Abstract

In this work, we present a procedure to modify the exchange bias (EB) properties of antiferromagnetic Mn/ferromagnetic NiFe bilayers by in-situ low energy Ar + bombardment of the Mn layer during sample deposition. We present structural and magnetic results for unassisted and Ar + assisted Mn/NiFe bilayers. X-ray diffraction, transmission electron microscopy and electron diffraction results establish different preferred Mn orientation directions between the two samples as a result of the Ar + bombardment process. Hysteresis loops taken over several temperatures reveal that samples assisted with Ar + ions during the Mn layer deposition had suppressed EB properties at low temperature as compared to samples grown without Ar + assistance. Abstract

Ion beam bombardment is a useful surface modification method in tailoring the microstructure and magnetic properties of ferromagnetic thin films. In this paper, Co thin films were bombarded by Ar + ion beam with oxygen and then capped with Al (to prevent oxidation). The oxygen content was altered from 0 to 41% to investigate the ion-beam bombardment effect. An oxide layer containing Al 2 O 3 , Co 3 O 4 and CoO was formed after the ion-beam bombardment with oxygen, as characterized by x-ray photoelectron spectroscopy, high-resolution transmission electron microscopy, selected area electron diffraction, and x-ray diffraction. Enhanced coercivities (H c 's) (from 4.14 kA/m to 7.96 kA/m) and loop squareness (M r /M s. from 0.5 to 0.8) was induced by pure Ar + ion-beam bombardment that resulted in domain wall pinning due to bombardment-induced defects and Co spin reordering. H c exhibited a strong dependence on the oxygen content in the ion beam, due to the nature of the cobalt oxide formation. Field cooling to 180 K resulted in increased squareness (M r /M s. = 0.9) and reduced H c . The low-field magnetization measured during zero field and field cooling to further investigate the exchange bias magnetism identified an irreversibility temperature at 320 K in a film treated by ion-beam bombardment with an O 2 /Ar ratio of 21%. Abstract

It has been well established that both in bulk at ambient pressure and for films under modest strains, cubic SrCoO3-δ (δ < 0.2) is a ferromagnetic metal. Recent theoretical work, however, indicates that a magnetic phase transition to an antiferromagnetic structure could occur under large strain accompanied by a metalinsulator transition. We have observed a strain-induced ferromagnetic-to-antiferromagnetic phase transition in SrCoO3-δ films grown on DyScO3 substrates, which provide a large tensile epitaxial strain, as compared to ferromagnetic films under lower tensile strain on SrTiO3 substrates. Magnetometry results demonstrate the existence of antiferromagnetic spin correlations and neutron diffraction experiments provide a direct evidence for a G-type antiferromagnetic structure with Neél temperatures between TN ~ 135 ± 10 K and ~325 ± 10 K, depending on the oxygen content of the samples. Therefore, our data experimentally confirm the predicted strain-induced magnetic phase transition to an antiferromagnetic state for SrCoO3-δ thin films under large epitaxial strain. Abstract

Depth-sensitive magnetic, structural, and chemical characterization is important in the understanding and optimization of physical phenomena emerging at the interfaces of transition metal oxide heterostructures. In a simultaneous approach we have used polarized neutron and resonant x-ray reflectometry to determine the magnetic profile across atomically sharp interfaces of ferromagnetic La0.67Sr0.33MnO3/multiferroic BiFeO3 bilayers with subnanometer resolution. In particular, the x-ray resonant magnetic reflectivity measurements at the Fe and Mn resonance edges allowed us to determine the element-specific depth profile of the ferromagnetic moments in both the La0.67Sr0.33MnO3 and BiFeO3 layers. Our measurements indicate a magnetically diluted interface layer within the La0.67Sr0.33MnO3 layer, in contrast to previous observations on inversely deposited layers [P. Yu et al., Phys. Rev. Lett. 105, 027201 (2010)]. Additional resonant x-ray reflection measurements indicate a region of altered Mn and O content at the interface, with a thickness matching that of the magnetic diluted layer, as the origin of the reduction of the magnetic moment. Abstract

Monte Carlo spin simulations were coupled to a Voronoi microstructure-generator to predict the magnitude and behavior of exchange bias in a ferromagnet/antiferromagnet (AF) thin film bilayer with a nanocrystalline microstructure. Our model accounts for the effects of irregular grain-shapes, finite-sized particles, and the possible presence of local random-fields originating from the antiferromagnet's grain-boundary regions. As the grain-boundary represents a crystal-structure distortion, we model the local effect on the exchange constants in the Gaussian approximation which can cause regions resembling a spin glass confined to an unusual 2D topology. Although an ensemble of completely disconnected AF grains isolated by non-magnetic barriers provides a small exchange bias, the introduction of a spin-glass network at the boundaries causes a four-fold enhancement in the magnitude of the loop-shift. Abstract

Powder neutron diffraction and magnetometry studies have been conducted to investigate the crystallographic and magnetic structure of Bi0.8La0.2Fe0.5Mn0.5O3. The compound stabilizes in the Imma orthorhombic crystal symmetry in the measured temperature range of 5 to 380 K, with a transition to antiferromagnetic order at TN ≈ 240 K. The spin cycloid present for BiFeO3 is found to be absent with 50% Mn3+ cation substitution, leading to G-type antiferromagnetic order with an enhanced out-of-plane canted ferromagnetic component, evident from measurable weak-ferromagnetic hysteresis. Structural modifications do not solely explain this behavior, indicating that modified electron exchange interactions must be taken into account. A classical spin simulation was developed to investigate the effect of random substitution in a disordered pseudocubic perovskite. The calculations took into account the nearest-neighbor, next-nearest-neighbor, and Dzyaloshinskii-Modya interactions, along with the local spin anisotropy. Using this framework to extend the established Hamiltonian model for BiFeO3, we show that only certain types of perturbations at a magnetic defect and the surrounding molecular fields trigger a simultaneous collapse of cycloidal order and the emergence of the long-range weak-ferromagnetic component. By adopting values for the Mn molecular fields appropriate for REMnO3 (RE = rare earth), simulations of BiMn0.5Fe0.5O3 exhibit the key magnetic properties of our experimental observations. Abstract

Polarized neutron reflectometry and x-ray reflectometry were used to determine the nanoscale magnetic and chemical depth profiles of the heavy rare-earth nitrides HoN, ErN, and DyN in the form of 15- to 40-nm-thick films. The net ferromagnetic components are much lower than the predictions of density-functional theory and Hund's rules for a simple ferromagnetic ground state in these 4f ionic materials, which points to the intrinsic contribution of crystal-field effects and noncollinear spin structures. The magnetic moment per rare-earth ion was determined as a function of temperature in the range 5-100 K at fields of 1-4 T. It is demonstrated that the films are stoichiometric within 1-3% and magnetically homogeneous on the nanometer scale. [ABSTRACT FROM AUTHOR]

Abstract

Abstract: In this manuscript we describe the major components of the Platypus time-of-flight neutron reflectometer at the 20MW OPAL reactor in Sydney, Australia. Platypus is a multipurpose spectrometer for the characterisation of solid thin films, materials adsorbed at the solid–liquid interface and free-liquid surfaces. It also has the capacity to study magnetic thin films using spin-polarised neutrons. Platypus utilises a white neutron beam (λ=2–20Å) that is pulsed using boron-coated disc chopper pairs; thus providing the capacity to tailor the wavelength resolution of the pulses to suit the system under investigation. Supermirror optical components are used to focus, deflect or spin-polarise the broad bandwidth neutron beams, and typical incident spectra are presented for each configuration. A series of neutron reflectivity datasets are presented, indicating the quality and flexibility of this spectrometer. Minimum reflectivity values of <10−7 are observed; while maximum thickness values of 325nm have been measured for single-component films and 483nm for a multilayer system. Off-specular measurements have also been made to investigate in-plane features as opposed to those normal to the sample surface. We present a new approach for dynamic energy illtering at spallation neutron sources, based on the original concept of the Drabkin spin-resonance flipper. The setup takes advantage of the neutron magnetic moment, and consists of a wavelength-selective magnetic resonator and a supermirror polarizer/analyzer system. We are proposing refinements (time dependence and revised magnetic field profiles) to the basic concepts of the setup, making it suitable for time-offlight experiments at spallation neutron sources. Abstract: When completed in 2006, the Spallation Neutron Source (SNS) will use an accelerator to produce the most intense beams of pulsed neutrons in the world. This unique facility is being built by a collaboration of six US Department of Energy laboratories and will serve a diverse community of users drawn from academia, industry, and government labs. The project continues on schedule and within budget, with commissioning and installation of all systems going well. Installation of 14 state-of-the-art instruments is under way, and design work is being completed for several others. These new instruments will enable inelastic and elastic-scattering measurements across a broad range of science such as condensed-matter physics, chemistry, engineering materials, biology, and beyond. Abstract

Abstract: The Portevin–Le Châtelier effect is an interesting phenomenon that afflicts the deformation working of ductile alloys. Characteristic of this effect are the so-called PLC bands, regions of concentrated deformation that usually follow each other in a regular manner. An optical extensometer is here presented as a new measurement set-up for the investigation of these PLC bands. The optical extensometer has been employed during tensile deformation of Al–3wt.%Mg specimens and the propagation of type-B PLC bands has been detected and its velocity measured. A real-time analysis of data has been realized up to 250scans/s, it permits to locate a deformation as soon as it emerges. Abstract

Intentional introduction of chemical disorder into mono-stoichiometric epitaxial FePt3 films allows to create a ferro-/antiferromagnetic two-phase system, which shows a pronounced and controllable exchange bias effect. In contrast to conventional exchange bias systems, granular magnetic interfaces are created within the same crystallographic structure by local variation of chemical order. The amount of the exchange bias can be controlled by the relative amount and size of ferromagnetic and antiferromagnetic volume fractions and the interface between them. The tailoring of the magnetic composition alone, without affecting the chemical and structural compositions, opens the way to study granular magnetic exchange bias concepts separated from structural artifacts. Abstract

Cellular organelles provide opportunities to relate biological mechanisms to disease. Here we use affinity proteomics, genetics and cell biology to interrogate cilia: poorly understood organelles, where defects cause genetic diseases. Two hundred and seventeen tagged human ciliary proteins create a final landscape of 1,319 proteins, 4,905 interactions and 52 complexes. Reverse tagging, repetition of purifications and statistical analyses, produce a high-resolution network that reveals organelle-specific interactions and complexes not apparent in larger studies, and links vesicle transport, the cytoskeleton, signalling and ubiquitination to ciliary signalling and proteostasis. We observe sub-complexes in exocyst and intraflagellar transport complexes, which we validate biochemically, and by probing structurally predicted, disruptive, genetic variants from ciliary disease patients. The landscape suggests other genetic diseases could be ciliary including 3M syndrome. We show that 3M genes are involved in ciliogenesis, and that patient fibroblasts lack cilia. Overall, this organelle-specific targeting strategy shows considerable promise for Systems Medicine., Mutations in proteins that localize to primary cilia cause devastating diseases, yet the primary cilium is a poorly understood organelle. Here the authors use interaction proteomics to identify a network of human ciliary proteins that provides new insights into several biological processes and diseases.

247. Feeding Performance of Argentine Stem Weevil Is Reduced by Peramine from Perennial Ryegrass Infected with Endophyte Fungus.

Abstract

One of the primary supports for extensive agriculture is pasture, which can suffer severe damage from insects including the Argentine stem weevil, Listronotus bonariensis . The main control method has been the infection of ryegrass with an endophyte fungus, forming a symbiotic association that produces alkaloids. In this study, we evaluated the impact of endophyte and peramine production on the weight of L. bonariensis across seven unnamed lines (LE161-LE167), and two Lolium perenne cultivars: Jumbo and Alto AR1. L. bonariensis adults fed on leaves from LE164, LE166, and ALTO AR1 showed weight losses of 13.3%, 17.1% and 18.2%, respectively. Similarly, the corresponding alkaloidal extract from LE164, LE166, and ALTO AR1 exhibited an antifeedant effect on L. bonariensis adults in laboratory assays, as observed through weight loss or low weight gain (-12.5%, 8.8% and 4.9%, respectively). Furthermore, one alkaloid, peramine, also elicited an antifeedant effect when incorporated into an artificial diet. Liquid chromatographic analysis of the alkaloid extract revealed that peramine was present in LE164, LE166 and ALTO AR1 in amounts ranging from 46.5-184.2 ng/g. Peramine was not detected in Jumbo and the remaining experimental lines. These data suggest that L. bonariensis were susceptible to peramine produced from endophyte infection in experimental lines LE164 and LE166, as well as ALTO AR1, affecting their feeding behavior.

Abstract

Alström syndrome (ALMS) is a very rare autosomal-recessive disorder, causing a broad range of clinical defects most notably retinal degeneration, type 2 diabetes, and truncal obesity. The ALMS1 gene encodes a complex and huge ∼0.5 MDa protein, which has hampered analysis in the past. The ALMS1 protein is localized to the centrioles and the basal body of cilia and is involved in signaling processes, for example, TGF-β signaling. However, the exact molecular function of ALMS1 at the basal body remains elusive and controversial. We recently demonstrated that protein complex analysis utilizing endogenously tagged cells provides an excellent tool to investigate protein interactions of ciliary proteins. Here, CRISPR/Cas9-mediated endogenously tagged ALMS1 cells were used for affinity-based protein complex analysis. Centrosomal and microtubule-associated proteins were identified, which are potential regulators of ALMS1 function, such as the centrosomal protein 70 kDa (CEP70). Candidate proteins were further investigated in ALMS1-deficient hTERT-RPE1 cells. Loss of ALMS1 led to shortened cilia with no change in structural protein localization, for example, acetylated and ɣ-tubulin, Centrin-3, or the novel interactor CEP70. Conversely, reduction of CEP70 resulted in decreased ALMS1 at the ciliary basal body. Complex analysis of CEP70 revealed domain-specific ALMS1 interaction involving the TPR-containing C-terminal (TRP-CT) fragment of CEP70. In addition to ALMS1, several ciliary proteins, including CEP135, were found to specifically bind to the TPR-CT domain. Data are available via ProteomeXchange with the identifier PXD046401. Abstract

Biological complexity is achieved through elaborate interactions between relatively few individual components. Affinity purification (AP) has allowed these networks of protein-protein interactions that regulate key biological processes to be interrogated systematically. In order to perform these studies at the required scale, easily transfectable immortalized cell lines have typically been used. Gene-editing now affords the systematic creation of isogenic mouse models carrying endogenous tags for affinity proteomics. This may allow protein-protein interactions to be characterized in the appropriate tissue for a particular biological process or disease phenotype under physiological conditions, and for interaction landscapes to be compared across tissues. Here we demonstrate application to intraflagellar transport (IFT) proteins, which are WD40-domain-containing proteins that are essential for the formation and function of all types of cilia. We describe a method to generate mice with an endogenous C-terminal streptavidin/FLAG tag, using Ift80 as an example, and demonstrate the successful implementation of AP in this model. This method can easily be adapted for N- and C-terminal tagging of many other proteins in vivo., (© 2024. 250. Paralog-specific TTC30 regulation of Sonic hedgehog signaling.

Abstract

The intraflagellar transport (IFT) machinery is essential for cilia assembly, maintenance, and trans-localization of signaling proteins. The IFT machinery consists of two large multiprotein complexes, one of which is the IFT-B. TTC30A and TTC30B are integral components of this complex and were previously shown to have redundant functions in the context of IFT, preventing the disruption of IFT-B and, thus, having a severe ciliogenesis defect upon loss of one paralog. In this study, we re-analyzed the paralog-specific protein complexes and discovered a potential involvement of TTC30A or TTC30B in ciliary signaling. Specifically, we investigated a TTC30A-specific interaction with protein kinase A catalytic subunit α, a negative regulator of Sonic hedgehog (Shh) signaling. Defects in this ciliary signaling pathway are often correlated to synpolydactyly, which, intriguingly, is also linked to a rare TTC30 variant. For an in-depth analysis of this unique interaction and the influence on Shh, TTC30A or B single- and double-knockout hTERT-RPE1 were employed, as well as rescue cells harboring wildtype TTC30 or the corresponding mutation. We could show that mutant TTC30A inhibits the ciliary localization of Smoothened. This observed effect is independent of Patched1 but associated with a distinct phosphorylated PKA substrate accumulation upon treatment with forskolin. This rather prominent phenotype was attenuated in mutant TTC30B. Mass spectrometry analysis of wildtype versus mutated TTC30A or TTC30B uncovered differences in protein complex patterns and identified an impaired TTC30A-IFT57 interaction as the possible link leading to synpolydactyly. We could observe no impact on cilia assembly, leading to the hypothesis that a slight decrease in IFT-B binding can be compensated, but mild phenotypes, like synpolydactyly, can be induced by subtle signaling changes. Our systematic approach revealed the paralog-specific influence of TTC30A KO and mutated TTC30A on the activity of PRKACA and the uptake of Smoothened into the cilium, resulting in a downregulation of Shh.
