The Drosophila central nervous system comprises an enormous diversity of neurons that are originated from neuronal stem cells, neuroblasts. They generate a specific series of ganglion mother cells, each of which is once cleaved into a pair of neurons. Among genes known to control neurogenesis, prospero (pros) was recently identified as a gene required for gene expression specifying properties of some identified neurons. Here we report that pros encodes a nuclear protein containing a homeodomain-like sequence. In neuronal lineages of the central nervous system, pros protein is specifically detected in ganglion mother cells, although their parental neuroblasts have begun expressing a significant level of pros transcripts, suggesting a post-transcriptional control of pros expression. Our results provoke that in neuronal cell differentiation ganglion mother cells might play a pivotal role associating with the pros function.

Mouse lymphoma L5178Y cells express at least two isoforms of beta-tubulin, designated M beta I and M beta II, as revealed by isoelectrofocusing, whereas two independently isolated normal T-cell clones, 3D10 and K23, express only M beta I. M beta II-tubulin is more acidic (pI, 5.10) than M beta I-tubulin (pI, 5.15). L5178Y cells were disrupted under the microtubule-stabilizing conditions, followed by centrifugation to separate fractions containing polymerized and unpolymerized tubulin. We found that a proportion of M beta II to total beta-tubulins is larger in the fraction containing unpolymerized tubulin than in that containing polymerized tubulin. In addition, when tubulin was purified from extracts of L5178Y cells by repeated cycles of polymerization-depolymerization, the M beta II-tubulin isoform was gradually lost during the successive purification steps. The low recovery of M beta II-tubulin was observed, irrespective of the presence or absence of MAPs, and even in the presence of an excess amount of essentially polymerizable porcine brain tubulin. These results indicate that M beta II-tubulin is less able to polymerize than is M beta I-tubulin, both in vivo and in vitro.

Effect of 12-O-tetradecanoyl-phorbol-13-acetate (TPA) on rat AH66 hepatoma cells was studied with a reference to that of insulin and the epidermal growth factor (EGF). In a short term cell incubation, TPA and EGF caused an approximately 2-fold increase in the production of alpha-fetoprotein (AFP) and other acid-precipitable materials, while the same concentration of insulin brought a 3-fold increase. In a long term culture using a low serum medium, TPA as well as insulin and EGF caused remarkable proliferation of AH66 cells, but the increase in cell number was not accompanied by a proportional increase in the levels of AFP of the culture media. These biological effects of TPA, insulin and EGF appeared to resemble each other, and subsequent hormone binding studies showed that TPA inhibited 125I-EGF binding to its membrane receptors without affecting 125I-insulin binding. Scatchard analysis of TPA effect on EGF binding indicated that TPA altered the affinity of the membrane receptors for EGF without changing the total number of available receptors per cell. From these data, it is suggested that some of the biological effects of TPA on AH66 cells may result from alterations in the functions of cell membrane.

A new human functional tumor cell line, designated as T3M-3, has been established from a xenotransplanted choriocarcinoma grown in nude mice. One of the biggest problems of the in vitro culture of these tumor cells using the xenotransplanted tumors had been the dense contamination of fibroblasts of host nude mouse origin. In the present study, these fibroblasts were completely removed by incubating the cells with antiserum raised against nude mouse spleen cells. The cell line established from the remaining tumor cells has been successfully propagated in vitro for as long as 4 years. These cells show the morphology of epithelioid cells containing a prominent nucleus with one or two large nucleoli. The cells grow in a monolayered sheet with the population-doubling time of 19 hr. The cells show perfect tumor takes when they are reinoculated into nude mice. Chromosomal analysis revealed that the cell is a human aneuploid one with a hypotriploid mode. These cultured cells maintained well the function of secreting large amounts of human chorionic gonadotropin, progesterone, and estrogen. The secretion of human chorionic gonadotropin and progesterone by these cells is enhanced by stimulation with tumor promoters, such as 12-O-tetradecanoylphorbol-13-acetate and teleocidin B, or with epidermal growth factor in a dose-and time-dependent manner. Interestingly, however, the tumor promoters did not exert a marked effect on the cellular binding of epidermal growth factor, indicating that the receptors for these reagents in T3M-3 cells are not shared by epidermal growth factor.

A highly sensitive procedure of solid-phase radioimmunoassay (RIA) was developed for the detection of measles IgG antibody. HeLa cells persistently infected with measles virus were used as a solid-phase antigen. This technique was applied to the detection of measles IgG antibody in patients with subacute sclerosing panencephalitis (SSPE) and multiple sclerosis. Normal subjects having experienced natural measles or measles vaccination and patients with various neurological diseases of non-virus nature were also examined as control groups. Measles antibody was detected at high titers in both the sera and cerebrospinal fluid of SSPE patients. Moreover, RIA/HI ratios of SSPE patients were significantly higher than those of normal subjects, suggesting the presence in the formers of antibodies to nucleocapsids at high titers as well as to viral envelopes. On the other hand, no significant difference was found in both RIA and HI titers between the sera of multiple sclerosis and those of various neurological diseases.

The 90-kDa heat-shock protein (HSP90) has been purified from mammalian tissues, mouse liver and porcine brain, with a good yield by a new method involving hydrophobic chromatography. Mouse liver HSP90 and porcine brain HSP90 were compared with mouse lymphoma HSP90 which was purified from T lymphoma cell line, L5178Y, by a modification of the previously reported method. These three HSP90s were indistinguishable from one another in amino acid composition, one-dimensional peptide mapping, elution pattern of proteolytic fragments (trypsin- or V8-protease-cleaved) in reverse-phase high-performance liquid chromatography, reactivity with the antibody against mouse T lymphoma HSP90 and the ability to bind to F-actin. The amino acid sequences of three portions (total 47 amino acid residues) of lymphoma HSP90 were determined and they were homologous to those of the corresponding portions of Drosophila HSP83A and yeast HSP90. These results suggest that HSP90 is a highly conserved protein during evolution.

Three beta-tubulin isoforms, M beta Ib, M beta Ia and M beta II, were detected by isoelectric focusing gel electrophoresis (IEF) in extracts of cultured cells of a mouse T cell lymphoma, L5178Y. To investigate the origin(s) of the isoforms, we have isolated cDNA clones encoding beta-tubulin from a cDNA library prepared from poly (A)+ RNA of L5178Y cells. Seventeen cDNA clones carrying the entire coding sequences of beta-tubulin were isolated and classified into two distinct isotypes, represented by two clones designated pMT27 and pMT49, according to the results of restriction mapping. On the basis of the nucleotide sequences of the two cDNAs, pMT27 and pMT49 were identified as mouse beta-tubulin isotypes 3 and 5, respectively. By using in vitro translation products of hybrid-selected mRNAs and of the SP6 in vitro transcripts of the cDNAs, polypeptides encoded by the two cDNA clones were analyzed by IEF. We found that pMT27 and pMT49 encode M beta Ib and M beta Ia, respectively. In addition, M beta II was detected in translation products of mRNA specifically hybridized to pMT49, but not in those of the in vitro transcript of pMT49 DNA. These results suggest that M beta II is the translation product of mRNA whose 3'-untranslated region is highly homologous to that of pMT49.

A peptide with insulin-like activity was isolated from human plasma. In the purification, insulin-like activity (ILA) was monitored by radioreceptor assay for insulin, using human placental membrane and [125I]-insulin. ILA was extracted from Cohn fraction III with acid-ethanol and chromatographed on Sephadex G-75 in 1% formic acid. When the active fractions were subjected to ion-exchange chromatography with CM-cellulose, the ILA was adsorbed to the column at pH 5.0 and was eluted with a gradient of ammonium acetate. The chromatographic behavior of the ILA was not identical to that of somatomedin A as determined by radioreceptor assay (RRA) for the latter. On isoelectric focusing of the ILA from the CM-cellulose column, insulin-like activity was distributed over a wide pH range. The ILA that was focused at pH 7.5-9.0 was further purified by gel filtration on Sephadex G-50 in 1 M acetic acid. The specific activity of the basic ILA was approximately 200 mU insulin equivalent /mg protein. The apparent molecular weight of the material was estimated to be 7,000. It stimulated [14C]-glucose oxidation in rat epididymal fat cells and had sulfation activity in chick chondrocytes. Furthermore, the basic ILA had a potent mitogenic activity in Balb/c-3T3 cells. Thus, the basic ILA is qualified as an 'insulin-like growth factor' (IGF). It is obviously different from somatomedin A, and may be closely related to IGF-I or somatomedin C. Subsequently, RRA for the basic ILA was developed in serum concentration of the basic ILA were determined. The serum concentrations of the basic ILA were high in acromegalics and low in patients with hypopituitarism. Thus, the basic ILA is entitled to be one of the IGFs.

Receptors specific for lactogenic hormones were solubilized by 1% (v/v) Triton X-100 from the crude particulate membrane fraction of livers of pregnant and non-pregnant female rats and the characteristics of both preparations were compared. Human (125)I-labelled somatotropin was used for binding studies of lactogenic hormone. The solubilized receptor retained most of the characteristics noted in the particulate fraction. The binding of human (125)I-labelled somatotropin to the solubilized receptor is a saturable process, depending on temperature and time. Scatchard analysis of displacement curves revealed similar affinity constants ranging from 1.02 x 10(9) to 1.20 x 10(9) 1/mol, while the binding capacity was 4.5 times greater in the pregnant rat livers than in the non-pregnant female rat livers. The receptors for human (125)I-labelled somatotropin from livers of non-pregnant and pregnant female rats were equally adsorbed onto a concanavalin-A-Sepharose column and were dissociated from the column with alpha-methyl-d-glucoside or alpha-methyl-d-mannoside in the same manner. By gel filtration on Sepharose 6B, however, the molecular sizes of the hepatic receptors were found to be different. The apparent M(r) value was approx. 270000 with a Stokes' radius of 5.5nm in the non-pregnant female rats and approx. 330000 with a Stokes' radius of 5.5nm in the pregnant rats. Furthermore, isoelectric-focusing experiments showed that a major part of the receptor from the non-pregnant female rat livers had a neutral pI (7.0-8.5), whereas that from pregnant-rat livers had an acidic pI (4.2-4.7). These data suggest that the increase in the lactogenic binding capacity in rat liver membranes during pregnancy may be associated with marked changes of the physicochemical properties of the receptors.

Troponin I (TnI) was reacted with a photosensitive heterobifunctional reagent, methyl 4-azidobenzimidate (ABI), and then troponin was reconstituted with the ABI-modified TnI. Flash irradiation of the reconstituted troponin resulted in the formation of cross-links between TnI and other components of troponin, troponin C (TnC) and troponin T (TnT), suggesting that TnI is in contact with TnC and TnT when troponin is free in solution. No effect of calcium on the cross-linking could be detected. When the reconstituted troponin was complexed with F-actin-tropomyosin, flash irradiation of the reconstituted then filament yielded the cross-linked products of TnC-TnI, TnI-TnT, and TnI-actin in the presence ans absence of calcium, indicating that TnI is in contact with TnC, TnT, and actin in the thin filament complex irrespective of calcium concentration. No cross-linking could be detected between TnI and tropomyosin. Calcium was found to affect the cross-linking of TnC-TnI and TnI-actin; when TnC was saturated with calcium, the extent of the TnC-TnI cross-linking increased, while that of the TnI-actin cross-linking decreased. Calcium did not affect the TnI-TnT cross-linking.

Both somatotrophic and lactogenic binding sites increase markedly in pregnant mouse liver membranes. The regulatory mechanisms of these binding sites involved in pregnancy are studied. The study of the effects of various hormone administration to female mice reveals that oestradiol increases and testosterone decreases lactogenic binding sites, and that other hormones affect neither somatotrophic nor lactogenic binding sites. Hysterectomy, adrenalectomy or foetectomy of pregnant mice cause a remarkable reduction of somatotrophic binding sites to the level observed in non-pregnant or post-partum animals. In contrast, the lactogenic binding sites are not influenced by these treatments. Replacement with corticosterone after adrenalectomy does not prevent the decrease of somatotrophic binding sites. The contents of placental lactogen from both adrenalectomized and foetectomized pregnant mice are similar to those of the normal pregnant group. These observations indicate that the regulatory mechanisms modulating somatotrophic and lactogenic binding activities are quite different and that increased somatotrophic binding sites during pregnancy are supported by the factor(s) generated in the foeto-placental unit and maternal adrenal gland.

We isolated a protein complex containing major cytoskeletal components from the Triton shell of bovine erythrocytes. This protein complex, which we called the 26-S complex, consisted of three major components, spectrin, band-4.1 protein and actin, and one minor component, band-4.9 protein. The molar ratio of spectrin heterodimer:band 4.1:actin was determined by sodium dodecyl sulfate (SDS) gel electrophoresis to be about 1:2:2, approximately the same as that for the Triton shell. By electron microscopic examinations of rotary-shadowed specimens, it was revealed that the 26-S complex had a "spider-like" morphology with a central core and several spectrin heterodimers radiating from it. The number of spectrin arms in the complex was not constant but was in the range between 3 and 6. The complexes with five spectrin heterodimers were the most numerous. The results showed that the 26-S complex contained on the average five spectrin heterodimers, ten band-4.1 polypeptides and ten actin monomers. As judged from the formation of oligomeric 26-S complexes through spectrin arms, the central core of the complex presumably contains band 4.1 and actin. Supporting this conclusion, the central core acted as a nucleus for actin polymerization when the 26-S complex was mixed with G-actin under an actin-polymerizing condition. The 26-S complex could form large aggregates under a certain condition that spectrin was promoted to associate from dimer to tetramer. We conclude that the 26-S complex is the structural unit of the erythrocyte cytoskeleton.
