202. Actuaciones para la eliminación del tapiz algal presente en los espeleotemas en la rehabilitación de las grutas visitables de La Paz y de La Viña en Ortigosa de Cameros, La Rioja

Abstract

This article reflects what has been done for the study, assessment and cleanup of exogenous algal communities that were deteriorating speleothems in the touristic caves of La Paz and La Viña in Ortigosa de Cameros (La Rioja) and growing in favour of artificial light sources installed for sightseeing. Based on the bibliography review, the possibility of using two general types of treatment in order to remove the underground plant was assessed. On the one hand the use of mechanical methods such as brushing, sanding or grinding or the other, the use of chemical methods, such as hypochlorites, peroxides or biocides. After relevant laboratory studies, the method of dilute hydrogen peroxide as the least harmful to the speleothems and caves fauna was chosen. In situ tests were performed and after observing the results, proceeded to apply the methodology tested with this product to all areas affected by the "lampenflora". The results were entirely satisfactory, removing all non-native flora of the underground speleothems, not producing any damage on the wildlife that inhabits these caves and without leaving any noticeable track for the visitor. The conclusion is that with an economical and friendly method to the subterranean environment, can remove the algal mat that covers the speleothems of many caves adapted for tourist visits owing to the artificial light sources., Este artículo recoge el trabajo llevado a cabo para el estudio, valoración y limpieza de las comunidades algales alóctonas subterráneas que estaban deteriorando los espeleotemas de las cuevas visitables de La Paz y La Viña en Ortigosa de Cameros (La Rioja) y que crecían a favor de los focos de luz artificial instalados para las visitas turísticas. Partiendo de la revisión bibliográfica, se valoró la posibilidad de utilizar dos tipos generales de tratamiento con la finalidad de remover la flora subterránea. Por un lado la utilización de métodos mecánicos tales como cepillado, lijado o desbastado y por otro lado, el uso de métodos químicos, como pueden ser los hipocloritos, peróxidos o biocidas. Tras los estudios de laboratorio pertinentes se eligió el método de peróxido de hidrógeno diluido como el menos perjudicial para los espeleotemas y la fauna cavernícola. Se realizaron ensayos in situ y tras observar los resultados, se procedió a aplicar, con la metodología ensayada, este producto a todas las áreas de las cavidades afectadas por el denominado “mal verde” o “lampenflora”. Los resultados fueron totalmente satisfactorios, removiendo la totalidad de la flora alóctona subterránea de los espeleotemas, no produciendo afección alguna sobre la fauna que habita estas cuevas visitables y sin dejar ningún rastro perceptible por el visitante. La conclusión es que con un método muy económico y amigable para el medio ambiente subterráneo, se puede eliminar el tapiz algal que, a favor de los focos de luz artificial, cubre los espeleotemas de muchas cuevas adaptadas para la visita turística.

Abstract

In agricultural ecosystems, pest insects, pathogens, and reduced soil fertility pose major challenges to crop productivity and are responsible for significant yield losses worldwide. Management of belowground pests and diseases remains particularly challenging due to the complex nature of the soil and the limited reach of conventional agrochemicals. Boosting the presence of beneficial rhizosphere organisms is a potentially sustainable alternative and may help to optimize crop health and productivity. Field application of single beneficial soil organisms has shown satisfactory results under optimal conditions. This might be further enhanced by combining multiple beneficial soil organisms, but this remains poorly investigated. Here, we inoculated wheat plots with combinations of three beneficial soil organisms that have different rhizosphere functions and studied their effects on crop performance. Plant beneficial Pseudomonas bacteria, arbuscular mycorrhizal fungi (AMF), and entomopathogenic nematodes (EPN), were inoculated individually or in combinations at seeding, and their effects on plant performance were evaluated throughout the season. We used traditional and molecular identification tools to monitor their persistence over the cropping season in augmented and control treatments, and to estimate the possible displacement of native populations. In three separate trials, beneficial soil organisms were successfully introduced into the native populations and readily survived the field conditions. Various Pseudomonas, mycorrhiza, and nematode treatments improved plant health and productivity, while their combinations provided no significant additive or synergistic benefits compared to when applied alone. EPN application temporarily displaced some of the native EPN, but had no significant long-term effect on the associated food web. 212. First molecular detection of Histoplasma capsulatum in a dog with an atypical cutaneous clinical presentation in Colombia.

Abstract

Anamnesis: a 4-year-old female dog‚ Pinscher breed‚ was presented to a small animal clinic in Colombia with a 2-month evolution of a skin and pruritic problem‚ alopecic skin lesions on the trunk and lesions with ulcers and crusts on the tips of the ears with serosanguineous discharge. Owner reported a previous treatment for scabies without a favorable response. Clinical findings: the canine patient presented round multifocal skin lesions on the trunk consistent with epidermal collarettes‚ as well as lesions with ulcers and crusts on the tips of the ears with serosanguinous discharge‚ consistent with ear tip vasculitis. The general examination of the patient was normal. Diagnostic: cutaneous histoplasmosis due to Histoplasma capsulatum was confirmed by cytology‚ histopathological analysis of skin biopsy that revealed structures consistent with fungal forms‚ Gomori-Grocott methenamine silver staining. For molecular diagnosis by nested PCR‚ DNA was extracted from skin and serum by E.Z.N.A.® Tissue DNA kit‚ and primers were designed to amplify 111 bp and 279 bp fragments from M antigen of H. capsulatum as a target. Amplicons were sequenced and confirmed as H. capsulatum by Sanger method. Treatment: after the diagnosis‚ the patient did not return for consultation‚ however‚ the treatment for histoplasmosis typically involves the use of an azole antifungal drug (e.g., ketoconazole‚ itraconazole‚ fluconazole‚ or posaconazole) which inhibits sterol 14α-demethylase‚ involved in ergosterol synthesis‚ a key component of the fungal cell wall. Results and conclusions: the diagnosis of cutaneous histoplasmosis in dogs can be difficult due to the resemblance of the intracellular structures to those of other pathogens. Therefore‚ it is important to confirm the diagnosis using complementary tests such as histopathology and PCR. To the authors’ knowledge‚ this is the first report of molecular detection of H. capsulatum as the causative agent of skin lesions in a canine patient in Colombia‚ emphasizing the need to consider this pathogen as a differential diagnosis in dogs with epidermal collarette-type lesions and/or lesions compatible with ear tip vasculitis. Abstract

The infectious bursal disease virus (IBDV) is a major health threat to the world’s poultry industry despite intensive controls including proper biosafety practices and vaccination. IBDV (Avibirnavirus, Birnaviridae) is a non-enveloped virus with a bisegmented double-stranded RNA genome. The virus is traditionally classified into classic, variant and very virulent strains, each with different epidemiological relevance and clinical implications. Recently, a novel worldwide spread genetic lineage was described and denoted as distinct (d) IBDV. Here, we report the development and validation of a reverse transcription-quantitative polymerase chain reaction (RT-qPCR) assay for the specific detection of dIBDVs in the global poultry industry. The assay employs a TaqMan-MGB probe that hybridizes with a unique molecular signature of dIBDV. The assay successfully detected all the assessed strains belonging to the dIBDV genetic lineage, showing high specificity and absence of cross-reactivity with non-dIBDVs, IBDV-negative samples and other common avian viruses. Using serial dilutions ofin vitro-transcribed RNA we obtained acceptable PCR efficiencies and determination coefficients, and relatively small intra- and inter-assay variability. The assay demonstrated a wide dynamic range between 103and 108RNA copies/reaction. This rapid, specific and quantitative assay is expected to improve IBDV surveillance and control worldwide and to increase our understanding of the molecular epidemiology of this economically detrimental poultry pathogen. Abstract

Los ensayos de toxicidad son pruebas diseñadas para evaluar la potencia relativa de un agente o una sustancia contaminante sobre una o más especies de organismos de los sistemas biológicos acuáticos y terrestres, al tiempo que permiten caracterizar la relación concentración-respuesta entre el agente contaminante y el organismo sometido a prueba. La toxicidad de una sustancia es la propiedad que tiene de causar daños a la salud humana o la muerte de un organismo vivo, mediante la comparación de efecto del agente con el de una solución patrón o estándar.Los resultados de las pruebas de toxicidad en sistemas acuáticos constituyen un criterio importante para determinar el efecto y el riesgo de la descarga de sustancias contaminantes sobre los ecosistemas acuáticos, así como para establecer valores límites permisibles para el control de sustancias tóxicas que son incorporadas a los ecosistemas acuáticos.Con la euge de la ecotoxicología ciencia que predice de los agentes potencialmente tóxicos en los ecosistemas naturales (Hoffman, 1995) se ha reforzado la idea de realizar monitoreos y pruebas de toxicidad para la detección, predicción y control de los efectos de diferentes tipos de sustancias sobre los ecosistemas, en particular, el acuático.

Abstract

We analyzed the main karyologic changes that have occurred during the dispersion of Triatoma infestans, the main vector of Chagas disease. We identified two allopatric groups, named Andean and non-Andean. The Andean specimens present C-heterochromatic blocks in most of their 22 chromosomes, whereas non-Andean specimens have only 4-7 autosomes with C-banding. These heterochromatin differences are the likely cause of a striking DNA content variation (approximately 30%) between Andean and non-Andean insects. Our study, together with previous historical and genetic data, suggests that T. infestans was originally a sylvatic species, with large quantities of DNA and heterochromatin, inhabiting the Andean region of Bolivia. However, the spread of domestic T. infestans throughout the non-Andean regions only involved insects with an important reduction of heterochromatin and DNA amounts. We propose that heterochromatin and DNA variation mainly reflected adaptive genomic changes that contribute to the ability of T. infestans to survive, reproduce, and disperse in different environments. Abstract

Nowadays, the disability approach is a perspective that proposes a way to deal with the incorporation of social dimension in mental health understanding. Its hegemony in Chile turns visible considering the design of mental health laws and public policies over the past 20 years. However, this approach to mental health as well as the legislations and policies developed and related to it, hardly take over the situation of justice and citizenship of this group of people. These difficulties are particularly relevant when the exercise of active citizenship is required, since citizenship has been proved to show a therapeutic effect on the deficiencies or mental disorders. The urgent need of a mental health law for Chile demands the challenge of designing a law which recognizes the particularities of mental disability, but it also offers the historical possibility to effectively include the perspective of people with mental disabilities and their families in the discussion for a new law.
