201. AGRONOMIC AND MOLECULAR ANALYSIS OF HETEROSIS IN ALFALFA

Abstract

Double 'free-hybrids' (DH) in alfalfa, a polyploid, allogamous and perennial forage crop, were obtained by crossing in a diallelic scheme six simple hybrids (SH), multiplied for two generations (2S2Syn3), derived from four partly inbred (S2) constituents (Rotili et al., 1999). Specific Combining Ability (SCA) source of variation for dry matter yield (DMY) resulted highly significant and larger than General Combining Ability (GCA) component and supported heterosis values of DHs vs the best parent of +45% on average, ranging from +76 to +5%. The investigation at the molecular level was carried out by means of SSR marker analysis on the 6 parental SHs and the 15 DH progenies and by comparison of gene expression profiles of a single DH and the respective parental plants. The parental genetic diversity, estimated by SSR markers, showed a significant relationship with heterosis and SCA effects (r= 0.70 and 0.76, respectively). The variation of heterozygosity estimates of the DHs explained a little part (about 20%) of their variation in DMY (r = 0.45 n.s.) while the number of alleles was significantly related to DM performance (r = 0.61 P < 0.05). A microarray analysis of the transcriptome was conducted in a highly performing hybrid (DH) compared to its parents (2S2Syn3). T-test enabled to identify genes with additive/non additive (according to the nomenclature as reviewed in Hochholdinger and Hoecker, 2007) value of expression in the hybrid compared to the parents (P < 0.05, Benjamini and Hochberg False discovery rate). Q-PCR validated the level of expression from microarray analysis for a few selected genes. In our experiment, most of the variation in gene expression was additive (87%). Interestingly among the genes with non-additive pattern of expression the greater proportion of probe sets (86%) fell outside the parental range, namely above/below the level of expression of the high/low parent; among such genes ontology classes highly represented are metabolism and genetic information processing. Abstract

OBJECTIVE: The aim of the study was to recognize clusters of diseases among hospitalized elderly and to identify groups of patients at risk of in-hospital death and adverse clinical events according to disease clustering. METHOD: This was a cross-sectional study conducted in 38 internal medicine and geriatric wards in Italy participating in the Registro Politerapie SIMI (REPOSI) study during 2008. The subjects were 1,332 inpatients aged 65 years or older. Clusters of diseases (i.e., two or more co-occurrent diseases) were identified using the odds ratios (OR) for the associations between pairs of conditions, followed by cluster analysis. Logistic regression models were used to evaluate the effect of disease clusters on in-hospital death and adverse clinical events. RESULTS: A total of 86.7% of the patients were discharged, 8.3% were transferred to another hospital unit, and 5.0% died during hospitalization; 36.4% of the patients had at least one adverse clinical event. Patients affected by the clusters, including heart failure (HF) and either chronic renal failure (CRF) or chronic obstructive pulmonary disease, had a significant association with in-hospital death (OR, 4.3;95% confidence interval [CI], 1.6-11.5; OR, 2.9; 95% CI, 1.1-8.3, respectively), as well as patients affected by CRF and anemia (OR, 6.1; 95% CI, 2.3-16.2). The cluster including HF and CRF was also associated with adverse clinical events (OR, 3.5; 95% CI, 1.5-7.8). The effect of both HF and CRF and anemia and CRF on in-hospital death was additive. CONCLUSION: Several groups of older patients at risk of in-hospital death and adverse clinical events were identified according to disease clustering. Knowledge of the relationship among co-occurring diseases may help developing strategies to improve clinical practice and preventative interventions.

206. Synergistic activity of everolimus and 5-aza-2'-deoxycytidine in medullary thyroid carcinoma cell lines.

Abstract

Medullary thyroid cancer (MTC) is a tumor highly resistant to chemo- and radiotherapy. Drug resistance can be induced by epigenetic changes such as aberrant DNA methylation. To overcome drug resistance, we explored a promising approach based on the use of 5-aza-2'-deoxycytidine (AZA), a demethylating agent, in combination with the mTOR inhibitor everolimus in MTC cells (MZ-CRC-1 and TT). This combined treatment showed a strong synergistic antiproliferative activity through the induction of apoptosis. The effect of everolimus and/or AZA on genome-wide expression profiling was evaluated by Illumina BeadChip in MZ-CRC-1 cells. An innovative bioinformatic pipeline identified four potential molecular pathways implicated in the synergy between AZA and everolimus: PI3K-Akt signaling, the neurotrophin pathway, ECM/receptor interaction, and focal adhesion. Among these, the neurotrophin signaling pathway was most directly involved in apoptosis, through the overexpression of NGFR and Bax genes. The increased expression of genes involved in the NGFR-MAPK10-TP53-Bax/Bcl2 pathway during incubation with AZA plus everolimus was validated by western blotting in MZ-CRC-1 cells. Interestingly, addition of a neutralizing anti-NGFR antibody inhibited the synergistic cytotoxic activity between AZA and everolimus. These results open a new therapeutic scenario for MTC and potentially other neuroendocrine tumors, where therapy with mTOR inhibitors is currently approved. [ABSTRACT FROM AUTHOR]

Abstract

Background: The aim of the present study was to evaluate the association between heart failure (HF) and chronic kidney disease (CKD) in tertiary care centers using the clinical records of patients enrolled in internal medicine departments.Patients and methods: We used the clinical records of 1380 elderly patients to identify patients with a history of HF and CKD using admission ICD codes and glomerular filtration rate (GFR) formulas. Magnitude and strength of such associations were investigated by univariable and multivariable analysis.Results: Of the 1380 patients enrolled, 27.9% had HF (age 80 ± 7, BMI 27 ± 6 kg/m2) and 17.4% CKD (age 81 ± 7, BMI 26.8 ± 6 kg/m2). Both groups were significantly older (P <' 0.0001) with BMI higher than the patients without those diagnosis (P < 0.02). Patients with a history of CKD showed higher non-fasting glycaemia (140 ± 86 vs. 125 ± 63 mg/dL, P < 0.001). CKD was significantly associated with HF (P < 0.0001). Patients with HF had an estimated GFR lower than patients without HF (P < 0.0001). Comorbidity and severity indices were significantly higher in subjects with HF (P < 0.0001) and CKD (P < 0.0001) than in those without. Multivariable analysis showed a significant association between HF and age (for five years increase OR 1.13, P < 0.009), BMI (for each 3 kg/m2 increase OR 1.15, P < 0.001), GFR (for each decrease of 10 mL/min increase OR 0.92, P < 0.002) and severity index (IS) (for each 0.25 units increase OR 1.43, P < 0.001).Conclusion: HF on admission is strongly associated with CKD, older age, BMI, and SI. These data focus the value of epidemiological studies such REPOSI in identifying and monitoring multimorbidity in elderly.

Abstract

The dramatic demographic changes that are occurring in the third millennium are modifying the mission of generalist professionals such as primary care physicians and internists. Multiple chronic diseases and the related prescription of multiple medications are becoming typical problems and present many challenges. Unfortunately, the available evidence regarding the efficacy of medications has been generated by clinical trials involving patients completely different from those currently admitted to internal medicine: much younger, affected by a single disease and managed in a highly controlled research environment. Because only registries can provide information on drug effectiveness in real-life conditions, REPOSI started in 2008 with the goal of acquiring data on elderly people acutely admitted to medical or geriatric hospital wards in Italy. The main goals of the registry were to evaluate drug prescription appropriateness, the relationship between multimorbidity/polypharmacy and such cogent outcomes as hospital mortality and re-hospitalization, and the identification of disease clusters that most often concomitantly occur in the elderly. The findings of 3-yearly REPOSI runs (2008, 2010, 2012) suggest the following pertinent tasks for the internist in order to optimally handle their elderly patients: the management of multiple medications, the need to become acquainted with geriatric multidimensional tools, the promotion and implementation of a multidisciplinary team approach to patient health and care and the corresponding involvement of patients and their relatives and caregivers. There is also a need for more research, tailored to the peculiar features of the multimorbid elderly patient.

213. Quantum Landauer erasure with a molecular nanomagnet

Abstract

The erasure of a bit of information is an irreversible operation whose minimal entropy production of kBln 2 is set by the Landauer limit1. This limit has been verified in a variety of classical systems, including particles in traps2,3and nanomagnets4. Here, we extend it to the quantum realm by using a crystal of molecular nanomagnets as a quantum spin memory and showing that its erasure is still governed by the Landauer principle. In contrast to classical systems, maximal energy efficiency is achieved while preserving fast operation owing to its high-speed spin dynamics. The performance of our spin register in terms of energy–time cost is orders of magnitude better than existing memory devices to date. The result shows that thermodynamics sets a limit on the energy cost of certain quantum operations and illustrates a way to enhance classical computations by using a quantum system.

Abstract

Pharmacological thromboprophylaxis (TP) is known to reduce venous thromboembolism (VTE) in medical inpatients, but the criteria for risk-driven prescription, safety and impact on mortality are still debated. We analyze data on elderly patients with multimorbidities admitted in the year 2010 to the Italian internal medicine wards participating in the REPOSI registry to investigate the rate of TP during the hospital stay, and analyze the factors that are related to its prescription. Multivariate logistic regression, area under the ROC curve and CART analysis were performed to look for independent predictors of TP prescription. Association between TP and VTE, bleeding and death in hospital and during the 3-month post-discharge follow-up were explored by logistic regression and propensity score analysis. Among the 1,380 patients enrolled, 171 (15.2 %) were on TP during the hospital stay (162 on low molecular weight heparins, 9 on fondaparinux). The disability Barthel index was the main independent predictor of TP prescription. Rate of fatal and non-fatal VTE and bleeding during and after hospitalization did not differ between TP and non-TP patients. In-hospital and post-discharge mortality was significantly higher in patients on TP, that however was not an independent predictor of mortality. Among elderly medical patients there was a relatively low rate of TP, that was more frequently prescribed to patients with a higher degree of disability and who had an overall higher mortality.

The article revisits the trajectory of this contraceptive in Brazil from its arrival through clinical trials to its eventual ban in 1986 by the Brazilian regulatory agency responsible for approving medications at the time. Its circulation generated controversies related to research practices, side effects and political uses of the drug as a birth control method. This article focuses its analysis on the divergences related to research practices. It uses a controversy analysis technique, reviewing the versions of those involved, investigating their understandings and the effects that this object generated in their networks. Norplant® provoked displacements and associations between civil society groups, State authorities, scientists and physicians, industry, pharmaceutical products, research procedures, bureaucratic instruments, and the female users of the contraceptives. Abstract

We conducted an investigation on the orientations and geometrical content of the Mithraea of Ostia Antica. A geometrical CAD study based on the most reliable plans from the Archives of the Sovrintendenza Speciale per il Colosseo, il MNR e l'Area Archeologica di Roma - Ostia Antica, aside with the identification of geometrical and numerical schemes and associated length units, allowed us to identify the axes of the Mithraea. Then the orientations of the axes were measured on field by means of a professional compass with ± 0.5° uncertainty. The distribution of these measured azimuths follows the topography of the city. On the contrary, the Decumanus, the main street of the town, presents an indubitable orientation toward the Winter Solstice Sunset. This was ascertained by measuring the coordinates of four significant points along the street with the help of a palmar Trimble GPS which averages 100 measures per point. The exceptional density of Mithraea in Ostia led us to suppose that a symbolic cosmic-solar value is to be searched in the orientation as a whole of the town itself, founded half a millennium before the first presence of Mithraism in Rome. The Decumanus could be considered the very holder of this symbolic value with its orientation toward the Winter Solstice Sunset, possibly making of Ostia a very special place for Mithraicism. Abstract

Embedding a magnetic electroactive molecule in a three-terminal junction allows for the fast and local electric field control of magnetic properties desirable in spintronic devices and quantum gates. Here, we provide an example of this control through the reversible and stable charging of a single all-organic neutral diradical molecule. By means of inelastic electron tunnel spectroscopy we show that the added electron occupies a molecular orbital distinct from those containing the two radical electrons, forming a spin system with three antiferromagnetically coupled spins. Changing the redox state of the molecule therefore switches on and off a parallel exchange path between the two radical spins through the added electron. This electrically controlled gating of the intramolecular magnetic interactions constitutes an essential ingredient of a single-molecule SWAPquantum gate.

232. A Generalized Approach for the Modeling of Goodwin-Type Cycles

Abstract

Goodwin’s celebrated growth cycle model has been widely studied since its introduction in 1967. In recent years several contributions have appeared with the aim of amending the original model so as to improve its economic coherence and enrich its structure. In this article we propose a new and generalized approach, within the theory of planar Hamiltonian systems, for the modeling of Goodwin-type cycles. This new approach, which includes and improves various attempts by the recent literature, is very general and fulfills the essential requirement that the orbits lie entirely in the economically feasible interval. We provide a necessary and sufficient condition for all solutions to be cycles lying entirely in the unit box. In addition, we study the period length of the cycles near the equilibrium and close to the boundary of the domain. Finally, we discuss an example of how small perturbations of the model may affect the qualitative behavior of the solutions.

233. Copper-alloy use in a Tyrrhenian medieval town: The case of Leopoli-Cencelle (Italy)

Abstract

This study focuses on the use of copper-alloys in the Tyrrhenian town of Leopoli-Cencelle (Italy) during the Middle Ages, and explores the technical aspects and sociocultural implications of metal consumption. Founded near the coast of the Tuscia region as a reaction to threats from Saracens, and located within the main trade routes linking Rome to the northern Tuscan powers of Pisa and Siena, Leopoli-Cencelle was an active town up to the 14th century. This was also facilitated by the proximity of the Tolfa Hills mining district and the important harbour of Corneto. This research concentrates on the technological and compositional study of copper-alloy artefacts such as everyday life objects, dress accessories, work instruments, jewellery, and structural and decorative items. The analytical data reported here includes pXRF, optical microscopy and SEM-EDS, which were employed to acquire information on copper-alloy composition and manufacturing processes. The results allow an assessment of the main alloys employed and their uses, which can be compared to information from medieval written sources, and they provide insight into the nature of the local workshops, and the skill and level of specialisation of metalworkers. Some interesting aspects are the relative scarcity of tin bronzes, which are almost exclusively reserved for bell casting, and the presence of gilded or silvered brasses, which denote influences from the eastern Mediterranean world. The comparison of metal surface and bulk compositions also provides a contribution to evaluate the potential of pXRF analyses on unprepared metal samples.

235. Chemical and rheological analysis of unaged and aged bio-extended binders containing lignin

Abstract

The use of alternative renewable sources to totally or partially replace bitumen is one of the most current challenges in the road pavement sector. The growing energy cost and environmental concerns lead to the necessity to find alternative solutions, by supporting at the same time sustainability and circular economy principles. Within this framework, this paper presents the application of a powder lignin, a natural bio-polymer deriving from by-products of wood pulp and paper industry, to replace part of bitumen. The bituminous blend consisting in 70% of bitumen and 30% of powder lignin (by weight) was made in laboratory through the use of a high shear stirring mixer, and a reference plain bitumen characterised by a similar consistency (i.e., same penetration grade) was used as comparison. Then, an extensive investigation on chemical and rheological properties of the bio-binder is presented. Fourier transform infrared spectroscopy (FTIR), saturates, aromatics, resins and asphaltenes (SARA), bending beam rheometer (BBR), frequency sweep tests and multiple stress creep recovery tests (MSCR) with a dynamic shear rheometer (DSR) were performed. Moreover, unaged, short- and long-term aging conditions were considered. Results indicated that powder lignin dominates the rheological behaviour of the bio-binder and, from chemical analysis, it seems that it partially acts as a filler and partially as a binder. This would result in improved performances at both low and high temperatures, leading to a wider temperature range of performance grade (PG). Moreover, despite a stiffening effect is recognised, lignin also offers an antioxidant potentiality, reducing the aging susceptibility of the investigated bio-binder.

237. 560 A RARE CASE OF PERICARDIAL SARCOMA

Abstract

A 47 years old man, with no previous medical history, presented to the emergency department for dyspnea increased over the prior weeks. The echocardiographic evaluation revealed a severe pericardial effusion. Moreover, a round-shaped, indeterminate mass was detected into the pericardium, partially enhanced after injection SonoVue R contrast. After the pericardiocentesis, to exclude a severe dilatation of the coronary sinus, the patient underwent to a coronary angiography that showed a diffuse vascularization of the region of the mass. A CT-scan were then performed, and confirmed the presence of a well-defined mass placed near to the left atrial-ventricular junction into the pericardium. The magnetic resonance revealed a limited area of infiltration of the lesion into the myocardium. Finally the patient underwent surgical asportation, and the biopsy showed an high-grade sarcoma. Primary cardiac tumors are rare diseases and in the majority of the cases are of benign nature. Among malignant tumor of the heart sarcoma is the most common. Despite their rarity, malignant heart cancers are associated with high mortality and the prognosis is mostly influenced by the localization of the tumor, its biological aggressiveness, and the difficulty in obtaining surgical radicality. Cardiac sarcomas are most likely to arise from the right atrium followed by right ventricle and less often from the epicardium. In these cases, the asportation of the mass can be challenging, and the risk of myocardial narrowing can affect the area of resection.We reported a rare case of primary cardiac sarcoma mainly localized into the pericardium. Due to its position, the surgical asportation of the tumor was considered complete and our patient was discharge from the hospital without further complication and with a program of chemotherapy.At six months follow-up no recurrence of disease was observed.

240. Supply chain risk management: creating an agenda for future research

Abstract

Supply chains are becoming globalised and resulting complex interaction between supply chain actors and increasing interdependency between supply chain risks pose a real challenge in modelling and managing these risks. The purpose of this paper is to conduct a comprehensive review of literature in the field of 'supply chain risk management' and identify important research gaps for future research. We have adopted the method of 'systematic literature review' using text mining software and examined peer-reviewed articles published over a period of last 15 years (2000-2014). Keeping in view the interdependent nature of interaction between supply chain risks, there is an urgent need of developing taxonomy of risks representing causal chains of risk sources, risk events and consequences instead of relying on the existing conventional classifications that assign risks to independent categories. Existing techniques have either considered risks as independent factors or focused on solving specific problems resulting in unrealistic and sub-optimal solutions respectively. We propose a new risk management framework that can help researchers model interdependency between supply chain risks and enable practitioners visualise dynamic and holistic interaction between these risks across multiple facets of a supply network.

245. Evaluation and Treatment of Post-Stroke Pusher Behaviour: literature review and case report.

Abstract

Background The Pusher Behaviour is a disorder exhibited by some stroke patients, who push themselves with their non-affected limbs towards the hemiplegic side and resist attempts at passive corrections of their tilted posture. The reported incidence varies widely from 10.4% to 63%, depending on the assessment methodology. A number of Authors have studied Pusher Behaviour, highlighting different correlations with neglect and brain damage localization. All of them found that this syndrome will slow down the functional recovery process and cause a long hospital stay. The literature shows three validated scales assessing pusher behaviour and only case reports or experts opinions about its treatment. Aim of this study was to describe in a case report a methodology for evaluation and treatment of pusher behaviour based on motor learning theories and 'experts opinions'. Methods Four patients showing symptoms were assessed. Three of them were not included because their Scale for Contraversive Pushing score was below the criteria of cut-off that had been established, or due to comorbidities. Only one patient received the mixed approaches treatment for 30 min. a day instead of equal time of classic physiotherapy. Results The patient reached a good functional recovery in six weeks of treatment. Comparison with the published case reports is not possible because of the lack of homogeneity in the reported data. Conclusions Further investigations using homogeneous assessment tools and involving physiotherapists are needed. [ABSTRACT FROM AUTHOR]

Abstract: We are dealing with the problem of counting the paths joining two points of a chessboard in the presence of a barrier. The formula for counting all the paths joining two distinct positions on the chessboard lying always over a barrier is well known (see for example Feller (1968) , Kreher and Stinson (1999) ). The problem is here extended to the calculation of all the possible paths of movements which stay exactly times, , over the barrier. Such a problem, motivated by the study of financial options of Parisian type, is completely solved by virtue of five different formulas depending on the initial and final positions and on the level of the barrier. Abstract

In the present paper the feasibility of the anodic dissolution of micrometric Cu and submicrometric Co powders in a fluidised bed electrode was investigated. The aqueous solution and fluidising gas were chosen to avoid any chemical contribution to the corrosion process. The process was carried out either galvanostatically or potentiostatically, and clean and oil contaminated powders were considered. Typical dissolution efficiencies of ∼ 15 mg h-1 were obtained in a laboratory reactor holding ∼ 10 g powder and ∼ 100 mL anolyte. A simple mathematical model is proposed and validated, accounting for the principal design and operating parameters of the fluidised bed electrode. [ABSTRACT FROM AUTHOR]
