This article presents a comparison between the performance obtained by using a spatial discretization of the Euler equations based on a high-order discontinuous Galerkin (dG) method and different sets of variables. The sets of variables investigated are as follows: (1) conservative variables; (2) primitive variables based on pressure and temperature; (3) primitive variables based on the logarithms of pressure and temperature. The solution is advanced in time by using a linearly implicit high-order Rosenbrock-type scheme. The results obtained using the different sets are assessed across several canonical unsteady test cases, focusing on the accuracy, conservation properties and robustness of each discretization. In order to cover a wide range of physical flow conditions, the test-cases considered here are (1) the isentropic vortex convection, (2) the Kelvin–Helmholtz instability and (3) the Richtmyer–Meshkov instability.

Copper is efficient, has a high conductivity (5.8 × 107 S/m), and is cost-effective. The use of copper-based catalysts is promising for the electrocatalytic reduction of nitrates. This work aims to grow and characterize copper micro-crystals on Screen-Printed Electrodes (SPEs) for NO3− reduction in water. Copper micro-crystals were grown by cyclic voltammetry. Different cycles (2, 5, 7, 10, 12, 15) of copper electrodeposition were investigated (potential ranges from −1.0 V to 0.0 V, scan rate of 0.1 V s−1). Electrodeposition generated different morphologies of copper crystals on the electrodes, as a function of the number of cycles, with various performances. The presence of numerous edges and defects in the copper micro-crystal structures creates highly reactive active sites, thus favoring nitrate reduction. The manufactured material can be successfully employed for environmental applications.

207. Characterization of Electric Field Fluctuations in the High-Latitude Ionosphere Using a Dynamical Systems Approach: CSES-01 Observations

We present an analysis of the ionospheric electric field dynamics at high latitudes during periods of quiet and disturbed geomagnetic activity by exploiting recent advancements in dynamical systems and extreme value theory. Specifically, we employed two key indicators: the instantaneous dimension d, which evaluates the degrees of freedom within the system, and the extremal index θ, which quantifies the system’s persistence in a given state. Electric field measurements were obtained from the CSES-01 satellite at mid- and high latitudes in the Southern Hemisphere. Our analysis revealed that the instantaneous dimension increases upon crossing specific ionospheric regions corresponding to the auroral oval boundaries. Outside these regions, the instantaneous dimension fluctuates around the state-space dimension, suggesting an ergodic nature of the system. As geomagnetic activity intensifies, differences in the properties of various ionospheric regions persist, albeit with an increased system instability characterized by higher θ values, thus indicating the externally driven nature of the electric field response to geomagnetic activity. This study provides new insights into the spatial and temporal variability of electric field fluctuations in the ionosphere, highlighting the complex interplay between geomagnetic conditions and ionospheric dynamics.

This study presents the first column-averaged dry-air mole fractions of carbon dioxide (XCO2), methane (XCH4) and carbon monoxide (XCO) in the coal mine aggregation area in Shanxi, China, using two portable Fourier transform infrared spectrometers (EM27/SUNs), in the framework of the Collaborative Carbon Column Observing Network (COCCON). The measurements, collected over two months, were analyzed. Significant daily variations were observed, particularly in XCH4, which highlight the impact of coal mining emissions as a major CH4 source in the region. This study also compares COCCON XCO with measurements from the TROPOspheric Monitoring Instrument (TROPOMI) onboard the Sentinel-5P satellite, revealing good agreement, with a mean bias of 7.15 ± 9.49 ppb. Additionally, comparisons were made between COCCON XCO2 and XCH4 data and analytical data from the Copernicus Atmosphere Monitoring Service (CAMS). The mean biases between COCCON and CAMS were −6.43 ± 1.75 ppm for XCO2 and 15.40 ± 31.60 ppb for XCH4. The findings affirm the stability and accuracy of the COCCON instruments for validating satellite observations and detecting local greenhouse gas sources. Operating COCCON spectrometers in coal mining areas offers valuable insights into emissions from these high-impact sources.

In this review, we tentatively tried to connect the most recent findings on the bat microbiome and to investigate on their microbial communities, that may vary even in conspecific hosts and are influenced by host physiology, feeding behavior and diet, social interactions, but also by habitat diversity and climate change. From a conservation perspective, understanding the potentially negative and indirect effects of habitat destruction on animal microbiota can also play a crucial role in the conservation and management of the host itself. According to the One Health concept, which recognizes an interdependence between humans, animals, and the environment, bat microbiota represents an indicator of host and environmental health, besides allowing for evaluation of the risk of emerging infectious diseases. We noticed that a growing number of studies suggest that animal microbiota may respond in various ways to changes in land use, particularly when such changes lead to altered or deficient food resources. We have highlighted that the current literature is strongly focused on the initial phase of investigating the microbial communities found in Chiroptera from various habitats. However, there are gaps in effectively assessing the impacts of pathogens and microbial communities in general in animal conservation, veterinary, and public health. A deeper understanding of bat microbiomes is paramount to the implementation of correct habitat and host management and to the development of effective surveillance protocols worldwide.

Turbulent flows present rich dynamics originating from non-trivial energy fluxes across scales, non-stationary forcings and geometrical constraints. This complexity manifests in non-hyperbolic chaos, randomness, state-dependent persistence and unpredictability. All these features have prevented a full characterization of the underlying turbulent (stochastic) attractor, which will be the key object to unpin this complexity. Here we propose a novel formalism to trace the evolution of the structural characteristics of phase-space trajectories across scales, providing a full characterization of the attractor. We demonstrate that the properties of the dynamically invariant objects depend on the scale we are focusing on. In the case of laboratory experiments on fluids we observe the emergence of an intrinsic timescale, solely determined by nonlinear interactions, controlling the geometrical and topological properties of phase-space trajectories. First person action recognition is becoming an increasingly researched area thanks to the rising popularity of wearable cameras. This is bringing to light cross-domain issues that are yet to be addressed in this context. Indeed, the information extracted from learned representations suffers from an intrinsic "environmental bias". This strongly affects the ability to generalize to unseen scenarios, limiting the application of current methods to real settings where labeled data are not available during training. In this work, we introduce the first domain generalization approach for egocentric activity recognition, by proposing a new audio-visual loss, called Relative Norm Alignment loss. It re-balances the contributions from the two modalities during training, over different domains, by aligning their feature norm representations. Our approach leads to strong results in domain generalization on both EPIC-Kitchens-55 and EPIC-Kitchens-100, as demonstrated by extensive experiments, and can be extended to work also on domain adaptation settings with competitive results., Comment: Accepted at WACV 2022. arXiv admin note: substantial text overlap with arXiv:2106.01689

Abstract Urban evolutionary ecology is inherently interdisciplinary. Moreover, it is a field with global significance. However, bringing researchers and resources together across fields and countries is challenging. Therefore, an online collaborative research hub, where common methods and best practices are shared among scientists from diverse geographic, ethnic, and career backgrounds would make research focused on urban evolutionary ecology more inclusive. Here, we describe a freely available online research hub for toolkits that facilitate global research in urban evolutionary ecology. We provide rationales and descriptions of toolkits for: (1) decolonizing urban evolutionary ecology; (2) identifying and fostering international collaborative partnerships; (3) common methods and freely‐available datasets for trait mapping across cities; (4) common methods and freely‐available datasets for cross‐city evolutionary ecology experiments; and (5) best practices and freely available resources for public outreach and communication of research findings in urban evolutionary ecology. We outline how the toolkits can be accessed, archived, and modified over time in order to sustain long‐term global research that will advance our understanding of urban evolutionary ecology.

218. Enabling People With Intellectual and Sensory Disabilities to Trigger a Tablet’s Delivery of Task Instructions by Walking to the Tablet: Proof-of-Concept Study

BackgroundPeople with intellectual and sensory or sensory-motor disabilities tend to have problems performing multistep tasks. To alleviate their problems, technological solutions have been developed that provide task-step instructions. Instructions are generally delivered at people’s request (eg, as they touch an area of a computer or tablet screen) or automatically, at preset intervals. ObjectiveThis study carried out a preliminary assessment of a new tablet-based technology system that presented task-step instructions when participants with intellectual and sensory disabilities walked close to the tablet (ie, did not require participants to perform fine motor responses on the tablet screen). MethodsThe system entailed a tablet and a wireless camera and was programmed to present instructions when participants approached the tablet, that is, when the camera positioned in front of the tablet detected them. Two instructions were available for each task step. One instruction concerned the object(s) that the participants were to collect, and the other instruction concerned the “where” and “how” the object(s) collected would need to be used. For 3 of the six participants, the two instructions were presented in succession, with the second instruction presented once the required object(s) had been collected. For the other 3 participants, the two instructions were presented simultaneously. Instructions consisted of pictorial representations combined with brief verbal phrases. The impact of the system was assessed for each of the 2 groups of participants using a nonconcurrent multiple baseline design across individuals. ResultsAll participants were successful in using the system. Their mean frequency of correct task steps was close to or above 11.5 for tasks including 12 steps. Their level of correct performance tended to be much lower during the baseline phase when they were to receive the task-step instructions from a regular tablet through scrolling responses. ConclusionsThe findings, which need to be interpreted with caution given the preliminary nature of the study, suggest that the new tablet-based technology system might be useful for helping people with intellectual and sensory disabilities perform multistep tasks.

ABSTRACT: Rabies is an invariably fatal infectious-contagious viral disease caused by viruses in the genus Lyssavirus, which affects the central nervous system of domestic and wild mammals. This study draws attention to the importance of insectivorous bats and domestic cats in the epidemiology of rabies. For this, two cases of rabies registered in domestic cats in the southeast region of Rio Grande do Sul were analyzed. Diagnoses were based on histological alterations and positive staining for the virus in direct immunofluorescence with confirmation by biological test, immunohistochemical evaluation and identification of the rabies virus variant by polymerase chain reaction. In Brazil, a change in the epidemiological profile of rabies has been observed, in which bats play a major role in the current chain of transmission of the disease. It is noteworthy, that the antigenic viral variant AgV-4, typical of the insectivorous bat Tadarida brasiliensis, was identified in the domestic cats affected in this study.

220. Effect of grain size on defect annealing in displacement-damaged tungsten

Pulsed laser deposition and subsequent thermal treatment were used to create compact tungsten layers with various grain sizes (1μm, 100 nm, and 10 nm). The layers were self-damaged at room temperature up to 0.23 dpa using 20.3 MeV W ions and annealed in vacuum at 473 K or 673 K prior to exposure to deuterium plasma to populate the surviving displacement defects. Thermal desorption spectroscopy was used to measure deuterium retention and study the desorption profile. Rate-equation modeling of the desorption spectra was performed to study the behavior of individual traps at both annealing temperatures. After annealing the 1μm and 100 nm samples at 473 K, the deuterium retention was found to decrease by 24% and 9%, respectively, whereas the total deuterium retention in the 10 nm sample did not change. Annealing at 673 K reduced total deuterium retention by additional 27%, 14%, and 34% in the case of the samples with a grain size of 1μm, 100 nm, and 10 nm, respectively.

Allelic variation at the Ptprc gene, which encodes the pan-leukocyte marker CD45/Ly5, is commonly exploited to track hematopoietic reconstitution by flow cytometry in mixed bone marrow chimera transplant experiments. Historically, this was accomplished using bone marrow from C57BL/6 (Ptprcb/CD45.2/Ly5.2) and congenic B6.SJL-PtprcaPepcb/Boy (Ptprca/CD45.1/Ly5.1) mice. Recently, the Jackson Laboratory directly CRISPR-engineered the Ptprca allele in C57BL/6J mice. This new isogenic strain, termed JAXBoy, differs from wild-type C57BL/6J mice by two nucleotides, compared to the biologically significant 37 megabase (Mb) SJL interval retained in B6.SJL-PtprcaPepcb/Boy/J mice. Currently, Ptprc/CD45 variants are identified by flow cytometry or allele-specific real-time PCR, both of which require specialized workflows and equipment compared to standard genotyping of endpoint PCR products by gel electrophoresis. Here, we employed allele-specific oligonucleotides in conjunction with differential incorporation of a long non-specific oligo 5′-tail to allow for simultaneous identification of the Ptprca and Ptprcb alleles using endpoint PCR and gel electrophoresis. This method allows for integration of Ptprc genotyping into standard genotyping workflows, which use a single set of thermocycling and gel electrophoresis conditions. Importantly, the strategy of primer placement and tail addition described here can be adapted to discriminate similar single- or multi-nucleotide polymorphisms at other genomic loci.

The paper describes the development and application of a screen-printed electrode cell with a graphite-ink working electrode modified by a molecularly imprinted electropolymerized polypyrrole for the voltammetric determination of the herbicide 4‑chloro-2-methylphenoxyacetic acid (MCPA). The method exploits the direct measurement of the analyte by applying the differential pulse voltammetry (DPV) technique, taking advantage of the irreversible oxidation peak at about +1.0 V vs. Ag/AgCl pseudo reference electrode. The presence of the molecularly imprinted polypyrrole enhances the sensor's selectivity and sensitivity. A chemometric approach has been crucial for quantitative analysis because of the peak's broad and not well-defined shape. Firstly, a proper pretreatment of the voltammetric signals is identified, proving the most effective is the first-derivative function transformation of the signal. The Partial Least Square regression (PLS) is the tool applied for MCPA quantification. A preliminary PLS model has been developed and validated in dihydrogen phosphate solution at pH 5.5, aiming to optimize the data treatment approach. Then, the same approach is used to develop a PLS model analyzing tap water samples fortified with MCPA and other pesticides as possible interferents to simulate contaminated natural waters. The model correctly predicted the analyte concentration in the range of 2.5–75 μM, assuring the reliability and robustness of the sensor for the possible quantification of MCPA in wastewater samples.

Each year thousands of babies are born with rare genetic disorders not identified by current NBS panels, due to programs which are not yet optimal. Next-generation sequencing technologies have the potential to overcome many NBS drawbacks and provide large amounts of molecular data, broadening the number of diseases investigated. Here, we design and set up an NGS-based approach to evaluate the feasibility of NGS from dried blood spot starting from 34 DBSs.After assessing gDNA yield and integrity, libraries were performed using three target enrichment approaches, sequenced on NS500 platform, and analyzed on commercial platform. Specifically, we focus on virtual gene panels related to highly actionable neonatal/pediatric disorders.WES show that amount and quality of DBS-extracted gDNA are suitable for high-throughput sequencing. We obtain 500–1500 ng for each specimen, 1.7–1.8 260/280 wavelength, and DIN of 7 resulting DNA integrity, on par with traditional venous blood collection. A high read depth with 94.3% coverage uniformity is achieved for all samples.Data results on mean coverage are comparable among the different workflows tested and demonstrate that DBS from newborn collected at birth is a suitable material for the developing of gNBS programs.

ABSTRACT: A retrospective study of skin diseases diagnosed in cattle from 2000 to 2022 was performed at the “Laboratório Regional de Diagnóstico” of the “Faculdade de Veterinária” of the “Universidade Federal de Pelotas” (LRD-UFPel) to determine the main skin diseases that affect cattle in southern Brazil. Information regarding epidemiological data, lesions and diagnosis was collected. The diseases were grouped into infectious/inflammatory, toxic, nonneoplastic proliferative and neoplastic. The macroscopic aspects, distribution, histopathology, and evolution of the lesions were obtained in the protocols, and the original diagnosis was preserved. The main skin diseases diagnosed during this period occurred during outbreaks. They were infectious, with an emphasis on dermatophilosis, papillomatosis and dermatophytosis, followed by hepatogenous photosensitization, which was included in the group of toxic diseases. Among the neoplasms, squamous cell carcinoma stood out, occurring in isolated cases, reflecting an irrelevant cause of economic losses.

225. Genetic determinants of serum bilirubin using inferred native American gene variants in Chilean adolescents

Gene variants in the UGT1A1 gene are strongly associated with circulating bilirubin levels in several populations, as well as other variants of modest effect across the genome. However, the effects of such variants are unknown regarding the Native American ancestry of the admixed Latino population. Our objective was to assess the Native American genetic determinants of serum bilirubin in Chilean admixed adolescents using the local ancestry deconvolution approach. We measured total serum bilirubin levels in 707 adolescents of the Chilean Growth and Obesity Cohort Study (GOCS) and performed high-density genotyping using the Illumina-MEGA array (>1.7 million genotypes). We constructed a local ancestry reference panel with participants from the 1000 Genomes Project, the Human Genome Diversity Project, and our GOCS cohort. Then, we inferred and isolated haplotype tracts of Native American, European, or African origin to perform genome-wide association studies. In the whole cohort, the rs887829 variant and others near UGT1A1 were the unique signals achieving genome-wide statistical significance (b = 0.30; p = 3.34 × 10−57). After applying deconvolution methods, we found that significance is also maintained in Native American (b = 0.35; p = 3.29 × 10−17) and European (b = 0.28; p = 1.14 × 10−23) ancestry components. The rs887829 variant explained a higher percentage of the variance of bilirubin in the Native American (37.6%) compared to European ancestry (28.4%). In Native American ancestry, carriers of the TT genotype of this variant averaged 4-fold higher bilirubinemia compared to the CC genotype (p = 2.82 × 10−12). We showed for the first time that UGT1A1 variants are the primary determinant of bilirubin levels in Native American ancestry, confirming its pan-ethnic relevance. Our study illustrates the general value of the local ancestry deconvolution approach to assessing isolated ancestry effects in admixed populations.

Abstract The objective of this work was to identify the most adapted and stable genotypes for dry mass and seed yield using the genotype plus environment interaction (GGE) biplot technique. The experiment was conducted in the municipality of Frederico Westphalen, in the state of Rio Grande do Sul, Brazil, in 2018, 2019, 2020, and 2021, using five lines and two cultivars of black oat, in a randomized complete block experimental design, with three replicates. The adaptability and stability of the genotypes, as well as their behavior in the environments, were evaluated using the GGE biplot. The seed yield of black oat genotypes is highly influenced by the environmental conditions over the four studied cultivation years. The UFSMFW 2-07 genotype stands out, but shows instability. The UFSMFW 2-07 and 'UPFA 21-Moreninha' genotypes show adaptability in specific years. Mega environments are formed for the dry mass trait in 2018 and 2021 and seed yield in 2018, 2020, and 2021, whereas, 2019 is considered the year for genotype discrimination.

Background: COVID-19 is primarily known as a respiratory illness; however, many patients present to hospital without respiratory symptoms. The association between non-respiratory presentations of COVID-19 and outcomes remains unclear. We investigated risk factors and clinical outcomes in patients with no respiratory symptoms (NRS) and respiratory symptoms (RS) at hospital admission. Methods: This study describes clinical features, physiological parameters, and outcomes of hospitalised COVID-19 patients, stratified by the presence or absence of respiratory symptoms at hospital admission. RS patients had one or more of: cough, shortness of breath, sore throat, runny nose or wheezing; while NRS patients did not. Results: Of 178,640 patients in the study, 86.4 % presented with RS, while 13.6 % had NRS. NRS patients were older (median age: NRS: 74 vs RS: 65) and less likely to be admitted to the ICU (NRS: 36.7 % vs RS: 37.5 %). NRS patients had a higher crude in-hospital case-fatality ratio (NRS 41.1 % vs. RS 32.0 %), but a lower risk of death after adjusting for confounders (HR 0.88 [0.83–0.93]). Conclusion: Approximately one in seven COVID-19 patients presented at hospital admission without respiratory symptoms. These patients were older, had lower ICU admission rates, and had a lower risk of in-hospital mortality after adjusting for confounders.

Rare subtypes of cutaneous T-cell lymphomas (CTCL) include four entities, primary cutaneous γδ T-cell lymphoma, primary cutaneous CD8+ aggressive epidermotropic cytotoxic T-cell lymphoma, and primary cutaneous CD4+ small/medium T-cell lymphoproliferative disorders, primary cutaneous acral CD8+ T-cell lymphoma, which were previously considered provisional and are now included in the new 5th World Health Organization classification of hematolymphoid tumors as distinct entities. An updated summary of the clinical, histological, and genomic characteristics of these uncommon CTCL subtypes is given in this review, with a focus on the growing body of knowledge regarding their classification and possible treatment strategies.

Introduction Breast milk is a dynamic type of nourishment that changes based on the needs of the child. An increasing amount of data suggests that mental health may be an important factor in such modulation. In addition, breast milk contains extracellular vesicles (EVs), which are currently considered an important dynamic system of communication between cells, even of different individuals. Objectives Purpose of this article is to investigate whether changes in breast milk in terms of EVs concentrations are related to maternal mental health. Methods This is a case-control study for which we enrolled mothers of infants with bronchiolitis (N=33) and mothers of healthy infants (N=13). Breast milk samples were taken and EVs concentrations were quantified. Maternal mental health was assessed by administration of five different psychometric scales: Edinburgh Postnatal Depression Scale (EPDS), State Trait Anxiety Inventory (STAI-S, STAI-T), Barkin Index of Maternal Functioning (BIMF), The Connor-Davidson Resilience Scale 10 items (CD-RISC). Subsequently, scale scores were related to evs concentrations by negative binomial regressions adjusted for case-control. Results As maternal resilience increases, the EVs of neutrophilic origin (p=0.0447) and those of endothelial origin (p=0.0078) decrease¹. In contrast, an increased EPDS score is associated with higher levels of B-lymphocyte EVs (p=0.0376). Mutations in the gene encoding MFN2 have been identified as associated with Charcot–Marie–Tooth disease type 2A (CMT2A), a neurological disorder characterized by a broad clinical phenotype involving the entire nervous system. MFN2, a dynamin-like GTPase protein located on the outer mitochondrial membrane, is well-known for its involvement in mitochondrial fusion. Numerous studies have demonstrated its participation in a network crucial for various other mitochondrial functions, including mitophagy, axonal transport, and its controversial role in endoplasmic reticulum (ER)-mitochondria contacts. Considerable progress has been made in the last three decades in elucidating the disease pathogenesis, aided by the generation of animal and cellular models that have been instrumental in studying disease physiology. A review of the literature reveals that, up to now, no definitive pharmacological treatment for any CMT2A variant has been established; nonetheless, recent years have witnessed substantial progress. Many treatment approaches, especially concerning molecular therapy, such as histone deacetylase inhibitors, peptide therapy to increase mitochondrial fusion, the new therapeutic strategies based on MF1/MF2 balance, and SARM1 inhibitors, are currently in preclinical testing. The literature on gene silencing and gene replacement therapies is still limited, except for a recent study by Rizzo et al.(Rizzo et al., 2023), which recently first achieved encouraging results in in vitro and in vivo models of the disease. The near-future goal for these promising therapies is to progress to the stage of clinical translation.

IntroductionAccording to various censuses, Europe has less than 1.5 million ha of old-growth forests (OGF). Most of them are in the boreal zone, while their presence in the temperate zone is residual and fragmented.In the framework of the EU biodiversity strategy, it has been adopted a broad definition of OGF which includes late-seral forests and forests with some management legacies. However, research purposes need to identify strictly defined OGFs characterized by structure, disturbance history, and processes typical and exclusive of the last stage of the forest dynamic.MethodsThe present paper wants to contribute to this debate by presenting a research network of four mixed (Fagus-Abies-Picea) montane OGFs in the Dinaric Alps (Lom, BiH; Janj, BiH; Perućica, BiH; Biogradska Gora, MNE), summarizing 20 years of multidisciplinary research by focusing on the structural characteristics and the disturbance history of the whole network and their coherency with strict OGF indicators. These sites were selected in relatively structurally uniform study areas, where 142 permanent plots have been established since 2002.Results and discussionThe study sites have a high living (747–1,201 m3 ha−1) and coarse woody debris (CWD) biomass (304–410 m3 ha−1), resulting in the highest forest carbon sink at the continental level (398–484 Mg C ha−1). The presence of large and old trees is one of the critical characteristics of the old-growth stage: in Lom and Perućica, there are 19 trees and 14 ha−1 larger than 1 m at breast height, respectively, and 14 trees and 15 trees ha−1 older than 400 years. In the last three centuries, continuous small-scale disturbances have driven forest dynamics, developing stands characterized by gap-phase dynamics and quasi-equilibrium structure. The Dinaric OGF network presents robust indicators of old-growthness, similar structural characteristics, and dynamic processes across all four sites. Identifying this sub-set of OGF using strict criteria is critical for recognizing conservation priorities and for quantifying, along an old-growthness chronosequence, the current structural differences of managed or recently abandoned forests. Besides, only OGF selected with rigorous criteria can act as a reliable reference for ecological restoration and sustainable forest management as a benchmark for carbon sink and for quantifying the impact of climate change on forests.

Summary: Gamma-aminobutyric acid (GABA) is the main inhibitory neurotransmitter in adults. Depolarizing GABA responses have been well characterized at neuronal-population average level during typical neurodevelopment and partially in brain disorders. However, no investigation has specifically assessed whether a mosaicism of cells with either depolarizing or hyperpolarizing/inhibitory GABAergic responses exists in animals in health/disease at diverse developmental stages, including adulthood. Here, we showed that such mosaicism is present in wild-type (WT) and down syndrome (DS) neuronal networks, as assessed at increasing scales of complexity (cultures, brain slices, behaving mice). Nevertheless, WT mice presented a much lower percentage of cells with depolarizing GABA than DS mice. Restoring the mosaicism of hyperpolarizing and depolarizing GABA-responding neurons to WT levels rescued anxiety behavior in DS mice. Moreover, we found heterogeneous GABAergic responses in developed control and trisomic human induced-pluripotent-stem-cells-derived neurons. Thus, a heterogeneous subpopulation of GABA-responding cells exists in physiological/pathological conditions in mouse and human neurons, possibly contributing to disease-associated behaviors.

The photoinduced ultrafast coherent inter-chromophore energy redistribution in a triarylamine trimer is explored using nonadiabatic excited state molecular dynamics followed by simulations of X-ray Raman signals. The nitrogencentered system ensures strong interchromophore interactions and, thus, the presence of coherences. Nevertheless, the multitude of non-deterministic photoinduced pathways during the ultrafast inter-branch migration of the excitation results in random confinement on some branches and, therefore, spatial exciton scrambling and loss of phase information at long times. We show that the vibronic coherence dynamics evolving into the incoherent scrambling mechanism on ultrafast 50 fs timescale, is accurately probed by the TRUECARS X-ray stimulated Raman signal. In combination with previous results, where the technique has revealed long-lived coherences in a rigid heterodimer, the signal is most valuable for detecting ultrafast molecular coherences or their absence. We demonstrate that X-ray Raman spectroscopy is a useful tool in the chemical design of functional molecular building blocks.

Fifth-generation (5G) systems are designed to enable convergent access-agnostic service availability. This means that 5G services will be available over 5G New Radio air interface and also through other non-Third Generation Partnership Project (3GPP) access networks, e.g., IEEE 802.11 (Wi-Fi). 3GPP has recently published the Release 16 that includes trusted non-3GPP access network concept and wireless wireline convergence. The main goal of this tutorial is to present an overview of access to 5G core via non-3GPP access networks specified by 3GPP until Release 16 (i.e., untrusted, trusted, and wireline access). The tutorial describes aspects of the convergence of a 5G system and these non-3GPP access networks, such as the authentication and authorization procedures and the data session establishment from the point of view of protocol stack and exchanged messages between the network functions. In order to illustrate several concepts and part of 3GPP specification, we present a basic but fully operational implementation of untrusted non-3GPP access using WLAN. We perform experiments that demonstrate how a Wi-Fi user is authorized in a 5G core and establishes user plane connectivity to a data network. In this report, we describe the technical details of our submission to the EPIC-Kitchens-100 Unsupervised Domain Adaptation (UDA) Challenge in Action Recognition. To tackle the domain-shift which exists under the UDA setting, we first exploited a recent Domain Generalization (DG) technique, called Relative Norm Alignment (RNA). It consists in designing a model able to generalize well to any unseen domain, regardless of the possibility to access target data at training time. Then, in a second phase, we extended the approach to work on unlabelled target data, allowing the model to adapt to the target distribution in an unsupervised fashion. For this purpose, we included in our framework existing UDA algorithms, such as Temporal Attentive Adversarial Adaptation Network (TA3N), jointly with new multi-stream consistency losses, namely Temporal Hard Norm Alignment (T-HNA) and Min-Entropy Consistency (MEC). Abstract Coastal vegetated ecosystems are acknowledged for their capacity to sequester organic carbon (OC), known as blue C. Yet, blue C global accounting is incomplete, with major gaps in southern hemisphere data. It also shows a large variability suggesting that the interaction between environmental and biological drivers is important at the local scale. In southwest Atlantic salt marshes, to account for the space occupied by crab burrows, it is key to avoid overestimates. Here we found that southern southwest Atlantic salt marshes store on average 42.43 (SE = 27.56) Mg OC·ha−1 (40.74 (SE = 2.7) in belowground) and bury in average 47.62 g OC·m−2·yr−1 (ranging from 7.38 to 204.21). Accretion rates, granulometry, plant species and burrowing crabs were identified as the main factors in determining belowground OC stocks. These data lead to an updated global estimation for stocks in salt marshes of 185.89 Mg OC·ha−1 (n = 743; SE = 4.92) and a C burial rate of 199.61 g OC·m−2·yr−1 (n = 193; SE = 16.04), which are lower than previous estimates.

Abstract There are several overlapping clinical practice guidelines in acute pancreatitis (AP), however, none of them contains suggestions on patient discharge. The Hungarian Pancreatic Study Group (HPSG) has recently developed a laboratory data and symptom-based discharge protocol which needs to be validated. (1) A survey was conducted involving all members of the International Association of Pancreatology (IAP) to understand the characteristics of international discharge protocols. (2) We investigated the safety and effectiveness of the HPSG-discharge protocol. According to our international survey, 87.5% (49/56) of the centres had no discharge protocol. One of the most important obstacles for taking advantage of forest resources in the Italian Alps is represented by the high level of private properties fragmentation and by their small size. Thus, there is an urgent need for tools to support single or multi-forest owners to gain reliable and updated information on their forest stands so that the proper silvicultural activities following all the existing regulations can be adopted. The present research was aimed at promoting a shared management of small private forest properties in the mountainous area of Friuli Venezia Giulia (NE Italy) through the implementation of a new WebGIS tool to support forest decisions and management at different spatial scales. This new tool was developed updating and merging together different available information sources (e.g., tree species composition, the presence of protected areas, forest roads, etc.) with ad-hoc elaborated layers (e.g., standing volume, annual increment of volume, forest accessibility, etc.), also elaborating a cost analysis related to the different possible harvesting methods. The tool allows queries at the level of either a single or a group of cadastral parcels to obtain data in a format, which can be used for filling in the planning document requested by the regional authorities.

Abstract Background In 2018, our center started a program to offer genetic diagnosis to patients with kidney and liver monogenic rare conditions, potentially eligible for organ transplantation. We exploited a clinical exome sequencing approach, followed by analyses of in silico gene panels tailored to clinical suspicions, obtaining detection rates in line with what reported in literature. However, a percentage of patients remains without a definitive genetic diagnosis. This work aims to evaluate the utility of NGS data re-analysis for those patients with an inconclusive or negative genetic test at the time of first analysis considering that (i) the advance of alignment and variant calling processes progressively improve the detection rate, limiting false positives and false negatives; (ii) gene panels are periodically updated and (iii) variant annotation may change over time. Methods 114 patients, recruited between 2018 and 2020, with an inconclusive or negative NGS report at the time of first analysis, were included in the study. Re-alignment and variant calling of previously generated sequencing raw data were performed using the GenomSys Variant Analyzer software. Results 21 previously not reported potentially causative variants were identified in 20 patients. In most cases (n = 19), causal variants were retrieved out of the re-classification from likely benign to variants of unknown significance (VUS). In one case, the variant was included because of inclusion in the analysis of a newly disease-associated gene, not present in the original gene panel, and in another one due to the improved data alignment process. Whenever possible, variants were validated with Sanger sequencing and family segregation studies. As of now, 16 out of 20 patients have been analyzed and variants confirmed in 8 patients. Specifically, in two pediatric patients, causative variants were de novo mutations while in the others, the variant was present also in other affected relatives. In the remaining patients, variants were present also in non-affected parents, raising questions on their re-classification. Conclusions Overall, these data indicate that periodic and systematic re-analysis of negative or inconclusive NGS data reports can lead to new variant identification or reclassification in a small but significant proportion of cases, with benefits for patients’ management.

Abstract We use analogues of atmospheric patterns to investigate changes in four devastating Acqua Alta (flooding) events in the lagoon of Venice associated with intense Mediterranean cyclones occurred in 1966, 2008, 2018 and 2019. Our results provide evidence that changes in atmospheric circulation, although not necessarily only anthropogenically driven, are linked to the severity of these events. We also evaluate the cost and benefit of the MoSE system, which was designed to protect against flooding. Our analysis shows that the MoSE has already provided protection against analogues of the most extreme event, which occurred in 1966. These findings have significant implications for the future of Venice and other coastal cities facing similar challenges from rising sea levels due to extreme events. This study also provides a pathway to evaluate the effectiveness of adaptation in a scenario more frequent and intense extreme events if higher global warming levels will be reached.

241. How will the Covid-19 pandemic shape the future of primary care undergraduate teaching? Understanding modifications and developments deployed by UK academic units of primary care, and their implications for the future

Abstract Background Primary care has been under-represented in its contribution to the academic literature base on Covid-19 developments. We sought to understand how teaching and learning was modified and developed by primary care academic leaders to support the continuation of primary care-orientated learning during the Covid-19 pandemic; and explore how these changes may shape future educational delivery in primary care. Methods We adopted a qualitative approach, using semi-structured interviews of seven General Practice Heads of Teaching (GP HoTs) from UK medical schools. We used mixed deductive and inductive coding to analyse interview transcripts. Modifications and developments were coded to four a priori themes (clinical off-site; clinical on-site; synchronous remote; asynchronous remote). We concurrently used inductive coding to identify developments that did not readily fit into these categories. To understand how participants perceived the developments may shape primary care teaching in the future, we carried out an inductive thematic analysis. Results A range of modifications and developments were described. Examples of developments include: GP practices being provided with increased flexibility to support ongoing provision of clinical placements (on-site clinical), examples of initiatives enabling students to consult remotely from their homes (off-site clinical), transfer of face-to-face teaching to remote formats (synchronous remote) and development of new, interactive on-line teaching materials (asynchronous remote). One additional theme arose inductively: collaboration and co-operation. For future implications, five themes arose: the evolution of flexible and hybrid clinical placement models; an increased role for telemedicine; increased networking and collaboration; increased active student involvement in patient care; and opportunities for community-based teaching afforded by the pandemic. Conclusion This study highlights how teaching was modified to support the continuation of primary care-based learning during the Covid-19 pandemic, and implications for the future. Collaboration and placement flexibility were notable features in the response. Participants perceived that flexible placement models containing a mixture of clinical on-site with remote synchronous and asynchronous teaching and learning activities, may persist into the post-Covid era. Further research is required to understand which developments become routinely embedded into primary care teaching in the post-Covid era and explain how and why this occurs.

Abstract Eutrophication usually impacts grassland biodiversity, community composition, and biomass production, but its impact on the stability of these community aspects is unclear. One challenge is that stability has many facets that can be tightly correlated (low dimensionality) or highly disparate (high dimensionality). Using standardized experiments in 55 grassland sites from a globally distributed experiment (NutNet), we quantify the effects of nutrient addition on five facets of stability (temporal invariability, resistance during dry and wet growing seasons, recovery after dry and wet growing seasons), measured on three community aspects (aboveground biomass, community composition, and species richness). Nutrient addition reduces the temporal invariability and resistance of species richness and community composition during dry and wet growing seasons, but does not affect those of biomass. Different stability measures are largely uncorrelated under both ambient and eutrophic conditions, indicating consistently high dimensionality. Harnessing the dimensionality of ecological stability provides insights for predicting grassland responses to global environmental change.

Urbanization transforms environments in ways that alter biological evolution. We examined whether urban environmental change drives parallel evolution by sampling 110,019 white clover plants from 6169 populations in 160 cities globally. Plants were assayed for a Mendelian antiherbivore defense that also affects tolerance to abiotic stressors. Urban-rural gradients were associated with the evolution of clines in defense in 47% of cities throughout the world. Variation in the strength of clines was explained by environmental changes in drought stress and vegetation cover that varied among cities. Sequencing 2074 genomes from 26 cities revealed that the evolution of urban-rural clines was best explained by adaptive evolution, but the degree of parallel adaptation varied among cities. Our results demonstrate that urbanization leads to adaptation at a global scale.

A non-equilibrium model for laser-induced plasmas is used to describe how nano-second temporal mode-beating affects plasma kernel formation and growth in quiescent air. The chemically reactive Navier-Stokes equations describe the hydrodynamics, and non-equilibrium effects are modeled based on a two-temperature model. Inverse Bremsstrahlung and multiphoton ionization are self-consistently taken into account via a coupled solution of the equations governing plasma dynamics and beam propagation and attenuation (i.e., Radiative Transfer Equation). This strategy, despite the additional challenges it may bring, allows to minimize empiricism and enables for more accurate simulations since it does not require an artificial plasma seed to trigger breakdown. The benefits of this methodology are demonstrated by the good agreement between the predicted and the experimental plasma boundary evolution and absorbed energy. The same goes for the periodic plasma kernel structures which, as suggested by experiments and confirmed by the simulations discussed here, are linked to the modulating frequency.

In this work, we consider the linear inverse problem $y=Ax+\epsilon$, where $A\colon X\to Y$ is a known linear operator between the separable Hilbert spaces $X$ and $Y$, $x$ is a random variable in $X$ and $\epsilon$ is a zero-mean random process in $Y$. This setting covers several inverse problems in imaging including denoising, deblurring, and X-ray tomography. Within the classical framework of regularization, we focus on the case where the regularization functional is not given a priori but learned from data. Our first result is a characterization of the optimal generalized Tikhonov regularizer, with respect to the mean squared error. We find that it is completely independent of the forward operator $A$ and depends only on the mean and covariance of $x$. Then, we consider the problem of learning the regularizer from a finite training set in two different frameworks: one supervised, based on samples of both $x$ and $y$, and one unsupervised, based only on samples of $x$. In both cases, we prove generalization bounds, under some weak assumptions on the distribution of $x$ and $\epsilon$, including the case of sub-Gaussian variables. Our bounds hold in infinite-dimensional spaces, thereby showing that finer and finer discretizations do not make this learning problem harder. The results are validated through numerical simulations.
