203. Phylogeography and Origin of Sheep Breeds in Northern China

Abstract

With the establishment of modern sheep production systems in China, various forms of hybridization with Western breeds and between native breeds have been utilized for genetic improvement. At the same time, the progressive destruction or deterioration of sheep habitat has accompanied urbanization in China. Together these factors have accelerated the loss of genetic diversity, or even resulted in the extinction of some indigenous breeds. It is therefore important that efficient strategies for surveillance, evaluation, conservation and utilization of available genetic resources are developed for this species. In this study, a total of 30 microsatellite markers were used to assess genetic diversity for 12 native breeds and one Western sheep breed in Northern China. The high polymorphism information contents at the 30 markers, varying from averages of 0.519 to 0.666 for the 13 breeds, imply the retention of natural variation from source populations in the domestic breeds from different geographic regions in China. Analysis of genetic differentiation revealed substantial divergence among these breeds. Neutrality tests indicated that more than one third of the 30 loci were in departure from neutrality, implying that some evolutionary forces (e.g. selection and migration) had acted on these populations. Phylogenetic and phylogeographic analyses displayed a remarkable degree of consistency between geographic origins, breeding histories and the pattern of genetic differentiation.

204. LC−MS/MS assay for the determination of a novel anti‐fibrotic candidate mefunidone in monkey plasma and its application to a pharmacokinetics study.

Abstract

Mefunidone (MFD) is a promising anti‐fibrotic candidate molecule with greater anti‐fibrotic activity than pirfenidone (PFD). However, there has been no report on the methodology used for the quantification of MFD or on any investigation of its pharmacokinetics. In this study, an efficient and reliable liquid chromatography–tandem mass spectrometry (LC–MS/MS) method was developed to assay MFD in monkey plasma. This assay method was validated and applied to a pharmacokinetics study in monkeys. The lower limit of quantification of this assay was 0.1 μg·mL−1, and the linear calibration curve was acquired with R2 > 0.99 between 0.1 and 60 μg·mL−1. The intra‐day and inter‐day precision were evaluated with coefficient of variations of 1.5%–5.8%, whereas the mean accuracy ranged from 91.7% to 106.9%. A negligible matrix effect and good recovery were obtained using this assay, with average extraction recoveries of MFD and the internal standard (IS) in the range of 85.5%–124.8% and 84.1%–94.0%, respectively. The precision of the absolute matrix effect of MFD and the IS was 1.2–3.0% and 1.2–7.3%, respectively. The samples were stable under all experimental conditions. Linear pharmacokinetics were observed for MFD in monkeys, where the exposures of MFD increased proportionally with increasing MFD doses at the range of 10–90 mg·kg−1. Moderate elimination of MFD from the body was observed, with t1/2 of 5–7 h, and the elimination rate of MFD was stable during multiple dosing. In conclusion, this method provides an reliable analytical approach for quantification of MFD in plasma and was successfully applied to a pharmacokinetics study in monkeys. [ABSTRACT FROM AUTHOR]

Abstract

Single nucleotide polymorphisms of the 5' regulatory region of the FSHR gene were detected in 3 goat breeds (Jining Grey, Boer, and Inner Mongolia Cashmere) by the polymerase chain reaction-single-strand conformation polymorphism method, and its effects on litter size in Jining Grey goats were also evaluated. Concerning primer P1, 2 genotypes (AA and BB) were detected in Boer goats, while only the AA genotype was identified in Jining Grey and Inner Mongolia Cashmere goats. Sequencing revealed a T->A transversion at the 26th position and an A->C transversion at the 61st position of the amplified region in genotype BB in comparison to genotype AA. Regarding primer P2, 3 genotypes (CC, CD, and DD) were identified in the 3 goat breeds mentioned above. Sequencing revealed a T->A transversion at the 70th position and a G->C transversion at the 130th position of the amplified region in genotype DD compared with CC. Does with genotype CC had 0.46 (P < 0.05) or 1.03 (P < 0.05) kids more than those with genotype CD or DD, respectively, while does with CD had 0.57 (P < 0.05) kids more than those with DD in Jining Grey goats. These results preliminarily showed that allele C of the FSHR gene was a potential marker for improving litter size in goats.

Abstract

Heteroplasmy is the presence of more than one mitochondrial DNA (mtDNA) variant within a cell, tissue, or individual. In this study, sequence variation was investigated in the control region (CR) of mitochondrial DNA (mtDNA) from 135 individuals belonging to five primary domestic Bactrian camel breeds in China and Mongolia. Due to variation of the repeat unit G(T/C)(AC)n, length heteroplasmy was detected within each camel by direct sequencing and fragment analysis. A high occurrence of mtDNA heteroplasmy, up to 100 percentages was observed in five camel populations. Our study provides the first evidence of extensive length heteroplasmy in Bactrian camels. Abstract

Using Jining Grey goat ear marginal tissues as experimental materials, we succeeded in establishing a fibroblast cell bank, including 32 samples, by means of a primary explant and cryopreservation technique. The results from the biological analysis suggested that the population doubling time of the cell line was approximately 48 h. The diploid accounted for 98.35%, the isozyme analysis of the lactic dehydrogenase and malic dehydrogenase disproved cross contamination, and the results of the bacterium, fungi, virus, and mycoplasma detection tests were all negative. The transfection rates of 3 fluorescent protein genes were high, which supported the opinion that exogenous genes could be effectively expressed in the cells. This research has not only preserved the precious germplasm resources of the Jining Grey goat at the somatic cell level, but it has also provided valuable materials for the study of genomics, postgenomics, and somatic cloning.

Abstract

Considering the characteristics of science and technology resources image, the system structure of image retrieval techniques from the Science and technology Resources image database is designed to rapidly and effectively retrieve the image from the database. Firstly, a novel image retrieval algorithm based on texture is proposed. The algorithm proposed takes fully the advantage of statistical attribution of image’s Nonsubsampled Contourlet transform (NSCT) coefficients to form the eigenvector for depicting textural feature and shape feature. Finally, the TBIR method is used to retrieve the image from the database. Secondly the CBIR method is used to retrieve the image from the result of the TBIR method. Moreover, the three key technologies, the image segmentation, the feature extraction and the similarity measurement, is researched and comprehensively used to retrieve the image.

246. Analysis of transmission spectra and cladding mode evolution of etched double-cladding fiber long-period grating.

Abstract

We demonstrate the inscription of long-period gratings (LPGs) in a fluorine-doped double-cladding fiber (DCF) with a low refractive index trench. The grating has a single resonance dip in the wide wavelength range from 1200 nm to 2400 nm. The transmission spectra and cladding mode evolution of the DCF-LPG during the cladding etching process are investigated theoretically and experimentally. The mode order of the cladding modes decreases with one order of every 20 μ m diameter etching, while the resonance wavelength of the DCF-LPG barely shifts when the fiber diameter decreases gradually. Only LP 11 mode can be excited in the etched DCF-LPG when the fiber diameter is less than 40 μ m. The characteristics of the DCF-LPGs are investigated experimentally. The torsion sensitivity was measured to be −0.154 nm/(rad/m) and −0.271 nm/(rad/m) at 1. 55 μ m and 2. 0 μ m wavebands, respectively, which are one order of magnitude higher than that of the conventional LPG. The proposed DCF-LPG has potential application as a tunable band-rejection filter in broad wavebands. • Only one resonance dip of the DCF-LPG can be observed in the wavelength range from 1200 nm to 2400 nm. • The mode order of the cladding modes decrease with diameter etching, while the resonance wavelength barely shifts. • The twist sensitivity of the DCF-LPGs at 1. 55 μ m and 2 μ m wavebands was one order of magnitude higher than that of the conventional LPG.
