- Abstract
OBJECTIVE: Leukocyte traffic from the blood to the joints is crucial in the pathogenesis of arthritis. A bifunctional endothelial cell-surface glycoprotein, AOC3 (amine oxidase, copper-containing 3; also known as vascular adhesion protein 1), has both adhesive and enzymatic properties. We undertook this study to determine the contribution of AOC3 and its oxidase activity to leukocyte trafficking into inflamed joints in vivo. METHODS: We used gene-modified animals, molecular modeling, an AOC3 enzyme inhibitor, oxidase assays, and arthritis models (adjuvant-induced arthritis [AIA] in rats and anti-type II collagen antibody-induced arthritis in mice) to dissect the importance of AOC3 in vivo. RESULTS: The AOC3 inhibitor fitted well with a covalent binding mode into the active site of the AOC3 crystal structure. It selectively blocked the oxidase activity of AOC3 in enzyme assays. Intraperitoneal and oral administration of the AOC3 inhibitor significantly ameliorated rat AIA. In anti-type II collagen antibody-induced arthritis in mice, the AOC3 inhibitor also improved the outcome of the joint inflammation. The acute semicarbazide-sensitive amine oxidase blockade by the inhibitor had even more pronounced effects than genetic deletion of AOC3. Enzymatic analyses showed that the inhibitor also blocked 2 other structurally very closely related AOCs, but not any of more than 100 other enzymes tested. CONCLUSION: These are the first data to demonstrate that the enzymatic activity of the atypical endothelial adhesion molecule AOC3, and possibly that of other closely related ecto-oxidases, is crucial for leukocyte exit from the vessels in inflamed joints in vivo. Planarized star-shaped oligothiophenes 1 have been synthesized by connecting short-chain oligothiophenes on a benzo[1,2-b:3,4-b:5,6-b ]trithiophene central core. Their electrochemical and optical properties have been characterized by cyclic voltammetry and UV−visible spectroscopy, respectively. These results associated with theoretical calculations show the advantage of benzotrithiophene as a central core in terms of π-electron delocalization.- Published

- Abstract
The X-ray crystal structures and physical characterizations of the first radical cation salts obtained from the symmetrical S-position isomer of bis(ethylenedithio)tetrathiafulvalene (BEDT-TTF), are presented in this work. The unusual feature of this new π-donor D1, involved in the described salts (D1)3·(ClO4)2, (D1)3·(BF4)2 and D1·Cu(SCN)2, is the presence of two disulfide bridges, allowing weak intermolecular S⋯S interactions. The influence of these outer sulfur atoms on the network of related materials is also discussed in relation to the first physical characterizations which have been carried out.

This paper considers the recent radiometric dating (14C-AMS, TL, ESR) of 76 late Middle and early Upper Paleolithic samples from Ortvale Klde Rockshelter, located in the Republic of Georgia. We present a critical evaluation of each date based on its stratigraphic and archaeological context, its pretreatment and contamination history, and its resulting accuracy and precision, the goal being to establish a sound chronology for the site. Only by systematically identifying aberrant dates within a data set and isolating them from further analysis can we hope to understand cultural and biological phenomena on an accurate temporal scale. Based on the strict discard protocol outlined here, we omit 25% of the dated samples from the analysis. The remaining data speak to the lengthy tenure of Neandertals in the region, but also to their relatively rapid demise and the establishment of modern human populations w38–34 ka 14C BP (42–39 ka cal BPHulu). We compare these chronometric data with those from the neighboring sites of Bronze and Dzudzuana caves, as well as Mezmaiskaya Cave, located in the northern Caucasus. While the lack of key contextual information limit our ability to subject these other data sets to the same critical evaluation procedure, they provide the first interregional temporal assessment of the Middle to Upper Paleolithic ‘‘transition,’’ the results of which suggest an initial expansion of modern humans into the southern Caucasus followed by expansion along the Black Sea coast and into the northern Caucasus., Anthropology

216. Thermoluminescence Dating of a Middle Palaeolithic Occupation at Sodmein Cave, Red Sea Mountains (Egypt)

- Abstract
The Palaeolithic sequence from Sodmein Cave in the Egyptian Eastern Desert, near Quseir, contains seven stratified archaeological levels from the Middle and Upper Palaeolithic. The lowest archaeological level (Middle Palaeolithic level 5) consists of huge fireplaces associated with a lithic assemblage in which Nubian Levallois technology and bifacial tools are present. From this level, six fragments from two large burned flint blocks have been dated by thermoluminescence. A mean age-estimate of 118±8ka for the six dates, all of which fall between 109 and 127ka, puts the use of this hearth at the beginning of Oxygen Isotope Stage 5, most likely during episodes e or d. This age is close to that of other Egyptian Middle Palaeolithic sites with very similar lithic assemblages.

Age-estimates ranging from 50 to 70ky were obtained for the Mousterian deposits of Amud Cave in Israel from thermoluminescence measurements performed on 19 burnt flints. The late dates obtained for the stratigraphic layers bearing hominid remains confirm the evidence for the late presence of Neanderthals in the Levant. The dates enable a more effective comparison of the lithic assemblages from Amud Cave with those of other contemporaneous sites and underline the variability within Mousterian lithic industries at the end of the Middle Palaeolithic in the Levant.

- Abstract
Abstract.: In many reptile species, sexual differentiation of gonads is sensitive to temperature during a critical period of embryonic development (thermosensitive period, TSP). Experiments carried out with different models among which turtles, crocodilians and lizards have demonstrated the implication of estrogens and the key role played by aromatase (the enzyme complex that converts androgens to estrogens) in ovary differentiation during TSP and in maintenance of the ovarian structure after TSP. In some of these experiments, the occurrence of various degrees of gonadal intersexuality is related to weak differences in aromatase activity, suggesting subtle regulations of the aromatase gene at the transcription level. Temperature could intervene in these regulations. Present studies deal with cloning (complementary DNAs) and expression (messenger RNAs) of genes that have been shown, or are expected, to be involved in gonadal formation and/or differentiation in mammals. Preliminary results indicate that homologues of AMH, DAX1, SF1, SOX9 and WT1 genes with the same function(s) as in mammals exist in reptiles. How these genes could interact with aromatase is being examined.

- Abstract
The optical study of the trivalent europium ion in polycrystalline K4Eu2(CO3)3F4is reported. Three main low symmetry sites for Eu3+are deduced from the emission spectra at 4.2 K. Extra lines are also present at low temperature, which may be assigned to a fourth site. When the temperature increases, these extra lines disappear without energy change of other lines. This reversible phenomenon is not yet explained. Above 8 K, site to site energy transfer becomes effective. The electrostatic crystal field effects on the 7F multiplet are also evaluated on the basis of the phenomenological crystal field theory.

- Abstract
Activation of the thrombin receptors, called PAR, leads to a remodeling of the actin cytoskeleton depending on the small G proteins of the Rho family. The main one, RhoA, controls platelet contractility. The Arhgef1 nucleotide exchange factor, responsible for the activation of RhoA, is not well known. Our goal is to identify the role of Arhgef1 in platelet activity and thrombin generation in response to thrombin-activated PAR receptors. Thrombin generation, platelet activation and aggregation are measured by fluorimetry, flow cytometry, and turbidimetry in the blood of Arhgef1+/+ and Arhgef1−/− mice. The formation of a thrombus in the carotid artery in response to the local application of FeCl3, is followed by Echo-Doppler. In response to thrombin, aggregation of washed platelets and intracellular calcium are similar in Arhgef1−/− and Arhgef1+/+ mice. However, the secretion of alpha granules, exposure of phosphatidylserine and release of microparticles associated with platelet activation are decreased compared to Arhgef1+/+ mice. The generation of whole blood thrombin was significantly reduced (25%) in Arhgef1−/− mice compared to Arhgef1+/+. These changes result in an increase in the time of an occlusive thrombus occurrence induced by FeCl3 in the carotid artery. Our results show the involvement of the platelet RhoA pathway in the self-amplification of thrombin generation by a mechanism dependent on Arhgef1 and PAR receptors. Changes in the distribution of membrane phospholipids suggest that this pathway could participate in the deleterious effects of thrombin in atherothrombosis. [ABSTRACT FROM AUTHOR]

Measuring the infrared radiofluorescence (IR-RF) signal is methodologically simpler than measuring the IRSL or TL signals. However, unlike classical luminescence measurements, the IR-RF signal is totally resetting when its highest value is reached. Previous studies (Buylaert et al., 2012; Trautmann et al., 2000) reported that the proper bleaching level of an IR-RF signal is difficult to define and seems to be dependent 1) on the wavelength used for bleaching and 2) on exposure duration. The IR-RF signals from K-feldspar samples have been measured with various bleaching wavelengths and exposure time using a Lexsyg research luminescence unit. In this study, we propose suitable bleaching parameters adapted to IR-RF measurements. [ABSTRACT FROM AUTHOR]

Abstract: Thermoluminescence (TL), which is commonly used to date burnt flints from Palaeolithic sites, sometimes yields ages which are younger than those obtained by other methods. Many reasons could explain such discrepancies and one concerns the evaluation of the dose-rate originating from the sample itself, i.e., the internal dose-rate. In order to refine the parameters used in the internal dose-rate estimation, we investigated the uranium (U) distribution by the induced fission track method in three archaeological flint specimens. This approach shows that the U distribution is roughly uniform on a scale of a millimetre but not on a smaller scale (<100μm), because of the presence of quartz grains whose size is sample-dependent and because of relatively high local U concentrations. Therefore, the assumption of a homogeneous U distribution currently used to estimate the beta and alpha dose-rates may not be fully appropriate for all samples; in this case, this could lead to an underestimate of the TL age. The IRSL dating method is applied to silt-sized alkali feldspar grains (40–60 μm) from Upper and Middle Pleistocene loess deposits of southeastern Romania (sites of Tuzla and Giurgiu Malu-Rosu), using both the multiple aliquot
γ dose technique and the SAR method. All samples show evidence of anomalous fading; the measured IRSL ages are in correct stratigraphic order but systematically underestimate the expected geological age (up to 40%). Three protocols of age correction for the observed fading have been tested: the correction of Huntley and Lamothe (Can. J. Earth Sci. 38 (2001) 1093), the correction of Mejdahl (Quat. Sci. Rev. 7 (1988) 357) and the correction model suggested by Wintle (Quat. Sci. Rev. 9 (1990) 385). The three protocols yield corrected IRSL ages that are in better agreement with the expected ages. This study demonstrates the potential of the IRSL dating method to provide chronological information on Upper and Middle Pleistocene loess deposits of Romania. [Copyright &y& Elsevier]- Published
