Background: The number of clinical applications of different Platelet-Rich Fibrin (PRF) membranes has increased in regenerative medicine including regenerative dentistry. Intact platelets, leukocytes and stem cells of PRF play a crucial role in the local bone augmentation releasing cytokines and growth factors. An integral part of the post-surgical management is the application of mouthwashes, especially chlorhexidine digluconate, which is recommended in order to prevent postoperative infections. In some cases, it is possible that there is contact between the mouthwash and PRF membrane. Therefore, the impact of mouthwashes on cell viability of folded F-PRF was tested.

This chapter investigates the conundrum of inadvertently characterizing Dutch spatial science as ‘lagging’. It focuses on an extensive literature review, archival sources, and interviews and correspondence with involved Dutch geographers between September 2019 and December 2020. Pre-1960s innovations in Dutch human geography were driven by the discipline’s engagement in spatial planning. In 1946, George Zeegers gives a lecture where he pleads to reform the educational program to make geographers ready for a career in planning. Mass planning in post-war reconstruction meant a high demand for speedy survey work. During the mid-1960s, Dutch geography is increasingly in dialogue with the anglophone literature in geography and regional science, and authors like Peter Haggett are actively debated in student circles. The aforementioned narrative essentially tells a story of continuity and change.

In some cases there is possibility that there is contact between the mouthwash and PRF membrane. The impact of mouthwashes on cell viability of folded F-PRF was tested. Methods: 3 mouthwash brands were tested: Chorsodyl, Listerine 6 in 1 and Elmex Sensitive Plus using MTT viability assay after 30 seconds treatment and 72 hours treatment (twice daily for 30 seconds). The membrane samples were incubated in cell culture conditions. Results: 30 seconds of mouthwash treatment diminished the fresh F-PRF viability significantly by 15-21% depending on the agent. After 72 hours of treatment the viability loss was ~50%. Conclusion: The decreased number of platelets and other blood cells can not launch optimal bone morphogenesis. The MTT assay is cheap, reliable and simple method to assess the platelet and cellular viability and potential regenerative capacity of F-PRF membrane, or any platelet-rich product. The isolation of the PRF membrane from oral liquids and/or application of less aggressive mouthwashes is recommended for at least 5-7 days after PRF surgery.

In this study, a series of free-standing as well as controlled size Pt nanoparticles-loaded mesoporous metal oxides (NiO, Co3O4, CeO2 and MnO2) with high surface area and designed pore structure were prepared by hard template method and used as catalysts for CO2 hydrogenation together with dry reforming of CO2 with methane. The physicochemical properties of catalysts were analysed by N2 adsorption-desorption isotherm, XRD, TEM, and H2-TPR. Pt-free and Pt-loaded mesoporous NiO and Co3O4 performed with high catalytic activity and selectivity for both CO2 activation reactions. Pt-free NiO exhibited the highest catalytic activity and also showing 100% CH4 selectivity between 473–673 K and ˜1:1 H2/CO2 ratio between 673–973 K in CO2 hydrogenation and dry reforming with methane, respectively. The enhanced catalytic properties can be due to the existence of metallic Ni as well as the optimal dynamics of Ni/NiOx structure under reaction conditions.

Introduction: Our goal is to improve gene fusion detection via RNA sequencing by combining multiple fusion callers through machine learning techniques. Background: Gene Fusion events are important drivers of malignancy. RNA sequencing (RNAseq) methods for detection of fusions have the advantage that multiple markers can be targeted at one time. Unlike DNA methods, in which it is challenging to capture fusion breakpoints, in RNA methods fusions are readily identified through chimeric transcripts. While many fusion calling algorithms exist for use on RNAseq data, sensitive fusion callers, needed for samples of low tumor content, often present high false positive rates - a result of aligning chimeric transcripts. Further, there currently is no single feature in NGS data that can be used to filter out false positive fusion calls. In order to achieve higher accuracy in fusion calls than can be achieved using individual fusion callers, we have weighted and combined the results of multiple fusion callers by systematic and objective means: an ensemble learning approach based on random forest models. Our method selects from data generated by three independent fusion callers supplemented by metrics obtained from in-house methods. It presents a metric that can be immediately interpreted as the probability that a candidate fusion call is a true fusion call. Methods: Random forest models were generated by use of the randomForest package in R, with tuning by the R caret package. Training data sets consisted of a balanced set of 394 fusion calls from clinical samples of solid tumors. For training, fusion calls with at least 10 supporting reads were deemed true or false based on manual review via IGV, and orthogonal methods including PCR with Sanger sequencing and the commercial Archer™ fusion CTL and Sarcoma panels. We present the results of training on data from the three well-known fusion callers Arriba, STAR-Fusion, and FusionCatcher, together with additional data from an in-house developed junction counting method, and fusion membership in a list of known fusions (a “white list”). Models were validated by 10-fold cross-validation. Results: In performance evaluations, false positive and false negative calls were presumed false based on orthogonal determinations. On that basis, our current best model has an accuracy of 94.9% (sensitivity 93.4%, specificity 96.7%). Currently, High Confidence fusion calls (calls with probability score greater than 70%) are the most common positive calls. These have been confirmed with 100% success. Conclusion: We have successfully integrated multiple fusion callers by means of random forest models. Our current model is validated for use on our solid tumor fusion calling pipeline. However, research examining sex ratio and human mating is limited in scope. Prior work has examined the relationship between sex ratio and desire for short-term, uncommitted mating as well as outcomes such as marriage and divorce rates. Less empirical attention has been directed towards the relationship between sex ratio and mate preferences, despite the importance of mate preferences in the human mating literature. To address this gap, we examined sex ratio’s relationship to the variation in preferences for attractiveness, resources, kindness, intelligence and health in a long-term mate across 45 countries (n = 14 487). We predicted that mate preferences would vary according to relative power of choice on the mating market, with increased power derived from having relatively few competitors and numerous potential mates. We found that each sex tended to report more demanding preferences for attractiveness and resources where the opposite sex was abundant, compared to where the opposite sex was scarce.
