Cat manure (CM) possesses high level of nutrients for growing food crop. However, animal manure may contain toxic elements that may contaminate food crop. Spent coffee ground (SCG) may be used to reduce mobility of heavy metals and reduce crop uptake. In this study, SCG was composted with CM for 31 days to produce a co-compost (SCG-CM) for growing spinach (Spinacia oleracea). The growth rate of spinach was assessed until its maturity, and the metal uptake of spinach shoot was determined thereafter using inductively coupled plasma-optical emission spectroscopy (ICP-OES). The effect of soil treatment with SCG-CM on the height and elemental composition of spinach were compared with that of chicken manure compost (CMC). The prepared composts were primarily organic matter (72.9–81.4 % w/w) with the rest are ash (13.3–23.4 % w/w) and moisture (1.2–2.6 % w/w). Zinc content in SCG-CM (1261 ± 0.1 mg/kg) is significantly higher than that of soil and CMC (p < 0.05) and has exceeded the maximum permissible limit set by European Union Standard (2002) and the Malaysian Compost Quality Standard and Guidelines (2000). Matured spinach reached maximum plant height after 33 days. The amendment of SCG-CM significantly increased the height of spinach (32 ± 6 cm) compared to that of CMC (13 ± 1 cm) (p < 0.05). However, contents of Zn, Cu, Pb and Cd were not increased for spinach grown in the SCG-CM-amended soil, and the level of those elements are below permissible limit set by the Malaysian Food Act 1983 and Food Regulations 1985. This study shows that SCG-CM is effective in improving yield without causing accumulation of toxic trace elements in spinach.

Background. Mesenchymal stem cells (MSCs) express growth factors and other cytokines that stimulate repair and control the immune response. MSCs are also immunoprivileged with low risk of rejection. Umbilical cord-derived MSCs (UCMSCs) are particularly attractive as an off-the-shelf allogeneic treatment in emergency medical conditions. We aim to determine the safety and efficacy of intravenous allogeneic infusion of UCMSCs (CLV-100) by Cytopeutics® (Selangor, Malaysia) in healthy volunteers, and to determine the effective dose at which an immunomodulatory effect is observed. Methodology. Umbilical cord samples were collected after delivery of full-term, healthy babies with written consent from both parents. All 3 generations (newborn, parents, and grandparents) were screened for genetic mutations, infections, cancers, and other inherited diseases. Samples were transferred to a certified Good Manufacturing Practice laboratory for processing. Subjects were infused with either low dose (LD, 65 million cells) or high dose (HD, 130 million cells) of CLV-100 and followed up for 6 months. We measured cytokines using ELISA including anti-inflammatory cytokines interleukin 1 receptor antagonist (IL-1RA), interleukin 10 (IL-10), pro-/anti-inflammatory cytokine interleukin 6 (IL-6), and the proinflammatory cytokine tumor necrosis factor-alpha (TNF-α). Results. 11 healthy subjects (LD, n=5; HD, n=6; mean age of 55±13 years) were recruited. All subjects tolerated the CLV-100 infusion well with no adverse reaction throughout the study especially in vital parameters and routine blood tests. At 6 months, the HD group had significantly higher levels of anti-inflammatory markers IL1-RA (705±160 vs. 306±36 pg/mL; p=0.02) and IL-10 (321±27 vs. 251±28 pg/mL; p=0.02); and lower levels of proinflammatory marker TNF-α (74±23 vs. 115±15 pg/mL; p=0.04) compared to LD group. Conclusion. Allogeneic UCMSCs CLV-100 infusion is safe and well-tolerated in low and high doses. Anti-inflammatory effect is observed with a high-dose infusion.

Background: Currently, drug-eluting balloon (DEB) appears to be an attractive alternative option for the treatment of in-stent restenosis (ISR). Nevertheless, the clinical outcomes of DEB have seldom been compared to those of new-generation drug-eluting stent (DES). Thus, this meta-analysis aimed to evaluate the safety and efficacy of DEB compared to those of new-generation DES in the treatment of ISR. Methods: A comprehensive search of electronic databases including PubMed, EMBASE, and Cochrane Library up to November 2, 2017 was performed to identify pertinent articles comparing DEB to new-generation DES for the treatment of ISR. In addition, conference proceedings for the scientific sessions of the American College of Cardiology, American Heart Association, European Society of Cardiology, Transcatheter Cardiovascular Therapeutics, and EuroPCR were also searched. The primary endpoint was target lesion revascularization (TLR) at the longest follow-up. Dichotomous variables were presented as risk ratios (RR s) with 95% confidence intervals (CI s), while the overall RR s were estimated using the Mantel-Haenszel random-effects model. Results: Five randomized controlled trials (RCTs) and eight observational studies involving 2743 patients were included in the present meta-analysis. Overall, DEB was comparable to new-generation DES in terms of TLR (RR = 1.24, 95% CI: 0.89–1.72, P = 0.21), cardiac death (RR = 1.55, 95% CI: 0.89–2.71, P = 0.12), major adverse cardiovascular event (RR = 1.21, 95% CI: 0.98–1.48, P = 0.07), myocardial infarction (RR = 1.12, 95% CI: 0.72–1.76, P = 0.62), and stent thrombosis (RR = 0.95, 95% CI: 0.38–2.42, P = 0.92). However, DEB was associated with higher risk of all-cause mortality than new-generation DES (RR = 1.65, 95% CI: 1.09–2.50, P = 0.02). This was especially true in the real-world observational studies (RR = 1.79, 95% CI: 1.12–2.88, P = 0.02). In RCTs, however, no significant difference was found between the two treatment strategies in the risk of all-cause mortality. Conclusions: The current meta-analysis showed that DEB and new-generation DES had comparable safety and efficacy for the treatment of ISR in RCTs. However, treatment with DEB was associated with higher risk of all-cause mortality in the real-world nonrandomized studies.

Objective: To determine the involvement and the modulatory effects of IL-33 during Plasmodium berghei ANKA (PbA) infection. Methods: PbA infection in male ICR mice was utilized as a model of malaria. Systemically circulating IL-33 levels were determined in blood plasma by enzyme-linked immunosorbent assay (ELISA). After 24 hours post-inoculation of PbA, recombinant IL-33 and ST2, and antibodies against IL-33 and IgG treatments were administered daily for 3 days. Tissue expression and localization of IL-33 were assessed in organs generally affected by malaria via immunohistochemistry. Moreover, histopathological examination was performed to assess the effects of the treatments. Results: The levels of systemic IL-33 were elevated at the critical phase of PbA infection. Likewise, immunohistochemical analysis revealed a significant upregulation of IL-33 expression at the critical phase in the brain, lungs, and spleen of PbA-infected mice as compared to healthy controls. Treatment with IL-33 protected against experimental cerebral malaria development and reduced pathological features in the brain and lungs of the PbA-infected mice. Conclusions: A potential critical role and involvement of IL-33 in PbA infection may hint at the resolution of immunopathological sequelae associated with malaria. [ABSTRACT FROM AUTHOR]

Mesoporous manganese dioxide (mMnO2) was first synthesized for the loading of methotrexate (MTX), and then dopamine was in situ polymerized on the surface of the MTX‐loaded mMnO2 (mMnO2‐MTX) in an alkaline solution to encapsulate the drug in the mesopores of mMnO2. Both low pH and glutathione (GSH) can result in the degradation of mMnO2 and poly(dopamine) (PDA), and thus the delivery of MTX from the mMnO2‐MTX‐PDA can be triggered by low pH and GSH. Near‐infrared (NIR) light‐responsive delivery of MTX can be achieved owing to the outstanding photothermal conversion capability of PDA; on the other hand, the mMnO2‐MTX‐PDA can be utilized for photothermal therapy under the irradiation of NIR light due to the elevated temperature. The results of cytotoxicity test demonstrate that the pH, GSH, and NIR light tri‐responsive drug‐controlled delivery system has excellent biocompatibility, while exhibits pronounced growth inhibition against murine breast tumor cell line 4T1.
