202. A nonlinear model for mammary gland growth and regression in lactating sows

Abstract

The objective was to propose an empirical mathematical model to describe mammary gland growth and regression in lactating sows. A nonlinear dynamic model based on the logistic function was constructed, and data from 61 sows were used to illustrate the model. Sows were fed four diets with two levels of energy and of protein during lactation, and individuals were slaughtered over a 30-d period to produce a cross sectional data set on weight and composition variables from suckled mammary glands. Data (y(x)) were obtained for each day of lactation (x) and fitted by nonlinear regression. The logistic distribution function was modified for different durations of growth (f; days/gram of weight or composition) and regression (g; days/gram of weight or composition): [formula in text] where y(max) is maximum weight or composition and x(max) is day of lactation at maximum. Based on results for wet weight, for example, individually suckled mammary glands grow until between Day 21 and 28 of lactation and reach a maximum of about 500 to 600 g, depending on diet. Growth pattern of mammary glands can be described well with an asymmetric nonlinear model, using different durations for growth and regression. From this model, it was possible to estimate directly biologically important parameters: maximum weight or composition, day of lactation at maximum weight or composition, and durations of growth and regression. This model can be applied to describe mammary gland growth patterns for other species and to describe similar growth or production patterns.

Abstract

The objective of this paper is to introduce fuzzy set theory and develop fuzzy mathematical models to assess sustainable development based on context-dependent economic, ecological, and societal sustainability indicators. Membership functions are at the core of fuzzy models, and define the degree to which indicators contribute to development. Although a decision-making process regarding sustainable development is subjective, fuzzy set theory links human expectations about development, expressed in linguistic propositions, to numerical data, expressed in measurements of sustainability indicators. In the future, practical implementation of such models will be based on elicitation of expert knowledge to construct a membership function. The fuzzy models developed in this paper provide a novel approach to support decisions regarding sustainable development.

Abstract

BACKGROUND: Mutation in the progranulin gene (GRN) can cause frontotemporal dementia (FTD). However, it is unclear whether some rare FTD-related GRN variants are pathogenic and whether neurodegenerative disorders other than FTD can also be caused by GRN mutations. OBJECTIVES: To delineate the range of clinical presentations associated with GRN mutations and to define pathogenic candidacy of rare GRN variants. DESIGN: Case-control study. SETTING: Clinical and neuropathology dementia research studies at 8 academic centers. PARTICIPANTS: Four hundred thirty-four patients with FTD, including primary progressive aphasia, semantic dementia, FTD/amyotrophic lateral sclerosis (ALS), FTD/motor neuron disease, corticobasal syndrome/corticobasal degeneration, progressive supranuclear palsy, Pick disease, dementia lacking distinctive histopathology, and pathologically confirmed cases of frontotemporal lobar degeneration with ubiquitin-positive inclusions (FTLD-U); and 111 non-FTD cases (controls) in which TDP-43 deposits were a prominent neuropathological feature, including subjects with ALS, Guam ALS and/or parkinsonism dementia complex, Guam dementia, Alzheimer disease, multiple system atrophy, and argyrophilic grain disease. MAIN OUTCOME MEASURES: Variants detected on sequencing of all 13 GRN exons and at least 80 base pairs of flanking introns, and their pathogenic candidacy determined by in silico and ex vivo splicing assays. RESULTS: We identified 58 genetic variants that included 26 previously unknown changes. Twenty-four variants appeared to be pathogenic, including 8 novel mutations. The frequency of GRN mutations was 6.9% (30 of 434) of all FTD-spectrum cases, 21.4% (9 of 42) of cases with a pathological diagnosis of FTLD-U, 16.0% (28 of 175) of FTD-spectrum cases with a family history of a similar neurodegenerative disease, and 56.2% (9 of 16) of cases of FTLD-U with a family history. 225. Safety and feasibility of liver-directed ex vivo gene therapy for homozygous familial hypercholesterolemia

Abstract

OBJECTIVE: The purpose of this report was to provide detailed information on the safety and feasibility of surgical procedures associated with the first ex vivo liver-directed gene therapy trial for the treatment of vivo gene therapy for homozygous familial hypercholesterolemia (FH). SUMMARY BACKGROUND DATA: Familial hypercholesterolemia is an autosomal dominant disease in which the gene encoding the low density lipoprotein receptor is defective. Patients homozygous for this mutation have extraordinarily high levels of cholesterol and accelerated atherosclerosis and die prematurely of myocardial infarction. The concept of liver-directed gene therapy was based on the report of normalization of cholesterol levels by orthotopic cardiac/liver transplant in a child with homozygous FH. METHODS: Five patients with homozygous FH were selected for inclusion in this trial. The patients underwent hepatic resection and placement of a portal venous catheter. Primary hepatocytes cultures were prepared from the resected liver and transduced with a recombinant retrovirus encoding the gene for the human low density lipoprotein receptor. The genetically modified cells were then transplanted into the liver through the portal venous catheter. RESULTS: Numerous clinical, laboratory, and radiologic parameters were analyzed. Elevations of the hepatic transaminases and leukocyte counts and a decline in hematocrit count were noted. Transient elevations of the portal pressure were observed during cell infusion. No major perioperative morbidity--specifically, myocardial infarct, perioperative hemorrhage, or portal vein thrombosis--or death occurred as a result of this protocol. CONCLUSION: Liver-directed ex vivo gene therapy can be accomplished safely in humans and is appropriate for selected patients.
