Abstract

It is now established that the faint radio population is a mixture of star-forming galaxies and faint active galactic nuclei (AGNs), with the former dominating below S(1.4GHz) \sim 100 muJy and the latter at larger flux densities. The faint radio AGN component can itself be separated into two main classes, mainly based on the host-galaxy properties: sources associated with red/early-type galaxies (like radio galaxies) are the dominant class down to \sim 100 muJy; quasar/Seyfert--like sources contribute an additional 10-20\%. One of the major open questions regarding faint radio AGNs is the physical process responsible for their radio emission. This work aims at investigating this issue, with particular respect to the AGN component associated with red/early-type galaxies. Such AGNs show, on average, flatter radio spectra than radio galaxies and are mostly compact (<= 30 kpc in size). Various scenarios have been proposed to explain their radio emission. For instance they could be core/core-jet dominated radio galaxies, low-power BL LACs, or advection-dominated accretion flow (ADAF) systems. Abstract

We report the detection of diffuse radio emission which might be connected to a large-scale filament of the cosmic web covering a 8deg x 8deg area in the sky, likely associated with a z~0.1 over-density traced by nine massive galaxy clusters. In this work, we present radio observations of this region taken with the Sardinia Radio Telescope. Two of the clusters in the field host a powerful radio halo sustained by violent ongoing mergers and provide direct proof of intra-cluster magnetic fields. In order to investigate the presence of large-scale diffuse radio synchrotron emission in and beyond the galaxy clusters in this complex system, we combined the data taken at 1.4 GHz obtained with the Sardinia Radio Telescope with higher resolution data taken with the NRAO VLA Sky Survey. We found 28 candidate new sources with a size larger and X-ray emission fainter than known diffuse large-scale synchrotron cluster sources for a given radio power. This new population is potentially the tip of the iceberg of a class of diffuse large-scale synchrotron sources associated with the filaments of the cosmic web. Abstract

Background: Among patients with advanced high-grade ovarian carcinoma (aHGOC) treated with poly (ADP-ribose) polymerase (PARP) inhibitors (PARPis), the presence of a germline BRCA pathogenic variant (gBRCA-PV) may increase the risk of bone marrow mutagenesis resulting in postcytotoxic therapy myelodysplastic neoplasms (MDS-pCT) or acute myeloid leukemia (AML-pCT), as it is expressed in heterozygosity also by hematopoietic progenitors. We aimed to investigate the occurrence of post-PARPi MDSs/AMLs-pCTs according to gBRCA-PV status. Patients and methods: We conducted a retrospective single-center study to evaluate MDS/AML-pCT in patients with aHGOC and a known gBRCA-PV status receiving at least 8 weeks of maintenance PARPi, in any line of therapy, from February 2017 to December 2022. The endpoint was the proportion of patients who experienced MDSs-pCT and AMLs-pCT during and after treatment with PARPi, in gBRCA-PV carriers and non-carriers. Results: A total of 166 patients were included: 95 (57%) had a gBRCA-PV and 72% received PARPi for recurrent disease. The number of lines of chemotherapies before and after PARPi, median overall survival, and median follow-up were comparable between gBRCA-PV carriers and non-carriers. After a median follow-up of 40.0 (95% confidence interval: 35.7-44.3) months, 10 (6%) patients were diagnosed with an MDS-pCT and 4 (2%) with an AML-pCT. A higher proportion of MDSs/AMLs-pCT (10% versus 2%; P = 0.16) and, in particular, of MDSs-pCT (9% versus 1%; P = 0.04) was observed among gBRCA-PV carriers. Conclusions: The presence of a gBRCA-PV is associated with a higher risk of MDS-pCT and possibly of myeloid neoplasms after PARPi in patients with aHGOC who received PARPi, especially in the setting of recurrent disease.

Abstract

The authors present an atypical case of an infant with unremarkable familiar, birth, and neonatal history who developed a mild/benign form of transient cholestasis. At admission, second- and third-level assessments were conducted, mitochondrial respiratory chain disorders were excluded. The patient was accurately and promptly diagnosed through a clinically driven genetic test. The genetic analysis evidenced a compound heterozygous mutation c.383A>G (p.Tyr128Cys) of maternal origin and c.835G>A (p.Val279Met) of paternal origin in the TRMU gene associated to transient infantile liver failure, a condition known for its progressive and sometimes fatal prognosis. Biochemical test and patient’s clinical evolution were both good and there was no evidence of liver failure or dysfunction of other organs. This is the first-ever reported case of a patient with a TRMU pathological compound mutation with such good clinical evolution.

Abstract

Background: Uterine leiomyosarcoma (uLMS) represents one of the most common sarcoma histotypes, demonstrating an overall dismal prognosis. Previous studies reported uLMS to carry recurrent somatic BRCA2 homozygous deletions, related to significant clinical benefits from the use of PARP inhibitors. Methods: To investigate the prevalence in uLMS of genomic alterations (alt) in BRCA2 and other homologous recombination (HR) and DNA damage response (DDR) genes, cBioPortal was accessed and data were retrieved from studies including pan-sarcoma histologies. HR-/DDR-genes included BRCA1, BRCA2, ATM, BARD1, BRIP1, CHEK1, CHEK2, FANCA, FANCB, FANCC, FANCD2, FANCE, FANCF, FANCG, FANCI, FANCL, FANCM, NBN, PALB2, RAD51C, RAD51D, RAD50, and ATR. Only oncogenic/likely oncogenic alterations were included according to OncoKB. Clinical Report and Results: We reported a clinical case of a patient affected by a highly pretreated uLMS discussed at the European Institute of Oncology Molecular Tumor Board. A targeted next-generation sequencing panel demonstrated a somatic BRCA2 homozygous deletion (homDel). Upon access to Niraparib, a remarkable response of 15 months was observed before experiencing disease progression. In the genomic query, among 2393 cases, uLMS (n = 193) displayed 9 of all 31 BRCA2alt observed, representing the only sarcoma histotype showing an enrichment in BRCA2alt (4.66%; q < 0.001). All of 9 BRCA2alt were represented by homDel, which related to a high fraction of genome altered. Conclusion: uLMS displays a significant frequency of somatic BRCA2alt homDel. Considering their dismal prognosis, further investigation is warranted to test the use of PARPi in uLMS, and particularly in the setting of BRCA1/2 alterations.

Abstract

Background: At the time of AtTEnd trial design, standard treatment for advanced or recurrent endometrial cancer included carboplatin and paclitaxel chemotherapy. This trial assessed whether combining atezolizumab with chemotherapy might improve outcomes in this population. Methods: AtTEnd was a multicentre, double-blind, randomised, placebo-controlled, phase 3 trial done in 89 hospitals in 11 countries across Europe, Australia, New Zealand, and Asia. Enrolled patients were aged 18 years or older, and had advanced or recurrent endometrial carcinoma or carcinosarcoma, an Eastern Cooperative Oncology Group performance status of 0–2, and received no previous systemic chemotherapy for recurrence. Patients were randomly assigned (2:1) using an interactive web response system (block size of six) to either atezolizumab 1200 mg or placebo given intravenously with chemotherapy (carboplatin at area under the curve of 5 or 6 and paclitaxel 175 mg/m2 intravenously on day 1 every 21 days) for 6–8 cycles, then continued until progression. Stratification factors were country, histological subtype, advanced or recurrent status, and mismatch repair (MMR) status. Participants and treating clinicians were masked to group allocation. The hierarchically tested co-primary endpoints were progression-free survival (in patients with MMR-deficient [dMMR] tumours, and in the overall population) and overall survival (in the overall population). Primary analyses were done in the intention-to-treat population, defined as all randomly assigned patients who gave their full consent to participation in the study and data processing. Safety was assessed in all patients included in the intention-to-treat population who received at least one dose of study treatment. Abstract

Alzheimer’s disease (AD) is a deadly brain degenerative disorder that leads to brain shrinkage and dementia. AD is manifested with hyperphosphorylated tau protein levels and amyloid beta (Aβ) peptide buildup in the hippocampus and cortex regions of the brain. The nervous tissue of AD patients also contains fungal proteins and DNA which are linked to bacterial infections, suggesting that polymicrobial infections also occur in the brains of those with AD. Both immunohistochemistry and next-generation sequencing (NGS) techniques were employed to assess fungal and bacterial infections in the brain tissue of AD patients and non-AD controls, with the most prevalent fungus genera detected in AD patients being Alternaria, Botrytis, Candida, and Malassezia. Interestingly, Fusarium was the most common genus detected in the control group. Both AD patients and controls were also detectable for Proteobacteria, followed by Firmicutes, Actinobacteria, and Bacteroides for bacterial infection. At the family level, Burkholderiaceae and Staphylococcaceae exhibited higher levels in the brains of those with AD than the brains of the control group. Accordingly, there is thought to be a viscous cycle of uncontrolled neuroinflammation and neurodegeneration in the brain, caused by agents such as the herpes simplex virus type 1 (HSV1), Chlamydophilapneumonia, and Spirochetes, and the presence of apolipoprotein E4 (APOE4), which is associated with an increased proinflammatory response in the immune system. Systemic proinflammatory cytokines are produced by microorganisms such as Cytomegalovirus, Helicobacter pylori, and those related to periodontal infections. These can then cross the blood–brain barrier (BBB) and lead to the onset of dementia. Here, we reviewed the relationship between the etiology of AD and microorganisms (such as bacterial pathogens, Herpesviridae viruses, and periodontal pathogens) according to the evidence available to understand the pathogenesis of AD. These findings might guide a targeted anti-inflammatory therapeutic approach to AD.

Abstract

A 12-month prospective consecutive case series study was conducted to evaluate the effectiveness of a novel porcine collagen matrix to correct moderate to severe buccogingival recession defects. A total of 10 healthy patients (8 women and 2 men; age range: 30 to 68 years) with 26 maxillary and mandibular recession gingival defects > 4 mm deep were included. Healthy maturation of gingival tissues with natural color and texture matching the adjacent soft tissue areas were observed at all reevaluation visits. Complete root coverage was not achieved in all cases, possibly due to severe buccal bone loss in most of the selected cases, which negatively impacted the results. Nevertheless, when a novel porcine collagen matrix was used, a mean root coverage of 63.15% was achieved, and gains in clinical attachment level and keratinized tissue height were seen. Future randomized clinical trials are needed to further explore the potential of porcine collagen matrix for treatment of localized gingival recession defects. [ABSTRACT FROM AUTHOR]

244. Factors influencing experiences of non-consensual sex: results from a mixed data cross-sectional online survey of Australian university students.

Abstract

Background: The high prevalence of non-consensual sex, including physical force, verbal threats, intimidation, and/or verbal coercion and rape among university students, has prompted urgent calls for action from governments, statutory, and university bodies. This research aims to identify key factors students see as contributing to non-consensual sex with a view to developing effective strategies to address these issues. Methods: An online cross-sectional survey was administered to 4291 university students attending universities in south-east Queensland, Australia. Participants were recruited via email, print media, and face-to-face invitations. The survey contained closed and open-ended questions on a range of sexual health knowledge, attitudes, and behaviours, including non-consensual sexual experiences. A subset of students (n = 199) who responded 'yes' to engaging in sexual acts against their consent were the primary participants for this paper. Descriptive statistical analysis and thematic analysis, employing the theory of planned behaviour, were conducted using the quantitative and qualitative data regarding non-consensual sex, respectively. Results: Immediate and longer-term fear and partner eagerness were the most commonly identified influences on non-consensual sex. An inability to give consent due to lack of consciousness or intoxication were also frequently cited factors contributing to non-consensual sex. Conclusions: Interventions acknowledging and addressing sociocultural factors are needed. Sexuality and relationship education encompassing effective communication, relationship boundaries, and safe use of alcohol and drugs could substantially reduce the incidence of non-consensual sex. Importantly, such education should be non-judgmental and place greater responsibility on perpetrators than victim-survivors. Recent studies demonstrate the concerning extent of sexual violence experienced by Australian university students and the resulting psychological, emotional, and physical harms. This study aims to understand experiences and contributing factors of non-consensual sex among university students in south-east Queensland, Australia. Findings demonstrate that interventions to reduce non-consensual sex and promote healthy sexual experiences need to reflect the complex, relational, and highly contextual nature of affirmative consent influenced by perceived behavioural control and subjective norms. Abstract

Neural machine translation (NMT) is a sophisticated technique that employs a large, singular neural network to learn and execute automatic translation tasks. Unlike statistical machine translation systems, NMT handles the entire translation process in an end-to-end manner, removing the need for additional components. This approach has shown significant promise in translation quality and has become the prevalent method. In this study, we apply sparsely factored NMT to English and several Indo-Aryan (Hindi, Bengali) and Dravidian (Tamil, Malayalam) language pairs. Specifically, we develop the machine translation system using an attention-based mechanism. A significant problem with traditional transformers is the huge memory requirement. Therefore, a sparsely factored NMT (SFNMT) is used to reduce the memory requirement but also improves the training time, thereby, reducing the computing time. In this paper, take inspiration from Vaswani transformer and modify it to get the best results. The system's performance was evaluated using the BLEU metric. The proposed model indtrl achieves a BLUE score of 32.13 (en→hi), 29.31 (en→be), 31.21 (en→ta), 21.12 (en→ml) and 32.67 (en→hi), 29.38 (en→be), 31.75 (en→ta), 21.17 (en→ml) without backtranslation and with backtranslation. To evaluate the performance of the system, we compared the results with those of existing systems. Abstract

This paper introduces an LLM-Enhanced Agent-Based Influence Diffusion Simulation (LLM-AIDSim) framework that integrates large language models (LLMs) into agent-based modelling to simulate influence diffusion in social networks. The proposed framework enhances traditional influence diffusion models by allowing agents to generate language-level responses, providing deeper insights into user agent interactions. Our framework addresses the limitations of probabilistic models by simulating realistic, context-aware user behaviours in response to public statements. Using real-world news topics, we demonstrate the effectiveness of LLM-AIDSim in simulating topic evolution and tracking user discourse, validating its ability to replicate key aspects of real-world information propagation. Our experimental results highlight the role of influence diffusion in shaping collective discussions, revealing that, over time, diffusion narrows the focus of conversations around a few dominant topics. We further analyse regional differences in topic clustering and diffusion behaviours across three cities, Sydney, Auckland, and Hobart, revealing how demographics, income, and education levels influence topic dominance. This work underscores the potential of LLM-AIDSim as a decision-support tool for strategic communication, enabling organizations to anticipate and understand public sentiment trends. Abstract

We observed the galaxy cluster CIZA J2242.8+5301 with the Sardinia Radio Telescope to provide new constraints on its spectral properties at high frequency. We conducted observations in three frequency bands centred at 1.4 GHz, 6.6 GHz and 19 GHz, resulting in beam resolutions of 14$^{\prime}$, 2.9$^{\prime}$ and 1$^{\prime}$ respectively. These single-dish data were also combined with archival interferometric observations at 1.4 and 1.7 GHz. From the combined images, we measured a flux density of ${\rm S_{1.4GHz}=(158.3\pm9.6)\,mJy}$ for the central radio halo and ${\rm S_{1.4GHz}=(126\pm8)\,mJy}$ and ${\rm S_{1.4GHz}=(11.7\pm0.7)\,mJy}$ for the northern and the southern relic respectively. After the spectral modelling of the discrete sources, we measured at 6.6 GHz ${\rm S_{6.6GHz}=(17.1\pm1.2)\,mJy}$ and ${\rm S_{6.6GHz}=(0.6\pm0.3)\,mJy}$ for the northern and southern relic respectively. Assuming simple diffusive shock acceleration, we interpret measurements of the northern relic with a continuous injection model represented by a broken power-law. This yields an injection spectral index ${\rm \alpha_{inj}=0.7\pm0.1}$ and a Mach number ${\rm M=3.3\pm0.9}$, consistent with recent X-ray estimates. Unlike other studies of the same object, no significant steepening of the relic radio emission is seen in data up to 8.35 GHz. By fitting the southern relic spectrum with a simple power-law (${\rm S_{\nu}\propto\nu^{-\alpha}}$) we obtained a spectral index ${\rm \alpha\approx1.9}$ corresponding to a Mach number (${\rm M\approx1.8}$) in agreement with X-ray estimates. Finally, we evaluated the rotation measure of the northern relic at 6.6 GHz.
