Three new β-triketone flavanone hybrids, cajuputones A-C were obtained from Melaleuca cajuputi (the Australian 'tea tree'). The structures of cajuputones A-C were elucidated by 1D/2D NMR spectroscopy and HR-ESI-MS analyses; and their absolute configurations were established by electric circular dichroism (ECD) calculations using TDDFT method. Structurally, cajuputones A-C feature a rare 6/6/6/6 oxatetracyclic ring system fused between an acylphloroglucinol-derived β-triketone and a pinocembrin or strobopinin moiety via an angle-type pyran-like motif. DFT-based conformational optimization in chloroform explained the similarity of the 1D NMR data of cajuputones B and C (C-2 epimers).

Onychomycosis is a disease that affects many adults, whose treatment includes both oral and topical therapies with low cure rates. The topical therapy is less effective but causes fewer side effects. This is why the development of an effective, easy to apply formulation for topical treatment is of high importance. We have used a nanotechnological approach to formulate Pickering emulsions (PEs) with well-defined properties to achieve site-specific delivery for antifungal drug combination of tioconazole and Melaleuca alternifolia essential oil. Silica nanoparticles with tailored size and partially hydrophobic surface have been synthesized and used for the stabilization of PEs. In vitro diffusion studies have been performed to evaluate the drug delivery properties of PEs. Ethanolic solution (ES) and conventional emulsions (CE) have been used as reference drug formulations. The examination of the antifungal effect of PEs has been performed on Candida albicans and Trichophyton rubrum as main pathogens. In vitro microbiological experimental results suggest that PEs are better candidates for onychomycosis topical treatment than CE or ES of the examined drugs. The used drugs have shown a significant synergistic effect, and the combination with an effective drug delivery system can result in a promising drug form for the topical treatment of onychomycosis.

Staphylococcus spp. is not only a commensal bacteria but also a major human pathogen that causes a wide range of clinical infections. Recent evidence suggests that Staphylococcus has the ability to colonize the reproductive system and to affect its structure and functions. The objective of this study was to determine the chemical properties and antibacterial effects of select essential oils (EOs): Amyris balsamifera L., Boswellia carterii Birdw., Canarium luzonicum (Blume) A. Gray, Cinnamomum camphora (L.) J. Presl., Cinnamomum camphora var. linaloolifera Y. Fuita, Citrus x aurantium L., Gaultheria procumbens L., Litsea cubeba (Lour.) Pers., Melaleuca ericifolia Smith., Melaleuca leucadendra L., Pogostemon cablin (Blanco) Benth., Citrus limon (L.) Osbeck, Santalum album L., and Vetiveria zizanoides (L.) Roberty against 50 Staphylococcus spp. cultures isolated from human semen, specifically Staphylococcus aureus, S. capiti, S. epidermidis, S. haemoliticus, and S. hominis. The disc diffusion and broth microdilution methods were used to assess the antimicrobial potential and to determine the minimum inhibitory concentration (MIC) of the selected EOs. The best anti-Staphylococcus activities were found with both methods for the essential oils of C. luzonicum (Blume) A. Gray, A. balsamifera, C. camphora, and P. cabli.

Tropical floodplain wetlands are found in low-lying areas that are periodically inundated. During wet periods, these wetlands can receive large amounts of suspended and dissolved material from the catchment, including many potential pollutants. In this study, we use traditional isotope tracers (&delta, 15N and &delta, 13C) along with soil eDNA to investigate the sources of transported materials and potential contaminants in seven forested floodplain wetlands in tropical Australia. We hypothesised that eDNA and isotope tracers in the soil would reflect the land use of the catchment. Our goal was to test whether eDNA could be used as a potential tool to identify and monitor pollutants in floodplain wetlands. The sampling sites were located within catchments that have a mosaic of land types, from well-conserved rainforests to intensive agricultural land uses, such as grazing, sugar cane, wood production, and horticulture. The soil eDNA was comprised of a mix of plant species consistent with the land use of the catchments. Most of the eDNA pool was derived from native trees, accounting for 46.2 &plusmn, 6.5% of the total, while cultivated species associated with agricultural activities contributed to 1&ndash, 24% of the total. From the cultivated species, highest contributions (&gt, 5%) were from Sorghum sp. used for grazing, banana (Musa ornata), melons (Cucumis melo), and Pinus radiata and Juniperus sp. grown for wood production. Interestingly, tropical wetlands on sites 15 km offshore had soil eDNA from agricultural activities of the mainland, highlighting the connectivity of these wetlands, probably during extensive floods. Overall, soil eDNA, more than isotopic tracers, showed promising results for tracing and monitoring potential pollutants in tropical floodplain wetlands that are highly connected and susceptible to environmental degradation.

Rhizoctonia solani Kuhn, the causal pathogen of sheath blight is second most damaging fungal disease in rice. While using chemical fungicides present high detriment to environment, the study investigate the efficacy of treatments composing five biofungicides in three different rates along with a biological agent, chemical check and untreated against the pathogen in in vitro level. In vitro efficacy showed that Melaleuca alternifolia + terpenes at 3.00 ml/L of H20 (T6), Aloe vera powder (Manopol) + Melaleuca oil at 3.00 ml/L of H20 (T15) and at 2.00 ml/L of H20 (T14) and Melaleuca alternifolia + terpenes at 2.00 ml/L of H20 (T5) as very effective (0-10 mm diameter zone of growth) treatments comparable to the chemical check (T17). Therefore, attaining high yield rice while having low risk to environment can always be done.

Wild teas are valuable genetic resources for studying domestication and breeding. Here we report the assembly of a high-quality chromosome-scale reference genome for an ancient tea tree. The further RNA sequencing of 217 diverse tea accessions clarifies the pedigree of tea cultivars and reveals key contributors in the breeding of Chinese tea. Candidate genes associated with flavonoid biosynthesis are identified by genome-wide association study. Specifically, diverse allelic function of CsANR, CsF3’5’H and CsMYB5 is verified by transient overexpression and enzymatic assays, providing comprehensive insights into the biosynthesis of catechins, the most important bioactive compounds in tea plants. The inconspicuous differentiation between ancient trees and cultivars at both genetic and metabolic levels implies that tea may not have undergone long-term artificial directional selection in terms of flavor-related metabolites. Australian tea tree (Melaleuca alternifolia) oil (TTO) and its monoterpene constituents such as terpinen-4-ol (T4O), 1,8-cineole, limonene, p-cymene, and α-terpinene have been shown to be effective in controlling a wide range of parasitic infections. The anti-parasitic effects of these compounds are mainly due to their anti-histamine and anti-acetylcholinesterase activities as well as their ability to modulate host inflammatory responses. This review attempts to summarize recent advances in the uses of TTO and its 15 major monoterpene constituents in treating parasitic infections in both humans and animals. Activities against parasitic protozoans (Plasmodium falciparum, Leishmania spp., Trypanosoma spp., Acanthamoeba castellanii, Trichomonas vaginalis, Eimeria, and Ichthyophthirius multifiliis), nematodes (Haemonchus contortus and Anisakis simplex), cestode (Echinococcus ortleppi), and monogeneans (Gasterosteus spp. and Dactylogyrus minutus) have been reported, showing good potentials in treating parasitic infections. Further studies are necessary for developing anti-parasite therapies using TTO or its monoterpenes constituents.

A viscous solution of low molecular weight chitosan (CH) at 5% w/v (10.2 kDa, 75 % deacetylated, 1451 cP at 25 °C) was associated with a microemulsion (ME) that undergoes a phase transition after water absorption in situ (≈28 % w/w), forming a more viscous liquid crystal, which was potentially evaluated as a topical vehicle. The ME was selected from a phase diagram, selecting a composition based on Tween® 80 (52 %), myristate isopropyl (28 %), and the aqueous phase (water and polyethylene glycol 400, 60:40 w/w) (20 %), which was after replaced by CH and herbal medicines (HM). HM are alternatives to treat candidiasis, and Stryphnodendron adstringens shell extract, characterized by molecular networking, and Melaleuca alternifolia Chell essential oil (46 % of terpinen-4-ol), showed in vitro activity against Candida albicans. Associating CH in ME improved the mechanical properties of the topical formulation, as adhesiveness, which is an advantageous feature for the topical treatment of vulvovaginal candidiasis.

Recently, there has been a demand for the replacement of chemical sunscreens with natural compounds that could prevent or restore UV-induced skin damage. Here, we investigated the photoprotective influence of the Melaleuca leucadendron ethanolic flower extract (EEMec) on factors involved in cellular and molecular UVB-induced oxidative stress in human skin keratinocytes (HaCaT). The phytochemical constituents, antioxidant potential by DPPH assay, content of total phenolic and flavonoid compounds in EEMec were evaluated. HaCaT cells were treated with EEMec followed by irradiation with UVB. CAT activity; GSH and ROS levels; and SOD1, GPx, CAT and COX-2 expression assays were employed to verify the oxidative stress, as well as EEMec effect on transmembrane transport, and pro-inflammatory and pro-apoptotic protein expression. EEMec reverted the viability loss of HaCaT cells after irradiation with UVB, exhibited significant antioxidant capacity and free radical scavenging activity in vitro, inhibited COX-2 expression and ensure protection of DNA-damage. EEMec shown a great photoprotective property to prevent keratinocytes damage induced by UV radiation and, thus a candidate potential to application as an adjuvant in sunscreen formulations as a strategy to reduce risk of sunburn and prevent skin diseases associated with UV-induced inflammation and cancer.

Staphylococcus aureus is able to rapidly develop mechanisms of resistance to various drugs and to form strong biofilms, which makes it necessary to develop new antibacterial drugs. The essential oil of Melaleuca alternifolia is used as an antibacterial, a property believed to be mainly due to the presence of terpinen-4-ol. Based on this, the objective of this study was to evaluate the antibacterial and antibiofilm potential of terpinen-4-ol against S. aureus. The Minimal Inhibitory and Minimal Bactericidal Concentrations (MIC and MBC) of terpinen-4-ol were determined, and the effect of its combination with antibacterial drugs as well as its activity against S. aureus biofilms were evaluated. In addition, an in silico analysis of its pharmacokinetic parameters and a molecular docking analysis were performed. Terpinen-4-ol presented a MIC of 0.25% (v/v) and an MBC of 0.5% (v/v) (bactericidal action); its association with antibacterials was also effective. Terpinen-4-ol has good antibiofilm activity, and the in silico results indicated adequate absorption and distribution of the molecule in vivo. Molecular docking indicated that penicillin-binding protein 2a is a possible target of terpinen-4-ol in S. aureus. This work highlights the good potential of terpinen-4-ol as an antibacterial product and provides support for future pharmacological studies of this molecule, aiming at its therapeutic application.

Mosquitoes are important vectors of several diseases, and control of these insects is imperative for human health. Insecticides have proven useful in controlling mosquito populations, but insecticide resistance and environmental concerns are increasing. Additionally, emerging and re-emerging microbial infections are problematic. Essential oils have been shown to be promising mosquito larvicidal agents as well as antimicrobial agents. In this work, the essential oils from four species of Myrtaceae (Baeckea frutescens, Callistemon citrinus, Melaleuca leucadendra, and Syzygium nervosum) growing wild in central Vietnam have been obtained by hydrodistillation and analyzed by gas chromatographic techniques. The essential oils have been screened for mosquito larvicidal activity against Aedes aegypti, Aedes albopictus, and Culex quinquefasciatus, and for antimicrobial activity against Enterococcus faecalis, Staphylococcus aureus, and Candida albicans. Callistemon citrinus fruit essential oil, rich in &alpha, pinene (35.1%), 1,8-cineole (32.4%), limonene (8.2%), and &alpha, terpineol (5.8%) showed good larvicidal activity with 24-h LC50 = 17.3 &mu, g/mL against both Ae. aegypti and Cx. quinquefasciatus, and good antibacterial activity against E. faecalis (minimum inhibitory concentration (MIC) = 16 &mu, g/mL) The 48-h larvicidal activities of M. leucadendra leaf essential oil, rich in &alpha, eudesmol (17.6%), guaiol (10.9%), linalool (5.1%), (E)-caryophyllene (7.0%), and bulnesol (3.6%) were particularly notable, with LC50 of 1.4 and 1.8 &mu, g/mL on Ae. aegypti and Cx. quinquefasciatus. Similarly, M. leucadendra bark essential oil, with &alpha, eudesmol (24.1%) and guaiol (11.3%), showed good antibacterial activity against. E. faecalis. Both B. frutescens and C. citrinus leaf essential oils demonstrated anti-Candida activities with MIC values of 16 &mu, g/mL. The results of this investigation suggest that essential oils derived from the Myrtaceae may serve as &ldquo, green&rdquo, alternatives for the control of mosquitoes and/or complementary antimicrobial agents.

Medicinal plants have long been used as an alternative in the treatment of diseases, whether using extracts, essential oils and / or bioactive components. Thus, the use of medicinal plants, with popularly known effects for the treatment of infections, has gained prominence, in research related to extracts and essential oils of medicinal plants, in an attempt to discover and future application of new molecules with antimicrobial and / or effect coadjuvant molecules to antimicrobials on the market. Studies with species of Melaleuca spp., Demonstrate wide application of its essential oil, without treating skin infections, microbial infections, herpes, diseases associated with pathogenic microorganisms of the oral cavity. The emerging microbial resistance to the drugs currently available on the market, has been affected by both researchers and the general population, so that the research experimented with was conducted revealing a high synergistic potential between drugs and natural drugs, present in drugs. In view of the above, this study aimed to present, through a bibliographic survey, a use of medicinal plants in the face of microbial resistance, as well as medicinal therapy based on the use of essential oils from alternative tea tree, leucadendron tea tree and quinquenervia tea tree, already inclusion in the literature.

The stable fly, Stomoxys calcitrans (Linnaeus 1758), is a hematophagous fly responsible for causing loss of performance in horses, causing losses in cattle productivity, and impacting the animals’ health through the spread of pathogenic microorganisms. The objective of this work was to investigate the insecticidal activity of essential oil obtained from Melaleuca alternifolia (Cheel), presenting high 1,8-cineole content, against S. calcitrans adults. Insecticidal activity was determined using surface application methods and exposure to oil impregnated paper. It was observed that treatments at 25 and 50 μg/cm2 (P < 0.05) present fumigant activity through exposure to the impregnated paper, and in the first 15 min of exposure, the mortality rates obtained for these treatments were, respectively (96.6 ± 3.3% and 100%), equivalent to the positive control. Using the superficial application method, the only treatment concentration presenting adulticidal action was 5% (w/v) (P < 0.05). Respective toxicities LC50 (%, w/v) and LC80 for the impregnated paper method were 1.06 ± 0.02 and 1.47 ± 0.17; for the superficial application method, they were 3.82 ± 0.65 and 5.53 ± 0.74. As demonstrated, M. alternifolia essential oil presents adulticidal potential against S. calcitrans.

Essential oils are potential antimicrobial agents and can be used as active ingredients in the pharmaceutical, food and cosmetics industries. This work intends to evaluate the antibacterial activity and design a strategy for the proposition of the mechanism of action of Melaleuca leucadendra essential oil. Optimum concentration of the bacteria and the phase where they had the highest pathogenic activity were determined. Results show that for each microorganism it is necessary to use a different concentration at the time of adjusting the initial inoculum, and that the time to achieve exponential growth phase varies from one to the other. M. leucadendra essential oil demonstrated in vitro antimicrobial properties. This oil was chemically characterized and the main compounds were evaluated by their mechanism of antibacterial action based on structure-activity analysis. The mechanism is related to the increase of bacteria cell membrane permeability. This indication was confirmed by flow cytometry and transmission electronic microscopy. Thus, in silico analysis is an important tool in the search for new antimicrobial agents and these results showed that M. leucadendra essential oil may be useful on the development of new chemotherapies or food preservation systems.

Background Phytotherapy is becoming a more and more common practice, not only for personal care but also for pet care. Nevertheless, we often have to deal with substances on which, in most cases, very little literature is available, even more so if the species of interest are the exotic ones. In particular, the essential oil from the Melaleuca leaves, because of its antinflammatory and antibacterial properties, is widely used and very little is known about its potential toxicity on pet birds. The present paper describes the first case of Tea tree oil intoxication in a pet bird. Case presentation A one-year-old, 80 g male cockatiel (Nymphicus hollandicus) was presented for clinical examination due to a serious despondency episode after the application of 3 drops of tea tree oil (Melaleuca alternifoglia) directly on the cutis of its right wing. The subject was urgently hospitalized and blood tests were performed.Serum biochemical values showed severe liver damage and slight renal involvement, complete blood count (CBC) parameters indicated a moderate neutrophilia a moderate neutropenia. Warm subcutaneous fluids and vitamin (VIT) B12 were administered, and after 8 h of fluid therapy the clinical condition of the patient improved. The subject was discharged after 48 h of hospitalization, in stable conditions. Conclusions Toxicosis are relatively common in bird pets and a number of cases are reported in literature, concerning heavy metals intoxications and toxic plants ingestion. However, in literature there are no described cases regarding Melaleuca oil intoxication in pet birds, but it has been reported in humans (mainly by ingestion) as well as in dogs, cats and rats. We hope that this first case report can be an initial aid in the knowledge of this potential toxicosis and therefore in the clinical veterinary practice of pet birds.

Grape berries are susceptible to Aspergillus niger (A. niger) infection during storage, leading to a significant reduction in its nutritional quality. However, most alternations in nutrient contents and related gene expression during fungal infection or treated with antimycotics remain unexplored. This work aimed to monitor and verify the metabolic changes in berries caused by A. niger or Melaleuca alternifolia oil (MAO) by using UHPLC-ESI-MS2 and Quantitative Real-time PCR (RT-qPCR). Results showed that sucrose, glucose, fructose, trans-resveratrol and pterostilbene levels were down and pentose phosphate pathway, glycolysis pathway and phenylpropanoid pathway were significantly down-regulated compared with healthy berries due to A. niger infection, all of which were alleviated by MAO treated. A. niger also induced down-regulation of key genes expression associated with metabolic pathways and magnitude of down-regulation was reduced by MAO. These results provide a theoretical basis for MAO used to control the risk of A. niger-mediated diseases.

Essential oils from the leaves, stems, flowers and fruits of Melaleuca leucadendra growing in the state of Pernambuco, Brazil, were analyzed using gas chromatography-mass spectrometry (GC-MS). The effects of the oils and their major constituent were evaluated on the agricultural pests Tetranychus urticae and Plutella xylostella in different stages of development. The analysis revealed a M. leucadendra chemotype rich in (E)-nerolidol (81.78 ± 0.90 to 95.78 ± 1.20%). P. xylostella was more susceptible to the oils and major constituent than T. urticae. The fruit oil was 1.5-fold more toxic than the leaf oil to T. urticae eggs. (E)-Nerolidol was 5.5-fold and 4.5-fold more toxic to T. urticae adults than the leaf and fruit oils, respectively. Azamax® used as the positive control was more efficient than the oils and (E)-nerolidol against T. urticae. However, the oils and (E)-nerolidol were more toxic to P. xylostella than Azamax®.

Essential oils (EOs) are known for their use in cosmetics, food industries, and traditional medicine. This study presents the chemical composition and therapeutic properties against kinetoplastid and eukaryotic cells of the EO from Melaleucaleucadendra (L.) L. (Myrtaceae). Forty-five compounds were identified in the oil by GC-MS, containing a major component the 1,8-cineole (61%). The EO inhibits the growth of Leishmania amazonensis and Trypanosoma brucei at IC50 values &lt, 10 &mu, g/mL. However, 1,8 cineole was not the main compound responsible for the activity. Against malignant (22Rv1, MCF-7, EFO-21, including resistant sublines MCF-7/Rap and MCF-7/4OHTAMO) and non-malignant (MCF-10A, J774A.1 and peritoneal macrophage) cells, IC50 values from 55 to 98 &mu, g/mL and from 94 to 144 &mu, g/mL were obtained, respectively. However, no activity was observed on Staphylococcus aureus, Enterococcus faecalis, Escherichia coli, Pseudomonas aeruginosa, Aspergillus niger, Candida parapsilosis, Microsporum canis, or Trypanosoma cruzi. The EO was able to control the lesion size and parasite burden in the model of cutaneous leishmaniasis in BALB/c mice caused by L. amazonensis compared to untreated animals (p &lt, 0.05) and similar with those treated with Glucantime&reg, (p &gt, 0.05). This work constitutes the first evidence of antiproliferative potentialities of EO from M. leucadendra growing in Cuba and could promote further preclinical investigations to confirm the medical value of this plant, in particular for leishmaniasis treatment.

The essential oil of the Melaleuca alternifolia (Maiden & Betche) (tea tree oil) has been effective in previous studies, in the treatment of infestation by Demodex mites in humans. The present study aimed at evaluating the in vitro acaricidal effect of this herbal medicine on Demodex canis. For the parasitological examination, samples were collected from a dog’s skin and examined using optical microscopy. Only samples with intact mites and with evident movement of chelicerae and tarsi were selected. Twenty-one samples were tested with the oil, in seven different concentrations: 100%; 50%; 25%; 12.5%; 6.25%; 5.0% and 3.13%. Three samples were tested with the positive control amitraz, and three with the negative control neutral shampoo. The interval between the time the solution was added and the moment the movement of the last mite ceased defined the survival time in the sample. By comparing the times at different concentrations and controls, the results showed that the higher the concentration of the oil, the more quickly its lethal effect occurred, and that the survival times in the controls were longer than in the different concentrations of melaleuca oil. Resumo O óleo essencial de Melaleuca alternifolia (Maiden & Betche) (óleo da árvore do chá) foi eficaz em estudos anteriores, no tratamento da infestação por ácaros Demodex em humanos. O presente estudo teve por objetivo avaliar o tempo do efeito acaricida, in vitro, desse fitoterápico sobre Demodex canis. Para o exame parasitológico, amostras foram coletadas da pele de um cão e examinadas por microscopia óptica. Apenas amostras com ácaros íntegros e com evidente movimentação de quelíceras e tarsos foram selecionadas. Vinte e uma amostras foram direcionadas para teste com o óleo, em sete concentrações diferentes: 100%; 50%; 25%; 12,5%; 6,25%; 5,0% e 3,13%. Três amostras foram testadas com o controle positivo, amitraz, e três com o controle negativo, xampu neutro. O intervalo entre o momento em que a solução foi adicionada e o instante em que cessou o movimento do último ácaro definiu o tempo de sobrevivência na amostra. Os tempos nas diferentes concentrações e nos controles foram comparados. Quanto mais alta a concentração do óleo, mais rapidamente ocorreu seu efeito letal. Os tempos de sobrevivência nos controles foram maiores que nas diferentes concentrações do óleo de melaleuca.

Melaleuca alternifolia essential oil (tea tree oil) is widely used as an ingredient in skin care products because of its recognized biological activities. The European Scientific Committee on Consumer Products constantly promotes research and collection of data on both skin distribution and systemic exposure to tea tree oil components after the application of topical formulations. This study quantitatively evaluates permeation, skin layer distribution (stratum corneum, epidermis, and dermis), and release into the surrounding environment of bioactive tea tree oil markers (i.e., α-pinene, β-pinene, α-terpinene, 1,8-cineole, γ-terpinene, 4-terpineol, α-terpineol) when a 5% tea tree oil formulation is applied at a finite dosing regimen. Permeation kinetics were studied in vitro on pig ear skin using conventional static glass Franz diffusion cells and cells ad hoc modified to monitor the release of markers into the atmosphere. Formulation, receiving phases, and skin layers were analyzed using a fully automatic and solvent-free method based on headspace solid-phase microextraction/gas chromatography-mass spectrometry. This approach affords, for the first time, to quantify tea tree oil markers in the different skin layers while avoiding using solvents and overcoming the existing methods based on solvent extraction. The skin layers contained less than 1% of each tea tree oil marker in total. Only oxygenated terpenes significantly permeated across the skin, while hydrocarbons were only absorbed at trace level. Substantial amounts of markers were released into the atmosphere.

The exotic tree Melaleuca quinquenervia (melaleuca) has invaded and formed monotypic forest stands in ecologically sensitive wetlands of Florida. We hypothesized that natural-enemy impact would influence melaleuca’s litter and seed fall dynamics via chronic attacks, and enhance native species diversity, which will be reflected in leaf litter composition. This hypothesis was tested during 1997–2013 by studying melaleuca stands in occasionally inundated habitats infested by its natural enemies. Results from our study showed: (1) an initial increase in the leaf and seed fall during the first-half of the study period, followed by about 70% decline during remainder of the study period, (2) a positive correlation between the amount of leaf and seed fall, (3) a negative correlation between the proportion of natural-enemy damaged melaleuca leaf litter and the quantity of fallen seeds, and (4) a slow but steady increase of the non-melaleuca leaf litter amount by 81% and with a corresponding decrease in melaleuca leaf litter by 15%. Of the 17 non-melaleuca species recovered, we recorded two perennials at the onset of the study, which increased to seven during the second half of the study period, one of which was non-native. Cladium and Myrsine species produced significantly more leaf-litter by the end of the study period. Over 80% of the melaleuca leaf litter manifested natural-enemy damage during the last 7-year of the study period. These results provide evidence of a negative influence of natural enemy attack on melaleuca leaf and seed fall dynamics and a positive influence on native species diversity in the fallen leaf litter.

Melaleuca alternifolia tea tree oil (TTO) is largely used in cutaneous infections. Clinical observations reported antibacterial, antifungal, and antiviral activities, whereas in vitro experiments ascribed most of biological properties to terpinen-4-ol. Since different plant chemotypes and storage conditions result in variations of chemical composition of commercially available TTO, in this study we investigated the antimicrobial activity and the chemical profile of ten commercially available TTO products. The antimicrobial activity was assessed against Candida glabrata, Herpes simplex virus type 1 (HSV-1), methicillin-resistant Staphylococcus aureus (MRSA), and Pseudomonas aeruginosa grown in planktonic mode or biofilms. Only five out of ten TTO batches reported significant antimicrobial activity. The identified TTO products reduced bacterial survival in biofilms, generated oxidative damage in C. glabrata, and diminished HSV-1 infectivity. GC-MS analysis revealed that all the analyzed TTO batches fitted into the terpinen-4-ol chemotype even if we reported great variability in composition of nine major ISO-specified TTO components. Overall, we were not able to ascribe the antimicrobial activity to the content in terpinen-4-ol. We therefore conclude that the antimicrobial activity of TTO results from complex interaction among different components.

Some evidence has demonstrated that Ichthyophthirius multifiliis, etiologic agent of "white spot disease", causes severe bioenergetics dysfunction in the spleen of naturally infected silver catfish (Rhamdia quelen), which contributes directly to disease pathogenesis. Recently, several studies have demonstrated the efficacy of Melaleuca alternifolia essential oil, popularly known as tea tree oil (TTO), in the treatment of freshwater fish naturally or experimentally infected with I. multifiliis. In this sense, the aim of this study was to evaluate whether TTO is capable of preventing or reducing splenic bioenergetics dysfunction in silver catfish naturally infected with I. multifiliis. Splenic cytosolic and mitochondrial creatine kinase (CK) and pyruvate kinase (PK) activities decreased in infected animals compared to uninfected animals, while adenylate kinase (AK) activity increased. Treatment with TTO was able to prevent the inhibition on splenic CK and PK activities but was not able to prevent the stimulation of AK activity. Based on this evidence, treatment with TTO prevents the impairment on energetic metabolism via improvement of enzymes belonging to the phosphotransfer network, such as CK and PK. In summary, this treatment can be considered an interesting approach to prevent the bioenergetics imbalance in spleen of silver catfish naturally infected with I. multifiliis.

Melaleucaeae is the second largest angiosperm tribe in Australia containing more than 330 species. Most Melaleuca species are endemic to Australia and are found in a wide variety of habitats. The Melaleuca leucadendra complex, including M. leucadendra, M. quinquenervia and M. viridiflora, are dominant components of the tropical and sub-tropical biota of Australia. All three species are known to be susceptible to Austropuccinia psidii (myrtle rust), which was first detected in Australia in 2010 and is now widespread in the eastern states and the Northern Territory. In this study we examined variability in susceptibility to A. psidii within populations of M. quinquenervia, M. leucadendra and M. viridiflora using in-vitro assessments of seedlings from provenances across the native range of the three species. We identified wide variation in susceptibility to A. psidii between M. quinquenervia, M. leucadendra and M. viridiflora, among provenances within these species as well as within provenances. Further studies on larger populations of these Melaleuca species addressing the long-term impacts of repeated infection in native ecosystems are needed to not only help predict but also limit impact. Regaining lost genetic diversity within some of these species populations may require human intervention.

The aim of this study was to evaluate the efficacy and safety of several anesthetics in the brachyuran crab Neohelice granulata, an emergent experimental model. The essential oils (EOs) of Lippia alba, Aloysia tryphilla, and Melaleuca alternifolia (tea tree oil; TTO), the isolated compounds eugenol, menthol, terpinen-4-ol, and the nanoencapsulated form of TTO, were administered in one or more of the following ways: added to the water (immersion), through an arthrodial membrane (injected), or by oral gavage. Unexpectedly, most EOs did not produce an anesthetic effect after immersion. Only TTO and eugenol induced anesthesia by immersion, with very long induction and recovery times compared to anesthesia of other crustaceans. However, a good anesthetic effect was observed with the injection of terpinen-4-ol and nanoencapsulated TTO in N. granulata; both demonstrated ideal induction and recovery times. These substances appear to be promising anesthetic alternatives for crustaceans.

Australian tea tree oil (TTO) and its extract terpinen-4-ol (T4O) are found to be effective in moderating demodex-related diseases. Their possible effects are lowering the mite counts, relieving the demodex-related symptoms and modulating the immune system especially the inflammatory response. This review summarizes the topical treatments of TTO and T4O in human demodicosis, their possible mechanism of actions, side-effects and potential resistance in treating this condition. Although current treatments other than TTO and T4O are relatively effective in controlling the demodex mite population and the related symptoms, more research on the efficacy and drug delivery technology is needed in order to assess its potential as an alternative treatment with minimal side-effect profile, low toxicity and low risk of demodex resistance.

Plants have developed complex defense mechanisms to protect themselves against pathogens. A wide-host-range fungus, Austropuccinia psidii, which has caused severe damage to ecosystems and plantations worldwide, is a major threat to Australian ecosystems dominated by members of the family Myrtaceae. In particular, the east coast wetland foundation tree species Melaleuca quinquenervia, appears to be variably susceptible to this pathogen. Understanding the molecular basis of host resistance would enable better management of this rust disease. We identified resistant and susceptible individuals of M. quinquenervia and explored their differential gene expression in order to discover the molecular basis of resistance against A. psidii. Rust screening of germplasm showed a varying degree of response, with fully resistant to highly susceptible individuals. We used transcriptome profiling in samples collected before and at 5 days postinoculation (dpi). Differential gene expression analysis showed that numerous defense-related genes were induced in susceptible plants at 5 dpi. Mapping reads against the A. psidii genome showed that only susceptible plants contained fungal-derived transcripts. Resistant plants exhibited an overexpression of candidate A. psidii resistance-related genes such as receptor-like kinases, nucleotide-binding site leucine-rich repeat proteins, glutathione S-transferases, WRKY transcriptional regulators, and pathogenesis-related proteins. We identified large differences in the expression of defense-related genes among resistant individuals.

Accumulation of fine grained sediment and natural regeneration of mangrove species are normally prioritized to control erosion on mangrove dominated muddy coasts. Current solutions to manage eroded mangrove dominated muddy coasts have been not as cost-effective as expected. Entrapping microsites, although mainly used for restoring eroded terrestrial resources, were first tested for restoring an eroded mangrove dominated muddy coast of Vam Ray, Hon Dat, Kien Giang, Vietnam. Entrapping microsites, constructed with a 50 cm layer of discarded Melaleuca small sticks that were overlain on the surface of the site, accumulated a high level of fine grained sediment and promoted robust natural regeneration or regrowth of local mangrove species after approximately two years. The effectiveness and efficiency of the Vam Ray Melaleuca entrapping microsites provide a good basis on which further research and ground works can proceed.

In Melaleuca minutifolia mature leaves are linear, in Melaleuca micromera scale-like and peltate. This peltation is caused by swollen basal mesophyll without meristematic fusions. Thus, a novel peltation strategy is described. The foliar change of two Australian Melaleuca species (Myrtaceae) was investigated in different ontogenetic stages from cotyledons up to the mature leaf types. In both taxa (M. minutifolia and M. micromera), a strong leaf reduction occurs. While in M. minutifolia the mature leaf type is linear, inverse bifacial and strongly adpressed to the shoot axis, in M. micromera it is scale-like, aequifacial, minute and peltate. The mature leaf type of M. micromera is one of the rare examples of peltation among woody species. The ontogenetic studies on the mature peltate leaf type of M. micromera showed that a shield-like leaf base and typical peltate leaf morphology can be formed, not exclusively by meristematic fusions, as is the case for peltate leaves in the classical sense, but instead by strongly swollen mesophyll, particularly in the basal part of the leaf. While most species with peltate leaves are herbaceous perennials from very moist to aquatic habitats and lack reduced leaves, the four Melaleuca species known with the M. micromera type of peltation are from habitats showing either seasonal water stress, soil infertility or both and all show extreme leaf reduction. Given our morpho-anatomical data, it is clear that, as well as the classical type of peltation, a novel, second strategy for achieving peltation needs to be distinguished. The two types are correlated with quite different environmental conditions.

The severity of the effects of herbivory on plant fitness can be moderated by the ability of plants to compensate for biomass loss. Compensation is an important component of the ecological fitness in many plants, and has been shown to reduce the effects of pests on agricultural plant yields. It can also reduce the effectiveness of biocontrol through introduced herbivores in controlling weedy invasive plants. This study used a modeling approach to predict the effect of different levels of foliage herbivory by biological control agents introduced to control the invasive tree Melaleuca quinquennervia (melaleuca) in Florida. It is assumed in the model that melaleuca can optimally change its carbon and nitrogen allocation strategies in order to compensate for the effects of herbivory. The model includes reallocation of more resources to production and maintenance of photosynthetic tissues at the expense of roots. This compensation is shown to buffer the severity of the defoliation effect, but the model predicts a limit on the maximum herbivory that melaleuca can tolerate and survive. The model also shows that the level of available limiting nutrient (e.g., soil nitrogen) may play an important role in a melaleuca’s ability to compensate for herbivory. This study has management implications for the best ways to maximize the level of damage using biological control or other means of defoliation.

During 2015–2016, wooden and herbaceous plants growing in parks, boulevards, fields, gardens and forests in Khuzestan province, southwestern Iran, were visually inspected for symptoms resembling phytoplasma. Fifty‐one symptomatic samples from nine different species and one symptomless sample from each plant were collected. Leaf midribs, petioles and the parts of stem cambium were separated and freeze‐dried. Total DNA was extracted using CTAB‐based method and tested for phytoplasma using a nested PCR assay. The expected size amplicons of 16S rDNA were sequenced and compared to those of reference phytoplasmas by BLASTn search and phylogenetic analysis. The consensus 16S rDNA sequence of the detected phytoplasma in narrow cattail related to reference phytoplasma group 16SrVI, “Candidatus Phytoplasma trifolii” while in the other plants were related to reference phytoplasma subgroup 16SrII–D, “Candidatus Phytoplasma aurantifolia.” All isolates showed 98%–99% sequence identity to members of their reference groups. To our knowledge, this is the first report of “Candidatus Phytoplasma aurantifolia”‐related strains infecting the plants of Acacia salicina, Alternanthera ficoidea, Melaleuca citrine, Citrus aurantium throughout the world and Celosia christata in Iran. Furthermore, this study is the first to report the association of a “Candidatus Phytoplasma trifolii”‐related strain with Typha angustifolia worldwide.

Ethnopharmacological relevance Medicines of the Yao ethnic group in China are a special branch of traditional Chinese medicine (TCM) and are well documented for use in disease prevention. According to an ethnopharmacological survey, there are 1392 species of medicinal plants that have been documented as Yao ethnomedicines and 104 of these species are used routinely. This study evaluated a partial collection of these 104 core plant species for their potential as anticancer agents. Material and methods A literature study of scientific journals and books in the local language was conducted. Based on an ethnopharmacological survey, 16 plant species widely used in Yao ethnomedicine were collected and 64 plant extracts were prepared from these plants. in vitro cytotoxicity screening was conducted with a panel of four human cancer cell lines, lung cancer A549, breast cancer BT20 and MCF-7, bone cancer U2OS. The potential toxicity of the extracts was evaluated using two normal human cell lines, human peripheral lung epithelial cells (HPL1A) and human umbilical vein endothelial cells (HUVEC). Additionally, the 10 extracts that demonstrated cytotoxicity in cancer cells with an IC50 of less than 25.0 µg/mL were examined for the ability to induce apoptosis in U2OS cells. Results The up-to-date information regarding the traditional uses, pharmacological and biological activities, as well as the chemical constituents of the 16 plants are presented. Extracts from all 16 plants showed cytotoxicity against one to four of the human cancer cell lines and the cytotoxic effects of extracts from Melaleuca leucadendra, Stephania longa, Microsorium fortune and Bidens biternata were demonstrated for the first time. The highest anticancer potential was observed for extracts prepared from Melaleuca leucadendra Linn against all tested cancer cells (BT20, A549, U2OS, and MCF7) with an IC50 range of 3.1–32.7 µg/mL. The selectivity index of the active samples varied from 0.1 to 25, and five extracts from Bidens biternata, Wedelia calendulacea, Stephania longa and Achras zapota showed significant selectivity against cancer cell lines versus normal cell lines. All tested extracts induced apoptosis in U2OS cells, and for the first time extracts from Melaleuca leucadendra and Microsorium fortune were shown to induce apoptosis. Conclusion We demonstrated the in vitro anticancer efficacy and safety of 16 medicinal plants that have been historically used in Yao ethnomedicine. This study provides evidence to assist the clinical practice of Yao ethnomedicine and the development of chemotherapeutic agents from extracts prepared from these plants.

Purpose: The aim of this study was to isolate and identify potentially pathogenic airborne fungi from Hospital das Clinicas Samuel Libanio in the city of Pouso Alegre-MG and evaluate their susceptibility to natural and industrial products. Methods: The air collection was performed by passive sedimentation in the morning during the autumn and winter seasons. Petri dishes were open as the location of air conditioning. The isolates were subjected to pathogenicity test. The identification of the fungi was performed according to the macroscopic evaluation and micromorphology. Potentially pathogenic isolates were susceptibility tested by disc diffusion method. The agents used were insecticides and industrial cleaning products and essential oils of citronella plants, lemon grass, eucalyptus and Melaleuca extracted by steam distillation method. Results: We obtained a total of 356 fungal isolates. The inside door environments were 126 (35.39%) and the outside environments were 230 (64.6%) isolates. The 22 (6.18%) isolates from the inside and 25 (6.18%) outside the hospital showed pathogenic potential. This isolates were identified as Acremonium spp., A. niger, A. terreus, A. versicolor, Curvularia sp., Penicillium sp. and Scopulariopsis sp. Susceptibility testing it was observed that most of the isolates were susceptible to the principle product containing sodium hypochlorite. Citronella oil showed enormous potential inhibition against all isolates. Already lemon grass oil was effective only against isolates of Penicillium spp. Conclusions: All genres identified are significant allergens, which can cause respiratory disease in both immunocompromised individuals such as asthmatics and people with any immune deficit. The monitoring of environmental sources should be performed, especially in special areas with immunocompromised patients. Despite efforts to try to reduce fungal infections hospital there are still flaws in the strategies employed.

Plasma polymer coatings fabricated from Melaleuca alternifolia essential oil and its derivatives have been previously shown to reduce the extent of microbial adhesion on titanium, polymers, and other implantable materials used in dentistry. Previous studies have shown these coatings to maintain their performance under standard operating conditions; however, when used in e.g., a dental implant, these coatings may inadvertently become subject to in situ cleaning treatments, such as those using an atmospheric pressure plasma jet, a promising tool for the effective in situ removal of biofilms from tissues and implant surfaces. Here, we investigated the effect of such an exposure on the antimicrobial performance of the Melaleuca alternifolia polymer coating. It was found that direct exposure of the polymer coating surface to the jet for periods less than 60 s was sufficient to induce changes in its surface chemistry and topography, affecting its ability to retard subsequent microbial attachment. The exact effect of the jet exposure depended on the chemistry of the polymer coating, the length of plasma treatment, cell type, and incubation conditions. The change in the antimicrobial activity for polymer coatings fabricated at powers of 20–30 W was not statistically significant due to their limited baseline bioactivity. Interestingly, the bioactivity of polymer coatings fabricated at 10 and 15 W against Staphylococcus aureus cells was temporarily improved after the treatment, which could be attributed to the generation of loosely attached bioactive fragments on the treated surface, resulting in an increase in the dose of the bioactive agents being eluted by the surface. Attachment and proliferation of Pseudomonas aeruginosa cells and mixed cultures were less affected by changes in the bioactivity profile of the surface. The sensitivity of the cells to the change imparted by the jet treatment was also found to be dependent on their origin culture, with mature biofilm-derived P. aeruginosa bacterial cells showing a greater ability to colonize the surface when compared to its planktonic broth-grown counterpart. The presence of plasma-generated reactive oxygen and nitrogen species in the culture media was also found to enhance the bioactivity of polymer coatings fabricated at power levels of 10 and 15 W, due to a synergistic effect arising from simultaneous exposure of cells to reactive oxygen and nitrogen species (RONS) and eluted bioactive fragments. These results suggest that it is important to consider the possible implications of inadvertent changes in the properties and performance of plasma polymer coatings as a result of exposure to in situ decontamination, to both prevent suboptimal performance and to exploit possible synergies that may arise for some polymer coating-surface treatment combinations.

Objetivo: Entre os diversos óleos essenciais, o óleo essencial de Melaleuca alternifolia se destaca diante das suas múltiplas atividades biológicas, sendo a principal a antimicrobiana, tornando-o um elemento atraente para tratamentos faciais. Dito isso, o objetivo deste estudo é avaliar a eficácia do óleo essencial de Melaleuca alternifolia no tratamento da acne. Métodos: O trabalho desenvolvido caracteriza-se como revisão sistemática da literatura. O levantamento bibliográfico foi realizado nas bases de dados Pubmed, ScienceDirect, Lilacs e Medline. Durante a etapa de busca, se estabeleceram como critérios de inclusão artigos publicados no período de 2011 a 2021, artigos com dados primários e disponíveis em língua inglesa, portuguesa e espanhola. Já os critérios de exclusão foram: artigos que apresentaram incompatibilidade com o tema, duplicidade nas bases e artigos de revisão. Resultados: Após a realização das buscas nas bases de dados, da aplicação dos critérios de inclusão e leitura do título e resumo, foram selecionados dez artigos que se enquadravam nos requisitos de elegibilidade. Os dez artigos foram lidos na íntegra, com análise acurada de suas metodologias e principais resultados. Considerações finais: O óleo de melaleuca apresenta um grande potencial no tratamento da acne devido sua significativa atividade antimicrobiana, capacidade de reduzir lesões, bem como auxiliar na cicatrização de feridas. Ademais, apresenta vantagens em relação ao uso de antibióticos devido ao baixo impacto no desenvolvimento de resistência e suscetibilidade aos antimicrobianos. Ainda que apresente estudos acerca das atividades farmacológicas deste óleo, é imprescindível a realização de pesquisas mais apuradas para consolidar esses apanhados.
