Pyroptosis is a novel proinflammatory programmed cell death process. This study was designed to investigate the functional mechanisms of long noncoding RNA growth arrest‐specific transcript 5 (lncRNA GAS5) on lipopolysaccharide (LPS)‐induced human bronchial epithelial cell (HBEC) pyroptosis. LPS was used to induce pyroptosis in HBECs, followed by the detection of the expression of GAS5, forkhead box O3 (FOXO3), and nuclear factor E2‐related factor 2/heme oxygenase 1 (Nrf2/HO‐1) signaling pathway‐related factors. Cell viability was evaluated using CCK‐8 assay, lactate dehydrogenase (LDH) release was assessed by LDH assay kit and caspase‐1 activity by flow cytometry. Furthermore, expression of NOD‐like receptor family pyrin domain containing 3 and pyroptosis‐related proteins was evaluated using Western blot analysis, while enzyme‐linked immunosorbent assay was used to determine the levels of inflammatory factors. The interaction between GAS5 and FOXO3 was confirmed using bioinformatic prediction, RNA immunoprecipitation assay, RNA pull‐down, and dual‐luciferase reporter gene assay. Treatment of HBECs with LPS upregulated the expression of GAS5 and FOXO3, resulting in the inactivation of the Nrf2/HO‐1 signaling pathway. On the other hand, inhibition of both GAS5 and FOXO3 promoted cell viability, reduced LDH release, pyroptosis, and inflammatory response in LPS‐induced HBECs. Furthermore, FOXO3 could interact with GAS5, while FOXO3 overexpression reversed the inhibitory effect of GAS5 knockdown on cell pyroptosis. Thus, mechanistically, inhibition of FOXO3 activates the Nrf2/HO‐1 pathway to suppress LPS‐induced pyroptosis in HBECs. This study revealed that GAS5 knockdown attenuates FOXO3 expression thereby activating the Nrf2/HO‐1 pathway to inhibit LPS‐induced pyroptosis in HBECs. Foot process effacement is an important feature of early diabetic nephropathy (DN), which is closely related to the development of albuminuria. Under certain nephrotic conditions, the integrity and function of the glomerular slit diaphragm (SD) structure were impaired and replaced by the tight junction (TJ) structure, resulting in so-called SD-TJ transition, which could partially explain the effacement of foot processes at the molecular level. However, the mechanism underlying the SD-TJ transition has not been described in DN. Here, we demonstrated that impaired autophagic flux blocked p62-mediated degradation of ZO-1 (TJ protein) and promoted podocytes injury via activation of caspase3 and caspase8. Interestingly, the expression of VDR in podocytes was decreased under diabetes conditions, which impaired autophagic flux through downregulating Atg3. Of note, we also found that VDR abundance was negatively associated with impaired autophagic flux and SD-TJ transition in the glomeruli from human renal biopsy samples with DN. Furthermore, VDR activation improved autophagic flux and attenuated SD-TJ transition in the glomeruli of diabetic animal models. In conclusion, our data provided the novel insight that VDR/Atg3 axis deficiency resulted in SD-TJ transition and foot processes effacement via blocking the p62-mediated autophagy pathway in DN. The SmFLS gene was cloned from eggplant and has an ORF of 1014 bp encoding 337 amino acids. The deduced protein sequence of SmFLS was 88.07% and 84.94% identical to homologs encoded by StFLS in Solanum tuberosum and SlFLS in S. lycopersicum, respectively. SmFLS contains typical DIOX_N and 2OG-Fe (II)_Oxy functional domains, as well as five strictly conserved amino acid residues (H223, D225, H279, R289, and S291) related to FLS enzyme activity. Phylogenetic tree analysis showed that SmFLS had the closest genetic relationship with the FLS genes in potato and tomato. At a high temperature of 35 °C, the expression level of SmFLS was higher than that of the control in the same period, and it reached extremely significant levels on 15DAF and 20DAF, at which the eggplant peel color became lighter accordingly. Upon overexpression of SmFLS in eggplant, the flavonol content of transformed plants was significantly higher than that of untransformed plants, and the peel color was lighter than that of the control. Introduction and objectives: Laparoscopic splenectomy and esophagogastric devascularization (LSED) are minimally invasive, effective, and safe in treating esophageal-fundic variceal bleeding with portal hypertension (PHT). The study aimed to assess the learning curve of LSED by cumulative summation (CUSUM) analysis. The 10-year follow-up data for LSED and open surgery were also examined. Patients and methods: Five hundred and ninety-four patients were retrospectively analyzed. Operation time, intraoperative blood loss, open operation conversion, and postoperative complications were selected as the evaluation indicators of surgical ability. The learning curve of LESD was assessed by the CUSUM approach. Patient features, perioperative indices, and 10-year follow-up data were examined. Results: Totally 236 patients underwent open surgery, and 358 underwent LSED. Patient characteristics were similar between groups. The LSED patients experienced less intraoperative blood loss, fewer complications, and faster recovery compared to the open surgery cohort. The learning curve of LESD was maximal for a case number of 50. Preoperative general characteristics were comparable for both stages. But the skilled stage had decreased operation time, reduced blood loss, less postoperative complications, and better recovery compared to the learning stage. The LSED group had higher recurrent hemorrhage-free survival rate and increased overall survival in comparison with cases administered open surgery in the 10-year follow-up. Free-liver cancer rates were similar between two groups. Conclusions: About 50 cases are needed to master the LSED procedure. Compared to open surgery, LSED is a safer, feasible, and safe procedure for PHT patients, correlating with decreased rebleeding rate and better overall survival. [ABSTRACT FROM AUTHOR]

Owing to their high capacity and low working potential, Si-based anodes are regarded as potential alternatives to graphite anodes to meet the higher requirements of Li-ion batteries (LIBs). However, high volume change causes the fracturing and pulverization of the bulk anode and continuous side reactions, which are more severe in large-particle Si anodes, limiting its practical application. Herein, to build a low-cost battery system, we chose a common industrial waste product, Al–Si microparticles (Al–SiMPs, ∼30 μm), as the anode for LIBs and coupled it with a 2.0 M LiFP6 2-MeTHF electrolyte to support its operation. The Al–SiMP anode showed a high specific capacity and a significantly improved electronic conductivity, ensuring high energy and power densities. An ultra-high initial coulombic efficiency (iCE) of 91.6% and a cycling CE of ∼99.9% were obtained in the half-cells, which delivered a capacity of 1300 mA h g−1 and maintained 95.3% after 100 cycles. Paired with low-cost and high-safety LiFePO4 as the cathode, the LFP‖‖Al–SiMP full cells showed decent cycling stability and exhibited a considerable cost advantage, demonstrating a competitive solution for stationary energy storage. • Estimation of actual evapotranspiration (ET) in forest vegetation using inverse simulation of soil parameters. • Eddy covariance measurements over grass were well-correlated to inverse ET values. • Four-year mean cumulative growing season ET estimates are reported for four vegetation species. • Annual cumulative ET in descending order: aspen > deep rooted conifer > shallow rooted conifer > sage > grass. Evapotranspiration (ET) is a major component of water balance and is crucial for understanding the hydrological processes, however due to highly heterogeneous distributions of soils and plants, accurate quantification of ET remains a major challenge in montane ecosystems. In this study, water use for four common vegetation communities was numerically modeled using Hydrus-1D over four continuous growing seasons (2009 to 2012), supported by a network of soil water content sensor measurements. Plant species included i) aspen (Populus tremuloides) with understory dominated by grass/forb (rudbeckia occidentalis, Bromus carinatus and Elymus trachycaulu), ii) conifer (Picea engelmannii and Abies lasiocarpa), iii) grass (dominated by B. carinatus and E. trachycaulu, et al.), and iv) sage (Artemisia tridentata). The numerical model simulated water transport within the soil and water loss from soil evaporation and plant transpiration processes. Simulations were compared with temporal and spatial dynamics of soil water content, measured at 0.1 m, 0.25 m and 0.5 m within each of 36 subplots at 30-minute intervals. Numerically simulated ET in the sage/grass meadow was compared with ET determined from a centrally-located eddy covariance tower. The simulations effectively predicted soil moisture and ET of the montane plant communities during summer dry down. Results indicate vegetation type showed no significant difference (<5 %) in ET comparing four growing seasons, except where abnormally wet conditions occurred. Four-year mean cumulative growing season ET estimates for aspen, deep-rooted conifer, shallow rooted conifer, sage, and grass were 43.0, 40.2, 28.7, 28.8 and 26.1 cm, respectively.
