Abstract Objective Prostate cancer (PC) is a significant disease affecting men’s health worldwide. More than 60% of patients over 65 years old and more than 80% are diagnosed with localized PC. The current choice of treatment modalities for localized PC and whether overtreatment is controversial. Therefore, we wanted to construct a nomogram to predict the risk factors associated with cancer-specific survival (CSS) and overall survival (OS) in elderly patients with localized PC while assessing the survival differences in surgery and radiotherapy for elderly patients with localized PC. Methods Data of patients with localized PC over 65 years were obtained from the Surveillance, Epidemiology, and End Results (SEER) database. Univariate and multivariate Cox regression models were used to determine independent risk factors for CSS and OS. Nomograms predicting CSS and OS were built using multivariate Cox regression models. The consistency index (C-index), the area under the subject operating characteristic curve (AUC), and the calibration curve were used to test the accuracy and discrimination of the prediction model. Decision curve analysis (DCA) was used to test the potential clinical value of this model. Results A total of 90,434 patients over 65 years and diagnosed with localized PC from 2010 to 2018 were included in the study. All patients were randomly assigned to the training set (n = 63,328) and the validation set (n = 27,106). Univariate and multivariate Cox regression model analysis showed that age, race, marriage, T stage, surgical, radiotherapy, prostate-specific antigen (PSA), and Gleason score (GS) were independent risk factors for predicting CSS in elderly patients with localized PC. Age, race, marriage, surgery, radiotherapy, PSA, and GS were independent risk factors for predicting OS in elderly patients with localized PC. The c-index of the training and validation sets for the predicted CSS is 0.802(95%CI:0.788–0.816) and 0.798(95%CI:0.776–0.820, respectively). The c-index of the training and validation sets for predicting OS is 0.712(95%:0.704–0.720) and 0.724(95%:0.714–0.734). It shows that the nomograms have excellent discriminatory ability. The AUC and the calibration curves also show good accuracy and discriminability. Conclusion We have developed new nomograms to predict CSS and OS in elderly patients with localized PC. After internal validation and external temporal validation with reasonable accuracy, reliability and potential clinical value, the model can be used for clinically assisted decision-making.

In this study, effects of four single factors which including slice thickness, frying temperature, frying time and deoilling time on the quality of persimmon chips under low-temperature vacuum frying were analyzed. According to the Box-Behnken test principle, the processing technology of persimmon chips under low-temperature vacuum frying was further optimized by taking crispness and chromatic aberration ΔE of persimmon chips as response values. Meanwhile, the physicochemical composition, microbiological index and microstructure of the final product were further researched. The results showed that all of frying temperature, frying time, deoiling time and slice thickness affected the crispness and chromatic aberration of persimmon chips, the optimal technology of vacuum frying persimmon chips at low temperature was determined by response surface optimization experiment and then actual correction were as follows: Deoiling time of 5 min, frying time of 50 min, frying temperature of 80 ℃ and thickness 5.5 mm. Under these conditions, the crispness, ΔE, water content, oil content, titrable acid and VC content of persimmon chips respective was 858.97 g, 17.28, 2.60%, 22.53%, 0.34 g/L and 31.20 mg/100 g. The persimmon chips were golden in color and complete in shape, and the inside of the chips presented a honeycomb-like porous and loose structure, the product had a crisp taste, and the microbial indicators met the health requirements. This research could provide technical support for the development of low-temperature vacuum frying persimmon chips.

A multiband microstrip-fed wideband (WB) antenna with filtering notches is described for $3 \mathrm {G} / 4 \mathrm {G} / 5 \mathrm {G}$ applications. The proposed antenna comprises three notch bands by etching a modified inverted U-slot, a square ring slot, and an interdigital inductor slot on the patch element and feedline. The antenna resonates at $1.9,2.3$ , and $3.5 \mathrm {~GHz}$ due to the resonant components’ mutual interaction, which eliminates interference at other frequencies. The antenna’s measured $S_{11}$ are $18.79 \mathrm {~dB}$ at $1.9 \mathrm {~GHz},-24.8 \mathrm {~dB}$ at $2.4 \mathrm {~GHz}$ , and $-40.6 \mathrm {~dB}$ at $3.5 \mathrm {~GHz}$ , showing a multiband function with a band-reject level of $0.4 \mathrm {~dB}$ . The VSWR is less than two at all resonant frequencies. The effects of altering the notch dimensions on the $S_{11}$ and VSWR were explored. The antenna was developed and tested using a Rogers RT/Duroid 5880 substrate. The agreement between measured and simulated results was satisfactory. The $S_{11}$ result was validated using the ADS schematic and machine learning techniques. The proposed triband-notched antenna offers encouraging results, with radiation patterns exhibiting omnidirectional characteristics and effective performance within the required frequencies. Current distribution analysis reveals how notches disrupt surface current and lower radiation at specific frequencies. The antenna’s gain and efficiency performed satisfactorily in the stated frequency ranges.

Development in industrial companies aims to digitize processes, ensure traceability and advance automation. Systems with automated guided vehicles (AGVs) can help meet these objectives. A core element of these systems is the higher-level system controller. When an AGV system is operated, some dynamic aspects appear. It is then to identify, that the previous specified rules for scheduling and routing of vehicles are not always proper and practical, which may result in waiting time of AGVs. That means the static performance requirements and the specifications based on them often lead to suboptimal results. A methodical problem-solving approach is to develop a digital twin as a reflection of reality and to carry out strategy analyses using simulation. For this purpose, a digital twin of the concerned AGV system with five vehicles and its technological environment is created using software of plant simulation. Various simulation scenarios are developed to simulate material flows by using different routing and scheduling rules and strategies. Drawing from the insights gained through the digital twin and simulation analyses, this study identifies novel scheduling and routing rules for the AGV system. These rules improve the overall efficiency and effectiveness of the system operations by easing traffic congestion, reducing transit time, and minimizing production downtime.

Modern industrial systems demand intricate connectivity and automation, especially in the realm of shop floor processes and intralogistics. Automated Guided Vehicle (AGV) systems are characterized by their potential for seamlessly networking value creation areas. However, failures and disruptions in AGV systems and adjacent facilities can lead to production halts, adversely affecting delivery reliability and quality. A substantial portion of the downtime stems from manual troubleshooting, underscoring the pivotal importance of the response time from maintenance staff. This paper introduces an approach employing a neural network with long short-term memory for forecasting and predictive maintenance to enhance AGV system reliability and availability in production environments. By analysing historical data, identifying patterns, and predicting potential failures or maintenance needs in AGV components and neighbouring facilities, the proposed AI-based forecasting ensures timely preventive measures. A case study shows the effectiveness of this approach in significantly improving AGV system performance, minimizing disruptions, and enhancing operational availability. This research contributes to smart manufacturing by providing a practical solution for optimizing availability of the concerned AGV system through advanced AI-based forecasting strategies.

ObjectiveTo analyze the clinical and epidemiological characteristics of confirmed cases of human monkeypox infection in Changning District， Shanghai， and to explore their clinical and epidemiological characteristics.MethodsClinical data from 10 reported cases of monkeypox in individuals residing in Changning District or identified by local medical institutions between July 20 and September 30， 2023， were collected. Epidemiological case investigations were conducted， and throat swabs， anal swabs， and rash swabs were collected by the treating medical institutions. Real-time fluorescence quantitative PCR was used for monkeypox virus nucleic acid testing， and descriptive epidemiological analysis was applied to analyze the epidemiological characteristics of the cases.ResultsAll 10 confirmed cases of human monkeypox infection were all young males with an average age of 35.4 years， all of whom belonged to the men who have sex with men （MSM） population， with no occupational clustering. The primary clinical symptoms included fever， rash， enlarged inguinal lymph nodes， and muscle soreness. Nine cases presented with a rash， and seven cases experienced fever symptoms. Among the 10 cases， one experienced fever， rash， enlarged lymph nodes， and muscle soreness； two had fever， rash， and enlarged lymph nodes； two had fever， rash， and systemic soreness； two had only a rash； one had fever or rash； and one was asymptomatic. Among the nine cases with a rash， the rash was mainly localized to the genital or anal area， with fewer cases presenting rashes on the limbs or trunk simultaneously. All cases reported a history of non-exclusive MSM behavior within 21 days before the onset of the disease. The interval between the last suspected high-risk exposure and the onset of symptoms was 4 to 10 days， with an average interval of 6.9 days. The time from the onset of fever to the appearance of a rash was 0 to 5 days， with an average of 1.87 days.ConclusionThe main clinical manifestations of human infection with monkeypox are fever， rash， and enlarged inguinal lymph nodes. The MSM population is a high-risk group for monkeypox infection， and its source of infection may be associated with MSM exposure. Early-stage symptoms are mild， leading to potential underdiagnosis. Additionally， patients may conceal information during the investigation process， which increases the difficulty of epidemic prevention and control.

Traditional assessment indexes could not fully describe offshore wind resources, for the meteorological properties of offshore are more complex than onshore. As a result, the uncertainty of offshore wind power projects would be increased and final economic benefits would be affected. Therefore, a study on offshore wind resource assessment is carried out, including three processes of “studying data sources, conducting multi-dimensional indexes system and proposing an offshore wind resource assessment method based on analytic hierarchy process (AHP)”. First, measured wind data and two kinds of reanalysis data are used to analyze the characteristics and reliability of data sources. Second, indexes such as effective wind speed occurrence, affluent level occurrence, coefficient of variation, neutral state occurrence have been proposed to depict availability, richness, and stability of offshore wind resources, respectively. Combined with existing parameters (wind power density, dominant wind direction occurrence, water depth, distance to coast), a multi-dimensional indexes system has been built and on this basis, an offshore wind energy potential assessment method has been proposed. Furthermore, the proposed method is verified by the annual energy production of five offshore wind turbines and practical operating data of four offshore wind farms in China. This study also compares the ranking results of the AHP model to two multi-criteria decision making (MCDM) models including weighted aggregated sum product assessment (WASPAS) and multi-attribute ideal real comparative analysis (MAIRCA). Results show the proposed method gains well in practical engineering applications, where the economic score values have been considered based on the offshore reasonable utilization hours of the whole life cycle in China.

SignificanceCrop production is related to national food security, economic development and social stability, so timely information on the growth of major crops is of great significance for strengthening the crop production management and ensuring food security. The traditional crop growth monitoring mainly judges the growth of crops by manually observing the shape, color and other appearance characteristics of crops through the external industry, which has better reliability and authenticity, but it will consume a lot of manpower, is inefficient and difficult to carry out monitoring of a large area. With the development of space technology, satellite remote sensing technology provides an opportunity for large area crop growth monitoring. However, the acquisition of optical remote sensing data is often limited by the weather during the peak crop growth season when rain and heat coincide. Synthetic aperture radar (SAR) compensates well for the shortcomings of optical remote sensing, and has a wide demand and great potential for application in crop growth monitoring. However, the current research on crop growth monitoring using SAR data is still relatively small and lacks systematic sorting and summarization. In this paper, the research progress of SAR inversion of crop growth parameters were summarized through comprehensive analysis of existing literature, clarify the main technical methods and application of SAR monitoring of crop growth, and explore the existing problems and look forward to its future research direction.Progress]The current research status of SAR crop growth monitoring were reviewed, the application of SAR technology had gone through several development stages: from the early single-polarization, single-band stage, gradually evolving to the mid-term multi-polarization, multi-band stage, and then to the stage of joint application of tight polarization and optical remote sensing. Then, the research progress and milestone achievements of crop growth monitoring based on SAR data were summarized in three aspects, namely, crop growth SAR remote sensing monitoring indexes, crop growth SAR remote sensing monitoring data and crop growth SAR remote sensing monitoring methods. First, the key parameters of crop growth were summarized, and the crop growth monitoring indexes were divided into morphological indicators, physiological and biochemical indicators, yield indicators and stress indicators. Secondly, the core principle of SAR monitoring of crop growth parameters was introduced, which was based on the interaction between SAR signals and vegetation, and then the specific scattering model and inversion algorithm were used to estimate the crop growth parameters. Then, a detailed summary and analysis of the radar indicators mainly applied to crop growth monitoring were also presented. Finally, SAR remote sensing methods for crop growth monitoring, including mechanistic modeling, empirical modeling, semi-empirical modeling, direct monitoring, and assimilation monitoring of crop growth models, were described, and their applicability and applications in growth monitoring were analyzed.Conclusions and ProspectsFour challenges exist in SAR crop growth monitoring are proposed: 1) Compared with the methods of crop growth monitoring using optical remote sensing data, the methods of crop growth monitoring using SAR data are obviously relatively small. The reason may be that SAR remote sensing itself has some inherent shortcomings; 2) Insufficient mining of microwave scattering characteristics, at present, a large number of studies have applied the backward scattering intensity and polarization characteristics to crop growth monitoring, but few have applied the phase information to crop growth monitoring, especially the application study of polarization decomposition parameters to growth monitoring. The research on the application of polarization decomposition parameter to crop growth monitoring is still to be deepened; 3) Compared with the optical vegetation index, the radar vegetation index applied to crop growth monitoring is relatively less; 4 ) Crop growth monitoring based on SAR scattered intensity is mainly based on an empirical model, which is difficult to be extended to different regions and types of crops, and the existence of this limitation prevents the SAR scattering intensity-based technology from effectively realizing its potential in crop growth monitoring. Finally, future research should focus on mining microwave scattering features, utilizing SAR polarization decomposition parameters, developing and optimizing radar vegetation indices, and deepening scattering models for crop growth monitoring.

Seeds are the most basic and important means of production for agriculture. During the production and processing of seeds, they may undergo potential mechanical damages and mildew alterations, which might jeopardize their germination viability. Hence, checking the quality of seeds before sowing is of paramount importance for the benefit of the sower and the safety of agricultural production. In order to achieve an efficient detection of maize seed quality, our experiment assembled a dataset composed of 2,128 seeds with four different health statuses of maize: healthy, broken, moth-eaten, and mildewed. In this paper, we proposed a lightweight maize seed quality detection model for small objects based on improved YOLOv8: I-YOLOv8. Firstly, we introduced a multi-scale attention mechanism called EMA to efficiently retain information across channels and reduce computational load. Next, we chosen the SPD-Conv module for low-resolution images and small objects, and applied it to the backbone, which addressed the loss of fine-grained information and the less efficient learning of feature representations present in YOLOv8. Lastly, we reduced the large detection layer, which directed the network to pay more attention to the location, channel, and dimensional information of smaller objects, and we also replaced the loss function with WIoUv3. We validated our model using ablation studies and compared it with YOLOv5, YOLOv6, and YOLOv8. The mAP (Mean Average Precision) of the improved model I_YOLOv8 reaches 98.5%, which is 6.7% higher than YOLOv8. The average recognition time per image was 163.9fps, a boost of 5.2fps compared to YOLOv8. This study lays a theoretical foundation for the efficient, convenient, and rapid detection of maize quality, while also offering a technical basis for advancing automated maize quality detection means.

Abstract Background In HIV-1 infection, more than 95% of CD4+T cells die of caspase-1 mediated pyroptosis. What governs the increased susceptibility of CD4+T cells to pyroptosis is poorly understood. Methods Blood samples were obtained from 31 untreated HIV-infected patients (UNT), 29 antiretroviral therapy treated HIV-infected patients (ART), and 21 healthy control donors (HD). Plasma levels of IL-18 and IL-1β, caspase-1 expression, mitochondrial mass (MM) and mitochondrial fusion/fisson genes of CD4+T subsets were measured. Results A significantly higher IL-18 level in plasma and MM level of CD4+T cells were found in HIV-infected patients (UNT and ART) compared to HD, and the MMhigh phenotype was manifested, related to increased caspase-1 expression. Moreover, the increased MM was more pronounced in the early differentiated and inactivated CD4+T cells. However, higher MM was not intrinsically linked to T cell differentiation disorder or excessive activation of the CD4+T cells. Mechanistically, the increased MM was significantly correlated with an elevated level of expression of the mitochondrial fusion gene mitofusin1. Conclusion An increase in MM was associated with heightened sensitivity of CD4+T cells to pyroptosis, even in early differentiated and inactivated CD4+T cells, in patients with HIV-1 infection, regardless of whether patients were on antiretroviral therapy or not. These new revelations have uncovered a previously unappreciated challenge to immune reconstitution with antiretroviral therapy.

Abstract Background Lynch syndrome (LS) is the most common hereditary colorectal cancer (CRC) syndrome. This condition is characterized by germline variants in DNA mismatch repair (MMR) genes, including MLH1, MSH2, MSH6, and PMS2. In this study, we analyzed the molecular defects and clinical manifestations of two families affected with CRC and proposed appropriate individual preventive strategies for all carriers of the variant. Methods We recruited two families diagnosed with CRC and combined their family history and immunohistochemical results to analyze the variants of probands and those of other family members by using whole exome sequencing. Subsequently, gene variants in each family were screened by comparing them with the variants available in the public database. Sanger sequencing was performed to verify the variant sites. An online platform ( https://www.uniprot.org ) was used to analyze the functional domains of mutant proteins. Results A novel frameshift variant (NM_001281492, c.1129_1130del, p.R377fs) in MSH6 and a known deleterious variant (NM_000249.4:c.1731G > A, p.S577S) in MLH1 were identified in the two families with CRC. Using bioinformatics tools, we noted that the frameshift variant reduced the number of amino acids in the MSH6 protein from 1230 to 383, thereby leading to no MSH6 protein expression. The silent variant caused splicing defects and was strongly associated with LS. 5-Fluorouracil-based adjuvant chemotherapy is not recommended for patients with LS. Conclusions The novel frameshift variant (MSH6, c.1129_1130del, p.R377fs) is likely pathogenic to LS, and the variant (MLH1, c.1731G > A, p.S577S) has been further confirmed to be pathogenic to LS. Our findings underscore the significance of genetic testing for LS and recommend that genetic consultation and regular follow-ups be conducted to guide individualized treatment for cancer-afflicted families, especially those with a deficiency in MMR expression.

Abstract Background Decitabine has been widely used to treat acute myeloid leukemia (AML); however as AML is a heterogeneous disease, not all patients benefit from decitabine. This study aimed to identify markers for predicting the response to decitabine. Methods An intersection of in vitro experiments and bioinformatics was performed using a combination of epigenetic and transcriptomic analysis. A tumor-suppressor gene associated with methylation and the response to decitabine was screened. Then the sensitivity and specificity of this marker in predicting the response to decitabine was confirmed in 54 samples from newly diagnosed AML patients treated with decitabine plus IA regimen in a clinical trial (ChiCTR2000037928). Results In vitro experiments showed that decitabine caused hypomethylation and upregulation of BTG1, while downregulation of BTG1 attenuated the inhibitory effect of decitabine. In newly diagnosed AML patients who received decitabine plus IA regimen, the predictive value of BTG1 to predict complete remission (CR) was assigned with a sensitivity of 86.7% and a specificity of 100.0% when BTG1 expression was

Abstract Drug-eluting stent implantation suppresses the excessive proliferation of smooth muscle cells to reduce in-stent restenosis. However, the efficacy of drug-eluting stents remains limited due to delayed reendothelialization, impaired intimal remodeling, and potentially increased late restenosis. Here, we show that a drug-free coating formulation functionalized with tailored recombinant humanized type III collagen exerts one-produces-multi effects in response to injured tissue following stent implantation. We demonstrate that the one-produces-multi coating possesses anticoagulation, anti-inflammatory, and intimal hyperplasia suppression properties. We perform transcriptome analysis to indicate that the drug-free coating favors the endothelialization process and induces the conversion of smooth muscle cells to a contractile phenotype. We find that compared to drug-eluting stents, our drug-free stent reduces in-stent restenosis in rabbit and porcine models and improves vascular neointimal healing in a rabbit model. Collectively, the one-produces-multi drug-free system represents a promising strategy for the next-generation of stents.

Abstract Atopic dermatitis (AD) is a chronic skin condition with intense pruritus, eczema, and dry skin. The recurrent intense pruritus and numerous complications in patients with AD can profoundly affect their quality of life. Obesity is one of its comorbidities that has been confirmed to be the hazard factor of AD and also worsen its severity. Nevertheless, the specific mechanisms that explain the connection between obesity and AD remain incompletely recognized. Recent studies have built hopes on various adipokines to explain this connection. Adipokines, which are disturbed by an obese state, may lead to immune system imbalances in people with AD and promote the development of the disease. This review focuses on the abnormal expression patterns of adipokines in patients with AD and their potential regulatory molecular mechanisms associated with AD. The connection between AD and obesity is elucidated through the involvement of adipokines. This conduces to the in-depth exploration of AD pathogenesis and provides a new perspective to develop therapeutic targets.

Abstract Objective Traditional observational research has suggested a connection between socioeconomic position, mental health, and sleep apnea (SA), but the specifics of this connection are still unclear. Using the Mendelian randomization approach, we intended to evaluate the potential causal link between mental health, socioeconomic status, and SA. Methods Our research employed summary statistics data from large-scale genome-wide association studies (GWAS) on mental health, socioeconomic status, and SA. In the main study, the connection between mental health, socioeconomic status, and SA was examined using the inverse variance weighted approach. In addition, as a supplement, we also used other Mendelian randomization methods, including MR Egger, weighted median, simple mode, and weighted mode. Results The primary analysis showed that educational attainment, including longer years of schooling, college or university degree, and higher intelligence was associated with a lower risk of SA (OR = 0.750, 95%CI = 0.653–0.862; OR = 0.558, 95%CI = 0.423–0.735; OR = 0.871, 95%CI = 0.760–0.999, respectively), while social deprivation was associated with a higher risk of SA (OR = 1.821, 95%CI = 1.075–3.085). And the income was not associated with the risk of sleep apnea (OR = 0.877, 95%CI = 0.682–1.129). In mental health exposure, major depressive disorder was associated with a higher risk of sleep apnea (OR = 1.196, 95%CI = 1.015–1.409), while attention-deficit hyperactivity disorder, bipolar disorder, and schizophrenia were not associated with the risk of sleep apnea (OR = 1.064, 95%CI = 0.958–1.181; OR = 1.030, 95%CI = 0.942–1.127; OR = 0.990, 95%CI = 0.957–1.025, respectively). Reverse MR analysis failed to find a causal effect from SA on mental health and socioeconomic status. Conclusions This MR investigation offers proof of a possible causal relationship between SA, socioeconomic level, and mental health.

Abstract Flexible information memory is the key component of flexible electronic devices and the core of intelligent wearable devices. In this paper, Ge/Sb multilayer phase change films of various thickness ratios were prepared using polyether ether ketone as substrate, and their flexible phase change properties and device conversion characteristics were studied. After bending for 100000 times and bending experiments with different bending radius, the film can still realize the transition from amorphous to crystalline states, and the resistance fluctuation was small. Bending, stretching and pressing of the film resulted in grain refinement and increasing of crystalline resistance. The flexible electronic devices using Ge/Sb multilayer films were prepared. The phase change memory device can realize reversible conversion between SET and RESET states with different pulse widths in flat, bent states and after bending many times. All findings show that Ge/Sb multilayer films on PEEK substrate have broad application prospects in high-performance flexible memory in the future.

Carotenoids are one of the important coloring substances in the ray florets of Zinnia elegans. Carotenoid cleavage dioxygenase (CCD) directly affects carotenoid contents by cleaving the double bond of carotenoids, thereby affecting the flower color. However, few reports have elucidated its function in Zinnia elegans. In the present study, through transcriptome homology searches, three ZeCCDs, ZeCCD1, ZeCCD4-1, and ZeCCD4-2 were screened from Zinnia elegans. The function of ZeCCDs was verified by bacterial pigment complementation system, and it revealed that all of the three ZeCCDs had the ability to cleave β-carotene and other carotenoids (ε-carotene, zeaxanthin and lycopene). Meanwhile, the expression levels of ZeCCD1 and ZeCCD4-2 in the petals of different cultivars were significantly negatively correlated with the carotenoid content. Furthermore, compared with the cytoplasmic localization of ZeCCD1, ZeCCD4-2 is localized in the plastid. Our results indicate that ZeCCD4-2 is the key gene responsible for the differential accumulation of carotenoids in the petals of different Zinnia elegans cultivars. This study lays a foundation for further research on the molecular mechanism of petal coloration and carotenoid metabolism of Zinnia elegans.

Triggering receptor expressed on myeloid cells (TREM)-1 is a transmembrane receptor embedded in the intrinsic immune cell membrane that amplifies inflammation by increasing inflammatory signalling triggered by toll-like receptors as well as transcript levels in the NF-κB signalling pathway. The soluble form of TREM-1 is obtained by cleavage of the extracellular portion of TREM-1 by matrix metalloprotease-9 and can be used as a biomarker for the diagnosis and prognosis of TREM-1 pathway activation in many acute and chronic inflammatory diseases. The present review aims to summarise the latest research and insights on TREM-1 in inflammatory and non-inflammatory diseases.

Objective To observe the effects of astragaloside （Ast）-incubated human adipose derived stem cell （hADSC） on diabetic kidney disease （DM） after caudal vein transplantation. examine the repair status of diabetic kidney disease （DKD） on renal injury in rats and elucidate its possible mechanism. Methods A total of 21 Sprague-Dawley rats were randomized into four groups of normal （n = 5）, model （n = 6）, hADSC （n = 5） and Ast-hADSC （n = 5）. DKD model was established in the last 3 groups. Both hADSC and Ast-hADSC groups were injected 6 times with hADSC or Ast-hADSC suspension through tail vein once biweekly. The model group received the same volume of normal saline. Renal function, blood lipid, urinary microalbumin/urinary creatinine and urinary protein quantification were detected. Renal pathology was observed after hematoxylin-eosin and Masson stains and renal colonization of hADSC by confocal laser. Immunohistochemistry and immunoblot were utilized for observing the voltage-dependent anion channel （VDAC） and microtubule-associated protein-light-chain-3. LC3Ⅱ/LC3Ⅰ, autophagy substrate P62 protein, phosphatase and tensin homology induced putative kinase 1 （Pink1） and E3 ubiquitin ligase （Parkin） protein expression. Mitochondria and autophagosomes were observed by transmission electron microscopy. Results （1） As compared with model group, total serum cholesterol [（2.65 ± 0.04） mmol/L, （2.30 ± 0.29） mmol/L vs （3.31 ± 0.91） mmol/L], ratio of urinary microalbumin/urinary creatinine [（58.91 ± 11.86） mg, （44.00 ± 23.82） mg vs （101.03 ± 46.88） mg] and 24 h urinary protein quantity [（321.18 ± 88.71） mg, （136.35 ± 66.01） mg vs （408.67 ± 113.95） mg] （P＜0.05） declined in hADSC and Ast-hADSC groups. Body weight gained [（240.20 ± 26.43） g, （274.00±17.89） g vs （193.60 ± 18.57） g] （P＜0.05）. The effects of body weight and 24 h urinary protein were more significant in Ast-hADSC group than those in hADSC group （P＜0.05）. （2） As compared with model group, no significant differences existed in triglyceride, low density lipoprotein, urea nitrogen or high density lipoprotein in hADSC and Ast-hADSC groups （P＞0.05）. （3） Both hADSC and Ast-hADSC groups had lesser renal injury while renal index and glomerular cross-sectional area spiked （P＜0.05）; Improvement was more significant in Ast-hADSC group. （4） Under laser confocal microscope, minimal red fluorescence was observed in hADSC and Ast-hADSC groups. （5） Immunohistochemistry and immunoblotting revealed that protein expressions of LC3, LC3Ⅱ/LC3Ⅰ, Pink1 and Parkin spiked in hADSC and Ast-hADSC groups （P＜0.05） while protein expression of P62 declined （P＜0.05）. The effect of Ast-hADSCs group was more significant. （6） In model group, mitochondria were swollen, deformed and fragmented and the number of autophagosomes was fewer. Both hADSC and Ast-hADSC groups showed more autophagosomes and fewer swollen deformed mitochondria and mitochondrial fragments. The effect of Ast-hADSC group was more significant. Conclusions hADSC may restore mitochondrial autophagy in kidney tissue of DKD rats and reduce kidney injury. The effect is more significant after Ast incubation.

An efficient and practical method for the synthesis of C5-brominated 8-aminoquinoline amides via a copper-promoted selective bromination of 8-aminoquinoline amides with alkyl bromides was developed. The reaction proceeds smoothly in dimethyl sulfoxide (DMSO) under air, employing activated and unactivated alkyl bromides as the halogenation reagents without additional external oxidants. This method features outstanding site selectivity, broad substrate scope, and excellent yields.

Homomorphic encryption (HE) enables third-party servers to perform computations on encrypted user data while preserving privacy. Although conceptually attractive, the speed of software implementations of HE is almost impractical. To address this challenge, various domain-specific architectures have been proposed to accelerate homomorphic evaluation, but efficiency remains a bottleneck. In this paper, we propose a homomorphic evaluation accelerator with heterogeneous reconfigurable modular computing units (RCUs) for the Brakerski/Fan-Vercauteren (BFV) scheme. RCUs leverage operator abstraction to efficiently perform basic sub-operations of homomorphic evaluation such as residue number system (RNS) conversion, number theoretic transform (NTT), and other modular computations. By combining these sub-operations, complex homomorphic evaluation operations like multiplication, rotation, and addition are efficiently executed. To address the high demand for data access and improve memory efficiency, we design a coordinate-based address encoding strategy that enables in-place and conflict-free data access. Furthermore, specific optimizations are performed on the core sub-operations such as NTT and automorphism. The proposed architecture is implemented on Xilinx Virtex-7 and UltraScale+ FPGA platforms and evaluated for polynomials of length 4096. Compared to state-of-the-art accelerators with the same parameter set, our accelerator achieves the following advantages: $1)\,\,2.04\times $ to $3.33\times $ reduction in the area-time product (ATP) for the key sub-operation NTT, $2)\,\,1.08\times $ to $7.42\times $ reduction in latency for homomorphic multiplication with higher area efficiency, and 3) support for a wider range of homomorphic evaluation operations, including rotation, compared to other BFV-based accelerators.

Abstract Background Glioblastoma (GBM) is the most aggressive malignant central nervous system tumor with a poor prognosis.The malignant transformation of glioma cells via epithelial-mesenchymal transition (EMT) has been observed as a main obstacle for glioblastoma treatment. Epithelial membrane protein 3 (EMP3) is significantly associated with the malignancy of GBM and the prognosis of patients. Therefore, exploring the possible mechanisms by which EMP3 promotes the growth of GBM has important implications for the treatment of GBM. Methods We performed enrichment and correlation analysis in 5 single-cell RNA sequencing datasets. Differential expression of EMP3 in gliomas, Kaplan–Meier survival curves, diagnostic accuracy and prognostic prediction were analyzed by bioinformatics in the China Glioma Genome Atlas (CGGA) database and The Cancer Genome Atlas (TCGA) database. EMP3-silenced U87 and U251 cell lines were obtained by transient transfection with siRNA. The effect of EMP3 on glioblastoma proliferation was examined using the CCK-8 assay. Transwell migration assay and wound healing assay were used to assess the effect of EMP3 on glioblastoma migration. Finally, quantitative real-time polymerase chain reaction (qRT-PCR) and western blot were used to detect the mRNA and protein expression levels of EMT-related transcription factors and mesenchymal markers. Results EMP3 is a EMT associated gene in multiple types of malignant cancer and in high-grade glioblastoma. EMP3 is enriched in high-grade gliomas and isocitrate dehydrogenase (IDH) wild-type gliomas.EMP3 can be used as a specific biomarker for diagnosing glioma patients. It is also an independent prognostic factor for glioma patients' overall survival (OS). In addition, silencing EMP3 reduces the proliferation and migration of glioblastoma cells. Mechanistically, EMP3 enhances the malignant potential of tumor cells by promoting EMT. Conclusion EMP3 promotes the proliferation and migration of GBM cells, and the mechanism may be related to EMP3 promoting the EMT process in GBM; EMP3 may be an independent prognostic factor in GBM.

Abstract Background Chemoradiotherapy is a critical treatment for patients with locally advanced and unresectable non-small cell lung cancer (NSCLC), and it is essential to identify high-risk patients as early as possible owing to the high incidence of radiation pneumonitis (RP). Increasing attention is being paid to the effects of endogenous factors for RP. This study aimed to investigate the value of computed tomography (CT)-based radiomics combined with genomics in analyzing the risk of grade ≥ 2 RP in unresectable stage III NSCLC. Methods In this retrospective multi-center observational study, 100 patients with unresectable stage III NSCLC who were treated with chemoradiotherapy were analyzed. Radiomics features of the entire lung were extracted from pre-radiotherapy CT images. The least absolute shrinkage and selection operator algorithm was used for optimal feature selection to calculate the Rad-score for predicting grade ≥ 2 RP. Genomic DNA was extracted from formalin-fixed paraffin-embedded pretreatment biopsy tissues. Univariate and multivariate logistic regression analyses were performed to identify predictors of RP for model development. The area under the receiver operating characteristic curve was used to evaluate the predictive capacity of the model. Statistical comparisons of the area under the curve values between different models were performed using the DeLong test. Calibration and decision curves were used to demonstrate discriminatory and clinical benefit ratios, respectively. Results The Rad-score was constructed from nine radiomic features to predict grade ≥ 2 RP. Multivariate analysis demonstrated that histology, Rad-score, and XRCC1 (rs25487) allele mutation were independent high-risk factors correlated with RP. The area under the curve of the integrated model combining clinical factors, radiomics, and genomics was significantly higher than that of any single model (0.827 versus 0.594, 0.738, or 0.641). Calibration and decision curve analyses confirmed the satisfactory clinical feasibility and utility of the nomogram. Conclusion Histology, Rad-score, and XRCC1 (rs25487) allele mutation could predict grade ≥ 2 RP in patients with locally advanced unresectable NSCLC after chemoradiotherapy, and the integrated model combining clinical factors, radiomics, and genomics demonstrated the best predictive efficacy.

Abstract Background Multiple distraction indicators have been applied to measure street-crossing distraction but their validities in predicting pedestrian safety are poorly understood. Methods Based on a video-based observational study, we compared the validity of four commonly used distraction indicators (total duration of distraction while crossing a street, proportion of distracted time over total street-crossing time, duration of the longest distraction time, and total number of distractions) in predicting three pedestrian safety outcomes (near-crash incidence, frequency of looking left and right, and speed crossing the street) across three types of distraction (mobile phone use, talking to other pedestrians, eating/drinking/smoking). Change in Harrell’s C statistic was calculated to assess the validity of each distraction indicator based on multivariable regression models including only covariates and including both covariates and the distraction indicator. Results Heterogeneous capacities in predicting the three safety outcomes across the four distraction indicators were observed: 1) duration of the longest distraction time was most predictive for the occurrence of near-crashes and looks left and right among pedestrians with all three types of distraction combined and talking with other pedestrians (Harrell’s C statistic changes ranged from 0.0310 to 0.0335, P

Abstract Background The bimolecular fluorescence complementation (BiFC) assay is commonly used for investigating protein–protein interactions. While several BiFC detection systems have been developed, there is a limited amount of research focused on using laser scanning confocal microscope (LSCM) techniques to observe protoplasts. Protoplasts are more susceptible to damage and instability compared to their original cell state due to the preparation treatments they undergo, which makes it challenging for researchers to manipulate them during observation under LSCMs. Therefore, it is crucial to utilize microscope techniques properly and efficiently in BiFC assays. Results When the target fluorescence is weak, the autofluorescence of chloroplast particles in protoplasts can interfere with the detection of BiFC signals localized in the nuclear region. Spectrum analysis revealed that chloroplast autofluorescence can be excited by lasers of various types, with the highest fluorescence signal observed at around 660 nm. Furthermore, our investigation into the impact of different pipette tips on the integrity of protoplast samples indicated that the utilization of cut tips with larger openings can mitigate cell breakage. We presented a workflow of LSCM techniques for investigating protoplast BiFC and discussed the microscopic manipulation involved in sample preparation and image capturing. Conclusion When the BiFC signals are weak, they may be affected by chloroplast autofluorescence. However, when used properly, the autofluorescence of chloroplasts can serve as an excellent internal marker for effectively distinguishing other signals. In combination with other findings, this study can provide valuable reference for researchers conducting BiFC assays and related studies.

Abstract With the emergence of intelligent manufacturing, new-generation information technologies such as big data and artificial intelligence are rapidly integrating with the manufacturing industry. One of the primary applications is to assist manufacturing plants in predicting product quality. Traditional predictive models primarily focus on establishing high-precision classification or regression models, with less emphasis on imbalanced data. This is a specific but common scenario in practical industrial environments concerning quality prediction. A SMOTE-XGboost quality prediction active control method based on joint optimization hyperparameters is proposed to address the problem of imbalanced data classification in product quality prediction. In addition, edge computing technology is introduced to address issues in industrial manufacturing, such as the large bandwidth load and resource limitations associated with traditional cloud computing models. Finally, the practicality and effectiveness of the proposed method are validated through a case study of the brake disc production line. Experimental results indicate that the proposed method outperforms other classification methods in brake disc quality prediction.

Abstract Background Neoadjuvant immunochemotherapy can effectively downstage tumors and reduce the risk of postoperative recurrence and distant metastasis in patients with non‐small cell lung cancer (NSCLC). In this study, we investigated the correlation between inflammatory biomarkers and tertiary lymphoid structure (TLS) expression. We also compared the predictive values of these inflammatory parameters, TLSs, and a combination of inflammatory parameters and TLSs for neoadjuvant efficacy in patients with NSCLC. Methods We retrospectively analyzed the clinical information of 106 patients with NSCLC who underwent neoadjuvant immunochemotherapy and radical surgery at Shandong Cancer Hospital between June 2020 and June 2022. Results TLS was evaluated using hematoxylin–eosin staining and immunohistochemically‐stained tissue sections. Logistic analysis was performed to determine the correlation between inflammatory parameters, TLSs, and the factors affecting major pathological response (MPR). Receiver operating characteristic curves and the C‐index were used to evaluate the predictive value of the nomogram models for MPR. The systemic immune‐inflammatory index (SII) was an independent predictor of high TLS abundance and maturity. Platelet‐to‐lymphocyte ratio (PLR) ≤201.8, TLS abundance, and TLS maturity were independent predictors of MPR. The PLR‐TLS combined model performed better in assessing the MPR in patients with NSCLC than models using single indicators. Conclusion Our study demonstrated that the SII is an independent predictor of both TLS abundance and maturity. Both TLSs and PLR can predict MPR rates in patients with NSCLC receiving neoadjuvant immunochemotherapy. However, assessing the MPR in patients with NSCLC using a combination of PLR and TLSs is more accurate than using either indicator alone.

Cooperative driving is crucial for improving traffic efficiency and safety for connected and automated vehicles (CAVs), especially in traffic bottlenecks. However, most of the state-of-the-art cooperative driving strategies neglect the issue of fairness. Fairness is essential to properly allocate road resources and improve the travel experience. In this paper, we focus on the fairness concerns in the on-ramp cooperative driving problem. First, we note that enhancing traffic efficiency usually leads to unfairness, but we propose solutions to balance both aspects. Using the fundamental relation in traffic flow theory, we illustrate the existence of the trade-off at congested on-ramps. We then make some modifications to the cooperative driving strategies to incorporate fairness considerations. Simulation results show that the modified strategies achieve trade-offs in agreement with the theoretical one, laying the foundation for implementing the trade-off in real-world scenarios. These findings are enlightening for the increasing research on fairness issues in cooperative driving, and contribute to the optimization of traffic management strategies.

Early corrosion behavior of 35CrMo low-alloy steel for high-strength bolt in the simulated shallow seawater and deep-sea environment was studied using mass loss measurement, electrochemical impedance spectroscopy (EIS), scanning electron microscope (SEM), energy dispersive spectrometer (EDS), Raman spectrometer, X-ray photoelectron spectrometer (XPS) and 3D microscope. The results show that the corrosion of 35CrMo steel initiates mainly at the steel matrix around the inclusions of CaO–MgO–Al2O3, (Ca, Mn)S and their mixed composite. The corrosion rate in the simulated deep-sea environment is lower than that in the shallow seawater, which is declined with immersion time. The corrosion products formed in the two environments are composed of γ-FeOOH, α-FeOOH, Fe3O4 and little Cr(III) oxide or/and hydroxide, but the rust layer produced in the deep-sea environment is thin and uniform. The steel has pits in a shallow dish shape and tends to develop uniform corrosion in the deep-sea environment.

Boron neutron capture therapy(BNCT), as a highly promising radiotherapy modality, has emerged as a significant treatment option for various malignant tumors, including high-grade gliomas, recurrent head and neck cancers, and melanoma. BNCT is characterized by the nuclear reactions that occur when nonradioactive 10B is irradiated by low energy thermal neutron to yield high linear energy transfer(LET) particles, such as 4He and 7Li. These particles have high relative biological efficacy and a short path length of approximately the diameter of a single cell(5-9 μm), which enables selective damage to tumour cells marked with 10B and preserves the adjacent normal tissue. Compared to classical radiotherapies, BNCT has garnered growing attention within the medical community due to its precision targeting, potent biological effects, and short course of treatment. The therapeutic efficacy of BNCT is based on accurately controlling radiation dose, which is mainly determined by the local 10B concentration of tumour and the neutron flux. Currently, BNCT is being implemented in clinical trials. Dynamic and quantitative monitoring of 10B concentration of boron-containing drugs in intratumor is the key element to realize the “efficiency and side-effect reduction” of BNCT. Consequently, there is an urgent need for the development of more accurate measurement techniques. In this review, we provide a concise overview of the principles underlying BNCT, and summarize various monitoring methods for boron-containing drugs during treatment, including physical, nuclear, chemical, and novel techniques utilizing advanced molecular imaging technologies such as positron emission tomography, magnetic resonance imaging, and optical imaging. Furthermore, we analyze the strengths and limitations of these approaches. We hope this review provides informative insights into the future development of the precise and efficient BNCT.

Abstract Background Intraductal papillary mucinous neoplasm (IPMN) is a cystic tumor of the pancreas arising from abnormal papillary proliferation of ductal epithelial cells, and is a precancerous lesion of pancreatic malignancy. This study aimed to evaluate associations between acute pancreatitis (AP) and histologic subtypes of IPMN. Methods In the clinical study, patients with IPMN confirmed by surgical resection specimens at our institute between 2009 and 2021 were eligible for inclusion. Associations and predictive accuracy of AP on the presence of HGD were determined by logistic regressions. In addition, a systematic review and meta-analysis was conducted through literatures upon search in PubMed, Embase, CENTRAL, China National Knowledge Infrastructure (CKNI), and Wanfang database, up to June, 2023. Pooled effects of the associations between AP and HGD and intestinal epithelial subtype subtype, shown as odds ratios (ORs) with 95% confidence intervals (CIs), were calculated using random effects model. Results The retrospective cohort study included 47 patients (32 males, 15 females) diagnosed with IPMN at our center between 2009 and 2021, including 11 cases with AP (median 62 years) and 36 cases (median 64.5 years) without. Accuracy, sensitivity, specificity, positive predictive value (PPV), and negative predictive value (NPV) of AP in predicting HGD were 78.7%, 57.1%, 82.5%, 36.4%, and 91.7%, respectively. Univariate logistic regression analysis showed that AP group had greater odds of presence of HGD (OR: 6.29,95% CI: 1.14–34.57) than non-AP group. Meta-analysis of five case-control studies in the literature included 930 patients and showed that AP-IPMN patients had higher odds for HGD (OR: 2.13, 95% CI 1.38–3.29) and intestinal epithelial subtype (OR: 5.38, 95% CI: 3.50–8.27) compared to non-AP IPMN. Conclusions AP is predictive of malignancy in patients with IPMN.

Abstract Background The scarcity of pluripotent stem cells poses a major challenge to the clinical application, given ethical and biosafety considerations. While germline stem cells commit to gamete differentiation throughout life, studies demonstrated the spontaneous acquisition of pluripotency by spermatogonial stem cells (SSCs) from neonatal testes at a low frequency (1 in 1.5 × 107). Notably, this process occurs without exogenous oncogenes or chemical supplementation. However, while knockout of the p53 gene accelerates the transformation of SSCs, it also increases risk and hampers their clinical use. Results We report a transformation system that efficiently and stably convert SSCs into pluripotent stem cells around 10 passages with the morphology similar to that of epiblast stem cells, which convert to embryonic stem (ES) cell-like colonies after change with ES medium. Epidermal growth factor (EGF), leukemia inhibitory factor (LIF) and fresh mouse embryonic fibroblast feeder (MEF) are essential for transformation, and addition of 2i (CHIR99021 and PD0325901) further enhanced the pluripotency. Transcriptome analysis revealed that EGF activated the RAS signaling pathway and inhibited p38 to initiate transformation, and synergically cooperated with LIF to promote the transformation. Conclusion This system established an efficient and safe resource of pluripotent cells from autologous germline, and provide new avenues for regenerative medicine and animal cloning.

Abstract Sodium‐ion batteries (SIBs) present great appeal in various energy storage systems, especifically for stationary grid storage, on account of the abundance of sources and low cost. Unfortunately, the commercialization of SIBs is mainly limited by available electrode materials, especially for the cathodes. Prussian blue analogs (PBAs), emerge as a promising alternative for their structural feasibility in the application of SIBs. Decreasing the defects (vacancies and coordinated water) is an effective strategy to achieve superior electrochemical performance during the synthetic processes. Herein, we summarize crystal structures, synthetic methods, electrochemical mechanisms, and the influences of synthesis conditions of PBAs in detail. This comprehensive overview on the current research progresses of PBAs will give guides and directions to solve the existing problems for their application in SIBs.

Abstract Background This study was conducted to develop and validate an individualized prediction model for spontaneous preterm birth (sPTB) in twin pregnancies. Methods This a retrospective cohort study included 3845 patients who gave birth at the Chongqing Maternal and Child Health Hospital from January 2017 to December 2022. Both univariable and multivariable logistic regression analyses were performed to find factors associated with sPTB. The associations were estimated using the odds ratio (OR) and the 95% confidence interval (CI). Model performance was estimated using sensitivity, specificity, accuracy, area under the receiver operating characteristic curve (AUC) and decision curve analysis (DCA). Results A total of 1313 and 564 cases were included in the training and testing sets, respectively. In the training set, univariate and multivariate logistic regression analysis indicated that age ≥ 35 years (OR, 2.28; 95% CI 1.67–3.13), pre-pregnancy underweight (OR, 2.36; 95% CI 1.60–3.47), pre-pregnancy overweight (OR, 1.67; 95% CI 1.09–2.56), and obesity (OR, 10.45; 95% CI, 3.91–27.87), nulliparity (OR, 0.58; 95% CI 0.41–0.82), pre-pregnancy diabetes (OR, 5.81; 95% CI 3.24–10.39), pre-pregnancy hypertension (OR, 2.79; 95% CI 1.44–5.41), and cervical incompetence (OR, 5.12; 95% CI 3.08–8.48) are independent risk factors for sPTB in twin pregnancies. The AUC of the training and validation set was 0.71 (95% CI 0.68–0.74) and 0.68 (95% CI 0.64–0.73), respectively. And then we integrated those risk factors to construct the nomogram. Conclusions The nomogram developed for predicting the risk of sPTB in pregnant women with twins demonstrated good performance. The prediction nomogram serves as a practical tool by including all necessary predictors that are readily accessible to practitioners.

Abstract To investigate the relationship of the middle cerebral artery (MCA) bifurcation aneurysms with patients’ age and sex, vascular angles at the bifurcation, and diameters of the M1 and two M2 arteries, patients with and without MCA aneurysms were retrospectively enrolled. The lateral angles, MCA bifurcation angle and arterial diameter were measured and analyzed. Totally, 121 (19.0%) patients with and 517 (81.0%) without MCA aneurysms were enrolled. Most (n = 88 or 72.7%) aneurysms were present in the age range of 40–70 years, and significantly (P = 0.01) more women than men had the bifurcation aneurysms. Abstract Cervical cancer is a leading cause of gynecological cancer death in the world. Human papillomavirus (HPV) is the most causative factor of cervical cancer. In addition, many genetic factors are involved in cervical cancer development. Most studies focus on cervical samples to do research work about cervical cancer and precancerous lesions, but no sensitive or specific biomarkers were found. High-throughput genomic technologies are able to capture information from tumors and precancerous lesions in blood, thus providing a new way for the early diagnosis of cervical precancer and cervical cancer. Blood is an ideal specimen for detecting cancer biomarkers because it contains a lot of information, such as circulating tumor cells and circulating tumor DNA (ctDNA). This article reviews the clinical use and challenges of blood ctDNA testing in patients with cervical precancer and cervical cancer.

Abstract The importance of uncommon/rare oncogenic drivers in non‐small cell lung cancer (NSCLC) was underscored during the 20th China Lung Cancer Summit. These drivers, while present in a significant proportion of NSCLC patients, remain a challenge for diagnosis and therapeutic targeting. In the never‐smokers/low smokers category with mutations such as EGFR and HER2, the efficacy of immune checkpoint inhibitors (ICIs) remains suboptimal, attributed to lower PD‐L1 expression and tumor mutation burden (TMB). However, heavy smokers, often with mutations like KRAS, may derive benefits from ICIs, as supported by trials like CheckMate‐057. With the complex landscape of these drivers and their clinical implications, the summit culminated in six pivotal consensus points, aiming to guide future research and clinical decisions. Despite the advancements, the detection, interpretation, and therapeutic strategies involving these drivers necessitate further exploration and standardization.

Glioma is characterized by rapid cell proliferation, aggressive invasion, altered apoptosis and a poor prognosis. β-Sitosterol, a kind of phytosterol, has been shown to possess anticancer activities. Our current study aims to investigate the effects of β-sitosterol on gliomas and reveal the underlying mechanisms. Our results show that β-sitosterol effectively inhibits the growth of U87 cells by inhibiting proliferation and inducing G2/M phase arrest and apoptosis. In addition, β-sitosterol inhibits migration by downregulating markers of epithelial-mesenchymal transition (EMT). Mechanistically, network pharmacology and transcriptomics approaches illustrate that the EGFR/MAPK signaling pathway may be responsible for the inhibitory effect of β-sitosterol on glioma. Afterward, the results show that β-sitosterol effectively suppresses the EGFR/MAPK signaling pathway. Moreover, β-sitosterol significantly inhibits tumor growth in a U87 xenograft nude mouse model. β-Sitosterol inhibits U87 cell proliferation and migration and induces apoptosis and cell cycle arrest in U87 cells by blocking the EGFR/MAPK signaling pathway. These results suggest that β-sitosterol may be a promising therapeutic agent for the treatment of glioma.

Abstract In the context of electricity market, the heavy load of some inter‐provincial and intra‐provincial lines limits available transfer capability (ATC). As a series compensation device, static synchronous series compensator (SSSC) can reallocate the power flow of adjacent lines. In order to maximize the inter‐regional ATC, a two‐stage optimal SSSC allocation method considering the uncertainty and correlation of multi‐wind farm is proposed. Firstly, the initial wind power scenarios are generated based on latin hypercube sampling (LHS) and reduced by the improved iterative self‐organizing data analysis techniques algorithm (I‐ISODATA). Then, a multi‐objective optimization model is constructed with the maximum expected power flow transfer entropy (PFTE) and minimum expected SSSC investment cost in all typical wind power scenarios, and non‐dominated sorting genetic algorithm‐II (NSGA‐II) method is used to determine the location of SSSC. In addition, based on the reduced wind power scenarios, the ATC calculation model containing SSSC is constructed, and the modern interior point method is used to determine the control parameters and configuration capacity of SSSC. Finally, based on the improved IEEE39 and IEEE118 node systems, case study proves that the proposed model can determine the optimal SSSC configuration scheme and significantly improves the ATC.

Metasomatic zircons in ultramafic rocks are crucial in tracking orogenic processes. Metasomatic zircons are conceived to form by local zircon saturation resulting from effective Zr transport during metamorphic events and incomplete Zr accommodation in mantle minerals. However, the accommodation capacity of Zr in mantle minerals remains unknown, hampering our understanding of zircon genesis in ultramafic rocks. Here we performed experiments at 2.0−6.0 GPa and 1050−1200 °C and determined Zr content (solubility) in the mantle minerals at zircon saturation (ZCZS). The results show that ZCZS values for the main mantle minerals are olivine (ol, 3.68 ± 0.27 ppm)

Coexisting or mixed type of colorectal tumors has been rarely reported. This study was designed to investigate clinicopathological characteristics of co-existing or mixed colorectal adenocarcinoma and highly differentiated neuroendocrine tumor (NET-G1). To do that, clinicopathological characteristics of five cases of co-existing or mixed colorectal adenocarcinoma and NET-G1 admitted to our institution between 2017 and 2021 were retrospectively analyzed and literature review was conducted. Four patients were male and one female, aged 62–75 years old. Among them, four cases were diagnosed with rectal cancer and one case of colon cancer. Gross examination found that one patient was diagnosed with multiple colon polyps including three malignant polyps, and the remaining four cases of ulcerous masses. The tumors infiltrated into the muscle layer in two cases, and three cases with tumors infiltrating into surrounding adipose tissues. Microscopic examination revealed one patient developed poorly differentiated adenocarcinoma and four cases of moderately differentiated adenocarcinoma. Four patients had adenocarcinoma and NET-G1 in colon, and one case of adenocarcinoma in colon and NET-G1 in appendix. To conclude, co-existing or mixed colorectal tumors are extremely rare in clinical settings. Clinicopathological characteristics of five cases of co-existing or mixed adenocarcinoma and NET-G1 are diverse and adenocarcinoma is more aggressive in most affected patients.

Abstract How aging affects cells of the human brain active milieu remains largely unknown. Here, we analyze astrocytes and neurons in the neocortical tissue of younger (22–50 years) and older (51–72 years) adults. Aging decreases the amount of reduced mitochondrial cytochromes in astrocytes but not neurons. The protein-to-lipid ratio decreases in astrocytes and increases in neurons. Aged astrocytes show morphological atrophy quantified by the decreased length of branches, decreased volume fraction of leaflets, and shrinkage of the anatomical domain. Atrophy correlates with the loss of gap junction coupling between astrocytes and increased input resistance. Aging is accompanied by the upregulation of glial fibrillary acidic protein (GFAP) and downregulation of membrane-cytoskeleton linker ezrin associated with leaflets. No significant changes in neuronal excitability or spontaneous inhibitory postsynaptic signaling is observed. Thus, brain aging is associated with the impaired morphological presence and mitochondrial malfunction of cortical astrocytes, but not neurons.

Abstract Mazdutide is a once-weekly glucagon-like peptide-1 (GLP-1) and glucagon receptor dual agonist. We evaluated the efficacy and safety of 24-week treatment of mazdutide up to 6 mg in Chinese overweight adults or adults with obesity, as an interim analysis of a randomised, two-part (low doses up to 6 mg and high dose of 9 mg), double-blind, placebo-controlled phase 2 trial (ClinicalTrials.gov, NCT04904913). Overweight adults (body-mass index [BMI] ≥24 kg/m2) accompanied by hyperphagia and/or at least one obesity-related comorbidity or adults with obesity (BMI ≥ 28 kg/m2) were randomly assigned (3:1:3:1:3:1) to once-weekly mazdutide 3 mg, 4.5 mg, 6 mg or matching placebo at 20 hospitals in China. The primary endpoint was the percentage change from baseline to week 24 in body weight. A total of 248 participants were randomised to mazdutide 3 mg (n = 62), 4.5 mg (n = 63), 6 mg (n = 61) or placebo (n = 62). Abstract The signal in the receiver is mainly a combination of different modulation types due to the complex electromagnetic environment, which makes the modulation recognition of the mixed signal a hot topic in recent years. In response to the poor adaptability of existing mixed signals recognition methods, this paper proposes a new recognition method for mixed signals based on cyclic spectrum projection and deep neural network. Firstly, through theoretical derivation, we prove the feasibility of using cyclic spectrum for mixed communication signal identification. Then, we adopt grayscale projections on the two-dimensional cyclic spectrum as identifying representation. And a new nonlinear piecewise mapping and directed pseudo-clustering method are used to enhance the above-mentioned grayscale images, which reduces the impact of energy ratios and symbol rates on signal identification. Finally, we use deep neural networks to extract deep abstract modulation information to achieve effective recognition of mixed signals. Simulation results show that the proposed method is robust against noise. When signal-to-noise ratio is not less than 0 dB, the average recognition rate is greater than 95%. Furthermore, this method exhibits good robustness towards the changes in signal symbol rates and energy ratios between mixed signals.

Abstract Pushing layered cathode to higher operating voltage can facilitate the realization of high-energy lithium-ion batteries. However, the released oxygen species initiate materials surface upon highly delithiated states will react severely with electrolyte, accelerating the structure deterioration and triggering the thermal degradation. Here we propose an inert phase of La2Mo2O9 with abundant oxygen vacancies (about 41%) by regulating the annealing temperature to engineer the cathode interface beyond conventional modifications. By employing LiNi0.8Co0.1Mn0.1O2 as a model system and extending to higher voltage-operated LiCoO2 and Li-rich cathode, we demonstrate that the introduced lanthanum and molybdenum ions will transfer electrons to enhance the surface oxygen electronegativities, thus served as “oxygen anchor” to alleviate oxygen evolution. Furthermore, the possible released oxygen can be operando captured and reserved by β-phase La2Mo2O9 depositor for the intrinsic high oxygen vacancy formation energy. The reaction involving oxygen species with electrolyte is fundamentally diminished, thus effectively mitigate the structure deterioration and elevate the electrochemical performances, enabling a 1.5-Ah pouch-type full cell to exhibit negligible 6.0% capacity loss after 400 cycles.

Abstract Background Fatigue is a common symptom of long COVID syndrome. Compared to male survivors, females have a higher incidence of post-COVID fatigue. Therefore, long-term follow-up is necessary to understand which groups of females are more vulnerable to post-COVID fatigue. Methods This is a nested case–control study of female COVID-19 survivors who were discharged from two designated hospitals in Wuhan, China in 2020, and received 2-year follow-up from March 1 to April 6, 2022. All patients completed the Checklist Individual Strength-subscale subjective fatigue (CIS-fatigue), a chronic obstructive pulmonary disease (COPD) assessment test (CAT), and the Hospital Anxiety and Depression Scale (HADS; including the HADS-Anxiety [HADS-A] and the HADS-Depression [HADS-D]). Individuals with CIS-fatigue scores of 27 or higher were classified as cases. The risk factors for fatigue was analysed with multivariable logistic regression analysis. Results A total of 899 female COVID-19 survivors were enrolled for analysis, including 47 cases and 852 controls. Compared with controls, cases had higher CAT, HADS-A and HADS-D scores, and showed a higher prevalence of symptoms, including anxiety (cases vs. controls, 44.7% vs. 4.0%, p
