Background Endometrial cancer (EC) is the 8* leading cause of cancer death amongst American women. Most ECs are endometrioid, serous, or clear cell carcinomas, or an admixture of histologies. Serous and clear ECs are clinically aggressive tumors for which alternative therapeutic approaches are needed. The purpose of this study was to search for somatic mutations in the tyrosine kinome of serous and clear cell ECs, because mutated kinases can point to potential therapeutic targets. Methods In a mutation discovery screen, we PCR amplified and Sanger sequenced the exons encoding the catalytic domains of 86 tyrosine kinases from 24 serous, 11 clear cell, and 5 mixed histology ECs. For somatically mutated genes, we next sequenced the remaining coding exons from the 40 discovery screen tumors and sequenced all coding exons from another 72 ECs (10 clear cell, 21 serous, 41 endometrioid). We assessed the copy number of mutated kinases in this cohort of 112 tumors using quantitative real time PCR, and we used immunoblotting to measure expression of these kinases in endometrial cancer cell lines. Results Overall, we identified somatic mutations in TNK2 (tyrosine kinase non-receptor, 2) and DDR1 (discoidin domain receptor tyrosine kinase 1) in 5.3% (6 of 112) and 2.7% (3 of 112) of ECs. Copy number gains of TNK2 and DDR1 were identified in another 4.5% and 0.9% of 112 cases respectively. Immunoblotting confirmed TNK2 and DDR1 expression in endometrial cancer cell lines. Three of five missense mutations in TNK2 and one of two missense mutations in DDR1 are predicted to impact protein function by two or more in silico algorithms. The TNK2P761Rfs*72 frameshift mutation was recurrent in EC, and the DDR1R570Q missense mutation was recurrent across tumor types. Conclusions This is the first study to systematically search for mutations in the tyrosine kinome in clear cell endometrial tumors. Our findings indicate that high-frequency somatic mutations in the catalytic domains of the tyrosine kinome are rare in clear cell ECs. We uncovered ten new mutations in TNK2 and DDR1 within serous and endometrioid ECs, thus providing novel insights into the mutation spectrum of each gene in EC. [ABSTRACT FROM AUTHOR]

In the United States, there is no comprehensive energy policy at the federal level. To address issues as diverse as climate change, energy security, and economic development, individual states have increasingly implemented Renewable Portfolio Standards (RPSs), which mandate that utility providers include a specified amount of electricity from renewable energy sources in their total energy portfolios. Some states have included incentives for individual energy technologies in their RPS, such as solar electric (also called photovoltaic or PV technology). Here, we use GIS to visualize adoption of RPSs and electricity generation from renewable energy sources in the US and examine changes in renewable electricity and solar electric generation over time with the goal of informing future policies aimed at promoting the adoption of renewable energy technologies. This case study describes how behavioural activation was used to treat a client with depression who had undergone previous psychological treatments including cognitive behaviour therapy. After assessment and formulation of problems and goals, a treatment plan was structured to increase contact with positive activities and previously avoided situations, people and pursuits. Collaborative recovery-orientated work on this plan resulted in improvements confirmed by self-rating scores and subjective reports. Conservation organizations rely on conservation easements for diverse purposes, including protection of species and natural communities, working forests, and open space. This research investigated how perpetual conservation easements incorporated property rights, responsibilities, and options for change over time in land management. We compared 34 conservation easements held by one federal, three state, and four nonprofit organizations in Wisconsin. They incorporated six mechanisms for ongoing land management decision-making: management plans (74 %), modifications to permitted landowner uses with discretionary consent (65 %), amendment clauses (53 %), easement holder rights to conduct land management (50 %), reference to laws or policies as compliance terms (47 %), and conditional use permits (12 %). Easements with purposes to protect species and natural communities had more ecological monitoring rights, organizational control over land management, and mechanisms for change than easements with general open space purposes. Forestry purposes were associated with mechanisms for change but not necessarily with ecological monitoring rights or organizational control over land management. The Natural Resources Conservation Service-Wetland Reserve Program had a particularly consistent approach with high control over land use and some discretion to modify uses through permits. Conservation staff perceived a need to respond to changing social and ecological conditions but were divided on whether climate change was likely to negatively impact their conservation easements. Many conservation easements involved significant constraints on easement holders' options for altering land management to achieve conservation purposes over time. This study suggests the need for greater attention to easement drafting, monitoring, and ongoing decision processes to ensure the public benefits of land conservation in changing landscapes. Endometrial cancer is the sixth most commonly diagnosed cancer in women worldwide, causing ?74,000 deaths annually. Serous endometrial cancers are a clinically aggressive subtype with a poorly defined genetic etiology. We used whole-exome sequencing to comprehensively search for somatic mutations within ?22,000 protein-encoding genes in 13 primary serous endometrial tumors. We subsequently resequenced 18 genes, which were mutated in more than 1 tumor and/or were components of an enriched functional grouping, from 40 additional serous tumors. We identified high frequencies of somatic mutations in CHD4 (17%), EP300 (8%), ARID1A (6%), TSPYL2 (6%), FBXW7 (29%), SPOP (8%), MAP3K4 (6%) and ABCC9 (6%). Overall, 36.5% of serous tumors had a mutated chromatin-remodeling gene, and 35% had a mutated ubiquitin ligase complex gene, implicating frequent mutational disruption of these processes in the molecular pathogenesis of one of the deadliest forms of endometrial cancer. People who are prone to depression have a tendency to hold negative core beliefs about themselves, others and the future. They may develop assumptions, rules and compensatory strategies to protect themselves from these difficult beliefs. When these no longer work, depression may occur. Cognitive behaviour therapy enables client and therapist to work on cycles of thought and behaviour that perpetuate low mood. In three experiments, we investigated the cognitive effects of linguistic prominence to establish whether focus plays a similar or different role in modulating language processing in healthy ageing. Information structuring through the use of cleft sentences is known to increase the processing efficiency of anaphoric references to elements contained with a marked focus structure. It also protects these elements from becoming suppressed in the wake of subsequent information, suggesting selective mechanisms of enhancement and suppression. In Experiment 1 (using self-paced reading), we found that focus enhanced (faster) integration for anaphors referring to words contained within the scope of focus; but suppressed (slower) integration for anaphors to words contained outside of the scope of focus; and in some cases, the effects were larger in older adults. In Experiment 2 (using change detection), we showed that older adults relied more on the linguistic structure to enhance change detection when the changed word was in focus. In Experiment 3 (using delayed probe recognition and eye-tracking), we found that older adults recognized probes more accurately when they were made to elements within the scope of focus than when they were outside the scope of focus. Although previous research indicates that audio computer-assisted self-interviewing (ACASI) yields higher reports of threatening behaviors than interviewer-administered interviews, very few studies have examined the potential effect of the gender of the ACASI voice on survey reports. Because the voice in ACASI necessarily has a gender, it is important to understand whether using a voice that is perceived as male or female might further enhance the validity associated with ACASI. This study examines gender-of-voice effects for a set of questions about sensitive behaviors administered via ACASI to a sample of young adults at high risk for engaging in the behaviors. Results showed higher levels of engagement in the behaviors and more consistent reporting among males when responding to a female voice, indicating that males were potentially more accurate when reporting to the female voice. Reports by females were not influenced by the voice's gender. Our analysis adds to research on gender-of-voice effects in surveys, with important findings on measuring sensitive behaviors among young adults. [ABSTRACT FROM AUTHOR]

Summary 1. Food web theory hypothesizes that trophic interaction strengths of consumers should vary with consumer metabolic body mass (mass0·75) rather than simply with consumer body mass (mass1·0) owing to constraints on consumption imposed by metabolic demand for and metabolic capacity to process nutrients and energy. Accordingly, species with similar metabolic body masses should have similar trophic interaction strengths. 2. We experimentally tested this hypothesis by assembling food webs comprised of species of arthropod predators, small sap-feeding and large leaf-chewing insect herbivores and herbaceous plants in a New England, USA meadow grassland. The experiment comprised of a density-matching treatment where herbivore species were stocked into field mesocosms at equal densities to quantify baseline species identity and metabolic body mass effects. The experiment also comprised of a metabolic biomass-matching treatment where smaller sap-feeding herbivore (SH) species were stocked into mesocosms such that the product of their density and metabolic body mass (metabolic biomass) was equal to the large herbivore (LH) species. We compared the magnitude of the direct effects of herbivore species on plants in the different treatments. We also compared the magnitude of indirect effects between predators and plants mediated by herbivores in the different treatments. 3. Consistent with the hypothesis, we found that increasing metabolic biomass translated into a 9-14-fold increase in magnitude of herbivore direct effects and up to a fivefold increase in indirect effects on plants. Moreover, metabolic biomass matching caused interaction strengths among herbivore species to converge. This result came about through increases in the herbivore mean effects as well as decreases in variation in effects among treatment replicates as herbivore metabolic biomass increased. 4. We found, however, that herbivore feeding mode rather than herbivore metabolic biomass explained differences in the sign of indirect effects in the different food webs. 5. We conclude that increasing herbivore metabolic biomass not only strengthened the direct and indirect effects on plants but also made those effects more consistent across space. Nevertheless, metabolic biomass alone could not completely explain variation in the nature of indirect effects in the food web, suggesting that additional consideration of consumer traits like feeding mode will provide a more nuanced understanding of trophic interaction strengths in food webs. By taking on the roles of observer, client and co-therapist in cognitive behaviour therapy (CBT) workshops, trainee psychological therapists can practise CBT methods on themselves. By doing so, they can gain insights into how they think and how their clients may respond to therapy. This article is a personal account of the author's experiences of such workshops, written in the first person to add immediacy (Webb 1992, Hamill 1999), that focuses on a specific aspect of her practice that she found difficult. ATAD5, the human ortholog of yeast Elg1, plays a role in PCNA deubiquitination. Since PCNA modification is important to regulate DNA damage bypass, ATAD5 may be important for suppression of genomic instability in mammals in vivo. To test this hypothesis, we generated heterozygous (Atad5+/m) mice that were haploinsuffficient for Atad5. Atad5+/m mice displayed high levels of genomic instability in vivo, and Atad5+/m mouse embryonic fibroblasts (MEFs) exhibited molecular defects in PCNA deubiquitination in response to DNA damage, as well as DNA damage hypersensitivity and high levels of genomic instability, apoptosis, and aneuploidy. Importantly, 90% of haploinsufficient Atad5 +/m mice developed tumors, including sarcomas, carcinomas, and adenocarcinomas, between 11 and 20 months of age. High levels of genomic alterations were evident in tumors that arose in the Atad5 +/m mice. Consistent with a role for Atad5 in suppressing tumorigenesis, we also identified somatic mutations of ATAD5 in 4.6% of sporadic human endometrial tumors, including two nonsense mutations that resulted in loss of proper ATAD5 function. The purpose of this qualitative descriptive study was to explore self-care production experiences in older African Americans who, despite some cognitive dysfunction, were able to produce hypertension-related self-care behaviors or blood pressure control successfully. Participants were 10 urban, community-dwelling older African Americans, 60-89 years of age, living in a Midwest region of the United States. A semi-structured interview was conducted in participants homes' using Kvale's 'conversational discourse' approach. Oral recordings were transcribed and analyzed for themes and codes. Elders' experiences with the production of self-care were characterized by three themes: preparation, monitoring, and evaluation. Self-care production was found to be cognitively challenging consistent with the finding that 60% of the participants had difficulty with a cognitive task requiring complex cognitive skills. This finding may explain why the production of self-care became a social phenomenon in which elders demonstrated resourcefulness in seeking assistance from surrounding support systems. Nurse practitioners can support better health outcomes in older adults with hypertension by using valid and reliable measures for assessing complex cognitive skills, assessing individuals' progress in self-care production, and identifying individuals' use of social and professional supports to produce self-care. Background: Medication errors commonly involve confusion between drugs with similar names. One possible method of reducing error is to emphasize differences between the names using 'Tall Man' (uppercase) letters (e.g. cefTAZidime vs cefUROxime). Previous studies investigating this issue have been conducted mainly on university students, and results have been mixed.Objective: To investigate the influence of Tall Man lettering on drug name confusion in other key participant groups.Study Design: Two separate experiments were conducted. In Experiment 1 (conducted at the University of Glasgow, Scotland, between January 2008 and May 2008), younger and older adults performed a same/different judgement task. In Experiment 2 (conducted at various sites in England between December 2008 and February 2009), healthcare practitioners performed a task based on electronic prescribing.Results: In Experiment 1, both younger and older adults made fewer name confusion errors when names contained Tall Man letters. Response times suggested that Tall Man lettering drew participants' attention to those letters, but that readers did not solely rely on these letters in making their response. In Experiment 2, healthcare practitioners made fewer name confusion errors when the names contained Tall Man letters.Conclusions: Overall, results showed that Tall Man lettering reduced drug name confusion errors in a series of laboratory-based tasks, in both younger and older adults, and healthcare practitioners. Thus, the current findings offer some support for the use of Tall Man letters as a possible systems change that could be made by both pharmacies and manufacturers in an effort to reduce error caused by drug name confusion. Information structuring through the use of cleft sentences increases the processing efficiency of references to elements within the scope of focus. Furthermore, there is evidence that putting certain types of emphasis on individual words not only enhances their subsequent processing, but also protects these words from becoming suppressed in the wake of subsequent information, suggesting mechanisms of enhancement and suppression. In Experiment 1, we showed that clefted constructions facilitate the integration of subsequent sentences that make reference to elements within the scope of focus, and that they decrease the efficiency with reference to elements outside of the scope of focus. In Experiment 2, using an auditory text-change-detection paradigm, we showed that focus has similar effects on the strength of memory representations. These results add to the evidence for enhancement and suppression as mechanisms of sentence processing and clarify that the effects occur within sentences having a marked focus structure. This article focuses on family processes and adolescent religious attendance and personal religiosity. We find that the closeness and quality of the marital relationship and relationship between adolescent and parents significantly contributes to the strength of adolescent religious conviction and practice. The study used data from the NLSY97 cohort. Predictors include parenting style, closeness, and parent-child closeness; family structure; income, employment, parental education, mother's age at first birth, and number of siblings; adolescent characteristics (e.g., age, gender, race/ethnicity, disability, lying or cheating); and environmental characteristics (e.g., region of country, urbanicity, and physical environment risk). Family religious attendance was dramatically influenced by race in adolescents aged 16 years. Adolescents living with married, biological parents in 1997 were 36% more likely to attend worship services than those living with stepfamilies. Adolescents living in more physically risky environments, with peers who belonged to gangs, cut classes, or had sex, were less likely to attend weekly worship services with their families. Finally, compared with adolescents whose parents had a high-quality marital relationship and who had good relationships with both parents, all other adolescents were less likely to attend weekly worship services with their families. Despite available diagnostic tests and recent advances, diagnosis of pulmonary invasive aspergillosis (IPA) remains challenging. We performed a longitudinal case-control pilot study to identify host-specific, novel, and immune-relevant molecular candidates indicating IPA in patients post allogeneic stem cell transplantation (alloSCT). Supported by differential gene expression analysis of six relevant in vitro studies, we conducted RNA sequencing of three alloSCT patients categorized as probable IPA cases and their matched controls without Aspergillus infection (66 samples in total). We additionally performed immunoassay analysis for all patient samples to gain a multi-omics perspective. Profiling analysis suggested LGALS2, MMP1, IL-8, and caspase-3 as potential host molecular candidates indicating IPA in investigated alloSCT patients. MMP1, IL-8, and caspase-3 were evaluated further in alloSCT patients for their potential to differentiate possible IPA cases and patients suffering from COVID-19-associated pulmonary aspergillosis (CAPA) and appropriate control patients. Possible IPA cases showed differences in IL-8 and caspase-3 serum levels compared with matched controls. Furthermore, we observed significant differences in IL-8 and caspase-3 levels among CAPA patients compared with control patients. With our conceptual work, we demonstrate the potential value of considering the human immune response during Aspergillus infection to identify immune-relevant molecular candidates indicating IPA in alloSCT patients. Routine infant immunization with live, oral rotavirus vaccines (LORVs) has had a major impact on severe gastroenteritis disease. Nevertheless, in high morbidity and mortality settings rotavirus remains an important cause of disease, partly attributable to the sub-optimal clinical efficacy of LORVs in those settings. Regardless of the precise immunological mechanism(s) underlying the diminished efficacy, the introduction of injectable next-generation rotavirus vaccines (iNGRV), currently in clinical development, could offer a potent remedy. In addition to the potential for greater clinical efficacy, precisely how iNGRVs are delivered (multiple doses to young infants; alongside LORVs or as a booster; co-formulated with Diphtheria-Tetanus-Pertussis (DTP)-containing vaccines), their pricing, and their storage and cold chain characteristics could each have major implications on the resultant health outcomes, on cost-effectiveness as well as on product preferences by national stakeholders and healthcare providers. To better understand these implications, we critically assessed whether there is a compelling public health value proposition for iNGRVs based on potential (but still hypothetical) vaccine profiles. Our results suggest that the answer is highly dependent on the specific use cases and potential attributes of such novel vaccines. Notably, co-formulation of iNGRVs with similar or greater efficacy than LORVs with a DTP-containing vaccine, such as DTP-Hib-HepB, scored especially high on potential impact, cost-effectiveness, and strength of preference by national stakeholders and health care providers in lower and middle income countries. The article examines the description of law-trained readers in first-year legal writing textbooks in the U.S. It points out that the textbooks are heavily inclined to over-generalizations. It notes that the textbooks tend to suggest that most lawyers are extraordinarily impatient, aggressively critical and formalistic in outlook. A book by Yang Costello argued that the simplest and most significant step to reduce identity dissonance among lawyers is to acknowledge that this problem exists.

Release rates of a blend of monoterpenes (spruce blend) and ethanol significantly affected mean trap catch of Tetropium fuscum (F.), Tetropium castaneum L., and Tetropium cinnamopterum Kirby. Addition of an ethanol lure to traps baited with the spruce blend lure was necessary to attract T. castaneuin and T. cinnamopterum and significantly increased attraction of T. fuscum. The combination of spruce blend and ethanol at high release rates had the highest mean catch of Tetropium spp. and was the only lure treatment that resulted in capture of T. fuscum and T. castaneum (in Poland) in every test block, suggesting it would be the best for detection surveys among the lures tested. The effect of trap design on mean catch of T. fuscum was inconsistent. In one experiment, the larger collapsible cross-vane Colossus trap caught about twice as many beetles as the IPM-Intercept trap, but in two other experiments, mean catch did not differ significantly. Type of killing agent in the collecting bucket significantly affected mean catch of T. fuscum. Traps with liquid killing agent (50/50 mixture of propylene glycol and deionized water plus 0.5 ml/liter of Kodak Photo-Flo 200 and 12.5 mg/liter of Bitrex) in the collecting bucket caught more beetles than traps with an insecticidal (dichlorvos) strip. Although any of cross-vane traps tested seem suitable for trapping several cerambycid species, the Colossus trap with liquid killing agent is recommended for use as a detection tool for T. fuscum because it caught similar or greater numbers than the other trap types. The article focuses on patient safety and links patient and family-centered care. At the 6th Annual National Patient Safety Foundation Congress held in Boston, Massachusetts, a presentation by nursing leaders and a patient from the Dana-Farber Cancer Institute challenged conference participants to think more broadly about how they could incorporate patients and families into their patient safety programs. A patient and family-centered model of care is based on mutually beneficial partnerships among health care providers, patients, and families and has profound implications for the planning, delivery and evaluation of care.

The state of New York has ambitious mandates for reducing greenhouse gas emissions and increasing renewable energy generation. Solar energy will play an important role in reducing greenhouse gas emissions from the electric energy sector. Concerns over solar installations' impacts to host communities and the environment have led to growing conflicts over solar energy siting on Long Island, in other parts of New York, and throughout the US. Understanding community members' perspectives is critical for reducing conflict. Solar energy can be deployed more quickly and at lower cost if projects are structured to address the concerns and meet the needs of the community. This paper presents the results of a survey of residential utility ratepayers that examined their perceptions, preferences, and priorities concerning mid- to large-scale solar development on Long Island (250 kW and larger). The survey asked respondents to consider specific installation types, financial models, and other aspects of solar development. Results indicate that respondents were overwhelmingly supportive of mid- to large-scale solar development in their communities. The most highly supported development types were solar systems on rooftops and solar systems that are co-located with other land uses (mixed use) at a particular site, such as parking canopies, landfills, or integration with agriculture. The most highly supported financial models included privately funded projects by local developers and community solar projects. The largest concern about solar development expressed by respondents did not involve tree removal or visibility (as initially hypothesized to be the most significant considerations) but rather the fairness of the distribution of economic benefits associated with solar development. The Long Island Solar Roadmap Project involves a collaborative research approach with multiple organizational entities and actors engaged in a stakeholder driven process. Long Island is a space-constrained region with a steep urban to rural gradient, resulting in a complex suite of local governments, development priorities, and utility, planning, and development actors. This project is integrating technological, economic, and social data into a spatial planning output that allows decision makers to see where mid-to-large scale solar development (capacity of 250 kW and larger) is technically, economically, and socially feasible. This spatial output involves innovative methods of evaluating site suitability based on criteria developed by stakeholders. The project's stakeholder and partnership driven approach allow the team to consider technological and economic feasibility across a wide variety of solar development forms and financial models. Social science data collected via a residential electric utility ratepayer survey is used to examine the perceptual barriers and opportunities for solar development as well as the sites and types of solar development that community members are most likely to support. Fungal diseases represent a considerable global health concern, affecting >1 billion people annually. In response to this growing challenge, the World Health Organization introduced the pivotal fungal priority pathogens list (FPPL) in late 2022. The FPPL highlights the challenges in estimating the global burden of fungal diseases and antifungal resistance (AFR), as well as limited surveillance capabilities and lack of routine AFR testing. Furthermore, training programs should incorporate sufficient information on fungal diseases, necessitating global advocacy to educate health care professionals and scientists. Established international guidelines and the FPPL are vital in strengthening local guidance on tackling fungal diseases. Future iterations of the FPPL have the potential to refine the list further, addressing its limitations and advancing our collective ability to combat fungal diseases effectively.
