-9 expression levels were increased at 6 hours after CLP in the hippocampus, while TNFR and caspase-8 amounts were not increased until 12 hours after CLP. Cell apoptosis increased at 12 hours after CLP, but was inhibited by IGF-1.Conclusion: Cognitive impairment in rats recovering from SE is associated with cell apoptosis in the hippocampus. Supplementation of IGF-1 reduces cell apoptosis by preventing the over-expression of cytochrome C and TNFR, and results in improved cognitive function. However, improvement only occurs when IGF-1 is administered at the early stage (within 6 hours) of sepsis. As cytochrome C activation occurs earlier than that of TNFR in this study, cytochrome C may be the main factor inducing apoptosis in early SE. Single-layer transition metal dichalcogenides (TMDCs) are showing promising thermoelectric applications due to their superior stability and electronic transport properties. Unfortunately, the intrinsic high lattice thermal conductivity prevents their further improvement of thermoelectric performance. Motivated by recent experimental synthesis of two-dimensional TMDC heterostructures and superlattices, we propose to minimize the lattice thermal conductivity of single-layer MoS
2 and MoSe2 using the lateral superlattice (LS) as building blocks. First-principles calculations with the phonon Boltzmann transport equation reveal a remarkably low lattice thermal conductivity of MoS2 -MoSe2 LS due to the enhanced anharmonic phonon scattering as compared to the individual single-layer. We also show that the strong phonon anisotropy of MoS2 -MoSe2 LS is primarily ascribed to the out-of-plane quadratic acoustic branch and orientation-dependent anharmonic scattering. Our calculations clearly demonstrate the advantages of LS structure in minimizing the lattice thermal conductivity of single-layer TMDCs and also accelerate their related applications in the field of renewable energy. Heavy metal-contaminated sediment may release harmful ions into the water column and seriously affect the growth of aquatic organisms. Biochar is a good adsorbent for water pollutants. In this study, a simulated water ecosystem with copper (Cu)-contaminated sediment was established to study the effects of biochar addition (CK: 0 g/L, T1: 3 g/L, T2: 6 g/L, T3: 12 g/L) on Cu content in water and the growth and accumulation of Cu in a submerged macrophyte, Vallisneria natans. Compared with the CK group, Cu contents in water decreased by 43.65%, 33.86%, and 54.10% in the T1, T2, and T3 groups, respectively. Cu contents in the aboveground part of V. natans decreased by 56.03%, 61.90%, and 52.38% in T1, T2, and T3, respectively, while in the belowground part Cu contents decreased by 17.46%, 11.94%, and 55.90%, respectively. The plant height, fresh weight, and chlorophyll content significantly increased with the increase of biochar addition, showing that biochar promoted the growth of V. natans. This study demonstrated that the addition of biochar in water could significantly reduce the Cu content in water and the accumulation of Cu in V. natans, thus reducing the harm caused by Cu to V. natans. The environmental behavior of arsenic (As) in soil is closely related to the composition of the soil microbial community and soil enzyme activity. In this paper, the effects of inorganic selenium (Se(IV), Se(VI)) and organic Se (Se-Cys, Se-Met) on enzyme activity and microbial community composition in As-contaminated soil were studied by a pot experiment under greenhouse conditions. The results showed that the application of inorganic Se and organic Se can significantly affect the activities of urease, catalase, invertase, fluorescein diacetate (FDA) hydrolase and dehydrogenase in As-contaminated soil. The total phospholipid fatty acid (PLFA) microbial content in the soil with organic Se treatments was higher than that in the control group (CK), while the inorganic Se treatments was opposite. Organic Se promoted the growth of microorganisms in the soil, while the inorganic Se treatments was opposite. From the perspective of the distribution and community structure of microorganisms, organic Se treatment enriches the microbial diversity and increases the content of gram-positive bacteria (G
+ ), gram-negative bacteria (G- ), and fungi, while the inorganic Se treatment group shows the opposite trend. This study can provide a reference for in-depth exploration of the mechanism of Se and microbial metabolism of As and its application in the treatment of As pollution. For investigating the response of chlorophyll fluorescence characteristics under single and combined of Cu or Zn stress, Vallisneria natans was selected as the experimental subject. V. natans was cultured 21 days under six concentration levels of Cu or Zn. The results showed that Fv/Fm, Fv/Fo, Y (II) and qP increased, and other indicators were almost unchanged in low- concentration treatment (Cu≤0.4mg/L, Zn≤4.0mg/L, Cu+Zn≤0.2+2.0 mg/L). V. natans could still maintain normal growth within such a range of heavy metal concentration, and it could maintain relatively a high photosynthetic activity. When heavy metal concentration was higher than the above concentration, the photosynthetic activity was inhibited. Fv/Fm, Fv/Fo, Y (II) and qP decreased gradually with the increasing stress concentration and stress time, while Y(NO), Y (NPQ), and qN showed different degrees of increase. The combined stress of Cu and Zn had a synergistic effect on the photosynthetic activity. V. natans can be used for ecological restoration of polluted water with low concentration of Cu and Zn. The food problem is a major problem of common concern in the world, and the prediction of irrigation area can promote the solution of food and agricultural problems. In this paper, the data of grain production and irrigation area in the world are analyzed. An improved Random Forest Regression model is proposed and applied to the prediction of irrigation area. Based on ordinary Random Forest and Limit Tree Regression algorithm, an improved random forest prediction model for irrigation area in China is proposed. Firstly, the arithmetic mean value (AMM) of mean square error (MSE) and mean absolute error (MAE) was used as the evaluation index of the improved impure function and irrigation area prediction effect. Then, the grid search method is used to determine the optimal number of decision trees (70 trees and 30 trees respectively) in ordinary random forest and limit tree regression, and a new improved random forest model is established. After following, the model is compared with other prediction models, and 10 fold cross validation shows the rationality of the model. Finally, the error analysis of the improved Random Forest model shows that the prediction error is small. It is expected to be applied in the annual analysis of irrigation area in China.

The aim of this study is to assess variation in levels of early secondary referral within European SNCs and to compare the outcomes of directly admitted and secondarily transferred patients. Methods: Patients with moderate and severe TBI (Glasgow Coma Scale < 13) from the prospective European CENTER-TBI study were included in this study. All participating hospitals were specialist neuroscience centers. First, adjusted between-country differences were analysed using random effects logistic regression where early secondary referral was the dependent variable, and a random intercept for country was included. Second, the adjusted effect of early secondary referral on survival to hospital discharge and functional outcome [6 months Glasgow Outcome Scale Extended (GOSE)] was estimated using logistic and ordinal mixed effects models, respectively. Results: A total of 1347 moderate/severe TBI patients from 53 SNCs in 18 European countries were included. Of these 1347 patients, 195 (14.5%) were admitted after early secondary referral. Secondarily referred moderate/severe TBI patients presented more often with a CT abnormality: mass lesion (52% vs. 34%), midline shift (54% vs. 36%) and acute subdural hematoma (77% vs. 65%). After adjusting for case-mix, there was a large European variation in early secondary referral, with a median OR of 1.69 between countries. Early secondary referral was not associated with functional outcome (adjusted OR 1.07, 95% CI 0.78–1.69), nor with survival at discharge (1.05, 0.58–1.90). Conclusions: Across Europe, substantial practice variation exists in the proportion of secondarily referred TBI patients at SNCs that is not explained by case mix. Within SNCs early secondary referral does not seem to impact functional outcome and survival after stabilisation in a non-specialised hospital. Future research should identify which patients with TBI truly benefit from direct transportation.

- Abstract
In March 2021, following the annual International Committee on Taxonomy of Viruses (ICTV) ratification vote on newly proposed taxa, the phylum Negarnaviricota was amended and emended. The phylum was expanded by four families (Aliusviridae, Crepuscuviridae, Myriaviridae, and Natareviridae), three subfamilies (Alpharhabdovirinae, Betarhabdovirinae, and Gammarhabdovirinae), 42 genera, and 200 species. Thirty-nine species were renamed and/or moved and seven species were abolished. This article presents the updated taxonomy of Negarnaviricota as now accepted by the ICTV.

- Abstract
The move from reading to writing the human genome offers new opportunities to improve human health. The United States National Institutes of Health (NIH) Somatic Cell Genome Editing (SCGE) Consortium aims to accelerate the development of safer and more-effective methods to edit the genomes of disease-relevant somatic cells in patients, even in tissues that are difficult to reach. Here we discuss the consortium's plans to develop and benchmark approaches to induce and measure genome modifications, and to define downstream functional consequences of genome editing within human cells. Central to this effort is a rigorous and innovative approach that requires validation of the technology through third-party testing in small and large animals. New genome editors, delivery technologies and methods for tracking edited cells in vivo, as well as newly developed animal models and human biological systems, will be assembled-along with validated datasets-into an SCGE Toolkit, which will be disseminated widely to the biomedical research community. We visualize this toolkit-and the knowledge generated by its applications-as a means to accelerate the clinical development of new therapies for a wide range of conditions.

- Abstract
This article is a multi‐vocal account, a form of writing differently, which captures our changing lives and livelihoods under the present global health crisis. Through the process of writing, we create a safe space to understand how the COVID‐19 pandemic exposes our gendered, intersectional lives. Our writing gives voice to suppressed thoughts and embodied affects as they surface in relation to entrenched structural inequalities where we witness the marginalization of intersectional difference, in our case women, the feminine, and race in academia and neoliberal society. By rendering visible the structural inequalities that have become amplified during the pandemic, and the ways in which these inequalities have affected our everyday lives, we are able to give witness to intersectional differences. Our multi‐vocal embodied text is offered as an emancipatory, affective mobilization of our lives, encompassing feelings of grief, loss, fear, anger, frustration, and vulnerability. This collective piece of writing gives rise to solidarity in a crisis‐stricken world where we choose to live with hope.

- Abstract
Background: Fluid therapy—the administration of fluids to maintain adequate organ tissue perfusion and oxygenation—is essential in patients admitted to the intensive care unit (ICU) with traumatic brain injury. We aimed to quantify the variability in fluid management policies in patients with traumatic brain injury and to study the effect of this variability on patients' outcomes. Methods: We did a prospective, multicentre, comparative effectiveness study of two observational cohorts: CENTER-TBI in Europe and OzENTER-TBI in Australia. Patients from 55 hospitals in 18 countries, aged 16 years or older with traumatic brain injury requiring a head CT, and admitted to the ICU were included in this analysis. We extracted data on demographics, injury, and clinical and treatment characteristics, and calculated the mean daily fluid balance (difference between fluid input and loss) and mean daily fluid input during ICU stay per patient. We analysed the association of fluid balance and input with ICU mortality and functional outcome at 6 months, measured by the Glasgow Outcome Scale Extended (GOSE). Patient-level analyses relied on adjustment for key characteristics per patient, whereas centre-level analyses used the centre as the instrumental variable. Findings: 2125 patients enrolled in CENTER-TBI and OzENTER-TBI between Dec 19, 2014, and Dec 17, 2017, were eligible for inclusion in this analysis. The median age was 50 years (IQR 31 to 66) and 1566 (74%) of patients were male. The median of the mean daily fluid input ranged from 1·48 L (IQR 1·12 to 2·09) to 4·23 L (3·78 to 4·94) across centres. The median of the mean daily fluid balance ranged from −0·85 L (IQR −1·51 to −0·49) to 1·13 L (0·99 to 1·37) across centres. In patient-level analyses, a mean positive daily fluid balance was associated with higher ICU mortality (odds ratio [OR] 1·10 [95% CI 1·07 to 1·12] per 0·1 L increase) and worse functional outcome (1·04 [1·02 to 1·05] per 0·1 L increase); higher mean daily flui
